Simple Summary: As a filter-feeding and sessile invertebrate living in estuaries and intertidal areas, Crassostrea gigas must cope with and adapt to a dynamic and changeable environment. Vibrio alginolyticus is a Gram-negative bacterium that is widespread in oceans and estuaries, and is one of the main Vibrio species that cause oyster disease. In this study, we used transcriptome sequencing to help us better understand how the giant oyster adapts to pathogen-rich environments. By focusing on the gills, which play a crucial role in the immune response, we aimed to shed light on the molecular processes underlying the interaction between the oyster and the pathogen. Over the years, oysters have faced recurring mass mortality issues during the summer breeding season, with Vibrio infection emerging as a significant contributing factor. Tubules of gill filaments were confirmed to be in the hematopoietic position in Crassostrea gigas, which produce hemocytes with immune defense capabilities. Additionally, the epithelial cells of oyster gills produce immune effectors to defend against pathogens. In light of this, we performed a transcriptome analysis of gill tissues obtained from C. gigas infected with Vibrio alginolyticus for 12 h and 48 h. Through this analysis, we identified 1024 differentially expressed genes (DEGs) at 12 h post-injection and 1079 DEGs at 48 h post-injection. Enrichment analysis of these DEGs revealed a significant association with immune-related Gene Ontology (GO) terms and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways. To further investigate the immune response, we constructed a protein–protein interaction (PPI) network using the DEGs enriched in immune-associated KEGG pathways. This network provided insights into the interactions and relationships among these genes, shedding light on the underlying mechanisms of the innate immune defense mechanism in oyster gills. To ensure the accuracy of our findings, we validated 16 key genes using quantitative RT-PCR. Overall, this study represents the first exploration of the innate immune defense mechanism in oyster gills using a PPI network approach. This paper discusses a type of mixed-delay quaternion-valued neural networks (QVNNs) under impulsive and stochastic disturbances. The considered QVNNs model are treated as a whole, rather than as complex-valued neural networks (NNs) or four real-valued NNs. Using the vector Lyapunov function method, some criteria are provided for securing the mean-square exponential stability of the mixed-delay QVNNs under impulsive and stochastic disturbances. Furthermore, a type of chaotic QVNNs under stochastic and impulsive disturbances is considered using a previously established stability analysis method. After the completion of designing the linear feedback control law, some sufficient conditions are obtained using the vector Lyapunov function method for determining the mean-square exponential synchronization of drive–response systems. Finally, two examples are provided to demonstrate the correctness and feasibility of the main findings and one example is provided to validate the use of QVNNs for image associative memory. Simple Summary: The health of Amphioctopus fangsiao, a species used in aquaculture or fish farming, can be greatly affected by infections. To understand how their immune system responds, we looked at the changes in their immune cells when exposed to substances that mimic these infections. By studying these responses, we were able to identify key parts of their immune system that change when faced with infection-causing threats. We also discovered important genes, including PKMYT1 (protein kinase, membrane associated tyrosine/threonine 1) and NAMPT (nicotinamide phosphoribosyltransferase), that play a crucial role in this response. Our study gives us a deeper understanding of the immune system of the Amphioctopus fangsiao. The primary influencer of aquaculture quality in Amphioctopus fangsiao is pathogen infection. Both lipopolysaccharides (LPS) and polyinosinic:polycytidylic acid (Poly I:C) are recognized by the pattern recognition receptor (PRR) within immune cells, a system that frequently serves to emulate pathogen invasion. Concurrently, we developed a Weighted Gene Co-expression Network Analysis (WGCNA) to identify key modules and genes. Further, we carried out Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses to investigate the primary modular functions. Co-expression network analyses unveiled a series of immune response processes following pathogen stress, identifying several key modules and hub genes, including PKMYT1 and NAMPT. The invaluable genetic resources provided by our results aid our understanding of the immune response in A. fangsiao hemolymph and will further our exploration of the molecular mechanisms of pathogen infection in mollusks. Background: The occurrence of compound hot extreme (CHE) events in China is increasingly frequent. This study aimed to investigate the association between CHEs and all-cause mortality in Shandong Province and to estimate the attributable excess deaths. Methods: We collected daily data on weather, air pollution, and all-cause mortality at the subdistrict level in Shandong Province from 2013 to 