Public sector entities are increasingly encouraged to implement enterprise risk management (ERM) to streamline procedures, create efficiencies, and safeguard delivery of public services. But, the ERM practices – as applied – adopt a confined concept of risk that subdues uncertain and unknown aspects of complex and (potentially) vast public exposures. We suggest that the management of uncertainty as opposed to risk – particularly in the public sector – requires different leadership approaches, organizing structures, and strategic response capabilities. It is argued that public institutions pursue distinctly different purposes and face unique challenges compared to commercial enterprise, which makes wholesale applications of ERM difficult in public sector entities. So, adoption of ERM in public settings requires a significant modification to the approach. This effort can take inspiration from strategic risk leadership in dealing with the complexity of public exposures vying for long-term solutions for society where successful outcomes are measured as sustainable resilience.

The capstone course for many Earth Science programs is a field course. To increase accessibility and to create a substitute in light of the cancellation of in-person courses due to the Covid-19 pandemic, we developed an online introductory field geology course. The course consisted of six different modules. The first was a virtual reality room, with 3-D rendered outcrops, designed to teach notebook skills and establish an effective learning community. This was followed by a module that taught orienteering and map datums. The third module taught field structural geology and utilized 3-D printed blocks and compasses sent to the participants. The next module consisted of a virtual field site in Google Earth built using a combination of videos, air photos, still photos, and hand samples sent to participants. These tools, along with provided structural measurements, were used by students to create a geologic map. The fifth module introduced core logging to students, a topic rarely covered in field mapping courses but nevertheless a critical skill for students pursuing a career in the mining and related industries. The final module introduced detailed geologic mapping, such as that done in pit, trench, or underground tunnel mapping. We assessed the success of these techniques in meeting the learning objectives using a combination of instructors’ observations, targeted rubrics, and two student feedback forms. While the course was generally deemed effective at meeting the learning objectives, some important potential improvements were identified, such as the addition of a group assignment using geologic maps at the beginning of the course. This additional module will give students an important background in geologic maps and encourage groupwork and idea sharing. Equipped with these skills, they will be better prepared and remain engaged for the remainder of the course. © 2021 National Association of Geoscience Teachers.

Background The number of forcibly displaced people globally has never been higher, with refugees from Syria constituting the largest displaced population worldwide. Many studies have documented elevated levels of mental health problems in refugee populations, though prevalence estimates of common mental disorders vary considerably between studies, explained both by methodological and contextual factors. Objective Using questionnaire-based screening checklists to approximate the prevalence of and investigate risk factors for post-traumatic stress disorder (PTSD), anxiety and depression among adult refugees from Syria resettled in Norway and to compare estimates with a sister-study in Sweden. Method Cross-sectional survey of a randomly selected sample from the National Population Register in Norway of adult refugees from Syria who were granted residency rights in Norway between 2015 and 2017 (Nsample = 9,990; nrespondents = 902). Above-threshold scores on the Harvard Trauma Questionnaire (HTQ) and Hopkins Symptoms Checklist (HSCL-25) defined caseness for PTSD (HTQ>2.06); anxiety (HSCLanxitey>1.75); and depression (HSCLdepression>1.80). Results Weighted, checklist-positive prevalence estimates for PTSD, anxiety and depression were 29.7% (25.4%-34.4%), 30.1% (25.7%-34.9%), and 45.2% (40.6%-49.8%), respectively. Cumulative exposure to potentially traumatic experiences before or during flight was a clear risk factor for all outcomes, and female gender was a risk factor for anxiety and depression, though only in adjusted analyses. The choice of HTQ cut-off to define PTSD caseness (2.5 vs. 2.06) had a notable effect on prevalence estimates. Conclusion In line with prior evidence, the present study suggests adult refugees from Syria resettled in Norway have higher rates of anxiety and depression and markedly higher rates of PTSD compared to general, non-refugee populations, and that this is clearly linked to past traumatic experiences. Prevalence estimates were highly consistent with estimates from the sister-study in Sweden, which used almost identical methodology. Findings underline the importance of screening for and intervening on mental health problems in newly arrived refugees., HIGHLIGHTS Nationwide, questionnaire survey of anxiety, depression and PTSD in adult refugees from Syria resettled in Norway estimated checklist-positive prevalence rates of: 29.7% (PTSD); 30.1% (anxiety); and 45.2% (depression).

