Abstract

International audience; The temporal and spatial evolution of dispersed media is a fundamental problem in a widerange of physicochemical systems, such as emulsions, suspensions or aerosols. These systems are multiphasic and involve compounds of different densities. They are therefore subject to the influence of gravity which determines the sedimentation rate of their dispersed phase. This effect can be dominant and prevent a detailed study of the phenomena occurring between the constituents themselves, such as the coalescence of drops in emulsions, the evaporation of droplets or the flocculation in suspensions. In this context, the Centre National d'Etudes Spatiales (CNES) has recently supported the development of a new instrument to produce populations of droplets, a few micrometers in radius, under controlled conditions with the objective of allowing a detailed study of their properties in microgravity conditions. The principle of this instrument is to generate, by a fast compression/expansion of air, populations of water droplets and to track their evolution by optical scanning tomography in transmission mode within a volume of approximately 2 mm3. Parabolic flight experiments have shown the possibility to generate and accurately follow the evolution of populations of several hundred droplets for more than 20 seconds. The first experimental results show that it is possible to study their evaporation kinetics or their motion when imposing Von Karman swirling flows to air. This work is part of the AEROSOL project of DECLIC-EVO supported by CNES and aims to help the understanding of cloud microphysics which remains a critical open problem in the context of global warming.

Abstract

International audience; This work experimentally addresses aluminum combustion in steam, pure or mixed with diluents, for aluminum particles in size range 40 ∼80 μm, using an electrodynamic levitator. High-speed videos unveil an unreported and complex mechanism in steam, not observed in other oxidizers. The detached flame is quite faint and very close to the surface. Alumina smoke around the droplet rapidly condenses and coalesces into a large, single orbiting alumina satellite. It eventually collides the main aluminum droplet while generating secondary alumina droplets. A unique feature is the presence of several distinct oxide lobes on the droplet, which merge only at the end of burning and encapsulate the remaining aluminum, possibly promoting an incomplete combustion. The measured burning times in pure water vapor are longer than expected and the efficiency of steam is found to be 30% that of oxygen, lower than the usually accepted value of 60%. A general correlation on burning time, including the major oxidizers, is proposed. Direct numerical simulations are conducted and are in line with experiments, in terms of burning rate or flame stand off ratio. Combustion in steam seems mostly supported by surface reactions, giving a faint flame with low gas temperatures and high hydrogen content. It is speculated that those two specific features could help explain the peculiarity of steam.

Abstract

International audience; Aluminum particles are widely used as an additive to increase energetic performances. Recently, Aluminum has also received a growing interest as a source of hydrogen for fuel cell applications, or in novel power generation concepts as it provides a carbon-free energy that meets current policies for reducing emissions of greenhouse gases. Our recent studies focused on the description of the combustion process varying ambient conditions (pressure and oxidizer). Only the second stage of the combustion process, after the establishment of a steady state, was scrutinized. The proposed work sheds light more specifically to the initial stages where the particle is warmed up from the initial state to a thermal runaway condition. Indeed, the question arises at what temperature do these condensed fuels burst into flames, knowing that the presence of a passive coat greatly influences the process.For this study, a dedicated set-up has been improved. It consists of an electrodynamic levitator in which a single particle, trapped by electrostatic forces, burns in a controlled atmosphere. A two-color ratio pyrometer has been specifically assembled with photomultipliers (PM) to obtain the time evolution of mean surface temperature. The PM signals are synchronised witha particle visualization device that is composed of a high-speed camera combined with a long-distance microscope. Subsequently, the temperature measurement of high temporal resolution can then be associated for the first time at a rate of 250 000 fps with the fast-apparent phenomenology of the pre-ignition processes. In addition, the spatial resolution of the visualization (1.92 μm/pix) allows the interpretation of the PM signals to be commented.

The idea of this work is to generate with a fast compression/expansion of air, populations of water drop let s and to track their evolution by optical scanning tomography in transmission mode. Parabolic flights experiments have shown the possibility to generate and accurately follow the evolution of several hundred droplets for more than 20 seconds of microgravity within a 2 mm 3 volume . First results show that it is possible to study their evaporation kinetics as well as their motion when moving in a Von Karman swirling flow. Abstract

Introduction Over the past 50 years, the prevalence of allergic respiratory diseases has been increasing. The Hygiene hypothesis explains this progression by the decrease in the bio-diversity of early microbial exposure. This study aims to evaluate the effect of early-life farm exposure on airway hyperresponsiveness and cough hypersensitivity in an allergic airway inflammation rabbit model. Method A specific environment was applied to pregnant rabbits and their offspring until six weeks after birth. Rabbits were housed in a pathogen-free zone for the control group and a calf barn for the farm group. At the end of the specific environmental exposure, both groups were then housed in a conventional zone and then sensitized to ovalbumin. Ten days after sensitization, the rabbit pups received ovalbumin aerosols to provoke airway inflammation. Sensitization to ovalbumin was assessed by specific IgE assay. Cough sensitivity was assessed by mechanical stimulation of the trachea, and bronchial reactivity was assessed by methacholine challenge. The farm environment was characterized by endotoxin measurement. Results A total of 38 rabbit pups were included (18 in the farm group). Endotoxin levels in the farm environment varied from 30 to 1854 EU.m-3. There was no significant difference in specific IgE values to ovalbumin (p = 0.826) between the two groups. The mechanical threshold to elicit a cough did not differ between the two groups (p = 0.492). There was no difference in the number of cough (p = 0.270) or the intensity of ventilatory responses (p = 0.735). After adjusting for age and weight, there was no difference in respiratory resistance before and after methacholine challenge. Conclusion Early exposure to the calf barn did not affect cough sensitivity or bronchial reactivity in ovalbumin-sensitized rabbits. These results suggest that not all farm environments protect against asthma and atopy. Continuous exposure to several sources of microbial diversity is probably needed.

