Oriented ligand immobilization is one of the most effective strategies used in the design and construction of a high-capacity protein A chromatography. In this work, cysteine was introduced as anchoring sites by substituting a specific residue on Helix I, II, and at C-terminus of antibody binding domain Z from protein A, respectively, to investigate structural evolution and binding behavior of protein A ligands at liquid-solid interfaces. Among the three affinity dextran-coated Fe3O4 magnetic nanoparticles (Fe3O4@Dx MNPs), affinity MNPs with the immobilized ligand via N11C on Helix I (Fe3O4@Dx-Z1 MNPs) had the highest helical content, and MNPs with the immobilized ligand via G29C on Helix II (Fe3O4@Dx-Z2 MNPs) had the lowest helical content at the same pHs. It was attributed to less electrostatic attraction of ligand to negatively charged surface on Fe3O4@Dx-Z1 MNPs because of less positive charged residues on Helix I (K6) than Helix II (R27/K35). Among the three affinity MNPs, moreover, the highest affinity to immunoglobulin G (IgG) binding was observed on Fe3O4@Dx-Z1 MNPs in isothermal titration calorimetry measurement, further validating greater structural integrity of the ligand on Fe3O4@Dx-Z1 MNPs. Finally, the study of IgG binding on MNPs and 96-well plates showed that anchoring sites for ligand immobilization had distinct influences on IgG binding and IgG-mediated antigen binding. This work illustrated that anchoring sites of the ligands had a striking significance for the molecular structure of the ligand at liquid-solid interfaces and raised an important implication for the design and optimization of protein A chromatography and protein A-based immunoassay analysis.

Nutritional screening tools should be sensitive, simple, and easy to use. Differing opinions among clinicians concern the simplicity of the three tools-the Global Leadership Initiative on Malnutrition (GLIM) criteria, Nutritional Risk Screening 2002 (NRS-2002), and Patient-Generated Subjective Global Assessment (PG-SGA). For each tool, we estimated prediction of overall survival (OS) in tumor staging, sensitivity, and specificity. The NRS-2002 is favored by clinicians because it is simple to use. We compared its sensitivity and specificity with the GLIM and PG-SGA.This is an analysis of data from 1,358 adult colorectal cancer patients recruited in a multicenter from July 2013 to July 2018.In Kaplan-Meier models, each tool was found to be significantly predictive of OS: NRS-2002 (1.28), GLIM (1.49), and PG-SGA (1.42). Use of any tool improved prediction of survival at tumor staging. NRS-2002 has superior specificity (0.90) to diagnose patients without nutritional deficits (GLIM = 0.62 and PG-SGA = 0.82).This study provides evidence for the superiority of NRS-2002 to accurately identify colorectal cancer patients without nutritional limitations. Compared with the complexity of the other tools, NRS-2002 is the simplest tool to use in routine nutritional screening in busy clinical practice.

Aim of the study To evaluate the efficacy and preliminary outcomes of using a postauricular-groove approach without endoscopic assistance for the excision of parotid tumors. Case series Patients who underwent parotidectomy using a postauricular-groove incision were selected for this study. For patients in which parotidectomy was difficult, namely, for tumors located in the deep lobe area, the parotid gland sternocleidomastoid space was fully utilized, and the tumor was resected from the posterior plane. A total of fifty-eight patients with parotid tumor were enrolled and divided into superior lobe group (n=46) and deep lobe group (n=12). The difference in operation time (94 vs 119 min) and postoperative drainage (20.18 vs 45.33 mL) was statistically significant between the tumors in the superficial and deep lobes. However, postoperative cosmetic VAS score was 10 (extremely satisfied) for all patients. The incidence of transient facial nerve paralysis was comparable (8.7% vs 16.7%), and all of them resolved spontaneously within 3 months. No recurrence of tumors was found in either group in the median follow-up interval of 26.45 months (range 22.2-35.3 months), which was comparable to the result using the conventional “S” approach. Conclusion After making full use of the parotid gland sternocleidomastoid space, the postauricular-groove approach demonstrated satisfactory facial nerve protection, as well as easy maneuverability without the risk of surgical complications for tumors located in the deep lobe area. Importantly, the postauricular-groove approach showed excellent cosmetic outcomes for all patients and should be considered an alternative approach for parotidectomy of selected cases.

