The systematic microbiological evaluation of endophthalmitis allows the confirmation of the infectious nature of the disease and the possible adaptation of treatment at the individual level and, at the collective level, the epidemiological characterization of the bacterial spectrum of endophthalmitis. Long reserved for research, the use of molecular biology techniques to complement conventional culture techniques has become important for the diagnosis of endophthalmitis in recent years. These new diagnostic techniques are particularly useful for the microbiological study of bacteria that are difficult or impossible to grow because of their intrinsic properties, their presence in only a small inoculum, their sequestration on prosthetic materials, or their inactivation by prior antibiotic treatment. These techniques are based on the polymerase chain reaction (PCR), which allows the amplification and detection of extracted bacterial deoxyribonucleic acid (DNA) that is initially present in minute quantities in an ocular sample. In practice, these conventional or real-time PCRs allow either the a priori detection of bacterial DNA (universal PCR) or the identification of a specific DNA fragment of a bacterial genus or species (specific PCR). We tested that the breath-by-breath V(O2) determination by an algorithm termed BR did not bias the kinetics parameters. It was compared to two other algorithms using a correction for changes in lung gas stores between two successive breaths (AU, W) and one without correction (NC). Ten healthy male subjects cycled 10 min at 15 and 30 W below and above ventilatory threshold (VT) after 3 min at 0 W. The breath-by-breath V(O2) variability was lower with BR than the other methods during the last 3 min at each power. V(O2) kinetics was described by a mono-exponential model at power below VT and a bi-exponential model above VT. Differences in parameter were only observed for the primary component between estimates using AU and NC. The between-subject variability in the parameters of the slow component at 15 W above VT was lower with AU and BR than W and NC. Platelets have long been confined to haemostasis only. However, novel functions for platelets have been identified recently. Those non-nucleated cells indeed participate to inflammation and also they produce and release numerous factors with known immunomodulatory functions. Among those factors are cytokines and chemokines and the like, such as soluble CD40-Ligand (CD154), which are key molecules in that they bridge innate and adaptative immunity; sCD40L is active on T cells, B cells, monocytes and macrophages, dendritic cells and endothelial cells lining the blood vessels. This means that when a platelet concentrate is transfused to a recipient, a huge amount of cytokines and chemokines is also infused. In this state of the art review, we will present arguments on the role of platelet secretory products in modulating cellular parameters of immunity, and--very likely--in altering functions of those immune cells upon encounters while infusing platelets in blood recipients. We aimed at summarizing data that have been made available on the issue of cytokines/chemokines released by stored platelets prior to delivery. We will focus on the suspected role of the CD40/CD40L tandem in postplatelet transfusion reactions or incidents. Many studies have demonstrated that contact time is a key factor affecting both the energetics and mechanics of running. The purpose of the present study was to further explore the relationships between contact time (t(c)), step frequency (f) and leg stiffness (k(leg)) in human running. Since f is a compound parameter, depending on both contact and aerial time, the specific goal of this study was to independently vary f and t(c) and to investigate their respective effects on spring-mass characteristics during running, seeking to determine if the changes in k(leg) observed when running at different f are mainly due to inherent changes in t(c). We compared three types of constant 3.33 m s(-1) running conditions in 10 male subjects: normal running at the subject's freely chosen f, running with decreased and increased f, and decreased and increased t(c) at the imposed freely chosen f. The data from the varied f trials showed that the variation of t(c) was strongly correlated to that of k(leg) (r(2)=0.90), and the variation of f was also significantly correlated to that of k(leg) (r(2)=0.47). Further, changes in t(c) obtained in various t(c) conditions were significantly correlated to changes in k(leg) (r(2)=0.96). These results confirm that leg stiffness was significantly influenced by step frequency variations during constant speed running, as earlier demonstrated, but our more novel finding is that compared to step frequency, the effect of contact time variations appears to be a stronger and more direct determinant of k(leg). Indeed, 90-96% of the variance in k(leg) can be explained by contact time, whether this latter parameter is directly controlled, or indirectly controlled through its close relationship with step frequency. In conclusion, from the comparison of two experimental procedures, i.e. imposing various step frequency conditions vs. asking subjects to intentionally vary contact time at their freely chosen step frequency, it appears that changes in leg stiffness are mainly related to changes in contact time, rather than to those in step frequency. Step frequency appears to be an indirect factor influencing leg stiffness, through its effect on contact time, which could be considered a major determinant of this spring-mass characteristic of human running. more...

