7. Characterization and genetic mapping of eceriferum-ym (cer-ym), a cutin deficient barley mutant with impaired leaf water retention capacity

Abstract

The cuticle covers the aerial parts of land plants, where it serves many important functions, including water retention. Here, a recessive cuticle mutant, eceriferum-ym (cer-ym), of Hordeum vulgare L. (barley) showed abnormally glossy spikes, sheaths, and leaves. The cer-ym mutant plant detached from its root system was hypersensitive to desiccation treatment compared with wild type plants, and detached leaves of mutant lost 41.8% of their initial weight after 1 h of dehydration under laboratory conditions, while that of the wild type plants lost only 7.1%. Stomata function was not affected by the mutation, but the mutant leaves showed increased cuticular permeability to water, suggesting a defective leaf cuticle, which was confirmed by toluidine blue staining. The mutant leaves showed a substantial reduction in the amounts of the major cutin monomers and a slight increase in the main wax component, suggesting that the enhanced cuticle permeability was a consequence of cutin deficiency. cer-ym was mapped within a 0.8 cM interval between EST marker AK370363 and AK251484, a pericentromeric region on chromosome 4H. The results indicate that the desiccation sensitivity of cer-ym is caused by a defect in leaf cutin, and that cer-ym is located in a chromosome 4H pericentromeric region.

9. Variation in the wheat AP2 homoeologs, the genes underlying lodicule development

Abstract

The bread wheat genome harbors three homoeologs of the barley gene HvAP2, which determines the cleistogamous/non-cleistogamous flowering. The three homoeologs, TaAP2-A, TaAP2-B and TaAP2-D, are derived from the A, B and D genomes. The importance of lodicule swelling in assuring non-cleistogamous flowering in a range of wild and domesticated wheat accessions of varying ploidy level was established. Re-sequencing of wheat AP2 homoeologous genes was carried out to identify natural variation at both the nucleotide and polypeptide level. The sequences of wheat AP2 homoeologs are highly conserved even across different ploidy levels and no functional variants at the key miR172 targeting site were detected. These results indicate that engineering of cleistogamous wheat will require the presence of a functional TaAP2 modification at each of the three homoeologs.

10. Construction of a High-density AFLP and SSR Map Using Recombinant Inbred Lines of Cultivated×Weedy Soybean

Abstract

A genetic linkage map of soybean was constructed by using a cross between a cultivar 'Keburi' and a weedy form of soybean 'Masshokutou Kou 502'. The parents differed in their ability in somatic embryogenesis. The mapping population consisted of 117 recombinant inbred lines (F_), and generated 30 linkage groups, which covered 2089 cM, including 515 amplified-fragment-length polymorphism (AFLP) markers and 85 simple sequence repeat (SSR) markers. For AFLP analysis, a simple technique using small polyacrylamide gels combined with silver staining was applied, which enabled a single person to construct a map in a short time at a reasonable cost. Comparison of this map with previously published maps revealed that 1) the level of polymer-phism was the same as in a cultivar×cultivar cross; and 2) the cultivar×weed map was 13% shorter than the cultivar×wild map. The high-density map constructed in this study would contribute to the molecular mapping of quantitatively and qualitatively inherited characters from weedy forms of soybean.

30. Mutations within the miR172 target site of wheat AP2 homoeologs regulate lodicule size and rachis internode length.

Abstract

Closed fertilization in flowers, or cleistogamy, reduces the risk of fungal infection in Triticeae crops. In barley ( Hordeum vulgare ), cleistogamy is determined by a single recessive gene, cly1 , which results from a single nucleotide polymorphism within the microRNA172 target site of the Apetala2 ( AP2 ) transcription factor gene. The recessive cly1 allele negatively regulates the development of lodicules, keeping florets closed at anthesis. However, cleistogamy is not evident in hexaploid wheat ( Triticum aestivum ) cultivars. This study aimed at identifying mutations in wheat AP2 orthologs by ethyl methane sulfonate-induced mutagenesis and high-resolution melt analysis. Although flowers of AP2 mutants induced in the A and D genomes opened at anthesis, their lodicule size was significantly smaller, especially in the direction of depth, than that of wild-type plants. One of the mutants that carried a nucleotide replacement in AP2 from the D genome produced a compact spike caused by a substantial decrease in rachis internode length, analogous to the barley dense spike.
