Summary: The all‐vanadium redox flow battery (VRFB) is emerging as a promising technology for large‐scale energy storage systems due to its scalability and flexibility, high round‐trip efficiency, long durability, and little environmental impact. As the degradation rate of the VRFB components is relatively low, less attention has been paid in terms of VRFB durability in comparison with studies on performance improvement and cost reduction. This paper reviews publications on performance degradation mechanisms and mitigation strategies for VRFBs in an attempt to achieve a systematic understanding of VRFB durability. Durability studies of individual VRFB components, including electrolyte, membrane, electrode, and bipolar plate, are introduced. Various degradation mechanisms at both cell and component levels are examined. Following these, applicable strategies for mitigating degradation of each component are compiled. In addition, this paper summarizes various diagnostic tools to evaluate component degradation, followed by accelerated stress tests and models for aging prediction that can help reduce the duration and cost associated with real lifetime tests. SUMMARY The microstructures of catalyst layers (CLs) in proton exchange membrane fuel cells determine cell performance and durability. Delicate ink preparation processes and coating/drying processes affect the resulting microstructures including active sites, pore networks, ionomer networks and Pt/C networks. This paper reports our recent experimental observations of the effect of ink dilution and evaporation condition on the microstructures. The microstructures of dried ink droplets are presented and compared among different dilution ratios and different evaporation conditions in terms of the spatial distributions of Pt/C particles, ionomers, and pores. The method through which the microstructures are visualized is also introduced in this paper. It is observed that ink dilution ratio and evaporation condition can significantly alter resulting microstructure patterns through affecting viscosity and particle flow patterns during the evaporation. More concentrated solution makes catalyst inks less spread out on a substrate surface, leading to larger droplet height and larger contact angle. Ambient relative humidity has a significant impact on catalyst deposition patterns. Under low relative humidity condition, catalyst particles are concentrated both near the central and the periphery of the droplet; while under high relative humidity, the central part is uniform, and the particles move towards the edge of the deposition, forming a stripe‐like structure. This indicates that ink dilution and evaporation is key to the CL microstructure formation and must be properly controlled in order to obtain the quality and consistency of the CLs in fabrication. Inflammatory bowel disease (IBD) is a chronic inflammatory disease with a high prevalence and canceration rate. The immune disorder is one of the recognized mechanisms. Acupuncture is widely used to treat patients with IBD. In recent years, an increasing number of studies have proven the effectiveness of acupuncture in the treatment of IBD, and some progress has been made in the mechanism. In this paper, we reviewed the studies related to acupuncture for IBD and focused on the immunomodulatory mechanism. We found that acupuncture could regulate the innate and adaptive immunity of IBD patients in many ways. Acupuncture exerts innate immunomodulatory effects by regulating intestinal epithelial barrier, toll-like receptors, NLRP3 inflammasomes, oxidative stress, and endoplasmic reticulum stress and exerts adaptive immunomodulation by regulating the balance of Th17/Treg and Th1/Th2 cells. Numerous control strategies of temperature regulation have been carried out for proton-exchange membrane fuel cell systems including a cooling fan in order to ensure operation at the desired condition and extend the lifetime of the fuel cell stack. However, most existing control strategies are developed without considering the efficiency limitation of the cooling system such that the cooling fan may be unable to eliminate the additional heat. Moreover, there are unknown modelling errors, external disturbance and noise during modelling and experiment processes for fuel cells. Due to those unknown dynamics, the conventional control strategies may fail to achieve the expectant results. To address this issue, an alternative control strategy is proposed in this paper, which consists of a composite proportional-integral (PI) controller with an unknown system dynamics estimator. First, the control strategy is developed by reducing the temperature of input air through the humidifier and simultaneously increasing the mass flow of air in order to eliminate the excess heat that a cooling fan cannot remove. Moreover, an unknown system dynamics estimator is proposed in order to compensate the effect of the unknown dynamics. The construction of the estimator is designed through finding an invariant manifold which implies the relation between known variables and the unknown manifold. The invariant manifold is derived by applying a simple low-pass filter to the system which is beneficial to avoid the requirement of the unmeasurable state derivative. Furthermore, the proposed estimator is easily merged into the proposed PI control strategy and ensures the exponential convergence of estimated errors. Besides, the estimator is further modified such that the derivative of the desired temperature is not required in the controller. The purpose of the paper is to derive a formula to describe the quantitative relationship among the number of the pacing cells required (NPR), the dimension i , and the diffusion coefficient D (electrical coupling or gap junction G). The relationship between NPR and G has been investigated in different dimensions, respectively. That is, for each fixed i , there is a formula to describe the relationship between NPR and G ; and three formulas are required for the three dimensions. However, there is not a universal expression to describe the relationship among NPR, G , and i together. In the manuscript, surveying and investigating the basic law among the existed data, we speculate the preliminary formula of the relationship among the NPR, i , and G ; and then, employing the cftool in MATLAB, the explicit formulas are derived for different cases. In addition, the goodness of fit ( R 2 ) is computed to evaluate the fitting of the formulas. Moreover, the 1D and 2D ventricular tissue models containing biological pacemakers are developed to derive more data to validate the formula. The results suggest that the relationship among the NPR, i , and the G (D) could be described by a universal formula, where the NPR scales with the i (the dimension) power of the product of the square root of G (D) and a constant b which is dependent on the strength of