Pipelines extend thousands of kilometers to transport and distribute oil and gas. Given the challenges often faced with corrosion, fatigue, and other issues in steel pipes, the demand for glass fiber-reinforced plastic (GFRP) pipes is increasing in oil and gas gathering and transmission systems. However, the medium that is transported through these pipelines contains multiple acid gases such as CO 2 and H 2 S, as well as ions including Cl - , Ca 2+ , Mg 2+ , SO 4 2- , CO 3 2- , and HCO 3 - . These substances can cause a series of problems, such as aging, debonding, delamination, and fracture. In this study, a series of aging damage experiments were conducted on V-shaped defect GFRP pipes with depths of 2 mm and 5 mm. The aging and failure of GFRP were studied under the combined effects of external force and acidic solution using acoustic emission (AE) techniques. It was found that the acidic aging solution promoted matrix damage, fiber/matrix desorption, and delamination damage in GFRP pipes over a short period. However, the overall aging effect was relatively weak. Based on the experimental data, the SSA-LSSVM algorithm was proposed and applied to the damage pattern recognition of GFRP. An average recognition rate of up to 90% was achieved, indicating that this method is highly suitable for analyzing AE signals related to GFRP damage.

This article systematically reviews the extraction and purification methods, structural characteristics, structure-activity relationship, and health benefits of C. speciosa polysaccharides, and their potential application in food, medicine, functional products, and feed, in order to provide a useful reference for future research. Chaenomeles speciosa (Sweet) Nakai. has attracted the attention of health consumers and medical researchers as a traditional Chinese medicine with edible, medicinal, and nutritional benefits. According to this study, C. speciosa polysaccharides have significant health benefits, such as anti-diaetic, anti-inflammatory and analgesic, anti-tumor, and immunomodulatory effects. Researchers determined the molecular weight, structural characteristics, and monosaccharide composition and ratio of C. speciosa polysaccharides by water extraction and alcohol precipitation. This study will lay a solid foundation for further optimization of the extraction process of C. speciosa polysaccharides and the development of their products. As an active ingredient with high value, C. speciosa polysaccharides are worthy of further study and full development. C. speciosa polysaccharides should be further explored in the future, to innovate their extraction methods, enrich their types and biological activities, and lay a solid foundation for further research and development of products containing polysaccharides that are beneficial to the human body.

Recombinant adeno-associated virus (rAAV) has emerged as a prominent vector for in vivo gene therapy, owing to its distinct advantages. Accurate determination of the rAAV genome titer is crucial for ensuring the safe and effective administration of clinical doses. The evolution of the rAAV genome titer assay from quantitative PCR (qPCR) to digital PCR (dPCR) has enhanced accuracy and precision, yet practical challenges persist. This study systematically investigated the impact of various operational factors on genome titration in a single-factor manner, aiming to address potential sources of variability in the quantitative determination process. Our findings revealed that a pretreatment procedure without genome extraction exhibits superior precision compared with titration with genome extraction. Additionally, notable variations in titration results across different brands of dPCR instruments were documented, with relative standard deviation (RSD) reaching 23.47% for AAV5 and 11.57% for AAV8. Notably, optimal operations about DNase I digestion were identified; we thought treatment time exceeding 30 min was necessary, and there was no need for thermal inactivation after digestion. And we highlighted that thermal capsid disruption before serial dilution substantially affected AAV genome titers, causing a greater than ten-fold decrease. Conversely, this study found that additive components of dilution buffer are not significant contributors to titration variations. Furthermore, we found that repeated freeze-thaw cycles significantly compromised AAV genome titers. In conclusion, a comprehensive dPCR titration protocol, incorporating insights from these impact factors, was proposed and successfully tested across multiple serotypes of AAV. The results demonstrate acceptable variations, with the RSD consistently below 5.00% for all tested AAV samples. This study provides valuable insights to reduce variability and improve the reproducibility of AAV genome titration using dPCR.

The aggregation-induced emission (AIE) effect exhibits a significant influence on the development of luminescent materials and has made remarkable progress over the past decades. The advancement of high-performance AIE materials requires fast and accurate predictions of their photophysical properties, which is impeded by the inherent limitations of quantum chemical calculations. In this work, we present an accurate machine learning approach for the fast predictions of quantum yields and wavelengths to screen out AIE molecules. A database of about 563 organic luminescent molecules with quantum yields and wavelengths in the monomeric/aggregated states was established. Individual/combined molecular fingerprints were selected and compared elaborately to attain appropriate molecular descriptors. Different machine learning algorithms combined with favorable molecular fingerprints were further screened to achieve more accurate prediction models. The simulation results indicate that combined molecular fingerprints yield more accurate predictions in the aggregated states, and random forest and gradient boosting regression algorithms show the best predictions in quantum yields and wavelengths, respectively. Given the successful applications of machine learning in quantum yields and wavelengths, it is reasonable to anticipate that machine learning can serve as a complementary strategy to traditional experimental/theoretical methods in the investigation of aggregation-induced luminescent molecules to facilitate the discovery of luminescent materials.

β-Farnesene is an advanced molecule with promising applications in agriculture, the cosmetics industry, pharmaceuticals, and bioenergy. To supplement the shortcomings of rational design in the development of high-producing β-farnesene strains, a Metabolic Pathway Design-Fermentation Test-Metabolomic Analysis-Target Mining experimental cycle was designed. In this study, by over-adding 20 different amino acids/nucleobases to induce fluctuations in the production of β-farnesene, the changes in intracellular metabolites in the β-farnesene titer-increased group were analyzed using non-targeted metabolomics. Differential metabolites that were detected in each experimental group were selected, and their metabolic pathways were located. Based on these differential metabolites, targeted strain gene editing and culture medium optimization were performed. The overexpression of the coenzyme A synthesis-related gene pantothenate kinase ( PanK ) and the addition of four mixed water-soluble vitamins in the culture medium increased the β-farnesene titer in the shake flask to 1054.8 mg/L, a 48.5% increase from the initial strain. In the subsequent fed-batch fermentation, the β-farnesene titer further reached 24.6 g/L. This work demonstrates the tremendous application value of metabolomics analysis for the development of industrial recombinant strains and the optimization of fermentation conditions.

The human visual attention system plays an important role in infrared target recognition because it can quickly and accurately recognize infrared small targets and has good scene adaptability. This paper proposes an infrared small target detection method based on an attention mechanism, which consists of three modules: a bottom-up passive attention module, a top-down active attention module, and decision feedback equalization. In the top-down active attention module, given the Gaussian characteristics of infrared small targets, the idea of combining knowledge-experience Gaussian shape features is applied to implement feature extraction, and quaternion cosine transform is performed to achieve multi-dimensional fusion of Gaussian shape features, thereby achieving complementary fusion of multi-dimensional feature information. In the bottom-up passive attention module, considering that the difference in contrast and motion between the target and the background can attract attention easily, an optimal fast local contrast algorithm and improved circular pipeline filtering are adopted to find candidate target regions. Meanwhile, the multi-scale Laplacian of the Gaussian filter is adopted to estimate the optimal size of the infrared small target. The fast local contrast algorithm based on box filter acceleration and structure optimization is employed to extract local contrast features, and candidate target regions can be obtained by using an adaptive threshold. Besides, the mean gray, target size, Gaussian consistency, and circular region constraint are used in pipeline filtering to extract motion regions, and the false-alarm rate is reduced effectively. Finally, decision feedback equalization is adopted to obtain real targets. Experiments are conducted on some real infrared images involving complex backgrounds with sea, sky, and ground clutters, and the experimental results indicate that the proposed method can achieve better detection performance than conventional baseline methods, such as RLCM, ILCM, PQFT, MPCM, and ADMD. Also, mathematical proofs are provided to validate the proposed method.