2018. A CHE was defined as both daily maximum and minimum temperatures being higher than their historical 90th percentiles during 2013–2018 hot seasons. A case time-series analysis with a distributed lagged non-linear model was applied to analyze the subdistrict-specific association between different hot extremes and mortality risk, which were then pooled at the province level using meta-analysis. Results: Hot nights (RR = 1.44, 95%CI: 1.35–1.53) and CHEs (RR = 1.77, 95%CI: 1.64–1.90) were significantly associated with an increased mortality risk. CHEs had a greater effect for females (RR = 1.99, 95%CI: 1.81–2.19) and the elderly (>74 years) (RR = 2.14, 95%CI: 1.93–2.38) than their counterparts, respectively. Cardiovascular and respiratory deaths were more susceptible to CHEs than other deaths. Each year, 4888 (95%CI: 4133–5811) excess deaths in Shandong Province were attributable to CHEs, accounting for 2.60% (95%CI: 2.20–3.10%) of all-cause deaths and equating to 50 (95%CI: 42–58) deaths per 1,000,000 residents. The CHE-related mortality burden varied across subdistricts, with the highest occurring in the southeastern area and the lowest occurring in the northeastern and southwestern regions. Conclusion: CHEs and hot nights were substantially associated with excess deaths in Shandong Province, especially for females, the elderly, and residents living in the southeastern area. In this study, diploid, triploid, and tetraploid Crassostrea gigas samples were subjected to gas chromatography and ion mobility (GC-IMS) to identify and analyze volatile compounds and flavor fingerprints under conditions of high-temperature incubation. The GC-IMS technology identified a total of 54 volatile components in C. gigas. The contents of 1-octen-3-ol, butyl pentanoate, p-methyl anisole, and 2-methyl-2-hepten-6-one in male oysters were significantly higher than in females, while the contents of phenylacetaldehyde, benzaldehyde, 2-ethyl-3-methylpyrazine, 2-ethylfuran, and 2,4-hexadienal in female oysters were significantly higher than in males. The contents of non-3-en-2-one-M and 1-pentanol in diploids were significantly higher than in triploids and tetraploids, while the content of 2,4-hexadienal in tetraploids was significantly higher than in diploids and tetraploids. The contents of ethyl acetate, ethyl-2-butenoate, and butanal in tetraploids were significantly higher than those in diploids and triploids. The results of a principal components analysis showed that different samples were relatively independently clustered, allowing the ability to distinguish different oyster samples. The chemical fingerprints of volatile compounds of C. gigas with different ploidy and gender under high-temperature incubation were established, and the volatile substance contours of C. gigas were visualized. Simple Summary: The cuttlefish, Sepia esculenta, is both a valuable economic resource and a creature of scientific interest. Our study investigated how changes in ocean salinity, a factor subject to environmental shifts, affect young, more responsive cuttlefish. We examined the gene expression of cuttlefish when exposed to lower levels of salinity. Using transcriptomic and enrichment analysis, we identified over a thousand genes that were differentially expressed under these circumstances. The images created from the results of these analyses can be used to visualize the content of the analyses. Interestingly, we discovered that low salinity could trigger harmful cell proliferation in juvenile cuttlefish, thus disrupting their normal functions. This information is valuable as it can inform potential measures to reduce the mortality rate of hatchling cuttlefish in artificially controlled conditions. Sepia esculenta is an economically important mollusk distributed in the coastal waters of China. Juveniles are more susceptible to stimulation by the external environment than mature individuals. The ocean salinity fluctuates due to environmental changes. However, there is a lack of research on the salinity adaptations of S. esculenta. Therefore, in this study, we investigated the differential expression of genes in S. esculenta larvae after stimulation by low salinity. RNA samples were sequenced and 1039 differentially expressed genes (DEGs) were identified. Then, enrichment analysis was performed using the Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) databases. Finally, a protein–protein interaction network (PPI) was constructed, and the functions of key genes in S. esculenta larvae after low-salinity stimulation were explored. We suggest that low salinity leads to an excess proliferation of cells in S. esculenta larvae that, in turn, affects normal physiological activities. As antimicrobial resistance continues to escalate, the exploration of alternative approaches to safeguard food safety becomes more crucial than ever. Phage endolysins are enzymes derived from phages that possess the ability to break down bacterial cell walls. They have emerged as promising antibacterial agents suitable for integration into food processing systems. Their application as food preservatives can effectively regulate pathogens, thus contributing to an overall improvement in food safety. This review summarizes the latest