Infectious plant diseases are a major threat to global agricultural productivity, economic development, and ecological integrity. There is widespread concern that these social and natural disasters caused by infectious plant diseases may escalate with climate change and computer modeling offers a unique opportu-nity to address this concern. Here, we analyze the intrinsic problems associated with current modeling strategies and highlight the need to integrate evolutionary principles into polytrophic, eco-evolutionary frameworks to improve predictions. We particularly discuss how evolutionary shifts in functional trade-offs, relative adaptability between plants and pathogens, ecosystems, and climate preferences induced by climate change may feedback to future plant disease epidemics and how technological advances can facilitate the generation and integration of this relevant knowledge for better modeling predictions.

The Chinese iron pan can function as a nonstick pan even without a polytetrafluoroethylene (PTFE) coating after a “Kitchen God blessing” seasoning process. We simulate this process and disclose the science behind the “Kitchen God blessing,” finding that through repeated oil-coating and heating, the reversible insertion and extraction of oxygen atoms split the surface of the iron pan, gradually producing Fe3O4 nanoballs. These balls give the iron pan a conditional hydrophobicity property, meaning the pan would be hydrophilic when the ingredients contain much water and hydrophobic when they contain less water. The former enables heat to be transferred rapidly through the nanoballs while the latter slows down the heat transference and prevents the pan from sticking. This discovery provides an approach of generating nanoballs on the surface of the metal and also discloses the secret of the fantastic taste produced by cooking with a Chinese iron pan.

Herein, a multistage induced electric field (IEF) combined with a continuous-flow reactor was utilized to assist the acid hydrolysis of corn, potato, and waxy corn starch for avoiding plate corrosion and heavy metal leakage. It was found that adding IEF stages was beneficial to improve the hydrolysis efficiency. Treating potato, corn, and waxy corn starch via continuous-flow IEF increased the reducing sugar contents up to 78.76 %, 57.86 %, and 66.18 %, respectively. The electrical conductivity of starch grew with the reaction stages, while starch yield demonstrated the opposite trend. Treated starch had higher solubility and gelatinization peak temperature than native starch, with the gelatinization enthalpy showing fluctuations. Meanwhile, the swelling power decreased as the number of IEF stages was increased. Observations of Fourier transform infrared spectroscopy, X-ray diffraction, and scanning electron microscopy indicated that the treated starch became more ordered, and crystalline regions were destroyed to various degrees with pores forming on particle surfaces. These variations could be attributed to acid hydrolysis and IEF.

The DNA damage response (DDR) pathway generally protects against genome instability, and defects in DDR have been exploited therapeutically in cancer treatment. We have reported that histone demethylase PHF8 demethylates TOPBP1 K118 mono-methylation (K118me1) to drive the activation of ATR kinase, one of the master regulators of replication stress. However, whether dysregulation of this physiological signalling is involved in tumorigenesis remains unknown. Here, we showed PHF8-promoted TOPBP1 demethylation is clinically associated with breast tumorigenesis and patient survival. Mammary gland tumors from Phf8 knockout mice grow slowly and exhibit higher level of K118me1, lower ATR activity, and increased chromosomal instability. Importantly, we found that disruption of PHF8-TOPBP1 axis suppresses breast tumorigenesis and creates a breast tumor-specific vulnerability to PARP inhibitor (PARPi) and platinum drug. CRISPR/Cas9 mutation modelling of the deleted or truncated mutation of PHF8 in clinical tumor samples demonstrated breast tumor cells expressing the mimetic variants are more vulnerable to PARPi. Together, our study supports the pursuit of PHF8-TOPBP1 signalling pathway as promising avenues for targeted therapies of PHF8-TOPBP1 proficient tumors, and provides proof-of-concept evidence for loss-of-function of PHF8 as a therapeutic indicator of PARPis.

