This paper reviews the published terminology, mathematical models, and the possible approaches used to characterise the risk of foodborne chemical hazards, particularly pesticides, metals, mycotoxins, acrylamide, and polycyclic aromatic hydrocarbons (PAHs). The results confirmed the wide variability of the nomenclature used, e.g., 28 different ways of referencing exposure, 13 of cancer risk, or 9 of slope factor. On the other hand, a total of 16 equations were identified to formulate all the risk characterisation parameters of interest. Therefore, the present study proposes a terminology and formulation for some risk characterisation parameters based on the guidelines of international organisations and the literature review. The mathematical model used for non-genotoxic hazards is a ratio in all cases. However, the authors used the probability of cancer or different ratios, such as the margin of exposure (MOE) for genotoxic hazards. For each effect studied per hazard, the non-genotoxic effect was mostly studied in pesticides (79.73%), the genotoxic effect was mostly studied in PAHs (71.15%), and both effects were mainly studied in metals (59.4%). The authors of the works reviewed generally opted for a deterministic approach, although most of those who assessed the risk for mycotoxins or the ratio and risk for acrylamide used the probabilistic approach. In addition, the studies also show the role of Epstein–Barr virus (EBV) reactivation and interactions with tested pathways of immune checkpoints involved in the immunopathogenesis of this disease. Simple Summary: Obesity has been linked to several common cancers including breast, colorectal, esophageal, kidney, gallbladder, uterine, pancreatic, and liver cancer. Obesity also increases the risk of dying from cancer and may influence the treatment choices. About 4–8% of all cancers are attributed to obesity. The underlying mechanism of obesity causing cancer is complex and is incompletely understood. Lifestyle changes that include diet, exercise, and behavior therapy are the mainstay of interventions. Drug therapy and weight reduction surgery result in a more rapid weight loss and may be used for a subgroup of cancer survivors with obesity. This review highlights the epidemiology and the risk associated with the development and recurrence of cancer in obese individuals and the management of obesity. Background: Obesity or excess body fat is a major global health challenge that has not only been associated with diabetes mellitus and cardiovascular disease but is also a major risk factor for the development of and mortality related to a subgroup of cancer. This review focuses on epidemiology, the relationship between obesity and the risk associated with the development and recurrence of cancer and the management of obesity. Methods: A literature search using PubMed and Google Scholar was performed and the keywords 'obesity' and cancer' were used. The search was limited to research papers published in English prior to September 2022 and focused on studies that investigated epidemiology, the pathogenesis of cancer, cancer incidence and the risk of recurrence, and the management of obesity. Results: About 4–8% of all cancers are attributed to obesity. Obesity is a risk factor for several major cancers, including post-menopausal breast, colorectal, endometrial, kidney, esophageal, pancreatic, liver, and gallbladder cancer. Excess body fat results in an approximately 17% increased risk of cancer-specific mortality. The relationship between obesity and the risk associated with the development of cancer and its recurrence is not fully understood and involves altered fatty acid metabolism, extracellular matrix remodeling, the secretion of adipokines and anabolic and sex hormones, immune dysregulation, and chronic inflammation. Obesity may also increase treatment-related adverse effects and influence treatment decisions regarding specific types of cancer therapy. Structured exercise in combination with dietary support and behavior therapy are effective interventions. Treatment with glucagon-like peptide-1 analogues and bariatric surgery result in more rapid weight loss and can be considered in selected cancer survivors. Conclusions: Obesity increases cancer risk and mortality. Weight-reducing strategies in obesity-associated cancers are important interventions as a key component of cancer care. Breast cancer (BC) risks imparted by CHEK2 c.1100delC ("1100delC") germline pathogenic/likely pathogenic variant (GPV) are 20–30%, compared to CHEK2 c.470T>C ("I157T") GPV with <20%, leading to different breast screening recommendations through MRI. We compared cancer risk management (CRM) across these two GPVs. Study participants were adult females with an 1100delC or I157T GPV drawn from the Inherited Cancer Registry (ICARE) across the United States. Cancer history, clinical characteristics, and CRM were compared using chi-squared tests, t-tests, and logistic regression. Of 150 CHEK2 carriers, 40.7% had BC, with a mean age of 50. Comparing 1100delC and I157T GPVs, there were no differences in rates of (1) breast MRI among those with (65.2% versus 55.6% of 23 and 9; p = 0.612) and without (44.0% versus 44.8% of 50 and 29; p = 0.943) BC; (2) risk-reducing mastectomy among those with (50% versus 38.9% of 46 and 15; p = 0.501) and without (13.8% versus 6.5% of 58 and 31; p = 0.296) BC; and (3) risk-reducing salpingo-oophorectomy among those with (24.2% versus 22.2% of 45 and 18; p = 0.852) and without (17.5% versus 16.7% of 57 and 30; p = 0.918) BC. Pathogenic mutations in BRCA1 (BReast CAncer gene 1) confer high risks of both breast (up to 70%) and ovarian (up to 40%) cancers. Zinc (Zn) and copper (Cu) are essential for various physiological functions, including antioxidant reactions. Their balance, reflected in the Zn/Cu ratio, plays a crucial role in maintaining redox homeostasis, which is vital for cancer prevention. This study examines the antioxidant properties of Zn and Cu, specifically focusing on the blood Zn/Cu ratio as a potential marker for cancer risk among BRCA1 mutation carriers. The study cohort consisted of 989 initially unaffected women, followed up for 7.5 years. Blood samples were analyzed using inductively coupled plasma mass spectrometry. Although individual Zn and Cu levels did not significantly correlate with overall cancer risk, those women with a Zn/Cu ratio above 6.38 experienced a significantly lower cancer risk than women with a ratio below this cut-off point. This suggests that the Zn/Cu ratio may be a valuable biomarker for cancer prevention in this high-risk group. Given the increased cancer risk in BRCA1 mutation carriers, optimizing Zn and Cu levels through dietary and active interventions could provide a preventive strategy. [ABSTRACT FROM AUTHOR]