techniques considering endolysins' potential for food safety. These techniques include native and engineered endolysins for controlling bacterial contamination at different points within the food production chain. However, we find that characterizing endolysins through in vitro methods proves to be time consuming and resource intensive. Alternatively, the emergence of advanced high-throughput sequencing technology necessitates the creation of a robust computational framework to efficiently characterize recently identified endolysins, paving the way for future research. Machine learning encompasses potent tools capable of analyzing intricate datasets and pattern recognition. This study briefly reviewed the use of these industry 4.0 technologies for advancing the research in food industry. We aimed to provide current status of endolysins in food industry and new insights by implementing these industry 4.0 strategies revolutionizes endolysin development. Background: Emotional mimicry, a phenomenon frequently observed in our everyday interactions, is the act of replicating another individual's facial expression. The Emotion Mimicry in Context View and the Correction Hypothesis underscore the critical role of context and intention within emotional mimicry. Methods: In two distinct studies, participants were presented with facial expressions of models (happiness and anger) within various contexts (affiliative, distancing, and neutral). Concurrently, we recorded electromyography (EMG) to index emotional mimicry, while participants explicitly rated the models' intentions. Results: We observed context swiftly influences emotional mimicry within 500 ms, notably when the intentions of contexts are opposing to the intentions of facial expressions, leading to weakened muscle responses and diminished perceived intention. Furthermore, a notable correlation was discovered in the mimicry of angry faces; the more distancing the context, the stronger the corrugator supercilii (CS) muscle activity after context processing. Conclusions: First, emotional mimicry should not be simply viewed as an output corresponding to the expresser's facial expressions but the dynamic process involving the active participation of the observer. Plastic products are widely used due to their lightness, durability, low cost, and malleability, and their threat to the environment is becoming increasingly apparent. However, the detailed mechanism of such an effect is unclear. Due to improper treatment, microplastic contamination is very much studied in the areas around sewage outfalls or some plastic manufacturers, and its content is high. To investigate the ecological impact of microplastics in this particular area, we evaluated the toxic effect of polystyrene microplastics (PS-MPs) on the microalgae Scenedesmus obliquus (S. obliquus). In particular, the cell growth, photosynthesis, antioxidant enzyme activity, membrane permeability, and morphology of the microalgae in the presence of different PS-MP concentrations were monitored. These new data revealed an increase in microalgae cell growth as an early response to low-dose PS-MP exposure with a maximum inhabitation rate of −3.33%. In contrast, a higher concentration of PS-MP solution leads to a significant inhibition within the test concentration, with a maximum inhabitation rate of 43.62%. Meanwhile, a decreased photosynthesis activity of S. obliquus was observed during the exposure. Moreover, PS-MPs induced oxidative damage to the algae cells, exhibiting decreased antioxidant activities and enhanced lipid peroxidation reactions, as evidenced by the reduction in the superoxide dismutase (SOD) level and increased malondialdehyde (MDA) content, as well as damaged cell membrane and soluble protein. Collectively, our study systematically investigates the toxicity of microplastics on microalgae, providing a deep insight into the possible influences of plastic pollution on the whole food web in the aquatic ecosystem. Artificial modification of Bacillus thuringiensis (Bt) proteins can effectively improve their resistance to target pests, but the effect of such modification on the diversity of rhizosphere microorganisms remains unclear. Transgenic maize 2A-7 contains two artificially modified Bt proteins, mCry1Ab and mCry2Ab. These proteins can enter soil and pose a potential threat to soil microbial diversity. To assess their impacts on rhizosphere bacteria communities, the contents of the two Bt proteins and changes in bacterial community diversity in the rhizosphere soils of transgenic maize 2A-7 and its control variety were analyzed at different growth stages in 2020. The results showed that the two Bt proteins were detected at low levels in the rhizosphere soils of 2A-7 plants. No significant differences in soil bacterial diversity were detected between 2A-7 and its control variety at any of the growth stages. Bioinformatics analysis indicated that the growth stage, rather than the cultivar, was the main factor causing changes in bacterial communities. This research provides valuable data for understanding the impact of Bt crops on the soil microbiome, and establishes a theoretical basis for evaluation of their safety. [ABSTRACT FROM AUTHOR]

