Abstract

Excavation of a superlarge diameter tunnel by tunnel boring machine (TBM) is different from that of a shield tunnel with normal dimension, in which the control system of the superlarge TBM is very complicated and difficult to operate. Hence, it is very important to focus on the control and management of significant parameters to ensure excavation stability under uncertainty. In this paper, we (i) utilize a BIM-based big data platform (BIM-BDP) to manage the essential construction data of tunnel project in digital format; (ii) adopt the global sensitivity analysis (SA) to recognize significant parameters for shield excavation based on polynomial chaos expansion (PCE)–extended Fourier amplitude sensitivity test (eFAST) model; and (iii) employ the uncertainty analysis (UA) to discover the correlation between significant parameters from the data of the BIM-BDP. This research contributes to (i) the body of knowledge of proposing a more appropriate research methodology that can cope with aleatory and epistemic uncertainty and support uncertainty and sensitivity analysis (UA/SA) processes based on data from BIM-BDP and (ii) the state of practice by providing a data-driven surrogate model to simulate system behaviors of shield excavation with high reliability and to reduce dependency on domain experts. Here, we pay close attention to the most influential parameters that require priority parameter control, which can help administrators optimize the management of shield parameters during tunnel excavation.

Abstract

Coffee beans roasting is an important step in coffee production because it enables the development of flavor, aroma and color. Most coffee beans are commercially roasted in large-scale, but some coffee drinkers prefer to precisely control the freshness and the special flavor by their own roasting method. This study was conducted to develop a novel approach to thermal decomposition that includes the heat reactivity properties of coffee beans. From roasting conditions versus thermal decomposition characteristics, we found the optimal roasting conditions and thermal decomposition characteristics, and discovered the control conditions. The parameters and reactivity properties could be applied to design during roasting and storage. Overall, we applied a nonisothermal kinetic model for the evaluation of kinetic parameters and storage conditions, compared the results to simulated thermal analysis, and then did a sensory evaluation to determine the suitable roasting condition. Practical Applications This approach led to the development of a novel procedure and an appropriate model for estimating the interesting relationship of roasting control process, thermal decomposition properties, storage conditions and sensory evaluation of coffee beans. A novel approach to discovering the heat reactivity properties of coffee beans. Coffee beab flavor with roasting conditions has a very close character of the relationship. Abstract

Lipoprotein lipase (LPL) has been identified as an anti-hepatitis C virus (HCV) host factor, but the cellular mechanism remains elusive. Here, we investigated the cellular mechanism of LPL involving in anti-HCV. The functional activation of peroxisome proliferator-activated receptor (PPAR) α signal by LPL transducing into hepatocytes was investigated in HCV-infected cells, primary human hepatocytes, and in HCV-core transgenic mice. The result showed that the levels of transcriptional transactivity and nuclear translocation of PPARα in Huh7 cells and primary human hepatocytes were elevated by physiologically ranged LPL treatment of either very-low density lipoprotein or HCV particles. The LPL-induced hepatic PPARα activation was weakened by blocking the LPL enzymatic activity, and by preventing the cellular uptake of free unsaturated fatty acids with either albumin chelator or silencing of CD36 translocase. The knockdowns of PPARα and CD36 reversed the LPL-mediated suppression of HCV infection. Furthermore, treatment with LPL, like the direct activation of PPARα, not only reduced the levels of apolipoproteins B, E, and J, which are involved in assembly and release of HCV virions, but also alleviated hepatic lipid accumulation induced by core protein. HCV-core transgenic mice exhibited more hepatic miR-27b, which negatively regulates PPARα expression, than did the wild-type controls. The induction of LPL activity by fasting in the core transgenic mice activated PPARα downstream target genes that are involved in fatty acid β-oxidation. Taken together, our study reveals dual beneficial outcomes of LPL in anti-HCV and anti-steatosis and shed light on the control of chronic hepatitis C in relation to LPL modulators.
