pH-Sensitive dextran derivatives having 3-methylglutarylated residues (MGlu-Dex) were prepared by reacting dextran with 3-methyl-glutaric anhydride. MGlu-Dex changed the protonation state and their characteristics from hydrophilic to hydrophobic in neutral and acidic pH regions. Surface modification of egg yolk phosphatidylcholine liposomes with MGlu-Dex produced highly pH-sensitive liposomes that were stable at neutral pH but which were destabilized strongly in the weakly acidic pH region. MGlu-Dex-modified liposomes were taken up efficiently by dendritic cells and delivered entrapped ovalbumin (OVA) molecules into the cytosol. When MGlu-Dex-modified liposomes loaded with OVA were administered subcutaneously to mice, the antigen-specific humoral and cellular immunity was induced more effectively than the unmodified liposomes loaded with OVA. Furthermore, administration of MGlu-Dex-modified liposomes loaded with OVA to mice bearing E.G7-OVA tumor significantly suppressed tumor growth and extended the mice survival. Results suggest that MGlu-Dex-modified liposomes are promising for the production of safe and potent antigen delivery systems that contribute to the establishment of efficient cancer immunotherapy., (Copyright © 2013 Elsevier Ltd. A new family of poly(anhydrides) (PA) has been developed which can be cured photochemically to produce degradable networks. These degradable anhydride networks may be useful in orthopaedics as bone cements and as matrices for drug delivery. This system, which is a semi-interpenetrating network (semi-IPN), has been evaluated for biocompatibility in subcutaneous tissue in rats and appears to undergo degradation primarily by surface erosion. The inflammatory response to the semi-IPN implants was minimal at both short (3 and 6 weeks) and long (28 weeks) time points and the fibrotic response was largely absent throughout the duration of this study. Furthermore, the OrthoCure implant material integrated well with the surrounding tissue and was invaded with vascularized connective tissue. For reference, linear PA controls were tested and showed a foreign body response culminating in the formation of relatively avascular fibrous capsule several cell layers thick, which became thicker over time, a response similar to what is typically observed in FDA approved implantable polymeric device systems.

A new approach for the in situ formation of porosity in a matrix based on differential polymer degradation has been studied. This approach exploits the differences in polymer properties such as molecular weight, hydrophilicity (hydrophobicity), and degradation to induce preferential degradation of one phase in a biphasic polymer system. Biphasic polymer systems polymers derived from alpha-hydroxy acids and poly(anhydrides), which vary in their erosion characteristics (surface vs. bulk) and hydrophobicities were studied. In addition to examining the generality of the approach, potential advantages of such systems in the context of tissue engineering and drug delivery are briefly discussed.

A novel biomaterial: poly(sebacic anhydride-co-ethylene glycol) was synthesized by introducing poly(ethylene glycol) (PEG) into a polyanhydride system. This copolymer was synthesized using sebacic acid and PEG via melt-condensation polymerization. The crystalline behavior of these synthesized products was studied, and compared to that of polymer blends of poly(sebacic anhydride) (PSA) and PEG. The crystallinity of PSA chain segments can be significantly enhanced by increasing chain mobility via the introduction of PEG. The crystallinity of the PSA component in copolymers was substantially greater than that of blends. However, the crystalline growth of the PEG segments was totally hindered by the presence of PSA chain segments, such that no crystal for PEG component was found in these copolymers. Besides, a dynamic mechanical analysis of these materials was also performed to provide additional information concerning visco-elastic behavior for other biomedical applications, where it was found that the viscous behavior in copolymers was more significant than in neat PSA and PEG., (Copyright 2002 Elsevier Science Ltd.)

Two classes of fluorescent copolyanhydrides were synthesized by melt copolycondensation of the fluorophoric diacid, p-(carboxyethylformamido)benzoic acid (CEFB), with sebacic acid (SA) or N-trimellitylimidoglycine (TMA-gly). Alternate copolyanhydride based on SA and CEFB was also prepared by solution polycondensation of CEFB and sebacoyl chloride. 'H NMR spectra of the copolymers confirm their structures. Fluorescence was observed for all the copolymers, the intensity of which increases with the CEFB fraction. Either blue (ca. 429 nm) or green (ca. 520 nm) light can be emitted from the copolymers with the excitation of UV (356 nm) and visible (470 nm) light, respectively. The polymers were further fabricated to microsphere formulation, which can be clearly visualized by fluorescent microscopy.

