14. Promising Performance: The Overestimation and Underestimation of Performance Targets

This study examines goal setting in the federally funded, state-implemented Weatherization Assistance Program (WAP). Specifically, using federally determined criteria that states are supposed to use to set expected performance targets, I model the predicted performance targets and compare them against the observed performance targets. The purpose is to examine the determinants of performance target overestimation and underestimation in the WAP. The findings suggest that, where credible commitment to achieving performance targets is important (i.e., when grantees want to show top performance to attract grant money and federal partners), grantee governments are more likely to underestimate goals. However, where the aspirational nature of performance targets is more important than the credible commitment of achieving them (i.e., in an election year or partisan congruence with federal government), grantee governments are more likely to overestimate goals.

Performance metrics are intended to reinforce accountability in the principal-agent relationship between administrators and elected officials. This form of accountability is often assumed to improve democratic responsiveness and policy outcomes. However, as many scholars have noted, performance measurement systems often result in the intractable problem of administrative gaming, which can dilute accountability in the principal-agent relationship. This qualitative case study generates theory on how and why administrative agents engage in gaming. It develops a typology and discusses the importance of understanding gaming from the perspective of administrators.

Rules are dynamic entities whose implementation is shaped by context, resources, and institutions. Variations in this implementation influence the functionality and effectiveness of rules. While the last decade has seen considerable scholarship on the determinants and effects of ineffective and effective rules, less is known about how rule implementation evolves over time. Particularly, what are the factors that influence administrative implementation intent over time? This study uses interview and archival data to examine administrative implementation intent of a single rule over a 25-year period. From qualitative data analysis, propositions are developed to explain why, although the text of the rule remained nearly the same, administrative intent of implement changed drastically.

The relationship between contract management capacity and implementation success in local government contracting is well established. However, less is known about how specific contract mechanisms are linked to this success. This study uses implementation of the Energy Efficiency and Conservation Block Grant Program to examine the use of formal and informal contract management techniques in the implementation of energy projects. The use of formal contract mechanisms, such as full and open competitive bidding, contact rescission and outcome-based performance measures, exhibits a statistically significant effect on implementation performance. The findings advance contracting theory and have practical implications for local government contracting and implementation success in fiscal federalism.

Two experiments examined the effect of film music on the narrative persuasion. In Experiment 1, participants viewed a short film with its original musical soundtrack or with soundtrack muted. In Experiment 2, musical soundtrack was added to a film that was originally produced without music. Findings indicated that participants reported greater transportation into the film and greater agreement with film-relevant beliefs when soundtrack was presented but only when music was congruent with the film's affective tone. In support of the transportation-imagery model (Green & Brock, 2000), the effect of film music on beliefs was mediated by transportation into the film.

A review of methodological inquiry shows that, from its inception, comparative public administration (CPA) scholarship brushed over important epistemological and ontological questions in favor of searching for broad-based, generalizable theories that subscribed to only objectivist ontologies. This article traces these problematic methodological assumptions in contemporary CPA research back to the mischaracterization of the terms idiographic and nomothetic in CPA research. Revisiting this history is important because the recognition of this distinction allows room in the field for research that highlights the uniqueness of different cultural contexts and constructs.

The goal of this study was to determine if there is any association between the DNA mutations in the insulin-like growth factor-1 receptor gene and litter size in Polish sows. The following reproductive traits were investigated: total number of piglets born (TNB), number of piglets born alive (NBA), and number of piglets weaned. The polymorphism in the IGF1R gene was analyzed by a PCR-RFLP method using the restriction enzymes SacII. Two different alleles of the IGF1R gene were identified. The statistical analysis in first parity showed that the BB genotype was superior and the differences were highly significant (p≤0.01). Later parities sows with the BB genotype still had the largest litter size compared to AB and AA sows, but the difference was statistically not significant. The preliminary study suggests a possibility of using the IGF1R gene polymorphisms to improvement of litter size of sows.