Pancreatic cancer is the third leading cause of cancer-related death in the United States. Pancreatic ductal adenocarcinoma (PDAC) is the major form of pancreatic cancer with the worst outcomes. Early detection is key to improving the overall survival rate of PDAC patients. Recent studies have demonstrated that microRNA (miRNA) signatures in plasma small extracellular vesicles (EVs) are potential biomarkers for the early detection of PDAC. However, published results are inconsistent due to the heterogeneity of plasma small EVs and the methods used for small EV isolation. We have recently refined the process of plasma small EV isolation using double filtration and ultracentrifugation. In the present study, we applied this protocol and analyzed plasma small EV miRNA signatures by small RNA sequencing and quantitative RT-PCR in a pilot cohort, consisting of patients with early-stage PDAC, and age- and gender-matched healthy subjects (n = 20). We found, via small RNA sequencing, that there are several miRNAs enriched in plasma small EVs of PDAC patients, and the levels of miR-18a and miR-106a were confirmed by quantitative RT-PCR to be significantly elevated in patients with early-stage PDAC compared with age- and gender-matched healthy subjects. Furthermore, using an immunoaffinity-based plasma small EV isolation approach, we confirmed that the levels of miR-18a and miR-106a in plasma small EVs were significantly higher in PDAC patients versus the healthy subjects. The leakage prediction calculation method for dynamic seal rings in underground equipment is presented in this paper. The framework of the method is given. The leakage prediction model is built. The non-Newtonian fluid interface element is brought in. The leakage prediction calculation method was developed based on the thermal–structural coupled method and the fluid–structural coupled method. A test is performed to validate the proposed method. It is proved that the film thickness of an O-ring made of nitrile rubber in pulling-in travel is thicker than that in pushing-out travel. The leakage of an O-ring made of fluororubber is larger than that of an O-ring made of nitrile rubber in the same environmental condition. The presented method is useful for predicting the sealing ability of dynamic seal rings in underground equipment. Honeycomb structure composites are taking an increasing proportion in aircraft manufacturing because of their high strength-to-weight ratio, good fatigue resistance, and low manufacturing cost. However, the hollow structure is very prone to liquid ingress. Here, we report a fast and automatic classification approach for water, alcohol, and oil filled in glass fiber reinforced polymer (GFRP) honeycomb structures through terahertz time-domain spectroscopy (THz-TDS). We propose an improved one-dimensional convolutional neural network (1D-CNN) model, and compared it with long short-term memory (LSTM) and ordinary 1D-CNN models, which are classification networks based on one dimension sequenced signals. The automated liquid classification results show that the LSTM model has the best performance for the time-domain signals, while the improved 1D-CNN model performed best for the frequency-domain signals. The oncogenic role of Ladinin-1 (LAD1), an anchoring filament protein, is largely unknown. In this study, we conducted a series of studies on the oncogenic role of LAD1 in lung adenocarcinoma (LUAD). Firstly, we analyzed the aberrant expression of LAD1 in LUAD and its correlation with patient survival, tumor immune infiltration, and the activation of cancer signaling pathways. Furthermore, the relationship between LAD1 expression and K-Ras and EGF signaling activation, tumor cell proliferation, migration, and colony formation was studied by gene knockout/knockout methods. We found that LAD1 was frequently overexpressed in LUAD, and high LAD1 expression predicts a poor prognosis. LAD1 exhibits promoter hypomethylation in LUAD, which may contribute to its mRNA upregulation. Single-sample gene set enrichment analysis (ssGSEA) showed that acquired immunity was negatively correlated with LAD1 expression, which was verified by the downregulated GO terms of "Immunoglobulin receptor binding" and "Immunoglobulin complex circulating" in the LAD1 high-expression group through Gene Set Variation Analysis (GSVA). Notably, the Ras-dependent signature was the most activated signaling in the LAD1 high-expression group, and the phosphorylation of downstream effectors, such as ERK and c-jun, was strongly inhibited by LAD1 deficiency. Moreover, we demonstrated that LAD1 depletion significantly inhibited the proliferation, migration, and cell-cycle progression of LUAD cells and promoted sensitivity to Gefitinib, K-Ras inhibitor, and paclitaxel treatments. We also confirmed that LAD1 deficiency remarkably retarded tumor growth in the xenograft model. Background: This study aimed to explore the correlation between body mass index (BMI) and dyslipidemia and the optimal cut-off point for BMI to distinguish the risk of dyslipidemia in lactating women. Methods: A total of 2295 lactating women subjects were included in this study, all within 2 years postpartum. All samples were from "China Children and Lactating Mothers Nutritional Health Surveillance (2016–2017)". BMI, blood lipids, demographic information, lifestyle habits, and other serum indicators were obtained in this survey. Generalized linear model, logistic regression, restricted cubic spline (RCS) and ROC curve analysis were used to evaluate the relationship among BMI, blood lipids, and dyslipidemia. Results: BMI in lactating women was positively correlated with total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), but negatively correlated with high-density lipoproteincholesterol (HDL-C) (p < 0.05). Higher BMI in lactating women was associated with higher ORs of dyslipidemia (hypercholesterolemia, hypertriglyceridemia, high-LDL-cholesterolemia, low HDL-cholesterolemia) (p < 0.05). These associations were stable across age groups, breastfeeding child age (months), parity, physical activity level, fasting plasma glucose (FPG), and hemoglobin. These factors did not interact with this relationship (p > 0.05). The optimal cut-off point for BMI was 24.85 kg/m2 determined by using ROC analysis, which can distinguish the risk of dyslipidemia. Conclusions: BMI was positively correlated with risk of dyslipidemia. Maintaining an ideal weight may prevent dyslipidemia in lactating women, and BMI is recommended to be controlled below 24.85 kg/m2. [ABSTRACT FROM AUTHOR]

During the process of gas production in high-pressure, high-temperature (HPHT) gas wells, the choke valve, as the most vital component of the surface control equipment, plays a significant role in regulating the output and reducing the fluid pressure to ensure the safety of surface gathering and transportation equipment. High-pressure, high-velocity fluid flow and solid-phase particles cause deterioration of the choke valve. With the enhancement of intelligent and digital oilfields, conventional choke valves have been progressively replaced by electric choke valves. Due to the complex structure of the throttle valve, the flow path and the velocity state of the fluid in the throttle valve, and the distribution law of the erosion fraction are quite distinctive from those in the ordinary throttle valve, meriting further research. In this paper, a simulation of computational fluid dynamics (CFD) was conducted to determine the effects of the pressure distribution, fluid state, divergent particle sizes, and sand volume on the erosion rate of the choke valve. Under various valve openings, the fluid state and the location of high-risk points can be ascertained. The large particle size (diameter greater than 6 mm) of sand and gravel is convenient for causing concentrated erosion in the position of the valve hole, which induces the channel diameter to expand. Fine silt sand (diameter from 0.1 mm to 1 mm) gives rise to relatively uniform abrasion to the choke's current-facing surface. This study can optimize the layout of the choke valve and reduce the cost and number of switching wells, thereby decreasing the frequency of maintenance and the pressure fluctuation's effect on the formation. It is well known that Zipf's rank-size law is powerful to investigate the rank-size distribution of tourist flow. Recently, widespread attention has been drawn to investigating the impacts of COVID-19 on tourism for its sustainability. However, little is known about the impacts of COVID-19 on the rank-size distribution of regional tourism central places. Taking Guangdong-Hong Kong-Macao Greater Bay Area as a research case, this article aims to examine the fractal characteristics of the rank-size distribution of regional tourism central places, revealing the impacts which COVID-19 has on the rank-size distribution of regional tourism central places. Based on the census data over the years from 2008 to 2021, this paper reveals that before COVID-19, the rank-size distribution of the tourism central places in Guangdong-Hong Kong-Macao Greater Bay Area appears monofractal, and the difference in the size of the tourism central places has a tendency to gradually decrease; in 2020, with the outbreak of COVID-19, the characteristic of the rank-size distribution shows that the original monofractal is broken into multifractal; in 2021, with COVID-19 becoming under control, the structure of tourism size distribution, changes into bifractal based on the original multifractal, showing that the rank-size distribution of tourism central places in Guangdong-Hong Kong-Macao Greater Bay Area becomes more ideal and the tourism order becomes better than the last year. The results obtained not only fill in the gap about the impacts of COVID-19 on tourism size distribution, but also contribute to the application of fractal theory to tourism size distribution. How to use bioinformatics methods to quickly and accurately locate the effective targets of traditional Chinese medicine monomer (TCM) is still an urgent problem needing to be solved. Here, we used high-throughput sequencing to identify the genes that were up-regulated after cells were treated with TCM monomers and used bioinformatics methods to analyze which transcription factors activated these genes. Then, the binding proteins of these transcription factors were analyzed and cross-analyzed with the docking proteins predicted by small molecule reverse docking software to quickly and