Twenty distinct endophytic fungi were isolated from the surface-sterilized plant parts of Nymphaea nouchali and were identified using morphological and molecular techniques. At 300 µg/disc concentration, eight of the 20 fungal extracts exhibited antimicrobial activities against Staphylococcus aureus (ATCC 25923) and Bacillus cereus (ATCC 11778) while two within the eight showed activity against Pseudomonas aeruginosa (ATCC 9027) and Escherichia coli (ATCC 35218). Furthermore, investigation of the crude extract of Chaetomium globosum resulted in the isolation of two known cytochalasans, chaetoglobosin A and C, and their structures were elucidated and confirmed by mass and nuclear magnetic resonance (NMR) (1H, 13C, COSY, HSQC, HMBC and tROESY) spectral data. Chaetoglobosin A showed antibacterial activities against Bacillus subtilis (MIC 16 µg mL−1), Staphylococcus aureus (MIC 32 µg mL−1) and methicillin-resistant Staphylococcus aureus (MRSA, MIC 32 µg mL−1). This is the first study to report the isolation, identification and antimicrobial properties of endophytic fungi of N. nouchali in Sri Lanka.

Dietary cholesterol has received a lot of attention in regards to its potential association with both heart disease and Type 2 diabetes from health professionals and the general public. There is ample evidence from epidemiological data, meta-analysis reports and clinical interventions on the lack of a relationship between dietary cholesterol and heart disease risk. However, current reports appear to indicate that increased dietary cholesterol may be associated with an increased risk for heart disease in diabetic patients. Further, the relationship between dietary cholesterol and diabetes risk has not reached a consensus in the current literature and clinical studies have failed to prove that increased dietary cholesterol affects glucose metabolism or insulin resistance, indicating the need for more prospective and clinical studies. An endophytic fungus was isolated from surface sterilized leaf segments of Anoectochilus setaceus, an orchid endemic to Sri Lanka, and was identified as Xylaria sp. by morphological characters and DNA sequencing. Bioassay-guided chromatographic fractionation of the organic extract of a laboratory culture of this fungus led to the isolation of the known antibacterial helvolic acid. Helvolic acid was active against the Gram-positive bacteria, Bacillus subtilis [minimal inhibitory concentrations (MIC), 2 μg mL−1] and methicillin-resistant Staphylococcus aureus (MIC, 4 μg mL−1).

Chronic stress is prevalent in patients with ischemic heart disease (IHD) and worsens the long-term prognosis. Chronic stress is vaguely defined, but is associated with depressive symptoms, reduced psychological wellbeing, and reduced quality of life (QOL). Stress seems to induce hyperalgesia.The aim of the present study was to evaluate hyperalgesia by pressure pain sensitivity (PPS) in patients with IHD, and compare PPS to questionnaires measuring depressive symptoms, reduced psychological wellbeing, and QOL as markers of stress. Design. A cross-sectional study of 361 subjects with IHD.PPS was measured on the sternum, and compared to the questionnaires: Clinical stress symptoms score (CSS), Major Depression Inventory (MDI), WHO-5 Wellbeing Index, and SF-36 QOL score.PPS correlated to CSS (r = 0.20, p0.001), MDI (r = 0.14, p = 0.02), SF-36 mental component summary score (MCS) (r = - 0.10, p = 0.049), SF-36 physical component summary score (PCS) (r = - 0.17, p = 0.001), and self-perceived stress level (r = 0.15, p = 0.006). CSS correlated similarly (r = 0.5-0.7, all p0.001). Comparing subjects within the lowest vs. highest tertiles of PPS and CSS, the mean MDI score was 4 vs. 15, WHO-5 was 77 vs. 53, SF-36 PCS was 53 vs. 43, and SF-36 MCS was 58 vs. 46; all p0.001.PPS reflected to a modest degree markers of chronic stress in IHD. PPS and CSS together might be useful as easy-to use tools for evaluating these markers in IHD patients.

