Several environmental stresses, including biotic and abiotic factors, adversely affect the growth and development of crops, thereby lowering their yield. However, abiotic factors, e.g., drought, salinity, cold, heat, ultraviolet radiations (UVr), reactive oxygen species (ROS), trace metals (TM), and soil pH, are extremely destructive and decrease crop yield worldwide. It is expected that more than 50% of crop production losses are due to abiotic stresses. Moreover, these factors are responsible for physiological and biochemical changes in plants. The response of different plant species to such stresses is a complex phenomenon with individual features for several species. In addition, it has been shown that abiotic factors stimulate multi-gene responses by making modifications in the accumulation of the primary and secondary metabolites. Metabolomics is a promising way to interpret biotic and abiotic stress tolerance in plants. The study of metabolic profiling revealed different types of metabolites, e.g., amino acids, carbohydrates, phenols, polyamines, terpenes, etc, which are accumulated in plants. Among all, primary metabolites, such as amino acids, carbohydrates, lipids polyamines, and glycine betaine, are considered the major contributing factors that work as osmolytes and osmoprotectants for plants from various environmental stress factors. In contrast, plant-derived secondary metabolites, e.g., phenolics, terpenoids, and nitrogen-containing compounds (alkaloids), have no direct role in the growth and development of plants. Nevertheless, such metabolites could play a significant role as a defense by protecting plants from biotic factors such as herbivores, insects, and pathogens. In addition, they can enhance the resistance against abiotic factors. Therefore, metabolomics practices are becoming essential and influential in plants by identifying different phytochemicals that are part of the acclimation responses to various stimuli. Hence, an accurate metabolome analysis is important to understand the basics of stress physiology and biochemistry. This review provides insight into the current information related to the impact of biotic and abiotic factors on variations of various sets of metabolite levels and explores how primary and secondary metabolites help plants in response to these stresses. Simple Summary: This review focuses on plant growth and development harmed by abiotic stress, primarily salt stress. Salt stress raises the intracellular osmotic pressure, leading to hazardous sodium buildup. Plants react to salt stress signals by regulating ion homeostasis, activating the osmotic stress pathway, modulating plant hormone signaling, and altering cytoskeleton dynamics and cell wall composition. Understanding the processes underlying these physiological and biochemical responses to salt stress could lead to more effective agricultural crop yield measures. In this review, researchers outline recent advances in plant salt stress control. The study of plant salt tolerance processes is essential, both theoretically and practically, to improve agricultural output, produce novel salt-tolerant cultivars, and make full use of saline soil. Based on past research, this paper discusses the adverse effects of salt stress on plants, including photosynthesis suppression, ion homeostasis disturbance, and membrane peroxidation. The authors have also covered the physiological mechanisms of salt tolerance, such as the scavenging of reactive oxygen species and osmotic adjustment. This study further identifies specific salt stress-responsive mechanisms linked to physiological systems. Based on previous studies, this article reviews the current methodologies and techniques for improving plant salt tolerance. Overall, it is hoped that the above-mentioned points will impart helpful background information for future agricultural and crop plant production. Salinity is significant abiotic stress that affects the majority of agricultural, irrigated, and cultivated land. It is an issue of global importance, causing many socio-economic problems. Salt stress mainly occurs due to two factors: (1) soil type and (2) irrigation water. It is a major environmental constraint, limiting crop growth, plant productivity, and agricultural yield. Soil salinity is a major problem that considerably distorts ecological habitats in arid and semi-arid regions. Excess salts in the soil affect plant nutrient uptake and osmotic balance, leading to osmotic and ionic stress. Plant adaptation or tolerance to salinity stress involves complex physiological traits, metabolic pathways, the production of enzymes, compatible solutes, metabolites, and molecular or genetic networks. Different plant species have different salt overly sensitive pathways and high-affinity K+ channel transporters that maintain ion homeostasis. However, little progress has been made in developing salt-tolerant crop varieties using different breeding approaches. This review highlights the interlinking of plant morpho-physiological, molecular, biochemical, and genetic approaches to produce salt-tolerant plant species. Most of the research emphasizes the significance of plant growth-promoting rhizobacteria in protecting plants from biotic and abiotic stressors. Plant growth, survival, and yield can be stabilized by utilizing this knowledge using different breeding and agronomical techniques. This information marks existing research areas and future gaps that require more attention to reveal new salt tolerance determinants in plants—in the future, creating genetically modified plants could help increase crop growth and the toleration of saline environments. Adaptation to unfavorable abiotic stresses is one of the key processes in the evolution of plants. Calcium (Ca2+) signaling is characterized by the spatiotemporal pattern of Ca2+ distribution and the activities of multi-domain proteins in integrating environmental stimuli and cellular responses, which are crucial early events in abiotic stress responses in plants. However, a comprehensive summary and explanation for evolutionary and functional synergies in Ca2+ signaling remains elusive in green plants. We review mechanisms of Ca2+ membrane transporters and intracellular Ca2+ sensors with evolutionary imprinting and structural clues. These may provide molecular and bioinformatics insights for the functional analysis of some non-model species in the evolutionarily important green plant lineages. We summarize the chronological order, spatial location, and characteristics of Ca2+ functional proteins. Furthermore, we highlight the integral functions of calcium-signaling components in various nodes of the Ca2+ signaling pathway through conserved or variant evolutionary processes. These ultimately