In order to improve the safety and quality of lactose-free milk (LFM) Maillard reaction products (MRPs), this study used raw cow's milk as raw material and lactase hydrolysis to prepare LFM, which was heat-treated using pasteurization and then placed in storage temperatures of 4 °C, 25 °C and 37 °C to investigate the changes in the Maillard reaction (MR). The results of the orthogonal test showed that the optimal conditions for the hydrolysis of LFM are as follows: the hydrolysis temperature was 38 °C, the addition of lactase was 0.03%, and the hydrolysis time was 2.5 h. Under these conditions, the lactose hydrolysis rate reached 97.08%, and the lactose residue was only 0.15 g/100 g as determined by high-performance liquid chromatography (HPLC), complying with the standard of LFM in GB 28050-2011. The contents of furoamic acid and 5-hydroxymethylfurfural were determined by high-performance liquid chromatography, the color difference was determined by CR-400 color difference meter, and the internal fluorescence spectrum was determined by F-320 fluorescence spectrophotometer. The test results showed that the variation range of furosine in lactose-free milk after pasteurization was 44.56~136.45 mg/100g protein, the range of 5-hydroxymethylfurfural (HMF) was 12.51~16.83 mg/kg, the color difference ranges from 88.11 to 102.53 in L*, from -0.83 to -0.10 in a*, and from 1.88 to 5.47 in b*. The furosine content of LFM during storage at 4, 25, and 37 °C ranged from 44.56 to 167.85, 44.56 to 287.13, and 44.56 to 283.72 mg/100 g protein, respectively. The average daily increase in protein content was 1.18-3.93, 6.46-18.73, and 15.7-37.66 mg/100 g, respectively. The variation range of HMF was 12.51~17.61, 12.51~23.38, and 12.51~21.1 mg/kg, and the average daily increase content was 0.03~0.07, 0.47~0.68, and 0.51~0.97 mg/kg, respectively. During storage at 4 °C, the color difference of LFM ranged from 86.82 to 103.82, a* ranged from -1.17 to -0.04, and b* ranged from 1.47 to 5.70. At 25 °C, color difference L* ranges from 72.09 to 102.35, a* ranges from -1.60 to -0.03, b* ranges from 1.27 to 6.13, and at 37 °C, color difference L* ranges from 58.84 to 102.35, a* ranges from -2.65 to 1.66, and b* ranges from 0.54 to 5.99. The maximum fluorescence intensity (FI) of LFM varies from 131.13 to 173.97, 59.46 to 173.97, and 29.83 to 173.97 at 4, 25, and 37 °C. In order to reduce the effect of the Maillard reaction on LFM, it is recommended to pasteurize it at 70 °C-15 s and drink it as soon as possible during the shelf life within 4 °C.

Carbon nanotubes (CNTs) and styrene-butadiene-styrene (SBS) are used as reinforcing modifiers in asphalt sealants due to their excellent properties, which can effectively improve the internal structure of the sealant and enhance its mechanical properties. Based on this background, two SBS/CNT-modified asphalt sealants were identified and selected by the orthogonal experimental method and compared with two commercially available sealants. The softening point, flow value, multi-temperature frequency scan test, and multiple stress creep recovery test were used to study the high-temperature rheological properties and aging resistance of four types of sealants. The overall evaluation shows that the proportion of the sealant compound's preparation material is 1% by weight of CNT doping, 5% by weight of SBS doping, and 5% by weight of furfural-extracted-oil doping. The results show that the addition of SBS and CNTs more significantly improves the fatigue resistance of the sealants. With the CAM model, C1.0S5F5 reflects a better relaxation property, which better avoids secondary cracking after the construction of the sealant. With the Burgers model, C1.0S5F5 shows excellent deformation resistance under heavy traffic conditions. In summary, conventional performance indicators, such as the softening point and flow value of SBS/CNT-modified asphalt sealants, can meet the specification requirements and show good high-temperature stability and anti-aging properties compared to commercially available sealants.

In addition to the essential pharmacological effects of opioids, situational cues associated with drug addiction memory are key triggers for drug seeking. CircRNAs, an emerging hotspot regulator in crown genetics, play an important role in central nervous system-related diseases. However, the internal mediating mechanism of circRNAs in the field of drug reward and addiction memory remains unknown. Here, we trained mice on a conditional place preference (CPP) model and collected nucleus accumbens (NAc) tissues from day 1 (T0) and day 8 (T1) for high-throughput RNA sequencing. QRT-PCR analysis revealed that circTmeff-1 was highly expressed in the NAc core but not in the NAc shell, suggesting that it plays a role in addiction memory formation. Meanwhile, the down-regulation of circTmeff-1 by adeno-associated viruses in the NAc core or shell could inhibit the morphine CPP scores. Subsequently, the GO and KEGG analyses indicated that circTmeff-1 might regulate the addiction memory via the MAPK and AMPK pathways. These findings suggest that circTmeff-1 in NAc plays a crucial role in morphine-dependent memory formation.

Chilling causes a significant decline in photosynthesis in tomato plants. Tomato tryptophan decarboxylase gene 1 ( SlTDC1 ) is the first rate-limiting gene for melatonin (MT) biosynthesis and is involved in the regulation of photosynthesis under various abiotic stresses. However, it is not clear whether SlTDC1 participates in the photosynthesis of tomato under chilling stress. Here, we obtained SlTDC1 overexpression transgenic tomato seedlings, which showed higher SlTDC1 mRNA abundance and MT content compared with the wild type (WT). The results showed that the overexpression of SlTDC1 obviously alleviated the chilling damage to seedlings in terms of the lower electrolyte leakage rate and hydrogen peroxide content, compared with the WT after 2 d of chilling stress. Moreover, the overexpression of SlTDC1 notably increased photosynthesis under chilling stress, which was related to the higher chlorophyll content, normal chloroplast structure, and higher mRNA abundance and protein level of Rubisco and RCA, as well as the higher carbon metabolic capacity, compared to the WT. In addition, we found that SlTDC1 -overexpressing seedlings showed higher W k (damage degree of OEC on the PSII donor side), φ Eo (quantum yield for electron transport in the PSII reaction center), and PI ABS (photosynthetic performance index) than WT seedlings after low-temperature stress, implying that the overexpression of SlTDC1 decreased the damage to the reaction center and donor-side and receptor-side electron transport of PSII and promoted PSI activity, as well as energy absorption and distribution, to relieve the photoinhibition induced by chilling stress. Our results support the notion that SlTDC1 plays a vital role in the regulation of photosynthesis under chilling stress.

Carbon xerogels co-doped with nitrogen (N) and phosphorus (P) or sulfur (S) were synthesized and employed as catalysts for the electrocatalytic reduction of p-nitrophenol (p-NP). The materials were prepared by first synthesizing N-doped carbon xerogels (NDCX) via the pyrolysis of organic gels, and then introducing P or S atoms to the NDCX by a vapor deposition method. The materials were characterized by various measurements including X-ray diffraction, N 2 physisorption, Transmission electron microscopy, Fourier Infrared spectrometer, and X-ray photoelectron spectra, which showed that N atoms were successfully doped to the carbon xerogels, and the co-doping of P or S atoms affected the existing status of N atoms. Cyclic voltammetry (CV) scanning manifested that the N and P co-doped materials, i.e., P-NDCX-1.0, was the most suitable catalyst for the reaction, showing an overpotential of -0.569 V (vs. Ag/AgCl) and a peak slop of 695.90 μA/V. The material was also stable in the reaction and only a 14 mV shift in the reduction peak overpotential was observed after running for 100 cycles.