To explore children's perceptions of their teachers' feelings in everyday classroom contexts, the authors conducted focus groups with New Zealand primary (elementary) schoolchildren to discuss what they observed about positive classroom teachers' interactional style and emotional behavior. Seventy-nine students between age 8 and 12 years, from low- and high-socioeconomic communities, were divided into small focus groups to answer questions about their school experiences using a novel game-like procedure to foster open communication. Verbatim statements from the children were recorded and organized into major topics using thematic analysis. Age-related developmental differences were noted in the children's ability to recognize teachers' feelings. Mitotic kinase enzymes regulate critical stages of mitosis and are amenable to pharmacological inhibition. Since natural products have been a rich source of antimitotic inhibitors, we postulated that natural products would also provide effective inhibitors of mitotic kinases.To explore unique marine and terrestrial natural product sources for new anticancer drug leads, we screened our natural product extract library for polo-like kinase-1 (Plk1) kinase inhibitors.Extracts of the lichen Parmotrema sp. (Parmeliaceae) exhibited in vitro inhibitory activity. Bioassay-guided fractionation of the Parmotrema sp. extract led to the isolation of depside inhibitors.A new depside 1 has been isolated from the Sri Lankan lichen Parmotrema sp. along with the known metabolites 2 (β-collatolic acid) and 3 (β-alectoronic acid). The structure of depside 1 was elucidated by spectroscopic analysis. The three depsides 1-3 exhibited moderate inhibition of purified recombinant Plk1 kinase with IC₅₀ of 2.8, 0.7, and 1.7 µM, respectively, at 1 µM ATP. Inhibitory activity was also observed at high concentrations of ATP, suggesting the potential for activity in a cellular environment. The depsides were also tested against a panel of 23 other recombinant kinases and were found to possess up to 30-fold selectivity toward Plk1.These data suggest that the depsides 1-3 may serve as core structures that can be further explored as potential inhibitors of Plk1 and other kinases.

Two novel sesterterpenes, 1 and 3, containing a scalarane-based framework, have been isolated from the sponge Hyrtios erectus collected in Papua New Guinea. Their structures and relative stereochemistry have been elucidated by analysis of their spectroscopic data.

Xylopia championii. Hook. f. & Thoms. (Annonaceae) is endemic to Sri Lanka. The antioxidant and antifungal activities of five alkaloids, oxopurpureine, (+)-laudanidine, (−)-discretine, nordicentrine, and dehydrocorytenchine, isolated from the stem bark and stem of X. championii., were studied. The alkaloids, (+)-laudanidine and (−)-discretine, at a concentration of 0.5 mg/mL, exhibited exceptionally high antioxidant activity, where as nordicentrine and dehydrocorytenchine showed moderate activity as compared with the standard antioxidant dl-α -tocopherol in the DPPH assay. All five alkaloids were subjected to an antifungal bioassay against Clodosporium cladosporioides.. Nordicentrine showed the most potent antifungal activity at 6 μ g/spot, and (-)-discretine showed moderate activity at 30.0 μg/spot.

An important factor in determining perceived scene layout is optical contact between objects and the ground surface (“ground contact”). The relation between ground contact and occlusion was investigated in four experiments with stationary and motion parallax scenes containing a pole that occluded or was occluded by one or more cylinders. Some judgements were inconsistent with occlusion in stationary scenes, but judgements were consistent with occlusion in motion parallax scenes. When the range of object positions consistent with occlusion did not include the positions specified by ground contract or by motion parallax, the ground contact and motion parallax information still had a quantitative effect on the perceived position of the object within the range consistent with occlusion. These results demonstrate a cooperative interaction between ground contact, occlusion, and motion parallax in determining perceived layout in 3-D scenes.