BRCA1 mutations predispose women to breast and ovarian cancer. The anticancer effect of zinc is typically linked to its antioxidant abilities and protecting cells against oxidative stress. Zinc regulates key processes in cancer development, including DNA repair, gene expression, and apoptosis. We took a blood sample from 989 female BRCA1 mutation carriers who were initially unaffected by cancer and followed them for a mean of 7.5 years thereafter. There were 172 incident cases of cancer, including 121 cases of breast cancer, 29 cases of ovarian cancers, and 22 cancers at other sites. A zinc level in the lowest tertile was associated with a modestly higher risk of ovarian cancer compared to women with zinc levels in the upper two tertiles (HR = 1.65; 95% CI 0.80 to 3.44; p = 0.18), but this was not significant. Among those women with zinc levels in the lowest tertile, the 10-year cumulative risk of ovarian cancer was 6.1%. Among those in the top two tertiles of zinc level, the ten-year cumulative risk of ovarian cancer was 4.7%. There was no significant association between zinc level and breast cancer risk. The region surrounding the Elbrus volcano in the Great Caucasus is a popular tourist destination, home to native people and an area of the richest ore mineral resources. Based on the chemical composition of surface water, mineral, and groundwater in the Elbrus area, as well as statistical and thermodynamic calculations, waters with high concentrations of Mn, Cu, Zn, Mo, Ba, W, Pb, and As were identified, possible mineral phases that may lead to these metal concentrations in water were estimated, and risks to human health were assessed. Two main sources of surface water pollution have been identified: (1) water from tailings that are enriched due to oxidation of sulfide minerals is considered a source of As (181 µg/L) and Mo (2070 µg/L) pollution in the area of the Tyrnyauz W-Mo deposit; and (2) CO2-rich waters of deep horizons discharge on the earth surface with high concentrations of As (828 µg/L) and W (504 µg/L). It has been shown that compared to all toxic elements studied, only Mo and As have adverse non-carcinogenic effects on adult health. The estimated carcinogenic risk of As in the mine area was 1.7 × 10−3 and exceeded the generally accepted range of 10−4 to 10−6. In order to minimize the health risks, regular monitoring of water quality is required when mining activities on the Tyrnyauz W-Mo deposit are renewed. [ABSTRACT FROM AUTHOR]

Urban vegetable farming in wetlands and riverbanks are common features of Ugandan cities. However, urbanization has led to various anthropogenic activities that can lead to the pollution of water resources, enrichment of pollutants in soils and, consequently, pollutant bioaccumulation in edible tissues of plants cultivated on such soils. In this study, we report on the levels of six trace metals (TMTs) in 75 samples of leafy vegetables (Brassica oleracea L., Spinacia oleracea L., Amaranthus hybridus L., Cucurbita pepo L. and Solanum nigrum L.) and soils (n = 75) grown on the banks of River Rwizi, the second longest river in Uganda only after the Nile River. The concentrations of TMTs (Mn, Zn, Cd, Pb, Cr and Cu) in edible vegetable tissues and soils were quantified using flame atomic absorption spectrometry. The mean concentrations (in mg kg−1) of the TMTs in the soil samples were 205–373.84 (Mn), 12.72–65.04 (Zn), 0.26–0.42 (Cd), 3.36–16.80 (Pb), 5.96–25.06 (Cr) and 2.83–35.27 (Cu). In vegetable samples, the concentrations ranged from 43.25 to 110.00 (Mn), 1.08 to 1.83 (Cd), 41.06 to 71.20 (Zn), 4.31 to 6.16 (Pb), 0.65 to 0.81 (Cr) and 5.70 to 14.35 (Cu). With the exception of Mn and Cr, the rest of the TMTs were bioaccumulated in the edible vegetable tissues (bioconcentration factors = 1.03 to 10.71). Considering chronic daily intake through ingestion, dermal contact and inhalation of the TMTs in soils from the banks of River Rwizi, there are no potential non-cancer and carcinogenic health effects that could be experienced in both adults and children. Consumption of leafy vegetables could pose both non-cancer health risks (from ingestion of Zn, Pb, Cr, Mn and Cd) and cancer health risks (due to intake of Cd) in both children and adults. Hispanics of Mexican descent have disproportionate rates of non-alcoholic fatty liver disease (NAFLD). The purpose of this work is to investigate the association between the traditional Mexican diet score (tMexS) and hepatic steatosis and fibrosis, two NAFLD-related clinical endpoints, in Hispanic adults of Mexican descent. Data from 280 Hispanic adults of Mexican descent (n = 102 men, 178 women) with overweight or obesity enrolled in a cross-sectional observational study were analyzed. The tMexS was calculated from 24 h dietary recalls. Hepatic steatosis and fibrosis measurements were assessed using transient elastography (Fibroscan®). Linear regression models testing the association between tMexS and hepatic steatosis and fibrosis were run individually and through the stratification of significant modifiers. Mean tMexS were 5.9 ± 2.1, hepatic steatosis scores were 288.9 ± 48.9 dB/m, and fibrosis scores were 5.6 ± 2.2 kPa. Among the US-born group, with every point increase in the tMexS, there was a statistically significant 5.7 lower hepatic steatosis point (95% CI: −10.9, −0.6, p-value = 0.07). Higher adherence to a traditional Mexican diet was associated with lower hepatic steatosis in US-born Hispanics of Mexican descent. The association between red meat consumption and cancer risk remains a controversy. In this study, we systematically collected and analyzed global data (from Our World in Data and Global Cancer Observatory) to investigate this association for the first time. Our results confirmed significant positive associations between red meat consumption (RMC) and overall cancer incidence (0.798, p < 0.001), or colorectal cancer incidence (0.625, p < 0.001). Several previously unreported cancer types linked to RMC were also unveiled. Gross domestic product (GDP) per capita were found to have an impact on this association. However, even after controlling it, RMC remained significantly associated with cancer incidence (0.463, p < 0.001; 0.592, p < 0.001). Meanwhile, after controlling GDP per capita, the correlation coefficients between white meat consumption and overall cancer incidence were found to be much lower and insignificant, at 0.089 (p = 0.288) for poultry consumption and at −0.055 (p = 0.514) for seafood and fish consumption. Notably, an interesting comparison was performed between changes of colorectal cancer incidence and RMC in many countries and regions. A lag of 15–20 years was found, implying causality between RMC and cancer risk. Our findings will contribute to the development of more rational meat consumption concept. [ABSTRACT FROM AUTHOR]

Simple Summary: Cystic fibrosis (CF) is a genetic condition that affects the lungs, digestion, and other body systems. People with CF have a higher chance of developing certain types of cancer. The reason for this is related to a gene called CFTR, which is altered in CF patients. This gene normally helps regulate the movement of substances in and out of cells. When it does not work properly, it can lead to changes in cells that make them more likely to become cancerous. The cancers most commonly associated with CF are colorectal, pancreatic, and respiratory cancers. By understanding how CFTR and cancer are connected, doctors can develop better ways to screen for and treat these cancers in people with CF. More research is needed to fully understand this link and improve care for CF patients. Cystic fibrosis (CF) is a genetic disorder affecting multiple organs, primarily the lungs and digestive system. Over the years, advancements in medical care and treatments have significantly increased the life expectancy of individuals with CF. However, with this improved longevity, concerns about the potential risk of developing certain types of cancers have arisen. This narrative review aims to explore the relationship between CF, increased life expectancy, and the associated risk for cancers. We discuss the potential mechanisms underlying this risk, including chronic inflammation, immune system dysregulation, and genetic factors. Additionally, we