From yeast to humans, pre-mRNA splicing occurs mainly co-transcriptionally, with splicing and transcription functionally coupled such that they influence one another. The recruitment model of co-transcriptional splicing proposes that core members of the transcription elongation machinery have the potential to influence co-transcriptional spliceosome assembly and pre-mRNA splicing. Here, we tested whether the transcription elongation kinases Bur1 and Ctk1 affect co-transcriptional spliceosome assembly and pre-mRNA splicing in the budding yeast Saccharomyces cerevisiae. In S. cerevisiae, Ctk1 is the major kinase that phosphorylates serine 2 of the carboxy-terminal domain of the largest subunit of RNA polymerase II, whilst Bur1 augments the kinase activity of Ctk1 and is the major kinase for elongation factor Spt5. We used the auxin-inducible degron system to conditionally deplete Bur1 and Ctk1 kinases, and investigated the effects on co-transcriptional spliceosome assembly and pre-mRNA splicing. Depletion of Ctk1 effectively reduced phosphorylation of serine 2 of the carboxy-terminal domain but did not impact co-transcriptional spliceosome assembly or pre-mRNA splicing. In striking contrast, depletion of Bur1 did not reduce phosphorylation of serine 2 of the carboxy-terminal domain, but reduced Spt5 phosphorylation and enhanced co-transcriptional spliceosome assembly and pre-mRNA splicing, suggesting a role for this kinase in modulating co-transcriptional splicing.

Rugby league carries a high injury incidence with 61% of injuries occurring at tackles. The ball carrier has a higher injury incidence than the defender, therefore understanding mechanisms occurring during injurious tackles are important. Given the dynamic, open nature of tackling, characteristics influencing tackle outcome likely encompass complex networks of dependencies. This study aims to identify important classifying characteristics of the tackle related to ball carrier injurious and non-injurious events in rugby league and identify the characteristics capability to correctly classify those events. Forty-one ball carrier injuries were identified and 205 matched non-injurious tackles were identified as controls. Each case and control were analysed retrospectively through video analysis. Random forest models were built to (1) filter tackle characteristics possessing relative importance for classifying tackles resulting in injurious/non-injurious outcomes and (2) determine sensitivity and specificity of tackle characteristics to classify injurious and non-injurious events. Six characteristics were identified to possess relative importance to classify injurious tackles. This included 'tackler twisted ball carrier's legs when legs were planted on ground', 'the tackler and ball carrier collide heads', 'the tackler used body weight to tackle ball carrier', 'the tackler has obvious control of the ball carrier' 'the tackler was approaching tackle sub-maximally' and 'tackler's arms were below shoulder level, elbows were flexed'. The study identified tackle characteristics that can be modified in attempt to reduce injury. Additional injury data are needed to establish relationship networks of characteristics and analyse specific injuries. Sensitivity and specificity results of the random forest were 0.995 and 0.525.

This systematic review aimed to identify and summarise associations between currently identified contextual factors and match running in senior male professional rugby league. Eligible articles included at least one contextual factor and used GPS to measure at least one displacement variable within competitive senior, male, professional rugby league matches. Of the 15 included studies, the identified contextual factors were grouped into factors related to individual characteristics (n = 3), match result (n = 4), team strength (n = 2), opposition strength (n = 3), match conditions (n = 6), technical and tactical demands (n = 6), spatial and temporal characteristics (n = 7), and nutrition (n = 1). Speed was the most commonly reported measure of match running (100%), followed by distance (47%), and acceleration (20%). Inconsistencies were found between studies for most contextual factors on match running. Higher speeds were generally associated with higher fitness, encountered earlier in the match and whilst defending. All 15 studies utilised a univariate approach to quantify associations of a contextual factor. The inconsistencies found in the associations of given contextual factors highlight the complex and multi-faceted nature of match running. Therefore, practitioners should consider contextual factors when analysing and interpreting GPS data.

