Chlamydia is a small gram-negative (G−) microorganism that can be dangerous to human and animals. In this study, we conducted a systematic review and meta-analysis of Chlamydia infection in swine in China. From PubMed, ScienceDirect, Chinese Web of knowledge (CNKI), VIP Chinese journal database, and Wanfang database, we collected a total of 72 publications reported in 1985–2020. The prevalence of Chlamydia was 22.48% in China. In the sampling year subgroup, the prevalence after 2011 was the highest (26.14%). In southern China, the prevalence was 30.97%. By contrast, the prevalence in northern China was only 10.79%. Also the difference was significant (p < 0.05). In the provincial level, Hubei had the highest rate of 36.23%. Boars had a higher prevalence (29.47%). The prevalence of Chlamydia detection in pigs with reproductive disorders (21.86%) was higher than that without reproductive disorders. Among the three age groups, finishing pigs (21.43%) had the highest prevalence. The prevalence in large-scale farmed pigs (28.58%) was the highest in the subgroup of feeding methods. The prevalence in farms was 24.29%, which was the highest in the survey areas. The prevalence in spring was the highest with 40.51%. Other methods had the highest prevalence (39.61%) than enzyme-linked immunosorbent assay (ELISA) and indirect hemagglutination assay. The prevalence of Chlamydia psittaci 18.41% was lower than the prevalence of Chlamydia abortus (41.35%). We also analyzed the impact of different climate factor subgroups (rainfall, temperature, and humidity) on the probability of pigs suffering from the disease. The results showed that Chlamydia was widespread in pigs in China. We suggest that we should strengthen the detection of Chlamydia in the semen of breeding pigs and pigs with reproductive disorders, and reasonably control the environment of large-scale pig farms, so as to reduce further infection of Chlamydia in pigs. Integrated analysis of accumulated data is an effective way to obtain reliable potential diagnostic molecular in gastric cancer (GC). The study aimed to identify potential lncRNAs associated with the pathogenesis and prognosis in GC. Raw noncoding RNA microarray data (GSE53137, GSE95667, and GSE111762) was downloaded from Gene Expression Omnibus (GEO) database. Differentially expressed genes between GC and adjacent normal gastric tissue samples were screened by an integrated analysis of multiple gene expression profile after gene reannotation and batch normalization. Differentially expressed genes were further confirmed by the cancer genome atlas (TCGA) database. Competing endogenous RNA (ceRNA) network, survival analysis, and gene set enrichment analysis (GSEA) were extensively applied to identify hub lncRNAs and discover potential biomarkers related to diagnosis and prognosis of GC. qPCR was applied to confirm hub lncRNA expression levels in GC tissues. In total, 17 integrated differential lncRNAs were obtained after intersections of differential genes between GEO and TCGA database. Four lncRNAs (HMGA1P4, UBE2Q1-AS1, MAGI2-AS3, MIR22HG) concentrated in ceRNA network were validated by qPCR in GC tissues, which were consistent with informatics results. The clinicopathological association revealed that four lncRNAs might be effective in GC progression. Further study revealed that GC patients with lower MAGI2-AS3 expression was evidently longer than those with higher MAGI2-AS3 expression (p = 0.015). Multivariate analysis revealed MAGI2-AS3 was independently associated with overall survival in GC. GSEA showed GC samples were differentially enriched in pyrimidine metabolism, RNA degradation, cell cycle, oxidative phosphorylation etc., and most significantly enriched in ribosome pathway in MAGI2-AS3 low expression phenotype. Four lncRNAs, including HMGA1P4, UBE2Q1-AS1, MAGI2-AS3, and MIR22HG may contribute to GC development, and MAGI2-AS3 might be associated with the prognosis of GC. [ABSTRACT FROM AUTHOR]

Brucellosis and chlamydiosis are important zoonotic diseases and bluetongue virus (BTV) is an arthropod-borne viral disease of ruminants. They are widely distributed around the world, cause large economic losses, and significant harmful effects on humans. However, epidemiological information relating to transmission from commercial sika deer in China is limited. Therefore, from 2016 to 2017, 458 sika deer blood samples were collected from three cities in Jilin Province in China. The Brucella antigen and specific antibodies to Chlamydia and BTV were examined using RT-PCR, indirect hemagglutination assay, and ELISA, respectively. The prevalence of Brucella was found to be 12.9% (59/458) and the seroprevalence of Chlamydia and BTV was 14.4% (66/458) and 17.0% (78/458), respectively. Seasonality was considered a risk factor for the presence of Brucella or BTV in sika deer and the region was considered a risk factor for Chlamydia infection. Aleutian mink disease (AD) is a chronic viral infection that causes autoimmune disorders in minks and presents a significant economic burden on mink farming. Despite the substantial challenges presented by AD, no effective vaccine is available and only partial protection has been achieved. We constructed a whole-gene nucleic acid vaccine from an isolated virulent Aleutian mink disease virus (ADV) strain (pcDNA3.1-ADV). Based on this whole-gene nucleic acid vaccine, we generated truncated mutant constructs by removing portions of the ADV VP2 gene using overlap extension polymerase chain reaction. pcDNA3.1-ADV-428 lacks nucleotides encoding VP2 amino acid residues 428-466, and pcDNA3.1-ADV-428-487 harbors additional deletion of nucleotides coding for VP2 amino acid residues 487-501. We also generated nucleic acid vaccines for the ADV NS1 gene, truncated ADV NS1 gene, ADV VS2 gene, and truncated ADV VS2 gene: pcDNA3.1-NS1, pcDNA3.1-NS1-D, pcDNA3.1-VP2, and pcDNA3.1-VP2-D, respectively. The immunogenicity of the seven DNA vaccines was confirmed by immunofluorescent evaluation. Sixty female minks were divided into 10 groups: seven groups were immunized with the DNA vaccines, one control group was injected with phosphate-buffered saline, one group was immunized with pcDNA3.1 empty vector, and one group was immunized with inactivated ADV-G virus. ADV antibody levels, percentage of CD8+ cells in blood, and levels of γ-globulin and circulating immune complexes in the serum were evaluated longitudinally over 36 weeks after ADV challenge. Minks that were immunized with the pcDNA3.1-ADV-428-487 nucleic acid vaccine produced ADV antibodies. After ADV challenge, the minks immunized with pcDNA3.1-ADV-428-487 nucleic acid vaccine had lower γ-globulin content and lower CIC in serum compared to other immunization groups. Although the pcDNA3.1-ADV-428-487 nucleic acid vaccine did not demonstrate complete protection against ADV, it demonstrated marked efficacy and could potentially be used as a vaccine to prevent losses in mink populations due to ADV. Discovery of effective means to vaccinate mink against ADV will not only improve overall health of mink populations but will also reduce the economic impact of ADV. [ABSTRACT FROM AUTHOR]
