Infant formulas (IFs) are used as substitutes for human milk and are mostly based on cow milk proteins. For sustainability reasons, animal protein alternatives in food are increasingly being considered, as plant proteins offer interesting nutritional and functional benefits for the development of innovative IFs. This study aimed to assess how a partial substitution (50%) of dairy proteins with faba bean and pea proteins influenced the digestibility of IFs under simulated dynamic in vitro digestion, which were set up to mimic infant digestion. Pea- and faba bean-based IFs (PIF and FIF, respectively) have led to a faster aggregation than the reference milk-based IF (RIF) in the gastric compartment; that did not affect the digesta microstructure at the end of digestion. The extent of proteolysis was estimated via the hydrolysis degree, which was the highest for FIF (73%) and the lowest for RIF (50%). Finally, it was apparent that in vitro protein digestibility and protein digestibility-corrected amino acid score (PDCAAS)-like scores were similar for RIF and FIF (90% digestibility; 75% PDCAAS), but lower for PIF (75%; 67%). Therefore, this study confirms that faba bean proteins could be a good candidate for partial substitution of whey proteins in IFs from a nutritional point of view, provided that these in vitro results are confirmed in vivo.

Infant formulas (IF) are the only source of nutrition for infants when breastfeeding is not possible. Usually, IF are spray-dried to a powder form and include cow milk proteins as well as whey protein concentrate. In the future, plant proteins could be an interesting alternative to the use of animal proteins for nutritional, functional and sustainable concerns. The objective of this work was to study the influence of the partial substitution of whey protein concentrate by plant proteins on the functional properties and the digestibility of IF. For this purpose, proteins from pea, faba bean, rice and potato replaced half of the proteins in plant-based IF and where compared to a reference IF based on whey proteins. The digestibility of these IF, manufactured at pilot scale then at semi-industrial scale, was assessed through in vitro digestion simulating infant physiological conditions using static and dynamic models. Although all IF showed a balanced nutritional profile compared to the infant needs, it was demonstrated that the protein source had a great impact on IF properties. Indeed, solubility and viscosity limits observed for rice and potato IF affected their manufacturing as well as their digestibility. On the contrary, pea and faba bean IF showed physicochemical properties and protein nutritional quality very close to the reference. These promising results highlight future innovation possibilities in infant nutrition field. However, they need to be validated at the real industrial scale and extended through in vivo studies.; Les formules infantiles (FI) sont la seule source nutritionnelle pour les nourrissons lorsque l’allaitement est impossible. Les FI en poudre sont obtenues par séchage par atomisation et sont composées de protéines de lait de vache, majoritairement des protéines du lactosérum. A l’avenir, les protéines végétales pourraient constituer une alternative nutritionnelle, fonctionnelle et durable à l’utilisation de protéines animales. L’objectif de ce travail était d’étudier l’influence de la substitution partielle des protéines laitières par des protéines végétales sur les propriétés fonctionnelles et la digestibilité de FI innovantes. A cet effet, des protéines de pois, féverole, riz et pomme de terre ont remplacé la moitié des protéines dans des FI végétalisées et ont été comparées à une FI de référence composée de protéines du lactosérum. La digestibilité de ces FI, fabriquées à échelle pilote puis semi-industrielle, a été étudiée en digestions in vitro statique puis dynamique simulant les conditions physiologiques du nourrisson. La source de protéine influence les propriétés des FI, qui étaient cependant toutes nutritionnellement adaptées aux besoins du nourrisson. Les limites de solubilité et de viscosité observées pour les FI à base de riz et de pomme de terre affectent leur fabrication et leur digestibilité. A l’inverse, les FI à base de pois et de féverole présentent des propriétés physicochimiques et nutritionnelles très proches de la référence. Ces résultats prometteurs éclairent les possibilités d’innovation en nutrition infantile. Ils devront toutefois être confirmés à échelle industrielle et complétés par des études in vivo.

