Natural product derived pesticides have increased in popularity worldwide because of their high efficacy, eco-friendly nature and favorable safety profile. The development of polyketide pesticides from actinomycetes reflects this increase in popularity in the past decades. These pesticides, which include avermectins, spinosyns, polynactins, tetramycin and their analogues, have been successfully applied in crop protection. Moreover, the advance of biotechnology has led to continuous improvement in the discovery and production processes. In this review, we summarize these polyketide pesticides, their activities and provide insight into their development. We also discuss engineering strategies and the current status of industrial production for these pesticides. Given that actinomycetes are known to produce a wide range of bioactive secondary metabolites, the description of pesticide development and high yield strain improvement presented herein will facilitate further development of these valuable polyketide pesticides from actinomycetes.

Dramatic decrease of sugar uptake is a general phenomenon in Streptomyces at stationary phase, when antibiotics are extensively produced. Milbemycins produced by Streptomyces bingchenggensis are a group of valuable macrolide biopesticides, while the low yield and titer impede their broad applications in agricultural field. Considering that inadequate sugar uptake generally hinders titer improvement of desired products, we mined the underlying sugar uptake systems and fine-tuned their expression in this work. First, we screened the candidates at both genomic and transcriptomic level in S. bingchenggensis. Then, two ATP-binding cassette transporters named TP2 and TP5 were characterized to improve milbemycin titer and yield significantly. Next, the appropriate native temporal promoters were selected and used to tune the expression of TP2 and TP5, resulting in a maximal milbemycin A3/A4 titer increase by 36.9% to 3321 mg/L. Finally, TP2 and TP5 were broadly fine-tuned in another two macrolide biopesticide producers Streptomyces avermitilis and Streptomyces cyaneogriseus, leading to a maximal titer improvement of 34.1% and 52.6% for avermectin B1a and nemadectin, respectively. This work provides useful transporter tools and corresponding engineering strategy for Streptomyces.

The emergence of bacterial resistance to common antibiotics poses a threat to human health and has rekindled an interest in antimicrobial peptides (AMPs). LHP7, a novel hybrid AMP containing 83 amino acid residues was designed on the basis of the LH28 and plectasin. LHP7 was expressed in Pichia pastoris, the total concentration of secreted protein reached 0.906 g/L after 108 h of methanol induction. Its antimicrobial activity was higher than that of the parent AMPs; the minimal inhibitory concentrations (MICs) of LHP7 against Staphylococcus aureus, Streptococcus pneumoniae and Streptococcus suis were 0.091, 0.023 and 0.18 μM, respectively. The antibacterial activity of LHP7 against clinical MRSA isolates (MICs = 0.73–2.91 μM) was enhanced over that of plectasin. The fractional inhibitory concentration (FIC) indicated a synergistic effect between LHP7 and ampicillin against MRSA (FIC = 0.375), and combinations of LHP7 with gentamicin, rifampin or tetracycline provided evidence of additive effects (FIC = 0.625–1.0). LHP7 exhibited a broad range of pH stability and thermostability, and a hemolytic activity of less than 5% below a concentration of 500 μg/mL. It was resistant to pepsin and papain digestion, but sensitive to trypsin digestion. These results suggest that LHP7 might have potential as a broadly applied and clinically useful antimicrobial agent.

Development of agents to overcome multidrug resistance (MDR) is important in cancer chemotherapy, and the overexpression of P-glycoprotein (P-gp) is one of the major mechanisms of MDR. In this paper, we evaluated the effects of two new milbemycin compounds, milbemycin β(14) and secomilbemycin D, isolated from fermentation broth of S. bingchenggensis on reversing MDR of adriamycin-resistant human breast carcinoma (MCF-7/adr) cells. We observed that the both milbemycins (5 μM) showed strong potency to increase adriamycin cytotoxicity toward MCF-7/adr cells with reversal fold (RF) of 13.5 and 10.59, respectively. In addition, the mechanisms of milbemycins on reversing P-gp-mediated MDR demonstrated that they significantly increased the accumulations of adriamycin and Rh123 via inhibiting P-gp efflux in MCF-7/adr cells. Furthermore, the results also revealed that milbemycin β(14) and secomilbemycin D could regulate down the expression of P-gp, but not affect the expression of MDR1 gene. In conclusion, our observations suggest that the two new milbemycin compounds probably represent the promising agents for reversing MDR in cancer therapy.

