This review analyzes methods for controlling plant viral infection. The high harmfulness of viral diseases and the peculiarities of viral pathogenesis impose special requirements regarding developing methods to prevent phytoviruses. The control of viral infection is complicated by the rapid evolution, variability of viruses, and the peculiarities of their pathogenesis. Viral infection in plants is a complex interdependent process. The creation of transgenic varieties has caused much hope in the fight against viral pathogens. The disadvantages of genetically engineered approaches include the fact that the resistance gained is often highly specific and short-lived, and there are bans in many countries on the use of transgenic varieties. Modern prevention methods, diagnosis, and recovery of planting material are at the forefront of the fight against viral infection. The main techniques used for the healing of virus-infected plants include the apical meristem method, which is combined with thermotherapy and chemotherapy. These methods represent a single biotechnological complex method of plant recovery from viruses in vitro culture. It widely uses this method for obtaining non-virus planting material for various crops. The disadvantages of the tissue culture-based method of health improvement include the possibility of self-clonal variations resulting from the long-term cultivation of plants under in vitro conditions. The possibilities of increasing plant resistance by stimulating their immune system have expanded, which results from the in-depth study of the molecular and genetic bases of plant resistance toward viruses and the investigation of the mechanisms of induction of protective reactions in the plant organism. The existing methods of phytovirus control are ambiguous and require additional research. Further study of the genetic, biochemical, and physiological features of viral pathogenesis and the development of a strategy to increase plant resistance to viruses will allow a new level of phytovirus infection control to be reached.

Heavy metal concentrations exceeding permissible limits threaten human life, plant life, and all other life forms. Different natural and anthropogenic activities emit toxic heavy metals in the soil, air, and water. Plants consume toxic heavy metals from their roots and foliar part inside the plant. Heavy metals may interfere with various aspects of the plants, such as biochemistry, bio-molecules, and physiological processes, which usually translate into morphological and anatomical changes. They use various strategies to deal with the toxic effects of heavy metal contamination. Some of these strategies include restricting heavy metals to the cell wall, vascular sequestration, and synthesis of various biochemical compounds, such as phyto-chelators and organic acids, to bind the free moving heavy metal ions so that the toxic effects are minimized. This review focuses on several aspects of genetics, molecular, and cell signaling levels, which integrate to produce a coordinated response to heavy metal toxicity and interpret the exact strategies behind the tolerance of heavy metals stress. It is suggested that various aspects of some model plant species must be thoroughly studied to comprehend the approaches of heavy metal tolerance to put that knowledge into practical use.

Diarrhea is a severe bovine disease, globally prevalent in farm animals with a decrease in milk production and a low fertility rate. Cryptosporidium spp. are important zoonotic agents of bovine diarrhea. However, little is known about microbiota and short-chain fatty acids (SCFAs) changes in yaks infected with Cryptosporidium spp. Therefore, we performed 16S rRNA sequencing and detected the concentrations of SCFAs in Cryptosporidium-infected yaks. Women entrepreneurs are significant contributors to the economic development of any country and their role becomes more vital in improving the economic condition of developing countries. This highlights the important role of women-owned small and medium enterprises (SMEs) and their entrepreneurial success. Therefore, the current study extends the entrepreneurship literature by examining the effects of factors like personality traits (PT), motivation and commitment (MC), availability of financial resources (AFR), and government support (GS) on entrepreneurial success (ES) of women-owned SMEs. Using a purposive sampling technique data from 255 women-owned SMEs were collected. A multi-analytical approach was employed to analyze the data. The Structural equation modeling (SEM) results indicated that PT, MC, AFR, and GS have a direct effect of ES whereas MC also mediated the link between PT and ES, and the results reveal that in presence of MC the effects of PT on ES become more significant. SEM results revealed that PT and AFR are the most important factors related to entrepreneurial success. On the contrary, ANN analysis revealed that “motivation and commitment” is the most influencing factor. These findings can guide business practitioners and policymakers in the envisioned strategy formulation to encourage women entrepreneurs who can contribute to their country’s sustainable economic growth.