To evaluate the association between proton pump inhibitor (PPI) use and in-hospital gastrointestinal (GI) bleeding in patients with acute coronary syndrome (ACS) taking dual antiplatelet therapy (DAPT).This study is based on the Improving Care for Cardiovascular Disease in China-ACS project, an ongoing collaborative registry and quality improvement project of the American Heart Association and the Chinese Society of Cardiology. A total of 25,567 patients with ACS taking DAPT from 172 hospitals from July 1, 2017, through December 31, 2018, were included. Multivariable Cox regression and propensity score-matched analyses were used to evaluate the association between PPI use and in-hospital GI bleeding.Of these patients with ACS, 63.9% (n=16,332) were prescribed PPIs within 24 hours of admission. Patients using PPIs had a higher rate of GI bleeding compared with those not using PPIs (1.0% vs 0.5%; P.001). In the multivariable Cox regression analysis, early PPI use was associated with a 58% higher risk of GI bleeding (hazard ratio, 1.58; 95% CI, 1.15 to 2.18; P=.005). Further propensity score matching attenuated the association but still showed that patients using PPIs had a higher rate of GI bleeding (0.8% vs 0.6%; P=.04).In China, PPIs are widely used within 24 hours of admission in patients with ACS taking DAPT. An increased risk of GI bleeding is observed in inpatients with early PPI use. Randomized trials on early use of PPIs in patients with ACS receiving DAPT are warranted.clinicaltrials.gov Identifier: NCT02306616.

Reader engagement has often been explored from a metadiscourse perspective, but little research has been conducted on the variation in engagement across participants, especially readers. Based on data from a collection of 120 Chinese letters of advice from governments and hospitals produced during the COVID-19 pandemic, a cross-interactant analysis of metadiscourse is conducted in this paper, and the interactants’ impacts on reader engagement are discussed. The findings reveal that (1) both governments and hospitals utilize engagement markers, boosters, attitude markers, and frame markers more than other types of metadiscourse items; (2) differences exist in the use of metadiscourse among the issuing agencies; (3) as reader identity varies from ingroup to outgroup membership, the probability of booster use by a hospital is higher than that by a government; and (4) the likelihood of using frame, transition and attitude markers is higher for governments than for hospitals. The paper concludes by discussing different communicative styles adopted by two agencies in a crisis context.

This study aims to investigate the effect of eccentric load on the seismic and fracture behavior of Q235B steel members with circular hollow section (CHS). To this end, uniaxial tension and ultra-low cycle fatigue tests on both un-notched and notched Q235B steel coupon specimens were conducted first to evaluate the material’s micromechanical fracture behavior. Next, 7 full-scale Q235B CHS steel members with different global slenderness were tested under cyclic axial loads with varying eccentricities. It is observed that: (1) the increase of the eccentricity of the axial load negatively affects the seismic and fracture resistance of CHS steel members, resulting in the deterioration of the strength, stiffness, energy dissipation, and ductility; (2) Large eccentricities lead to the reduction in the fracture life of Q235B CHS steel members; and (3) High global slenderness results in the reduction in both compression bearing capacity and energy dissipation of CHS steel members. In addition to the experimental program, finite element analyses are performed to simulate the seismic and fracture behavior of CHS steel members. The results from the finite element analyses are in good agreement with those from the experiment.

Diamide compounds such as chlorantraniliprole, a famous anthranilic diamide insecticide targeting the insect ryanodine receptor (RyR), have received continuous attention in pesticide research during the past 15 years owing to their excellent insecticidal potentials. With the aim of discovering new heterocyclic pesticides used for crop protection, based on the structural information of compound M from the reported pharmacophore-based virtual screening for RyR insecticides and diamide compound, a series of new heterocyclic mono-, di-, and tri-amide derivatives containing piperazine moiety have been synthesized in this paper. The new compounds were identified and confirmed by melting point, 1H NMR, 13C NMR and HRMS. Compound M was firstly validated for insecticidal activities, and the new synthesized compounds were all made comprehensive insecticidal evaluations against diamondback moth and oriental armyworm. The bioassay results showed that some of the compounds exhibit favorable insecticidal potentials, particularly some novel piperazine-containing heterocyclic mono-/di-/tri-amide derivatives such as 8g, 14a, 15a, 15g, 15i, 15j, 15k, 15l, and 15m could be used as new insecticidal leading structures for further study (e.g., towards diamondback moth, 15i-15m LC50: 0.0022−0.0081 mg/L). The structure-activity relationships of the compounds discussed in detail provide useful guidance for further design and development of new insecticides.

