Unfortunately, adverse environments in early life are frequently found in most human populations. Early life stress leads to diverse cognitive impairments, some of them related to learning and memory and executive functions such as working memory (WM). We employ an animal model of early stress using repeated maternal separation (MS) for 4 h a day on 21 consecutive days, pre-weaning. In adulthood, we tested their spatial WM using the Morris water maze. MS subjects showed a marked delay in the acquisition of the task. In addition, we explored brain energy oxidative metabolism and found an increase in cytochrome c oxidase (CCO) activity in the cingulate cortex, anterior thalamus, and supramammillary areas, indicating an intense effort to successfully solve the WM task. However, decreased CCO activity was found in the medial-medial mammillary nucleus in MS animals, which would partially explain the delayed acquisition of the WM task. Further studies are needed to explore the long-term alterations produced by early stress. LAY SUMMARY A stressful environment caused by the separation of baby rats from the mother for several hours a day in the first stages of postnatal life can be devastating to brain cells, making them look for alternative sources of energy, among other changes. These alterations in brain functional networks would lead to cognitive impairments such as the delayed acquisition of new learning and strategies.

Praziquantel (PZQ), a broad spectrum anthelmintic drug, cannot be found in acceptable dosage forms for elderly patients, paediatric patients, and for veterinary use. In fact, very little has been done up to now in the formulation of liquid dosage forms, being they always formulated for parenteral administration. To beat this important challenge, it was accomplished a comprehensive analysis of the influence of two elementary physicochemical aspects, i.e. surface thermodynamic and electrokinetic properties, on the colloidal stability of PZQ nanosuspensions. The hydrophobic character of the drug, intensely determining the flocculation curves, was confirmed by the thermodynamic characterization. The electrophoretic characterization, in combination with the sedimentation and relative absorbance versus time curves, highlighted that the electrical double layer thickness and the surface charge can play an essential role in the stability of the pharmaceutical colloid. Finally, it was demonstrated that controlling the pH values and the incorporation of electrolytes can help in formulating PZQ aqueous nanosuspensions with appropriate stability and redispersibility behaviours for pharmaceutical use.

Introduction : Pharmacotherapy is limited by the inefficient drug targeting of non-healthy cells/tissues. In this pharmacological landscape, liposomes are contributing to the impulse given by Nanotechnology to optimize drug therapy. Areas covered : The analysis of the state-of-the-art in liposomal formulations for drug delivery purposes have underlined that lately published patents (since 2014) are exploring alternative compositions and ways to optimize the stability and drug loading content/release profile. These improvements are complemented by improved long-circulating structures and further liposome functionalizations, which have definitively opened the road for the (co-)delivery of therapeutics to the site of action. Liposomes are also contributing to new drug delivery approaches involving the generation of extracellular vesicles by targeted cells, while opening new ways to combine disease diagnosis and therapy (theranosis). Expert opinion : Patent publications on liposomal formulations have expanded new ways in drug delivery. New lipid compositions and strategies to optimize stability and drug vehiculization capabilities have settle solid pillars in liposome fabrication. Despite, their architecture has been satisfactorily adapted for combining passive and active drug targeting concepts, new inputs of liposomes into the disease arena should answer for: a simple/scalable/cost-effective formulation; a safe/stable/controllable formulation meeting quality control regulations; and, a confirmed therapeutic efficiency in clinical investigations.

Childhood maltreatment and neglect lead to a wide range of mental disorders highlighted by hormone and immune alterations in neglected children. This social-health challenge has led to the creation of early stress models such as maternal separation (MS) in rodents. We performed a MS model (4 h per day, 21 days; n  = 16 MS and n  = 16 control), and then measured three parameters in adult male rat brains, in order to look for long-term effects of early life stress. We used immunocytochemistry to mark glial fibrillary acidic protein (GFAP)-positive cells, which indicates changes in astroglia, and ionized calcium binding adaptor molecule 1 (Iba-1)-positive cells, which inform about reactive microglia. In order to study mRNA levels of some immune mediators, interleukin determination (interleukin-6, IL-6; tumor necrosis factor, TNFα) mRNAs were evaluated by real-time polymerase chain reaction (rt-PCR) in discrete brain regions. Measurements of numbers of GFAP-positive cells, and expression of Iba-1, IL-6 and TNFα mRNAs were performed in prefrontal cortex (PFC): cingulate cortex (CG), prelimbic cortex (PL) and infralimbic cortex (IL), striatal areas (dorsal striatum, STD; and nucleus accumbens, ACC), and dorsal hippocampus (HC: CA1, CA3 and dentate gyrus (DG)). We found that MS produces a dramatic and sustained decrease in the astroglial population in all the areas measured (from -25% in CA1 to -85.7% in ACC), whereas increased numbers of microglia were found, in more restricted regions: STD (72.6%), ACC (31%) and CA3 (33.3%) areas. Regarding mRNA measurements, we found increased IL-6 mRNA expression in HC (104.2%), and after MS.