The effects of Steam Flash-Explosion (SFE) on the physicochemical properties and molecular structure of high-temperature denatured defatted rice bran protein isolate (RBPI) were investigated. The mechanism of SFE treatment on high-temperature denatured defatted RBPI was revealed. The analysis of the physical and chemical properties of RBPI showed that the surface hydrophobicity, characteristic viscosity, and thermal stability of rice bran protein isolate were significantly affected by the pressure of saturated steam and pressure holding time. Under the conditions of 2.1 MPa and 210 s, the surface hydrophobicity index decreased significantly from 137.5 to 17.5, and the characteristic viscosity increased significantly. The peak temperature of denaturation decreases from 114.2 to 106.7 °C, and the enthalpy of denaturation decreases from 356.3 to 231.4 J/g. The higher structure (circular dichroic spectrum and endogenous fluorescence spectrum) of rice bran protein isolate was analyzed by volume rejection chromatography (SEC). The results showed that steam flash treatment could depolymerize and aggregate RBPI, and the relative molecular weight distribution changed greatly. The decrease in small molecules with poor solubility was accompanied by the increase in macromolecules (>550 kDa) soluble aggregates, which were the products of a Maillard reaction. The contents of free sulfhydryl and disulfide bonds in high-temperature rice bran meal protein isolate were significantly increased, which resulted in the increase in soluble aggregates containing disulfide bonds. Circular dichroism (CD) analysis showed that the α-helix content of the isolated protein was significantly decreased, the random curl content was increased, and the secondary structure of the isolated protein changed from order to disorder. The results of endogenous fluorescence spectroscopy showed that the high-temperature rice bran meal protein isolate was more extended, tryptophan was in a more hydrophilic microenvironment, the fluorescence intensity was reduced, and the tertiary structure was changed. In addition, the mean particle size and net surface charge of protein isolate increased in the aqueous solution, which was conducive to the development of the functional properties of the protein.

Carbon materials with pseudocapacitive performance have attracted emerging interest in the energy storage and conversion field. Reduced graphene oxide (r-GO) with superior conductivity and electrochemical stability has been extensively investigated as an efficient capacitive electrode material. In this study, three-dimensional carbon dots (CDs)@r-GO hydrogel electrode was successfully in situ prepared by the one-pot method, where the CDs play a critical role in serving as both reduction agent and electrochemical active sites. With prolonged reaction time, the oxygen content of the CDs@r-GO nanocomposite material could be effectively reduced to ensure better electric conductivity, and the nitrogen content, which provides pseudocapacitance, was gradually increased. The representative two pairs of fast and reversible current peaks appeared in cyclic voltammetry curves, with around three times higher specific capacitance of CDs@r-GO hydrogel electrode (290 F g -1 at the current density of 1 A g -1 in 1 M H 2 SO 4 electrolyte). This simple and mild approach is promising and it is believed it will shed more light on the preparation of high-efficiency and high-performance energy storage materials based on functional reductive CDs., Competing Interests: The authors declare no conflict of interest.

Grafting is widely applied to enhance the tolerance of some vegetables to biotic and abiotic stress. Salicylic acid (SA) is known to be involved in grafting-induced chilling tolerance in cucumber. Here, we revealed that grafting with pumpkin ( Cucurbita moschata , Cm ) as a rootstock improved chilling tolerance and increased the accumulation of SA, abscisic acid (ABA) and hydrogen peroxide (H 2 O 2 ) in grafted cucumber ( Cucumis sativus/Cucurbita moschata, Cs/Cm ) leaves. Exogenous SA improved the chilling tolerance and increased the accumulation of ABA and H 2 O 2 and the mRNA abundances of CBF1 , COR47 , NCED , and RBOH1 . However, 2-aminoindan-2-phosphonic acid (AIP) and L-a-aminooxy-b-phenylpropionic acid (AOPP) (biosynthesis inhibitors of SA) reduced grafting-induced chilling tolerance, as well as the synthesis of ABA and H 2 O 2 , in cucumber leaves. ABA significantly increased endogenous H 2 O 2 production and the resistance to chilling stress, as proven by the lower electrolyte leakage (EL) and chilling injury index (CI). However, application of the ABA biosynthesis inhibitors sodium tungstate (Na 2 WO 4 ) and fluridone (Flu) abolished grafting or SA-induced H 2 O 2 accumulation and chilling tolerance. SA-induced plant response to chilling stress was also eliminated by N,N'-dimethylthiourea (DMTU, an H 2 O 2 scavenger). In addition, ABA-induced chilling tolerance was attenuated by DMTU and diphenyleneiodonium (DPI, an H 2 O 2 inhibitor) chloride, but AIP and AOPP had little effect on the ABA-induced mitigation of chilling stress. Na 2 WO 4 and Flu diminished grafting- or SA-induced H 2 O 2 biosynthesis, but DMTU and DPI did not affect ABA production induced by SA under chilling stress. These results suggest that SA participated in grafting-induced chilling tolerance by stimulating the biosynthesis of ABA and H 2 O 2 . H 2 O 2 , as a downstream signaler of ABA, mediates SA-induced chilling tolerance in grafted cucumber plants.

Handwriting plays an important role in written communication, reading, and academic success. However, little is known about the neural correlates of handwriting in children. Using functional magnetic resonance imaging (fMRI) and a copying task, we investigated regional brain activation and functional lateralization associated with Chinese handwriting in children (N = 36, 9-11 years old), as well as their relations to reading skills. We found significant activation of the bilateral frontal motor cortices, somatosensory cortex, intraparietal sulcus (IPS), fusiform gyrus (FuG), and cerebellum during handwriting, suggesting that an adult-like brain activation pattern emerges by middle childhood. Moreover, children showed left-lateralized and bilateral activation of motor regions and right-lateralized activation of the FuG and cerebellum during handwriting, suggesting that functional lateralization of handwriting is not fully established by this age. Finally, the activation of Exner's area and the lateralization of the IPS and cerebellum during handwriting were correlated with reading skills, possibly representing a neural link between handwriting and reading in children. Collectively, this study reveals the brain correlates of handwriting and their relation to reading development in Chinese children, offering new insight into the development of handwriting and reading skills.

Photodynamic therapy (PDT) has become an emerging cancer treatment method. Choosing the photosensitizer (PS) compounds is one of the essential factors that can influence the PDT effect and action. Carbon dots (CDs) have shown great potential as photosensitizers in PDT of cancers due to their excellent biocompatibility and high generation of reactive oxygen species (ROS). Here, we used tea polyphenol as raw material for synthesized tea polyphenol carbon dots (T-CDs) that show dual emission bands of red and blue fluorescence and can efficiently generate hydroxyl radicals (OH) under mildly visible irradiation with a LED light (400-500 nm, 15 mW cm -2 ). The extremely low cytotoxicity and excellent biocompatibility of T-CDs without light irradiation were tested using MTT and hemolytic assay. Further, T-CDs have been shown by in vivo experiments, using a mouse breast cancer cell line (4T1) subcutaneously injected in the back of the mouse buttock as a model, to effectively inhibit the tumor cell proliferation in solid tumors and show an excellent PDT effect. In addition, pathological sections of the mice tissues after further treatment showed that the T-CDs had no apparent impact on the major organs of the mice and did not produce any side effect lesions. This work demonstrates that the as-synthesized T-CDs has the potential to be used as a PS in cancer treatment.

