Calculations of heating of membrane-forming holes in Teflon film exposed to decimeter waves were performed. The dependence of the temperature increment in holes on the geometry of holes, electrolyte concentration, and decimeter wave frequency was studied. The kinetics of heating depending on the hole diameter was also obtained. It was concluded that the observed in the experiment effects of the decimeter wave on bilayer lipid membranes resulted from the elevated concentration of decimeter electromagnetic waves in membrane-forming hole that led to selective heating of electrolyte in hole and bilayer lipid membranes.

In the present review, we tried to evaluate the known properties of gas hydrates and gases participating in the formation of gas hydrates from the point of view of the mechanisms of cryoinjury and cryoprotection, to consider the papers on freezing biological materials in the presence of inert gases, and to analyze the perspectives for the development of this direction. For the purpose, we searched for the information on the physical properties of gases and gas hydrates, compared processes occured during the formation of gas hydrates and water ice, analyzed the influence of the formation and growth of gas hydrates on the structure of biological objects. We prepared a short review on the biological effects of xenon, krypton, argon, carbon dioxide, hydrogen sulfide, and carbon monoxide especially on hypothermal conditions and probable application of these properties in cryopreservation technologies. The description of the existing experiments on cryopreservation of biological objects with the use of gases was analyzed. On the basis of the information we found, the most perspective directions of work in the field of cryopreservation of biological objects with the use of gases were outlined. An attempt was made to forecast the potential problems in this field.

The effect of the antioxidant α-tocopherol on rat brain protein kinase C activity as a model to study bimodal dose-dependent effect has been investigated. Enzyme activity has been monitored photometrically with a luciferase reporter assay that measures ADP produced by posphorylation. The inhibition of protein kinase C activity by α-tocopherol was found at the concentration range from 10(-3) to 10(-6) M with no effect of ultra low doses of the antioxidant (below. 10(-12) M). The absence of bimodal dose-dependent effect may be associated with the enzyme source.

It is shown that the exposure of heparinized venous human blood diluted in phosphate buffer saline to extremely weak alternating magnetic fields of the ultralow-frequency (1 Hz, 600 nT; 4.4 Hz, 100 nT; 16.5 Hz, 160 nT) in combination with a collinear static magnetic field of 42 microT at physiological temperatures, causes a sharp 3-4 fold increase in its chemiluminescence after addition of luminol.

It is shown that an exposure of pupae of the mealworm beetle Tenebrio molitor to the combined static (42 μT) and very weak alternating (250 nT) magnetic fields exerts different influence, depending on the frequency of the alternating magnetic field, on duration of metamorphosis processes in these insects. For instance, an exposure of pupae to weak combined magnetic fields, adjusted to the frequency of ion cyclotron resonance for glutaminic acid (4,4 Hz), stimulates metamorphosis process--a transitional stage from pupae to imago lasts shorter. An inhibiting effect was observed when adjusted to the frequency of ion cyclotron resonance for Ca2 (32,2 Hz). At some frequencies this effect is not seen. For instance, an exposure at a frequency of ion cyclotron resonance for K+ (16,5 Hz) exerts no noticeable effect on the duration of the pupal metamorphosis stage.

It was demonstrated that fibroblast growth factor bFGF influences the process of heme synthesis, the proliferation activity and viability of bone marrow mesenchymal stem cells in culture under hypoxic conditions. The addition of fibroblast growth factor bFGF (7 ng/ml) to the medium under above conditions led to the accumulation of aminolevulinic acid--an early porphyrin and heme precursor, an increase in CD 71 expression--a transferrin receptor, and also a decrease in porphyrin pigments and heme contents--a late precursor and end products of heme synthesis, respectively. It was found that cultivation of the cells under hypoxic conditions and bFGF is an optimum to maintain high viability and proliferation capacity of the mesenchymal stem cells.

In this paper we derive a formula to calculate the amount of free water molecules in solution. Physical values in this formula may be obtained by analyzing the spectra of aqueous solutions in the terahertz frequency range. Formula is derived on the basis of considering water polarization process in electric field. It is shown that without processes of shielding the electric field in the water calculations lead to very high estimation of a share of free water molecule.

We analyzed spectra of light and heavy water at temperatures from 4 up to 50 degrees C in a frequency range of 0.15 to 6.5 THz. It was shown that the amplitude of high-frequency relaxation absorption band with its maximum at 0.5 THz extends with increasing, temperature and this temperature dependence for light water has a marked feature at 35-40 degrees C as a sharp growth. This fact is noteworthy because this range corresponds to physiological values of a body temperature of the warm-blooded organisms. At the same time, the analogous temperature dependence for heavy water in the considered temperature range lacks this particular feature. Thus, the water with its properties differs significantly not only from other fluids, but also from its own isotopologues.

