Abstract

During processing of molten polymers the thermal and mechanical stress acting on the melt in presence of oxygen can induce degradation with a modification of the chemical structure of the polymers. This picture can become even more relevant if the melt is a multiphasic polymer system. In this last case, the effects of the degradation depend also on the presence of a second phase and on the interactions between the two phases. In this work, the effect of the processing conditions, temperature and time, have been considered in order to investigate the thermo-mechanical and thermo-oxidative degradation of nanocomposites made by polypropylene and different contents of graphene nanoplatelets. Graphene nanoplatelets improve the stability of the polymer matrix and the sample with the lower content of nanofiller shows a lower level of degradation. The better resistance of the filled samples has been interpreted with a barrier effect to the oxygen of the nanofiller that hinders the transport of oxygen necessary for the oxidation of the polypropylene. The unexpected effect of the concentration of graphene nanoplatelets has been correlated with the agglomeration of the nanoplatelets that reduces the surface area of the filler and then the barrier effect to the oxygen.

Abstract

PLA/PCL co-mingled nanofibrous mats were prepared via multi-jet electrospinning. The concentration of PLA and PCL in the co-mingled mats were controlled by changing the flow rate of the two polymer solutions. The amount of PLA and PCL in the co-mingled nanofibrous mats was monitored by UV–Vis measurements through a colored dye added to PLA and by FTIR-ATR analysis. Morphology and mechanical properties of the nanofibrous mats were respectively examined by scanning electron microscopy (SEM) and tensile tests. Water contact angles measurements were also carried out in order to investigate the wettability of the materials. Finally, the hydrolytic degradation of the mats in buffer solution at pH 4, pH 7 and pH 10 were studied up to 50 days.

Abstract

A novel floatable and biodegradable sponge for the selective absorption of oil from water and potentially useful as cell carrier for bioremediation treatments was prepared in polycaprolactone (PCL). The eco-friendly process for fabricating the PCL sponge does not involve either synthetic routes or organic solvents, thus minimizing environmental hazard. In particular, the 3D porous materials have been prepared by mixing in the melt the polymer matrix with two water-soluble porogen agents (NaCl and PEG) and thereafter leaching the obtained PCL/NaCl/PEG composites in water. The PCL sponges here proposed are capable to remove different types of oily pollutants (up to 500wt%), and were successfully tested as carrier materials of hydrocarbon(HC)-degrading bacteria for bioremediation. The bioremediation efficiency of the scaffold-bacteria system was analyzed on crude oil and n-alkanes using two highly performant HC-degrading bacterial strains: the marine hydrocarbonoclastic model strain Alcanivorax borkumensis SK2 and the soil long-chain n-alkane degrader Nocardia sp. SoB. Morphological analysis highlighted a high capacity of adhesion and proliferation of bacterial cells within the whole interconnected three-dimensional structure. HC degradation rates, evaluated by GC-FID analysis, demonstrated the higher degrading performance of immobilized-cells if compared with conventional submerged liquid culture.

Abstract

The investigation of the thermal stability up to 400 °C of Graphene Oxide (GO) and GO-silica nanoparticles (n-SiO2) composites prepared by direct mixture of GO and n-SiO2 is reported. Using Scanning Electron Microscopy, X-ray Photoelectron Spectroscopy, Energy Dispersive X-ray analysis, Atomic Force Microscopy, Raman and Infrared absorption measurements a thorough characterization of the prepared materials is carried out. By deepening the changes induced in the 2D Raman spectral region of GO at about 2900 cm−1 the comprehension of an interplay, driven by the thermally induced changes of the material, between inter-valley and intra-valley vibrational transitions is elucidated. This result enables to find out that the major thermally induced changes are related to the GO counterpart of the nanocomposite and shows that the deposited silica nanoparticles are not interfering with the GO principal features exploiting the potentiality of nanocomposites preparation. Furthermore, silica nanoparticles are found to act as solid exfoliant agents, thus inhibiting GO lamellae re-aggregation even at high temperatures.