the pacing cells and so on. Purpose. The purpose of this paper is investigating the effect and mechanism of Shenfu injection (a Traditional Chinese Medicine injection form) on prevention and treatment of paclitaxel chemotherapy in peripheral nerve injury. Methods. Wistar rat dorsal root ganglion cells were cultured in vitro and divided into groups of MOCK, PT, PT + LD, and PT + HD. Each group was cultured at a total serum concentration of 10%, including 10% blank serum in the MOCK group, 0.73 (IC30) μmol/L paclitaxel + 10% blank serum in the PT group, and 10% and 5% drug-containing serum and equal amount of paclitaxel were added into the high- and low-dosage groups, respectively. After culturing for 24 hours, the following tests were performed: (1) cell proliferation detected by using CCK-8 and a microplate reader; (2) axon length detected by cellular immunostaining and detection analysis on antibody β-tubulin III; and (3) changes in mitochondrial membrane potential by analyzing immunofluorescence staining with JC-1 probe. Results. (1) Cell proliferation: OD values of the MOCK group and PT group were 0.43 ± 0.02 and 0.25 ± 0.03, respectively (P < 0.05), while OD values of groups PT + LD and PT + HD were 0.41 ± 0.05 and 0.46 ± 0.03, respectively, higher than group PT (P < 0.05). (2) Axon length: (171.19 ± 9.2) μm and (59.2 ± 6.1) μm (P < 0.05) in the MOCK group and PT group, respectively; and (124.2 ± 18.3) um and (154.5 ± 22.9) um in the PT + LD and PT + HD groups, respectively, higher than the PT group (P < 0.05). (3) Changes in mitochondrial membrane potential: the MOCK group showed red fluorescence. Compared with the MOCK group, the green fluorescence of the PT group was enhanced and the red fluorescence was weakened. The red fluorescence of the PT + LD and PT + HD groups was partially enhanced compared with the PT group. Conclusion. Summary: Droplet behaviors play a major role in water management of proton exchange membrane fuel cells. Contact angle, as one of the critical parameters in the boundary conditions for the droplet dynamics, can greatly affect the simulation results for droplet deformation and evolvement. Recently, the dynamic contact angle (DCA) model implemented with Hoffman function has been successfully validated in the simulation of droplet impact on surfaces. In this paper, the Hoffman function is further applied to simulate liquid water slug flow in a straight microchannel with the volume of fluid (VOF) method. It is found that the numerical results are difficult to well match the corresponding experimental results under the same reported experimental conditions. However, the numerical results with lower gas inlet velocity can significantly improve the comparison. It is indicated that the DCA model coupled with Hoffman function has limitations in the simulation of liquid water behaviors with surrounding flows and needs to be further developed. In addition, a series of numerical simulations are conducted with different air inlet velocities, surface tensions, and viscosities to investigate the effects of these factors on the droplet behaviors. Adducin (ADD) is a family of membrane skeleton proteins including ADD1, ADD2, and ADD3 that are encoded by distinct genes on different chromosomes. Adducin is primarily responsible for the assembly of spectrin-actin network that provides physical support to the plasma membrane and mediates signal transduction in various cellular physiological processes upon regulation by protein kinase C-dependent and calcium/calmodulin-dependent pathways. Abnormal phosphorylation, genetic variations, and alternative splicing of adducin may contribute to alterations in cellular functions involved in pathogenic processes. These alterations are associated with a wide range of diseases including cancer. This paper begins with a discussion on how adducin partakes in the structural formation of membrane skeleton, its regulation, and related functional characteristics, followed by a review on the pathogenesis of hypertension, biliary atresia, and cancer with respect to increased disease susceptibility mediated by adducin polymorphism and/or dysregulation. Given the functional diversity of adducin in different cellular compartments, we aim to provide a knowledge base whereby its pathophysiological roles can be better understood. Mercury is an environmental poison, and the damage to living system is generally severe. The severity of mercury poisoning is consequent from the fact that it targets the thiol-containing enzymes, irreversibly oxidizing their critical thiol groups, consequently leading to an inactivation of the enzyme. The Na+/K+-ATPase is a sulfhydryl protein that is sensitive to Hg2+ assault. On the other hand, organoseleniums are a class of pharmacologically promising compounds with potent antioxidant effects. While Hg2+ oxidizes sulfhydryl groups of Na+/K+-ATPase under in vitro and in vivo conditions, the organoselenium compounds inhibit Na+/K+- ATPase in vitro but enhance its activities under in vivo conditions with concomitant increase in the level of endogenous thiols. Paradoxically, it appears that these two thiol oxidants can be used to counteract one another under in vivo conditions, and this hypothesis serves as the basis for this paper. [ABSTRACT FROM AUTHOR]

Cell-like recognizing membrane systems are computational devices in the framework of membrane computing inspired from the structure of living cells, where biological membranes are arranged hierarchically. In this paper tissue-like recognizing membrane systems are presented. The idea is to consider that membranes are placed in the nodes of a graph, mimicking the cell intercommunication in tissues. In this context, polynomial complexity classes associated with recognizing membrane systems can be defined. We recall the definition for cell-like systems, and we introduce the corresponding complexity classes for the tissue-like case. Inflammatory and antimicrobial diseases constitute a major burden for society, and fighting them is a WHO strategic priority. Most of the treatments available to fight inflammatory diseases are anti-inflammatory drugs, such as corticosteroids or immunomodulators that lack cellular specificity and lead to numerous side effects. In addition to suppressing undesired inflammation and reducing disease progression, these drugs lessen the immune system protective functions. Furthermore, treating infectious diseases is more and more challenging due to the rise of microbial resistance to antimicrobial drugs. Thus, controlling the inflammatory process locally without compromising the ability to combat infections is an essential feature in the treatment of inflammatory diseases. We isolated three forms (DRS-DA2N, DRS-DA2NE, and DRS-DA2NEQ) of the same peptide, DRS-DA2, which belongs to the dermaseptin family, from the Mexican tree frog Pachymedusa dacnicolor. Interestingly, DRS-DA2N and