A high amount of CO2 causes numerous health effects, including headaches, restlessness, difficulty in breathing, increased heart rate, high blood pressure, asphyxia, and dizziness. This issue of increasing atmospheric CO2 can only be solved via above-ground and below-ground carbon sequestration (CS). This study was designed to determine the relationship between CS with the crown area (CA), diameter at breast height (DBH), height (H), species richness (SR), and elevation in different forest types of Pakistan with the following specific objectives: (1) to quantify the direct and indirect relationship of carbon sequestration with CA, DBH, H, and SR in various natural forest types and (2) to evaluate the effect of elevation on the trees functional traits and resultant CS. We used the linear structural equation model (SEM) for each conceptual model. Our results confirmed that the highest CS potential was recorded for dry temperate conifer forests (DTCF) i.e., 52.67%, followed by moist temperate mix forests (MTMF) and sub-tropical broad-leaved forests (STBLF). The SEM further described the carbon sequestration variation, i.e., 57, 32, 19, and 16% under the influence of CA (β = 0.90 and P-value < 0.001), H (β = 0.13 and p-value = 0.05), DBH (β = 0.07 and p-value = 0.005), and SR (β = −0.55 and p-value = 0.001), respectively. The individual direct effect of SR on carbon sequestration has been negative and significant. At the same time, the separate effect of CA, DBH, and H had a positive and significant effect on carbon sequestration. The remaining 20% of CS variations are indirectly influenced by elevation. This means that elevation affects carbon sequestration indirectly through CA, DBH, H, and SR, i.e., β = 0.133 and P-value < 0.166, followed by β = 0.531 and P-value < 0.001, β = 0.007 and P-value < 0.399, and β = −0.32 and P-value < 0.001, respectively. It is concluded that abiotic factors mainly determined carbon sequestration in forest ecosystems along with the elevation gradients in Pakistan. Quantifying the role of various forest types in carbon dioxide (CO2) reduction leads to improved air quality, which positively impacts human health. This is an imperative and novel study that links the dynamics of the biosphere and atmosphere.

Staphylococcus aureus is recognized as commensal as well as opportunistic pathogen of humans and animals. Methicillin resistant strain of S. aureus (MRSA) has emerged as a major pathogen in hospitals, community and veterinary settings that compromises the public health and livestock production. MRSA basically emerged from MSSA after acquiring SCCmec element through gene transfer containing mecA gene responsible for encoding PBP-2α. This protein renders the MRSA resistant to most of the β-lactam antibiotics. Due to the continuous increasing prevalence and transmission of MRSA in hospitals, community and veterinary settings posing a major threat to public health. Furthermore, high pathogenicity of MRSA due to a number of virulence factors produced by S. aureus along with antibiotic resistance help to breach the immunity of host and responsible for causing severe infections in humans and animals. The clinical manifestations of MRSA consist of skin and soft tissues infection to bacteremia, septicemia, toxic shock, and scalded skin syndrome. Moreover, due to the increasing resistance of MRSA to number of antibiotics, there is need to approach alternatives ways to overcome economic as well as human losses. This review is going to discuss various aspects of MRSA starting from emergence, transmission, epidemiology, pathophysiology, disease patterns in hosts, novel treatment, and control strategies.

This study assessed the floristic composition, biological spectrum, and phytogeographical elements of the Bin Dara western boundary of Pakistan. The flora consisted of 140 plant species belonging to 47 families. The most dominant family was Poaceae (with 14 species), followed by Asteraceae (11 species). The biological spectrum showed the therophytes (71 species, 50.71%) as the dominant life form class, followed by mega-phanerophytes (27 species, 19.28%), nano-phanerophytes (15 species, 10.71%), geophytes (10 species, 7.14%), chamaephytes (9 species, 6.42%), and hemi-cryptophytes (8 species, 5.71%). Leaf size classes comprised of nanophylls (48 species, 34.28%), microphylls (32 species, 22.85%), mesophylls (22 species, 15.71%), macrophylls (17 species, 12.14%), leptophylls (12 species, 8.57), megaphylls (8 species, 5.75%), and aphyllous (1 species, 0.71%). Cosmopolitan was the most prominent form of phytogeographic elements (with 17 species, 12.14%), followed by pantropical and Euro-Siberian Mediterranean Irano-Turanian (14 species, 10%) each. The pluriregional represented (13 species, 9.28%), Holarctic and Mediterranean Irano-Turanian (10 species, 7.14%), Irano-Turanian and Western Himalayan (9 species, 6.42%) each. It is recommended that further study is needed to map the vegetation, its indicators, and rare species that face a huge threat of endangerment.

