Hemicellulose and its derivatives have attracted extensive attention as packaging materials and various methods have been utilized to improve its film formation properties. To make use of the byproduct in dissolving pulp production, hemicellulose collected from waste water was modified by carboxymethylation and blended with polyvinyl alcohol (PVA) to prepare composite film by solution casting method. Potassium cinnamate (PC) was further incorporated to endow the film with antibacterial activity. The properties of the composite films were characterized. Alginate oligosaccharide (AOS) is widely used in agriculture because of its many excellent biological properties. However, the possible beneficial effects of AOS and their underlying mechanisms are currently not well known in grass carp (Ctenopharyngodon idellus). Here, grass carp were fed diets supplemented with 5, 10, or 20 g/kg AOS for six weeks. HE and PAS staining showed that the diets of AOS significantly increased the number of goblet cells in the intestinal. According to transcriptome and quantitative real-time PCR (qRT-PCR) data, AOS-supplemented diets activated the expression of fat metabolism-related pathways and genes. The 16S rRNA sequencing results showed that supplementation with AOS affected the distribution and abundance of the gut bacterial assembly. qRT-PCR and activity assays revealed that the AOS diets significantly increased the antioxidant resistance in gut of grass carp, and down-regulated the expression of inflammatory and up-regulated anti-inflammatory cytokines. Finally, the Aeromonas hydrophila infection assay suggested that the mortality in the groups fed dietary AOS was slightly lower than that in the control. Therefore, supplementing the diet of grass carp with an appropriate amount of AOS can improve fat metabolism and immune responses and alter the intestinal bacterial community, which may help to fight bacterial infection.

This was a single-center, randomized, double-blind, parallel control study evaluating the immunogenicity and safety of a two-dose schedule of serogroups ACYW meningococcal polysaccharide conjugate vaccine with tetanus toxoid (TT) conjugate protein, in infants and toddlers of 3-35 months old.720 participants were stratified according to the age of 3-5 months old, 6-11 months old, and 12-35 months old and randomly assigned with an equal ratio to two different dose groups, i.e., 40- and 20-μg doses. Blood samples were taken from all participants before the first vaccination and 30 days after the full-course vaccination to detect the serogroups ACYW meningococcal antibodies. All adverse events occurred within 30 days after vaccination of each dose, and serious adverse events occurred within six months after full-course vaccination were collected for safety evaluation. This study was registered at the China drug trial registration with the identifier CTR 20182031.After 30 days of full-course vaccination, 92.78 % (95 % CI: 85.70 %-100.00 %) showed the immune response against all serogroups in both high-dose and low-dose groups by rabbit serum bactericidal antibody assay (rSBA) and the geometric mean titer (GMT) of all serogroups showed a high level (74.6-505.8, 95 % CI: 56.4-615.7). However, no significant difference between different dose groups was observed (P 0.05). The common local and systemic adverse events in both groups were redness (3 %-7%), and fever (26 %-65 %), respectively. In addition, the grade 3 adverse event related to the vaccine was fever (1.67 %-12.50 %). No serious adverse event was reported to be associate with the vaccination.The serogroups ACYW meningococcal polysaccharide conjugate vaccine was safe and effective in the population aged 3-35 months. The vaccine efficacy and safety of the 20-μg dose group were not less than that of the 40-μg dose group.

Postsurgical treatment is of great importance to combat tumor recurrence and metastasis. Anti-CD47 antibodies (aCD47) can block the CD47-signal regulatory protein-alpha (CD47-SIRPα) pathway to restore immunity. Here, an in-situ gel implantation was engineered by crosslinking chitosan (CS) and pullulan (Pul) for postsurgical treatment. A highly selected chemotherapeutic, cyclopamine (Cyc), encapsulated in liposomes (Cyc-Lip) was co-loaded with aCD47 in gels for chemoimmunotherapy. Importantly, a sequential drug release kinetics can be achieved. Nanotherapeutics were confirmed to be released prior to aCD47 in a burst-release manner, which was benefit for immediately killing residual tumor cells followed by releasing tumor antigens. Meanwhile, aCD47 was released in a sustained-release manner to restore macrophage functions and exert anti-tumor immune responses. Afterwards, the efficacy of in-situ chemoimmunotherapy was confirmed on 4T1 mouse breast cancer models, which could not only efficiently augment anti-tumor effect to inhibit tumor recurrence but also establish a long-term immune memory to combat tumor metastasis.

This paper designs formation–containment control algorithms for a class of second-order nonlinear multi-agent systems governed by Euler–Lagrange dynamics with communication delays. The formation–containment problem consists of leader agents’ formation control and follower agents’ containment control. Firstly, to make the leaders form a desired formation and move collectively with a constant velocity, a coordinated formation control algorithm is designed and the variable-gain technique is used to eliminate the effect of communication delays on the leaders’ formation control. Secondly, considering that only the leaders have access to the desired moving velocity, we propose distributed velocity estimators for followers in which the communication delays also exist in the followers’ information interaction. By using the estimated velocity information, coordinated containment control laws are designed for the followers to drive them asymptotically converge to the convex hull spanned by all leaders. Furthermore, to increase the system robustness against uncertainties and external disturbances, the adaptive updating laws are designed for all agents. Finally, simulations are given to demonstrate these obtained results.