Cabraleadiol monoacetate, 4-O.-methylcryptochlorophaeic acid, lichexanthone, and 3,6-dimethyl-2-hydroxy-4-methoxybenzoic acid showed mosquito larvicidal activity against the second instar larvae of Aedes aegypti.. Lichexanthone exhibited human sperm motility enhancing activity.

In the present study we assessed the use of landmarks and scene layout information for the control of locomotion. Observers were presented displays simulating forward locomotion through a random dot field with the horizontal position perturbed by a sum-of-sines function and were asked to steer and null out the horizontal disturbance of the path of locomotion. The results indicate greater control gain and accuracy when presented with a repeating layout of landmarks as compared to a changing layout of landmarks. Debriefing responses suggest that observers may have implicitly learned the layout of the repeating pattern. These results suggest that observers use an allocentric representation of the scene for steering control. A model for the control of locomotion is discussed that utilizes both scene-based information and optic flow.

Meng and Sedgwick (2001, 2002) found that the perceived distance of an object in a stationary scene was determined by the position at which it contacted the ground in the image, or by nested contact relations among intermediate surfaces. Three experiments investigated whether motion parallax would allow observers to determine the distance of a floating object without intermediate contact relations. The displays consisted of one or more computer-generated textured cylinders inserted into a motion picture or still image of an actual 3-D scene. In the motion displays, both the cylinders and the scene translated horizontally. Judged distance for a single cylinder floating above the ground was determined primarily by the location at which the object contacted the ground in the projected image (“optical contact”), but was altered in the direction indicated by motion parallax. Using a task switching paradigm, we investigated age effects on switch costs as a function of the number of sets to be switched. In Experiment 1, younger and older subjects determined a color or shape of an object presented on the computer screen, responding either by moving the joystick or by pressing a button on the joystick. The switch costs were assessed with differences between switch trials (task-set switch, response-set switch, and double switch) and non-switch trials. Contrary to the prediction that age would negatively influence performance on the double switch trials, age effects on switch costs were observed only for the single switch trials (i.e., response-set switch condition). Additionally, both younger and older adults were capable of preparing for task and response-set switches in parallel. While the clinical significance of terminal transferase (TdT) detection in acute lymphoblastic leukemia (ALL) is uncontested, conflicting evidence exists for the prognostic relevance of this enzyme in blast cells from patients with acute myeloid leukemia (AML). The technical challenge of determining TdT in myeloid blast cells may be primarily responsible for those discrepancies in the literature. We present here the relative values of various standard approaches to the detection of TdT in ALL versus AML, including the slide staining technique, which served as the reference method, flow cytometry, and the biochemical enzyme assay.

The novel sesquiterpene hydroquinone isoarenarol (1) and the known compound arenarol (2) were isolated from extracts of the marine sponge Dysidea arenaria Bergquist as part of a search for new protein kinase inhibitors. Both 1 and 2 showed potent and selective protein kinase inhibition in vitro.

The purpose of the study was to examine the effect of stimulation and inhibition of duodenal mucosal bicarbonate secretion on pancreatic and hepatic bicarbonate secretion in response to acid.The effect of inhibition (indomethacin) or stimulation (misoprostol) of duodenal mucosal bicarbonate secretion on pancreatic and biliary bicarbonate secretion in response to intraduodenal infusion of HCl or intravenous infusion of secretin was studied in anaesthetized pigs.The hepatic and pancreatic response to exogenous secretin was not significantly altered by stimulation/inhibition of duodenal bicarbonate secretion. However, pancreatic and biliary bicarbonate secretion in response to duodenal acidification was significantly augmented by inhibition of duodenal mucosal bicarbonate secretion; conversely, it was reduced by stimulation of duodenal bicarbonate secretion. The increase in plasma secretin levels in response to duodenal acidification was reduced by stimulation and augmented by inhibition of duodenal mucosal bicarbonate secretion.Duodenal mucosal bicarbonate secretion can serve as a modulator of both pancreatic and biliary bicarbonate secretion in response to luminal acidification, possibly through regulation of the release of secretin.

