Pear scab caused by Venturia nashicola has been reported as an important disease of pear resulting in lowering the quality of pear fruits. In this study, it was conducted to investigate the relationship between resistance of V. nashicola and mutation of β-tubulin gene and the fungicide resistance in field isolate group in benzimidazole fungicides. Responce of V. nashicola to carbendazim could be classified into 3 groups as sensitive that does not grow at all on PDA amended with 0.16 μg/ml of carbendazim, low resistance that could not grow in 4.0 μg/ml medium, and high resistance that can grow even at 100 μg/ml. Thirty isolates of V. nashicola collected from 3 regions as Wonju, Naju, and Okcheon were highly resistant to carbendazim. Analysis of the nucleotide sequence of β-tubulin gene of V. nashicola showed that there was no difference in the nucleotide sequence between the sensitive and the low-resistant isolate, but GAG at codon 198 (glutamic acid) was replaced with GCG (alanine) in the high-resistant isolate. Among 10 isolates obtained from the Okcheon, 5 isolates showed the substitution of glycine for glutamic acid, which were resistant to carbendazim, but more sensitive to the mixture of carbendazim and diethofencarb than others. Through these results, all isolates of V. nashicola isolated in pear orchard were found to be resistant to benzimidazoles. Also, mutants E198A and E198G at β-tubulin were found to be important mechanisms of V. nashicola resistance against benzimidazole fungicides.

A total of 23 isolates of Botrytis cinerea causing the grey mold were collected from infected ginseng in several fields of Korea. The sensitivity to carbendazim and the mixture of carbendazim plus diethofencarb was determined through a mycelial inhibition test on PDA amended with or without fungicides. B. cinerea isolates were classified as 3 phenotypes, which were the first phenotype resistant to both of carbendazim and the mixture (), the second one resistant to carbendazim and sensitive to the mixture (), and the last one sensitive to both of them (). Carbendazim resistance correlated with a single mutation -tubulin gene of B. cinerea amplified with primer pair tubkjhL and tubkjhR causing a change of glutamate to alanine at amino acid position 198. Furthermore, the substitution of valine for glutamate led the resistance to carbendazim and the mixture at the same position of amino acid. PCR-restriction fragment length polymorphism (PCR-RFLP) analysis using the restriction endonuclease, Tsp451 and BstUI allowed differentiation of the PCR fragment of -tubulin gene of isolates from that of and isolates. This method will aid in a fast detection of resistance of carbendazim and the mixture of carbendazim plus diethofencarb in B. cinerea in ginseng field.

Mixed-planting of two rice cultivars, HP (`Hopyeongbyeo`) and NP (`Nampyeongbyeo`), having a dissimilar susceptibility to rice blast was practiced for chemical-free control of rice blast in the field. The HP/NP combination was selected for applying under mechanized agricultural conditions. Because they have similar genetic characteristics such as seed germination and heading time, culm length, rice quality and size of rice grains except susceptibility to blast. Incidence of panicle blast was reduced 50.4 % compare with supposed blast incidence by HP/NP mixed-planting when the seeds of two cultivars were combined 1 to 1 as weight. Supposed blast incidence was estimated from reduction of rice blast caused by addition of a resistant cultivar NP. Race diversity of Magnaporthe oryzae was examined for correlation with control effect of HP/NP mixed-planting on rice blast. The population of dominant race KJ-101 was diminished and replaced with various co-existing races and eleven new races were appeared in mixed-planting plot. Total number of race isolated from mixed-planting plot was not largely different from mono-culture. However, detection frequency of the new race was increased and variation of the population size of each race was decreased in mixed-planting plots. It was shown that a biased community with a dominant race (KJ-101 or KI-181) was altered to a balanced one of coexisting races. From these results, it was supposed that the balanced diversity among co-existing races within a community might be correlated to control effect by HP/NP mixed-planting on rice blast. Further more, it should be studied that genetic characteristics of the individual race including a virulence on cv. HP and NP was examined for verifying a correlation of mixed-planting effect and race diversity.

In this experiment, 236 isolates of Botrytis cinerea isolated from the lesions of ginseng grey mold in 2005 and 2006 were examined for their sensitivity to fungicide inhibiting assembly. The baselines of fungicide resistance were determined as 10.0 and of values for carbendazim and the mixture of carbendazim and diethofencarb, respectively. The ratios of isolates resistant to carbendazim in 2005 and 2006 was investigated to be 87.6 and 96.6%, respectively. In the case of the mixture of carbendazim and diethofencarb, the ratio of the resistant isolates was 23.6% for 2005 and 24.5% for 2006. The ratio of the resistant isolates to the mixed fungicide was fluctuated according to regions where isolates of B. cinerea were obtained. In Yeoncheon of Gyeonggi Province, 4.3% of the isolates used in the experiment was resistant in 2005 and no resistant isolate was obtained in 2006. Among 5 regions, the ratio of resistant isolates was the highest as 70.0% in Yecheon of Gyeongbuk Province.