Marine fungi-derived secondary metabolites are still an important source for the discovery of potential antimicrobial agents. Here, five new polyketides ( 1 , 2 , and 6 - 8 ) and seven known compounds ( 3 - 5 and 9 - 12 ) were obtained from the culture of the marine-derived fungus Trichoderma sp. JWM29-10-1. Their structures were identified by extensive spectrographic data analyses, including 1D and 2D NMR, UV, IR, and HR-ESI-MS. Further, the absolute configurations of new compounds were determined by circular dichroism (CD) spectrum and alkali-hydrolysis in combination with the in situ dimolybdenum CD method. Subsequently, the antimicrobial effects of these isolated compounds were assessed by examining the minimal inhibition concentration (MIC) with the broth microdilution assay. Compounds 1 and 2 exhibited potent antimicrobial activity against Helicobacter pylori , including multidrug-resistant strains, with MIC range values of 2-8 µg/mL. Moreover, compound 1 showed significant inhibitory effects on the growth of Gram-positive pathogens, including methicillin-resistant Staphylococcus aureus (MRSA), Enterococcus faecalis , and vancomycin-resistant Enterococcus faecium , which greatly threaten human health. This study demonstrates that chromone derivatives 1 - 2 , especially for 1 , could be potential lead compounds for the development of new antimicrobial agents and provides insight for future medicinal chemistry research.

To improve the wear resistance and fatigue life of Cu alloys, surface modification by combining the magnetron sputtering of Ti film followed by vacuum thermal diffusion is always applied, where the structure and composition of the fabricated film play a determinant role on the mechanical properties. In the present work, the evolution of the layered structure and the element distribution of the formed multi-phases coating on C17200 Cu alloy are investigated by mathematical calculation based on Fick's law, and the experimental verification by the thermal diffusion of the gradient Cu-Ti film was undertaken under different temperatures and durations. The results show that the layered structure of the fabricated coating is dependent on the Cu-Ti atom concentration, the increasing time and the temperature, where a single or stratified layer is formed due to the generated Cu-Ti intermetallics for the inter-diffusion between the Cu and Ti atoms. The atom distribution by the proposed simulation method based on Fick's law corresponds to the experimental results, which can be applied to designing the structure of the modification layer.

Spodoptera frugiperda (J. E. Smith), an emerging invasive pest worldwide, has posed a serious agricultural threat to the newly invaded areas. Although somatic sex differentiation is fundamentally conserved among insects, the sex determination cascade in S. frugiperda is largely unknown. In this study, we cloned and functionally characterized Doublesex ( dsx ), a "molecular switch" modulating sexual dimorphism in S. frugiperda using male- and female-specific isoforms. Given that Lepidoptera is recalcitrant to RNAi, CRISPR/Cas9-mediated mutagenesis was employed to construct S. frugiperda mutants. Specifically, we designed target sites on exons 2, 4, and 5 to eliminate the common, female-specific, and male-specific regions of S. frugiperda dsx ( Sfdsx ), respectively. As expected, abnormal development of both the external and internal genitalia was observed during the pupal and adult stages. Interestingly, knocking out sex-specific dsx variants in S. frugiperda led to significantly reduced fecundity and fertility in adults of corresponding sex. Although graphitic carbon nitride (g-C 3 N 4 ) has been reported for several decades, it is still an active material at the present time owing to its amazing properties exhibited in many applications, including photocatalysis. With the rapid development of characterization techniques, in-depth exploration has been conducted to reveal and utilize the natural properties of g-C 3 N 4 through modifications. Among these, the assembly of g-C 3 N 4 with metal oxides is an effective strategy which can not only improve electron-hole separation efficiency by forming a polymer-inorganic heterojunction, but also compensate for the redox capabilities of g-C 3 N 4 owing to the varied oxidation states of metal ions, enhancing its photocatalytic performance. Herein, we summarized the research progress on the synthesis of g-C 3 N 4 and its coupling with single- or multiple-metal oxides, and its photocatalytic applications in energy production and environmental protection, including the splitting of water to hydrogen, the reduction of CO 2 to valuable fuels, the degradation of organic pollutants and the disinfection of bacteria. Aiming at the problems of single detection target of existing distributed denial of service (DDoS) attacks, incomplete detection datasets and privacy caused by shared datasets, we propose a trusted multi-domain DDoS detection method based on federated learning. Firstly, we divide the types of DDoS attacks into different sub-attacks, design the federated learning dataset for DDoS detection in each domain, and use them to realize a more comprehensive detection method of DDoS attacks on the premise of protecting the data privacy of each domain. Secondly, in order to improve the robustness of federated learning and alleviate poisoning attack, we propose a reputation evaluation method based on blockchain, which estimates interaction reputation, data reputation and resource reputation of each participant comprehensively, so as to obtain the trusted federated learning participants and identify the malicious participants. In addition, we also propose a combination scheme of multi-domain detection and distributed knowledge base and design a feature graph of malicious behavior based on a knowledge graph to realize the memory of multi-domain feature knowledge. The experimental results show that the accuracy of most categories of the multi-domain DDoS detection method can reach more than 95% with the protection of datasets, and the reputation evaluation method proposed in this paper has a higher ability to identify malicious participants against the data poisoning attack when the threshold is set to 0.6., Competing Interests: The authors declare no conflict of interest.

Sex determination is an important and traditional biological process. In Lepidoptera, Masculinizer ( Masc ) and doublesex ( dsx ) are the essential genes for sex determination and play critical roles in sexual differentiation and development. The functions of Masc and dsx have been characterized in several model insect species. However, the molecular mechanism and sex determination functions of Masc and dsx in Ostrinia furnacalis , an agricultural pest, are still unknown. Here, we successfully used the CRISPR/Cas9 genome editing system to knock out OfMasc and Ofdsx . Mutation of OfMasc induced male external genital defects and sterility. Disruptions of the Ofdsx common region caused sex-specific defects in the external genitals and adult sterility. In addition, we found that OfMasc and Ofdsx can regulate the pigmentation genes that control wing pigmentation patterns. These results demonstrate that OfMasc and Ofdsx play key roles in the sex determination of O. furnacalis , and suggest novel genetic control approaches for the management of pests, including O. furnacalis .

Flexible sensor arrays are widely used for wearable physiological signal recording applications. A high density sensor array requires the signal readout to be compatible with multiple channels. This paper presents a highly-integrated remote health monitoring system integrating a flexible pressure sensor array with a multi-channel wireless readout chip. The custom-designed chip features 64 voltage readout channels, a power management unit, and a wireless transceiver. The whole chip fabricated in a 65 nm complementary metal-oxide-semiconductor (CMOS) process occupies 3.7 × 3.7 mm 2 , and the core blocks consume 2.3 mW from a 1 V supply in the wireless recording mode. The proposed multi-channel system is validated by measuring the ballistocardiogram (BCG) and pulse wave, which paves the way for future portable remote human physiological signals monitoring devices.

The misuse of many types of broad-spectrum antibiotics leads to increased antimicrobial resistance. As a result, the development of a novel antibacterial agent is essential. Photodynamic antimicrobial chemotherapy (PACT) is becoming more popular due to its advantages in eliminating drug-resistant strains and providing broad-spectrum antibacterial resistance. Carbon dots (CDs), zero-dimensional nanomaterials with diameters smaller than 10 nm, offer a green and cost-effective alternative to PACT photosensitizers. This article reviewed the synthesis methods of antibacterial CDs as well as the recent progress of CDs and their nanocomposites in photodynamic sterilization, focusing on maximizing the bactericidal impact of CDs photosensitizers. This review establishes the base for future CDs development in the PACT field.