Two biofilm-based column reactors with walnut shell (WS) as carrier media were applied for fermentative hydrogen production using glucose as substrate by mixed microbial cultures at the temperature of 35 °C. Pure hydrogen producing bacteria Ethanoligenens harbinense B49 was supplemented into the reacting system periodically or continuously to enhance hydrogen production ability at the startup period. The results showed that the bioreactors supplemented with E. harbinense B49 performed better than the reactor without bacteria addition. Continuous addition mode was recommended, since the hydrogen production performance was better and the operation was easily to be accomplished. The optimal addition amount of pure bacteria was also investigated. The optimal bacterial addition amount was found to be 2.5% which led a better hydrogen production rate. In addition, the bioreactor supplemented with pure bacteria continuously presented a high hydrogen production ability as the specific hydrogen production rate (SHPR) maximized at 1.36 L/g-VSS·d; whereas, the bioreactor without bacteria addition obtained a maximum specific hydrogen production rate of 1.10 L/g-VSS·d. The addition of E. harbinense B49 favored the transformation to ethanol type fermentation in the bioreactor. Thereby, the startup period had been accelerated remarkably.

The effects of milbemycin A(4) (MB A(4)), milbemycin oxime A(4) (MBO A(4)) and milbemycin beta(1) (MB beta(1)) on reversing multidrug resistance (MDR) of tumor cells were firstly conducted according to the following research, including MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide] assay, the accumulation of adriamycin, the accumulation and efflux of rhodamine 123 (Rh123), the regulations of MDR1 gene, and expression of P-gp. The three milbemycins (5muM) showed strong potency to increase adriamycin cytotoxicity toward adriamycin-resistant human breast carcinoma cells MCF-7/adr with reversal fold (RF) of 21.42, 19.06 and 14.89, respectively. In addition, the mechanisms of milbemycins on P-glycoprotein (P-gp)-mediated MDR demonstrated that the milbemycins significantly increased the intracellular accumulations of adriamycin and Rh123 via inhibiting P-gp transport function. Based on the analysis of the P-gp and MDR1 gene expression using flow cytometry and RT-PCR, the results revealed that milbemycin compounds, particularly MB A(4), could regulate down the expression of the P-gp and MDR1 gene. These findings suggest that the milbemycins probably represent promising agents for overcoming MDR in cancer therapy, and especially MB A(4) is better modulator with the lowest toxicity.

Milbemectin is a widely used veterinary antiparasitic agent. A high-performance liquid chromatography with fluorescent detection (HPLC-FLD) method is described for the determination of milbemectin in dog plasma. The derivative procedure included mixing 1-methylimizole [MI, MI-ACN (1:1, v/v), 100 microL], trifluoroacetic anhydride [TFAA, TFAA-ACN (1:2, v/v), 150 microL] with a subsequent incubation for 3s at the room temperature to obtain a fluorescent derivative, which is reproducible in different blood samples and the derivatives proved to be stable for at least 80 h at room temperature. HPLC method was developed on C18 column with FLD detection at an excitation wavelength of 365 nm and emission wavelength of 475 nm, with the mobile phase consisting of methanol and water in the ratio of 98:2 (v/v). The design of an optimum and cost-efficient medium for high-level production of hydrogen by Ethanoligenens harbinense B49 was attempted by using response surface methodology (RSM). Based on the Plackett-Burman design, Fe(2+) and Mg(2+) were selected as the most critical nutrient salts. Subsequently, the optimum combination of the selected factors and the sole carbon source glucose were investigated by the Box-Behnken design. Results showed that the maximum hydrogen yield of 2.21 mol/mol glucose was predicted when the concentrations of glucose, Fe(2+) and Mg(2+) were 14.57 g/L, 177.28 mg/L and 691.98 mg/L, respectively. The results were further verified by triplicate experiments. The batch reactors were operated under an optimized condition of the respective glucose, Fe(2+) and Mg(2+) concentration of 14.5 g/L, 180 mg/L and 690 mg/L, the initial pH of 6.0 and experimental temperature of 35+/-1(o)C. Without further pH adjustment, the maximum hydrogen yield of 2.20 mol/mol glucose was obtained based on the optimized medium with further verified the practicability of this optimum strategy.

The biofilm-based expanded granular sludge bed (EGSB) reactor was developed to treat starch-containing wastewater and simultaneously recovery hydrogen by mixed microbial culture. Granular activated carbon (GAC) was used as the support media. Operating at the temperature of 30 °C for over 400 days (data not shown), the EGSB reactor presented high efficiency in hydrogen production and COD removal ability. The maximum hydrogen production rate (HPR) was found to be 1.64 L/L.d under the organic loading rate (OLR) of 1.0 g-starch/L.d, pH of 4.42 and HRT of 4 h. The hydrogen yield (HY) peaked at 0.11 L/g-COD, under the OLR of 0.5 g-starch/L.d, pH of 3.95 and HRT of 8 h. Hydrogen volume content was estimated to be 35–65% of the total biogas. The average COD removal rate was 31.1% under the OLR of 0.125 g-starch/L.d and HRT of 24 h. The main dissolved fermentation products were ethanol, acetate and butyrate. The average attached biofilm concentration was estimated to be 8.26 g/L, which favored hydrogen production and COD removal. It is speculated that the low pH operation in the present system would contribute significantly to lower the cost of alkaline amount required for pH control in the continuous operation, especially in the scale-up biohydrogen producing system. A model, built on the back propagation neural network (BPNN) theory and linear regression techniques, was developed for the simulation of EGSB system performance in the biodegradation of starch synthesis-based wastewater and simultaneous hydrogen production. The model well fitted the laboratory data, and could well simulate the removal of COD and the production of hydrogen in the EGSB reactor.