We have evaluated the morphology of the mouse preimplantation embryos at developmental stages from morula to late-blastocyst after two different impacts: microinjection of modified Witten's medium and osmotic stress in physiological osmolarity (310 mOsM), in 5% glucose (560 mOsM) at high concentration of NaCl (614 mOsM). Results of our research showed that these stresses caused similar changes in embryo morphology: volume was reduced followed by its recovery in culture medium (osmolality was less than a physiological value, 260 MOsM). The ability of embryos to recover the volume and morphology up to the initial level depends on a stage of embryo development and consequently competence of TB cells. In this study it was revealed that a key role in regulation of volume homeostasis after microinjection and after short-time (30-60 min) osmotic stress belongs to TB cells. Both physical effects induce the further embryo development in vitro up to the formation of primary colonies of embryonic and trophoblastic cells. These data could be used to develop the morphological criteria for a prediction of blastocyst-stage embryonic implantation potential.

The influence of FGF2 on the efficiency of cardiomyocytes differentiation of mesenchymal stem cells (MSC) of bone marrow induced by 5-azacetidine (5-aza) was studied. The effect of FGF2 developing by the 14th day after the combined action of a differentiating agent and growth factor was manifested in an increase in Mef2A, Mef2D and gene transcription and a rise of ionized Ca2+ concentration in cytoplasm keeping cell viability and proliferation activity. In the presence of FGF2 this approach provided cardiomyogenesis and the increase in the formation of early precursors of cardiomyocytes.

In this paper we present the results of spectral studies of water solutions and water at different temperatures. The calculation method for estimation of the portion of free water molecules is offered. The method is based on the analysis of transmissions spectra features in the terahertz region. The calculation using the proposed method shows that the portion of free water molecules at room temperature ranges from 10 to 16% for a variety of water solutions. In case of pure water, the temperature increase from 20 to 50 degrees C results in the increase of the free molecule fraction from 12% to 19%. Moreover, the increase in the number of free water molecules within a specified temperature range exhibits significant nonuniformity: the most intense growth was observed in the range of 30-40 degrees C. This is particularly interesting because this temperature range includes the optimum temperatures for the existence of warm-blooded animals.

In this paper, we analyzed spectra of liquid water and water solutions in a frequency domain, characteristic of the collective dynamics of water molecules (from 0 to 200 cm(-1)). Particular attention is paid to the relaxation processes, one of which is observed in the terahertz region of the spectrum (approximately 5-50 cm(-1)). The physical essence of this process at the molecular level is still unclear. Based on data obtained this process is strongly suggested to interpret as a monomolecular relaxation of unbound water molecules.

Effects of four inhibitors of NF-kappaB, SAPK/JNK and TLR4 signaling, namely, inhibitor XII, SP600125, CLI-095 and Oxpapc on a macrophage response to low dose ammonium were studied in RAW 264.7 cells. Low dose ammonium induced pro-inflammatory response in cells as judged from enhanced production of TNF-alpha, IF-gamma, and IL-6, and by activation of signal cascades. The increase in production of cytokines, namely TNF, IFN, and IL-6, demonstrated that low-dose ammonium induced a pro-inflammatory cellular response. In addition, an activation of NF-kappaB and SAPK/JNK cascades, as well as enhancement of TLR4 expression was shown. Each of used inhibitors reduced to a variable degree the pro-inflammatory response of RAW 264.7 cells on chemical toxin by decreasing cytokine production. The inhibitor of NF-kappaB cascade, IKK Inhibitor XII, was more effective, and not only prevented the development of pro-inflammatory response induced by ammonium, but also decreased cytokine production below control values. The inhibitor of extra cellular domains of TLR2 and TLR4 (OxPAPC) had almost the same anti-inflammatory effect, and an addition of the inhibitor of JNK cascade (SP600125) to cell culture practically neutralized effect of ammonium ions by decreasing cytokine production to control level. Inhibitory analysis showed that activation of RAW 264.7 cells induced by chemical toxin coincide incompletely with intracellular signaling pathways that were early determined regarding macrophage's response to toxin from gram-negative bacteria. Nevertheless, application of the inhibitors defended RAW 264.7 from toxic effect of the low dose ammonium.

In this paper we studied experimentally the frequency dependence of heating of human skin exposed to millimeter waves. Theoretical modeling of obtained data was performed using the hybrid bio-heat equation. It was found that the skin heating and SAR increased with increasing the exposure frequency. The frequency dependence of heating was entirely resulted from that of reflection from the skin. Unlike temperature, the frequency dependence of the SAR was due to the increased absorption of millimeter wave energy within the thin surface layer of the skin.

Two different genes of peroxiredoxin 6 are encoded in the genome of Xenopus laevis: xen1 (Acc.no. EMBL Data Bank - BCO54278) and xen2 (Acc.no. EMBL Data Bank - BC540309). Both genes were cloned and expressed in Escherichi coil. Proteins were purified and analyzed. The amino acid sequences of the enzymes Xen1 and Xen2 are 95% identical with the same peroxidase activity, pH and temperature optimums, as well as thermostability, being approximately equal. The expression of peroxiredoxin 6 genes significantly differ during ontogenesis of X. laevis. The expression of xen1 starts on a later stage of development 47-48, while the gene xen2 is expressed on all stages of development with the same increase starting from stage 0-5. The level of xen2 expression in embryos increased after incubation in presence of hydrogen peroxide. The comparison of amino acid sequences of proteins Xen1 and Xen2 shows that only the enzyme Xen2 may have phospholipase activity, since it has residues of phospholipase A2 active center: Ser31, His25, Asp139.