Abstract

International audience; The activation of phagocytic cells is a hallmark of many neurological diseases. Imaging them in their 3-dimensional cerebral environment over time is crucial to better understand their role in disease pathogenesis and to monitor their potential therapeutic effects. Phagocytic cells have the ability to internalize metal-based contrast agents both in vitro and in vivo and can thus be tracked by magnetic resonance imaging (MRI) or computed tomography (CT). In this review article, we summarize the different labelling strategies, contrast agents, and in vivo imaging modalities that can be used to monitor cells with phagocytic activity in the central nervous system using MRI and CT, with a focus on clinical applications. Metal-based nanoparticle contrast agents such as gadolinium, gold and iron are ideal candidates for these applications as they have favourable magnetic and/or radiopaque properties and can be fine-tuned for optimal uptake by phagocytic cells. However, they also come with downsides due to their potential toxicity, especially in the brain where they might accumulate. We therefore conclude our review by discussing the pitfalls, safety and potential for clinical translation of these metal-based neuroimaging techniques. Early results in patients with neuropathologies such as multiple sclerosis, stroke, trauma, cerebral aneurysm and glioblastoma are promising. If the challenges represented by safety issues are overcome, phagocytic cells imaging will be a very valuable tool for studying and understanding the inflammatory response and evaluating treatments that aim at mitigating this response in patients with neurological diseases.

Abstract

International audience; Bone marrow adipocytes (BMAds) accrue in various states of osteoporosis and interfere with bone remodeling through the secretion of various factors. However, involvement of the extracellular matrix (ECM) produced by BMAds in the impairment of bone marrow mesenchymal stromal cell (BM-MSC) osteoblastogenesis has received little attention. In type 2 diabetes (T2D), skeletal fragility is associated with several changes in bone quality that are incompletely understood, and BMAd quantity increases in relationship to poor glycemic control. Considering their altered phenotype in this pathophysiological context, we aimed to determine the contribution of the ECM of mature BMAds to osteoblastogenesis and mineralization quality in the context of chronic hyperglycemia. Human BM-MSCs were differentiated for 21 days in adipogenic medium containing either a normoglycemic (LG, 5.5 mM) or a high glucose concentration (HG, 25 mM). The ECM laid down by BMAds were devitalized through cell removal to examine their impact on the proliferation and differentiation of BM-MSCs toward osteoblastogenesis in LG and HG conditions. Compared to control plates, both adipocyte ECMs promoted cell adhesion and proliferation. As shown by the unmodified RUNX2 and osteocalcin mRNA levels, BM-MSC commitment in osteoblastogenesis was hampered by neither the hyperglycemic condition nor the adipocyte matrices. However, adipocyte ECMs or HG condition altered the mineralization phase with perturbed expression levels of type 1 collagen, MGP and osteopontin. Despite higher ALP activity, mineralization levels per cell were decreased for osteoblasts grown on adipocyte ECMs compared to controls. Raman spectrometry revealed that culturing on adipocyte matrices specifically prevents type-B carbonate substitution and favors collagen crosslinking, in contrast to exposure to HG concentration alone. Moreover, the mineral to organic ratio was disrupted according to the presence of adipocyte ECM and the glucose concentration used for adipocyte or osteoblast culture. HG concentration and adipocyte ECM lead to different defects in mineralization quality, recapitulating contradictory changes reported in T2D osteoporosis. Our study shows that ECMs from BMAds do not impair osteoblastogenesis but alter both the quantity and quality of mineralization partly in a glucose concentration-dependent manner. This finding sheds light on the involvement of BMAds, which should be considered in the compromised bone quality of T2D and osteoporosis patients more generally.

International audience; Since 2001, the SPIRAL1 facility has been providing post-accelerated radioactive beams by the ISOL method. Over the last decade, SPIRAL1 has been improved to provide beams of condensable elements, by using a combination of a FEBIAD-type ion source (to produce 1+ ions) and a PHOENIX ECR charge breeder (to transform 1+ ions into N+ ions for post-acceleration). Both these elements have been improved in recent years: the FEBIAD source has been upgraded to improve its efficiency and reliability over time and the charge breeder was modified to extend the range of available charge states. In addition, a krypton beam was recently sent in a FEBIAD target ion-source system (TISS), allowing to produce medium mass radioactive 1+ ions that had not been previously produced in SPIRAL1. Abstract

International audience; Aerosols are divided systems in which liquid and/or solid particles evolve in a continuous gas phase. They are naturally subject to the influence of gravity, which determines the settling speed of the particles they contain. Aerosols have many applications in the pharmaceutical and cosmetic industries (inhalers, sprays), in metallurgy (spray drying) and in agriculture (pesticides). From an academic point of view, they are the subject of intense research activity. The description of their evolution is, for example, a major issue in climatology. With global warming, more water is evaporatingin the air resulting in more cloud formation and a higher importance of cloud microphysics in the understanding of the climate evolution. In this context, CNES has recently supported the development of a new instrument allowing not only to produce aerosols under controlled thermodynamicconditions but also to follow their evolution in microgravity. This work presents results of parabolic flight experiments. It demonstrates the possibility of producing aerosols and to investigate their properties with optical microscopy for periods of about twenty seconds.