Infant formulas (IF) are the key nutritional source for infants who cannot be breastfed. There is currently a growing interest for these sensitive products, in order to control their quality and to design their composition with regard to nutritional balance and sustainable development concerns.This work aimed to study how plant proteins influence the digestibility of IF.Five infant formulas were developed with identical composition except that the protein source were whey proteins in the “reference IF”, pea, faba bean, rice or potato proteins in the four “plant IF” tested. The five IF were first evaluated using an in vitro static gastrointestinal model simulating infant conditions. Protein hydrolysis rate (PHR) and amino acid bioaccessibility (AAB) were measured using free amino group and free amino acid analyses, respectively. The pea and faba bean IF were then submitted to an in vitro dynamic digestion model closer to the physiological conditions, and compared to the reference IF. Protein digestibility (PHR and AAB) as well as the digesta microstructure were followed.Results showed that both PHR and AAB were very similar between the reference IF and pea and faba bean IF, but significantly lower for rice and potato IF. In dynamic conditions, the structure disintegration and the PHR showed differences among the reference, pea and faba bean IF. These differences were more pronounced during the gastric phase compared to the end of the intestinal digestion. This could result from the structure of proteins obtained from the processing steps that is known to influence the protein hydrolysis.This study provides new insights on the impact of protein source on the digestibility of IF. It especially showed that some plant proteins could be appropriate candidates for the composition of IF from a nutritional point of view.

Introduction et but de l’étude Les protéines généralement utilisées dans les formules infantiles proviennent du lait de vache et sont majoritairement des protéines de lactosérum afin de se rapprocher de la composition du lait maternel. Cependant, certaines protéines végétales offrent également de bonnes propriétés nutritionnelles et pourraient constituer d’intéressants substituts dans un contexte de compétition d’usage des protéines de lactosérum. Le but de cette étude était de comprendre l’impact de la substitution des protéines de lactosérum par des protéines végétales sur le degré d’hydrolyse et sur la bioaccessibilité des acides aminés. Matériels et Méthode Quatre protéines végétales ont été sélectionnées (PV1 à PV4) en raison de leur composition en acides aminés essentiels adaptée aux besoins du nourrisson. Elles ont été utilisées pour développer des formules infantiles de composition identique à la référence (R), à ceci près que les protéines de lactosérum étaient remplacées par une des quatre protéines végétales. Les cinq formules infantiles ont ensuite été soumises à un modèle de digestion in vitro gastro-intestinale en conditions statiques (Ménard et al. 2018). Le degré d’hydrolyse des protéines était mesuré par la méthode OPA (O-phtaldialdehyde). Les acides aminés libres ont été quantifiés par chromatographie échangeuse de cations. Résultats Tableau 1. AAS (acides aminés soufrés) ; AAA (acides aminés aromatiques) Les formules PV2 et PV3 libèrent autant d’acides aminés que la référence (R) et présentent un degré d’hydrolyse équivalent (p>0.05). En revanche, la formule PV4 se distingue de R par une moindre libération d’isoleucine et de leucine et présente également un degré d’hydrolyse plus faible. Enfin, la formule PV1 conduit à des valeurs très inférieures à celles obtenues avec R pour toutes les variables étudiées. Conclusion Il est établi que la cinétique de digestion des protéines dans une matrice et par conséquence, la libération des acides aminés et leur absorption peuvent être influencées notamment par sa composition en protéines (Le Feunteun et al., 2014). De manière concordante, nous montrons ici que la nature des protéines des formules infantiles aurait un effet sur la protéolyse. Ainsi, parmi les quatre formules « végétalisées » testées, seules PV2 et PV3 sembleraient être nutritionnellement comparables à la référence. Des digestions in vitro en conditions dynamiques (plus proches des conditions physiologiques) seront réalisées prochainement afin de compléter ces résultats quant à la qualité nutritionnelle de ces formules infantiles innovantes.