A survival benefit from esophagectomy was observed in elderly patients. But it's unclear how to identify specific patients who can benefit. Thus, we aimed to establish a predictive model to identify optimal candidates for esophagectomy.Patients (age ≥75 years) with esophageal cancer in Surveillance, Epidemiology and End Results (SEER) database were used to establish the predictive model. Propensity-score matching (PSM) was applied to eliminate the imbalance between esophagectomy group and non-esophagectomy group. We hypothesized that elderly patients could benefit from esophagectomy with longer cancer specific survival (CSS) time than those who did not receive esophagectomy. Patients received surgery were divided into beneficial group and non-beneficial group according to the median CSS time of non-esophagectomy group. Prognostic factors affecting patients' long-term survival were identified. Among esophagectomy group, a logistic regression model based on these factors was established to build a nomogram.A total of 7,025 eligible patients were extracted from the SEER database, with 831 patients received esophagectomy. Surgery was independently associated with better long-term survival (median CSS time in the matched population: 35 vs. 8 months, p 0.001). As a result, 361 (68.6%) patients were divided into beneficial group (CSS8 months). Factors including age, tumor site, histology, differentiation grade, TNM stage, and tumor size were used to formulate the nomogram, which was named as esophagectomy candidates screening score (ECSS). The validation from two aspects showed the model a useful and stable one.A predictive model was established to distinguish optimal candidates for esophagectomy among elderly patients with EC.

Stem cell-derived exosomes (SC-EXO) was an emerging therapeutic agent in regenerative medicine. Intratunical injection of SC-EXO is considered as a prospective approach for erectile dysfunction (ED) treatment. However, high vascularization of cavernous body makes effective retention a major challenge for SC-EXO intratunical injection. In this study, a Polydopamine nanoparticles (PDNPs) incorporated poly (ethylene glycol)-poly(ε-caprolactone-co-lactide) (PDNPs-PELA) thermosensitive hydrogels were fabricated by a facile in situ polymerization for intratunical administration of adipose stem cell-derived exosomes (EXO). The hydrogels exhibited sol-gel transition at body temperature. Moreover, the in-situ polymerization of PDNPs using poly (ethylene glycol)-poly(ε-caprolactone-co-lactide) (PELA) block copolymer as a template was found to be more stable dispersion in the gel system. After being encapsulated into the hydrogel, EXO shows sustained release behavior within two weeks. In vivo animal experiments revealed that exosomes released from hydrogel lead to the healing of endothelial cells and neurons, increase of the cavity's pressure, thereby restoring the erectile function. In particular, since the PDNPs in thermosensitive gels have excellent photoacoustic performance, the hydrogel can be accurately delivered into the tunica albuginea by the guidance of real-time photoacoustic imaging. These results suggest that the as-prepared PDNPs-PELA has a promising future as an injectable exosome carrier for ED treatment., Graphical abstract Image 1, Highlight • A temperature-sensitive hydrogel with photoacoustic activity was developed for intratunical injection by in-situ polymerization. •The exosomes encapsulated in the hydrogel can be slowly released and effectively restore damaged nerve and vascular endothelial cells. •The injection guided by photoacoustic images realizes accurate puncture and real-time filling detection of hydrogel in the corpus cavernosum.

Fast and accurate identification of pulsar signals is important for X-ray pulsar-based navigation (XNAV). Traditional pulsar signal detection technology based on FFT search and epoch folding (EF) requires a very long time to obtain an appropriate signal-to-noise ratio (SNR) gain, especially for weak pulsar signals with low photon fluxes. This paper proposes an adaptive stochastic resonance (SR) method based on the grey wolf optimizer (GWO) algorithm for fast pulsar identification. The GWO algorithm can optimize the SR parameters to obtain the optimal SR output that matches the input signal. Meanwhile, the initial population of GWO is generated by Chebyshev chaotic maps, which guarantees the diversity of the initial population and enhances the global search capability of GWO. To further improve the performance of SR, we add an extra denoising process, including a high-pass filter and singular value decomposition (SVD) that determines the effective singular components through dispersion entropy (DE), to suppress the noise to a great extent. We use simulation data and Rossi X-ray Timing Explorer (RXTE) observation data to verify the proposed method. The output SNR is greatly improved, and the method can identify pulsars in a short time. The results indicated that the proposed method has great practical value in engineering. Further experiments reveal the importance of SVD, DE and the Chebyshev chaotic map for the algorithm.