The possibility of using modified peroxyredoxins as powerful antioxidant agents has been considered. Peroxyredoxins immobilized on perfluorocarbon emulsions and PTD-modified peroxyredoxins have been studied. It has been shown that peroxyredoxins efficiently bind to particles of perfluorocarbon emulsions, while maintaining their antioxidant properties. A panel of PTD-modified peroxyredoxins has been created and peroxyredoxins most effective both in antioxidant properties and the ability to penetrate cells have been selected. The modified peroxyredoxins obtained may serve as the basis for the design of drug with powerful antioxidant action.

The possibility of using thermocouples for the artifact-free measurements of skin temperature during millimeter wave exposure was studied. The distributions of the specific absorption rate (SAR) in the human skin were calculated for different orientations of the thermocouple relative to the E-field of exposure. It was shown that, at the parallel orientation of a thermocouple relative to the E-field, SAR significantly increased at the tip of the thermocouple. This can result in an overheating of the thermocouple. At the perpendicular orientation of a thermocouple, the distortions of the SAR were insignificant. The data obtained confirm that the skin temperature can be measured with a thermocouple during exposure under the condition that the thermocouple is located perpendicular to the E-vector of the electromagnetic field. For the accurate determination of SAR from the rate of the initial temperature rise, it is necessary to fit the temperature kinetics measured with the thermocouple to the solution of the bio-heat transfer equation.

A number of effects of weak combined (static and alternating) magnetic fields with an alternating component of tens and hundreds nT at a collinear static field of 42 microT, which is equivalent to the geomagnetic field, have been found: the activation of fission and regeneration of planarians Dugesia tigrina, the inhibition of the growth of the Ehrlich ascites carcinoma in mice, the stimulation of the production of the tumor necrosis factor by macrophages, a decrease in the protection of chromatin against the action of DNase 1, and the enhancement of protein hydrolysis in systems in vivo and in vitro. The frequency and amplitude ranges for the alternating component of weak combined magnetic fields have been determined at which it affects various biological systems. Thus, the optimal amplitude at a frequency of 4.4 Hz is 100 nT (effective value); at a frequency of 16.5 Hz, the range of effective amplitudes is broader, 150-300 nT; and at a frequency of 1 (0.5) Hz, it is 300 nT. The sum of close frequencies (e.g., 16 and 17 Hz) produces a similar biological effect as the product of the modulating (0.5 Hz) and carrying frequencies (16.5 Hz), which is explained by the ratio A = A0sin omega1t + A0sin omega2t = A0sin(omega1 + omega2)t/2cos(omega1 - omega2)t/2. The efficiency of magnetic signals with pulsations (the sum of close frequencies) is more pronounced than that of sinusoidal frequencies. These data may indicate the presence of several receptors of weak magnetic fields in biological systems and, as a consequence, a higher efficiency of the effect at the simultaneous adjustment to these frequencies by the field. Even with consideration of these facts, the mechanism of the biological action of weak combined magnetic fields remains still poorly understood.

Effects of three chemical compounds: ammonia, diethyl ether, and acetic acid, known as common environmental contaminants in technogenic accidents, were investigated in vivo and in vitro in low concentrations. When added in cultivation media, each of the chemicals has affected peritoneal macrophages and spleen lymphocytes isolated from male NMRI mice and led to a rise in the production of several cytokines, particularly the tumor necrosis factor-alpha and interferon-gamma, as well as the expression of the inducible form of heat shock proteins (HSP72 and HSP90-alpha) and in the activation of signal cascades NF-kappaB and SAPK/JNK. The increase of the nitric oxide (NO) production in macrophages has been observed only when ammonia was added in cultivation media. Also, low concentrations of all compounds investigated led to the activation of the expression of receptor protein TLR4. When mice were exposed to airborne toxic contaminants in a hermetically sealed experimental chamber, an increase in the concentrations of cytokines, heat shock proteins, and signal proteins in immune cells was also observed in response to low concentrations of all chemicals investigated. Similarly to in vitro experiments, the NO production was augmented only in the presence of the airborne ammonia. The results indicate the environmental hazard of chemical contaminants even in rather low concentrations, which nevertheless lead to the stress response.

The parameters of the low-frequency (1, 4.4, 16.5 Hz or the sum of these frequencies) extremely weak (300, 100, 150-300 nT, according to frequencies) alternating component of combined magnetic fields have been found, which in combination with a weak collinear static constant field of 42 nT (the induction corresponds to the range of the geomagnetic field) has a marked antitumor activity. The exposure to these magnetic fields inhibits the tumor growth in mice with the intraperitoneally transplanted Ehrlich ascites carcinoma. The effect manifests itself in an increase in the life of tumor-bearing animals and in the content of damaged tumor cells. It was found that the death of tumor cells by the action of weak fields occurs predominantly by the mechanism of necrosis.