Abstract

International audience; The temporal and spatial evolution of dispersed media is a fundamental problem in a wide range of physicochemical systems, such as emulsions, suspensions and aerosols. These systems are multiphasic and involve compounds of different densities. They are therefore subject to the influence of gravity which determines the sedimentation rate of their dispersed phase. This effect can be dominant and prevent a detailed study of the phenomena occurring between the constituents themselves, such as the coalescence of drops in emulsions, the evaporation of droplets or the flocculation in suspensions. In this context, the Centre National d'Etudes Spatiales (CNES) has recently supported the development of a new instrument to produce populations of droplets, a few micrometers in radius, under controlled conditions with the objective of allowing a detailed study of their properties in microgravity conditions. The principle of this instrument is to generate, by a fast compression/expansion of air, populations of water droplets and to track their evolution by optical scanning tomography in transmission mode within a volume of approximately 2 mm 3. Parabolic flight experiments have shown the possibility to generate and accurately follow the evolution of populations of several hundred droplets for more than 20 seconds. The first experimental results show that it is possible to study their evaporation kinetics or their motion when imposing Von Karman swirling flows. This work is part of the AEROSOL project of DECLIC-EVO supported by CNES and aims to help the understanding of cloud microphysics which remains a critical open problem in the context of global warming.

The European Spallation Source will be driven by a 5 MW linear accelerator, producing 2.86 ms long proton beam pulses with a peak current of 62.5 mA at 14 Hz. RFQ at ESS has been successfully gone through RF conditioning, RF re-conditioning and low duty cycle beam commissioning. RFQ fulfills required functions and overall performance is satisfactory. RF conditioning, three RF re-conditionings after LEBT intervention and beam commissioning will be reported and RFQ performance during these periods will be described. RFQ performance in a large extent is reflected by dynamics and interactions between RF, cavity and beam. Thanks to advanced hardware capabilities and intelligent software intelligence, observation of those dynamics and interactions are done in detailed level. Analysis of those dynamics and interaction will be introduced. Abstract

International audience; Pattern recognition receptors (PRRs) protect against microbial invasion by detecting specific molecular patterns found in pathogens and initiating an immune response. Although microbial-derived PRR ligands have been extensively characterized, the contribution and relevance of endogenous ligands to PRR activation remains overlooked. Here, we characterize the landscape of endogenous ligands that engage RIG-I-like receptors (RLRs) upon infection by different RNA viruses. In each infection, several RNAs transcribed by RNA polymerase III (Pol3) specifically engaged RLRs, particularly the family of Y RNAs. Sensing of Y RNAs was dependent on their mimicking of viral secondary structure and their 5′-triphosphate extremity. Further, we found that HIV-1 triggered a VPR-dependent downregulation of RNA triphosphatase DUSP11 in vitro and in vivo, inducing a transcriptome-wide change of cellular RNA 5′-triphosphorylation that licenses Y RNA immunogenicity. Overall, our work uncovers the contribution of endogenous RNAs to antiviral immunity and demonstrates the importance of this pathway in HIV-1 infection.

International audience; Background: Systemic inflammation is the main factor underlying secondary osteoporosis in patients with rheumatoid arthritis (RA). Janus kinase inhibitors (JAKi), such as tofacitinib (Tofa), can control systemic inflammation and may have beneficial effects on bone in various models. This might be due to direct effects on the bone microenvironment and not exclusively based on their anti-inflammatory function. Bone marrow adipocytes (BMAds) are abundant in the bone microenvironment. The effect of JAKi on BMAds is unknown, but evidence suggests that there is competition between human bone marrow-derived stromal cell (hBMSC) differentiation routes towards BMAds and osteoblasts (Ob) in osteoporosis. Objectives: The aims of the study are to determine whether Tofa influences BMAds and Ob derived from hBMSCs and to investigate the potential effects of Tofa on bone marrow adiposity in RA patients. Methods: To determine the effect of Tofa on cellular commitment, hBMSCs were differentiated to BMAds or OBs for 3 days together with Tofa at 200, 400, or 800 nM and TNFa. This study was also conducted using differentiated BMAds. The impact of Tofa was determined by gene and protein expression analysis and cell density monitoring. In parallel, in a pilot study of 9 RA patients treated with Tofa 5 mg twice a day (NCT04175886), the proton density fat fraction (PDFF) was measured using MRI at the lumbar spine at baseline and at 6 months. Abstract

International audience; In human, bone loss is associated with increased marrow adipose tissue and recent data suggest that medullary adipocytes could play a role in osteoporosis by acting on neighboring bone-forming osteoblasts. Supporting this hypothesis, we previously showed, in a coculture model based on human bone marrow stromal cells, that factors secreted by adipocytes induced the conversion of osteoblasts towards an adipocyte-like phenotype. In this work, we employed an original integrative bioinformatics approach connecting proteomic and transcriptomic data from adipocytes and osteoblasts, respectively, to investigate the mechanisms underlying their crosstalk. Our analysis identified a total of 271 predicted physical interactions between adipocyte-secreted proteins and osteoblast membrane protein coding genes and proposed three pathways for their potential contribution to osteoblast transdifferentiation, the PI3K-AKT, the JAK2-STAT3 and the SMAD pathways. Our findings demonstrated the effectiveness of our integrative omics strategy to decipher cell-cell communication events.