Three kinds of disease occurring on ginseng leaves, such as grey mold, Alternaria blight, and anthracnose, started at the beginning of June, July, and August, respectively. The disease incidence was rapidly increased from the beginning of rainy season. To develop the control system with fungicides, 6 fungicides were selected and applied on ginseng at the indicated time. Calculating the control value by using the area under the disease progressing curve (AUDPC), the control activities of the supervised control system with fungicides were 61.7, 78.8 and 70.5% against grey mold, Alternaria blight, and anthracnose, respectively. The application of the mixture of carbendazim and diethofencarb on first of June was very important in control system with fungicides. If it was deleted in control system, control value was decreased against grey mold. In the case of Alternaria blight and anthracnose, the application of difenoconazole on July 18, and trifloxystrobin on August 7 were indispensible. If difenoconazole and trifloxystrobin were not applied on July 18, and August 7, the control activities against Alternaria blight and anthracnose, respectively, were decreased to 28.9% and 44.4%.

Effects of fungicides on the mycelial growth of Botrytis cinerea isolated from ginseng leaves were investigated by an agar dilution method. By using a agar dilution method, it was investigated the effect of fungicides, procymidone, carbendazim and the mixture with both of carbendazim and diethofencarb, on the mycelial growth of Botrytis cinerea isolates, which were isolated from infected leaves of ginseng in 2005 and 2006. With MIC (minimum inhibiton concentration) of procymidone against B. cinerea, pathogens were divided into two groups. While one showed the low MIC between 0.8 and , the other showed higher MIC above . In terms of the inhibition ratio of mycelial growth at the indicated concentration of procymidone, isolates of B. cinerea were divided into three groups; the sensitive, the intermediate resistant, and the resistant group. Each group was differentiated by ; the sensitive group showed below , the intermediate resistant group between 2.0 to , and resistant group above . Compared with the ratio of resistant isolates of B. cinerea in 2005, the ratio in 2006 increased from 19.3% to 27.5%. Furthermore, the average value of them increased from in 2005 to in 2006. The ratio of isolates showing the multiple resistance between procymidone and carbendazim was 40.2%, whereas the ratio was 4.0% showing the multiple resistance in the mixture.

The anthracnose disease on pepper fruits in Korea was caused by Colletotrichum acutatum as well as C. gloeosporioides. Since C. acutatum showed less sensitivity to benomyl, it was analyzed whether the less sensitivity was given by the same mechanism for the fungicide resistance of C. gloeosporioides. The isolates of C. acutatum were less sensitive to the three benzimidazole fungicides tested, benomyl, carbendazim, and thiophanate-methyl. However, the of C. acutatum isolates were different from the resistant isolates of C. gloeosporioides in their response to diethofencarb, one of N-phenyl-carbamates; the former was still less sensitive to diethofencarb than the latter. The differences in the resistance mechanisms in two species were conspicuous in sequence analysis of the tub2 genes. The genes from C. acutatum did not show any non-synonymous base substitutions at the regions known to be correlated with the benzimidazole-resistance. All of these data may indicate that the less sensitivity of C. acutatum to benomyl is based on different mechanism(s) from that of C. gloeosporioides.

It was confirmed that anthracnose pathogen, Colletotrichum acutatum, could specifically grow on PDA amended with /ml of ampicillin and tetracycline, and 100 /ml of mixture with carbendazim and diethofencarb. There was a positive correlation between the number of colony enumerated on semi-selective media and the disease severity on pepper fruits caused by C. acutatum. Using semi-selective media for C. acutatum, the number of pathogen on soil and plant debris infected by anthracnose pathogen was investigated. In plant debris, the colony number of C. acutatum was more than in soil. For the identification of colony appeared on semi-selective media, 10 isolates were selected randomly. They were identified as C. acutatum through PCR using C. acutatum-specific primer.

The control activity of isoprothiolane and tricyclazole mixed with carbosulfan, and probenazole by the nursery treatment was performed against rice leaf and neck blast caused by Magnaporthe grisea. In the paddy field, three fungicides showed good activities against leaf blast 3 months after nursery treatment. Especially the activity of tricyclazole against leaf blast gradually increased by the laps of time to 85.5％, which was assessed at 6 September,2003. Although the control value of isoprotholane and tricyclazole mixed with carbosulfan against neck blast was 47.5％ and 61.1％, respectively, probenazole showed a very high activity against not only leaf blast but also neck blast, of which that was 91.2％. No phytotoxicity was observed in all the treatments after transplanting rice seedling in the paddy field. Based on these results, three systemic fungicides tested in this study showed such a good potential that they might be used to formulate the nursery treating granule.
