Abstract

International audience; This article presents an hybrid and hierarchical model in which two modeling and simulation approaches, discrete event system specification simulation (DEVS) and semantic technologies, were used together in order to help in the analysis of a major healthcare problem, the severe acute respiratory syndrome-coronavirus 2 (SARS-CoV-2). Indeed, the complexity of the SARS-CoV-2 replication process, and the range of hierarchical scales over which it interacts with cellular components (extending from genomic and transcriptomic to proteomic and metabolomic scales), and the intricate way in which they are interwoven, make its understanding very challenging. It is therefore crucial to model the different scales of the replication process, by taking into account all interactions with the infected cell. By combining the advantages of both DEVS simulation and ontological modeling, we propose a hierarchical ontology-based DEVS simulation model of the SARS-CoV-2 viral replication at both the micro-molecular (proteomic and metabolomic) and macro-molecular (genomic and transcriptomic) scales. First, we demonstrate the usefulness of combining DEVS simulation and semantic technologies in a common modeling framework to face the complexity of the SARS-CoV-2 viral replication at different scales. Second, the modeling and simulation of the SARS-CoV-2 replication process on different levels provide valuable information on the different stages of the virus’s life cycle and lays the foundation for a system to anticipate future mutations selected by the virus.

Abstract

International audience; Handwriting with connected pen becomes one of the major human-computer easy interaction methods. In comparison with traditional touch screen handwriting systems, the pen-based interaction method has the advantage of producing the online handwriting signal without surface constraints. Indeed, people who write on a paper obtain the corresponding pen trajectory coordinates that represent the online handwriting signal. Furthermore, the feeling of writing on paper is important particularly for the children during the learning of writing. In this work which is part of ANR Franco-German KIHT project with Stabilo, we introduce a domain adaptation-based approach that reconstructs the paper handwriting traces of the digital stylus Digipen of STABILO which is equipped with a wireless trajectory tracking system based on kinematic sensors. We use unsupervised domain adaptation method, to pass from the tablet domain where the ground truth (the online trace of the writing on the tablet) is known, to the paper domain where only the input sensors data are known.

Abstract

International audience; Handwriting with digital pens is a common way to facilitate human-computer interaction through the use of Online Handwriting (OH) trajectory reconstruction. In this work, we focus on a digital pen equipped with sensors from which one wants to reconstruct the OH trajectory. Such a pen allows to write on any surface and to get the digital trace, which can help learning to write, by writing on paper, and can be useful for many other applications such as collaborative meetings, etc. In this paper, we introduce a novel processing pipeline that maps the sensor signals of the pen to the corresponding OH trajectory. Notably, in order to tackle the difference of sampling rates between the pen and the tablet (which provides ground truth information), our preprocessing pipeline relies on Dynamic Time Warping to align the signals. We introduce a dedicated neural network architecture, inspired by a Temporal Convolutional Network, to reconstruct the online trajectory from the pen sensor signals. Finally, we also present a new benchmark dataset on which our method is evaluated both qualitatively and quantitatively, showing a notable improvement over its most notable competitor.

Abstract

International audience; CUPID-Mo, located in the Laboratoire Souterrain de Modane (France), was a demonstrator for the next generation $0\nu\beta\beta$ decay experiment, CUPID. It consisted of an array of 20 enriched Li$_{2}$$ ^{100}$MoO$_4$ bolometers and 20 Ge light detectors and has demonstrated that the technology of scintillating bolometers with particle identification capabilities is mature. Furthermore, CUPID-Mo can inform and validate the background prediction for CUPID. In this paper, we present a detailed model of the CUPID-Mo backgrounds. This model is able to describe well the features of the experimental data and enables studies of the $2\nu\beta\beta$ decay and other processes with high precision. We also measure the radio-purity of the Li$_{2}$$^{100}$MoO$_4$ crystals which are found to be sufficient for the CUPID goals. Finally, we also obtain a background index in the region of interest of 3.7$^{+0.9}_{-0.8}$(stat)$^{+1.5}_{-0.7}$(syst)$\times10^{-3}$counts/$\Delta$E$_{FWHM}$/mol$_{iso}$/yr, the lowest in a bolometric $0\nu\beta\beta$ decay experiment.