The addition of urea to sodium dodecyl sulfate (SDS)-polyacrylamide gels has allowed the identification and characterization of the small nuclear ribonucleoprotein particle (snRNP) D' protein and has also improved resolution of the E, F, and G snRNP core proteins. In standard SDS-polyacrylamide gels, the D' and D snRNP core proteins comigrate at approximately 16 kilodaltons. The addition of urea to the separating gel caused the D' protein to shift to a slower electrophoretic mobility that is distinct from that of the D protein. The shift to a slower electrophoretic mobility in the presence of urea suggests that the D' protein has extensive secondary structure that is not totally disrupted by SDS alone. Both N-terminal sequencing and partial peptide maps indicate that the D and D' proteins are distinct gene products, and the sequence data have identified the faster moving of the two proteins as the previously cloned D protein (L. A. Rokeach, J. A. Haselby, and S. O. Hoch, Proc. Natl. Acad. Sci. USA 85:4832-4836, 1988). In the cytoplasm, the D protein is found primarily in the small-nuclear-RNA-free 6S protein complexes, while the D' protein is found primarily in the 20S protein complexes. Like the D protein, the D' protein is an autoantigen in patients with systemic lupus erythematosus and is recognized by some of the Sm class of autoimmune antisera.

Treatment of metastatic breast cancer based on the estrogen receptor content (ER) of the primary tumor builds on the assumption that the ER status of the primary tumor and the metastases are largely equal. Studies addressing this question have used ligand-binding assays for ER determination and have consequently been subject to the limitations of this technique. Reported disparity rates have been 20%. In order to avoid some of these limitations, we used an immunohistochemical assay in paraffin-embedded tissue. Among a total of 92 examined regional lymph node metastases, ER status was equal with that of their 37 primaries in 84 cases (91%). Semiquantified ER content was significantly correlated in primary tumor and the metastases (r = 0.67, p less than 0.001). Among a total of 51 distant metastases, equal ER status was found in 44 (86%) cases and a quantitative relationship could not be established. Disparities can be due to methodological errors in the histochemical assay or tumor heterogeneity.

We have previously found reduced absorption of vitamin B12 in rats with choledochocolic fistulae. To investigate whether the reduction is caused by epithelial dysfunction or mucosal hypoplasia, choledochocolic fistulae were made in 11 rats, whereas 10 rats were sham-operated. The epithelial function was evaluated 9 days later by measuring the uptake of 57CoB12 and glucose in perfused intestinal segments and by determining the activities of 11 mucosal enzymes. Hypoplasia was investigated by performing morphometric measurements of the villus surface area and by measuring the weight, protein, and DNA in mucosal scrapings. The results suggest that choledochocolic fistulae in rats do not impair the epithelial function or cause mucosal hypoplasia. The urinary excretion of indican was increased in the fistula-operated rats, but further studies are needed to establish the significance of this observation.

The distribution of seed-borne N in shoot and root of pea and field bean was studied using three methods: 1) determination of the N content in shoot and root of plants grown in sand culture without other N sources, 2) 15N isotope dilution in plants grown in Rhizobium-free medium supplied with 15N-labelled nitrate and 3) determination of the 15N-enrichment in shoot and root of plants in which the seed-borne N was labelled with 15N. The results from the three methods were concordant, showing that from 44 to 50% of the seed-borne N in the two species was located in above-ground plant parts. The effect of corrections for seed-borne N in studies of nitrogen fixation in legumes is discussed.

SummaryTwo hundred and sixty antenatal cardiotocograms on 68 ‘at risk’ antenatal patients were evaluated against 3 definitions of fetal outcome. Reactivity was defined by a single fetal heart rate acceleration of at least 15 beats per minute above the baseline lasting at least 15 seconds. The low sensitivity of a non-reactive antenatal cardiotocogram precludes its application as a screening procedure or as confirmation of a compromised fetus. The excellent specificity of a reactive antenatal cardiotocogram reassures the clinician that the fetus is not compromised. Cardiotocography performed closer to the onset of labour predicts the likelihood of intrapartum fetal distress more accurately.

