Tesis para obtener el grado de Doctor en Ciencias Agrarias, de la Universidad Nacional de Mar del Plata, en noviembre de 2022 La bacteria Campylobacter fetus, es uno de los principales agentes etiológicos que provocan pérdidas reproductivas en bovinos en Argentina. Hay dos subespecies de C. fetus relevantes para la salud del bovino: C. fetus subsp. fetus (Cff) que habita el tracto gastrointestinal y causa abortos ocasionales tras bacteriemia materna, y C. fetus subsp. venerealis (Cfv) (incluye el biovar intermedius; Cfvi), que es huésped exclusivo del tracto genital y causante de la campilobacteriosis genital bovina (CGB), una enfermedad de transmisión sexual que causa bajos porcentajes de preñez. Se estudió la infección de las 3 subespecies de C. fetus (Cff, Cfv y Cfvi) y el comportamiento de técnicas diagnósticas convencionales y moleculares en un modelo experimental bovino. En el mismo se infectaron experimentalmente vaquillonas con la subespecie de C. fetus respectiva, y se realizó servicio natural con toros sanos. Se recolectaron muestras de mucus cérvico vaginal (MCV) de las vaquillonas y esmegma prepucial (EP) de toros durante un período de 9 meses, y se procesaron por cultivo bacteriológico, IFD y qPCR (dos métodos de extracción de ADN; In-house y kit comercial), con el objetivo de evaluar el método de diagnóstico adecuado y clarificar la patogénesis de las subespecies. El 87% de las vaquillonas resultó infectada tras inoculación experimental, y al menos una por cada subespecie se mantuvo infectada durante todo el ensayo (285 días). En MCV el cultivo bacteriológico fue la técnica más adecuada (43%). Su consistencia viscosa fue una limitación para la qPCR, donde la recuperación de C. fetus con el método de extracción de ADN con kit comercial fue baja (29%), mientras el método de lisis In-house de lisis resultó inadecuada (8%). Todos los toros se infectaron tras servicio natural con respectiva subespecie. La qPCR con extracción de ADN In-house (75%) resultó una alternativa prometedora en EP, comparado con la IFD (63%) actualmente utilizada de rutina. En casos naturales de 7 establecimientos, la detección de C. fetus de MCV (n=53) resultó baja siendo el cultivo la técnica más adecuada (13%) seguida de qPCR (7,5%) e IFD (5,6%). Esto se asocia a la capacidad autolimitante de la bacteria y el tiempo de muestreo. De estos establecimientos se analizaron seis fetos abortados, todos identificados como Cff. La diferenciación de subespecie por métodos moleculares y pruebas bioquímicas fueron inconsistentes siendo de escaso valor, por lo que es necesario avanzar en estudios que permitan diferenciar las mismas de forma correcta, y que confirmen la relevancia clínico-epidemiológica de cada subespecie en bovinos. Por primera vez se reporta el aislamiento de Cfv desde vesículas seminales de un toro naturalmente infectado. Este hallazgo pone de manifiesto otro probable nicho que permita la persistencia de la bacteria y posible contaminación del semen. Se realizaron otros dos ensayos experimentales con Cff para estudiar su patogénesis, tras la alta persistencia observada en el modelo experimental, comprobándose la transmisión venérea entre vaquillonas y toros. El uso de qPCR en muestras clínicas resultan prometedoras en EP. Más estudios sobre los métodos de extracción de ADN y diagnóstico molecular son necesarios en muestras de MCV para mejorar su eficiencia. La capacidad venérea, la colonización y la persistencia de Cff en el tracto reproductor de vaquillonas y toros, y alta presencia en fetos bovinos abortados evidencia un alto riesgo en la salud reproductiva de los bovinos, sugiriendo sea incluida en la CGB junto al Cfv. The bacterium Campylobacter fetus is one of the main etiological agents that cause reproductive losses in cattle in Argentina. There are two subspecies of C. fetus relevant to bovine health: C. fetus subsp. fetus (Cff) which inhabits the gastrointestinal tract and causes occasional abortions after maternal bacteremia, and C. fetus subsp. venerealis (Cfv) (includes the biovar intermedius; Cfvi), which is an exclusive host of the genital tract and causes bovine genital campylobacteriosis (BGC), a sexually transmitted disease that causes low pregnancy rates. The infection of the 3 subspecies of C. fetus (Cff, Cfv and Cfvi) and the use of conventional and molecular diagnostic techniques in a bovine experimental model were studied. In it, heifers were experimentally infected with the respective subspecies of C. fetus, and natural mating was carried out with healthy bulls. Samples of cervical vaginal mucus (CVM) from heifers and preputial smegma (PS) from bulls were collected over a 9-month period, and processed by bacteriological culture, DIF and qPCR (two DNA extraction methods; In-house and commercial kit), with the aim of evaluating the appropriate diagnostic method and clarifying the pathogenesis of the subspecies. An 87% of the heifers were infected after experimental inoculation, and at least one for each subspecies remained infected throughout the trial (285 days). In CVM, bacteriological culture was the most appropriate technique (43%). Its viscous consistency was a limitation for qPCR, where the recovery of C. fetus with the DNA extraction method with a commercial kit was low (29%), while the In-house lysis method of lysis was inadequate (8%). All bulls were infected after natural mating with their respective subspecies. The qPCR with in-house DNA extraction (75%) was a promising alternative for detection of C. fetus in PS, compared to DIF (63%) currently used routinely. Diagnostic techniques were evaluated in natural cases from 7 cattle farms, where CVM (n=53) and aborted fetuses (n=6) were analyzed. The detection of C. fetus from CVM was low, with culture being the most appropriate technique (13%) followed by qPCR (7.5%) and DIF (5.6%). This is associated with the self-limiting capacity of the bacteria and the sampling time. All fetuses analyzed were identified as Cff. The differentiation of subspecies by molecular methods and biochemical tests were inconsistent and of low value, so it is necessary to advance in studies that allow them to be correctly differentiated, and that confirm the clinical-epidemiological relevance of each subspecies in cattle. For the first time, the isolation of Cfv from the seminal vesicles of a naturally infected bull is reported. This finding highlights another possible niche that allows the persistence of the bacteria and possible contamination of the semen. Two other experimental tests with Cff were carried out to study its pathogenesis, after the high persistence observed in the experimental model, verifying the venereal transmission between heifers and bulls. The use of qPCR in clinical samples is promising in PS samples. More studies on DNA extraction and molecular diagnostic methods are needed on CVM samples to improve their efficiency. The venereal transmissión, and colonization and persistence of Cff in the reproductive tract of heifers and bulls, with the high presence in aborted bovine fetuses show a high risk to bovine reproductive health, suggesting that it be included in the BGC along with Cfv.