Introduction: Iron oxide nanoparticles (IO NPs) exhibit remarkable properties, including inherent magnetism, biocompatibility, high surface to volume ratio, and versatility of engineering, making them ideal candidates for a variety of clinical applications., Areas Covered: The review provides an in-depth discussion on recent patents and developments related to IO NPs in Biomedicine from the last 7 years. It covers innovations in the chemical synthesis, surface coating and functionalization, and biomedical applications, including MRI and multimodal imaging, molecular imaging, cell labeling, drug delivery, hyperthermia, hyperphosphatemia, and antibacterial activity. A brief outline of the important properties of IO NPs is also presented., Expert Opinion: The main focus of current research is the development of new approaches to generate high-quality IO NPs with optimal properties in terms of particle geometry, crystal structure, surface functionalities, stability, and magnetization. Among chemical synthesis methods, thermal decomposition and hydrothermal synthetics processes allow fine control of the particle properties. Plenty of coating materials have been successfully used as shells for these NPs to provide colloidal stability, even enabling the formulation of nanotheranostics for simultaneous disease diagnosis and therapy. However, long-term toxicity and pharmacokinetic studies are necessary before magnetic nanosystems can be approved for clinical use.

To address the problem, liposome-based medicines have tried to improve pharmacotherapy., Areas Covered: The review provides an updated revision of the lately published patents covering recent advances in liposome-based drug delivery. They are principally related to the control of drug biodistribution by using stealth, stimuli-sensitive and/or liposomal structures surface modified for ligand-mediated delivery. The contribution further highlights liposome-based theranosis., Expert Opinion: Liposomes have received great attention given their biocompatibility, biodegradability and targetability. From 2007 to present date, patent publications related to their use in drug delivery have shown the move towards more stable structures with optimized drug delivery capabilities, further combining passive and active targeting concepts to gain control of the in vivo fate. However, the introduction of all these liposomal structures in the disease arena is still a challenge. Two key aspects are the difficulty of identifying easy and economic synthesis conditions which can be scaled up in the pharmaceutical industry, and the need for complementary investigations illustrating risks of toxicity/immunogenicity.

Introduction: Recent advances in nanoparticle synthesis engineering have made it possible to combine disease diagnosis and therapy. This progress could help to open the door to 'personalized' medicines., Areas Covered: This review highlights the significant applications of theragnostic nanoparticles in therapy. The basic elements to be included in the formulation of theragnostic nanotools are briefly compiled and explained. Special attention is given to the analysis of current formulation strategies from case studies in the literature published after 2000 for simultaneous selective disease imaging and efficient image-guided drug (gene) delivery. This contribution provides a systematic overview of important features in the formulation of theragnostic nanoparticulate systems. Special insight is given to the introduction of passive and active targeting concepts in the engineering of such multifunctional nanoplatforms to gain control of their biological fate., Expert Opinion: Theragnostic nanotechnologies will optimize the way of delivering therapeutic and imaging molecules to disease sites; as a consequence, combined selective diagnosis and effective pharmacotherapy could be used in unison to combat severe diseases. Nanotoxicity investigations, which illustrate the risks of toxicity/immunogenicity associated with the use of such nanoplatforms, will determine their introduction into the clinic.

Context: Oral administration of insulin is severely limited by very low bioavailability. Biocompatible polymeric nanocarriers have been investigated to overcome this problem. Flow focusing (FF) has revolutionized current engineering of poly(D,L-lactide-co-glycolide) (PLGA) based micromedicines. This technique has never been used to formulate insulin-loaded PLGA microparticles., Objective: Investigation of the benefits rising from the synthesis of insulin-loaded PLGA microplatforms by FF, compared to double emulsion/solvent evaporation method., Materials and Methods: Both synthesis methodologies were compared in terms of geometry, surface physicochemical properties and insulin vehiculization capabilities. The stability of the peptide during the formulation procedure was further analysed., Results: FF permitted the preparation of insulin-loaded microcarriers with better geometry and physicochemical properties for the oral route, along with greater insulin loading capabilities and sustained insulin release kinetics., Discussion and Conclusion: Results have lead to the identification of the best formulation conditions for the engineering of insulin-loaded PLGA microparticles against diabetes.