Abstract

Homogeneous and heterogeneous U1-xCexO2 (with 0≤ x≤ 0.25) materials were prepared via wet and dry chemistry routes, respectively before being submitted to dynamic leaching experiments. The feeding solution containing 0.20 mmol.L−1 H2O2 was kept under air and renewed to guarantee the stability of H2O2 during the experiment. Normalized alteration rates were determined from U concentration in the leachates. For homogeneous (U,Ce)O2 materials, the dissolution rate was divided by a factor of 3 when increasing the Ce content from 0.08 to 0.25. Surface characterizations revealed that studtite precipitated all over UO2 pellet surface and only on the UO2 grains of heterogeneous U0.92Ce0.08O2 samples. The behaviour of this heterogeneous material was similar to that observed for (U,Pu)O2 in the same conditions, which revealed the reliability of cerium as a plutonium analogue.

Abstract

International audience; Hydrazone compounds represent an important area of research that includes, among others, synthetic approaches and biological studies. A series of 17 hydrazones have been synthesized by mechanochemical means. The fragments chosen were phenolic and furanyl aldehydes coupled with 12 heterocyclic hydrazines or hydrazinamides. All compounds can be obtained quantitatively when operating on a planetary ball mill and a maximum reaction time of 180 min (6 cycles of 30 min each). Complete spectroscopic analyses of hydrazones revealed eight compounds (3–5, 8–11, 16) present in one geometric form, six compounds (1, 2, 13–15) present in two isomeric forms, and three compounds (6, 7, 12) where one rotation is restricted giving rise to two different forms. The single crystal X-ray structure of one of the hydrazones bearing the isoniazid fragment (8) indicates a crystal lattice consisting of two symmetry-independent molecules with different geometries. All compounds obtained were tested for anti-infectious and antibacterial activities. Four compounds (1, 3, 5 and 8) showed good activity against Mycobacterium tuberculosis, and one (7) was very potent against Staphylococcus aureus. Most interesting, this series of compounds displayed very promising antileishmanial activity. Among all, compound 9 exhibited an IC50 value of 0.3 µM on the Leishmania donovani intramacrophage amastigote in vitro model and a good selectivity index, better than miltefosine, making it worth evaluating in vivo.

Abstract

International audience; Random coincidences of events could be one of the main sources of background in the search for neutrino-less double-beta decay of $^{100}$Mo with macro-bolometers, due to their modest time resolution. Scintillating bolometers as those based on Li$_2$MoO$_4$ crystals and employed in the CROSS and CUPID experiments can eventually exploit the coincident fast signal detected in a light detector to reduce this background. However, the scintillation provides a modest signal-to-noise ratio, making difficult a pile-up pulse-shape recognition and rejection at timescales shorter than a few ms. Neganov-Trofimov-Luke assisted light detectors (NTL-LDs) offer the possibility to effectively increase the signal-to-noise ratio, preserving a fast time-response, and enhance the capability of pile-up rejection via pulse shape analysis. In this article we present: a) an experimental work performed with a Li$_2$MoO$_4$ scintillating bolometer, studied in the framework of the CROSS experiment, and utilizing a NTL-LD; b) a simulation method to reproduce, synthetically, randomly coincident two-neutrino double-beta decay events; c) a new analysis method based on a pulse-shape discrimination algorithm capable of providing high pile-up rejection efficiencies. We finally show how the NTL-LDs offer a balanced solution between performance and complexity to reach background index $\sim$$10^{-4}$ counts/keV/kg/year with 280~g Li$_2$MoO$_4$ ($^{100}$Mo enriched) bolometers at 3034 keV, the Q-value of the double-beta decay, and target the goal of a next generation experiment like CUPID.