The 352.21 MHz Radio Frequency Quadrupole (RFQ) for the European Spallation Source ERIC (ESS) has been delivered by the end of 2019 by CEA/IRFU. The RFQ is designed to accelerate a 70 mA proton beam from 75 keV up to 3.62 MeV. It consists of a 4-vane resonant cavity with a total length of 4.6 m. Two coaxial power loop couplers feed the RFQ with the 1.4 MW of RF power required for beam operation. This paper first presents the main systems required for the RFQ conditioning. Abstract

We study the processes $\gamma \gamma \to \eta_c \to \eta' K^+ K^-$, $\eta' \pi^+ \pi^-$, and $\eta \pi^+ \pi^-$ using a data sample of 519 $fb^{-1}$ recorded with the BaBar detector operating at the SLAC PEP-II asymmetric-energy $e^+e^-$ collider at center-of-mass energies at and near the $\Upsilon(nS)$ ($n = 2,3,4$) resonances. This is the first observation of the decay $\eta_c \to \eta' K^+ K^-$ and we measure the branching fraction $\Gamma(\eta_c \to \eta' K^+ K^-)/(\Gamma(\eta_c \to \eta' \pi^+ \pi^-)=0.644\pm 0.039_{\rm stat}\pm 0.032_{\rm sys}$. Significant interference is observed between $\gamma \gamma \to \eta_c\to \eta \pi^+ \pi^-$ and the non-resonant two-photon process $\gamma \gamma \to \eta \pi^+ \pi^-$. A Dalitz plot analysis is performed of $\eta_c$ decays to $\eta' K^+ K^-$, $\eta' \pi^+ \pi^-$, and $\eta \pi^+ \pi^-$. Combined with our previous analysis of $\eta_c \to K \bar K \pi$, we measure the $K^*_0(1430)$ parameters and the ratio between its $\eta' K$ and $\pi K$ couplings. The decay $\eta_c \to \eta' \pi^+ \pi^-$ is dominated by the $f_0(2100)$ resonance, also observed in $J/\psi$ radiative decays. A new $a_0(1700) \to \eta \pi$ resonance is observed in the $\eta_c \to \eta \pi^+ \pi^-$ channel. We also compare $\eta_c$ decays to $\eta$ and $\eta'$ final states in association with scalar mesons as they relate to the identification of the scalar glueball., Comment: 25 pages, 18 figures

Abstract

Dark photons are hypothetical massive vector particles that could mix with ordinary photons. The simplest theoretical model is fully characterised by only two parameters: the mass of the dark photon m$$_{\gamma ^{\mathrm {D}}}$$ γ D and its mixing parameter with the photon, $$\varepsilon $$ ε . The sensitivity of the SHiP detector is reviewed for dark photons in the mass range between 0.002 and 10 GeV. Different production mechanisms are simulated, with the dark photons decaying to pairs of visible fermions, including both leptons and quarks. Exclusion contours are presented and compared with those of past experiments. The SHiP detector is expected to have a unique sensitivity for m$$_{\gamma ^{\mathrm {D}}}$$ γ D ranging between 0.8 and 3.3$$^{+0.2}_{-0.5}$$ - 0.5 + 0.2 GeV, and $$\varepsilon ^2$$ ε 2 ranging between $$10^{-11}$$ 10 - 11 and $$10^{-17}$$ 10 - 17 .

Abstract

While numerous transgenic mouse strains have been produced to model the formation of amyloid-β (Aβ) plaques in the brain, efficient methods for whole-brain 3D analysis of Aβ deposits are lacking. Moreover, standard immunohistochemistry performed on brain slices precludes any shape analysis of Aβ plaques. The present study shows how in-line (propagation-based) X-ray phase-contrast tomography (XPCT) combined with ethanol-induced brain sample dehydration enables hippocampus-wide detection and morphometric analysis of Aβ plaques. Performed in three distinct Alzheimer mouse strains, the proposed workflow identified differences in signal intensity and 3D shape parameters: 3xTg displayed a different type of Aβ plaques, with a larger volume and area, greater elongation, flatness and mean breadth, and more intense average signal than J20 and APP/PS1. As a label-free non-destructive technique, XPCT can be combined with standard immunohistochemistry. XPCT virtual histology could thus become instrumental in quantifying the 3D spreading and the morphological impact of seeding when studying prion-like properties of Aβ aggregates in animal models of Alzheimer’s disease. Abstract

International audience; BACKGROUND: Whole-bowel irrigation (WBI) is a strategy of gastrointestinal decontamination, recommended by several European and American learned societies, which may be used in the management of the poisoned patients. OBJECTIVES: The objectives of this study were to describe the feasibility and tolerability of this technique and to compare the clinical outcome of a group of poisoned patients treated with WBI versus that of an untreated group. METHODS: This was a retrospective and observational study of data recorded by the Angers Poison Control Centre (PCC) between 2012 and 2018. All cases for which the PCC advised WBI were included. The association between outcomes (clinical deterioration after WBI advised by a PCC, length of hospitalisation), WBI treatment, and relevant associated risk factors was determined using univariate and multivariate logistic regression. RESULTS: A total of 257 patients were included. One hundred forty-one patients were treated with WBI with clearly successful induction of diarrhoea in 47 cases (31%). WBI was not initiated in 89 patients. WBI was initiated but unsuccessful (no diarrhoea) in nine cases. The median age is 46 years (interquartile range: 32-55 years), with a sex ratio (M/F) of 1.3. A total of 27 of 150 patients (18%) who underwent WBI had adverse effects possibly linked to WBI, mainly vomiting (n=23). Abstract