Background: Despite the very efficient antitumor activity of conventional chemotherapy, generally high doses of anticancer molecules must be administered to obtain the required therapeutic action, simultaneously leading to severe side effects. This is frequently a consequence of the development of multidrug resistance by cancer cells and of the poor pharmacokinetic profile of these agents., Objective: In Order to improve the antitumor effect of tegafur and overcome their important drawbacks, we have investigated its incorporation into a drug nanoplatform based on the biodegradable polymer chitosan., Materials and Methods: Two tegafur loading methods were studied: (i) absorption into the polymeric network (entrapment procedure); and (ii) surface deposition (adsorption procedure) in already formed chitosan nanoparticles., Results: Tegafur entrapment into the polymeric matrix has yielded higher drug loading values and a slower drug release profile, compared to single surface adsorption. The main factores determining the drug loading to chitosan were identified., Discussion and Conclusion: Such polymeric colloid present very interesting properties for efficient tegafur delivery to cancer.

Even though 5-fluorouracil has been demonstrated to display antitumor activity against a wide variety of cancers, it is needed to be administered at high doses to elicit the required therapeutic activity, simultaneously leading to severe side effects. We hypothesized that the efficient delivery of 5-fluorouracil to tumors using a magnetic colloid could reduce the dose required to bring out sufficient therapeutic response. Thus, we have formulated a 5-fluorouracil-loaded magnetic nanomedicine consisting of a magnetic core (iron) and a biocompatible polymeric shell (ethylcellulose), suitable for parenteral administration. These core/shell nanoparticles were synthesized by an emulsion solvent evaporation process. Two drug loading methods were analyzed: the first one based on 5-fluorouracil surface adsorption onto the preformed nanoparticles, and the second method being drug addition prior to the emulsion solvent evaporation process leading to drug entrapment into the polymeric network. 5-Fluorouracil entrapment into the polymeric matrix yielded a higher drug loading and a slower drug release profile as compared with drug adsorption. Finally, as a proof of concept, Prussian blue staining has demonstrated the considerable accumulation of these magnetically guided composite nanoparticles in the tumors, suggesting the potential of this stimuli-sensitive drug carrier for the efficient treatment of cancer by active targeting.

Gemcitabine hydrochloride is an anticancer nucleoside analogue indicated in clinic for the treatment of various solid tumors. Although this drug has been demonstrated to display anticancer activity against a wide variety of tumors, it is needed to be administered at high doses to elicit the required therapeutic response, simultaneously leading to severe adverse effects. We hypothesized that the efficient delivery of gemcitabine to tumors using a biodegradable carrier system could reduce the dose required to elicit sufficient therapeutic response. Thus, we have developed a nanoparticle formulation of gemcitabine suitable for parenteral administration based on the biodegradable polymer poly(octylcyanoacrylate) (POCA). The nanoparticles were synthesized by anionic polymerization of the corresponding monomer. Two drug loading methods were analyzed: the first one based on gemcitabine surface adsorption onto the preformed nanoparticles, and the second method being gemcitabine addition before the polymerization process leading to drug entrapment in the polymeric network. A detailed investigation of the capabilities of the polymer particles to load this drug is described. Gemcitabine entrapment into the polymer matrix yielded a higher drug loading and a slower drug release profile as compared with drug adsorption procedure. The main factors determining the gemcitabine incorporation to the polymer network were the nanoparticles preparation procedure, the monomer concentration, the surfactant concentration, the pH, and the drug concentration. The release kinetic of gemcitabine was found to be controlled by the pH and the type of drug incorporation. The cytotoxicity studies performed on L1210 tumor cells revealed a similar anticancer activity of the gemcitabine-loaded POCA (GPOCA) nanoparticle as free gemcitabine. Following intravenous administration into the mice bearing L1210 wt subcutaneous tumor, the GPOCA nanoparticles displayed significantly greater anticancer activity compared to free gemcitabine; this has been additionally confirmed by histology and immunohistochemistry studies, suggesting the potential of GPOCA for the efficient treatment of cancer.