Diarrhea is a word-widely severe disease coupled with gastrointestinal dysfunction, especially in cattle causing huge economic losses. However, the effects of currently implemented measures are still not enough to prevent diarrhea. Previously we found that dropped short-chain fatty acids in diarrhea yaks, and butyrate is commonly known to be related to the epithelial barrier function and intestinal inflammation. However, it is still unknown whether sodium acetate/sodium butyrate could alleviate diarrhea in animals. The present study is carried out to explore the potential effects of sodium acetate/sodium butyrate on lipopolysaccharide-induced diarrhea in mice. Fifty ICR mice were randomly divided into control (C), LPS-induced (L), and sodium acetate/sodium butyrate (D, B, A)-treated groups. Serum and intestine samples were collected to examine inflammatory cytokines, antioxidant levels, relative gene expressions via real-time PCR assay, and gut microbiota changes through high-throughput sequencing. Results indicated that LPS decreased the villus height (p < 0.0001), increased the crypt depth (p < 0.05), and lowered the villus height to crypt depth ratio (p < 0.0001), while sodium acetate/sodium butyrate supplementation caused a significant increase in the villus height (p < 0.001), decrease in the crypt depth (p < 0.01), and increase in the villus height to crypt depth ratio (p < 0.001), especially. In mice treated with LPS, it was found that the serum level of IL-1β, TNF-α (p < 0.001), and MDA (p < 0.01) was significantly higher; however, sodium acetate/sodium butyrate supplementation significantly reduced IL-1β (p < 0.001), TNF-α (p < 0.01), and MDA (p < 0.01), respectively. A total of 19 genera were detected among mouse groups; LPS challenge decreased the abundance of Lactobacillus, unidentified F16, unidentified_S24-7, Adlercreutzia, Ruminococcus, unclassified Pseudomonadales, [Ruminococcus], Acetobacter, cc 1, Rhodococcus, unclassified Comamonadaceae, Faecalibacterium, and Cupriavidus, while increased Shigella, Rhodococcus, unclassified Comamonadaceae, and unclassified Pseudomonadales in group L. Interestingly, sodium acetate/sodium butyrate supplementation increased Lactobacillus, unidentified F16, Adlercreutzia, Ruminococcus, [Ruminococcus], unidentified F16, cc 115, Acetobacter, Faecalibacterium, and Cupriavidus, while decreased Shigella, unclassified Enterobacteriaceae, unclassified Pseudomonadales, Rhodococcus, and unclassified Comamonadaceae. LPS treatment upregulated the expressions of ZO-1 (p < 0.01) and NLRP3 (p < 0.0001) genes in mice; however, sodium acetate/sodium butyrate solution supplementation downregulated the expressions of ZO-1 (p < 0.05) and NLRP3 (p < 0.05) genes in treated mice. Also, the LPS challenge clearly downregulated the expression of Occludin (p < 0.001), Claudin (p < 0.0001), and Caspase-1 (p < 0.0001) genes, while sodium acetate/sodium butyrate solution supplementation upregulated those gene expressions in treated groups. The present study revealed that sodium acetate/sodium butyrate supplementation alleviated LPS-induced diarrhea in mice via enriching beneficial bacterium and decreasing pathogens, which could regulate oxidative damages and inflammatory responses via NLRP3/Caspase-1 signaling. The current results may give insights into the prevention and treatment of diarrhea.

Arisaema jacquemontii Blume is a highly medicinal and poisonous plant belong to the family Araceae. It is used to treat several deadly diseases, including viral infections. It has antioxidant, anti-cancerous, antimalarial, anti-vermicidal, and antiviral activities. Therefore, five parts of the Arisaema jacquemontii Blume plant, such as leaf, seed, stem, pulp, and rhizome extract, were evaluated for metabolic and in silico characterization of probable compounds using gas chromatography-mass spectrometry (GC-MS) analysis. A total of 22 compounds were isolated from the methanolic extracts of A. jacquemontii Blume. A selected antiviral COVID-19 protein i.e., protease (6LU7) was docked against the obtained compounds. Different affinities were obtained through various compounds. The best results were shown by three different compounds identified in the rhizome. The maximum binding affinity of these compounds is 8.1 kJ/mol. Molecular docking (MD) indicate that these molecules have the highest binding energies and hydrogen bonding interactions. The binding mode of interaction was discovered to be reasonably effective for counteracting the SARS virus COVID-19. The findings of this study could be extremely useful in the development of more phytochemical-based COVID-19 therapeutics.