review studies that have examined the incidence and types of cancers seen in CF patients, with a focus on gastrointestinal, breast, and respiratory malignancies. We also explore the impact of CFTR modulator therapies on cancer risk. In the gastrointestinal tract, CF patients have an elevated risk of developing colorectal cancer, pancreatic cancer, and possibly esophageal cancer. The underlying mechanisms contributing to these increased risks are not fully understood, but chronic inflammation, altered gut microbiota, and genetic factors are believed to play a role. Regular surveillance and colonoscopies are recommended for early detection and management of colorectal cancer in CF patients. Understanding the factors contributing to cancer development in CF patients is crucial for implementing appropriate surveillance strategies and improving long-term outcomes. Consumption of maize and maize-based products contributes a significant percentage to the total food energy intake in Uganda. However, the production of maize-derived foodstuffs is performed traditionally or by small- and medium-scale processors using different processing techniques. This can lead to differences in the quality of these products from processors, raising food safety concerns. In this study, the effects of mechanical processing (milling) methods on deposition of heavy metals into milled maize flour and the associated consumption health risks were assessed. Atomic absorption spectrophotometry was used to quantitatively establish the concentration of iron (Fe), manganese (Mn), zinc (Zn), cadmium (Cd), lead (Pb), chromium (Cr), copper (Cu), cobalt (Co) and nickel (Ni) in 100 samples of maize milled using a wooden mortar (n = 2), a metallic mortar (n = 2), diesel engine−powered mills (n = 48) and electric motor−powered mills (n = 48). Results showed that the mean concentrations of heavy metals in mg/kg were Fe (11.60–34.45), Cu (0.50–8.10), Ni (0.50–1.60), Mn (0.70–25.40), Zn (4.40–15.90), Pb (0.53–10.20), Cd (0.51–0.85), Cr (0.50–1.53) and Co (0.50–1.51). The highest concentrations were found in flour milled using a traditional metallic mortar while the lowest levels were in those samples milled using a wooden mortar. The Fe, Pb and Cd contents of flours produced using the metallic mortar and some commercial mills was found to be higher than the permissible limits set by WHO/FAO. Human health risk assessment showed that there are potential carcinogenic health risks from adults' intake of heavy metals in maize flour milled using a metallic mortar. Therefore, processing of maize flour needs to be monitored by the relevant statutory bodies in Uganda to minimize the possibility of heavy metal contamination of food products and animal feeds. Simple Summary: Colorectal cancer is the second most deadly tumor worldwide, despite the availability of screening plans and advanced treatment strategies. Molecular biomarkers predictive of prognosis have been identified, although there is currently limited knowledge of biomarkers predictive of the response to pharmacological treatment. In recent years, molecular classifications mainly able to predict colorectal cancer prognosis and recurrence risk have been proposed. The "consensus molecular subtype (CMS) classification" is the most known and has contributed to the understanding of genomic and epigenomic landscapes of colorectal cancer for better patient management. This classification categorizes colorectal cancer into four CMS categories (CMS1–4) that display different prognosis. This manuscript contextualized the CMS classification in different settings, discussing its relationships with precursor lesions, tumor immunophenotype, and gut microbiota, as well as its role in predicting prognosis and/or response to pharmacological treatments, as a crucial step towards precision medicine. Colorectal cancer (CRC) is the second cause of cancer-related deaths in both sexes globally and presents different clinical outcomes that are described by a range of genomic and epigenomic alterations. Despite the advancements in CRC screening plans and treatment strategies, the prognosis of CRC is dismal. In the last two decades, molecular biomarkers predictive of prognosis have been identified in CRC, although biomarkers predictive of treatment response are only available for specific biological drugs used in stage IV CRC. Translational clinical trials mainly based on "omic" strategies allowed a better understanding of the biological heterogeneity of CRCs. These studies were able to classify CRCs into subtypes mainly related to prognosis, recurrence risk, and, to some extent, also to treatment response. Accordingly, the comprehensive molecular characterizations of CRCs, including The Cancer Genome Atlas (TCGA) and consensus molecular subtype (CMS) classifications, were presented to improve the comprehension of the genomic and epigenomic landscapes of CRCs for a better patient management. The CMS classification obtained by the CRC subtyping consortium categorizes CRC into four consensus molecular subtypes (CMS1–4) characterized by different prognoses. Background: Contamination of groundwater could result in serious and irreparable health problems for consumers. This study assessed the water quality and human health implications of metals in packaged groundwater from Abeokuta and Sagamu, southwestern Nigeria. Methods: One hundred and forty bags of the packaged groundwater were purchased and analyzed for physical and chemical parameters using standard procedures, while metals were determined using inductively coupled plasma optical emission spectrophotometry. Risk assessment of metals was estimated using hazard index (HI), hazard quotient (HQ), and cancer risk (CR). Water quality index (WQI) was also evaluated. Results: The observed physical and chemical parameters, except Mn and Fe, in the packaged groundwater samples were observed to be within the permissible limits of the World Health Organization. The WQI data indicated suitability for drinking purposes. The health risk data indicated high HQs > 1.0 for Ca (for adults and children at all the monitoring sites), Mn (children at all the sites, and adults at six sites), Mo (children at four sites, and adults at three sites), and Cu (children at three sites and adults at one site). Conclusion: The CRs of the packaged groundwater for Co and Ni at many sites revealed values > the acceptable limit of 1 × 10−4, indicating possible development of cancer by the consumers. Colorectal cancer is one of the most common tumors, and genetic predisposition is one of the key risk factors in the development of this malignancy. Lynch syndrome and familial adenomatous polyposis are the best-known genetic diseases associated with hereditary colorectal cancer. However, some other genetic disorders confer an increased risk of colorectal cancer, such as Li–Fraumeni syndrome (TP53 gene), MUTYH-associated polyposis (MUTYH gene), Peutz–Jeghers syndrome (STK11 gene), Cowden syndrome (PTEN gene), and juvenile polyposis syndrome (BMPR1A and SMAD4 genes). Moreover, the recent advances in molecular techniques, in particular Next-Generation Sequencing, have led to the identification of many new genes involved in the predisposition to colorectal cancers, such as RPS20, POLE, POLD1, AXIN2, NTHL1, MSH3, RNF43 and GREM1. In this review, we summarized the past and more recent findings in the field of cancer predisposition genes, with insights into the role of the encoded proteins and into the associated genetic disorders. Experimental