It has been found that the recombinant protein LIF induces jumps of current in bilayer lipid membranes, which indicates the formation of ionic channels. Some properties of these channels (dependence on voltage, sing of potential, ionic strength of solution, and lipid composition) were studied. A difference between the effects of protein LIF of eukaryotic and prokaryotic origin was shown.

The distributions of specific abcorption rate (SAR) and E-field in a membrane-forming hole of Teflon film and surrounding electrolyte were calculated for 0.9 GHz exposure. It was found that the specific absorption rate in the membrane-forming hole increased greatly with increasing thickness of the Teflon film, and electrolyte concentration and decreasing diameter of the hole. The previously demonstrated significant changes in the conductivity of modified bilayer lipid membranes induced by microwave exposure can be explained by a local increase in specific absorption rate and subsequent elevation of temperature in the membrane-forming hole of the Teflon film.

The effect of low-intensity lases light (0.2 mW/cm2, 632.8 nm, exposure time 1 min) or centimeter waves (8.15-18 GHz, 1 W/cm2, exposure time 1 h) on PhosphoSAPK/JNK production in mice lymphocytes was investigated. Normal isolated spleen lymphocytes or cells incubated previously with geldanamycin, an inhibitor of heat shock protein 90 (HSP90), were used in the experiments. A significant stimulation of PhosphoSAPK/JNK production in lymphocytes after treatment with laser light or microwaves has been shown in both cell models. It was proposed that the activation of SAPK/JNK signal pathway plays one of the central roles in cellular stress response to low-power nonionizing radiation.

The effects of thymopentin, a synthetic analogue of the active center of the thymus hormone thymopoietin, on the immune status of mice with two different models of inflammation induced by injection of lipopolysaccaride (LPS) from gram-negative bacteria were studied. Acute inflammation was induced by a single injection of LPS in a dose of 250 microg/100 g of body weight, and chronic inflammation (sepsis) was modeled by a daily injection of LPS for 11 days with a gradual increase in the dose range from 25 to 250 microg/100 g of body weight. Under acute inflammation, a preliminary injection of thymopentin did not induce any additional stimulation in cytokine production increased by LPS. On the contrary, whereas the chronic introduction of LPS was characterized by a depressed production of several cytokines, thymopentin produced an immunostimulating effect. Thus, an increase in the production of nitric oxide, interferon-gamma, and Hsp70 was demonstrated. In addition, a more effective restoration of the number of thymus cells, as well as an increase in macrophage tumor necrosis factor-alpha production were observed after repeal of LPS + hormone injections. The results show that preliminary application of thymopentin promotes the regulation of immune cell activity under acute and chronic inflammation.

The effect of the stem cell factor on the state of membranes and functional activity of mouse embryonic stem cells cultivated in LIF (Leukemia Inhibitory Factor) cytokine-free and LIF-containing media has been studied. It was shown that the stem cell factor induces changes in the viscosity of membrane lipid bilayer and increases the respiration rate, the ATP level, and the proliferation activity of embryonic stem cells. An intricate character of the LIF-dependent modification of biological effects of the stem cell factor was revealed.

The present investigation reveals that a 250-fold screening of the geomagnetic field ("zero" geomagnetic fields, 200 nT) is a biologically active factor that adversely affects embryonic cells and the processes of early embryogenesis as a whole. In particular, the cultivation of primary embryonic fibroblasts in "zero" geomagnetic fields causes reduces the capacity for adhesion and proliferation, changes the monolayer morphology and increases cell death. In a more highly organized experimental model, two-celled mouse embryos, the exposure to the "zero" field results in an increase of plasma membrane permeability for dyes, a reorganization of the cytoskeleton because of alpha-actin redistribution, and the disturbance of the spatial orientation of blastomeres. As a result, the development of two-celled mouse embryos stops, and they do not reach the stage of blastocyst. These data show the significant role of geomagnetic fields in the normal growth of embryonic cells in vitro and the regulation of mammalian embryogenesis.

In vitro effects of low-level electromagnetic waves (8.18 GHz, frequency swings within 1 s, intensity 1 microW/cm, exposure for 1 h) and low-energy laser light (He-Ne laser with 632.8 nm, 0.2 mW/cm, dose 1.2 x 10(-2) J/cm2) on the expression of receptor protein TLR4, which is known as a part of the system for microbal toxin recognition, were studied in mouse lymphocytes. In addition, TLR4 expression was examined in situations when stress responses to low-level nonionizing radiation were modified by the antibiotic geldanamycin, which suppresses the activity of the heat shock protein Hsp90. It was found that low-level microwaves significantly raised the amount of TLR4; in contrast, laser light decreased the expression of the receptor in lymphocytes. In cells pretreated with geldanamycin, the TLR4 expression in irradiated cells was reduced to minimum levels, much lower than control values. The results showed that TLR4, which is involved in specific binding of toxin from gram-negative bacteria, can regulate cell responses to signals of other origin, in particular to nonionizig radiation, including low-level microwaves and laser light.