It is critical to characterize changes in the structure and composition of the host fungal community in natural Cryptosporidium infection, because it gives the possible overview of gut microbiome in host homeostasis and disease progression. A total of 168 rectal fecal samples were collected and examined using nPCR. The positive samples were double-checked using 18S rDNA high-throughput sequencing. After confirmation, ITS high-throughput sequencing was utilized to investigate the fungal community’s response to natural Cryptosporidium infection. Results showed that a total of three positive samples (1.79%) were identified with an increased abundance of fungi associated with health hazards, such as class Dothideomycetes, families, i.e., Cladosporiaceae, Glomerellaceae, and genera, i.e., Wickerhamomyces, Talaromyces, Cladosporium, Dactylonectria, and Colletotrichum. On the contrary, taxa associated with favorable physiological effects on the host were shown to have the reverse impact, such as families, i.e., Psathyrellaceae, Pseudeurotiaceae and genera (Beauveria, Nigrospora, and Diversispora). For the first time, we evaluated the condition of natural Cryptosporidium infection in horses in Wuhan, China, and discovered distinct variations in the fungal microbiome in response to natural infection. It might prompt a therapy or prevention strategy to apply specific fungal microorganisms that are probably responsible for decreased susceptibility or increased resistance to infection.

Staphylococcus aureus (S. aureus) has become a leading animal and public health pathogen that keeps on transferring from one host to other, giving rise to newer strains by genetic shifts. The current study was designed to investigate the epidemiology and genetic relatedness of mecA gene in S. aureus isolated from pets, immediate individuals in contact with pets, and veterinary clinic environments. A total of n = 300 samples were collected from different veterinary hospitals in Pakistan using convenience sampling. The collected samples were subjected to microbiological and biochemical examination for the isolation of S. aureus. Methicillin resistance was investigated by both phenotypically using oxacillin disk diffusion assay and by genotypically targeting mecA gene by PCR. PCR amplicons were subjected for sequencing by Sanger method of sequencing, which were subsequently submitted to NCBI GenBank under the accession numbers MT874770, MT874771, and MT874772. Sequence evolutionary analysis and mecA gene characterization was done using various bioinformatics tools. Overall, 33.66% mecA genes harboring S. aureus strains were isolated from all sources (33.33% from pets, 46.0% from surrounding, and 28.0% from immediate contact individuals). The bioinformatics analysis noted that one SNP was identified at position c.253C>A (Transvertion). The phylogenetic tree (two clades) of S. aureus mecA revealed a possibility of inter-transmission of disease between the environment and pets. Frequency of adenine and thymine nucleotide in motifs were found to be the same (0.334). Cytosine and guanine frequency were also the same (0.166). Threonine was replaced by asparagine (p.T84D) in each sample of cat, environment, and human. On the other hand, protein structures ofcat-1 and cat-2 proteins were found identical while cat-3, environmental, and human proteins shared identical structures. The study thus concludes rising circulation of methicillin-resistant S. aureus (MRSA) strains in animal-human-environment interfaces, forecasting the development of novel strains withmodified range of resistance.

Burns cause many significant changes in metabolism and inflammatory reactions, leading to poor regeneration in animals and humans. A list of medicines to treat burns is available in the market. But due to the high cost of these medicines, these are unaffordable, especially for farmers of middle-class families of Africa and Asia. Therefore, a low-cost complementary treatment has always been a topic of many researchers, and there is a dire need of time for the welfare of animals to save them. The current study was planned to scrutinize the therapeutic effects of Manuka honey and Nitrofurazone ointments on full-thickness burn wounds in the rabbit model. The healing efficacy was performed through wound contraction rate, hematological analysis, the thickness of dermis and epidermis, and collagen content percentage. Histopathology was performed after taking biopsy samples at the end of the research. Based on statistical analysis using wound healing time (days, D), the combination (MO + NT) resulted in a shorter period (27 D ± 1) than the average healing time of controlled (36 ± 2), Manuka ointment (31.33 D ± 1.52), and Nitrofurazone ointment (32 ± 1). A significant decrease in the count of red blood cell (RBC), mean corpuscular volume (MCV), and mean corpuscular hemoglobin (MCH) in all treatments was noticed mainly in MO + NT. Furthermore, burns induced a significant difference (p < 0.05) in the white blood cells (WBCs) count levels in the MO-treated group. While the level of platelets (PLTs) was not significantly different from the healthy control group. Histopathological assessment (epithelialization, fibrosis, and angiogenesis) of skin showed burn healing to be better in MO and MO + NT groups. In conclusion, the composite of Manuka honey with Nitrofurazone led to the faster recovery than other treatments.