accurately determine the monomer's targets. Followeding this method, we predicted that the TCM monomer Daphnoretin (DT) directly binds to JAK2 with a binding energy of −5.43 kcal/mol, and activates the JAK2/STAT3 signaling transduction pathway. Subsequent Western blotting and in vitro binding and kinase experiments further validated our bioinformatics predictions. Our method provides a new approach for quickly and accurately locating the effective targets of TCM monomers, and we also have discovered for the first time that TCM monomer DT is an agonist of JAK2. Transgenic maize 2A-7 expressing mCry1Ab and mCry2Ab has excellent resistance to lepidopteran pests. Previous studies have investigated the effects of several Bacillus thuringiensis (Bt) proteins on the soil. However, the effects of artificially modified Bt proteins on soil ecosystems are still unclear. To evaluate the effects of transgenic maize 2A-7 on soil, the physicochemical properties, enzyme activities and functional diversities of the microbial communities in rhizosphere soils from 2A-7 and its near-isogenic non-transgenic control Dongdan 6531 were analyzed at different developmental stages under field conditions. The alteration of six physicochemical properties (pH, total nitrogen, total phosphorus, organic matter, available phosphorus and alkali-hydrolyzed nitrogen) and six functional enzymes (catalase, alkaline phosphatase, sucrase, acid phosphatase, urease and alkaline protease) activities in the rhizosphere soils between the two maize cultivars were drastically correlated with plant growth stage, but not affected by the artificially modified Bt transgenes. An analysis of time-course Biolog data revealed that the functional diversity of microbial communities in the rhizosphere soil of 2A-7 and its control were similar at each developmental stage. The results suggest that transgenic maize 2A-7 has no significant impact on the soil ecosystem and provide valuable information on scientific safety assessments of 2A-7 and its commercial applications. Simple Summary: Feather is one of the most valuable and economical products in goose farming and plays a crucial physiological role in birds. For avian biology and the poultry industry, it is essential to comprehend and regulate how skin and feather follicles develop during embryogenesis. This study showed that several key regulatory genes (FOXO3, CTGF, and PTCH1, among others) and miRNAs (miR-144-y) participated in the developmental process of the skin and feather follicles in Zhedong white goose. Our findings are particularly important because they will serve as a valuable resource for upcoming studies on down feathers in agricultural economic growth regarding complex molecular mechanisms and breeding techniques. Skin and feather follicle development are essential processes for goose embryonic growth. Transcriptome and next-generation sequencing (NGS) network analyses were performed to improve the genome of Zhedong White goose and discover the critical genes, miRNAs, and pathways involved in goose skin and feather follicle morphogenesis. Sequencing output generated 6,002,591,668 to 8,675,720,319 clean reads from fifteen libraries. There were 1234, 3024, 4416, and 5326 different genes showing differential expression in four stages, E10 vs. E13, E10 vs. E18, E10 vs. E23, and E10 vs. E28, respectively. The differentially expressed genes (DEGs) were found to be implicated in multiple biological processes and pathways associated with feather growth and development, such as the Wnt signaling pathway, cell adhesion molecules, ECM–receptor interaction signaling pathways, and cell cycle and DNA replication pathways, according to functional analysis. In total, 8276 DEGs were assembled into twenty gene profiles with diverse expression patterns. The reliability of transcriptome results was verified by real-time quantitative PCR by selecting seven DEGs and five miRNAs. The localization of forkhead box O3 (FOXO3), connective tissue growth factor (CTGF), protein parched homolog1 (PTCH1), and miR-144-y by in situ hybridization showed spatial-temporal expression patterns and that FOXO3 and miR-144-y have an antagonistic targeting relationship. The correlation coefficient of FOXO3 and miR-144-y was -0.948, showing a strong negative correlation. Dual-luciferase reporter assay results demonstrated that miR-144-y could bind to the expected location to suppress the expression of FOXO3, which supports that there is a targeting relationship between them. The detections in this report will provide critical insight into the complex molecular mechanisms and breeding practices underlying the developmental characteristics of skin and feather follicles in Zhedong white geese. Simple Summary: Plumage color is one of the economic characteristics of poultry. The most common plumage colors in adult geese are white and grey, so research on goose plumage color is relatively scarce compared with the molecular basis of research on chicken plumage color inheritance. Nevertheless, whether the formation of a black phenotype on the Holdobaggy geese newborn dorsal plumage color is associated with gender needs further profound study. Our study researched the gender identification of the newborn Holdobaggy geese