Abstract

Legumes, and especially pulses, are an important source of protein for food and feed, and are appreciated for their positive impact on the “one health”. However, their unstable yields and their susceptibility to biotic and abiotic stresses highlight the need for varietal improvement in order to increase the cultivated areas and productivity. With the advent of sequencing technologies, a large pool of genetic and -omics resources, heterogeneous at the inter- and intra-species scale, is emerging. Thus, it is important to capitalize on these scattered heterogeneous data to develop translational research to boost breeding projects and crop diversification. To meet this need, we undertook the development of the Orthology-driven knowledge base framework for translational research (Ortho_KB). For a set of species of interest, it infers orthologous relationships between genes, proposes associated syntenic blocks between chromosomes and creates a graph database linking genetic and RNA-seq data. To explore the possibilities of this framework, we populated Ortho_KB to obtain OrthoLegKB, an instance dedicated to legumes. This database includes four cultivated crops, namely Pisum sativum, Vicia faba, Lens culinaris and Vigna radiata, and the model legume Medicago truncatula. Abstract

International audience; The ability to visualize RNA in its native subcellular environment by using single-molecule fluorescence in situ hybridization (smFISH) has reshaped our understanding of gene expression and cellular functions. A major hindrance of smFISH is the difficulty to perform systematic experiments in medium- or high-throughput formats, principally because of the high cost of generating the individual fluorescent probe sets. Here, we present high-throughput smFISH (HT-smFISH), a simple and cost-efficient method for imaging hundreds to thousands of single endogenous RNA molecules in 96-well plates. HT-smFISH uses RNA probes transcribed in vitro from a large pool of unlabeled oligonucleotides. This allows the generation of individual probes for many RNA species, replacing commercial DNA probe sets. HT-smFISH thus reduces costs per targeted RNA compared with many smFISH methods and is easily scalable and flexible in design. We provide a protocol that combines oligo pool design, probe set generation, optimized hybridization conditions and guidelines for image acquisition and analysis. The pipeline requires knowledge of standard molecular biology tools, cell culture and fluorescence microscopy. It is achievable in ~20 d. In brief, HT-smFISH is tailored for medium- to high-throughput screens that image RNAs at single-molecule sensitivity.

Abstract

A scintillating bolometer based on a large cubic Li$_{2}$$^{100}$MoO$_4$ crystal (45 mm side) and a Ge wafer (scintillation detector) has been operated in the CROSS cryogenic facility at the Canfranc underground laboratory in Spain. The dual-readout detector is a prototype of the technology that will be used in the next-generation $0\nu2\beta$ experiment CUPID. The measurements were performed at 18 and 12 mK temperature in a pulse tube dilution refrigerator. This setup utilizes the same technology as the CUORE cryostat that will host CUPID and so represents an accurate estimation of the expected performance. The Li$_{2}$$^{100}$MoO$_4$ bolometer shows a high energy resolution of 6 keV FWHM at the 2615 keV $\gamma$ line. The detection of scintillation light for each event triggered by the Li$_{2}$$^{100}$MoO$_4$ bolometer allowed for a full separation ($\sim$8$\sigma$) between $\gamma$($\beta$) and $\alpha$ events above 2 MeV. The Li$_{2}$$^{100}$MoO$_4$ crystal also shows a high internal radiopurity with $^{228}$Th and $^{226}$Ra activities of less than 3 and 8 $\mu$Bq/kg, respectively. Abstract

International audience; Three lanthanide complexes (Ln=Gd, Eu) based on a DO3 A ([Ln(L$_1$)]) or DO2 A ([Ln(L$_{2-3}$)]$^+$) platform appended by a redox active TEMPO-based arm were prepared. Complex [Ln(L$_2$)]$^+$) shows an alkyne arm, offering the possibility of postfunctionalization by click reaction to yield [Ln(L$_3$)]$^+$). The complexes demonstrate a redox response whereby the hydroxylamine, nitroxide and oxoammonium forms of the arm can be obtained in turn. Luminescence measurements on the europium complexes support an octadentate (L$_1$, L$_3$) or heptadentate (L$_2$) chelation by the ligand, with one water molecule in the inner coordination sphere. The relaxivity was determined from 20 kHz to 30 MHz by fast-field cycling NMR. The three Gd-III complexes under their hydroxylamine form [Gd(L$_1$)] and [Gd(L$_{2-3}$)]$^+$) show $r_1$ values of 7.0, 5.1 and 5.0 mM$^{-1}$ s$^{-1}$ (30 KHz), which increase to 8.8, 5.5 and 6.1 mM$^{-1}$ s$^{-1}$ in the nitroxide form. The radical complexes are not toxic against M21 cell lines, at least up to 40 $\mu$M. By using EPR spectroscopy we establish that they do not penetrate the cells with the exception of [Eu(L$_2$)]$^+$.