This article presents a nonlinear least square estimation method based on the continuous TEC values at single stations from long-time recorded dual-frequency GPS data of International GNSS Service (IGS) stations to facilitate single-station empirical total electron content (TEC) modelling. The Nonlinear Polynomial GPS-TEC Model (NPGTM) at each of the four different stations (namely Ellesmere Island (alrt), Canada; Lhasa (lhaz), China; New Norcia (nnor), Australia; and East Ongle Island (syog) over the Antarctica) was constructed by the nonlinear least square estimation method using GPS-TEC data from 2005 to 2016 and validated with 2017–2018 datasets over the same locations in the dependencies of universal time (UT), solar activity (PI index), geomagnetic activity (Ap index) and day of year (DOY). The model was designed for quiet and moderate geomagnetic conditions (i.e. Ap ≤ 30 and F10.7 ≤ 160 sfu). Compared with IRI-2016 model values in 2017 to 2018, the accuracy of NPGTM model values is improved by 132% in high latitude area, 84% in medium latitude area and 88% in low latitude area. Evaluation results show that the NPGTM model values are in good agreement with GPS-TEC observations in high, medium and low latitude regions, which is much better than the IRI-2016 model values.

Long-term prediction parameters of polar motion (PM) with high precision have an important significance in understanding Earth geophysical processes in the future. The multilayer perceptron (MLP) is introduced into PM prediction and we try for the first time to combine MLP with autoregressive moving average model (ARMA) and singular spectrum analysis (SSA) for the mid-long-term prediction of PM. The principal components of PM were extracted and predicted by SSA first. Then, the combined MLP and ARMA were used to predict the residual term. The PM predictions were obtained by adding the principal components and the residual predictions. Applying this proposed method, 157 500-day and 12 5-year lead time predictions of PM were made based on IERS 14 C04 product. The results showed that the proposed method performed well in the mid-long-term period of PM predictions, which was superior to IERS Bulletin A.

Regenerable methods for phosphate (P) recycling have received intense attention due to their potential environmental and economic benefits. In this study, to improve the electrosorptive removal of P in membrane capacitive deionization, an activated carbon (AC) electrode was coated with a heterogeneous anion-exchange resin layer, and named the AE-AC composite electrode. It was shown that the AE-AC electrode exhibited a good capacitive behavior for electrical double-layer charging. The batch-mode experiments indicted that when the solution pH changed from 5 to 8, the predominant P species shifted from monovalent H2PO4− to divalent HPO42− that was preferentially electroadsorbed for competitive electrosorption with Cl−. Importantly, the AE-AC composite electrode significantly increased the selectivity coefficient of P over Cl− to 0.56 that was 2.24-fold greater than that of the uncoated AC electrode, at 1.2 V in single-pass mode operation. This improvement can be ascribed to the preferential transport of P through the thin coating layer containing quaternary amine functional groups. The permselectivity of the coating also significantly increased the electrosorption capacity of P from 0.031 to 0.101 mmol/g with a high charge efficiency (97%) by the reduction in the co-ion repulsion effect. When the reverse voltage (−1.2 V) was applied, electroadsorbed P was reversibly desorbed from the AE-AC electrode in repeated operation. This work suggests that coating an anion-exchange resin layer on the surface of a carbon electrode shows great potential to improve the selective removal of P through electrosorption.

Glucose metabolism reprogramming is one of the hallmarks of cancer cells, although functional and regulatory mechanisms of long noncoding RNA (lncRNA) in the contribution of glucose metabolism in lung adenocarcinoma (LUAD) remain incompletely understood. The aim of this study was to uncover the role of GAS6-AS1 in the regulation of progression and glucose metabolism in LUAD. We discovered that overexpression of GAS6-AS1 suppressed tumor progression of LUAD both in vitro and in vivo. Metabolism-related assays revealed that GAS6-AS1 inhibited glucose metabolism reprogramming. Mechanically, GAS6-AS1 was found to repress the expression of glucose transporter GLUT1, a key regulator of glucose metabolism. Ectopic expression of GLUT1 restored the inhibition effect of GAS6-AS1 on cancer progression and glucose metabolism reprogramming. Further investigation identified that GAS6-AS1 directly interacted with transcription factor E2F1 and suppressed E2F1-mediated transcription of GLUT1, and GAS6-AS1 was downregulated in LUAD tissues and correlated with clinicopathological characteristics and survival of patients. Taken together, our results identified GAS6-AS1 as a novel tumor suppressor in LUAD and unraveled its underlying molecular mechanism in reprogramming glucose metabolism.