Gelatin, a natural macromolecular substance formed by the hydrolysis of collagen, offers the advantages of abundant resources and low cost. It is a good precursor for the synthesis of N, O-doped carbon materials. Although some gelatin-derived carbon materials have been used for electrochemical energy storage, there have been few studies for the mechanism of gelatin carbonization. In addition, the influence of heteroatoms and defects on the K-ion storage performance remains unclear. In this work, in situ and ex situ characterization methods have been used to study the mechanism of gelatin carbonization. The pathways leading to formation of the carbon framework, heteroatom doping and defect generation in gelatin-based carbons during the pyrolysis process have been clearly identified. Moreover, by means of DFT calculations and experimental measurements, it has been confirmed that heteroatoms and defects both play important roles in improving the storage performance of potassium ions. By virtue of its abundance of both heteroatoms and defects, our optimized anode shows better electrochemical performance than graphite anodes. This study not only clarifies our understanding of carbonization mechanism and mechanism of K-ion storage in carbon materials, but also provides pathways for the synthesis and applications of new carbon-based anode materials for alkali-ion batteries.

The quorum-sensing (QS) system between the phages and their hosts is important for the phage lysis-lysogeny decision. In Vibrio cholerae, the QS system consists of a LuxR-type receptor VqmA (VqmAVc) and an autoinducer molecule 3,5-dimethylpyrazin-2-ol (DPO). A VqmA homolog encoded by vibriophage VP882 (VqmAPhage) can intervene the host QS system via binding to both the host-produced DPO and its cognate promoter (Pqtip) to induce the phage lysogeny-to-lysis transition, whereas VqmAVc cannot influence the VqmAPhage-induced pathway, suggesting an asymmetry regulation. In this study, we report the crystal structure of VqmAPhage-DPO complex at 2.65 A and reveal that the mechanism of DPO recognition is conserved in VqmA homologs. Besides, we identify a non-classical palindrome sequence in Pqtip, which can be effectively recognized by VqmAPhage but not VqmAVc. The sequence contains an interval longer than that in the vqmR promoter recognized by VqmAVc. In addition, the two DBD regions in the VqmAPhage dimer exhibit more relaxed architecture than that of the reported VqmAVc, which is likely to be in the conformation that may easily bind to target promoter containing a longer interval. In summary, our findings provide a structural and biochemical basis for the DBD-dependent DNA recognition in different promoter regions in the phage lysogeny-to-lysis decision communication system, and provide clues for developing phage therapies against Vibrio cholerae infection.

Nowadays, developing separation materials for oil-water mixture and even emulsion have drawn extensive interest, with increasing oily wastewater pollution. Among kinds of separation materials, hydrophobic magnetic particles can rapidly and high-efficiently separate oil from either oil-water mixtures or emulsions. However, they not only commonly suffer from complicated preparation procedures and poor resistance to harsh conditions, but also are almost unable to satisfy the demand of multitasking separations towards oil-water mixtures and micro/nanoemulsions with surfactant. Herein, a facile, simple and scalable approach is introduced to prepare superhydrophobic magnetic hollow carbon microspheres (SMHCMs) with micro/nano hierarchical surface. Thanks to the special wettability properties, micro/nano hierarchical surface, nano-porous shell and hollow cores, the as-prepared SMHCMs show an unprecedented absorption capacity of over 10 g/g for oil-water mixtures within an absorption time of 60 s as well as a superior separation efficiency of near 99% for surfactant-free and surfactant-stabilized micro/nanoemulsions at an extremely short time of as low as 10 s. In addition, the SMHCMs also possess stable performance, good reusability, durability and resistance to harsh environments (e.g., acidic, alkaline and high-saline). This work proposes a new prospect for designing advanced materials applied to the fast and highly-efficient oil-water separation.