DRS-DA2NEQ exhibit a dual activity by inducing the death of leukocytes as well as that of Gram-negative and Gram-positive bacteria, including multiresistant strains, without affecting other cells such as epithelial cells or erythrocytes. We showed that the death of both immune cells and bacteria is induced rapidly by DRS-DA2 and that the membrane is permeabilized, leading to the loss of membrane integrity. We also validated the capacity of DRS-DA2 to regulate the pool of inflammatory cells in vivo in a mouse model of noninfectious peritonitis. After the induction of peritonitis, a local injection of DRS-DA2N could decrease the number of inflammatory cells locally in the peritoneal cavity without inducing a systemic effect, as no changes in the number of inflammatory cells could be detected in blood or in the bone marrow. Collectively, these data suggest that this peptide could be a promising tool in the treatment of inflammatory diseases, such as inflammatory skin diseases, as it could reduce the number of inflammatory cells locally without suppressing the ability to combat infections. The author reflects on the behavior of amyloid beta-protein on nueronal membranes in patients with Alzheimer's disease (AD). He stresses the emerging evidences on the aggregation and toxic potentials of amyloidogenic proteins through its interaction with neuronal and/or glial membranes. This paper focuses on the role that mitogen-activated protein kinases (MAPKs) play in drug-induced kidney injury. The MAPKs, of which there are four major classes (ERK, p38, JNK, and ERK5/BMK), are signalling cascades which have been found to be broadly conserved across a wide variety of organisms. MAPKs allow effective transmission of information from the cell surface to the cytosolic or nuclear compartments. Cross talk between the MAPKs themselves and with other signalling pathways allows the cell to modulate responses to a wide variety of external stimuli. The MAPKs have been shown to play key roles in both mediating and ameliorating cellular responses to stress including xenobiotic-induced toxicity. Therefore, this paper will discuss the specific role of the MAPKs in the kidney in response to injury by a variety of xenobiotics and the potential for therapeutic intervention at the level of MAPK signalling across different types of kidney disease. HIV-1 particle assembly is driven by the structural protein Gag. Gag binds to and multimerizes on the inner leaflet of the plasma membrane, eventually resulting in formation of spherical particles. During virus spread among T cells, Gag accumulates to the plasma membrane domain that, together with target cell membrane, forms a cell junction known as the virological synapse. While Gag association with plasma membrane microdomains has been implicated in virus assembly and cell-to-cell transmission, recent studies suggest that, rather than merely accumulating to pre-existing microdomains, Gag plays an active role in reorganizing the microdomains via its multimerization activity. In this paper, we will discuss this emerging view of Gag microdomain interactions. Together with the Gag protein, the Env glycoprotein is a major retroviral structural protein and is essential for forming infectious virus particles. Env is synthesized, processed, and transported to certain microdomains at the plasma membrane and takes advantage of the same host machinery for its trafficking as that used by cellular glycoproteins. Incorporation of Env into progeny virions is probably mediated by the interaction between Env and Gag, in some cases with the additional involvement of certain host factors. Although several general models have been proposed to explain the incorporation of retroviral Env glycoproteins into virions, the actual mechanism for this process is still unclear, partly because structural data on the Env protein cytoplasmic tail is lacking. Tetherin/BST-2 is an important host restriction factor that limits the replication of HIV and other enveloped viruses. Tetherin is a type II membrane glycoprotein with a very unusual domain structure that allows it to engage budding virions and retain them on the plasma membrane of infected cells. Following the initial report identifying tetherin as the host cell factor targeted by the HIV-1 Vpu gene, knowledge of the molecular, structural, and cellular biology of tetherin has rapidly advanced. This paper summarizes the discovery and impact of tetherin biology on the HIV field, with a focus on recent advances in understanding its structure and function. The relevance of tetherin to replication and spread of other retroviruses is also reviewed. Tetherin is a unique host restriction factor that is likely to continue to provide new insights into host-virus interactions and illustrates well the varied ways by which host organisms defend against viral pathogens. The role of membrane aquaporin-1 (APQ-1) in the photobio modulation (PBM) on erythrocyte deformability will be studied in this paper with human dehydrated erythrocytes as echinocytic shape alterations lead to decreased cellular deformability. Human dehydrated erythrocytes were irradiated with low intensity He-Ne laser irradiation (LHNL) at 0.9, 1.8, 2.7, and 4.4mW/cm² for 5, 15, and 30min, respectively, and APQ-1 inhibitor, 0.2 µmol/L HgCl2, was used to study the role of APQ-1 in mediating PBM with LHNL at 4.4mW/cm² for 5min. Comprehensive morphological parameters of an intact cell such as contact area, perimeter, roundness and erythrocyte elongation index (EEI) were measured to characterize erythrocyte deformability with fast micro multi-channel spectrophotometer. It was observed that the dosage of LHNL improvement of themorphological parameters of dehydrated erythrocytes was morphological-parameter-dependent, but the Bunsen-Roscoe rule did not hold for roundness. The LHNL at 4.4mW/cm² for 5min significantly improved the contact area (P< 0.05) and EEI (P< 0.05) of the dehydrated erythrocytes, but the improvement was significantly inhibited by 0.2 µmol/L HgCl2 (P< 0.05). It was concluded that AQP-1 might mediate the effects of LHNL on erythrocyte deformability, which supports the membranotropic mechanism of PBM. [ABSTRACT FROM AUTHOR]