International audience; ABSTRACT Background An increasing number of infant and follow-on formulas are enriched with probiotics and/or prebiotics; however, evidence for health effects of such enrichment in early childhood remains inconclusive. Objectives The present study aimed to assess whether the consumption of formula enriched with probiotics or prebiotics was associated with the risk of infection and allergic diseases in early childhood. Methods Analyses involved data for 8389 formula-fed children from the Etude Longitudinale Française depuis l'Enfance (ELFE) cohort. Enrichment of the formula with probiotics or prebiotics that was consumed from the age of 2–10 mo was identified by the formula ingredient list. Lower respiratory tract infection (LRTI), upper respiratory tract infection (URTI), gastrointestinal infection, wheezing, asthma, food allergy, and itchy rash were prospectively reported by parents up to the age of 5.5 y. Adjusted logistic regression models were used to assess associations between the consumption of enriched formula and risk of infection and allergic diseases. Results Aged 2 mo, more than half of formula-fed infants consumed the probiotic-enriched formula and only 1 in 10 consumed the prebiotic-enriched formula. Consumption of the Bifidobacterium lactis-enriched formula at 2 mo was associated with a lower risk of LRTI [OR (95% CI) = 0.84 (0.73–0.96)]. Consumption of the Bifidobacterium breve-enriched formula up to 6 mo was associated with a higher risk of LRTI [OR (95% CI) = 1.75 (1.29–2.38)] and asthma [OR (95% CI) = 1.95 (1.28–2.97)], whereas its consumption from 6 to 10 mo was associated with a lower risk of LRTI [OR (95% CI) = 0.64 (0.48–0.86)] and asthma [OR (95% CI) = 0.59 (0.40–0.88)]. Moreover, the consumption of Streptococcus thermophilus from 6 to 10 mo was associated with a higher risk of asthma [OR (95% CI) = 1.84 (1.29–2.63)]. No significant association was found for gastrointestinal infection, food allergy, and itchy rash. Overall, the consumption of prebiotic-enriched formula was not significantly associated with infection and allergy risk. Conclusions Associations between the consumption of probiotic-enriched formula and risk of respiratory symptoms differ according to the strain considered and consumption period. Further well-designed studies are needed to confirm these results.

Abstract

Purpose The prototypical TSPO radiotracer (R)-[11C]PK11195 has been used in humans for more than thirty years to visualize neuroinflammation in several pathologies. Alternative radiotracers have been developed to improve signal-to-noise ratio and started to be tested clinically in 2008. Here we examined the scientific value of these “(R)-[11C]PK11195 challengers” in clinical research to determine if they could supersede (R)-[11C]PK11195. Methods A systematic MEDLINE (PubMed) search was performed (up to end of year 2020) to extract publications reporting TSPO PET in patients with identified pathologies, excluding studies in healthy subjects and methodological studies. Results Of the 288 publications selected, 152 used 13 challengers, and 142 used (R)-[11C]PK11195. Over the last 20 years, the number of (R)-[11C]PK11195 studies remained stable (6 ± 3 per year), but was surpassed by the total number of challenger studies for the last 6 years. In total, 3914 patients underwent a TSPO PET scan, and 47% (1851 patients) received (R)-[11C]PK11195. The 2 main challengers were [11C]PBR28 (24%—938 patients) and [18F]FEPPA (11%—429 patients). Only one-in-ten patients (11%—447) underwent 2 TSPO scans, among whom 40 (1%) were scanned with 2 different TSPO radiotracers. Conclusions Generally, challengers confirmed disease-specific initial (R)-[11C]PK11195 findings. However, while their better signal-to-noise ratio seems particularly useful in diseases with moderate and widespread neuroinflammation, most challengers present an allelic-dependent (Ala147Thr polymorphism) TSPO binding and genetic stratification is hindering their clinical implementation. As new challengers, insensitive to TSPO human polymorphism, are about to enter clinical evaluation, we propose this systematic review to be regularly updated (living review).

Maisons-Alfort : Anses, 109 p.; L’Agence française de sécurité sanitaire de l’alimentation, de l’environnement et du travail (Anses) s’est autosaisie le 29 mars 2017 afin d’évaluer les risques liés aux niveaux d’activité physique et de sédentarité des adultes de 18 à 64 ans, hors femmes enceintes et ménopausées, à partir des données recueillies en 2014-2015 de l’étude individuelle et nationale sur les consommations alimentaires Inca3.Cette autosaisine s’inscrit dans la continuité du rapport de l’Anses publié en février 2016, intitulé « Actualisation des repères du PNNS - Révisions des repères relatifs à l’activité physique et à la sédentarité » (Anses 2016).Le PNNS (Programme national de nutrition santé) est un programme national de santé publique qui vise à améliorer l’état de santé de la population en agissant sur la nutrition, déterminant majeur de l’état de santé. S’agissant de l’importance de l’activité physique, le PNNS mentionne notamment « les repères nutritionnels du PNNS visant à promouvoir une alimentation et une activité physique favorables à un état nutritionnel et un état de santé optimaux. » Depuis la publication du rapport de l’Anses (2016), de nombreuses études de cohorte ont été publiées, apportant de nouveaux éclairages sur l'effet de différents comportements d’activité physique et de sédentarité sur la santé. L'évaluation des niveaux d'activité physique (NAP) et de temps de sédentarité a permis d'obtenir des données plus objectives permettant d'étudier la répartition de différents comportements dans la journée et les liens de ces comportements entre eux et avec des facteurs de santé.Cette autosaisine repose sur le fonds scientifique suivant :- le rapport « Actualisation des repères du PNNS - Révisions des repères relatifs à l’activité physique et à la sédentarité » (Anses 2016) ;- les données issues de l’Etude individuelle et nationale sur les consommations alimentaires Inca 3 (Anses 2017) ;- les données sur les risques associés aux niveaux d'activité physique et de sédentarité issues de la littérature ; celles-ci portent sur l’activité physique de loisir c’est-à-dire hors du contexte professionnel.