studies support the hypothesis that GH/IGF-1 status may influence neoplastic tissue growth. Epidemiological studies suggest a link between GH/IGF-1 status and cancer risk. However, several studies regarding GH replacement safety in childhood cancer survivors do not show a prevalence excess of de novo cancers, and several reports on children and adults treated with GH have not shown an increase in observed cancer risk in these patients. Due to insufficient healthcare facilities for the fight against cancer, a large percentage of individuals die. Utilizing computational tools inside the health and medical system helps to minimize fatalities. Timely cancer detection enhances the likelihood of effective therapy. Cancer risk assessment is important for legal and regulatory reasons, for cancer prevention, and to avoid the risks. The approach for assessing cancer risk based on the q-rung orthopair fuzzy set (q-ROFS) is described. The technique is predicated on a multifactor evaluation of the likelihood of a cancerous. q-ROFS is a robust approach for modeling uncertainties in multicriteria decision making (MCDM). The combinative distance-based assessment (CODAS) technique integrates two separate approaches, namely the "simple additive weighting" (SAW) method and the "weighted product method (WPM)". In this study, the CODAS approach is extended to the q-rung orthopair fuzzy framework with application to cancer risk assessment. Our study aimed to identify pathways from the source of information to the uptake of cancer genetic testing, with consideration of intermediate variables including perceptional, attitudinal and psychosocial factors. We used the Health Information National Trends Survey (2020 database) and constructed a structural equation model for pathway analysis (using SPSS version 24). Variables for socio-demographic, lifestyle and health information were also collected and used for confounding adjustment. A total of 2941 participants were analyzed (68.5%, non-Hispanic white; 59.7%, females; 58 years, median age; and 142 (4.8%) had undertaken genetic testing for cancer risk previously). Our pathway analysis found that only information from particular sources (i.e., healthcare providers and genetic counsellors) had positive and significant effects on people's perceptions of cancer regarding its prevention, detection and treatment (standardized β range, 0.15–0.31, all p-values < 0.01). Following the paths, these perceptional variables (cancer prevention, detection and treatment) showed considerable positive impacts on the uptake of genetic testing (standardized β (95% CIs): 0.25 (0.20, 0.30), 0.28 (0.23, 0.33) and 0.12 (0.06, 0.17), respectively). Pathways involving attitudinal and psychosocial factors showed much smaller or insignificant effects on the uptake of genetic testing. The accumulation of polyaromatic hydrocarbons in plants is considered one of the most serious threats faced by mankind because of their persistence in the environment and their carcinogenic and teratogenic effect on human health. The concentrations of sixteen priority polycyclic aromatic hydrocarbons (16 PAHs) were determined in four types of edible vegetables (tomatoes, zucchini, eggplants, and cucumbers), irrigation water, and agriculture soil, where samples were collected from the Jordan Valley, Jordan. The mean total concentration of 16 PAHs (∑16PAHs) ranged from 10.649 to 21.774 µg kg−1 in vegetables, 28.72 µg kg−1 in soil, and 0.218 µg L−1 in the water samples. The tomato samples posed the highest ∑16PAH concentration level in the vegetables, whereas the zucchini samples had the lowest. Generally, the PAHs with a high molecular weight and four or more benzene rings prevailed among the studied samples. The diagnostic ratios and the principal component analysis (PCA) revealed that the PAH contamination sources in soil and vegetables mainly originated from a pyrogenic origin, traffic emission sources, and biomass combustion. The bioconcentration factors (BCF) for ∑16PAHs have been observed in the order of tomatoes > cucumbers and eggplants > zucchini. A potential cancer risk related to lifetime consumption was revealed based on calculating the incremental lifetime cancer risk of PAHs (ILCR). The history of gold mining in the Witwatersrand Basin has led to exponential growth in the economy, residential development, and the abundance of radionuclides in the environment, including the water system. This study aimed to evaluate the radionuclide levels in drinking water (municipal water and groundwater) and the health risks associated with the ingestion of the water in the communities of the West Rand region of Gauteng Province. The activity concentrations of uranium, radium, and thorium radioisotopes were established through alpha spectrometry and the activities were subsequently used to assess the health impacts. The results indicated that the groundwaters contain elevated activities of most radionuclides owing to prolonged periods of water–rock interactions. Similarly, the highest annual effective doses were recorded in groundwaters with a range of 0.0237–0.3106 mSv/yr, with most samples exceeding the WHO- and EU-prescribed limits of 0.1 mSv/yr. Cancer morbidity and mortality risks were higher in females than in males due to the higher life expectancy of females. Nonetheless, all morbidity and mortality risks were well below the USEPA radiological risk limit of 0.001. Hypothyroidism in patients with solid non-thyroid cancer is a tantalizing entity, integrating an intriguing thyroid hormones (THs)–cancer association with the complexity of hypothyroidism itself. The present narrative review provides a comprehensive overview of the clinical relevance of hypothyroidism in solid non-thyroid cancer. Hypothyroidism in patients with solid non-thyroid cancer is reminiscent of hypothyroidism in the general population, yet also poses distinct challenges due to the dual role of THs in cancer: promoting versus inhibitory. Close collaboration between oncologists and endocrinologists will enable the prompt and personalized diagnosis and treatment of hypothyroidism in patients with solid non-thyroid cancer. Clinical data indicate that hypothyroidism is a predictor of a decreased or increased risk of solid non-thyroid cancer and is a prognostic factor of favorable or unfavorable prognosis in solid non-thyroid cancer. However, the impact of hypothyroidism with respect to the risk and/or prognosis of solid non-thyroid cancer is not a consistent finding. To harness hypothyroidism, or THs replacement, as a personalized anticancer strategy for solid non-thyroid cancer, four prerequisites need to be fulfilled, namely: (i) deciphering the dual THs actions in cancer; (ii) identifying interventions in THs status and developing agents that block tumor-promoting THs actions and/or mimic anticancer THs actions; (iii) appropriate patient selection; and (iv) counteracting current methodological limitations. Poor and inadequate sanitation systems have been considered not only a human health issue, but also an environmental threat that instigates climate change. Nine heavy metals—arsenic (As), cadmium (Cd), cobalt (Co), chromium (Cr), iron (Fe), manganese (Mn), nickel (Ni), lead (Pb), and zinc (Zn)—were evaluated in influent and effluent water samples from four wastewater treatment plants in the Durban metropolis, KwaZulu-Natal, South Africa. The results indicate that the mean concentrations of all the heavy metals in the influent samples ranged from 0.122 to 1.808 mg/L, while the