Chromium (VI) in tannery effluent is one of the major environmental concerns for the environmentalists due to the hazardous nature of Cr(VI) ions. To reduce Cr(VI) to Cr(III) as an innocuous moiety, pure and I-doped ZnO was grafted over the etched surface of glass beads by successive ionic layer adsorption and reaction (SILAR). Powdered, pure, and I-doped ZnO scrapped from the surface of glass beads was characterized for crystallinity, morphology, and elemental composition by XRD, SEM, TEM, and EDX. The optical properties of both photocatalysts revealed that owing to optimized iodine doping of ZnO, reduction in the bandgap was observed from 3.3 to 2.9 eV. The crystalline nano-bricks of I:ZnO adhered to glass beads were investigated to have remarkable capability to harvest sunlight in comparison to intrinsic ZnO nanodiscs. The thermal stability of I:ZnO was also found to be much improved due to doping of ZnO. The photocatalytic activities of ZnO/GB and I:ZnO/GB were compared by extent of reduction of Cr(VI) under direct natural sunlight (600–650 KWh/m2). The disappearance of absorbance peaks associated with Cr(VI) after treatment with I:ZnO/GB confirmed higher photocatalytic activity of I:ZnO/GB. The reaction parameters of solar photocatalytic reduction, i.e., initial pH (5–9), initial concentration of Cr(VI) (10–50 ppm), and solar irradiation time (1–5 h) were optimized using response surface methodology. The solar photocatalytic reduction of Cr(VI) to Cr(III) present in real tannery effluent was examined to be 87 and 98%, respectively, by employing ZnO/GB and I:ZnO/GB as solar photocatalysts. The extent of reduction was also confirmed by complexation of Cr(VI) and Cr(III) present in treated and untreated tannery waste with 1, 5-diphenylcarbazide. The results of AAS and UV/vis spectroscopy for the decrease in concentration of Cr also supported the evidence of higher efficiency of I:ZnO/GB for reduction of Cr(VI) in tannery effluent. Reusability of the fabricated photocatalyst was assessed for eight cycles, and magnificent extent of reduction of Cr(VI) indicated its high efficiency. Conclusively, I:ZnO/GB is a potential and cost-effective candidate for Cr(VI) reduction in tannery effluent under natural sunlight.

Background: The driving force behind osteoarthritis (OA) pathogenesis is an anabolic-catabolic (a/c) imbalance. Melatonin (MT) is a key player in maintaining a/c stability and mitigates OA pathogenesis, but mechanisms underlying its effects remain poorly understood.Objectives: We performed a systematic review analyzing the experimental data that support the clinical applicability of MT in the treatment of OA pathogenesis, placing particular emphasis on the regulation of circadian rhythms and a/c balance.Methods: Major electronic databases and grey literature were used to identify related original articles. Methodological quality of all selected studies was evaluated using the SYRCLE risk of bias tool. Pooled mean differences (MDs)/standardized mean differences (SMDs) with 95% confidence intervals (CIs) were calculated to estimate the effect size.Results: Eleven trials were included in this systematic review. Compared with the control group, MT significantly decreased the levels of interleukin-1β (IL-1β; SMD = −5.45; 95% CI [−6.78, −4.12]; p < 0.00001, and histological grading scale (SMD = −3.46; 95% CI, [−5.24, −1.68]; p < 0.0001). MT significantly increased the transforming growth factor-β1 (TGF-β1; SMD = 1.17; 95% CI [0.31, 2.03]; p < 0.0007). Furthermore, core circadian clock genes Per2 and Cry1 mRNA levels were regulated by MT treatment in OA progression.Conclusion: MT may maintain a/c balance and regulate circadian rhythms during OA development. MT could be used in as adjunct with other interventions to manage pain and OA severity.

The influence of Artificial Light at Night (ALAN) is a severe interference in the biological rhythm of organisms. Feeding dysregulation might be the main factor responsible for developing metabolic diseases. The molecular basis of these physiological dysregulations is yet to be elucidated. The effect of light on appetite-regulating signal in the brain of zebrafish is still unknown. Therefore, the present study aimed to find the daily mRNA expression profile of two significant appetite and energy-balancing peptide hormone (leptin) and enzyme (goat) in the brain in various illumination conditions, LD (12Light:12Dark), LL (24L), and DD (24D). Moreover, Ga-SI, the protein level of the two appetite-regulating peptides, and brain and serum melatonin are measured after 72 h of incubation under three different photic conditions. The immunohistochemical localization of the primary mediators of appetite regulators, leptin and goat, in the brain of zebrafish, under different photic conditions, is reported for the first time in zebrafish. The study revealed that in continuous light after 72 h of incubation, feeding (Ga-SI) is found the highest and the translational level of two appetite-related genes (leptin and goat) is the lowest compared to LD. In continuous dark, this relation becomes opposite. The daily variation of mRNA expression of leptin and goat in LD shows peak expression at the light and dark phase, respectively. This pattern was abolished in continuous conditions. Thus, our study suggests that the photoperiod (zeitgeber) may influence the expression pattern (daily variation) of appetite-regulating peptide hormone and enzyme in the brain of zebrafish.