Melatonin (MT), as a signaling molecule, plays a vital role in regulating leaf senescence in plants. This study aimed to verify the antioxidant roles of MT in delaying dark- or age-induced leaf senescence of cucumber plants. The results showed that endogenous MT responds to darkness and overexpression of CsASMT , the key gene of MT synthesis, and delays leaf senescence stimulated by darkness, as manifested by significantly lower malonaldehyde (MDA) and reactive oxygen species (ROS) contents as well as higher activities and gene expression of antioxidant enzymes compared to the control. Moreover, MT suppressed both age- or dark-induced leaf senescence of cucumber, as evidenced by a decrease in senescence-related gene SAG20 and cell-death-related gene PDCD expression and ROS content and an increase in antioxidant capacity and chlorophyll biosynthesis compared with the H 2 O-treated seedlings. Meanwhile, the suppression of age-induced leaf senescence by melatonin was also reflected by the reduction in abscisic acid (ABA) biosynthesis and signaling pathways as well as the promotion of auxin (IAA) biosynthesis and signaling pathways in cucumber plants in the solar greenhouse. Combining the results of the two separate experiments, we demonstrated that MT acts as a powerful antioxidant to alleviate leaf senescence by activating the antioxidant system and IAA synthesis and signaling while inhibiting ABA synthesis and signaling in cucumber plants.

Environmental stresses, especially heat and drought, severely limit plant growth and negatively affect wheat yield and quality worldwide. Heat shock factors (Hsfs) play a central role in regulating plant responses to various stresses. In this study, the wheat heat shock factor gene TaHsfA2e-5D on chromosome 5D was isolated and functionally characterized, with the goal of investigating its role in responses to heat and drought stresses. Gene expression profiling showed that TaHsfA2e-5D was expressed constitutively in various wheat tissues, most highly in roots at the reproductive stage. The expression of TaHsfA2e-5D was highly up-regulated in wheat seedlings by heat, cold, drought, high salinity, and multiple phytohormones. The TaHsfA2e-5D protein was localized in the nucleus and showed a transcriptional activation activity. Ectopic expression of the TaHsfA2e-5D in yeast exhibited improved thermotolerance. Overexpression of the TaHsfA2e-5D in Arabidopsis results in enhanced tolerance to heat and drought stresses. Furthermore, RT-qPCR analyses revealed that TaHsfA2e-5D functions through increasing the expression of Hsp genes and other stress-related genes, including APX2 and GolS1 . Collectively, these results suggest that TaHsfA2e-5D functions as a positive regulator of plants' responses to heat and drought stresses, which may be of great significance for understanding and improving environmental stress tolerance in crops.

To discover sources for novel anti-influenza drugs, we evaluated the antiviral potential of nine extracts from eight medicinal plants and one mushroom ( Avena sativa L., Hordeum vulgare Linn. var. nudum Hook. f., Hippophae rhamnoides Linn., Lycium ruthenicum Murr., Nitraria tangutorum Bobr., Nitraria tangutorum Bobr. by-products, Potentilla anserina L., Cladina rangiferina (L.) Nyl., and Armillaria luteo-virens ) from the Qinghai-Tibetan plateau against the influenza A/H3N2 virus. Concentrations lower than 125 μg/mL of all extracts demonstrated no significant toxicity in MDCK cells. During screening, seven extracts ( A. sativa , H. vulgare , H. rhamnoides , L. ruthenicum , N. tangutorum , C. rangiferina , and A. luteo-virens ) exhibited antiviral activity, especially the water-soluble polysaccharide from the fruit body of the mushroom A. luteo-virens . These extracts significantly reduced the infectivity of the human influenza A/H3N2 virus in vitro when used at concentrations of 15.6-125 μg/mL. Two extracts ( N. tangutorum by-products and P. anserina ) had no A/H3N2 virus inhibitory activity. Notably, the extract obtained from the fruits of N. tangutorum and N. tangutorum by-products exhibited different anti-influenza effects. The results suggest that extracts of A. sativa , H. vulgare , H. rhamnoides , L. ruthenicum , N. tangutorum , C. rangiferina , and A. luteo-virens contain substances with antiviral activity, and may be promising sources of new antiviral drugs.

Understanding the way in which a decline in ocean pH can affect calcareous organisms could enhance our ability to predict the impacts of the potential decline in seawater pH on marine ecosystems, and could help to reconstruct the paleoceanographic events over a geological time scale. Planktic foraminifera are among the most important biological proxies for these studies; however, the existing research on planktic foraminifera is almost exclusively based on their geochemical indices, without the inclusion of information on their biological development. Through a series of on-board experiments in the western tropical Pacific (134°33'54″ E, 12°32'47″ N), the present study showed that the symbiont-bearing calcifier Trilobatus sacculifer -a planktic foraminifer-responded rapidly to a decline in seawater pH, including losing symbionts, bleaching, etc. Several indices were established to quantify the relationships between these biological parameters and seawater pH, which could be used to reconstruct the paleoceanographic seawater pH. We further postulated that the loss of symbionts in planktic foraminifera acts as an adaptive response to the stress of low pH. Our results indicate that an ongoing decline in seawater pH may hinder the growth and calcification of planktic foraminifera by altering their biological processes. A reduction in carbonate deposition and predation could have profound effects on the carbon cycle and energy flow in the marine food web.

Six novel Ir(C^N) 2 (L^X)-type heteroleptic iridium complexes with deep-red and near-infrared region (NIR)-emitting coverage were constructed through the cross matching of various cyclometalating (C^N) and ancillary (LX) ligands. Here, three novel C^N ligands were designed by introducing the electron-withdrawing group CF 3 on the ortho (o-), meta (m-), and para (p-) positions of the phenyl ring in the 1-phenylisoquinoline (piq) group, which were combined with two electron-rich LX ligands (dipba and dipg), respectively, leading to subsequent iridium complexes with gradually changing emission colors from deep red (≈660 nm) to NIR (≈700 nm). Moreover, a series of phosphorescent organic light-emitting diodes (PhOLEDs) were fabricated by employing these phosphors as dopant emitters with two doping concentrations, 5% and 10%, respectively. They exhibited efficient electroluminescence (EL) with significantly high EQE values: >15.0% for deep red light0 ( λ max = 664 nm) and >4.0% for NIR cases ( λ max = 704 nm) at a high luminance level of 100 cd m -2 . This work not only provides a promising approach for finely tuning the emission color of red phosphors via the easily accessible molecular design strategy, but also enables the establishment of an effective method for enriching phosphorescent-emitting molecules for practical applications, especially in the deep-red and near-infrared region (NIR).

