Pikeperch (Sander lucioperca) is a freshwater species and an internationally highly demanded fish in aquaculture. Despite intensive research efforts on this species, fundamental knowledge of skeletal muscle biology and structural characteristics is missing. Therefore, we conducted a comprehensive analysis of skeletal muscle parameters in adult pikeperch from two different origins, wild-caught specimens from a lake and those reared in a recirculating aquaculture system. The analyses comprised the biochemical characteristics (nucleic acid, protein content), enzyme activities (creatine kinase, lactate dehydrogenase, NADP-dependent isocitrate dehydrogenase), muscle-specific gene and protein expression (related to myofibre formation, regeneration and permanent growth, muscle structure), and muscle fibre structure. The findings reveal distinct differences between the skeletal muscle of wild and farmed pikeperch. Specifically, nucleic acid content, enzyme activity, and protein expression varied significantly. The higher enzyme activity observed in wild pikeperch suggests greater metabolically activity in their muscles. Conversely, farmed pikeperch indicated a potential for pronounced muscle growth. As the data on pikeperch skeletal muscle characteristics is sparse, the purpose of our study is to gain fundamental insights into the characteristics of adult pikeperch muscle. The presented data serve as a foundation for further research on percids' muscle biology and have the potential to contribute to advancements and adaptations in aquaculture practices., (© 2024. A study was conducted to evaluate the gonad differentiation of juvenile yellow perch (YP, Perca flavencens) and determine the latest labile period related to hormone treatment. Juvenile fish were subjected to two dietary concentrations of methyltestosterone (MT; 20 and 50 mg/kg feed) for 60 days in three (3) age groups of 38-, 46-, and 67-days post-hatching (dph), where control group were fed with standard commercial feed. Following a 10-month on-growing period, sex phenotypes were determined by gross and histological gonad morphology. Results showed the juvenile YP responded to the exogenous hormone when it was applied at 38 dph for both 20 and 50 mg/kg feed resulting in 100% males. At 46 dph, only 50 mg/kg feed resulted in 100% males. Both MT-treated at 38 and 46 dph significantly differed (P < 0.01) from the expected normal population of male:female (1:1). MT-treated at 67 dph resulted in 37% and 25% intersex fish for both 20 and 50 mg/kg feed dosage groups, respectively. MT-treated at 38 and 46 dph promoted growth and showed significantly heavier mean body weight (P < 0.05) compared to control. The gonadosomatic index (GSI) of MT-treated at 38 and 46 dph was significantly lower than that in control. This study provides the first evidence that juvenile YP can be successfully masculinized when the treatment is initiated at the age of up to 46 dph. An 8-week feeding trial was conducted to evaluate the effects of dietary carbohydrate to lipid (CHO:L) ratios on growth performance, body composition, serum biochemical indexes, lipid metabolism, and gene expression of central appetite regulating factors in Chinese perch (Siniperca chuatsi) (mean initial weight: 12.86 ± 0.10 g). Five isonitrogenous and isoenergetic diets (fish meal, casein as main protein sources) were formulated to contain different graded CHO:L ratio diets ranging from 0.12, 0.86, 1.71, 3.29, and 7.19. Each diet was assigned to triplicate groups of 18 experimental fish. Our results revealed that final body weight (FBW), weight gain rate (WGR), specific growth rate (SGR), and protein efficiency ratio (PER) increased with dietary CHO:L ratio from 0.12 to 1.71 and then decreased with further increases in dietary CHO:L ratio. A two-slope broken-line regression analysis based on WGR showed that the optimal dietary CHO:L level for maximum growth performance of fish was 1.60. Crude lipid and crude protein content in the liver and glycogen concentration in the muscle and liver were significantly influenced by the dietary CHO:L ratios (P < 0.05). The lowest crude lipid content in the liver was observed in fish fed the diet with a CHO:L ratio of 1.71(P < 0.05). Dietary CHO:L ratios significantly induced the glucose concentration of serum (P < 0.05). The relative expression levels of genes involved in lipid metabolism, such as srebp1 and fas in the liver, showed a trend of first decreased and then increased with the increase of dietary CHO:L ratio levels. Appropriate CHO:L ratio in the diet can effectively reduce the accumulation of liver fat. We observed in fish fed the 1.71 CHO:L ratio diet showed higher feed intake, up-regulated mRNA expression of neuropeptide Y (npy) and agouti gene-related protein (agrp), and down-regulated mRNA expression of cocaine- and amphetamine-regulated transcript (cart) and pro-opiomelanocorticoid (pomc) significantly as compared to control group. The effect of a constant photoperiod on the inhibition of male and female reproductive cycles was studied in pikeperch Sander lucioperca. Over a 153-day period, batches of pikeperch (2 years, 950 g) breeders were kept under either under natural or artificial photoperiod conditions (24L:0D) (30-35 fish/tank, triplicate) and sampled in late June (start of the photoperiod decrease in natural conditions), late August (start of temperature decrease) and late November (exogenous vitellogenesis) (7-10 fish/tank/sampling date). Morphological parameters, sexual steroids, alkaline-labile phosphate (µg/mL) levels and gamete developmental stages were investigated. Gonado-somatic index (%), developmental stages and sexual steroid levels (17β-estradiol, testosterone and 11-ketotestosterone, ng/mL) in both sexes and oocyte diameter (µm) and plasma alkaline-labile phosphate (µg/mL) in females were lower in response to a continuous lighting (24L:0D). In both sexes, continuous lighting applied in June for 153 days totally inhibited or delayed the onset of the reproductive cycle. In conclusion, photoperiod manipulation can be used to delay the pikeperch reproductive cycle, even if temperature decreases. This is the first report of the inhibitory effect of photoperiod on the onset of the reproductive cycle in pikeperch.

Morphometric characteristics, proximal composition and fatty acids profiles of eggs and larvae at hatching and mouth opening were studied as indicators for assessing the potential differences between spawns obtained from wild or captive females of pikeperch (Sander lucioperca). Captive females were acclimated to hatchery conditions and fed to satiation with chopped marine fish for more than a year, guaranteeing that captivity covered the full cycle of gonad maturation. The number of larvae obtained from the spawnings did not differ between captive and wild females, but both mean diameter and mean weight of the eggs collected from the captive females were significantly larger. Proximal compositions were similar in eggs and larvae from both groups, but fatty acids (FA) profiles showed striking differences: polyunsaturated FA consisted mainly of linoleic acid in eggs and larvae from wild females, while they consisted mainly of long chains of n-3 series in eggs and larvae from captive females. The diet of captive females largely modified the FA profiles of eggs and of hatching or mouth opening larvae, but did not affect the growth potential of early larvae, demonstrating that they were able to satisfy their early physiological needs regardless of the striking differences in their FA.