Abstract

International audience; We report on the first search for electron-muon lepton flavor violation (LFV) in the decay of a b quark and b antiquark bound state. We look for the LFV decay ϒ(3S)→e±μ∓ in a sample of 118 million ϒ(3S) mesons from 27 fb-1 of data collected with the BABAR detector at the SLAC PEP-II e+e- collider operating with a 10.36 GeV center-of-mass energy. No evidence for a signal is found, and we set a limit on the branching fraction B[ϒ(3S)→e±μ∓]<3.6×10-7 at 90% C. L. This result can be interpreted as a limit ΛNP/gNP2>80 TeV on the energy scale ΛNP divided by the coupling-squared gNP2 of relevant new physics (NP).

Presentation, translation and interpretation of an unpublished demotic contract dated April-May251 BC, concluded between an Egyptian employee at a temple of the goddess Thoueris and a Greek ‘born inEgypt’, called Stephanos son of Xenon. The latter agrees to keep a house which belongs to the former, located atCrocodilopolis-Arsinoe, the metropole of the Fayum, until the owner will indicate his desire to recover the useof his property, giving him 5 days to leave. The lack of a precise lease term and of any financial compensationis problematic. The unique character of this guarding contract is certainly linked to the difference in socioeconomicstatus of the two partners. Abstract

International audience; The current COVID-19 pandemic illustrates the importance of obtaining reliable methods for the rapid detection of SARS-CoV-2. A highly specific and sensitive diagnostic test able to differentiate the SARS-CoV-2 virus from common human coronaviruses is therefore needed. Coronavirus nucleoprotein (N) localizes to the cytoplasm and the nucleolus and is required for viral RNA synthesis. N is the most abundant coronavirus protein, so it is of utmost importance to develop specific antibodies for its detection. In this study, we developed a sandwich immunoassay to recognize the SARS-CoV-2 N protein. We immunized one alpaca with recombinant SARS-CoV-2 N and constructed a large single variable domain on heavy chain (VHH) antibody library. After phage display selection, seven VHHs recognizing the full N protein were identified by ELISA. These VHHs did not recognize the nucleoproteins of the four common human coronaviruses. Hydrogen Deuterium eXchange-Mass Spectrometry (HDX-MS) analysis also showed that these VHHs mainly targeted conformational epitopes in either the C-terminal or the N-terminal domains. All VHHs were able to recognize SARS-CoV-2 in infected cells or on infected hamster tissues. Moreover, the VHHs could detect the SARS variants B.1.17/alpha, B.1.351/beta, and P1/gamma. We propose that this sandwich immunoassay could be applied to specifically detect the SARS-CoV-2 N in human nasal swabs.

Abstract

International audience; Peculiar and unique growth mechanisms involved in semiconductor nanowires (NWs) pave the way to the achievement of new crystallographic phases and remarkable material properties, and hence, studying polytypism in semiconductor NWs arouses a strong interest for the next generation of electronic and photonic applications. In this context, the growth of ZnS nanowires has been investigated, as bulk ZnS compound exhibits numerous unstable polytypes at high temperatures, but their stable occurrence is highly anticipated in a nanowire due to its special quasi-dimensional shape and growth modes. In this work, the idea is to provide a change in the growth mechanism via the physical state of catalyst droplet (liquid or solid) and hence, study the induced structural modifications in ZnS nanowires. The HRTEM images of VLS (via liquid alloyed catalyst) grown ZnS NWs show periodic stacking faults, which is precisely identified as a stacking sequence of cubic or hexagonal individual planes leading to an astonishing 15R crystal polymorph. This crystallographic phase is observed for the first time in nanowires. Contrastingly, NWs grown with VSS (via solid catalyst) show crystal polytypes of zinc blende and wurtzite. We calculate and discuss the role of cohesive energies in the formation of such ZnS polytypes. Further, we present the selection rules for the crystallization of such 15R structure in NWs and discuss the involved VLS and VSS growth mechanisms leading to the formation of different crystal phases.