The effects of low-level electromagnetic waves (8.15-18 GHz, 1 microW/cm2, 1 h) on the production of heat shock proteins, several cytokines, and nitric oxide in isolated mouse macrophages and lymphocytes were examined both under normal conditions and after the treatment of the cells with geldanamycin (GA), a depressor of activity of the heat shock protein 90 (Hsp90). The irradiation of cells without GA induced the production of Hsp70, nitric oxide (NO), interleukin-1beta (IL-1beta), interleukin-10 (IL-10), and the tumor necrosis factor -alpha (TNF-alpha). No changes in the production of Hsp90 in irradiated cells were observed, but intracellular locations of Hsp25 and Hsp70 altered. The preliminary treatment of cells with GA did not remove the effects of microwaves: in these conditions, the synthesis of all cytokines tested, nitric oxide, as well as total and membrane amount of Hsp70, and the amount of Hsp25 in the cytoplasm and cytoskeleton increased. Moreover, the exposure of cells incubated with GA resulted in the reduction of Hsp90-alpha production.

It has been shown that, in the range of 03000 nT, a biological system differently responds to the treatment with a weak static magnetic field. Thus, at a practically complete compensation of the field (induction 5 nT), the intensity of fission of planarian does not differ from control values. As the field intensity is successively increased to 300 nT, a marked and statistically significant stimulating effect is observed (SC 1.51.8), which disappears in the intensity range of 400 and 600 nT. At intensities from 800 to 1500 nT, the stimulating effect manifests itself again. Further increase of field intensity to 3000 nT leads to the disappearance of the effect.

The effects of centimeter waves (8.15-18 GHz, 1 microW/cm2, 1 h daily for 10 days; MW) on the production of the tumor necrosis factor alpha, interleukin-lalpha, interleukin-1beta, interleukin-2, and the expression of interleukin-6, interleukin-10, interferon-gamma, nitric oxide and HSP27, HSP72 and HSP90alpha in mice irradiated before or after LPS injection were studied. An acute endotoxic model was produced by a single LPS injection. The effects of microwaves on nitric oxide, interleukin-6, tumor necrosis factor-alpha, and interferon-gamma were dependent on the functional status of exposed animals. Thus, an exposure of healthy mice to microwaves for 10 days was followed by a decrease in nitric oxide and interferon-gamma production, and an increase in the production of the tumor necrosis factor-alpha and interleukin-6. On the contrary, an exposure to MW before intoxication resulted in an increase in the synthesis of nitric oxide and interferon-gamma as well as a decrease in the concentration of the tumor necrosis factor-alpha and interleukin-6 in blood of mice in endotoxic shock. When microwave exposure was used after LPS injection, it did not provide any protective effect, and preliminary irradiation enhanced the resistance of the organism to endotoxic shock.

The effects of low-intensity laser radiation (LILR, 632.8 nm, 0.2 mW/cm2) and low-intensity electromagnetic waves (LIEW, 8.15 - 18 GHz, 1 MW/cm2) on the production of transcription factors in lymphocytes from NMRI male mice were examined. The total level of NF-KB and its phosphorylated metabolite Phospho-NF-kappaB, as well as the regulatory protein IkappaB-alpha were determined in spleen lymphocytes subjected to laser or microwave radiations. The proteins were determined by immunoblotting. Laser light induced a lowering in the level of NF-kappaB and IkappaB-alpha. By contrast, irradiation with electromagnetic waves resulted in a significant increase in the amount of NF-kappaB and IkappaB-alpha. The phosphorylated form of NF-kappaB did not noticeably change under either of the two kinds of radiation. The results showed that electromagnetic waves activate the production of both NF-kappaB and the regulatory protein IkappaB-alpha and these data confirm the stress character of the response of spleen lymphocytes to low-level microwaves of the centimeter range.

Peroxiredoxins (Prx) are a family of antioxidant proteins with peroxidase activity. The ability of 1-Cys Prx to self-associate was studied with the use of native PAGE and Western blotting. Two protein bands corresponding to monomeric and dimeric forms were detected in the preparation of the recombinant 1-Cys Prx subjected to native PAGE, with dimers being more abundant. The third band corresponding to the oligomeric form was detected after incubation of the recombinant 1-Cys Prx with DTT, although monomers and dimers were also observed. These results indicate that monomeric, dimeric, and oligomeric states of the protein are likely to be interchangeable. Native PAGE in combination with Western blot analysis revealed that self-association of 1-Cys Prx also occurred at physiologically relevant concentrations in vivo. The native 1-Cys Prx existed in the monomeric and dimeric forms in rat olfactory epithelium, with monomers being more common. The structural sensitivity of the recombinant 1-Cys Prx to imidazole was shown.