The amino sugar N-acetylglucosamine (GlcNAc) is well known for the important structural roles that it plays at the cell surface. It is a key component of bacterial cell wall peptidoglycan, fungal cell wall chitin, and the extracellular matrix of animal cells. Interestingly, recent studies have also identified new roles for GlcNAc in cell signaling. For example, GlcNAc stimulates the human fungal pathogen Candida albicans to undergo changes in morphogenesis and expression of virulence genes. Pathogenic E. coli responds to GlcNAc by altering the expression of fimbriae and CURLI fibers that promote biofilm formation and GlcNAc stimulates soil bacteria to undergo changes in morphogenesis and production of antibiotics. Studies with animal cells have revealed that GlcNAc influences cell signaling through the posttranslational modification of proteins by glycosylation. O-linked attachment of GlcNAc to Ser and Thr residues regulates a variety of intracellular proteins, including transcription factors such as NF?B, c-myc, and p53. In addition, the specificity of Notch family receptors for different ligands is altered by GlcNAc attachment to fucose residues in the extracellular domain. GlcNAc also impacts signal transduction by altering the degree of branching of N-linked glycans, which influences cell surface signaling proteins. During cortical development, proliferating neural progenitors exhibit polarized apical and basolateral membranes that are maintained by tightly controlled and membrane-specific vesicular trafficking pathways. Disruption of polarity through impaired delivery of proteins can alter cell fate decisions and consequent expansion of the progenitor pool, as well as impact the integrity of the neuroependymal lining. Loss of neuroependymal integrity disrupts radial glial scaffolding and alters initial neuronal migration from the ventricular zone. Vesicle trafficking is also required for maintenance of lipid and protein cycling within the leading and trailing edge of migratory neurons, as well as dendrites and synapses of mature neurons. Defects in this transport machinery disrupt neuronal identity, migration, and connectivity and give rise to a malformation of cortical development termed as periventricular heterotopia (PH). PH is characterized by a reduction in brain size, ectopic clusters of neurons localized along the lateral ventricle, and epilepsy and dyslexia. These anatomical anomalies correlate with developmental impairments in neural progenitor proliferation and specification, migration from loss of neuroependymal integrity and neuronal motility, and aberrant neuronal process extension. Genes causal for PH regulate vesicle-mediated endocytosis along an actin cytoskeletal network. This paper explores the role of these dynamic processes in cortical development and disease. Transporters are important mediators of specific cellular uptake and thus, not only for effects, but also for side effects, metabolism, and excretion of many drugs such as cisplatin. Cisplatin is a potent cytostatic drug, whose use is limited by its severe acute and chronic nephro-, oto-, and peripheral neurotoxicity. For this reason, other platinum derivatives, such as carboplatin and oxaliplatin, with less toxicity but still with antitumoral action have been developed. Several transporters, which are expressed on the cell membranes, have been associated with cisplatin transport across the plasma membrane and across the cell: the copper transporter 1 (Ctr1), the copper transporter 2 (Ctr2), the P-type copper-transporting ATPases ATP7A and ATP7B, the organic cation transporter 2 (OCT2), and the multidrug extrusion transporter 1 (MATE1). Some of these transporters are also able to accept other platinum derivatives as substrate. Since membrane transporters display a specific tissue distribution, they can be important molecules that mediate the entry of platinum derivatives in target and also nontarget cells possibly mediating specific effects and side effects of the chemotherapeutic drug. This paper summarizes the literature on toxicities of cisplatin compared to that of carboplatin and oxaliplatin and the interaction of these platinum derivatives with membrane transporters. A dual-mode circular polarization selective surface (CPSS) cell based on a modification of the Pierrot cell is presented for single-layer dual-band circularly polarized (CP) reflectarray antenna design. To achieve dual-mode CP discrimination performance, the improved three-dimensional geometry and the high-order resonances of the CPSS cell are utilized. The required phase shifts in both modes are obtained by varying the cell rotation angle. A single-layer dual-band reflectarray operating with orthogonal CPs has been fabricated and tested. Measured results show the peak gains of 25.2 dBi at 11.6 GHz with 31% 3 dB axial ratio (AR) bandwidth and 26.5 dBi at 14.6 GHz with 32% 3 dB AR bandwidth, respectively. In the last twenty years, the general view of the plasma membrane has changed from a homogeneous arrangement of lipids to a mosaic of microdomains. It is currently thought that islands of highly ordered saturated lipids and cholesterol, which are laterally mobile, exist in the plane of the plasma membrane. Lipid rafts are thought to provide a means to explain the spatial segregation of certain signalling pathways emanating from the cell surface. They seem to provide the necessary microenvironment in order for certain specialised signalling events to take place, such as the innate immune recognition. The innate immune system seems to employ germ-lined encoded receptors, called pattern recognition receptors (PRRs), in order to detect pathogens. One family of such receptors are the Toll-like receptors (TLRs), which are the central "sensing" apparatus of the innate immune system. In recent years, it has become apparent that TLRs are recruited into membrane microdomains in response to ligands. These nanoscale assemblies of sphingolipid, cholesterol, and TLRs stabilize and coalesce, forming signalling platforms, which transduce signals that lead to innate immune activation. In the current paper, we will investigate all past and current literature concerning recruitment of extracellular and intracellular TLRs into lipid rafts and how this membrane organization modulates innate immune responses. It is widely accepted that the conversion of the soluble, nontoxic amyloid β-protein (Aβ) monomer to aggregated toxic Aβ rich in β-sheet structures is central to the development of Alzheimer's disease. However, the mechanism of the abnormal aggregation of Aβ in vivo is not well understood. Accumulating evidence suggests that lipid rafts (microdomains) in membranes mainly composed of sphingolipids (gangliosides and sphingomyelin) and cholesterol play a pivotal role in this process. This paper summarizes the molecular mechanisms by which Aβ aggregates on membranes containing ganglioside clusters, forming amyloid fibrils. Notably, the toxicity and physicochemical properties of the fibrils are different from those of Aβ amyloids formed in solution. Furthermore, differences between Aβ-(1-40) and Aβ-(1-42) in membrane interaction and amyloidogenesis are also emphasized. [ABSTRACT FROM AUTHOR]