In the era of cost containment of healthcare, the rising incidence of BCC and the high cost of excision require a continuous search for efficient and cost-effective management.Examine the feasibility of the diagnosis and treatment of low-risk sBCC in a single visit.Retrospective chart review of sBCCs diagnosed and treated in a single visit.The study identified 151 histologically confirmed sBCCs in 86 patients over a 5-year period, 93 (61.6%) cases of which were diagnosed as low-risk sBCC and treated in a single appointment. The majority of the cases (Diagnosis and treatment of sBCC in a single visit is an efficient and cost-effective management option for those who are proficient in identifying low-risk sBCC.

Background: Intervention differential effects (IDEs) occur where changes in an outcome depend upon the initial values of that outcome. Although methods to identify IDEs are well documented, there remains a lack of understanding about the circumstances under which these methods are robust. One context that has not been explored is the identification of intervention differential effect in studies where sample selection is based on the initial value of the outcome being evaluated. We hypothesise that, in such settings, established methods for detecting IDEs will struggle to discriminate these from regression to the mean. Methods: Using simulated datasets of weight-loss intervention programmes that recruit according to initial body mass index, we explore the reliability of Oldham's method and multilevel modelling (MLM) to detect IDEs. Results: In datasets simulated with no IDE, Oldham's method and MLM yield Type I error rates >90%, confirming that threshold selection/truncation leads to bias due to regression to the mean. Type I error rates return close to 5% for both methods when a control group is introduced. Conclusions: Oldham's method and MLM can robustly detect IDEs in this setting, but only if analyses incorporate a control group for comparison.

Pre-messenger RNA splicing involves multi-step assembly of the large spliceosome complexes that catalyse the two consecutive trans-esterification reactions, resulting in intron removal. There is evidence that proof-reading mechanisms monitor the fidelity of this complex process. Transcripts that fail these fidelity tests are thought to be directed to degradation pathways, permitting the splicing factors to be recycled. While studying the roles of splicing factors in vivo, in budding yeast, we performed targeted depletion of individual proteins, and analysed the effect on co-transcriptional spliceosome assembly and splicing efficiency. Unexpectedly, depleting factors such as Prp16 or Prp22, that are known to function at the second catalytic step or later in the splicing pathway, resulted in a defect in the first step of splicing, and accumulation of arrested spliceosomes. Through a kinetic analysis of newly synthesized RNA, we observed that a second step splicing defect (the primary defect) was rapidly followed by the first step of splicing defect. Our results show that knocking down a splicing factor can quickly lead to a recycling defect with splicing factors sequestered in stalled complexes, thereby limiting new rounds of splicing. We demonstrate that this ‘feed-back’ effect can be minimized by depleting the target protein more gradually or only partially, allowing a better separation between primary and secondary effects. Our findings indicate that splicing surveillance mechanisms may not always cope with spliceosome assembly defects, and suggest that work involving knock-down of splicing factors or components of other large complexes should be carefully monitored to avoid potentially misleading conclusions.

Purpose:This study investigated sources of variability in the overall and phase-specific running match characteristics in elite rugby league. Methods:Microtechnology data were collected from 11 Super League (SL) teams, across 322 competitive matches within the 2018 and 2019 seasons. Total distance, high-speed running (HSR) distance (>5.5 m·s−1), average speed, and average acceleration were assessed. Variability was determined using linear mixed models, with random intercepts specified for player, position, match, and club. Results:Large within-player coefficients of variation (CV) were found across whole match, ball-in-play, attack and defence for total distance (CV range = 24% to 35%) and HSR distance (37% to 96%), whereas small to moderate CVs (≤10%) were found for average speed and average acceleration. Similarly, there was higher between-player, -position, and -match variability in total distance and HSR distance when compared with average speed and average acceleration across all periods. All metrics were stable between-teams (≤5%), except HSR distance (16% to 18%). The transition period displayed the largest variability of all phases, especially for distance (up to 42%) and HSR distance (up to 165%). Conclusion:Absolute measures of displacement display large within-player and between-player, -position, and -match variability, yet average acceleration and average speed remain relatively stable across all match-periods.