effluent samples had a concentration ranging from 0.118 to 0.854 mg/L. Iron was found to be in the highest concentration and the concentration of Co was the lowest across the wastewater treatment plants. The levels for most of the heavy metals in this study were found to be above the recommended maximum concentrations in surface and effluent waters as stipulated by the World Health Organization, United States Environmental Protection Agency, Food and Agriculture Organization, and the Department of Water Affairs and Forestry of South Africa. According to the toxicity effect due to non-carcinogenic risks, As, Pb, Cr, and Cd are considered to be of medium risk in this study, indicating that a probable adverse health risk is very likely to occur. Additionally, the cancer risk (RI) values were lower than 10−3, which shows that cancer development is very likely in individuals who are exposed. Featured Application: More appropriate decision making on how to manage a land contaminated by hazardous materials would be possible by application of the site-specific approach for health risk assessment presented herewith. After the Fukushima Daiichi accident, there have been long controversial discussions on "how safe is safe?" between the authorities and the residents in the affected area. This controversy was partly attributable to the way the authorities made a judgement based on the annual effective dose rate; meanwhile, many of the local residents have serious concerns about future consequences for their health caused by chronic radiation exposure, particularly of small children. To settle this controversy, the author presents an approach based on long-term cancer risk projections of female infants, i.e., the most radiosensitive group, following land contamination by radiocesium deposition into ground with different surface conditions; the land was classified into three categories on the basis of decaying patterns of radiation dose rate: "Fast", "Middle", and "Slow". From the results of analyses with an initial dose rate of 20 mGy per year, it was predicted that the integrated lifetime attributable risk (LAR) of cancer mortality of a female person ranged by a factor of 2 from 1.8% (for the Fast area) to 3.6% (for the Slow area) that were clearly higher than the nominal risk values derived from effective dose estimates with median values of environmental model parameters. These findings suggest that accurate site-specific information on the behavioral characteristics of radionuclides in the terrestrial environment are critically important for adequate decision making for protecting people when there is an event accompanied by large-scale radioactive contamination. Mycotoxin contamination in rice can create a health risk for the consumers. In this study, the measurement of 23 mycotoxins in rice samples (n = 180) was performed using a validated LC-MS/MS method. A food frequency questionnaire was used to get rice consumption data for the assessment of mycotoxin dietary exposure, before calculating the health risk in adults and children of north and south regions of the Pakistani Punjab province. The prevalence of aflatoxin B1 (56%), aflatoxin B2 (48%), nivalenol (28%), diacetoxyscirpenol (23%), fumonisin B1 (42%), zearalenone (15%), HT-2 toxin (10%), deoxynivalenol (8%), and ochratoxin A (6%) was estimated in samples with a mean concentration range between 0.61 and 22.98 µg/kg. Aflatoxin degradation by traditional Pakistani cooking recipes was evaluated and observed to be 41-63%. The dietary exposure to aflatoxins exceeded the tolerable daily intake at all levels, and ochratoxin A and zearalenone posed health risk at high contamination and high consumption levels. The margin of aflatoxin B1 exposure ranged between 10 and 69 in adults and 10 and 62 in children. The mean cancer risk by aflatoxin B1 exposure was 0.070 (adults) and 0.071 (children) cases/year/100,000 people in South Punjab population, and 0.122 (adults) and 0.127 (children) cases/year/100,000 people in North Punjab population. In this study, 24 h PM2.5 (particles with an equivalent diameter equal to or below 2.5 μm) samples were collected in winter and summer in Xi'an, Northwestern China to characterize the seasonal variations of eleven elements (As, Cd, Cr, Fe, K, Mn, Mo, Pb, Ni, Zn, and Cu) and to evaluate their health risks by using the US EPA (U.S. Environmental Protection Agency) method. Mass concentrations of the elements (except Ni) in winter were much higher than those in summer, with similar variations for both seasons. The levels of elements followed a decreasing order of K > Zn > Fe > Pb > Cr > As > Mn > Cu > Mo > Ni > Cd. According to the enrichment factor (EF) analysis, the highest EF value for Cd inferred that it should be linked with the metal smelting and other anthropogenic sources. In contrast, the EF values of K and Mn (1 < EF < 5) suggested that they were influenced by both natural and anthropogenic sources. The daily average exposure dose for children and adults by different exposure pathways were both ingestion > dermal contact > inhalation. The non-cancer risks for different exposure pathways showed different orders. The non-cancer risks (hazard quotients) were lower than the average risk threshold (1.0) except for As, Pb, and Cr, which require greater attention. Aflatoxins (AFTs), as a group 1 carcinogen, could lead to hepatocellular carcinoma (HCC). Dietary intake is the primary way of AFT exposure in humans. However, the contribution of foodborne AFT intake to the HCC burden remains unknown in recent years in China. Hence, the present study was conducted to estimate the burden of HCC attributed to foodborne AFT exposure by using disability-adjusted life years (DALYs). The risk assessment was used to estimate the incidence of HCC related to AFT exposure. Concentrations of AFTs in peanuts, peanut oil, corn, and corn products were retrieved from literature published between 2010 and 2020 in China. Corresponding daily food consumption data were obtained from two nationwide Chinese surveys. A direct approach was used to calculate DALY and DALY rates to quantify the HCC burden attributed to dietary AFT exposure. The total amount of AFT intake through peanut, peanut oil, corn, and corn products was 4.018 ng/kg bw/day resulting in 0.125 extra HCC cases per year/100,000 persons, corresponding to a DALY number and DALY rate of 21,625.08 and 1.53 per 100,000 population, respectively. Regionally, DALYs were high in Guangxi and Guangdong provinces, corresponding to 5948 and 5595 DALYs. A total of 1.5 DALYs/100,000 were lost due to the AFT exposure. DALYs per 100,000 population were higher in several coastal areas. Though the disease burden of HCC caused by dietary AFTs was low in the Chinese population, a high health risk was found in the residents of some areas with high AFT exposure. Simple Summary: Formaldehyde is a chemical compound present in many working activities and indoor workplaces. Occupational exposure occurs primarily by inhaling airborne formaldehyde, but it can also be absorbed through the skin or ingested. The International Agency for Research on Cancer (IARC) classified formaldehyde as a Group 1 carcinogen for humans in 2004, based on toxicological data and epidemiological evidence obtained in workplaces, all published before that year. Over the last two decades, many new studies in this field have been published, providing updated findings. The aim of the present systematic review was to synthetize the results of epidemiological studies in occupational