The interaction and survival of pathogens in hostile environments and in confrontation with host immune responses are important mechanisms for the establishment of infection. Ectophosphatases are enzymes localized at the plasma membrane of cells, and their active sites face the external medium rather than the cytoplasm. Once activated, these enzymes are able to hydrolyze phosphorylated substrates in the extracellular milieu. Several studies demonstrated the presence of surface-located ectophosphatases in a vast number of pathogenic organisms, including bacteria, protozoa, and fungi. Little is known about the role of ecto-phosphatases in host-pathogen interactions. Caveolin-2 is one of the major protein components of cholesterol- and glycosphingolipid-rich flask-shaped invaginations of plasma membrane caveolae. A new body of evidence suggests that caveolin-2 plays an important, and often more direct, role than caveolin- 1 in regulating signaling and function in a cell- and tissue type-specific manner. Cigarette smoking bears a strong etiological association with many neovascularization-related diseases, including cancer, cardiovascular disease, and age-related macular degeneration. Cigarette smoke is a complex mixture of many compounds, including nicotine, which is the major active and addictive component of tobacco. Nicotine and its specific metabolized carcinogens directly bind to nicotinic acetylcholine receptors (nAChRs) on cell membranes and trigger the nAChR signal cascade. The nAChRs were originally thought to be ligand-gated ion channels that modulate physiological processes ranging from neurotransmission to cancer signaling. For several decades, the nAChRs served as a prototypic molecule for neurotransmitter receptors; however, they are now important therapeutic targets for various diseases, including Alzheimer's and Parkinson's diseases, schizophrenia, and even cancer. The investigations of the interaction between phospholipid bilayer and short-chain alcohols are relevant for the potential of lipid bilayer membranes to serve as model systems for studies of various biological processes including permeability of the plasma membrane and molecular mechanisms of anesthesia. Because the hydrophobic portion of an alcohol favorably interacts with lipid hydrocarbon chains, the polar hydroxyl group remains free to form hydrogen bonds with polar lipid atoms that are located near the water/lipid interface. Experiments on phospholipid membranes have shown that alcohols can induce an interdigitated phase and at high concentration even promote the assembly of some lipids into non-bilayer structures within the membrane interior. In this paper we have investigated the DPPC:hexanol system at high alcohol concentration (two molecules per phospholipid) by means of calorimetric, Nuclear Magnetic Resonance, X-ray diffraction and density measurements. We have found that the presence of a high alcohol concentration shifts the membrane transition temperature to lower values, and has a disordering effect on the phospholipid acyl chains in the gel phase. Freeze-fracture electron microscopy enabled us to observe the molecular architecture of the biological membranes. We were studying the myofiber plasma membranes of health and disease by using this technique and were interested in the special assembly called orthogonal arrays (OAs). OAs were present in normal myofiber plasma membranes and were especially numerous in fast twitch type 2 myofibers; while OAs were lost from sarcolemmal plasma membranes of severely affected muscles with dystrophinopathy and dysferlinopathy but not with caveolinopathy. In the mid nineties of the last century, the OAs turned out to be a water channel named aquaporin 4 (AQP4). Since this discovery, several groups of investigators have been studying AQP4 expression in diseased muscles. This review summarizes the papers which describe the expression of OAs, AQP4, and other AQPs at the sarcolemma of healthy and diseased muscle and discusses the possible role of AQPs, especially that of AQP4, in normal and pathological skeletal muscles. One of the major instigators leading to neuronal cell death and brain damage following cerebral ischemia is calcium dysregulation. The neuron's inability to maintain calcium homeostasis is believed to be a result of increased calcium influx and impaired calcium extrusion across the plasma membrane. The need to better understand the cellular and biochemical mechanisms of calcium dysregulation contributing to neuronal loss following stroke/cerebral ischemia is essential for the development of new treatments in order to reduce ischemic brain injury. Expression of sphingosine kinase-1 (SphK-1) correlates with a poor survival rate of tumor patients. This effect is probably due to the ability of SphK-1 to be released into the extracellular medium where it catalyzes the biosynthesis of sphingosine-1-phosphate (S1P), a signaling molecule endowed with profound proangiogenic effects. SphK-1 is a leaderless protein which is secreted by an unconventional mechanism. In this paper, we will show that in human hepatocarcinoma Sk-Hep1 cells, extracellular signaling is followed by targeting the enzyme to the cell surface and parallels targeting of FGF-2 to the budding vesicles. We will also show that SphK-1 is present in a catalitycally active form in vesicles shed by SK-Hep1 and human breast carcinoma 8701-BC cells. The enzyme substrate sphingosine is present in shed vesicles where it is produced by neutral ceramidase. Human beings have been exposed to airborne nanosized particles throughout their evolutionary stages, and such exposures have increased dramatically over the last century. The rapidly developing field of nanotechnology will result in new sources of this exposure, through inhalation, ingestion, and injection. Although nanomaterials are currently being widely used in modern technology, there is a serious lack of information concerning the human health and environmental implications of manufactured nanomaterials. Since these are relatively new particles, it is necessary to investigate their toxicological behavior. The objective of this review was to trace the cellular response to nanosized particle exposure. Therapeutic application of selected nanoparticles together with their range of toxic doses was also reviewed. Effect of therapeutically used nanoparticles on cell membrane, mitochondrial function, prooxidant/antioxidant status, enzyme leakage, DNA, and other biochemical endpoints was elucidated. Flavonoids can exert beneficial health effects through multiple mechanisms. In this paper, we address the important, although not fully understood, capacity of flavonoids to interact with cell membranes. The interactions of polyphenols with bilayers include: (a) the partition of the more non-polar compounds in the hydrophobic interior of the membrane, and (b) the formation of hydrogen bonds between the polar head groups of lipids and the more hydrophilic flavonoids at the membrane interface. The consequences of these interactions are discussed. The induction of changes in membrane physical properties can affect the rates of membrane lipid and protein oxidation. The partition of certain flavonoids in the hydrophobic core can result in a chain breaking antioxidant activity. We suggest