An expanded granular sludge bed (EGSB) process with granular activated carbon (GAC) was developed for fermentative hydrogen production from molasses-containing wastewater by mixed microbial cultures. No pH regulation was performed during the whole operation period. Running at the temperature of 35 °C, the EGSB reactor presented a high hydrogen production ability as the hydrogen production rate (HPR) maximized at 0.71 L/L h. At the same time, the hydrogen yield (HY) peaked at 3.47 mol/mol sucrose and the maximum specific hydrogen production rate (SHPR) was found to be 3.16 mmol H 2 /g VSS h. Hydrogen volume content was estimated to be 30–53% of the total biogas and the biogas was free of methane throughout the study. Dissolved fermentation products were predominated by acetate and ethanol, with smaller quantities of propionate, butyrate and valerate. It was found that high hydrogen yield was always associated with a high level of ethanol production. When the pH value and alkalinity ranged from 4.2–4.4 mg CaCO 3 /L to 280–340 mg CaCO 3 /L, respectively, stable ethanol-type fermentation was formed with the sum of ethanol and acetate concentration ratio of 89.1% to the total liquid products. The average attached biofilm concentration was estimated to be 17.1 g/L, which favored hydrogen production efficiently. With high biomass retention at high organic loading rate (OLR), this EGSB system showed to be a promising high-efficient bioprocess for hydrogen production from high-strength wastewater.

In order to enrich hydrogen producing bacteria and to establish high-efficient communities of the mixed microbial cultures, inoculum needs to be pretreated before the cultivation. Four pretreatment methods including heat-shock pretreatment, acid pretreatment, alkaline pretreatment and repeated-aeration pretreatment were performed on the seed sludge which was collected from a secondary settling tank of a municipal wastewater treatment plant. In contrast to the control test without any pretreatment, the heat-shock pretreatment, acid pretreatment and repeated-aeration pretreatment completely suppressed the methanogenic activity of the seed sludge, but the alkaline pretreatment did not. Employing different pretreatment methods resulted in the change in fermentation types as butyric-acid type fermentation was achieved by the heat-shock and alkaline pretreatments, mixed-acid type fermentation was achieved by acid pretreatment and the control, and ethanol-type fermentation was observed by repeated-aeration pretreatment. Denaturing gradient gel electrophoresis (DGGE) profiles revealed that pretreatment method substantially affected the species composition of microbial communities. The highest hydrogen yield of 1.96 mol/mol-glucose was observed with the repeated-aeration pretreatment method, while the lowest was obtained as the seed sludge was acidified. It is concluded that the pretreatment methods led to the difference in the initial microbial communities which might be directly responsible for different fermentation types and hydrogen yields.

A wheat cytochrome P450 cDNA (CYP71C6v1) was cloned by RT-PCR and heterologously expressed in yeast. The microsomal fractions derived from this strain could catalyze the metabolism of some sulfonylurea herbicides such as chlorsulfuron, triasulfuron, metsulfuron-metyl, bensulfuron-metyl, and tribenuron-rnetyl, but not sulfonylurea herbicides such as thifensulfuron and pyrazosulfuron. Kinetic parameters K-m for chlorsulfuron and triasulfuron were 57 (+/- 15)mu M and 38 (+/- 16) mu M in vitro, respectively. Analysis of the metabolites demonstrated that the CYP71C6v1 functioned as a 5-phenyl ring hydroxylase when chlorsulfuron and triasulfuron were the substrates. (c) 2005 Elsevier Inc. Streptomyces cyaneogriseus ssp. noncyanogenus is thermotolerant bacterium producing commercially important nemadectin, a kind of macrolide antibiotics, which has been widely used as a biopesticide. Here, we report the complete genome sequence of S. cyaneogriseus ssp. noncyanogenus , which consists of one chromosome (7,762,396 bp) with no plasmid. Genome sequence information contributes to understanding the biosynthesis of nemadectin better and provides the basis for analysis of its thermotolerance and biosynthetical potential.