It has been shown that exposure to weak combined permanent (42 mT) and low-frequency (3-60 Hz) alternating (0.001-0.06 mT) magnetic fields changes the intensity of fluorescence of water and water-salt solutions. The gel filtration of solutions of inorganic salts treated with magnetic fields gives rise to intensively fluorescing fractions. Control solutions not exposed to electromagnetic waves do not exhibit these effects. The results obtained suggest that treatment with weak electromagnetic fields induces structural changes of water solutions, and the manifestations of these changes depend on the conditions of chromatography and chemical composition of solutions under study.

It has been shown that exposure to weak combined collinear magnetic fields (permanent component 42 mT; amplitude of alternating component 40 nT, frequency 3.7 Hz) or millimeter waves with a frequency of 36 GHz and power density of 100 mT/cm2 substantially stimulates the growth of the regeneration blastema in the tail fragment of planaria when the exposure to fields precedes the cutting of the planaria body. This effect is more clearly pronounced during the treatment of planaria with magnetic field. If the treatment with weak physical factors is carried out after the cutting of planaria, the effect of the field is two times less pronounced (exposure to magnetic waves) or is not evident at all (exposure to electromagnetic radiation).

The effect of preliminary short-term irradiation with He-Ne laser light (632.8 nm, 0.2 mW/cm2) of the thymus zone projection of male NMRI mice subjected to acute toxic stress on the responses of immune cells was studied. Stress was modeled by lipopolysaccharide injection, 250 mg/100 g of body weight, which induced a significant increase in the production of several macrophage cytokines, IL-1alpha, IL-1beta, IL-6, IL-10 and TNF-alpha. A single irradiation with laser light did not provoke considerable variations in NO production in cells but induced an enhancement in the production of heat shock proteins Hsp25, Hsp70, and Hsp90. Nevertheless, when irradiation with red laser light was applied prior to toxic stress, considerable normalization of production of nearly all cytokines studied and nitric oxide was observed. Moreover, the normalization of production of heat shock proteins has been shown in these conditions. Thus, preliminary exposure of a small area of animal skin surface provoked a significant lowering in the toxic effect of lipopolysaccharide.

The problems of a low efficiency of mammalian cloning are discussed with emphasis on the necessity of the expertise of each step of single cell reconstruction, beginning with microsurgical manipulations. The fact of cell content leakage when the cell is held during microsurgery or microinjections with the help of the conventional method using negative pressure in the holding micropipette was demonstrated in experiments on murine embryos. It was shown that the rate of cell content efflux depends on the value of negative pressure generated in the holding micropipette, and is directly proportional to the dimensions of its orifice and the duration of micromanipulations. An alternative method of cell fixation using the capillary forces of the holding micropipette was proposed. The method optimizes the process of cell fixation, reducing the holding effort by two orders of magnitude. As a result, 92% of embryos remain viable after fixation of embryo, as compared with 39% in the conventional technique. In order to diminish the cell damage produced by the tip of a microinstrument, a new technique of fabricating micropipettes was proposed. The improved method of filling the micropipette with viscous liquids, including DNA, which is described in details in the paper, enabled constant (non-stop) microinjection of more than 1000 cells by hand, without any special automatic device.

The effect of in vitro exposure to low-power laser light with a power density of 0.2 mW/cm2 and a wavelength of 632.8 nm induced by helium-neon laser on the functional activity of macrophages and splenic lymphocytes was studied. If the exposure period did not exceed 60 sec, the stimulation in interleukin-2 (IL-2) and nitric oxide (NO) production, as well as an increase in the activity of natural killer cells were observed. The increase of irradiation dose by prolongation of the exposure duration up to 180 s induced a significant decrease in NO production and natural killer cell activity, but IL-2 production was not different from the control level. A remarkable decrease in interferon-gamma (IFN-gamma) production was observed following laser light exposure of cells for 60 or 180 s, whereas under lower doses (exposure for 5 or 30 sec) IFN-gamma production increased. Irradiation of isolated macrophages induced a significant stimulation of cellular tumor necrosis factor-alpha (TNF- alpha) production at all dboes used, and, what is more important, an enhancement in both TNF-a phaand interleukin-6 (IL-6) production was revealed as early as after a 5-s exposure. In this case, more prolonged exposure periods, 60 and 180 s, either did not induce changes in IL-6 production (in macrophages) or decreased IL-6 production (in lymphocytes). Thus, upon in vitro exposure of cells to extremely low-power laser light, a basic tendency was observed: short-term irradiation predominantly induced stimulation in secretory activity of cells, whereas prolongation of exposure mainly induced immunosuppression. The only exception to the rule was a change in interleukin-3 (IL-3) production, which decreased after short-time exposure, but, on the opposite, increased when the cells were exposed for 180 s. In addition, a high sensitivity to extremely low-power laser light was supported by expression of the inducible heat shock protein, Hsp70, the effect being observed at all doses used, including the exposure for 5 s. At the same time, expression of another heat shock protein, Hsp90, was somewhat reduced after irradiation of cells with laser light.