As an important gas signaling molecule, hydrogen sulfide (H 2 S) plays a crucial role in regulating cold tolerance. H 2 S cooperates with phytohormones such as abscisic acid, ethylene, and salicylic acid to regulate the plant stress response. However, the synergistic regulation of H 2 S and auxin in the plant response to cold stress has not been reported. This study showed that sodium hydrosulfide (NaHS, an H 2 S donor) treatment enhanced the cold stress tolerance of cucumber seedlings and increased the level of auxin. CsARF5 , a cucumber auxin response factor (ARF) gene, was isolated, and its role in regulating H 2 S-mediated cold stress tolerance was described. Transgenic cucumber leaves overexpressing CsARF5 were obtained. Physiological analysis indicated that overexpression of CsARF5 enhanced the cold stress tolerance of cucumber and the regulation of the cold stress response by CsARF5 depends on H 2 S. In addition, molecular assays showed that CsARF5 modulated cold stress response by directly activating the expression of the dehydration-responsive element-binding (DREB)/C-repeat binding factor (CBF) gene CsDREB3 , which was identified as a positive regulator of cold stress. Taken together, the above results suggest that CsARF5 plays an important role in H 2 S-mediated cold stress in cucumber. These results shed light on the molecular mechanism by which H 2 S regulates cold stress response by mediating auxin signaling; this will provide insights for further studies on the molecular mechanism by which H 2 S regulates cold stress. The aim of this study was to explore the molecular mechanism of H 2 S regulating cold tolerance of cucumber seedlings and provide a theoretical basis for the further study of cucumber cultivation and environmental adaptability technology in winter.

Hydrogen sulfide (H 2 S) plays a crucial role in regulating chilling tolerance. However, the role of hydrogen peroxide (H 2 O 2 ) and auxin in H 2 S-induced signal transduction in the chilling stress response of plants was unclear. In this study, 1.0 mM exogenous H 2 O 2 and 75 μM indole-3-acetic acid (IAA) significantly improved the chilling tolerance of cucumber seedlings, as demonstrated by the mild plant chilling injury symptoms, lower chilling injury index (CI), electrolyte leakage (EL), and malondialdehyde content (MDA) as well as higher levels of photosynthesis and cold-responsive genes under chilling stress. IAA-induced chilling tolerance was weakened by N, N'-dimethylthiourea (DMTU, a scavenger of H 2 O 2 ), but the polar transport inhibitor of IAA (1-naphthylphthalamic acid, NPA) did not affect H 2 O 2 -induced mitigation of chilling stress. IAA significantly enhanced endogenous H 2 O 2 synthesis, but H 2 O 2 had minimal effects on endogenous IAA content in cucumber seedlings. In addition, the H 2 O 2 scavenger DMTU, inhibitor of H 2 O 2 synthesis (diphenyleneiodonium chloride, DPI), and IAA polar transport inhibitor NPA reduced H 2 S-induced chilling tolerance. Sodium hydrosulfide (NaHS) increased H 2 O 2 and IAA levels, flavin monooxygenase (FMO) activity, and respiratory burst oxidase homolog ( RBOH1 ) and FMO-like protein ( YUCCA2 ) mRNA levels in cucumber seedlings. DMTU, DPI, and NPA diminished NaHS-induced H 2 O 2 production, but DMTU and DPI did not affect IAA levels induced by NaHS during chilling stress. Taken together, the present data indicate that H 2 O 2 as a downstream signal of IAA mediates H 2 S-induced chilling tolerance in cucumber seedlings.

Tissue microenvironments are rich in signaling molecules. However, factors in the tissue matrix that can serve as tissue-specific cues for engineering pancreatic tissues have not been thoroughly identified. In this study, we performed a comprehensive proteomic analysis of porcine decellularized pancreatic extracellular matrix (dpECM). By profiling dpECM collected from subjects of different ages and genders, we showed that the detergent-free decellularization method developed in this study permits the preservation of approximately 62.4% more proteins than a detergent-based method. In addition, we demonstrated that dpECM prepared from young pigs contained approximately 68.5% more extracellular matrix proteins than those prepared from adult pigs. Furthermore, we categorized dpECM proteins by biological process, molecular function, and cellular component through gene ontology analysis. Our study results also suggested that the protein composition of dpECM is significantly different between male and female animals while a KEGG enrichment pathway analysis revealed that dpECM protein profiling varies significantly depending on age. This study provides the proteome of pancreatic decellularized ECM in different animal ages and genders, which will help identify the bioactive molecules that are pivotal in creating tissue-specific cues for engineering tissues in vitro.

Fish species substitution and fraud has become a worldwide economic issue in the seafood industry. In this study, an ultraviolet-visible (UV-Vis) spectroscopy-based method was developed for the identification of fish samples. Sixty fish samples from twelve commonly consumed fish species in China were analyzed as models to testify the protocol. The obtained results show that UV-Vis spectroscopy combined with chemometric analysis, such as principal component analysis (PCA), can accurately distinguish two fish species by boiling fish tissue sample in trifluoroacetic acid (TFA) solution for 2 min and analyzing the resultant samples using a UV-Vis spectrometer. The developed strategy was successfully applied to the classification and identification of fish samples on the market. It is a promising strategy that can be applied to the classification and authenticity testing of closely related fish species in order to detect and recognize fish substitution.

Crack is the main distress of asphalt pavement. Sealant is one of the most commonly used crack repair materials, and its performance is the key to affect the service life of asphalt pavements. In order to find an efficient modifier and optimize the performances of crack sealants. In this paper, carbon nanotubes (CNTs) and styrene-butadiene-styrene (SBS) were used as modifiers to prepare CNTs/SBS composite-modified asphalt crack sealant. The properties of the sealant were tested to evaluate its suitability for crack repair, which included the viscosity, softening point, resilience recovery, cone penetration, flow value, penetration, aging resistance, and fatigue resistance. The results showed that the conventional properties of the sealants meet the requirements of the specification. In addition, after heating aging, the elastic recovery rate of the sealant containing more CNTs decreased only slightly. The sealant containing 1 wt% CNTs exhibited a higher viscosity, fatigue resistance, thermal aging resistance.

The current statistics on cancer show that 90% of all human cancers originate from epithelial cells. Breast and prostate cancer are examples of common tumors of epithelial origin that would benefit from improved drug treatment strategies. About 90% of preclinically approved drugs fail in clinical trials, partially due to the use of too simplified in vitro models and a lack of mimicking the tumor microenvironment in drug efficacy testing. This review focuses on the origin and mechanism of epithelial cancers, followed by experimental models designed to recapitulate the epithelial cancer structure and microenvironment, such as 2D and 3D cell culture models and animal models. A specific focus is put on novel technologies for cell culture of spheroids, organoids, and 3D-printed tissue-like models utilizing biomaterials of natural or synthetic origins. Further emphasis is laid on high-content imaging technologies that are used in the field to visualize in vitro models and their morphology. The associated technological advancements and challenges are also discussed. Finally, the review gives an insight into the potential of exploiting nanotechnological approaches in epithelial cancer research both as tools in tumor modeling and how they can be utilized for the development of nanotherapeutics.

Acute lung injury (ALI) afflicts approximately 200,000 patients annually and has a 40% mortality rate. The COVID-19 pandemic has massively increased the rate of ALI incidence. The pathogenesis of ALI involves tissue damage from invading microbes and, in severe cases, the overexpression of inflammatory cytokines such as tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β). This study aimed to develop a therapy to normalize the excess production of inflammatory cytokines and promote tissue repair in the lipopolysaccharide (LPS)-induced ALI. Based on our previous studies, we tested the insulin-like growth factor I (IGF-I) and BTP-2 therapies. IGF-I was selected, because we and others have shown that elevated inflammatory cytokines suppress the expression of growth hormone receptors in the liver, leading to a decrease in the circulating IGF-I. IGF-I is a growth factor that increases vascular protection, enhances tissue repair, and decreases pro-inflammatory cytokines. It is also required to produce anti-inflammatory 1,25-dihydroxyvitamin D. BTP-2, an inhibitor of cytosolic calcium, was used to suppress the LPS-induced increase in cytosolic calcium, which otherwise leads to an increase in proinflammatory cytokines. We showed that LPS increased the expression of the primary inflammatory mediators such as toll like receptor-4 (TLR-4), IL-1β, interleukin-17 (IL-17), TNF-α, and interferon-γ (IFN-γ), which were normalized by the IGF-I + BTP-2 dual therapy in the lungs, along with improved vascular gene expression markers. The histologic lung injury score was markedly elevated by LPS and reduced to normal by the combination therapy. In conclusion, the LPS-induced increases in inflammatory cytokines, vascular injuries, and lung injuries were all improved by IGF-I + BTP-2 combination therapy.