The ‘woman question’ played a central role in the rhetoric and debates surrounding the Islamic Revolution in Iran, yet the precise outcome of the woman question is still debated. While some scholars argue that the revolution has repressed women by forcing them into traditional roles, others argue that the revolution has liberated women by mobilising them in the public sphere. This article claims that the revolution tried to create a female subject who was simultaneously pious and politically active, and that the creation of this particular subjectivity exceeds and defies the categories and dichotomies of earlier scholars. To make this claim, the article utilises both an intellectual history of ‘the woman question’ and the interpretivist trends of women themselves, showing how the ideal, revolutionary Islamist women was discursively produced and enforced. An analysis of the metastability of silicon-on-insulator (SOI) complementary metal-oxide-silicon (CMOS) latches is presented, using partially-depleted SOI devices with various body-connection topologies and an unbuffered latch. The metastability window, resolution time and time interval between the clock edge and the time t meta are evaluated as functions of power supply and the type of body-connection topology. Simulations using SOISPICE show improved metastability behaviour for SOI specific body-connection topologies.

To investigate the interaction between vascular endothelial growth factor (VEGF) and its receptor, we have constructed a chimeric protein consisting of the extracellular ligand-binding domain of the human VEGF receptor subtype KDR fused to a human IgG1 Fc domain (KDR-Fc). KDR-Fc was expressed in human 293 kidney epithelial cells as a 300-kDa secreted, dimeric glycoprotein that bound 125I-VEGF165 with high affinity (Kd = 150 pM). Unlike the full length cellular receptor, KDR-Fc did not require heparin for 125I-VEGF165 binding, although heparin did stimulate 125I-VEGF165 binding approximately 50 to 100%. Similar results were observed for KDR-Fc expressed in yeast cells. Since yeast do not synthesize heparan sulfate proteoglycans, we conclude that cellular heparan sulfates do not account for the lack of a heparin requirement for 125I-VEGF165 binding to KDR-Fc. The polycationic protein protamine, which inhibits (IC50 = 1 microgram/ml) 125I-VEGF165 binding to bovine aortic endothelial cells and other KDR-expressing cells by blocking heparin interactions, had no effect on the heparin independent component of 125I-VEGF165 binding to KDR-Fc. Protamine does inhibit (IC50 = 1 microgram/ml) the heparin dependent component of 125I-VEGF165 binding to KDR-Fc. KDR-Fc bound VEGF121 with the same affinity as VEGF165. Heparin had no effect on 125I-VEGF121 binding to KDR-Fc, indicating that heparin interaction with the 44 amino acids contained in VEGF165 but not VEGF121 allow for maximal VEGF165 binding. Deletion analysis of KDR-Fc demonstrated that the determinants required for high affinity VEGF binding are located in the three aminoterminal Ig-domains of the protein. Heparin had no effect on 125I-VEGF165 binding to the three Ig-domain receptor, suggesting that there are heparin binding determinants located in KDR Ig-domains 4 to 7.

Vascular endothelial growth factor (VEGF), a potent and specific activator of endothelial cells, is expressed as multiple homodimeric forms resulting from alternative RNA splicing. VEGF121 does not bind heparin while the other three isoforms do, and it has been documented that the binding of VEGF165 to its receptor is dependent upon cell surface heparin sulfate proteoglycans. Little is known about the biochemical mechanism that allows for heparin regulation of growth factor binding. For example, it is not clear whether heparin interactions with growth factor or with cell surface receptors or both are essential for VEGF binding to its receptor. In this manuscript we provide results which are consistent with the hypothesis that an interaction between heparin and a site on the KDR receptor subtype is essential for VEGF165 binding. First, we demonstrate that expression of KDR into a CHO cell line deficient in heparan sulfate biosynthesis does not allow VEGF165 binding unless heparin is exogenously added during the binding assay. Secondly, we show that a ten amino acid synthetic peptide, corresponding to a sequence from the extracellular domain of the KDR, both inhibits VEGF165 binding to the receptor and also binds heparin with high avidity. Third, affinity purification of heparin molecules on a KDR-derived peptide affinity column, together with capillary electrophoresis and polyacrylamide electrophoresis analysis, was used to show that the KDR-derived peptide interacts with a specific subset of polysaccharide chains contained in the unfractionated heparin. Taken together, these results are consistent with the hypothesis that interactions between cell surface heparan sulfate proteoglycans and the VEGF receptor contribute to allowing maximal VEGF binding.