A new family of alternate poly(ester-anhydrides) containing aliphatic and aromatic diacids were synthesized. The dicarboxylic acids were obtained by derivatization of p-hydroxy benzoic acid at the hydroxy terminus with cyclic anhydride (adipic anhydride and succinic anhydride) and subsequently polymerized via the corresponding mixed anhydrides by melt polycondensation. DSC traces revealed that the polymers had low Tg (< 40 degrees C) and no crystallinity. The static contact angle measurements indicated that the poly(ester-anhydrides) were more hydrophobic than poly(D,L-lactide) and poly(adipic anhydride). In vitro degradation of the polymers was also investigated in pH 7.4 PBS at 37 degrees C. It was found that degradation rate of the poly(ester-anhydrides) increased with p-carboxy phenyl adipic monoester (CPA) content in the polymers and the degradation duration could be adjusted from ca. 20 days to ca. 2 months. Erosion curve of poly(p-carboxy phenyl adipic monoester anhydride) (PCPA) was characterized by a linear region of weight loss at nearly constant rate in the first 7 days (ca. 80% of weight loss) followed by a gradual decrease region. IR and SEM analysis showed that significant erosion of PCPA occurred in the outer layer and no apparent erosion could be seen in the inner layer of the degrading sample after 7-day degradation. The poly(ester-anhydrides) may be used as either anti-infective polymeric prodrugs or matrices for drug delivery.

A biodegradable poly(anhydride-ester) was synthesized by melt condensation polymerization of the acetylated monomer to yield a novel polymeric prodrug. The polymer we have synthesized is composed of alkyl chains linked by ester bonds to aromatic moieties, specifically salicylic acid--the active component of aspirin. With the medicinal properties attributed to salicylic acid and the ease of metabolism, the incorporation of this compound into a polymer backbone yields a polymeric prodrug that may have potential in a variety of applications (i.e., inflammatory bowel disease). For these reasons, we have designed a synthetic scheme that yields the desired poly(anhydride-ester). The in vitro hydrolytic degradation of these polymers has been performed and results indicate that the polymer degradation rate is pH-dependent.

Polymer microspheres capable of the controlled release of macromolecules for periods ranging from days to over a month were developed. The microspheres were made using a new family of anhydride polymers: tyrosine-containing poly(anhydride-co-imides), specifically poly[trimellitylimido-L-tyrosine-co-sebacic acid-co-1,3-bis(carboxphenoxy)propane] anhydrides [poly(TMA-Tyr:SA:CPP)]. These polymers may be of particular interest for controlled delivery of vaccine antigens due to the incorporation of an immunological adjuvant, L-tyrosine, into their backbone. Microspheres were produced from a variety of polymer compositions using a double-emulsion solvent-evaporation technique, and tested for their ability to provide controlled release of a model protein, bovine serum albumin, in vitro. The microspheres are spherical with smooth surfaces and encapsulate greater than 70% of the protein. Protein release rates from polymers of identical composition could be varied from 0.3 to over 125 micrograms per mg spheres per month by changing the amount of protein encapsulated. This effect can be magnified by using polymers with various monomer ratios. A close correlation between protein release and polymer weight loss was observed, suggesting a release mechanism controlled mainly by polymer erosion. Bovine serum albumin release from poly(TMA-Tyr:SA:CPP) microspheres is also pH sensitive, being enhanced at high pH and depressed under acidic conditions.

The aim of this work was to study the cross-linkage of xerogels made of Carbopol of different molecular weight (polymers deriving from polyacrylic acid), and the influence exerted by different percentages of beta-cyclodextrin upon the cross-linkage percentage. Xerogels were obtained by high-pressure compression and cross-linking was studied by measuring the weight loss of different formulations over a temperature range of 70 to 190 degrees C. Kinetics of anhydride formation were found to follow a second-order mechanism with an activation energy of approximately 25 kcal mol-1.

The objective of the present study was to investigate the properties of branched polyanhydrides and compare them to the corresponding linear polymers. Sebacic acid was polymerized with 1,3,5 benzenetricarboxylic acid and poly(acrylic acid) to yield random and graft-type branched polyanhydrides. The polymerization was followed until the gel point and the resulting polymers were evaluated for their physico-chemical properties and degradation behaviour. Drug release from these polymers was studied using morphine as a model drug. The experiment showed that the molecular weights of branched polyanhydride were significantly higher (mol wt 250,000) than the molecular weight of linear poly(sebacic anhydride) (mol wt 80,000). In the case of poly(acrylic acid) branched polymers, the molecular weight increased linearly with increasing concentration of poly(acrylic acid). The specific viscosities of the branched polyanhydrides were lower than linear polyanhydrides with similar molecular weights. Except for the difference in molecular weights, there were no noticeable changes in the physico-chemical or thermal properties of the branched polymers and the linear poly(sebacic anhydride). The degradation of the branched polyanhydride was triphasic and the degradation rates were faster than for linear poly(sebacic anhydride). The release of morphine from the branched polymers was lower than the corresponding poly(sebacic anhydride). Release of morphine was much higher from the poly(acrylic acid) branched polymers compared to the 1,3,5 benzenetricarboxylic acid branched polymers and increased with increasing concentrations of the branching agent. However, in both cases the release rates and the total amounts of morphine released approached that of poly(sebacic anhydride).