Abstract

De nombreuses méta-analyses mettent en avant les bénéfices de l’allaitement sur la santé et le développement de l’enfant. Néanmoins, peu d’études considèrent l’alimentation du nourrisson au cours de la première année dans sa globalité, ceci alors que les pratiques d’alimentation (alimentation lactée puis diversifiée) sont liées entre elles. Ainsi, dans une première étape, nous avons cherché à caractériser l’alimentation des nourrissons en tenant compte de la durée d’allaitement ou du recours aux formules infantiles, de l’âge d’introduction et de la fréquence de consommation des différents groupes d’aliments puis des morceaux et enfin, de la consommation d’aliments infantiles et d’aliments issus de l’agriculture biologique. Dans la cohorte nationale ELFE (Charles et al., 2020), 8922 nourrissons avaient des données exploitables pour répondre à cet objectif. Nos analyses ont montré que, au-delà de la durée de l’allaitement et de l’âge à la diversification alimentaire, la description des pratiques d’alimentation, nécessite de considérer l’âge d’introduction de certains groupes alimentaires spécifiques tels que les boissons sucrées, le lait de vache ou les céréales infantiles, et d’introduction des morceaux (Camier et al., 2020). Par contre, le type d’aliments consommés (infantiles ou issus de l’agriculture biologique) participait peu à la définition des différents types de pratiques identifiés en 2011-12. Concernant les caractéristiques familiales associées à ces pratiques, comme dans des études précédentes, les pratiques d’allaitement et de diversification plus en accord avec les recommandations sont liées avec un âge ou un niveau d’études maternels plus élevés. Nous montrons également que la migration et un faible niveau de revenus sont associés à un allaitement plus long. Enfin, la présence d’enfants plus âgés dans le foyer est associée à un allaitement plus long mais aussi à une introduction précoce du lait de vache.Lorsque nous regardons les liens entre ces pratiques d’alimentation et la croissance des enfants, nous constatons qu’une durée plus longue de l'allaitement était liée à un IMC plus faible à 1 an mais à un IMC plus élevé à 7 ans (Camier et al., soumis). À l'âge de 7 ans, les enfants allaités pendant plus de 6 mois présentaient un risque plus faible d'insuffisance pondérale que les enfants allaités pendant 1 à 3 mois. Par rapport à l'âge recommandé pour la diversification alimentaire (4-6 mois), une diversification précoce ( 6 mois) était liée à un risque plus élevé d'allergie alimentaire entre 1 et 5,5 ans. Un faible score de diversité à 10 mois était lié à un risque plus élevé de rhinoconjonctivite avant 5,5 ans. Enfin, plus le nombre d'aliments allergènes (parmi lait, œuf, blé et poisson) non introduits à 10 mois était élevé, plus le risque de maladies allergiques avant 5,5 ans était important, en particulier l'allergie alimentaire et la rhinoconjonctivite.Nous avons enfin analysé le lien entre les pratiques d’alimentation et le neurodéveloppement lors des suivis à 1 an et 3,5 ans, respectivement (Somaraki et al., en préparation). Nos résultats confirment l’association positive entre la durée d’allaitement et les scores de neurodéveloppement. Au-delà de l’allaitement, l'introduction de morceaux était associée à la motricité globale à l'âge d'un an : une introduction précoce, avant 8 mois, était associée à une probabilité moindre de faire partie du groupe " score de motricité globale inférieur " et une introduction tardive (après 10 mois) à une probabilité plus élevée, par rapport à une introduction entre 8 et 10 mois. Ces associations n'ont pas été observées à 3,5 ans. Les associations avec le langage expressif présentaient un schéma similaire, avec une probabilité plus faible de faire partie du groupe " score de langage expressif plus faible " chez les enfants auxquels on a introduit des morceaux d'aliments tôt, et probabilité plus élevée chez ceux auxquels on a présenté des morceaux d'aliments tard. Cette association est également observées à l'âge de 3,5 ans. Ainsi ceci suggère une association positive entre l'introduction lors d’une période sensible (entre 8 et 10 mois) d'aliments texturés et le développement de l'enfant, plus particulièrement la motricité globale et le langage expressif. Ces premières analyses seront poursuivies en intégrant d’autres dimensions de l’alimentation précoce, notamment sa dimension environnementale.

Abstract

International audience; Older female drivers could be considered victims of negative stereotypes for two reasons: being a woman and belonging to the older category; but there are no studies specifically in this double context. Two psychosocial questionnaires were created, one assessing the perception of driving abilities in different contexts (PDADC), the second, the perception of the attributes of driving (PAD) with a box for the activation of the stereotype threat. A population of 98 women, aged 65 ¬years or older, comprised the experimental and control groups (quasi-experimental design). This study provides knowledge about the driving feelings of this population who appears, globally, as comfortable at the wheel and "immune" to stereotyping.

Abstract

TULIP (Target Ion Source for Short‐Lived Isotope Production) is an original Isotope Separator On Line project dedicated to the production of short lived neutron deficient ions. It was tested for the first time in March 2022 at GANIL (Grand Accélérateur National d’Ions Lourds ) / SPIRAL1 (Système de Production d’Ions Radioactifs Accélérés en Ligne, phase 1) with the production of Rb isotopes using a primary beam of 22Ne at an energy of 4.5 MeV/A. 76Rb and 78Rb isotope rates of ~4.E3 and ~6.E4 pps were measured for a primary beam intensity of 6.E12 pps and may be improved by a factor between 5 to10. A remarkably short atom-to-ion transformation (AIT) time close to 200 µs was achieved. This result demonstrates how beneficial the design of the TULIP target ion source system is in term of response time and allows us to confidently look forward 74Rb+ production, which is one of the goal of the TULIP project.The results obtained on Rb production suggests testing the principle for the production of more sticky elements. Ions from metallic isotopes around 100Sn will be the next objective., TULIP (Target Ion Source for Short‐Lived Isotope Production) est un projet original de séparateur d'isotopes en ligne dédié à la production d'ions de faible durée de vie et déficients en neutrons. Des taux d'isotopes de 76Rb et de 78Rb de ~4.E3 et de ~6.E4 pps ont été mesurés pour une intensité de faisceau primaire de 6.E12 pps, et pourraient être améliorés d'un facteur 5 à 10.Un temps de transformation remarquablement court proche de 200 µs a été obtenu. Ces résultats démontrent combien la conception du système cible source TULIP peut être bénéfique en termes de temps de réponse et nous permet sereinement d'envisager la production d'ions 74Rb+, qui est un des objectifs du projet TULIP.Les résultats obtenus pour la production de Rb suggèrent de tester le principe pour la production d'éléments plus collants. La production d'ions à partir d'isotopes métalliques autour de l'étain 100 sera le prochain objectif.