Intragastric pH was continuously measured over 24 h with a monocrystalline antimony electrode system and was compared with pH measured in simultaneously aspirated gastric juice and with pH measured by using a conventional intragastric glass electrode. There was a marked correlation between the pH readings obtained with the monocrystalline antimony electrode and the pH values measured in aspirated gastric juice (r = 0.92, p less than 0.001) and with the pH readings obtained with the intragastric glass electrode (r = 0.92, p less than 0.001). Both readings of pH with glass electrode and of pH after aspiration can be predicted by readings of pH with antimony electrode by using linear regression lines with slopes close to 1. Intragastric pH measurement is an alternative to aspiration of gastric juice, and the result obtained with an electrode of monocrystalline antimony is comparable to that obtained with a conventional glass electrode.

The Rous sarcoma virus (RSV) pp60v-src protein was expressed in Saccharomyces cerevisiae cells either from a plasmid vector carrying the v-src gene or in yeast cells containing a single-copy v-src gene chromosomally integrated. In both yeast strains, v-src gene transcription is regulated by the galactose-inducible GAL10 promoter. Growth in galactose-containing medium resulted in constitutive expression of pp60v-src in the integrated strain and transient expression of higher levels of pp60v-src in the plasmid-bearing strain. The concentration of pp60v-src in the plasmid-bearing strain at its peak of expression was approximately threefold lower than that found in RSV-transformed mammalian cells. pp60v-src synthesized in yeast cells was phosphorylated in vivo on sites within the amino and carboxyl halves of the molecule. In immune complex kinase assays, the yeast pp60v-src was autophosphorylated on tyrosine and was able to phosphorylate exogenous substrates such as casein and enolase. The specific activity of pp60v-src synthesized in yeast cells was approximately 5- to 10-fold higher than that made in mammalian cells. Induction of pp60v-src caused the death of the plasmid-bearing yeast strain and transient inhibition of growth of the single-copy strain. Concomitantly, this induction resulted in high levels of tyrosine phosphorylation of yeast cell proteins. This indicates that pp60v-src functions as a tyrosine-specific phosphotransferase in yeast cells and suggests that hyperphosphorylation of yeast proteins is inimical to cell growth.

A series of 50 patients treated with total cystectomy for transitional cell carcinoma is reported. The cases are grouped according to the histologic type and stage of the carcinoma. Forty-three patients had anaplastic carcinomas, of which 32 had invaded more than half of the bladder musculature (Stages P3 and P4). Twelve patients came to operation despite the presence of recognised distant metastases. Nine primary deaths occurred among 38 patients treated with radical surgery. When five years had elapsed, 8 of 29 patients were alive (28 per cent), of whom one had a recurrence in loco. The conclusion is drawn that cystectomy is best suited to tumours which have invaded to a depth of more than half the bladder musculature, or those with perivesical infiltration (Stage P3). However, the results hitherto recorded with supervoltage radiation are somewhat better than those obtained with cystectomy. Pure nickel and nickel-based binary and ternary materials containing tungsten carbide and graphite dispersions have been studied. Three types of carbide powder and six types of graphite, different in size, shape, and structure (one in each group being nickel-coated) were studied to varying degrees. The smallest carbide powder (0·35 μm) possessed the best wear-resistance and the lowest coefficient of friction. The larger and coated types of graphite are somewhat superior; however, considerable amounts (∼ 40 vol.-%) are needed to improve wear-resistance substantially and reduce the coefficient. Use of the coated type of carbide leads to rather high coefficients (0·58-0·76) under dry testing conditions. In a consecutive series of 28 patients with various gastro-intestinal disorders, a close relationship was shown between the secretion of hydrochloric acid, of intrinsic factor, and of the uropepsinogen excretion. The study suggests a possible close correlation between gastric acid production and the genetically determined ABH blood group secretor status.