Abstract

Basal forebrain neurons increase cortical blood flow by releasing acetylcholine (Ach), which stimulates endothelial cells (ECs) to produce the vasodilating gasotransmitter, nitric oxide (NO). Surprisingly, the mechanism whereby Ach induces NO synthesis in brain microvascular ECs is unknown. An increase in intracellular Ca2+ concentration recruits a multitude of endothelial Ca2+-dependent pathways, such as Ca2+/calmodulin endothelial NO synthase (eNOS). The present investigation sought to investigate the role of intracellular Ca2+ signaling in Ach-induced NO production in bEND5 cells, an established model of mouse brain microvascular ECs, by conventional imaging of cells loaded with the Ca2+-sensitive dye, Fura-2/AM, and the NO-sensitive fluorophore, DAF-DM diacetate. Ach induced dose-dependent Ca2+ oscillations in bEND5 cells, 300 μM being the most effective dose to generate a prolonged Ca2+ burst. Pharmacological manipulation revealed that Ach-evoked Ca2+ oscillations required metabotropic muscarinic receptor (mAchR) activation and were patterned by a complex interplay between repetitive ER Ca2+ release via inositol-1,4,5-trisphosphate receptors (InsP3Rs) and store-operated Ca2+ entry (SOCE). A comprehensive real time-polymerase chain reaction analysis demonstrated the expression of the transcripts encoding for M3-mAChRs, InsP3R1 and InsP3R3, Stim1-2 and Orai2. Next, we found that Ach-induced NO production was hindered by L-NAME, a selective NOS inhibitor, and BAPTA, a membrane permeable intracellular Ca2+ buffer. Moreover, Ach-elicited NO synthesis was blocked by the pharmacological abrogation of the accompanying Ca2+ spikes. Overall, these data shed novel light on the molecular mechanisms whereby neuronally-released Ach controls neurovascular coupling in blood microvessels.

Abstract

Biopolymeric porous devices exhibiting graded properties can play a crucial role in several fields, such as tissue engineering or controlled drugs release. In this context, the gradient of a specific property can be achieved by developing porous laminates composed by different types of materials. This work presents for the first time a multi-phasic porous laminate based on polycaprolactone (PCL) and polylactic acid (PLA) prepared by combining melt mixing, compression molding and particle leaching. All the materials were characterized from a morphological and a mechanical point of view. The results put into evidence the possibility to tune and to predict the mechanical properties by controlling the process parameters together with geometrical features.

Abstract

Nanocomposites are a new class of polymer composites that exhibit an interesting combination of chemical, physical, thermal and mechanical properties. Only small amounts of nanofiller are sufficient to generate great variations of many properties. This work focuses on the rheological behaviour of the intercalated polymer nanocomposites in shear flow. The increase of the viscosity and the more pronounced non-Newtonian behaviour is interpreted simply considering the increase of volume of the inert phase caused by the intercalation of the macromolecules. Indeed, the volume of the intercalated tactoids increases with increasing the interlayer distance. On the other hand, the interlayer distance can be correlated with the increase of the viscosity or with the more pronounced non-Newtonian behaviour. This simple method cannot be applied to exfoliated nanocomposites or when interactions between the matrix and the nanoclay occur, thus leading to a modification of the polymer chains, e.g., degradation of the matrix due to the interaction with the organo-modifier of the clay.

Abstract

Background: Overall survival after cancer is frequently used when assessing a health care service’s performance as a whole. It is mainly used by the public, politicians and the media, and is often dismissed by clinicians because of the heterogeneous mix of different cancers, risk factors and treatment modalities. Here we give survival details for all cancers combined in Europe, correlating it with economic variables to suggest reasons for differences. Methods: We computed age and cancer site case- mix standardised relative survival for all cancers combined (ACRS) for 29 countries participating in the EUROCARE-5 project with data on more than 7.5 million cancer cases from 87 population-based cancer registries, using complete and period approach. Results: Denmark, United Kingdom (UK) and Eastern European countries had lower survival than neighbouring countries. Five-year ACRS has been increasing throughout Europe, and substantial increases, between 1999–2001 and 2005–2007, have been achieved in countries where survival was lower in the past. Five-year ACRS for men and women are positively correlated with macro-economic variables like the Gross Domestic Product (GDP) and Total National Expenditure on Health (TNEH) (R2 about 70%). Countries with recent larger increases in GDP and TNEH had greater increases in cancer survival. Conclusions: ACRS serves to compare all cancer survival in Europe taking account of the geographical variability in case-mixes. The EUROCARE-5 data on ACRS confirm previous EUROCARE findings. Survival appears to correlate with macro-economic determinants, particularly with investments in the health care system.