The origin of the coronavirus disease 2019 (COVID-19) pandemic is zoonotic. The circadian day–night is the rhythmic clue to organisms for their synchronized body functions. The “development for mankind” escalated the use of artificial light at night (ALAN). In this article, we tried to focus on the possible influence of this anthropogenic factor in human coronavirus (HCoV) outbreak. The relationship between the occurrences of coronavirus and the ascending curve of the night-light has also been delivered. The ALAN influences the physiology and behavior of bat, a known nocturnal natural reservoir of many Coronaviridae. The “threatened” and “endangered” status of the majority of bat species is mainly because of the destruction of their proper habit and habitat predominantly through artificial illumination. The stress exerted by ALAN leads to the impaired body functions, especially endocrine, immune, genomic integration, and overall rhythm features of different physiological variables and behaviors in nocturnal animals. Night-light disturbs “virus–host” synchronization and may lead to mutation in the genomic part of the virus and excessive virus shedding. We also proposed some future strategies to mitigate the repercussions of ALAN and for the protection of the living system in the earth as well.

Amongst various environmental constraints, abiotic stresses are increasing the risk of food insecurity worldwide by limiting crop production and disturbing the geographical distribution of food crops. Millets are known to possess unique features of resilience to adverse environments, especially infertile soil conditions, although the underlying mechanisms are yet to be determined. The small diploid genome, short stature, excellent seed production, C4 photosynthesis, and short life cycle of foxtail millet make it a very promising model crop for studying nutrient stress responses. Known to be a drought-tolerant crop, it responds to low nitrogen and low phosphate by respective reduction and enhancement of its root system. This special response is quite different from that shown by maize and some other cereals. In contrast to having a smaller root system under low nitrogen, foxtail millet enhances biomass accumulation, facilitating root thickening, presumably for nutrient translocation. The low phosphate response of foxtail millet links to the internal nitrogen status, which tends to act as a signal regulating the expression of nitrogen transporters and hence indicates its inherent connection with nitrogen nutrition. Altogether, the low nitrogen and low phosphate responses of foxtail millet can act as a basis to further determine the underlying molecular mechanisms. Here, we will highlight the abiotic stress responses of foxtail millet with a key note on its low nitrogen and low phosphate adaptive responses in comparison to other crops.

Foxtail millet [Setaria italica (L.) Beauv.], a widely cultivated food and fodder crop, develops a smaller root system while enlarges the root diameter facilitating nutrient transport under nitrogen limitation. How foxtail millet responds to phosphate limitation (LP) remains unaddressed. LP seedlings of the sequenced variety Yugu1 had significantly lower P concentrations in both shoots and roots and displayed higher levels of anthocyanin accumulation in leaves, indicating that the seedlings suffered from P limitation under hydroponic culture. One obvious and adaptive phenotype of LP plants was the larger root system mostly as the result of stimulation of lateral root proliferation in terms of the number, density, and length. Preferential biomass accumulation in the root under LP ensured carbon provision for root expansion and resulted in significant increases in the total and specific root length, which substantially extended the absorptive surface of P in the growth medium. Elevation of auxin and gibberellin concentrations might serve as an internal boost underpinning root architectural re-patterning under LP. Not just morphological adaptation, up-regulation of expression of SiPHT1;1 and SiPHT1;4 in roots and that of SiPHT1;2 in roots and shoots preconditioned adaptive enhancement of P uptake and translocation under LP. Interestingly, internal nitrogen surpluses occurred as indicated by dramatic increases in free amino acids in LP shoots and roots and higher concentrations of nitrogen in roots. Such nitrogen surplus ‘signals’ tended to switch down expression of nitrate transporters SiNRT2.1 and SiNAR2.1 in the root and that of SiNRT1.11 and SiNRT1.12 in the shoot to reduce nitrate mobilization toward or within the shoot. Together, our work provided new insights into adaption of a critical cereal crop to LP and its innate connection with nitrogen nutrition.