This study aimed to identify which physical and technical-tactical performance indicators (PI) can classify between levels of rugby league match-play. Data were collected from 46 European Super League (ESL) and 36 under-19 Academy (Academy) level matches over two seasons. Thirty-one ESL players and 41 Academy players participated. Microtechnology units were used to analyse the physical PI and matches were videoed and coded for individual technical-tactical PI, resulting in 157 predictor variables. Data were split into training and testing datasets. Random forests (RF) were built to reduce the dimensionality of the data, identify variables of importance and build classification models. To aid practical interpretation, conditional inference (CI) trees were built. Nine variables were identified as most important for backs, classifying between levels with 83% (RF) and 78% (CI tree) accuracy. The combination of variables with the highest classification rate was PlayerLoad2D, PlayerLoadSLOW per Kg body mass and high-speed running distance. Four variables were identified as most important for forwards, classifying with 68% (RF) and 64% (CI tree) accuracy. Defensive play-the-ball losses alone had the highest classification rate for forwards. The identified PI and their unique combinations can be developed during training to aid in progression through the rugby league playing pathway.

Greenhood orchids are most diverse in New Zealand and Australia, with at least some species pollinated by sexual deception of male fungus gnats. Preliminary investigation of endemic New Zealand Pterostylis brumalis via field phenology, hand-pollination and insect trapping revealed P. brumalis is self-compatible but relies on insects for cross pollination. Natural fruit-set was low, averaging 2.6% across two populations. High fruit-set in hand-pollinated flowers (66.7%) indicates severe pollinator limitation. In comparison, only one collected insect, a female Mycetophila vulgaris, bore orchid pollinia. Potential pollinator attractants (colour and scent) were analysed. Leukocyte chemotactic factor 2 (LECT2) amyloidosis is one of the most recently described types of amyloidosis. Since its description, it has been found to be one the most common types of amyloidosis in large series of amyloid cases involving the kidney and liver in the United States, where it primarily affects patients of Hispanic ethnicity. We sought to investigate the prevalence of this disease among Hispanic adult decedents who had an autopsy performed at the New Mexico Office of the Medical Investigator and determine the organ distribution of amyloid deposition. LECT2 amyloid deposits were identified within the kidney in 3.1% of Hispanic decedents. It was consistently deposited in the liver, spleen, adrenals, and lungs but did not involve the myocardium or brain. LECT2 amyloidosis is likely not rare among Hispanics in the Southwest United States and could represent an important but under-recognized etiology of chronic kidney disease in this population.

This cross-sectional study examined motivation for participation in physical activity at different adult life stages. T test analysis revealed no differences based on gender. Analysis of variance (ANOVA) indicated differences based on life stage. Individuals in the 18–24 age range were highly motivated by competence mastery factors. In addition, participants older than age 65 were more highly motivated by social aspects of leisure. These results suggest that leisure professionals should seek to offer young adults options that help develop skills while older adult options should be focused less on competency and more on building social environments.

Kauri forest represents a major ecosystem in northern New Zealand; however, the invertebrate fauna and their ecological diversity in these forests is very poorly known. This article investigates the composition and diversity of beetle communities in two kauri forest remnants, sampled by pitfall traps each month for one year. In total 4777 beetles were caught, representing 28 families, 84 genera and 107 species. Estimates of species richness indicate there were 173 species at both sites combined. The five most abundant species contributed 71%, and the top 10 species contributed 88% of all beetles caught. The abundance, richness and composition of the beetle community were consistent throughout the year. Patterns of niche overlap also show that the same set of species co-occur throughout the year, rather than temporal partitioning of the environment on an annual time scale. This data suggests that seasonality plays a limited role in explaining the composition and diversity of beetle communities in kauri forest.

Introduced pollinator species may exacerbate the problems faced by pollinator communities, so the detection in 2006 of a new solitary bee, Anthidium manicatum, to New Zealand was concerning. We assess whether establishment of this wool-carder bee presents potential risks to native flora and fauna by recording its current distribution, which plant species it visits, which insects co-occur at these plants, and identifying which species this territorial bee attacks. Anthidium manicatum is now widespread in urban New Zealand. More than 80% of the plants visited by A. manicatum were exotic species (predominantly Lamiaceae and Plantaginaceae), although it also visited native species of Veronica (Plantaginaceae) and Lobelia (Campanulaceae). Anthidium manicatum primarily interacted with other introduced bees. Native Lasioglossum bees were seldom attacked and only for short durations.