that interactions of polyphenols at the surface of bilayers through hydrogen bonding, can act to reduce the access of deleterious molecules (i.e. oxidants), thus protecting the structure and function of membranes. Shewanella putrefaciens (S. putrefaciens) is frequently detected as a serious threat to food safety because it causes spoilage of aquatic products during cold chain transportation. Here, we studied phenotypic changes in S. putrefaciens at low temperatures to discuss the specific behavior of its spoiled aquatic products. S. putrefaciens was found to grow considerably through the growth curve at 0–30°C. In order to investigate the strong putrefaction of aquatic significant amounts under cold storage at 0–10°C, SEM was used to examine the microstructure of S. putrefaciens that had been cultivated at various temperatures. At low temperatures, it was discovered that the cell membrane surface of S. putrefaciens was smoother. FITC-labeled fluorescence microscope was used to view the results. Analysis of S. putrefaciens cultivated at various temperatures revealed that the integrity and permeability of the cell membrane deteriorated with the drop of culture temperature; however, the downward trend slowed down when the temperature was below 10°C. In addition, as the temperature continued to drop below 10°C, the capacity of S. putrefaciens to build biofilms did not diminish noticeably. The findings demonstrate that S. putrefaciens's cold adaptation mechanism at low temperatures involves modifications to the morphological and phenotypic characteristics of cell membranes, plays a significant role in the organism's ability to corrode aquatic products at low temperatures, and provides an important target for prevention and control of S. putrefaciens' ability to contaminate aquatic products and cause financial losses. Biological and engineering studies of Hess-Murray’s law are focused on assemblies of tubes with impermeable walls. Blood vessels and airways have permeable walls to allow the exchange of fluid and other dissolved substances with tissues. Should Hess-Murray’s law hold for bifurcating systems in which the walls of the vessels are permeable to fluid? This paper investigates the fluid flow in a porous-walled T-shaped assembly of vessels. Fluid flow in this branching flow structure is studied numerically to predict the configuration that provides greater access to the flow. Our findings indicate, among other results, that an asymmetric flow (i.e., breaking the symmetry of the flow distribution) may occur in this symmetrical dichotomous system. To derive expressions for the optimum branching sizes, the hydraulic resistance of the branched system is computed. Here we show the T-shaped assembly of vessels is only conforming to Hess-Murray’s law optimum as long as they have impervious walls. Findings also indicate that the optimum relationship between the sizes of parent and daughter tubes depends on the wall permeability of the assembled tubes. Recently, the use of nanometer liposomes as nanocarriers in drug delivery systems mediated by nanoelectroporation has been proposed. This technique takes advantage of the possibility of simultaneously electroporating liposomes and cell membrane with 10-nanosecond pulsed electric fields (nsPEF) facilitating the release of the drug from the liposomes and at the same time its uptake by the cells. In this paper the design and characterization of a 10 nsPEF exposure system is presented, for liposomes electroporation purposes. The design and the characterization of the applicator have been carried out choosing an electroporation cuvette with 1 mm gap between the electrodes. The structure efficiency has been evaluated at different experimental conditions by changing the solution conductivity from 0.25 to 1.6 S/m. With the aim to analyze the influence of device performances on the liposomes electroporation, microdosimetric simulations have been performed considering liposomes of 200 and 400 nm of dimension with different inner and outer conductivity (from 0.05 to 1.6 S/m) in order to identify the voltage needed for their poration. During the recent years, multiple studies demonstrated that C-peptide is not an inert peptide, but exerts important physiological effects. C-peptide binds to cell membranes, stimulates the Na, K-ATPase and the endothelial nitric oxide (NO) synthase. Moreover, there is evidence that C-peptide decreases glomerular hyperfiltration and increases glucose utilisation. Nevertheless, there is still limited knowledge concerning mechanisms leading to an increased glucose utilisation either in rats or in humans. Summary: Although hydrocarbon‐based proton exchange membranes have gained popularity, their durability needs further evaluation. In this study, a dynamic model is developed for a membrane electrode assembly (MEA) working with sulfonated poly (ether ether ketone) (SPEEK) membrane to predict the concentration profiles of reactants inside the MEA, their cross‐over in the membrane, and hydrogen peroxide formation via chemical and electrochemical modes as a function of voltage and membrane thickness. Results show that reactants cross‐over, especially oxygen is accelerated under high voltages, promoting the chemical mode of H2O2 formation, whereas the voltage driving force and oxygen concentration simultaneously control the electrochemical mode. Moreover, a 50% reduction in SPEEK membrane thickness yields about 90%, 200%, and up to 50% rise in reactant cross‐over, chemical, and electrochemical H2O2 formation modes, respectively. The figures for the chemical mode of H2O2 formation are five orders of magnitude greater than their electrochemical counterparts. This numerical study shows that the most attention should be paid to the anode/membrane interface to mitigate the chemical degradation of hydrocarbon‐based proton exchange membranes. Quantitative analysis of the dynamic behavior about membrane-bound secretory vesicles has proven to be important in biological research. This paper proposes a novel approach to automatically identify the elusive fusion events between VAMP2-pHluorin labeled GLUT4 storage vesicles (GSVs) and the plasma membrane. The differentiation is implemented to detect the initiation of fusion events by modified forward subtraction of consecutive frames in the TIRFM image sequence. Spatially connected pixels in difference images brighter than a specified adaptive threshold are grouped into a distinct fusion spot. The vesicles are located at the intensity-weighted centroid of their fusion spots. To reveal the true in vivo nature of a fusion event, 2D Gaussian fitting for the fusion spot is used to derive the intensity-weighted centroid and the spot size during the fusion process. The fusion event and its termination can be determined according to the change of spot size. The method is evaluated on real experiment data with ground truth annotated by