Abstract

Background This work presents relative survival estimates regarding urinary tract tumours among adult patients (age ⩾ 15 years) diagnosed in Europe. It reports on survival estimates of cases diagnosed in 2000–2007, and on survival time trends from 1999–2001 to 2005–2007. Methods Data on 677,340 adult urinary tract tumour patients, (429,154 cases of invasive and non-invasive bladder and 248,186 cases of invasive kidney cancers) diagnosed between 2000 and 2007 were provided by 86 population-based cancer registries from 29 European countries. The complete approach was used to estimate survival in 2000–2007; the period approach was used to estimate survival over time. Results The age-standardised 5-year relative survival for patients with kidney tumours diagnosed in Europe during 2000–2007 was 60%. The best prognosis was observed in Southern and Central Europe and prognosis improved in all regions along the time period. For invasive and non-invasive patients with bladder tumours combined the age-standardised 5-year relative survival in Europe was 68%. The best prognosis was observed in Southern and Northern Europe. However, in Scotland and The Netherlands the relative survival was significantly lower, although the survival estimates for these two countries were based on invasive tumours only. Conclusions Differences in registration practices affect comparisons of survival values between European countries, especially in patients with urinary bladder cancers. The between-country variation in survival is influenced by the varying use of diagnostic investigation in urinary tract tumours. Further data on stage at diagnosis can help to elucidate the influence of diagnostic intensity or early diagnosis on the survival patterns.

Abstract

Polymer nanocomposites have continually attracted increasing interest over the last decade, due to significant improvements they can offer compared to neat polymer matrices. However, the final morphology of a nanocomposite, determined by several variables, can significantly influence the macroscopic properties of the final product. Therefore, it is important to study the interactions between processing, morphology, structure and rheological properties, and the suitability of existing models in order to predict the system's behaviour with change of the main processing variables. In this work, the applicability of a predictive theory based on the Wu model was formulated and proposed in order to predict the morphology of nanocomposite systems. This theory allows description of a polymer matrix - lamellar filler system, provided that the interactions between the two components and the processing are known. In particular, a lamellar filler has been considered as a deformable second component of a blend, with its behaviour depending on the processing and on the interactions with the polymer matrix. This allowed analyzing different behaviour for the systems, due to the different polarity of the matrix, which may lead to a classification of polymer nanocomposites in two or more families, according to their different matrix polarities.

Abstract

Background Since 25 years the EUROCARE study monitors the survival of cancer patients in Europe through centralised collection, quality check and statistical analysis of population-based cancer registries (CRs) data. The European population covered by the study increased remarkably in the latest round. The study design and statistical methods were also changed to improve timeliness and comparability of survival estimates. To interpret the EUROCARE-5 results on adult cancer patients better here we assess the impact of these changes on data quality and on survival comparisons. Methods In EUROCARE-5 the survival differences by area were studied applying the complete cohort approach to data on nearly nine million cancer patients diagnosed in 2000–2007 and followed up to 2008. Survival time trends were analysed applying the period approach to data on about 10 million cancer cases diagnosed from 1995 to 2007 and followed up to 2008. Differently from EUROCARE-4, multiple primary cancers were included and relative survival was estimated with the Ederer II method. Results EUROCARE-5 covered a population of 232 million resident persons, corresponding to 50% of the 29 participating countries. The population coverage increased particularly in Eastern Europe. Cases identified from death certificate only (DCO) were on average 2.9%, range 0–12%. Microscopically confirmed cases amounted to over 85% in most CRs. Compared to previous methods, including multiple cancers and using the Ederer II estimator reduced survival estimates by 0.4 and 0.3 absolute percentage points, on average. Conclusions The increased population size and registration coverage of the EUROCARE-5 study ensures more robust and comparable estimates across European countries. This enlargement did not impact on data quality, which was generally satisfactory. Estimates may be slightly inflated in countries with high or null DCO proportions, especially for poor prognosis cancers. The updated methods improved the comparability of survival estimates between recently and long-term established registries and reduced biases due to informative censoring.