Islands are one of the most sensitive interfaces between global changes and land and sea dynamic effects, with high sensitivity and low stability. Therefore, under the dynamic coupling effect of human activities and frequent natural disasters, the vulnerability of the ecological environment of islands shows the characteristics of complexity and diversity. For the protection of island ecosystems, a system for the assessment of island ecosystems and studies on the mechanism of island ecological vulnerability are highly crucial. In this study, the North and South Changshan Islands of China were selected as the study area. Considering various impact factors of island ecological vulnerability, the geographical information systems (GIS) spatial analysis, field surveys, data sampling were used to evaluate island ecological vulnerability. The Bayesian network model was used to explore the impact mechanism of ecological vulnerability. The results showed that the ecological vulnerability of the North Changshan Island is higher than that of the South Changshan Island. Among all the indicators, the proportion of net primary productivity (NPP) and the steep slope has the strongest correlation with ecological vulnerability. This study can be used as references in the relevant departments to formulate management policies and promote the sustainable development of islands and their surrounding waters.

This study was undertaken to test two therapies for acute kidney injury (AKI) prevention, IGF-1, which is renal protective, and BTP-2, which is a calcium entry (SOCE) inhibitor. We utilized lipopolysaccharide (LPS) IP, as a systemic model of AKI and studied in five groups of animals. Three experiments showed that at 7 days: (1) LPS significantly reduced serum IGF-1 and intramuscular IGF-I in vivo gene therapy rescued this deficiency. (2) Next, at the 7-day time point, our combination therapy，compared to the untreated group,caused a significant increase in survival, which was noteworthy because all of the untreated animals died in 72 hrs. (3) The four pathways associated with inflammation, including (A) increase in cytosolic calcium, (B) elaboration of proinflammatory cytokines, (C) impairment of vascular integrity, and (D) cell injury, were adversely affected in renal tissue by LPS, using a sublethal dose of LPS. The expression of several genes was measured in each of the above pathways. The combined therapy of IGF-1 and BTP-2 caused a favorable gene expression response in all four pathways. Our current study was an AKI study, but these pathways are also involved in other types of severe inflammation, including sepsis, acute respiratory distress syndrome, and probably severe coronavirus infection.

Glucose metabolism plays an important role in many normal and pathological physiological processes in the body. The breakdown and synthesis of muscle glycogen provides ATP for muscle activities. A genome-wide association study for muscle glycogen was performed in 473 Jingxing yellow chickens to identify significant single nucleotide polymorphisms (SNPs) and insertions and deletions (INDELs) involved in muscle glycogen metabolism. A total of nine SNPs ( p < 1/699341) and three INDELs ( p < 1/755733) reached a significant level of potential association. The following results were obtained through a series of analyses, including additive effects and gene function annotation. Two significant SNPs were found in introns 12 and 13 of copine 4 ( CPNE4 ) on chromosome 2. The wild-type and mutant individuals had significant differences in glycogen metabolism at two loci ( p < 0.01 for both). Individuals carrying two mutations had increased muscle glycogen content. According to the gene annotation of chromosome 11, there is a significant INDEL in intron 6 of naked cuticle homolog 1 ( NKD1 ). After the INDEL mutation, the glycogen content increased significantly. There was a significant difference between wild-type and mutant individuals ( p < 0.01). These mutations likely affecting two genes ( CPNE4 and NKD1 ) may affect glycogen storage in a pleiotropic manner. Gene annotation indicates that CPNE4 and NKD1 may affect the process of glucose metabolism. Our findings contribute to understanding the genetic regulation of muscle glycogen metabolism and provide theoretical support.

Abiotic stresses are major constraints limiting crop growth and production. Heat shock factors (Hsfs) play significant roles in mediating plant resistance to various environmental stresses, including heat, drought and salinity. In this study, we explored the biological functions and underlying mechanisms of wheat TaHsfA6f in plant tolerance to various abiotic stresses. Gene expression profiles showed that TaHsfA6f has relatively high expression levels in wheat leaves at the reproductive stage. Transcript levels of TaHsfA6f were substantially up-regulated by heat, dehydration, salinity, low temperature, and multiple phytohormones, but was not induced by brassinosteroids (BR). Subcellular localization analyses revealed that TaHsfA6f is localized to the nucleus. Overexpression of the TaHsfA6f gene in Arabidopsis results in improved tolerance to heat, drought and salt stresses, enhanced sensitivity to exogenous abscisic acid (ABA), and increased accumulation of ABA. Furthermore, RNA-sequencing data demonstrated that TaHsfA6f functions through up-regulation of a number of genes involved in ABA metabolism and signaling, and other stress-associated genes. Collectively, these results provide evidence that TaHsfA6f participates in the regulation of multiple abiotic stresses, and that TaHsfA6f could serve as a valuable gene for genetic modification of crop abiotic stress tolerance.

Wound dressings can accelerate wound healing. The degradable polymer poly(lactic acid) (PLA) shows good mechanical properties and biocompatibility. Sodium alginate (SA) holds good biocompatibility, hemostasis, and high hygroscopicity. Poly(vinyl alcohol) (PVA) has good spinnability as a pharmaceutical excipient. Herein, we carried out a comparison study of electrospun PLA and PLA/PVA/SA fiber membranes for wound healing in vitro and in vivo. In this study, PLA and PLA/PVA/SA nanofiber membranes were fabricated through electrospinning to produce a highly porous and large specific surface area that could promote wound healing. In vitro experiments showed that PLA and PLA/PVA/SA nanofiber membranes could all provide good support for the growth of rat fibroblasts (L929). Moreover, rat fibroblasts displayed slightly better adhesion and proliferation on PLA/PVA/SA than on the PLA fiber membranes. The in vivo potentiality of the PLA and PLA/PVA/SA fiber membranes was assessed in rat models of skin defects in which the PLA and PLA/PVA/SA fiber membranes significantly improved wound healing compared to commercially available gauzes. No significant differences in wound healing were observed between PLA and PLA/PVA/SA fiber membranes in our study. Furthermore, Masson staining and PCR displayed the PLA fiber membrane promoted protein deposition compared to the PLA/PVA/SA fiber membrane. In addition, IHC suggested that PLA/PVA/SA dressing reduced the inflammatory response during early wound healing compared to the PLA fiber membrane. The growing demand for lightweight, renewable, and excellent thermal insulation materials has fueled a search for high performance biomass materials with good mechanical compressibility and ultralow thermal conductivity. We propose a fabrication method for making a lightweight, anisotropic, and compressible wood aerogel with aligned cellulose fibers by a simple chemical treatment. The wood aerogel was mainly composed of highly aligned cellulose fibers with a relative crystallinity of 77.1%. The aerogel exhibits a low density of 32.18 mg/cm 3 and a high specific surface area of 31.68 m 2 /g due to the removal of lignin and hemicellulose from the wood. Moreover, the multilayer structure of the aerogel was formed under the restriction of wood rays. Combined with a nanoscale pore, the aerogel presents good compressibility and an ultralow thermal conductivity of 0.033 W/mK. This study was aimed at improving the conductivity of polylactic acid (PLA)-based composites by incorporating carbonaceous fillers. The composites with the addition of graphene nanoplatelets (rGO) or multi-walled carbon nanotubes (MWCNTs) were fabricated by the masterbatch melting method in order to improve the dispersion of the two kinds of nano-fillers. The results showed that, with the addition of 9 wt % rGO, the volume electrical resistivity of the composite reached the minimum electrical resistance of 10 3 Ω·m, at which point the conductive network in the composites was completely formed. The interfacial compatibility, apparent viscosity, and the thermal stability of the composite were also good. The rGO functionalized by sodium dodecylbenzene sulfonate (SDBS) was an efficient method to further improve the electrical conductivity of the composite, compared with tannic acid and MWCNTs. The resistivity was reduced by an order in magnitude. Patterns printed onto different baseplates by fused deposition modeling illustrated that the functionalized composite had certain flexibility and it is suitable for printing complex shapes.