settings carried out in the last 20 years and to evaluate whether the IARC classification was confirmed by further studies. Our results show that the evidence of correlation between formaldehyde occupational exposure and the occurrence of cancer is limited. Background: Formaldehyde, classified as a carcinogen in 2004, as of today is widely used in many work activities. From its classification, further studies were performed to evaluate its carcinogenicity. The aim of the systematic review is to update the evidence on occupational exposure to formaldehyde and cancer onset. Methods: The review, in accordance with the PRISMA statement, includes articles in English reporting original results of studies conducted on workers exposed to formaldehyde, considering all types of cancer, published from 1 January 2000 to 30 July 2021 and selected from the Pubmed and Scopus databases. The studies' quality was assessed by the Newcastle–Ottawa Scale. Results: A total of 21 articles were included, conducted in different European, American, and Asian countries. The most investigated occupational areas are those characterized by a deliberate use of formaldehyde. Some studies evaluated all types of cancer, whereas others focused on specific sites such as thyroid and respiratory, lymphohematopoietic, or central nervous systems. The results showed weak associations with lung cancer, nasopharyngeal cancer, leukemia, and non-Hodgkin's lymphoma. Conclusions: The results demonstrate the need for further original studies carried out on representative samples of workers exposed to measured levels of FA. This study assessed the concentration of heavy metal, such as lead (Pb), cadmium (Cd), Chromium (Cr), iron (Fe), Nickel (Ni), and Silver (Ag), in Vernonia amygdalina Delile and agricultural soils of three university farms located in Port Harcourt, Nigeria. The soils and plants were taken randomly to form composite samples and analyzed for heavy metals by the use of atomic absorption spectroscopy (AAS) and X-ray fluorescence (XRF). The study stations were agricultural soils and Vernonia amygdalina Delile from the Ignatius Ajuru University of Education (I), River State University (R) and University of Port Harcourt (U). The soil samples recorded mean concentration ranges for Fe as 19.71 ± 1.77 (I)–27.24 ± 3.56 mg/kg (R) in soils and 12.95 ± 1.68 (R)–18.18 ± 2.02 mg/kg (U) for the bitter leaf samples. The mean range for Pb concentration in the soil and bitter leaf were 4.35 ± 0.87–6.80 ± 0.86 mg/kg and 0.24 ± 0.64–2.19 ± 0.74 mg/kg, while Cd concentration in the soil and bitter leaf were 0.46 ± 0.28–1.42 ± 0.40 mg/kg and 0.17 ± 0.22–0.42 ± 0.08 mg/kg, respectively. The respective mean ranges for Cr concentration in the soil and bitter leaf were 5.91 ± 1.14–8.77 ± 0.88 mg/kg and 4.04 ± 0.64–5.92 ± 0.69 mg/kg, while Ni in soil and bitter leaf were 0.54 ± 3.38–10.26 ± 3.50 mg/kg and 0.042 ± 1.42–3.30 ± 0.88 mg/kg, while Ag was negligible. Heavy metal levels in soils and Vernonia amygdalina followed the order Fe > Cr > Pb > Ni > Cd and Fe > Cr > Ni > Pb > Cd, respectively, and were lower than WHO/FAO and EPA, except Cd, which was higher in soil and in Vernonia amygdalina. The ecological risk factor (ErF) was comparatively lower in soils than in the plant, while pollution load index (PLI) showed high heavy metal retention capacities in Vernonia amygdalina due to more anthropogenic influences. The metal transfer factor (TF) was highest in Fe, followed by Cr > Cd > Ni > Pb, while Pb had the highest chances of cancer risks from the incremental lifetime cancer risk (ILCR), especially in both soil and plant (mean ILCR, 2.07 × 10−2 and 2.45 × 10−3), while Cd had the least (mean ILCR, 9.64 × 10−5 and 3.36 × 10−5). Road and industrial origin particulate matters (PM) are a significant source of potentially toxic elements (PTEs), with health risks to the surrounding residents. In Korea for 60 years, although industries, roads and automobiles have increased aggressively, there are still few PTEs data in PM in road-deposited sediment (RDS) of industrial complexes (ICs). Therefore, this study aimed to investigate the PTE composition of on-road PM10 from nine major ICs and its pollution degree in Korea and evaluate its human health risks. The geo-accumulation index (Igeo) and pollution load index (PLI) elucidated that on-road PM10 were severely polluted by Sb, Zn, Ag and Pb. A combination of principal component analysis (PCA) and chemical tracers was used to define the PTEs sources. The results showed that non-exhaust emission from vehicles' activity is the primary source of PTEs in on-road PM10, and industrial emissions are the secondary source. The riskiest pathway on carcinogenic and non-carcinogenic by on-road PM10 with PTEs was in-gestion. Traffic origin PTEs including Pb, As, Sb and Cd had a more significant impact on carcinogenic and non-carcinogenic health than those of industrial origins. In the present study, we determined the levels of chlorinated pesticide residues in surface water samples collected from the Charsadda district (KPK, Pakistan). SPME-GC-ECD with COMBI PAL CTC autosampler was used for extraction and analysis of 20 organochlorine pesticides in the collected water samples. For maximum efficiency of the SPME procedure, several parameters were studied, including the extraction and desorption time of the fiber, solution pH, agitation of samples, and stirring speed, etc. This method showed good liner response, with R2 values in the range of 0.9887 to 0.9999 for all pesticides. This method also provided good percent recoveries at 1 µg L−1 (87.5 to 106.0%) and at 2 µg L−1 (88.5 to 109.2%). Lower limits of detection for all 20 chlorinated pesticides were found to be lower than their maximum permissible contamination levels. Approximately 50% of the surface water samples collected from the Charsadda district were found to be contaminated with the pesticides γ-BHC, heptachlor, aldrin and dieldrin, with maximum concentrations of 0.023, 0.108, 0.014 and 0.013 µg L−1, respectively. For adults and children, the cancer risk from water due to contamination by various pesticides ranged from 0 to 33.29 × 10−6. The non-carcinogenic risk from each pollutant in the water samples of the Charsadda district was found to be in the order of heptachlor > aldrin > dieldrin > γ-BHC. However, the pesticides α-BHC, β-BHC, heptachlor epoxide, chlordane, endrin, 4,4′-DDD, endrin ketone, 4,4′-DDT, endosulfan sulfate and methoxychlor were not detected in any of the surface water samples of investigated in the present study. Vitamin A and carotenoids are fat-soluble micronutrients that play important role as powerful antioxidants modulating oxidative stress and cancer development. Breast cancer is the most common malignancy in women. As the risk of breast cancer is dependent on various lifestyle factors such as dietary modifications, there is increasing interest surrounding the anti-cancerous properties of vitamin A and carotenoids. Despite the suggested protective roles of vitamin A and carotenoids in breast cancer development, their clinical application for the prevention and treatment of breast cancer is limited. In this narrative review, we discuss the roles of vitamin A and carotenoids along with the evaluation method of vitamin A status. We also exhibit the association of genetic variations involved in metabolism of vitamin A and carotenoids with cancers and other diseases. We demonstrate the epidemiological evidence for the relationship of vitamin A and carotenoids with breast cancer risk, their