Les recommandations de l’OMS pour l’alimentation du nourrisson et du jeune enfant sont un allaitement exclusif jusqu’à l’âge de 6 mois, puis un allaitement partiel jusqu’à l’âge de 2 ans. Le taux et la durée de l’allaitement maternel en France restent cependant plus bas que les autres pays d’Europe. En 2017 en France, seulement 38% des enfants sont allaités jusqu’à l’âge de 6 mois (WHO/UNICEF, 2017). Lorsque l’allaitement maternel n’est pas possible ou non souhaité pour diverses raisons culturelles, économiques ou personnelles, les formules infantiles restent l’alternative la plus adaptée à la nutrition du nourrisson si leurs compositions respectent les besoins nutritionnels du jeune enfant. Ces formules doivent participer au développement optimale du nourrisson tout en respectant ses contraintes physiologiques, liées notamment à une immaturité digestive et rénale. Les formules infantiles doivent donc se rapprocher le plus possible du lait maternel. Leur composition nutritionnelle est globalement très proche, mais la composition fine et la structure diffèrent de par l’organisation des gouttelettes lipidiques et de la nature des protéines. En terme de digestion, étant donné l’immaturité du tube digestif à la naissance, des conditions de spécificités digestives adaptées aux nourrissons sont à prendre en considération afin de mettre en place des modèles de digestion in vitro pertinents (Bourlieu et al., 2014). Ces modèles sont utilisés aujourd’hui pour des raisons éthique, économique et de praticité pour les études de digestibilité. L’amélioration des formules infantiles, via la recherche et l’innovation, permet de répondre aux besoins nutritionnels et aux enjeux économiques. Des formules infantiles ont été développées pour répondre aux difficultés telles que les allergies, les intolérances, les problèmes de régurgitation, les troubles digestifs ou encore une production plus respectueuse de l’environnement. L’incorporation de probiotiques pour reproduire une partie du microbiote du lait maternel fait également partie de ces nouvelles générations de formules infantiles, mais également des formules infantiles de structures lipidiques et de composition en protéines plus proche du lait maternel ont été mises au point. De nouvelles perspectives visent à rechercher des protéines alternatives aux protéines de lactosérum du lait de vache via l’introduction de nouvelles sources de protéines d’origines végétales. L’objectif est de rechercher et d’étudier l’apport de nouvelles sources protéiques et d’aller jusqu’à la faisabilité technologique et la mesure de digestibilité en vue de s’assurer de la valeur nutritionnelle de ces nouvelles sources protéiques.

However this population has specific nutritional needs and often, oral impairments such as loss of tooth and/or decrease of salivary flow. Therefore it is necessary to develop foods adapted for the elderly, in terms of organoleptic properties and nutritional composition. In this context, the purpose of this work is to study the impact of oral impairments of elderly persons on food comfortability and bolus properties during the consumption of dairy products.76 elderly persons (ages 66 to 88, 42 women and 34 men), with or without oral health problems in terms of dental and/or salivary status were recruited. Four dairy products with identical composition but different textures (soft, hard, pasty and dry) were studied. Food comfortability of the four dairy products was assessed by means of a questionnaire based on bolus formation, pain, and texture and flavor perception. The time and number of cycle required to form the bolus for elderly person consuming each product was measured (video) as rheological properties (compression test) and saliva incorporated (dry extract) in each bolus. Statistical analysis (correlation, ANOVA, multiblock analysis) of the data were performed.Despite a wide array of texture, dairy products were comfortable for the elderly whatever their oral health. According to the food comfortability questionnaire, neither bolus formation, nor pain or organoleptic properties were significantly affected by the dental and/or salivary status of the elderly. The stimulated saliva flow was positively correlated with the amount of saliva incorporated in the bolus. Moreover dental status was correlated with the hardness of the bolus and results suggested that the dental status slightly affected the texture perception.Finally, food comfortability of our products was not affected by the oral health contrary to the bolus properties.