Objectives Our aim was to generalize the available evidence and evaluate the effectiveness of acupuncture for urinary incontinence in middle-aged and elderly women. Methods Six databases including VIP, CNKI, Wan Fang, Web of Science, PubMed and The Cochrane Library were systematically searched to retrieve similar studies updated to December 2019 to gather RCTs regarding the effectiveness of acupuncture for middle-aged and elderly women with urinary incontinence. Two researchers independently performed the whole process of retrieving the studies, extracting the data and assessing the risk of bias of the included studies. The current meta-analysis was performed using RevMan 5.3 software. Results A total of eight studies with 607 patients were included in the evaluation. The current meta-analysis showed that Compared with rehabilitation exercise or medication, acupuncture intervention significantly improved the clinical effectiveness (OR = 5.52, 95 % CI, 3.13–9.73), reduced the urine leakage in pad test (SMD = −2.67, 95 % CI, −4.05 to −1.29) and decrease the ICIQ-SF score (MD = −3.46, 95 % CI, −3.69 to −3.22). The results indicated that acupuncture intervention can help the patients alleviate the symptoms effectively. Conclusion Based on this study, acupuncture intervention of stress urinary incontinence in middle-aged and elderly women can improve the clinical effectiveness, reduce the urine leakage in pad test and ICIQ-SF score. More high-quality studies with large sample size are required for further verification.

Fuzheng Xiaoai Decoction 1 (FZXAD1) is a clinical experience prescription for the treatment of cancer patients at an advanced stage. FZXAD1 has been used for more than 10 years in the clinic and can effectively improve the deficiency syndrome of cancer patients. However, its mechanisms need further clarification.To check the effects of FZXAD1 in colon 26 (C26) cancer cachexia mice and try to clarify the mechanisms of FZXAD1 in ameliorating cancer cachexia symptoms.An animal model of cancer cachexia was constructed with male BALB/c mice bearing C26 tumor cells. Food intake, body weight and tumor size were measured daily during the animal experiment. Tissue samples in different groups including tumor and gastrocnemius muscle, were dissected and weighed at the end of the assay. Serum biochemical indicators such as total protein (TP), glucose (GLU) and alkaline phosphatase (ALP) were also detected. Network pharmacology-based analysis predicted the possible targets and signaling pathways involved in the effects of FZXAD1 on cancer cachexia therapy. Western blotting assays of the gastrocnemius muscle tissues from C26 tumor-bearing mice were then used to confirm the predicted possible targets of FZXAD1.The results of animal experiments showed that FZXAD1 could ameliorate cancer cachexia by alleviating the muscle wasting as well as kidney atrophy and increasing the body weight of cancer cachexia mice. AKT1, MTOR, MAPK3, HIF1A and MAPK1 were predicted as the core targets of FZXAD1. Western blotting confirmed the prediction that FZXAD1 increased the expression levels of phosphorylated Akt and mTOR in the muscle tissues. In addition, FZXAD1 treatment obviously ameliorated the increased levels of HIF-1α and phosphorylated Erk1/2 in C26 tumor-bearing mice.FZXAD1 effectively ameliorated cancer cachexia in an animal model of mice, which is consistent with its efficacy in the treatment of cancer patients. The mechanisms of FZXAD1 might be mainly based on its alleviating effects on muscle atrophy by activating the Akt-mTOR pathway and thus helping to maintain body weight.