Abstract

International audience; Introduction: We aimed to describe patients with coexisting infective endocarditis (IE) and bacterial meningitis (BM).Methods: We merged two large prospective cohorts, an IE cohort and a BM cohort, with only cases of definite IE and community-acquired meningitis. We compared patients who had IE and BM concurrently to patients with IE only and BM only.Results: Among the 1030 included patients, we identified 42 patients with IE-BM (4.1%). Baseline characteristics of patients with IE-BM were mostly similar to those of patients with IE, but meningitis was the predominant presentation at admission (39/42, 92.3%). Causative pathogens were predominantly Streptococcus pneumoniae (18/42, 42.9%) and Staphylococcus aureus (14/42, 33.3%). All pneumococcal IE were associated with BM (18/18). BM due to oral and group D streptococci, Streptococcus agalactiae, and S. aureus were frequently associated with IE (14/30, 46.7%). Three-month mortality was 28.6% in patients with IE-BM, 20.5% in patients with IE, and 16.6% in patients with BM.Conclusions: Patients with pneumococcal IE or altered mental status during IE must be investigated for BM. Patients with S. aureus, oral and group D streptococcal or enterococcal BM, or unfavorable outcome in pneumococcal meningitis would benefit from an echocardiography. Patients with the dual infection have the worst prognosis. Their identification is mandatory to initiate appropriate treatment.

Abstract

Depuis les années 1980, on assiste à un remplacement des professionnel·les de l’éducation par des travailleuses techniciennes faiblement qualifiées et rémunérées, pour la gestion de l’hétérogénéité des élèves. Cette situation permet à l’État de mettre en place une politique publique à bas coût. Les AESH, en charge de l’inclusion des élèves à besoins particuliers, en sont un exemple. Malgré leur situation précaire, ces travailleuses développent des marges d’autonomie dans leur quotidien, dans la gestion du temps, des activités et des modalités de gestion de crises. À rebours de l’image préconçue de leur travail, cet article invite à questionner plus finement le rapport entre précarité objective et vécue. The period since the 1980s has witnessed numerous educational professionals’ replacement by less qualified and lower paid technical workers hired to help manage student heterogeneity – an arrangement enabling the implementation of low-cost public policy. The trend is exemplified by special needs assistants responsible for disabled pupils’ overall inclusion. Despite the precarity of their situation, however, these actors have developed margins of autonomy in their daily work through the way they manage time, activities and crises. Undermining preconceived notions of the jobs they do, the article calls for a more granular analysis of the relationship between objective and subjective precariousness. Desde los años ochenta, observamos un remplazo de los profesionales de la educación por trabajadoras técnicas poco calificadas y remuneradas para el manejo de la heterogeneidad de los alumnos. Esta situación permite al Estado establecer una política pública a bajo costo. Los AESH, a cargo de la inclusión de los alumnos con necesidades especiales, son un ejemplo de ello. A pesar de su situación precaria, estas trabajadoras desarrollan márgenes de autonomía en su trabajo diario, en la gestión del tiempo, actividades y modalidades de manejo de crisis. A la inversa de la imagen preconcebida de su trabajo, este artículo invita a cuestionar más finamente la relación entre precariedad objetiva y vivida.

International audience; The impact of pre-transplant anti-severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) vaccine in 20 recipients of allogeneic hematopoietic stem cell transplantation (Allo-HSCT) and/or their donors is reported here, showing that the persistence of anti-SARS-CoV-2 antibodies can be detected in almost all patients, whatever the type of vaccine used, and up to 9 months post transplant. Also, an anti-SARS-CoV-2 spike glycoprotein CD3+ T-cell response could be detected in six (35%) of 17 evaluable patients. This study provides a rationale to consider anti-SARS-CoV-2 vaccination of both recipients and donors before Allo-HSCT.