bridge the Ca2+ cascade reactions into regulatory networks, particularly in the hormonal signaling pathways. In summary, this review provides new perspectives towards a better understanding of the evolution, interaction and integration of Ca2+ signaling components in green plants, which is likely to benefit future research in agriculture, evolutionary biology, ecology and the environment. Drought and salinity are two of the most frequently co-occurring abiotic stresses. Despite recent advances in the elucidation of the effects of these stresses individually during the vegetative stage of plants, significant gaps exist in our understanding of the combined effects of these two frequently co-occurring stresses. Here, Tibetan wild barley XZ5 (drought tolerant), XZ16 (salt tolerant), and cultivated barley cv. CM72 (salt tolerant) were subjected to drought (D), salinity (S), or a combination of both treatments (D+S). Protein synthesis is one of the primary activities of the green part of the plant. Therefore, leaf tissue is an important parameter to evaluate drought and salinity stress conditions. Sixty differentially expressed proteins were identified by mass spectrometry (MALDI-TOF/TOF) and classified into 9 biological processes based on Gene Ontology annotation. Among them, 21 proteins were found to be expressed under drought or salinity alone; however, under D+S, 7 proteins, including S-adenosylmethionine synthetase 3 (SAMS3), were exclusively upregulated in drought-tolerant XZ5 but not in CM72. HvSAMS3 carries both N-terminal and central domains compared with Arabidopsis and activates the expression of several ethylene (ET)-responsive transcription factors. HvSAMS3 is mainly expressed in the roots and stems, and HvSAMS3 is a secretory protein located in the cell membrane and cytoplasm. Barley stripe mosaic virus-based virus-induced gene silencing (BSMV-VIGS) of HvSAMS3 in XZ5 severely compromised its tolerance to D+S and significantly reduced plant growth and K+ uptake. The reduced tolerance to the combined stress was associated with the inhibition of polyamines such as spermidine and spermine, polyamine oxidase, ethylene, biotin, and antioxidant enzyme activities. Furthermore, the exogenous application of ethylene and biotin improved the tolerance to D+S in BSMV-VIGS:HvSAMS3-inoculated plants. Gleditsia sinensis Lam. is a woody species that can tolerate various drought conditions and has been widely used in all aspects of life, including medicine, food, cleaning products, and landscaping. However, few reports have focused on the regulatory mechanism of the drought response in G. sinensis. To understand the metabolic basis of the Gleditsia sinensis drought response, different tissues were subjected to a rehydration/dehydration treatment and subsequently analyzed using untargeted and targeted metabolomics profiling depending on gas chromatography-mass spectrometry (GC-MS) and liquid chromatography-tandem mass (LC-MS) analytical platforms, respectively. Eight sugars, twelve amino acids, and twenty phenolic compounds were characterized. Metabolites showing a significant increase or decrease under drought stress were considered to be the key metabolites of interest for a better understanding of the drought tolerance mechanisms. The GC-MS-identified compounds were shown to undergo tissue-specific regulation in response to drought stress. Moreover, the C6C3C6 and C6C3 structures were identified by LC-MS as phenolic metabolites, which revealed their drought-response association. Significant physiological parameters were measured, including overall plant development, and the results showed that antioxidant systems could not be completely restored, but photosynthetic parameters could be recovered. Rice (Oryza sativa L.) is one of the most important staple food crops worldwide, while its growth and productivity are threatened by various abiotic stresses, especially salt stress. Unraveling how rice adapts to salt stress at the transcription level is vital. It can provide valuable information on enhancing the salt stress tolerance performance of rice via genetic engineering technologies. Here, we conducted a meta-analysis of different rice genotypes at the seedling stage based on 96 public microarray datasets, aiming to identify the key salt-responsive genes and understand the molecular response mechanism of rice under salt stress. In total, 5559 genes were identified to be differentially expressed genes (DEGs) under salt stress, and 3210 DEGs were identified during the recovery process. The Gene Ontology (GO) enrichment results revealed that the salt-response mechanisms of shoots and roots were different. A close-knit signaling network, consisting of the Ca2+ signal transduction pathway, the mitogen-activated protein kinase (MAPK) cascade, multiple hormone signals, transcription factors (TFs), transcriptional regulators (TRs), protein kinases (PKs), and other crucial functional proteins, plays an essential role in rice salt stress response. In this study, many unreported salt-responsive genes were found. Besides this, MapMan results suggested that TNG67 can shift to the fermentation pathway to produce energy under salt stress and may enhance the Calvin cycle to repair a damaged photosystem during the recovery stage. Sugars are critical for plant growth and development as suppliers of carbon and energy, as signal molecules, or as solute molecules for osmotic homeostasis. Monosaccharide transporter (MST) genes are involved in various processes of plant growth and development as well as in response to abiotic stresses. However, the evolution and their roles of MST genes in growth and development and in coping with abiotic stresses in rice are poorly known. Here, we identified 64 MST genes in rice genome, which are classified into seven subfamilies: STP, PLT, AZT, ERD, pGlcT, INT, and XTPH. MST genes are not evenly distributed between chromosomes (Chrs) with a bias to Chr 3, 4, 7, and 11, which could be a result of duplication of fragments harboring MST genes. In total, 12 duplication events were found in the rice MST family, among which, two pairs were derived from fragmental duplications and ten pairs were from tandem duplications. The synonymous and nonsynonymous substitution rates of duplicate gene pairs demonstrated that the MST family was under a strong negative selection during the evolution process. Furthermore, a comprehensive expression analysis conducted in 11 different tissues, three abiotic stresses, five hormone treatments, and three sugar treatments revealed different expression patterns of MST genes and indicated diversified functions of them. Our results suggest that MST genes play important roles not only in various abiotic stresses but also in hormone and sugar responses.