expert cell biologists. Sphingolipids are important bioactive lipids that not only play an important role in maintaining the barrier function and fluidity of cell membranes but also regulate multiple processes in cancer development by controlling multiple signaling pathways in the signal transduction network. Background. The bacterial membrane plays a critical role in the survival of bacteria and the effectiveness of antimicrobial peptides in protecting the host. The lipid constituents of the bacterial membrane are not evenly distributed, and they could be affected by clustering anionic lipids with cationic peptides with multiple positive charges. That could be harmful to bacteria because it prevents lipids from interacting with other molecular components of the cell membrane, disrupts existing natural domains, or creates phase boundary defects between the clustered lipids and the bulk of the membrane. This preliminary quantitative study is aimed at assembling a correlation between antibiotic resistance and bacterial lipid composition in E. coli, based on the function and arrangement of the bilipid coating of the bacterial cell, intimately associated with the path of antimicrobials through membranes. Methods. Fifteen multiresistant E. coli samples are collected from swine with enterocolitis tested for resistance levels using the disc diffusimetric method (Kirby-Bauer disc diffusion). Pathogen identification completed using the API 20E multitest system revealed the E. coli presence in 11 samples. In these samples, bacterial membrane detection of fatty acid methyl esters (FAME) operating a 240 MS Ion Trap (Varian) GC/MS (Agilent Technologies, Santa Clara, CA, USA) was performed, using the MIDI Sherlock recognition software model. Results. Interpreting the descriptive statistical method, the correlation matrix, and regression curves and after ANOVA analysis, we ascertained that the studied E. coli population statistically confirmed different degrees of resistance in most of the samples analyzed in this test. Conclusions. In one case, the methyl-(Z)-11-tetradecenoate acid was observed to have a relationship with the susceptibility evaluation by using the disc diffusimetric method, which has revealed the lowest rate of antimicrobial resistance, so it has importance in further resistance evaluation studies. Currently, there are many effective pharmacological treatments for generalized anxiety disorder (GAD), formulated herbal granule is also an alternative way. Our research intends to construct a pharmacological network on genetic targets and pathways between Jiu Wei Zhen Xin Formula (JWZXF) and GAD. Through the TCMSP database, we collected the active ingredients of JWZXF and potential targets of the active ingredients. The GAD-related proteins collected from GeneCards database and DisGeNET database were combined. Component-target protein networks were constructed and visualized using Cytoscape 3.8.2 software to comprehensively clarify the relationships between ingredients, components, and targets. The intersection targets were imported into the STRING database, and the protein-protein interaction (PPI) network was constructed. We constructed and analyzed the visualized "drug-target-disease" network. Gene Ontology (GO) enrichment together with Kyoto encyclopedia of genes and genomes (KEGG) enrichment analysis were conducted on the common target through R language. Forty-one effective components and 106 potential targets of JWZXF were found. There were top ten hub genes and multiple important signaling pathways involved in the treatment of GAD with the JWZXF. This study expounded the pharmacological actions and molecular mechanisms of the JWZXF in treating GAD from a holistic perspective. The potential pharmacological effects of the JWZXF are closely related to regulation because not only does it comprehensively analyze the possible mechanism of JWZXF treatment of GAD but it can also facilitate further in-depth research and provide a theoretical basis for the clinical expansion of its application. The diseased cage-cultured cobia (Rachycentron canadum) displayed clinical signs, haemorrhagic eyes, dorsal darkness and gross pathological lesions, enlargement of spleen and liver. Haemorrhages were found in brain, heart and liver with cumulative mortality rates ranging from 20% to 50%. Extensive congestion in the heart, liver, spleen, kidney and brain was observed histopathologically. Epicarditis and meningitis were also revealed in diseased cobia. All isolates recovered from the organs (liver, spleen, head kidney, posterior kidney, brain and muscle) of cobia were found to be gram-positive, non-motile, ovoid cocci, short-chain-forming (diplococci) and a-haemolytic. The API 32 strep system together with the polymerase chain reaction assay for species-specific primers (pLG1 and pLG2) and the internal transcribed spacer (ITS) region (G1 and L1 primers) confirmed all four selected isolates as Lactococcus garvieae. Partial 16S rDNA nucleotide sequence (~1,100 bp) of one representative L. garvieae isolate AOD109191 (GenBank accession number, MW328528.1) shared 99.9% identities with the 16S rDNA nucleotide sequence of L. garvieae (GenBank accession numbers: MT604790.1). Transmission electron microscopy (TEM) evaluation of one representative L. garvieae isolate (AOD109191) and the results of multiplex PCR did not reveal the presence of the capsular gene cluster (CGC), thus categorizing the isolate as the KG+ phenotype. Capsule staining and TEM observations confirmed the presence of a hyaluronic acid-like capsule, a possible virulence factor in KG+ phenotype L. garvieae isolates. The pathogenic potential of the representative isolate (AOD109191) was assessed through intraperitoneal injection challenges in cobia. The gross lesions and histopathological changes found in experimentally infected cobia were similar to those seen in naturally infected fish. The paper investigates quasisynchronization in a communication system, which consists of cells communicating through quorum sensing. With the help of Lyapunov function method and Lur'e system approach, some sufficient conditions for quasisynchronization are presented, and a bound on the synchronization errors is derived. The obtained theoretical results show that the synchronization quality is influenced by two parameters detrimentally: the error bound depends almost linearly on the mismatches between cells and depends sensitively on the diffusion rates of the signals inward the cell membrane. Summary: This study describes the fundamentals of hydrogen‐separation membranes and H2 separation modeling through Pd‐based membranes. Furthermore, an assessment of hydrogen separation through a Pd‐Cu 40% membrane was performed using two different feed gas concentrations (99.999% H2 and 75% H2−25% N2) and a dielectric barrier discharge (DBD) plasma in a cylindrical type reactor (CTR). The plasma was