Consolidation processes were studied in the rat by using functional inactivation techniques. Previous results showed that unilateral hippocampal inactivation alters consolidation. It is not clear if bilateral treatments increase the impairment. Wistar rats were trained in the Morris water maze during 4 consecutive days. Subjects received saline or tetrodotoxin in the dorsal hippocampus 1 min after training. Results showed that bilateral as well as unilateral treatments impair consolidation to the same degree, as shown by the mean latency to reach the platform. In both cases, the impairment is only visible in the first trial of the session following the blockade.

The existence of efferent connections between tuberomammillary (TM) and the supraoptic (SON) nuclei led us to study the morphological changes at these levels during dehydration induced in rats by repeated administration of hypertonic saline. The dehydration effects are evaluated by three parameters: nucleus area, argyrophilic nucleolar organizer regions (Ag-NOR) area, and mean number of Ag-NORs. These parameters reflect the level of transcriptional activity of the rRNA genes, which in turn is closely related to the protein synthesis activity of the cell. The technique used is argyrophilic staining which labels the nucleolar organizer regions (NORs) of the nucleoli in the neurons and their area, permitting their quantification with an image analysis system. The statistical analysis reflects the sensitivity to dehydration of these structures at TM level. Our results support the regulatory role of vasopressin secretion by SON through the histaminergic pathways arising from TM in rat dehydration processes.

Type X collagen is a transient and developmentally regulated collagen that has been postulated to be involved in controlling the later stages of endochondral bone formation. However, the role of this collagen in these events is not yet known. In order to understand the function of type X collagen, if any, in the process of biomineralization, the properties of type X collagen in eggshell membranes were further investigated. Specifically, calvaria-derived osteogenic cells were tested for their ability to mineralize eggshell membranes in vitro. Immunohistochemistry with specific monoclonal antibodies was used to correlate the presence or absence of type X collagen or its propeptide domains with the ability of shell membranes to be mineralized. The extent of mineralization was assessed by Von Kossa staining, scanning electron microscopy and energy-dispersive spectroscopy. The results indicate that the non-helical domains of type X collagen must be removed to facilitate the cell-mediated mineralization of eggshell membranes. In this tissue, intact type X collagen does not appear to stimulate or support cell-mediated mineralization. We postulate that the non-helical domains of type X collagen function in vivo to inhibit mineralization and thereby establish boundaries which are protected from mineral deposition.

The avian eggshell is a complex, extracellularly assembled structure which contains both mineralized and non-mineralized regions. The composition of the hen eggshell organic matrix was examined by immunohistochemistry with antibodies to different extracellular matrix molecules. Type I collagen is found in the shell membranes, but only after treatment of the tissue sections with pepsin. When incomplete eggshells are removed from the oviduct and immunostained, type I collagen can be detected in the shell membranes without pepsin treatment. The shell membranes, which are non-mineralized, also contain type X collagen, and this immunostaining does not require pepsin treatment. The occurrence of type X collagen in the shell membranes is surprising, since this collagen has not been found in any tissue other than hypertrophic cartilage. Immunostaining for various glycosaminoglycans shows the presence of keratan sulfate and dermatan sulfate. Several different antibodies to keratan sulfate stain different regions of the eggshell; one keratan sulfate epitope is prominent in the calcium reserve assemblies. Dermatan sulfate staining is very intense in the palisade region. Demineralized matrix from the palisade region was extracted with guanidine and fractionated by ion exchange chromatography. A approximately 200-kDa dermatan sulfate proteoglycan is found in these extracts, along with a number of protein components. This preparation was tested for its ability to affect calcium carbonate crystal formation in vitro. Pieces of demineralized shell membranes were used as a substrate for crystal formation and various amounts of the palisade matrix dermatan sulfate proteoglycan preparation were added to the solution from which the crystals were formed. This material causes a concentration-dependent change in crystal morphology to one in which the crystals are smaller and more rounded, which more closely approximates the crystals normally observed in eggshells. These results suggest that the dermatan sulfate proteoglycans may be important in modulating crystal morphology in the hen eggshell and correlate with mineralization-modulating biomolecules from other calcified tissue, which are generally anionic.

An immunohistochemical analysis of the eggshell membranes shows the occurrence of type X collagen while type I collagen was not detected by using an appropriate monoclonal antibody with untreated shell membranes. A positive immuno-reaction for type I collagen was obtained after digestion of the shell membranes with pepsin. These observations indicate the possibility that type I collagen epitope was masked by type X collagen and that type X collagen may serve as an inhibitory boundary for biomineralization.