Abstract

Background Muscle strength decreases as kidney failure progresses. Low muscle strength affects more than 50% of hemodialysis patients and leads to daily life activities impairment. In the general population, numerous studies have linked low 25OH-vitamin D (25OHD) concentrations to the loss of the muscle strength and low physical performances. Data on native vitamin D and muscle function are scarce in the chronic kidney disease (CKD) population, but low 25OHD levels have been associated with poor muscle strength. We present in this article the protocol of an ongoing study named VITADIAL testing if cholecalciferol supplementation in hemodialysis patients with low 25OHD improves their muscle strength. Methods/design VITADIAL is a prospective open randomized French multicenter study. All patients will have 25OHD levels ≤50nmol/L at randomization. One group will receive 100,000 UI cholecalciferol once a month during 6 months; the other group will receive no treatment during 6 months. In order to randomize patients with 25OHD ≤50nmol/L, supplemented patients will undergo a 3 months wash-out period renewable 3 times (maximum of 12 months wash-out) until 25OHD reaches a level ≤50nmol/L. The main objective of this study is to analyze if a 6-month period of oral cholecalciferol (i.e., native vitamin D) supplementation improves muscle strength of hemodialysis patients with low 25OHD vitamin D levels. Muscle strength will be assessed at 0, 3, and 6 months, by handgrip strength measured with a quantitative dynamometer. Secondary objectives are (1) to analyze 25OHD plasma levels after vitamin D wash-out and/or supplementation, as well as factors associated with 25OHD lowering speed during wash-out, and (2) to analyze if this supplementation improves patient’s autonomy, reduces frailty risk, and improves quality of life. Fifty-four patients are needed in each group to meet our main objective. Discussion In the general population, around 30 randomized studies analyzed the effects of vitamin D supplementation on muscle strength. These studies had very different designs, sizes, and studied population. Globally, these studies and the meta-analysis of studies favor a beneficial effect of vitamin D supplementation on muscle strength, but this effect is mainly found in the subgroup of aged patients and those with the lowest 25OHD concentrations at inclusion. We reported a positive independent association between 25OHD and handgrip strength in a population of 130 hemodialysis patients in a dose-dependent manner. In our cohort, a plateau effect was observed above 75 nmol/L. Only two randomized studies analyzed the effect of native vitamin D supplementation on muscle strength in hemodialysis patients, but unfortunately, these two studies were underpowered. VITADIAL is a trial specifically designed to assess whether cholecalciferol might benefit to hemodialysis patient’s muscle strength. Trial registration ClinicalTrials.govNCT04262934. Abstract

Hypothesis. Isoniazid is one of the primary drugs used in tuberculosis treatment. Isoniazid encapsulation in liposomal vesicles can improve drug therapeutic index and minimize toxic and side effects. In this work, we consider mixtures of hydrogenated soy phosphatidylcholine/phosphatidylglycerol (HSPC/DPPG) to get novel biocompatible liposomes for isoniazid pulmonary delivery. Our goal is to understand if the entrapped drug affects bilayer structure. Experiments. HSPC-DPPG unilamellar liposomes are prepared and characterized by dynamic light scattering, $\zeta$-potential, fluorescence anisotropy and Transmission Electron Microscopy. Isoniazid encapsulation is determined by UV and Laser Transmission Spectroscopy. Calorimetry, light scattering and Surface Pressure measurements are used to get insight on adsorption and thermodynamic properties of lipid bilayers in the presence of the drug. Findings. We find that INH-lipid interaction can increase the entrapment capability of the carrier due to isoniazid adsorption. The preferential INH-HSPC dipole-dipole interaction promotes modification of lipid packing and ordering and favors the condensation of a HSPC-richer phase in molar excess of DPPG. Abstract

We present a search for signatures of neutrino mixing of electron anti-neutrinos with additional hypothetical sterile neutrino flavors using the Double Chooz experiment. The search is based on data from 5 years of operation of Double Chooz, including 2 years in the two-detector configuration. The analysis is based on a profile likelihood, i.e.\ comparing the data to the model prediction of disappearance in a data-to-data comparison of the two respective detectors. The analysis is optimized for a model of three active and one sterile neutrino. It is sensitive in the typical mass range $5 \cdot 10^{-3} $ eV$^2 \lesssim \Delta m^2_{41} \lesssim 3\cdot 10^{-1} $ eV$^2$ for mixing angles down to $\sin^2 2\theta_{14} \gtrsim 0.02$. No significant disappearance additionally to the conventional disappearance related to $\theta_{13} $ is observed and correspondingly exclusion bounds on the sterile mixing parameter $\theta_{14} $ as function of $ \Delta m^2_{41} $ are obtained., Comment: accepted for publication by EPJC

Abstract

With the aim of designing a preclinical study evaluating an intracerebral cell-based therapy for stroke, an observational study was performed in the rat suture model of ischemic stroke. Objectives were threefold: (i) to characterize neurofunctional and imaging readouts in the first weeks following transient ischemic stroke, according to lesion subtype (hypothalamic, striatal, corticostriatal); (ii) to confirm that intracerebral administration does not negatively impact these readouts; and (iii) to calculate sample sizes for a future therapeutic trial using these readouts as endpoints. Our results suggested that the most relevant endpoints were side bias (staircase test) and axial diffusivity (AD) (diffusion tensor imaging). Hypothalamic-only lesions did not affect those parameters, which were close to normal. Side bias in striatal lesions reached near-normal levels within 2 weeks, while rats with corticostriatal lesions remained impaired until week 14. AD values were decreased at 4 days and increased at 5 weeks post-surgery, with a subtype gradient: hypothalamic < striatal < corticostriatal. Intracerebral administration did not impact these readouts. After sample size calculation (18-147 rats per group according to the endpoint considered), we conclude that a therapeutic trial based on both readouts would be feasible only in the framework of a multicenter trial.

Abstract

International audience; Flame speed is a sizing metric of a fuel to be considered in designing energetic industrial systems such as IC engines and gas turbines. Indeed, this physicochemical property of a combustible mixture notably influences the heat release rate and flame stabilization. Moreover, its significance makes it a major target for the validation of kinetic mechanisms. For example, the flame speed of ammonia or ethanol is around one fourth compared to traditional hydrocarbon fuels. Flame stabilization difficulties, possibly encountered in industrial systems, can be solved by technical adjustments (i.e. air preheating, compression ratio increase, oxygen-enriched air, co-firing with methane or syngas).The accuracy of experimental flame speed determination (target data for numerical schemes) for slow flames is the main issue to be addressed here.