Poria cocos polysaccharide (PCP) is one of the main active components of Poria cocos that is extensively used in the world. PCP can be divided into intro-polysaccharides and exopolysaccharides. PCP is mainly composed of glucose, galactose and mannose. There are many methods to exact PCP, and methods can affect its yield. PCP and its derivatives exhibit diverse biological functions such as antitumour, antioxidant, anti-inflammatory, immune-regulatory, hepatoprotective, etc. There is the potential application of PCP as drug carriers. The review provides a comprehensive summary of the latest extraction and purification methods of PCP, its chemistry, synthesis of PCP derivates, their pharmacological activities and their applications as drug carriers. This review provides comprehensive information on PCP, which can be used as the basis for further research on PCP and its derivates.

Tumor recurrence and metastasis after surgery remain challenges for tumor treatment. Strategy that can promote the immunogenicity, activate adaptative immune response and eliminate post-operative immunosuppression would be a promising way to achieve a desired clinical benefit. In this study, immunogenic cell death (ICD) priming anti-tumor adaptive immune response was executed to potentiate immune checkpoint blockade (ICB) therapy through the PD1/PDL1 pathway for postsurgical treatment. Here, we present a bio-responsive and cargo-catchable gel depot composed of pullulan and chitosan cross-linking through matrix metalloproteinase (MMP) sensitive peptide for co-delivery of anti-programmed death-ligand 1 antibody (aPDL1) and doxorubicin -encapsulated liposomes (DOX-Lip). This drug carrier showed expected ability to respond to the highly expressed MMP in postsurgical tumor microenvironment (TME). In vivo studies on 4T1 breast tumor mouse model demonstrated that the gel depot could efficiently prolong the mouse survival after tumor resection by preventing tumor recurrence and metastasis. The results suggested that ICD combining with PD1/PDL1 blockade based on the bio-responsive and cargo-catchable gel depot could facilitate the maturation of DCs and reverse the immunosuppressive environment in tumor resection area, thus amplifying the systemic anti-tumor immune response.

Superoxide dismutases (SODs) are the main antioxidant enzymes involved in alleviating oxidative stress. Although mitochondrial manganese SOD (mMnSOD) has been reported to be correlated with the immune response in crustaceans, its biological properties and role in the immune response remain unclear. Here, we cloned the Macrobrachium rosenbergii mMnSOD (MrmMnSOD), analyzed its activity and expression pattern under Staphylococcus aureus and Vibrio parahaemolyticus infection, and further explored its possible mechanism during antibacterial immune response. The results showed that both enzyme activity and the expression of MrmMnSOD were significantly up-regulated by bacterial infection. MrmMnSOD knockdown made the prawn susceptible to Vibrio infection, which increased the mortality rate and the number of bacteria in haemocytes. The bacterial agglutination assay confirmed that MrmMnSOD decreases bacterial abundance via agglutination. Overall, this work identified antibacterial function of MrmMnSOD in the immune response. In addition to contributing to immunological theory, these findings aid disease prevention and control in crustacean aquaculture.

Hematopoietic stem cells (HSCs) and lineage committed hematopoietic progenitor cells (HPCs) undergo profound shifts in gene expression during neonatal and juvenile stages of life. Temporal changes in HSC/HPC gene expression underlie concomitant changes in self-renewal capacity, lineage biases and hematopoietic output. Moreover, they can modify disease phenotypes. For example, childhood leukemias have distinct driver mutation profiles relative to adult leukemias, and they may arise from distinct cells of origin. The putative relationship between neonatal HSC/HPC ontogeny and childhood blood disorders highlights the importance of understanding how, at a mechanistic level, HSCs transition from fetal to adult transcriptional states. In this perspective piece, we summarize recent work showing that the transition is uncoordinated and imprecisely timed. We discuss implications of these findings, including mechanisms that might enable neonatal HSCs and HPCs to acquire adult-like properties over a drawn-out period of time, in lieu of precise gene regulatory networks. The transition from fetal to adult transcriptional programs coincides with a pulse of type I interferon signaling that activates many genes associated with the adult-like state. This pulse may sensitize HSCs/HPCs to mutations that drive leukemogenesis shortly after birth. If we can understand how developmental switches modulate HSC and HPC fate after birth – both under normal circumstances and in the setting of disease-causing mutations – we can potentially reprogram these switches to treat or prevent childhood leukemias.

Adding hydrocolloids into native starch is a secure and effective method of physical modification. In this study, the effect of sodium alginate (AG) on the gelatinization, rheological, and retrogradation properties of rice starch (RS) was investigated by measuring the pasting parameters, melting enthalpy (ΔH), rheological characteristic parameters, intensity ratio of 1047 cm−1 to 1022 cm−1 (R1047/1022), and relative crystallinity (RC) of RS–AG blends. Rapid visco analysis shows that AG could significantly change the gelatinization parameters of RS. Differential scanning calorimetry results show that the ΔH values of RS initially decreased in the low AG concentration range (0.10%–0.30%), but increased in the high AG concentration range (0.30%–0.50%). Dynamic rheological analysis reveals that the modulus (G′, G′′) and the loss tangent (tan δ) increased with the rise of the AG concentration from 0.10% to 0.50%. Fourier transform infrared spectroscopy and X-ray diffraction patterns collectively prove that the crystallinity of RS decreased with the addition of AG during the retrogradation periods. The interactions between AG and starch molecules in RS–AG blends were hypothesized to correlate with the aforementioned results.