effects on cancer mechanism, and the recent updates in clinical practice of vitamin A or carotenoids as a potential therapeutic agent against breast cancer. Simple Summary: Individuals with abnormal alterations in mitochondrial DNA copy number (mtDNA-CN) and telomere length are at higher risk of developing certain types of cancer. This report suggests that mtDNA-CN and relative telomere length measured in peripheral blood have potential clinical applications for risk prediction of different cancers and that mtDNA-CN could be used as a prognostic biomarker in malignancy. This comprehensive work strengthens several previous relevant findings in certain types of cancer and broadens our understanding of the link between mtDNA-CN, telomere length and future risk of many cancer types. The translational implication of our findings is that postmenopausal genital organ cancer patients with lower levels of baseline mtDNA-CN or shorter telomere length can be identified for early adjustment of lifestyle and hormone replacement therapy. Changes in mitochondrial DNA copy number (mtDNA-CN) and telomere length have, separately, been proposed as risk factors for various cancer types. However, those results are conflicting. Here, mtDNA-CN and relative telomere length were measured in 3225 middle-aged women included in a large population-based prospective cohort. The baseline mtDNA-CN in patients with prevalent breast cancer was significantly higher (12.39 copies/µL) than cancer-free individuals. During an average of 15.2 years of follow-up, 520 patients were diagnosed with cancer. Lower mtDNA-CN was associated with decreased risk of genital organ cancer (hazard ratio (HR), 0.84), and shorter telomere length was associated with increased risk of urinary system cancer (HR, 1.79). Furthermore, mtDNA-CN was inversely associated with all-cause (HR, 1.20) and cancer-specific mortality (HR, 1.21) when considering all cancer types. Surprisingly, shorter telomere length was associated with decreased risk of cancer-specific mortality when considering all cancer types (HR, 0.85). Finally, lower mtDNA-CN and shorter telomere length were associated with increased risk of both all-cause and cancer-specific mortality in genital organ cancer patients. In this study population, we found that mtDNA-CN and telomere length were significantly associated with prevalent and incident cancer and cancer mortality. Numerous studies are focused on endemic regions of high exposure, and its effect on human health. Several authors suggest that As may be an important endocrine disruptor, mainly through estrogen-like activity. In our previous study on subjects without germline mutations in BRCA1, we reported that increased blood arsenic levels are significantly associated with high breast-cancer risk. The aim of this study was to assess if an association between As levels and cancer risk also exists among women harboring mutations in BRCA1. We found that women with As blood levels above the median (0.85 µg/L) had a significant 2-fold increased risk of developing breast cancer. This raises the possibility that lowering the blood arsenic level by dietary means might reduce cancer risk for BRCA1 mutation carriers. An important group of breast cancers is those associated with inherited susceptibility. In women, several predisposing mutations in genes involved in DNA repair have been discovered. Women with a germline pathogenic variant in BRCA1 have a lifetime cancer risk of 70%. As part of a larger prospective study on heavy metals, our aim was to investigate if blood arsenic levels are associated with breast cancer risk among women with inherited BRCA1 mutations. A total of 1084 participants with pathogenic variants in BRCA1 were enrolled in this study. Subjects were followed from 2011 to 2020 (mean follow-up time: 3.75 years). During that time, 90 cancers were diagnosed, including 67 breast and 10 ovarian cancers. The group was stratified into two categories (lower and higher blood As levels), divided at the median (<0.85 µg/L and ≥0.85 µg/L) As level among all unaffected participants. Cox proportional hazards models were used to model the association between As levels and cancer incidence. A high blood As level (≥0.85 µg/L) was associated with a significantly increased risk of developing breast cancer (HR = 2.05; 95%CI: 1.18–3.56; p = 0.01) and of any cancer (HR = 1.73; 95%CI: 1.09–2.74; p = 0.02). Simple Summary: Some patients carry a mutated copy of the CDH1 gene that can lead to a very rare form of hereditary gastric cancer called signet-ring cell adenocarcinoma (SRCC). SRCCs rarely form visible tumors prior to spreading. Hence, patients are recommended to have prophylactic gastrectomies at a young age. Many patients wish to avoid surgery and thus have regular checks with upper endoscopy with biopsies to rule out cancer. Further, these patients may also be at risk of other cancers beyond the already known breast cancer risks, but this is not known. In this study, we show that despite systematic biopsy protocols, many early cancers might be missed on endoscopy. Therefore, patients should not rely on endoscopy to delay surgery. These patients may also be at increased risk of colorectal SRCC, which has very poor survival outcomes. To confirm this, we need a central database that captures outcomes for this patient population. Hereditary diffuse gastric cancer (HDGC) is a rare signet-ring cell adenocarcinoma (SRCC) linked to CDH1 (E-cadherin) inactivating germline mutations, and increasingly other gene mutations. Female CDH1 mutation carriers have additional risk of lobular breast cancer. Risk management includes prophylactic total gastrectomy (PTG). The utility of endoscopic surveillance is unclear, as early disease lacks macroscopic lesions. The current systematic biopsy protocols have unknown efficacy, and other secondary cancer risks are postulated. We conducted a retrospective study of consecutive asymptomatic HDGC patients undergoing PTG, detailing endoscopic, pathologic, and outcome results. A systematic review compared endoscopic biopsy foci detection via random sampling versus Cambridge Protocol against PTG findings. A population-level secondary-cancer-risk postulation among sporadic gastric SRCC patients was completed using the Surveillance, Epidemiology, and End Results database. Of 97 patients, 67 underwent PTG, with 25% having foci detection on random endoscopic biopsy despite 75% having foci on final pathology. There was no improvement in the endoscopic detection rate by Cambridge Protocol. The postulated hazard ratio among sporadic gastric SRCC patients for a secondary colorectal SRCC was three-fold higher, relative to conventional adenocarcinoma patients. Overall, HDGC patients should not rely on endoscopic surveillance to delay PTG, and may have secondary SRCC risks. Airborne particulate matter (PM) pollution often occurs in the wintertime in northern China, posing a potential threat to human health. To date, there are limited studies about the metals and inorganic ions to link source apportionments and health risk assessments in the different size-segregated PM samples. In this study, our samples were collected by a high-volume air sampler from 26 December 2018 to 11 January 2019 in a high population residential area (Beijing). Water-soluble inorganic ions, metal elements in the different size-segregated PM samples were determined for health risk assessments by inhalation of PM. During the sampling period in Beijing, the average concentrations of PM1.1 and PM1.1–2.0 