Abstract

International audience; Objectives: The specific roles of remission status, lupus low disease activity state (LLDAS), and damage accrual on the prognosis of pregnancies in women with systemic lupus erythematosus (SLE) are unknown. We analysed their impact on maternal flares and adverse pregnancy outcomes (APOs).Methods: We evaluated all women (≥18 years) with SLE enrolled in the prospective GR2 study with an ongoing singleton pregnancy at 12 weeks (one pregnancy/woman). Several sets of criteria were used to define remission, disease activity, and damage. APOs included: fetal/neonatal death, placental insufficiency with preterm delivery, and small-for-gestational-age birth weight. First trimester maternal and disease features were tested as predictors of maternal flares and APOs.Results: The study included 238 women (98.3% on hydroxychloroquine) with 230 live births. Thirty-five (14.7%) patients had at least one flare during the second/third trimester. At least one APO occurred in 34 (14.3%) women.Hypocomplementemia in the first trimester was the only factor associated with maternal flares later in pregnancy (p = 0.02), while several factors were associated with APOs. In the logistic regression models, damage by SLICC-Damage Index (OR 1.8, 95% CI: 1.1-2.9 for model 1 and OR 1.7, 95% CI: 1.1-2.8 for model 2) and lupus anticoagulant (LAC, OR 4.2, 95% CI: 1.8-9.7 for model 1; OR 3.7, 95% CI: 1.6-8.7 for model 2) were significantly associated with APOs.Conclusion: LAC and damage at conception were predictors of APOs, and hypocomplementemia in the first trimester was associated with maternal flares later in pregnancy in a cohort of pregnant patients with well-controlled SLE.Clinical trial registration number: ClinicalTrials.gov, https://clinicaltrials.gov, NCT02450396.

Abstract

National audience; La recherche clinique en médecine de ville, notamment en médecine générale, celle qui en 1re ligne de rencontre avec la population, est aujourd’hui un enjeu de santé publique. L’exercice de cette recherche a des spécificités qui la différencie de la recherche clinique conduite en milieu hospitalier ce qui nécessite une adaptation de ses conditions d’exercice : – le développement de la recherche en médecine de ville suppose la maîtrise de ses fondamentaux par les investigateurs, et donc leur présentation, en amont, dès le cycle initial de formation, suivi, puisqu'il s'agit d'un domaine de connaissance complexe et évolutif, d'une participation des praticiens à des modules de formation adaptées à la recherche en soins primaires à l’exemple de ce que plusieurs Groupement interrégional de recherche clinique et d’innovation (GIRCI) ont déjà organisés. Pour compenser le mode éclaté de leur localisation, sur le modèle des équipes mobiles de recherche clinique en cancérologie (EMRC) en cancérologie, des équipes mobiles de recherche doivent mettre les cabinets de médecine générale en capacité de participer aux essais cliniques. Ceci suppose, d’une part, l’allocation de financements fléchés permettant de pérenniser un socle de personnels dédiés, et, d’autre part, l’impulsion d’une dynamique nationale au travers de la mise en place d’un Institut thématique multi-organisme de « Recherche en soins primaires » associé, au plan opérationnel, à un réseau d’investigation d’envergure nationale porté par une plateforme d’excellence ; – le recours aux outils et innovations digitales (télémédecine ; recueil des données via des outils connectés ; e-consentement ; signature électronique) qui permettent de dématérialiser et délocaliser tout ou partie des actes de la recherche que ce soit pour le participant ou les équipes d’investigations ; – une adaptation du cadre juridique afin de rapprocher le lieu de recherche du patient et non l’inverse, ce qui revient à déplacer les matériels et les investigations au plus près du patient ; – la prise en compte de l’acceptabilité du patient, avec donc le souci de limiter la perturbation que peut représenter sa participation à un protocole de recherche et la motivation du praticien en valorisant sa contribution et en apportant toutes les garanties de pertinence scientifique et d’indépendance d’exercice. Au vu de l’analyse contextuelle, des retours d’expérience positifs et de la disponibilité de points d’appui organisationnels et digitaux facilitant la délocalisation et dématérialisation de la conduite de l’activité de recherche au plus près du patient et de son médecin, la table ronde a conclu que des opportunités existent aujourd’hui qui favorisent le développement de la recherche clinique en médecine générale. Il convient de s’en saisir et de profiter, sans tarder, de ce moment propice.