Foxtail millet (FM) [Setaria italica (L.) Beauv.] is a grain and forage crop well adapted to nutrient-poor soils. To date little is known how FM adapts to low nitrogen (LN) at the morphological, physiological, and molecular levels. Using the FM variety Yugu1, we found that LN led to lower chlorophyll contents and N concentrations, and higher root/shoot and C/N ratios and N utilization efficiencies under hydroponic culture. Importantly, enhanced biomass accumulation in the root under LN was in contrast to a smaller root system, as indicated by significant decreases in total root length; crown root number and length; and lateral root number, length, and density. Enhanced carbon allocation toward the root was rather for significant increases in average diameter of the LN root, potentially favorable for wider xylem vessels or other anatomical alterations facilitating nutrient transport. Lower levels of IAA and CKs were consistent with a smaller root system and higher levels of GA may promote root thickening under LN. Further, up-regulation of SiNRT1.1, SiNRT2.1, and SiNAR2.1 expression and nitrate influx in the root and that of SiNRT1.11 and SiNRT1.12 expression in the shoot probably favored nitrate uptake and remobilization as a whole. Lastly, more soluble proteins accumulated in the N-deficient root likely as a result of increases of N utilization efficiencies. Such “excessive” protein-N was possibly available for shoot delivery. Thus, FM may preferentially transport carbon toward the root facilitating root thickening/nutrient transport and allocate N toward the shoot maximizing photosynthesis/carbon fixation as a primary adaptive strategy to N limitation.

Under the background of the shortage of traditional energy and the increasingly serious environmental problems, this paper proposes a bi-level optimization configuration method for microgrids based on stepped carbon trading and price-based demand response. The stepped carbon trading mechanism is introduced into the planning model to clarify the impact of construction processes on land carbon emissions. Simultaneously, price-based demand response during system operation is introduced, guiding users to change the electricity consumption strategies to promote the consumption of renewable energy. Then, a bi-level optimal configuration framework is constructed. The upper level is optimized to allocate the capacity of the microgrid, with the lowest annual equivalent comprehensive cost as the objective function. The lower level is optimized for microgrid operation, with the minimum sum of the microgrid operation cost and carbon emission cost as the objective function. In view of the advantages of enhanced whale optimization algorithm (E-WOA) in solving multidimensional feature selection problems, E-WOA and CPLEX solver are combined to solve the bi-level optimization model for the first time. Simulation results show that after introducing a stepped carbon trading mechanism, the system carbon emissions decreased by 23.09%. Considering price-based demand response under the premise of introducing a stepped carbon trading mechanism, the total cost decreased by 3.57% and the carbon emissions decreased by 19.83%. Staphylococcus aureus is recognized as commensal as well as opportunistic pathogen of humans and animals. Methicillin resistant strain of S. aureus (MRSA) has emerged as a major pathogen in hospitals, community and veterinary settings that compromises the public health and livestock production. MRSA basically emerged from MSSA after acquiring SCCmec element through gene transfer containing mecA gene responsible for encoding PBP-2α. This protein renders the MRSA resistant to most of the β-lactam antibiotics. Due to the continuous increasing prevalence and transmission of MRSA in hospitals, community and veterinary settings posing a major threat to public health. Furthermore, high pathogenicity of MRSA due to a number of virulence factors produced by S. aureus along with antibiotic resistance help to breach the immunity of host and responsible for causing severe infections in humans and animals. The clinical manifestations of MRSA consist of skin and soft tissues infection to bacteremia, septicemia, toxic shock, and scalded skin syndrome. Moreover, due to the increasing resistance of MRSA to number of antibiotics, there is need to approach alternatives ways to overcome economic as well as human losses. This review is going to discuss various aspects of MRSA starting from emergence, transmission, epidemiology, pathophysiology, disease patterns in hosts, novel treatment, and control strategies. Background: The driving force behind osteoarthritis (OA) pathogenesis is an anabolic-catabolic (a/c) imbalance. Melatonin (MT) is a key player in maintaining a/c stability and mitigates OA pathogenesis, but mechanisms underlying its effects remain poorly understood. Objectives: We performed a systematic review analyzing the experimental data that support the clinical applicability of MT in the treatment of OA pathogenesis, placing particular emphasis on the regulation of circadian rhythms and a/c balance. Methods: Major electronic databases and grey literature were used to identify related original articles. Methodological quality of all selected studies was evaluated using the SYRCLE risk of bias tool. Pooled mean differences (MDs)/standardized mean differences (SMDs) with 95% confidence intervals (CIs) were calculated to estimate the effect size. Results: Eleven trials were included in this systematic review. Compared with the control group, MT significantly decreased the levels of interleukin-1β (IL-1β; SMD = −5.45; 95% CI [−6.78, −4.12]; p < 0.00001, and histological grading scale (SMD = −3.46; 95% CI, [−5.24, −1.68]; p < 0.0001). MT significantly increased the transforming growth factor-β1 (TGF-β1; SMD = 1.17; 95% CI [0.31, 2.03]; p < 0.0007). Furthermore, core circadian clock genes Per2 and Cry1 mRNA levels were regulated by MT treatment in OA progression. Conclusion: MT may maintain a/c balance and regulate circadian rhythms during OA development. MT could be used in as adjunct with other interventions to manage pain and OA severity. The origin of the coronavirus disease 2019 (COVID-19) pandemic is zoonotic. The circadian day–night is the rhythmic clue to organisms for their synchronized body functions. The "development for mankind" escalated the use of artificial light at night (ALAN). In this article, we tried to focus on the possible influence of this anthropogenic factor in human coronavirus (HCoV) outbreak. The relationship between the occurrences of coronavirus and the ascending curve of the night-light has also been delivered. The ALAN influences the physiology and behavior of bat, a known nocturnal natural reservoir of many Coronaviridae. The "threatened" and "endangered" status of the majority of bat species is mainly because of the destruction of their proper habit and habitat predominantly through artificial illumination. The stress exerted by ALAN leads to the impaired body functions, especially endocrine, immune, genomic integration, and overall rhythm features of different physiological variables and behaviors in nocturnal animals. Night-light disturbs "virus–host" synchronization and may lead to mutation in the genomic part of the virus and excessive virus shedding. Investigating prior methodologies, it has come to our knowledge that in smart cities, a disaster management system needs an autonomous reasoning mechanism to efficiently enhance the situation awareness of disaster sites and reduce its after-effects. Disasters are unavoidable events that occur at anytime and anywhere. Timely response to hazardous situations can save countless lives. Therefore, this paper introduces a multi-agent system (MAS) with a situation-awareness method utilizing NB-IoT, cyan industrial Internet of things (IIOT), and edge intelligence to have efficient energy, optimistic planning, range flexibility, and handle the situation promptly. We introduce the belief-desire-intention (BDI) reasoning mechanism in a MAS to enhance the ability to have disaster information when an event occurs and perform an intelligent reasoning mechanism to act efficiently in a dynamic environment. Moreover, we illustrate the framework using a case study to determine the working of the proposed system. We develop ontology and a prototype model to demonstrate the scalability of our proposed system.