applied by combining zeolite catalyst materials (SA‐600A and 330‐HUDIA) in two different hydrogen concentration experiments. Scanning electron microscopy indicated that the SA‐600A zeolite exhibited a larger surface area than the 330‐HUDIA zeolite. This may have increased the ability of the former material to diffuse hydrogen molecules. The hydrogen separation results showed that N2 gas has a positive effect on separation through a Pd‐Cu membrane. The most interesting result from the plasma‐catalyst experiment was that the CTR H2 permeation rate reached a maximum of 100% at most input gas flow rates. The maximum plasma‐catalyst energy efficiencies at feed gas concentrations of 99.999% and 75% H2‐25% N2 were 81% and 54%, respectively. Propionibacterium acnes (P. acnes) is a major pathogen of acne vulgaris. The traditional Chinese medicine (TCM) compound prescription, Dian Dao San (DDS), is effective for treating P. acnes. Previous clinical work by our team demonstrated that improved Dian Dao San (IDDS) has better antibacterial effects. However, the mechanism of IDDS inhibition of P. acnes is still unknown. Hence, the isobaric tags for relative and absolute quantitation (iTRAQ) technology was applied to explore the antibacterial mechanism of IDDS against P. acnes. Our results suggested that the antibacterial mechanism of IDDS was related to the glycolytic pathway. gap, pgk, and tpiA enzymes were found to be potential target proteins in the bacterial glycolytic pathway as an antibacterial mechanism of inhibition. In addition, SEM and TEM analyses revealed that IDDS may destruct bacterial plasma membrane and cell wall. Increasing outbreaks of new pathogenic viruses have promoted the exploration of novel alternatives to time-consuming vaccines. Thus, it is necessary to develop a universal approach to halt the spread of new and unknown viruses as they are discovered. One such promising approach is to target lipid membranes, which are common to all viruses and bacteria. The ongoing severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic has reaffirmed the importance of interactions between the virus envelope and the host cell plasma membrane as a critical mechanism of infection. Metadichol®, a nanolipid emulsion of long-chain alcohols, has been demonstrated as a strong candidate that inhibits the proliferation of SARS-CoV-2. Naturally derived substances, such as long-chain saturated lipid alcohols, reduce viral infectivity, including that of coronaviruses (such as SARS-CoV-2) by modifying their lipid-dependent attachment mechanism to human host cells. The receptor ACE2 mediates the entry of SARS-CoV-2 into the host cells, whereas the serine protease TMPRSS2 primes the viral S protein. In this study, Metadichol® was found to be 270 times more potent an inhibitor of TMPRSS2 (E C 50 = 96 ng / mL) than camostat mesylate (E C 50 = 26000 ng / mL). Additionally, it inhibits ACE with an EC50 of 71 ng/mL, but it is a very weak inhibitor of ACE2 at an EC50 of 31 μg/mL. Furthermore, the live viral assay performed in Caco-2 cells revealed that Metadichol® inhibits SARS-CoV-2 replication at an EC90 of 0.16 μg/mL. Moreover, Metadichol® had an EC90 of 0.00037 μM, making it 2081 and 3371 times more potent than remdesivir (E C 50 = 0.77 μ M) and chloroquine (E C 50 = 1.14 μ M), respectively. To date the genus Corynebacterium comprises 88 species. More than half of these are connected to human and animal infections, with the most prominent member of the pathogenic species being Corynebacterium diphtheriae, which is also the type species of the genus. Corynebacterium species are characterized by a complex cell wall architecture: the plasma membrane of these bacteria is followed by a peptidoglycan layer, which itself is covalently linked to a polymer of arabinogalactan. Bound to this, an outer layer of mycolic acids is found which is functionally equivalent to the outer membrane of Gram-negative bacteria. As final layer, free polysaccharides, glycolipids, and proteins are found. To address the challenges associated with differential expression proteomics, label-free mass spectrometric protein quantification methods have been developed as alternatives to array-based, gel-based, and stable isotope tag or label-based approaches. In this paper, we focus on the issues associated with label-free methods that rely on quantitation based on peptide ion peak area measurement. These issues include chromatographic alignment, peptide qualification for quantitation, and normalization. In addressing these issues, we present various approaches, assembled in a recently developed label-free quantitative mass spectrometry platform, that overcome these difficulties and enable comprehensive, accurate, and reproducible protein quantitation in highly complex protein mixtures from experiments with many sample groups. As examples of the utility of this approach, we present a variety of cases where the platform was applied successfully to assess differential protein expression or abundance in body fluids, in vitro nanotoxicology models, tissue proteomics in genetic knock-in mice, and cell membrane proteomics. The usual way to find a solution for the problems with exponential time complexity with Membrane Computing techniques is by P System with active membrane using division rule. It makes an exponential workspace and solves the problems with exponential complexity in a polynomial (even linear) time. On the other hand, searching is currently one of the most used methods for finding solution for problems in real life, that the blind search algorithms are accurate, but their time complexity is exponential such as breadth-first search (BFS) algorithm. In this paper, we proposed a new approach for implementation of BFS by using P system with division rule technique for first time. Gangliosides are integral to the structure and function of cell membranes. Ganglioside composition of the intestinal brush border and apical surface of the colon influences numerous cell processes including microbial attachment, cell division, differentiation, and signaling. Accelerated catabolism of ganglioside in intestinal disease results in increased proinflammatory signaling. Restoring proper structure and function to the diseased intestine can resolve inflammation, increase resistance to infection, and improve gut integrity to induce remission of conditions like necrotizing enterocolitis (NEC) and Crohn's disease (CD). Maintaining inactive state of disease may be achieved by reducing the rate that gangliosides are degraded or by increasing intake of dietary ganglioside. Collectively, the studies outlined in this paper indicate that the amount of gangliosides GM3 and GD3 in intestinal mucosa is decreased with inflammation, low level of GM3 is associated with higher production of proinflammatory signals, and ganglioside content of intestinal mucosa can be increased by dietary ganglioside.