La crise migratoire est-elle réellement une crise ?« Crise migratoire », « invasion », « grand remplacement », « ruée » : autant d’expressions employées pour décrire l’immigration vers la France, l’Europe ou les « pays du Nord ». Ni anodins ni complètements nouveaux, ces termes font des étrangers une menace.Mais sommes-nous réellement au cœur d’une crise migratoire exceptionnelle ? Ou s’agit-il là d’un discours politique et non étayé, alimenté par la peur de l’Autre ? Un discours qui se répète depuis plus d’un siècle et dont seules changent les cibles, et qui se durcit à la faveur de chaque nouvelle crise économique.Il est temps de lever le voile sur le processus de fabrication de ce discours sur l’immigration, plus que jamais instrumentalisé au profit de différents acteurs – politiques, entreprises et institutions –, et d’interroger le modèle de société que nous souhaitons bâtir.

Abstract

Legumes, and especially pulses, are an important source of protein for food and feed, and areappreciated for their positive impact on the “one health”. However, their unstable yields andtheir susceptibility to biotic and abiotic stresses highlight the need for varietal improvement inorder to increase the cultivated areas and productivity. With the advent of sequencingtechnologies, a large pool of genetic and -omic resources, heterogeneous at the inter- and intraspecies scale, is emerging. Thus, it is important to capitalize on these scattered heterogeneousdata to develop translational research and boost breeding projects and crop diversification.To meet this need, we undertook the development of NeoLeg, a NoSQL graph-based databaseusing Neo4j, dedicated for translational research in pulses. Starting from genome sequencesand annotation files, we inferred orthologous relationships between genes, and proposedassociated syntenic blocks between the chromosomes of four cultivated crops, namely Pisumsativum, Vicia faba, Lens culinaris and Vigna radiata, and model legume Medicago truncatula.Available information on quantitative trait loci (QTL) for multiple traits are being included aswell as expression data. The proposed modeling was evaluated in studying the conservation ofa flowering-promoting gene. The main achievements as well as remaining challenges andperspectives will be discussed

International audience; Let-7e precursor microRNA has the potential to adopt a G-quadruplex (rG4) structure and recently, its roles in oncology have been the focus of much attention, as it is now known that let-7e pre-miRNA is frequently dysregulated in cancers. Therefore, it is crucial to unveil and fully characterize its ability to adopt a rG4 structure, which could be stabilized or destabilized by small molecules and proteins such as nucleolin, a protein that is deeply associated with miRNA biogenesis. Herein, by combining a set of different methods such as circular dichroism (CD), nuclear magnetic resonance (NMR), UV spectroscopy (thermal difference spectra (TDS) and isothermal difference spectra (IDS)) and polyacrylamide gel electrophoresis (PAGE), we demonstrate the formation of the rG4 structure found in let-7e pre-miRNA sequence in the presence of K+ (5'-GGGCUGAGGUAGGAGG-3'). The ability of eight small molecules (or ligands) to bind to and stabilize this rG4 structure was also fully assessed. The dissociation constants for each RNA G-quadruplex/ligand complex, determined by surface plasmon resonance (SPR), ranged in the 10-6 to 10-9 M range. Lastly, the binding of the rG4 structure to nucleolin in the presence and absence of ligands was evaluated via CD, SPR, PAGE and confocal microscopy. The small molecules 360 A and PDS demonstrated attractive properties to targetthe rG4 structure of let-7e pre-miRNA and control its biology. Our findings also highlighted that the interaction of TMPyP4 with the G-quadruplex of let-7e precursor miRNA could block the formation of the complex between the rG4 and nucleolin. Overall, this study introduces an approach to target the rG4 found in let-7e pre-miRNA which opens up a new opportunity to control the microRNA biogenesis.