Vascular endothelial growth factor (VEGF) is an angiogenic growth factor which binds to two structurally related tyrosine kinase receptors denoted KDR and FLT1. To compare the interaction of VEGF with each receptor, cell lines which express individual receptor subtypes were identified using Northern blot hybridization. Bovine aortic endothelial (ABAE) cells and WM35 melanoma cells were found to express KDR, while FLT1 was primarily expressed on SK-MEL-37. Both receptor subtypes were detected on another melanoma cell line (WM9). Heparin augmented VEGF binding to KDR-expressing cells (ABAE and WM35), but inhibited VEGF binding to FLT1-expressing cells (SK-MEL-37 and WM9). The concentration of heparin required for half maximal stimulation of VEGF binding to KDR-expressing cells (500 ng/ml) was 25 times greater than that required for half maximal inhibition of binding to FLT1-expressing cells (20 ng/ml). In WM9 cells, the effect of heparin was bimodal; low concentration inhibited, while higher concentrations stimulated binding of 125I-VEGF. Placenta growth factor (PIGF-1) is a recently described growth factor structurally similar to VEGF. PIGF-1 had a negligible or no effect on 125I-VEGF binding to KDR-expressing cells (ABAE, WM35), but did complete for binding to FLT1-expressing cells (SK-MEL-37 and WM9). Addition of heparin had no effect on its ability to compete for binding with 125I-VEGF. The data indicate differential regulation of the two VEGF receptors by heparin and extended specificity of FLT1 receptor, but not KDR, for binding PIGF-1 growth factor.

The results axe applied to prove the existence of traveling wave solutions of the FitzHugh-Nagumo equations and a system of reaction diffusion equations which arise as a model for a two step combustion process.

Transforming growth factor-beta 1 (TGF beta 1) and basic fibroblast growth factor (bFGF) are known to be potent inhibitors and stimulators, respectively, of endothelial cell growth in vitro. In the present study we examined the effect of bFGF on endothelial cell growth inhibitory activity of TGF beta 1 and on the binding of (125I)-TGF beta 1 to these cells. The concentration of TGF beta 1 required for half-maximal inhibition of endothelial cell growth was increased in a dose-dependent manner by bFGF (a 20-100 fold increase at 1 ng/ml of bFGF). A 24 h-pretreatment of cells with bFGF resulted in abolition of the TGF beta 1 inhibitory effect on DNA synthesis. Moreover, the binding of (125I)-TGF beta 1 to the endothelial cell surface was decreased in a concentration-dependent and time-dependent manner after a preincubation of these cells with bFGF. Analysis of the binding parameters showed that bFGF decreased by two-fold the number of TGF beta receptors (to approximately 6000 receptors per cell). Cross-linking experiments with disuccinimidyl suberate demonstrated the presence of two TGF beta receptor subtypes, a predominant 85 kDa form and a minor 65 kDa form. Basic FGF decreased selectively the labeling of the 85 kDa TGF beta receptor subtype. These findings suggest that the growth stimulator bFGF can attenuate the cell's response to the growth inhibitor TGF beta 1.

The aim of the experiment was to detect polymorphism in the GPX5 gene and to determine associations between individual genotypes and selenium concentration in liver and kidney of wild boars from West Pomerania province in Poland. The polymorphism in GPX5 gene was detected using the PCR-RFLP method with specific primers and the restriction enzyme HinfI. Two different alleles of the GPX5 gene were identified - allele 1B (0.37) and 2B (0.63). The relationships between the GPX5 genotypes and concentrations of selenium in liver and kidney were analyzed, revealing statistically significant (P≤0.05) differences between wild boars carrying different GPX5 genotypes in both genders. Obtained results indicated that wild boars with the 2B2B genotype had the highest selenium concentration in livers and kidneys compared to 1B1B and 1B2B animals and these differences were statistically significant (P≤0.05).