The aim of this study was to propose and validate a post-hoc correction method to obtain maximal power values taking into account inertia of the flywheel during sprints on friction-loaded cycle ergometers. This correction method was obtained from a basic postulate of linear deceleration-time evolution during the initial phase (until maximal power) of a sprint and included simple parameters as flywheel inertia, maximal velocity, time to reach maximal velocity and friction force. The validity of this model was tested by comparing measured and calculated maximal power values for 19 sprint bouts performed by five subjects against 0.6-1 N kg(-1) friction loads. Non-significant differences between measured and calculated maximal power (1151+/-169 vs. 1148+/-170 W) and a mean error index of 1.31+/-1.20% (ranging from 0.09% to 4.20%) showed the validity of this method. Furthermore, the differences between measured maximal power and power neglecting inertia (20.4+/-7.6%, ranging from 9.5% to 33.2%) emphasized the usefulness of power correcting in studies about anaerobic power which do not include inertia, and also the interest of this simple post-hoc method. more...

Antibodies are characteristically induced in many parasitic infection processes. The class and subclass of the antibody response is instrumental because each isotype has a distinct biological function. It is thus crucially important for an infected individual to mount the most appropriate secondary antibody response--that is the response that has the best chance of clearing the infection and/or controlling disease. This represents a fundamental of vaccine strategies. Immuno-epidemiological surveys and in vitro models of antibody production have helped to understand some of the goals which should be achieved when designing antiparasitic vaccines. more...

Background: Mab 5-D8/1 is a monoclonal antibody (Mab) that was shown to be directed towards a conserved epitope of the capsid protein VP1 among the genus enterovirus. The use of this Mab for the routine detection of enteroviruses in clinical specimens led to the observation that several strains of echovirus type 11 (EV-11) could not be detected on spontaneously detached cells from 26-h cultures using a two-step immunofluorescence (IF) assay. Conversely, these strains were detected positive with the same Mab when tested on adherent or trypsinizated cells., Objectives: The aim of this study was to understand the misrecognition of some strains of EV-11 by this Mab., Study Design and Results: IF tests at different times of the viral cycle brought evidence that the detection of a variant strain of EV-11 decreased rapidly with time, becoming undetectable 26 h post-infection, since the reference strain remained positive up to 46 h post-infection. The infective titres of the variant strains were shown to be high in comparison with those of well-recognised strains. Sequencing the Mab binding epitope confirmed that the variant strains exhibited no antigenic shift., Conclusion: These results suggest that the poor recognition of some strains of EV-11 by Mab 5-D8/1 is due to a rapid decrease of the expression of the binding epitope in the cell, maybe in relation with the high lytic power of these strains. From a practical point of view, our data indicate that a negative result when Mab 5-D8/1 is used for enterovirus typing must be interpreted cautiously with highly replicative strains and that detached cells should not be used for enterovirus identification under these circumstances. more...

Clodronate can be used in animals to prevent the effects of immobilization on bone. For this reason we have studied the effects of this agent on immobilisation bone loss in man. We administered clodronate by mouth to 14 paraplegic patients (400 mg/d, n = 7; 1600 mg/day, n = 7), and compared its effect with a placebo (n = 7). Treatment was given for 100 days, 5-29 days after spinal cord injury. Our results suggest that clodronate, given early after immobilization, prevents the acute bone loss observed in immobilization as judged by its effects on serum and urine calcium and hydroxyproline, bone mineral content, trabecular bone volume, and the number of osteoclasts present in bone. No mineralization defect or other side-effects were observed during or after treatment. In addition, a total of 70 patients, with comparable degrees of immobilization, were studied with a variety of antiosteoclastic drugs comprising controls (n = 16), etidronate (n = 20), salmon calcitonin (n = 20) and clodronate 400 mg/d (n = 7) or 1600 mg/d (n = 7). Clodronate, at the dose of 1600 mg/d appeared the most effective drug on bone resorption, together with calcitonin. Unlike calcitonin, clodronate can be administered orally. The mineralisation defects observed during prolonged treatment with etidronate at high doses were not observed with clodronate. We conclude that clodronate 1600 mg/d is a promising agent for the treatment of bone loss and the resorptive hypercalcaemia and hypercalciuria noted in immobilisation. more...