An experimental model of differentiated mouse embryonic stem cells with retractive activity similar to that of cardiomyocytes without preliminary formation of embryoid bodies was obtained. The basic factor that induced in vitro embryonic cell differentiation into cardial type is the recombinant cytokine LIF under prolonged cultivation. The positive reaction of the cells with retractive activity to isoproterenol indicates the presence of the beta-adrenergic receptor activity characteristic only for terminal differentiated mammalian cardiomyocytes.

The effect of helium-neon laser light of extremely low power of 0.2 mW/cm2 and wavelength 632.8 nm on the immune status of mice bearing solid tumors was studied. The evaluation of the status of tumor-bearing animals was provided by taking into account the number of immune cells, cytokine concentration (tumor necrosis factor, interleukin 2, production of nitric oxide, expression of heat shock proteins (Hsp70 and Hsp90), and activity of natural killers. The model of a solid tumor was formed by subcutaneous injection of Ehrlich carcinoma cells, and average life span of tumor-bearing mice achieved about 55 days. Different areas of the skin of tumor-bearing mice were subjected either to a single (1 min, dose 0.012 J/cm3) or repeated exposure to laser light (1 min, 48-h intervals, 30 days). Two different areas were irradiated: the thymus projection area or a hind limb with solid tumors. The results showed that chronic exposure of tumor-bearing mice in the thymus projection area, and especially, hind limb, reduced the resistance, which manifested itself in the acceleration of tumor growth and a tendency of mouse life span to decrease. On the contrary, a single exposure stimulated the antitumor immunity for several days after the exposure. The results show the expediency of further investigation of the immunomodulative effects of low-power laser light and the necessity of monitoring the immune system during laser therapy.

The effect of low-intensity laser light on the activity of natural killer cells from healthy and tumor-bearing mice was studied. Skin in the zone of the thymus or hind limb was illuminated, the remaining body surface being screened. The illumination was carried out for 30 days, with the duration of a single exposure being 1 min and intervals between the exposures being 48 h. The effect of laser light depended on the location of the illuminated area. It was shown that the exposure of the thymus of healthy animals for 20 and 30 days leads to a significant decrease in the activity of natural killer cells. On the contrary, the illumination of the limb for 10 or 20 days increased the activity of natural killer cells; but when hind limbs were treated for 30 days, the activity of natural killer cells decreased. Whereas tumor growth increased the natural killer cell activity, the illumination of tumor-bearing mice lowered the adaptive antitumoral resistance by decreasing the activity of natural killer cells.

The effects of millimeter (40 GHz) and centimeter (8.15-18.00 GHz) low-intensity waves on the production of tumor necrosis factor (TNE) in macrophages and lymphocytes from exposed mice as well as in exposed isolated cells were compared. It was found that the dynamics of TNF secretory activity of cells varies depending on the frequency and duration of exposure. The application of millimeter waves induced a nonmonotonous course of the dose-effect curve for TNF changes in macrophages and splenocytes. Alternately, a stimulation and a decrease in TNF production were observed following the application of millimeter waves. On the contrary, centimeter waves provoked an activation in cytokine production. It is proposed that, in contrast to millimeter waves, the single application of centimeter waves to animals (within 2 to 96 h) or isolated cells (within 0.5 to 2.5 h) induced a much more substantial stimulation of immunity.

It was found that single total-body exposure to electromagnetic centimeter waves (8.15-18 GHz, 1 microW/cm2, 5 h) stimulated the proliferation of mouse T and B splenic lymphocytes. The same effects were observed upon in vivo treatment of rats for 5 h with millimeter waves (42.2 GHz, amplitude modulation 10 Hz, 1 microW/cm2). The whole-body irradiation with centimeter or millimeter waves did not cause any significant changes in natural activity of killer cells. The cellular responses induced by the irradiation of isolated animal cells in vitro did not coincide with those revealed after the total-body irradiation of animals. Thus, the in vitro irradiation of natural killer cells to millimeter waves for 1 h increased their cytotoxic activity whereas, after treatment to centimeter waves for the same time, the activity of killer cells did not change. On the contrary, irradiation of T and B lymphocytes with millimeter waves (42.2 GHz, amplitude modulation 10 Hz, 1 microW/cm2, 1 h) suppressed the blasttransformation of cells. The results show a higher immunostimulative potential of centimeter waves as compared to millimeter waves.

The effect of low-intensity centimeter electromagnetic waves (8.15-18 GHz, 1 microW/cm2, 1.5 h daily, 20 days) on the production of tumor necrosis factor, intreleukin-2, and interleukin-3 and the expression of the heat shock protein 72 in healthy and tumor-bearing mice was measured. A significant increase in tumor necrosis factor production and a slight reduction of interleukin-2 concentration were observed after exposure to microwaves; we consider these effects as adaptive response. The interleukin-3 production in healthy mice was not affected by microwaves. Low-intensity centimeter waves induced antitumoral resistance in tumor-bearing mice. Thus, exposure of tumor-bearing mice led to a significant rise in the tumor necrosis factor production and the normalization of both interleukin-2 and interleukin-3 concentration. We assume that the significant immunomodulating effect of low-density centimeter microwaves can be used for immunocorrection and suppression of tumor growth.