Magnetic resonance imaging is the most generally utilized imaging methodology that permits radiologists to look inside the cerebrum using radio waves and magnets for tumor identification. However, it is tedious and complex to identify the tumorous and nontumorous regions due to the complexity in the tumorous region. Therefore, reliable and automatic segmentation and prediction are necessary for the segmentation of brain tumors. This paper proposes a reliable and efficient neural network variant, i.e., an attention-based convolutional neural network for brain tumor segmentation. Specifically, an encoder part of the UNET is a pre-trained VGG19 network followed by the adjacent decoder parts with an attention gate for segmentation noise induction and a denoising mechanism for avoiding overfitting. The dataset we are using for segmentation is BRATS’20, which comprises four different MRI modalities and one target mask file. The abovementioned algorithm resulted in a dice similarity coefficient of 0.83, 0.86, and 0.90 for enhancing, core, and whole tumors, respectively.

In the last decade, smart computing has garnered much attention, particularly in ubiquitous environments, thus increasing the ease of everyday human life. Users can dynamically interact with the systems using different modalities in a smart computing environment. The literature discussed multiple mechanisms to enhance the modalities for communication using different knowledge sources. Among others, Multi-context System (MCS) has been proven quite significant to interlink various context domains dynamically to a distributed environment. MCS is a collection of different contexts (independent knowledge sources), and every context contains its own set of defined rules and facts and inference systems. These contexts are interlinked via bridge rules. However, the interaction among knowledge sources could have the consequences such as bringing out inconsistent results. These issues may report situations such as the system being unable to reach a conclusion or communication in different contexts becoming asynchronous. There is a need for a suitable framework to resolve inconsistencies. In this article, we provide a framework based on contextual defeasible reasoning and a formalism of multi-agent environment is to handle the issue of inconsistent information in MCS. Additionally, in this work, a prototypal simulation is designed using a simulation tool called NetLogo, and a formalism about a Parkinson's disease patient's case study is also developed. Both of these show the validity of the framework.