Calcium bioaccessibility depends mainly on solubility. Furthermore in vitro digestion allows to determine bioaccessibility and to identify the key parameters affecting it. This study focused on the influence of the gastro-intestinal pH, the food structure and the calcium source on the bioaccessibility of the nutrient. Four calcium sources (calcium carbonate, calcium citrate malate, dairy calcium phosphate and calcium bisglycinate) were added at 0.5% (w/w) in water (control), and in whey protein matrices as the liquid form or with heating to induce a gel. These matrices were digested under static conditions in duplicate, with the harmonized INFOGEST protocol [1]. Soluble and ionic calcium, and pH kinetics were followed during in vitro digestion. The kinetics were analyzed by nonparametric tests with repeated measurements. Results showed that all calcium sources were soluble in the matrices, mainly as ionic form (Ca2+), during the gastric phase at pH 3, but that calcium become insoluble in the intestinal compartment at pH 7. This rise in calcium insolubility led to a fall of the nutrient bioaccessibility from 20 to 80%. Dairy calcium phosphate had a distinct behavior with an important fall of solubility in the intestinal compartment due to strong ionic interactions between calcium and phosphate. In addition, the organization of whey proteins in the food matrix had no effect on solubilisation of calcium and therefore on its bioaccessibility. These results provide new insights to better understand bioavailability differences between calcium sources and matrices. Further in vivo digestion experiments on a pig model will be achieved in order to confirm bioaccessibility and to measure the bioavailability of those different sources of calcium. It will finally provide a relation between the results from in vitro and in vivo models of digestion.

Dairy calcium phosphate insolubilisation was quicker and more intense than for other calcium sources; its concentration of soluble calcium is 17+/-3%, instead of more than 60% for the three other calcium sources (p0.1). These results provide new insights to better understand bioavailability differences between calcium sources and matrices. Further in vivo digestion experiments on a pig model will be achieved in order to confirm bioaccessibility and to measure the bioavailability of those different sources of calcium. It will finally provide a relation between the results from in vitro and in vivo models of digestion.

National audience; Discipline : Expérimental/mécanismes cellulaires et moléculaires Introduction et but de l’étude : Le calcium est un élément essentiel pour le développement et le maintien de la masse osseuse chez l’Homme. Pourtant, en France, seulement 40% des seniors ont un apport suffisant en calcium [1]. En moyenne, la biodisponibilité du calcium est de 35% et varie en fonction des sources de calcium [2] et de la composition de l’aliment. Ce protocole permet d'estimer la bioaccessibilité du calcium pouvant être absorbé par l’organisme. Matériel et méthodes : Quatre sources de calcium (carbonate de calcium, citrate malate de calcium, bisglycinate de calcium et phosphate de calcium laitier contenant respectivement 40 %, 23 %, 13 % et 28 % de calcium) ont été ajoutées à 0,5% dans l’eau (contrôle), et dans deux matrices constituées de protéines sériques [20 % lipides +24% protéines dont 23 % protéines sériques], sous forme liquide ou gélifiée par traitement thermique. Les matrices ont été digérées en double en suivant le protocole harmonisé INFOGEST de digestion in vitro statique. Les concentrations en calcium sous forme soluble et ionique, ainsi que le pH, ont été mesurées au cours de la digestion. L’ensemble des cinétiques ont été analysées à l’aide d’un test non paramétrique en mesures répétées. Résultats et Analyse statistique : Dans les matrices alimentaires, la totalité du calcium est sous forme soluble et majoritairement sous forme ionique en phase gastrique à pH 3. Puis en début de phase intestinale, le calcium s’insolubilise. Le phosphate de calcium laitier s’insolubilise plus rapidement que les autres sources lors du passage en phase intestinale (p0,1).Conclusion : Le pH gastro-intestinal joue un rôle majeur sur la solubilité du calcium avec des différences selon les sources utilisées. En revanche, la gélification de la matrice protéique (majoritairement constituée de protéines sériques) n’a pas eu d’impact sur la solubilité des calciums testés. Cette description de la bioaccessibilité des sources de calcium au cours du parcours digestif, présentant des variations de pH très importantes, permet d’apporter des éléments de compréhension aux différences de biodisponibilité du calcium en fonction du segment intestinal. Il serait nécessaire de pouvoir corréler ces bioaccessibilités mesurées aux biodisponibilités évaluées in vivo.
