The role of cryptochrome 1 in photosynthetic processes and pro-/antioxidant balance in the Arabidopsis thaliana plants was studied. Wild type (WT) and hy4 mutant deficient in cryptochrome 1 grown for 20 d under red (RL, 660 nm) and blue (BL, 460 nm) light at an RL:BL = 4:1 ratio were kept for 3 d in different lights: RL:BL = 4:1, RL:BL:GL = 4:1:0.3 (GL - green light, 550 nm), and BL, then were exposed to high irradiance (4 h). Activity of PSII and the rate of photosynthesis in WT and hy4 decreased under the high irradiance in all spectral variants but under BL stronger decrease in the activity was found in the hy4 mutant than in WT. We assumed that lowered resistance of photosynthetic apparatus in the hy4 mutant may be associated with the low activity of the main antioxidant enzymes and reduced content of low-molecular-mass antioxidants in the mutant compared to the WT. The changes in photosynthetic activity, as well as the activity of nitrogen-metabolism enzymes, the intensity of lipid peroxidation, and proline content were studied in Triticum aestivum L. plants after their incubation at a low CO2 concentration in a sealed chamber for 10 d. CO2 deficiency (-CO2) compared to normal CO2 concentration (control) led to a decrease in the rate of O2 gas exchange at the plateau of the light curve and quantum yield of photosynthesis. The maximum and effective quantum photochemical yields also decreased. CO2 deficiency reduced the activity of nitrate reductase, but increased the activities of nitrite reductase, glutamine synthetase, and glutamate dehydrogenase, and promoted proline accumulation. It is assumed that with a lack of CO2, an excess of nitrogen-containing compounds occurs, which must be removed from metabolic processes. The light spectral composition acting through a set of photoreceptors, such as cryptochromes and phytochromes, plays an important role in maintaining sustainable photosynthesis. An impact of cryptochrome 1 deficiency and additions of green light (GL) against the background of red (RL) and blue (BL) (different ratios of RL:BL:GL) on the activity of the photosynthetic apparatus, the content of photosynthetic pigments, pro-/antioxidant balance, and expression of some genes in the leaves of 23-d-old Arabidopsis thaliana hy4 mutant plants was studied. The deficiency of cryptochrome 1 at RL/BL ratio of 4:1 led to a decrease in the rate of photosynthesis, photosystem II activity, and activity of ascorbate peroxidase and total peroxidase but to an increase in the content of products reacting with thiobarbituric acid. However, in the presence of additional GL, this difference for photosynthetic parameters either decreased or was absent, likely due to a GL-induced decrease in the content of active cryptochrome. Light-induced changes in miRNAs, morphogenesis, and photosynthetic processes in phytochrome-deficient mutant plants grown under different light qualities were studied. miRNA activity in many processes is regulated by phytochromes and phytochrome-interacting factors (PIFs). The reduced content of photoreceptors in phytochrome mutants affects the PIF-microRNA interaction. In plants grown under red light (RL) and white light (WL), the phenotype of phyb mutant was distorted; however, under blue light (BL) conditions, the phyb phenotype was normalized. The photosynthetic rates of both the mutants and wild type were higher under BL than under RL and WL. The expression of most studied miRNAs increased in phyaphyb mutants under BL conditions, which is probably one of the reasons for the normalization of the phenotype, the increase in PSII activity, and the photosynthetic rate. MicroRNAs under BL can partially improve photosynthesis and phenotype of the mutants, which indicates the conjugation of the functioning of phytochromes in miRNA formation. [ABSTRACT FROM AUTHOR]

The effect of UV-A radiation (365 nm) and the protective effect of preillumination with red light (RL, 664 nm, 10 min) or with a combination of red and far-red light (FRL, 727 nm, 10 min) on the activity of the PSII as well as the expression levels of selected genes, especially those encoding chloroplast proteins (sAPX, tAPX, CAB1, and D1), were studied in leaves of the 26-d-old hy3 mutant of Arabidopsis thaliana, which is deficient in the phytochrome B apoprotein. The effects were compared with corresponding effects observed in the hy2 mutant of A. thaliana, which is deficient in the phytochrome chromophore. Illumination with UV-A decreased the photosynthetic pigment content, the maximum photochemical quantum yield of PSII (F/F), and the effective quantum yield of PSII (Φ). The reduction of the F/F ratio and Φ was more pronounced in the mutants as compared to wild-type plants (WT). The preillumination of the leaves with RL caused a significant reduction in the inhibitory effect of UV-radiation on the PSII activity in the WT plants, but it caused only a small decrease in the hy3 mutant. The preillumination of leaves with RL and FRL combination compensated the protective effect of RL on the UV-induced decrease of the fluorescence parameters in the WT. Such reversibility is typical for involvement of red/far-red reversible phytochromes at low intensity light. The results suggest an important role of red/far-red reversible phytochromes (phytochrome B) in the resistance of PSII to UV-A radiation caused by changes in contents of either carotenoids or other UV-absorbing pigments probably through biosynthesis of these pigments. The data also demonstrated that phytochrome B and other phytochromes can affect the PSII stress resistance by the fast regulation of the expression of genes encoding antioxidant enzymes and transcription factors at the step of gene transcription.