Abstract

The White-breasted Thrasher (Ramphocinclus brachyurus) is the smallest passerine bird in the Mimids family. Ranked as “endangered” in the IUCN list, the demographic distribution of this species is spread unevenly across two islands in the Lesser Antilles. The Saint-Lucian subspecies, Ramphocinclus brachyurus sanctaeluciae, was estimated to number between 1200 and 1700 individuals, whereas the Martinique subspecies, Ramphocinclus brachyurus brachyurus, was estimated at between 200 and 400 individuals. As an endemic rare species with a high risk of extinction, the Martinique subspecies should be regarded as a conservation priority. The small size of the Martinique subspecies population, its restricted geographical range, and its narrow habitat tolerance make it quite difficult to study. The variability of morphological traits in this subspecies has not been biostatistically analyzed recently. In order to update knowledge and fill in any gaps, we have used the morphometric approach to study the morphological variability of the Martinique subspecies. The study site is a small peninsula located in the northeast of the island of Martinique (French West Indies) known as “la Caravelle”. We captured 63 White-breasted Thrashers which were marked, measured, and sexed before being released. We did not observe any significant differences between the sexes, signifying an absence of sexual dimorphism. However, we were able to observe a significant variation in morphological traits between individuals measured inside and outside the national protected area of Caravelle. The body mass-tarsus ratio was equal at both sampled sites, something which could potentially be a clue to the bird’s adaptation to different habitats.

Abstract

PD/BD/142851/2018 PD/00065/2013 MIT-EXPL/BIO/0008/2017 IF/00959/2015 One of the most significant challenges in capturing and detecting biomarkers is the choice of an appropriate biomolecular receptor. Recently, RNA G-quadruplexes emerged as plausible receptors due to their ability to recognize with high-affinity proteins. Herein, we have unveiled and characterized the capability of the precursor microRNA 149 to form a G-quadruplex structure and determined the role that some ligands may have in its folding and binding capacity to nucleolin. The G-quadruplex formation was induced by K+ ions and stabilized by ligands, as demonstrated by nuclear magnetic resonance and circular dichroism experiments. Surface plasmon resonance measurements showed a binding affinity of precursor microRNA 149 towards ligands in the micromolar range (10−5–10−6 M) and a strong binding affinity to nucleolin RNA-binding domains 1 and 2 (8.38 × 10−10 M). Even in the presence of the ligand PhenDC3, the binding remains almost identical and in the same order of magnitude (4.46 × 10−10 M). The molecular interactions of the RNA G-quadruplex motif found in precursor miRNA 149 (5′-GGGAGGGAGGGACGGG- 3′) and nucleolin RNA-binding domains 1 and 2 were explored by means of molecular docking and molecular dynamics studies. The results showed that RNA G-quadruplex binds to a cavity between domains 1 and 2 of the protein. Then, complex formation was also evaluated through polyacrylamide gel electrophoresis. The results suggest that precursor microRNA 149/ligands and precursor microRNA 149/nucleolin RNA-binding domains 1 and 2 form stable molecular complexes. The in vitro co-localization of precursor microRNA 149 and nucleolin in PC3 cells was demonstrated using confocal microscopy. Finally, a rapid and straightforward microfluidic strategy was employed to check the ability of precursor microRNA 149 to capture nucleolin RNA-binding domains 1 and 2. The results revealed that precursor microRNA 149 can capture nucleolin RNA-binding domains 1 and 2 labeled with Fluorescein 5-isothiocyanate in a concentration-dependent manner, but PhenDC3 complexation seems to decrease the ability of precursor microRNA 149 to capture the protein. Overall, our results proved the formation of the G-quadruplex structure in the precursor microRNA 149 and the ability to recognize and detect nucleolin. This proof-of-concept study could open up a new framework for developing new strategies to design improved molecular receptors for capture and detection of nucleolin in complex biological samples. publishersversion published