based on plumage color and the expression levels of relative genes for melanin, which offers a theoretical basis for gender identification. In production practice, we have found that the gray and black down on the backs of the Holdobaggy goslings is usually darker in females than in males. Melanin is the key pigment affecting the color of poultry plumage. Therefore, to determine whether the darkness of the dorsal plumage of the Holdobaggy goslings is related to sex, we study the melanin in the feather follicles of the dorsal skin during the embryonic period. The feather follicle structure and melanin distribution on the dorsal surface of the goose embryo is observed by HE staining and melanin-specific staining. The melanin content in the feather follicles of the dorsal skin of goslings is determined by ELISA. The results showed that the melanin content is higher in female geese than in males (p < 0.05). In addition, we also analyze the mRNA and protein expression levels of melanin-related genes (TYRP1 and ASIP) by quantitative real-time PCR and Western blotting analysis. The results show that the mRNA expression level of TYRP1 is significantly higher in the females' dorsal skin feather follicles (p < 0.05), while the mRNA expression level of ASIP is significantly higher in the dorsal skin feather follicles of male geese (p < 0.05). In conclusion, the difference between males and females in the color of the black feathers on the dorsal track of the Holdobaggy goslings is verified, and it is feasible to identify the sex by the initial plumage color. Featured Application: Energy storage providing auxiliary service Energy storage providing auxiliary service at the user-side has broad prospects in support of national polices. Three auxiliary services are selected as the application scene for energy storage participating in demand management, peak shaving and demand response. Considering the time value of funds, the user-side energy storage economy model is built. The model comprehensively considers the delayed transformation income, the government subsidy income, the auxiliary service income and the whole-life-cycle cost factor. According to the cost and benefit analysis, an energy storage optimization configuration model is proposed. The model takes maximum revenue of industrial user in energy storage's whole-life-cycle as the objective function. Then, the Cplex solver is employed to solve the model. In addition, four indexes are utilized to evaluate the financial effect brought by the user-side energy storage. Finally, the revenue and configuration results of the four types of battery energy storage are calculated to verify the validity of the proposed model. In comparison to the value of evaluation index, planning suggestions are provided for the user-side energy storage providing different auxiliary services. Simple Summary: In this study, we found that the novel small molecule kinase inhibitor DCLK1-IN-1 not only inhibited DCLK1 phosphorylation, stemness, and EMT-related properties of RCC cells but also revealed its potential as an immunotherapy agent and potential combination therapy with anti-PD1 against RCC in immune co-culture experiments. The approval of immune checkpoint inhibitors has expanded treatment options for renal cell carcinoma (RCC), but new therapies that target RCC stemness and promote anti-tumor immunity are needed. Previous findings demonstrate that doublecortin-like kinase 1 (DCLK1) regulates stemness and is associated with RCC disease progression. Herein, we demonstrate that small-molecule kinase inhibitor DCLK1-IN-1 strongly inhibits DCLK1 phosphorylation and downregulates pluripotency factors and cancer stem cell (CSC) or epithelial-mesenchymal transition (EMT)-associated markers including c-MET, c-MYC, and N-Cadherin in RCC cell lines. Functionally, DCLK1-IN-1 treatment resulted in significantly reduced colony formation, migration, and invasion. Additionally, assays using floating or Matrigel spheroid protocols demonstrated potent inhibition of stemness. An analysis of clinical populations showed that DCLK1 predicts RCC survival and that its expression is correlated with reduced CD8+ cytotoxic T-cell infiltration and increases in M2 immunosuppressive macrophage populations. The treatment of RCC cells with DCLK1-IN-1 significantly reduced the expression of immune checkpoint ligand PD-L1, and co-culture assays using peripheral blood monocytes (PBMCs) or T-cell expanded PBMCs demonstrated a significant increase in immune-mediated cytotoxicity alone or in combination with anti-PD1 therapy. Together, these findings demonstrate broad susceptibility to DCLK1 kinase inhibition in RCC using DCLK1-IN-1 and provide the first direct evidence for DCLK1-IN-1 as an immuno-oncology agent. Heavy metal contamination affects lives with concomitant environmental pollution, and seaweed has emerged as a remedy with the ability to save the ecosystem, due to its eco-friendliness, affordability, availability, and effective metal ion removal rate. Heavy metals are intrinsic toxicants that are known to induce damage to multiple organs, especially when subjected to excess exposure. With respect to these growing concerns, this review presents the preferred sorption material