It was found that weak magnetic fields (constant 42 microT; variable 0.05 microT; summation of frequencies in the range of 3.58-4.88 Hz) substantially (approximately fourfold) accelerate the hydrolysis of the amyloid beta-protein in aqueous solution. A region of the amyloid beta-protein most sensitive to the action of weak magnetic fields was determined. It is localized between Asp and Ser amino acid residues in positions 7 and 8 of the peptide sequence. It is in this region that the hydrolysis of the molecule of the amyloid beta-protein by the action of magnetic fields predominantly takes place.

The literature data and our own data on the synaptic plasticity and remodeling of synaptic organelles in the central nervous system are reviewed. Modern techniques of laser scanning confocal microscopy and serial thin sectioning for in vivo and in vitro studies of dendritic spines, including the relationship between morphological changes and the efficacy of synaptic transmission, are discussed using, in particular, a model of long-term potentiation. The organization of dendritic spines and postsynaptic densities of different categories as well as the role of filopodia in spine genesis were analyzed. It was shown that the method of serial ultrathin sections is the most effective for unbiased quantitative stereological analysis and 3D reconstructions. By using the refined method of serial ultrathin sections with subsequent three-dimensional reconstructions, the presence of giant mitochondria in hippocampal neuronal dendrites was demonstrated. It was shown that smooth endoplasmic reticulum forms a unified continuum with the outer membrane of the mitochondrial envelope within dendrites. It was suggested that this continuum provides calcium tunneling, which makes possible intracellular signal transduction during synaptic transmission. Evidence is presented indicating the presence of gap junctions ("electrical synapses") in the synapses of mammalian brain, as well as between glial processes, and between glial cells and neurons. Our data and the data of other authors show that glial cell processes form a structural and functional glial network, which modulates the functioning of the neuronal network. The connection of dendritic spines with the glial network is shown on 3D reconstructions by analyzing the neuropil volume in CA1 hippocampal area of ground squirrels in three functional states: normothermia, provoked arousal, and hibernation when brain temperature falls below 6 degrees C. The own data of the authors are discussed indicating the formation of more than five presynaptic boutons (multiple synapses) on both CA1 mushroom-like dendritic spines and CA3 thorny excrescences. On the basis of the analysis, new ideas of the organization and functioning of synapses were suggested.

The results of the study of the temperature dependence of water absorption spectra in the visible region and in the region of vibrational overtones are discussed.

The effect of low-level millimeter fractionated radiation on the production of tumor necrosis factor, intreleukin-2, interleukine-3, and nitric oxide and on the activity of natural killer cells and proliferation of T and B lymphocytes in mice was studied. Cell activity was measured in four groups of male Balb/c mice (control, exposed, tumor-bearing unexposed, and exposed tumor-bearing animals) within 30 days of tumoral growth and microwave exposure (42.2 GHz, 10 Hz amplitude modulation, 0.5 microW/cm2, 1.5 h daily). A significant increase in the production of tumor necrosis factor and nitric oxide and in the activity of natural killer cells was observed at the early stage of tumor development; this effect was considered as adaptive response. In healthy mice, millimeter radiation produced both stimulating and immunodepressive effects. The changes were nonmonotonous; as the exposure duration was increased, the stimulating effect became weaker and on day 30 it was not observed. Irradiation of tumor-bearing mice did not induce any significant changes in the activity of cells compared to unirradiated tumor-bearing animals. Moreover, exposure to millimeter waves impaired some characteristics of cell immunity in tumor-bearing mice. It was concluded that low-intensity millimeter waves do not increase the resistance against tumor as it was shown earlier in our experiments with centimeter waves.

It was shown that the stimulating effect of weak combined magnetic fields (constant component 42 microT, frequency of the variable component 3.7 Hz) on the division of planarians depends on the amplitude of the variable component of the field. The effect is particularly pronounced at 40 (the main maximum), 120, 160, and 640 nT. Narrow ranges of effective amplitudes alternate in some cases with equally narrow ranges in which the system does not respond to he treatment. In the range of super weak amplitudes of the variable field (0.1 and 1 nT), the stimulating effect is poorly pronounced. The data obtained indicate the presence of narrow amplitude windows in the response of the biological systems to weak and super weak magnetic fields. In a special series of experiments, it was shown that the effect of fields on planarians is partially mediated via aqueous medium preliminarily treated with weak magnetic fields. It is noteworthy that in experiments with water treated with weak magnetic fields, there were no pronounced maxima and minima in the magnitude of the effect in the range of amplitude of the variable magnetic field from 40 to 320 nT.

The mechanism of interaction of an external magnetic field with liquid was proposed. The statistical integral and configurational contributions for a free energy, entropy and specific heat for the soliton model of bulk knitted structures in a magnetic field were calculated. It was shown that the concentration of solitons depends on the effect of external fields. In the specific case of bulk knitted structures (liquid water without magnetic field), the theoretical data are consistent with experimental. The memory effects in systems with hydrogen bonds in magnetic field was explained in the framework of the continuum soliton concept.