Abstract

The MMX infrared spectrometer (MIRS) is an imaging spectrometer onboard MMX JAXA mission. MMX (Martian Moon eXploration) is scheduled to be launched in 2024 with sample return to Earth in 2029. MIRS is built at LESIA-Paris Observatory in collaboration with four other French laboratories, collaboration and financial support of CNES and close collaboration with JAXA and MELCO. The instrument is designed to fully accomplish MMX’s scientific and measurement objectives. MIRS will remotely provide near-infrared spectral maps of Phobos and Deimos containing compositional diagnostic spectral features that will be used to analyze the surface composition and to support the sampling site selection. MIRS will also study Mars atmosphere, in particular spatial and temporal changes such as clouds, dust and water vapor. Graphical Abstract

Abstract

Purpose Digital PET involving silicon photomultipliers (SiPM) provides an enhanced time-of-flight (TOF) resolution as compared with photomultiplier (PMT)-based PET, but also a better prevention of the count-related rises in dead time and pile-up effects mainly due to smaller trigger domains (i.e., the detection surfaces associated with each trigger circuit). This study aimed to determine whether this latter property could help prevent against deteriorations in TOF resolution and TOF image quality in the wide range of PET count rates documented in clinical routine. Methods Variations, according to count rates, in timing resolution and in TOF-related enhancement of the quality of phantom images were compared between the first fully digital PET (Vereos) and a PMT-based PET (Ingenuity). Single-count rate values were additionally extracted from the list-mode data of routine analog- and digital-PET exams at each 500-ms interval, in order to determine the ranges of routine PET count rates. Results Routine PET count rates were lower for the Vereos than for the Ingenuity. For Ingenuity, the upper limits were estimated at approximately 21.7 and 33.2 Mcps after injection of respectively 3 and 5 MBq.kg-1 of current 18F-labeled tracers. At 5.8 Mcps, corresponding to the lower limit of the routine count rates documented with the Ingenuity, timing resolutions provided by the scatter phantom were 326 and 621 ps for Vereos and Ingenuity, respectively. At higher count rates, timing resolution was remarkably stable for Vereos but exhibited a progressive deterioration for Ingenuity, respectively reaching 732 and 847 ps at the upper limits of 21.7 and 33.2 Mcps. The averaged TOF-related gain in signal/noise ratio was stable at approximately 2 for Vereos but decreased from 1.36 at 5.8 Mcps to 1.14 and 1.00 at respectively 21.7 and 33.2 Mcps for Ingenuity. Conclusion Contrary to the Ingenuity PMT-based PET, the Vereos fully digital PET is unaffected by any deterioration in TOF resolution and consequently, in the quality of TOF images, in the wide range of routine PET count rates. This advantage is even more striking with higher count-rates for which the preferential use of digital PET should be further recommended (i.e., dynamic PET recording, higher injected activities). Abstract

Pseudomonas aeruginosa possesses one of the most complex bacterial regulatory networks, which largely contributes to its success as a pathogen. However, most of its transcription factors (TFs) are still uncharacterized and the potential intra-species variability in regulatory networks has been mostly ignored so far. Here, we used DAP-seq to map the genome-wide binding sites of all 55 DNA-binding two-component systems (TCSs) response regulators (RRs) across the three major P. aeruginosa lineages. The resulting networks encompass about 40% of all genes in each strain and contain numerous new regulatory interactions across most major physiological processes. Strikingly, about half of the detected targets are specific to only one or two strains, revealing a previously unknown large functional diversity of TFs within a single species. Three main mechanisms were found to drive this diversity, including differences in accessory genome content, as exemplified by the strain-specific plasmid in IHMA87 outlier strain which harbors numerous binding sites of conserved chromosomally-encoded RRs. Additionally, most RRs display potential auto-regulation or RR-RR cross-regulation, bringing to light the vast complexity of this network. International audience; La métropolisation et la périurbanisation à l'œuvre en France depuis l'après-guerre ont structuré le paysage socio-spatial de la mégarégion de Paris selon des processus complexes opérant à des rythmes et des échelles divers. Si la tertiarisation de l'emploi et la croissance des cadres et professions intellectuelles supérieures y ont été plus importantes que dans le reste de la France, le phénomène ne s'est pas répandu de manière uniforme dans cet espace, mais bien au profit de Paris et des villes les plus grandes et les plus proches de la capitale. Nous assistons donc à un renforcement des divisions sociales déjà présentes tant à l'échelle des villes qu'entre les villes elles-mêmes.