among the many natural sorption materials. The use of seaweeds to treat contaminated solutions has demonstrated outstanding results when compared to other materials. The sorption of metal ions using dead seaweed biomass offers a comparative advantage over other natural sorption materials. This article summarizes the impact of heavy metals on the environment, and why dead seaweed biomass is regarded as the leading remediation material among the available materials. Metal organic frameworks (MOFs) have been rapidly developed in the application of electrode materials due to their controllable morphology and ultra-high porosity. In this research, flower-like layered nickel-based bimetallic MOFs microspheres with different metal central ions were synthesized by solvothermal method. Compared with Ni-MOFs, the optimization of the specific capacitance of NiCo-MOFs and NiMn-MOFs was been confirmed. For example, the specific capacitance of NiCo-MOFs can reach 882 F·g−1 at 0.5 A·g−1 while maintaining satisfactory cycle life (the specific capacity remains 90.1% of the initial value after 3000 charge-discharge cycles at 5 A·g−1). In addition, the NiCo-MOFs//AC HSCs, which are composed of NiCo-MOFs and activated carbon (AC), achieved a maximum energy density of 18.33 Wh·kg−1 at a power density of 400 W·kg−1, and showed satisfactory cycle life (82.4% after 3000 cycles). These outstanding electrochemical properties can be ascribed to the synergistic effect between metal ions, the optimized conductivity, and the unique layered stacked flower structure, which provides a smooth transmission channel for electrons/ions. Functional characterization of plant agrichemical transporters provided an opportunity to discover molecules that have a high mobility in plants and have the potential to increase the amount of pesticides reaching damage sites. Agrobacterium-mediated transient expression in tobacco is simple and fast, and its protein expression efficiency is high; this system is generally used to mediate heterologous gene expression. In this article, transient expression of tobacco nicotine uptake permease (NtNUP1) and rice polyamine uptake transporter 1 (OsPUT1) in Nicotiana benthamiana was performed to investigate whether this system is useful as a platform for studying the interactions between plant transporters and pesticides. The results showed that NtNUP1 increases nicotine uptake in N. benthamiana foliar discs and protoplasts, indicating that this transient gene expression system is feasible for studying gene function. Moreover, yeast expression of OsPUT1 apparently increases methomyl uptake. Overall, this method of constructing a transient gene expression system is useful for improving the efficiency of analyzing the functions of plant heterologous transporter-encoding genes and revealed that this system can be further used to study the functions of transporters and pesticides, especially their interactions. To coordinate multiple power sources properly, this paper presents an optimal control strategy for a fuel cell/battery/supercapacitor light rail vehicle. The proposed strategy, which uses the firefly algorithm to optimize the equivalent consumption minimization strategy, improves the drawback that the conventional equivalent consumption minimization strategy takes insufficient account of the global performance for the vehicle. Moreover, the strategy considers the difference between the two sets of optimized variables. The optimization objective is to minimize the daily operating cost of the vehicle, which includes the total fuel consumption, initial investment, and cycling costs of power sources. The selected case study is a 100% low-floor light rail vehicle. The advantages of the proposed strategy are investigated by comparison with the operating mode control, firefly algorithm-based operating mode control, and equivalent consumption minimization strategy. In contrast to other methods, the proposed strategy shows cost reductions of up to 39.62% (from operating mode control), 18.28% (from firefly algorithm-based operating mode control), and 13.81% (from equivalent consumption minimization strategy). In this study, ethylene diamine phosphate (EDP) was employed as an activator to improve the sulfidization and flotation of chrysocolla. The micro-flotation experiment results indicated that EDP could greatly increase the flotation recovery of chrysocolla. BET and TEM measurements confirmed that the porous structure of the chrysocolla’s surface would lead to large amounts of the reagents. ICP-AES analysis revealed that the addition of EDP caused more active Cu sites formed on the chrysocolla’s surface, enhancing the adsorption of S2− on its surface. Meanwhile, a redox reaction occurred between the S2− and [Cu(en)2]2+ ions causing the Cu, S, and N in the solution to counter-adsorb onto the chrysocolla’s surface by forming new complexes. During this reaction, the Cu(II) species reduced to Cu(I) species and the sulfide ions in the form of S2−, S22−, Sn2−, and SO42− appeared on the mineral surface. The zeta potential measurements further revealed that the EDP-activated chrysocolla surfaces adsorbed more sulfide species and xanthate species, thereby improving the floatability of the chrysocolla.