were 39.67 ± 10.66 μg m−3 and 32.25 ± 6.78 μg m−3. Distinct distribution profiles characterized the different elements. The markers of coal combustion Pb, As, and Se had >52% of their mass concentration in PM1.1. The average mass ratios of (NO3− + NO2−)/SO42−, Cl−/Na+, Cl−/K+, and Cl−/(NO3− + NO2−) were 1.68, 6.58, 6.18, and 0.57, which showed that coal combustion and vehicle emissions were the main anthropogenic sources of PM in Beijing in winter. PM1.1 was the major contributor of Pb, Cd, and As for carcinogenic risks (CR) and hazard quotient (HQ). It was indicated that PM1.1 is more harmful than coarse PM. The toxic elements of Cr (VI) (1.12 × 10−6), V (0.69 × 10−6), and As (0.41 × 10−6) caused higher CR for children than Ni, Cd, Co, and Pb. Meanwhile, Pb (35.30 × 10−6) and Ni (21.07 × 10−6) caused higher CR for adults than As, Cr (VI), V, Co, and Cd, especially PM1.1. Non-nutritive sweeteners (NNS) are popular sugar substitutes that can help in weight and diabetes management, but concerns regarding their use have been raised by the public. This study aimed to investigate knowledge, benefits and safety perceptions of NNS in a sample of UK adults. The impact of knowledge dissemination on the change in perceptions was also examined. An online survey was distributed through social media platforms and UK Universities and was completed by 1589 participants aged 18 years and above. Results showed a high-risk perception of NNS and a lack of knowledge in regulations in nearly half the population sample. The artificial attributes of NNS further limited their acceptance. Risk perception has been significantly linked to a lower consumption of sweeteners (p < 0.001) and was affected by gender, occupation, education levels, age and body weight status. Information dissemination significantly reduced risk perception and increased awareness of the benefits of NNS. Results suggest that developing effective communication strategies to educate consumers, potentially through trusted health government agencies and professional bodies, can help them to make informed choices. Cancer is predominantly considered as an environmental disease caused by genetic or epigenetic alterations induced by exposure to extrinsic (e.g., carcinogens, pollutants, radiation) or intrinsic (e.g., metabolic, immune or genetic deficiencies). Over-exposure to antibiotics, which is favored by unregulated access as well as inappropriate prescriptions by physicians, is known to have led to serious health problems such as the rise of antibiotic resistance, in particular in poorly developed countries. In this review, the attention is focused on evaluating the effects of antibiotic exposure on cancer risk and on the outcome of cancer therapeutic protocols, either directly acting as extrinsic promoters, or indirectly, through interactions with the human gut microbiota. The preponderant evidence derived from information reported over the last 10 years confirms that antibiotic exposure tends to increase cancer risk and, unfortunately, that it reduces the efficacy of various forms of cancer therapy (e.g., chemo-, radio-, and immunotherapy alone or in combination). Alternatives to the current patterns of antibiotic use, such as introducing new antibiotics, bacteriophages or enzybiotics, and implementing dysbiosis-reducing microbiota modulatory strategies in oncology, are discussed. The information is in the end considered from the perspective of the most recent findings on the tumor-specific and intracellular location of the tumor microbiota, and of the most recent theories proposed to explain cancer etiology on the notion of regression of the eukaryotic cells and systems to stages characterized for a lack of coordination among their components of prokaryotic origin, which is promoted by injuries caused by environmental insults. Objective: To evaluate the incidence of cancer within the first 15 years of life in children who underwent cardiac catheterization under the age of one year. Methods: In this retrospective, single center study, 2770 infants (7.8% with trisomy 21) were studied. All infants underwent cardiac catheterization under one year of age between January 1980 and December 1998. Newly diagnosed cancer in the first 15 years of life was assessed through record linkage to the German Childhood Cancer Registry (GCCR). Cancer risk in study patients was compared to the GCCR population of children less than 15 years. Patients with trisomy 21 were compared to the Danish Cytogenic Register for trisomy 21. Effective radiation doses were calculated for each tumor patient and 60 randomly selected patients who did not develop cancer. Results: In total, 24,472.5 person-years were analyzed. Sixteen children developed cancer, while 3.64 were expected (standardized incidence ratio (SIR) = 4.4, 95% confidence interval (CI): 2.5–7.2, p < 0.001). There was no preferred cancer type. The observed incidence of leukemia and solid tumors in trisomy 21 was only slightly higher (1 in 476 py) than expected (1 in 609 py, p = 0.64). There was no direct relationship between the radiation dose and the incidence of cancer. DNA adducts are believed to play a central role in the induction of cancer in cigarette smokers and are proposed as being potential biomarkers of cancer risk. We have summarized research conducted since 2012 on DNA adduct formation in smokers. A variety of DNA adducts derived from various classes of carcinogens, including aromatic amines, polycyclic aromatic hydrocarbons, tobacco-specific nitrosamines, alkylating agents, aldehydes, volatile carcinogens, as well as oxidative damage have been reported. The results are discussed with particular attention to the analytical methods used in those studies. Mass spectrometry-based methods that have higher selectivity and specificity compared to 32P-postlabeling or immunochemical approaches are preferred. Multiple DNA adducts specific to tobacco constituents have also been characterized for the first time in vitro or detected in vivo since 2012, and descriptions of those adducts are included. We also discuss common issues related to measuring DNA adducts in humans, including the development and validation of analytical methods and prevention of artifact formation. [ABSTRACT FROM AUTHOR]

The general population is exposed to the genotoxic carcinogen glycidol via food containing refined edible oils where glycidol is present in the form of fatty acid esters. In this study, internal (in vivo) doses of glycidol were determined in a cohort of 50 children and in a reference group of 12 adults (non-smokers and smokers). The lifetime in vivo doses and intakes of glycidol were calculated from the levels of the hemoglobin (Hb) adduct N-(2,3-dihydroxypropyl)valine in blood samples from the subjects, demonstrating a fivefold variation between the children. The estimated mean intake (1.4 μg/kg/day) was about two times higher, compared to the estimated intake for children by the European Food Safety Authority. The data from adults indicate that the non-smoking and smoking subjects are exposed to about the same or higher levels compared to the children, respectively. The estimated lifetime cancer risk (200/105) was calculated by a multiplicative risk model from the lifetime in vivo doses of glycidol in the children, and exceeds what is considered to be an acceptable cancer risk. The results emphasize the importance to further clarify exposure to glycidol and other possible precursors that could give a contribution to the observed adduct levels. [ABSTRACT FROM AUTHOR]