In central lymphoid tissues, mature lymphocytes are generated and pathogenic autoreactive lymphocytes are deleted. However, it is currently known that a significant number of potentially pathogenic autoreactive lymphocytes escape the deletion and populate peripheral lymphoid tissues. Therefore, peripheral mechanisms are present to prevent these potentially pathogenic autoreactive lymphocytes from harming one's own tissues. One such mechanism is dictated by regulatory T (Treg) cells. So far, the most extensively studied Treg cells are CD4 + Foxp3 + Treg cells. However, recent clinical trials for the treatment of immune-mediated diseases using CD4 + Foxp3 + Treg cells met with limited success. Accordingly, it is necessary to explore the potential importance of other Treg cells such as CD8 + Treg cells. In this regard, one extensively studied CD8 + Treg cell subset is Qa-1(HLA-E in human)-restricted CD8 + Treg cells, in which Qa-1(HLA-E) molecules belong to a group of non-classical major histocompatibility complex Ib molecules. This review will first summarize the evidence for the presence of Qa-1-restricted CD8 + Treg cells and their regulatory mechanisms. Major discussions will then focus on the potential clinical translation of Qa-1-restricted CD8 + Treg cells. At the end, we will briefly discuss the current status of human studies on HLA-E-restricted CD8 + Treg cells as well as potential future directions.

A new pyrazine derivative, trypilepyrazinol ( 1 ), a new α-pyrone polyketide, (+)-neocitreoviridin ( 2 ), and a new ergostane analogue, 3β-hydroxyergosta-8,14,24(28)-trien-7-one ( 3 ), were isolated and characterized along with five known compounds from the marine-derived fungus Penicillium sp. IMB17-046. The structures of these new compounds were determined using spectroscopic data analyses (HRESIMS, 1D- and 2D-NMR), X-ray crystallography analysis, and TDDFT ECD calculation. Compounds 1 and 3 exhibited broad-spectrum antiviral activities against different types of viruses, including human immunodeficiency virus (HIV), hepatitis C virus (HCV), and influenza A virus (IAV), with IC 50 values ranging from 0.5 to 7.7 μM. Compounds 1 and 2 showed antibacterial activities against Helicobacter pylori , a causative pathogen of various gastric diseases, with minimum inhibitory concentration (MIC) values of 1-16 μg/mL.

The aim of the study was to improve the electrical and thermal conductivity of the polylactic acid/wood flour/thermoplastic polyurethane composites by Fused Deposition Modeling (FDM). The results showed that, when the addition amount of nano-graphite reached 25 pbw, the volume resistivity of the composites decreased to 10⁸ Ω·m, which was a significant reduction, indicating that the conductive network was already formed. It also had good thermal conductivity, mechanical properties, and thermal stability. The adding of the redox graphene (rGO) combined with graphite into the composites, compared to the tannic acid-functionalized graphite or the multi-walled carbon nanotubes, can be an effective method to improve the performance of the biocomposites, because the resistivity reduced by one order magnitude and the thermal conductivity increased by 25.71%. Models printed by FDM illustrated that the composite filaments have a certain flexibility and can be printed onto paper or flexible baseplates.

Sparse signal processing has already been introduced to synthetic aperture radar (SAR), which shows potential in improving imaging performance based on raw data or a complex image. In this paper, the relationship between a raw data-based sparse SAR imaging method (RD-SIM) and a complex image-based sparse SAR imaging method (CI-SIM) is compared and analyzed in detail, which is important to select appropriate algorithms in different cases. It is found that they are equivalent when the raw data is fully sampled. Both of them can effectively suppress noise and sidelobes, and hence improve the image performance compared with a matched filtering (MF) method. In addition, the target-to-background ratio (TBR) or azimuth ambiguity-to-signal ratio (AASR) performance indicators of RD-SIM are superior to those of CI-SIM in down-sampling data-based imaging, nonuniform displace phase center sampling, and sparse SAR imaging model-based azimuth ambiguity suppression.

In plants, the HAK (high-affinity K⁺)/KUP (K⁺ uptake)/KT (K⁺ transporter) family represents a large group of potassium transporters that play important roles in plant growth and environmental adaptation. Although HAK/KUP/KT genes have been extensively investigated in many plant species, they remain uncharacterized in wheat, especially those involved in the response to environmental stresses. In this study, 56 wheat HAK/KUP/KT (hereafter called TaHAK s) genes were identified by a genome-wide search using recently released wheat genomic data. Phylogenetic analysis grouped these genes into four clusters (Ι, II, III, IV), containing 22, 19, 7 and 8 genes, respectively. Chromosomal distribution, gene structure, and conserved motif analyses of the 56 TaHAK genes were subsequently performed. In silico RNA-seq data analysis revealed that TaHAK s from clusters II and III are constitutively expressed in various wheat tissues, while most genes from clusters I and IV have very low expression levels in the examined tissues at different developmental stages. qRT-PCR analysis showed that expression levels of TaHAK genes in wheat seedlings were significantly up- or downregulated when seedlings were exposed to K⁺ deficiency, high salinity, or dehydration. Furthermore, we functionally characterized TaHAK1b-2BL and showed that it facilitates K⁺ transport in yeast. Collectively, these results provide valuable information for further functional studies of TaHAKs , and contribute to a better understanding of the molecular basis of wheat development and stress tolerance.

In this study, a series of heat-induced shape memory composites was prepared by the hot-melt extrusion and three-dimensional (3D) printing of thermoplastic polyurethane (TPU) using wood flour (WF) with different contents of EPDM- g -MAH. The mechanical properties, microtopography, thermal property analysis, and heat-induced shape memory properties of the composites were examined. The results showed that, when the EPDM- g -MAH content was 4%, the tensile elongation and tensile strength of the composites reached the maximum value. The scanning electron microscopy and dynamic mechanical analysis results revealed a good interface bonding between TPU and WF when the EPDM- g -MAH content was 4%. The thermogravimetric analysis indicated that the thermal stability of TPU/WF composites was enhanced by the addition of 4% EPDM- g -MAH. Heat-induced shape memory test results showed that the shape memory performance of composites with 4% EPDM- g -MAH was better than that of unmodified-composites. The composites' shape recovery performance at a temperature of 60 °C was higher than that of the composites at ambient temperature. It was also found that, when the filling angle of the specimen was 45°, the recovery angle of the composites was larger.

We have previously reported that a porous membrane of polyethylene terephthalate (PET) enables significant augmentation of human pluripotent stem cell (hPSC) proliferation and differentiation. The interaction between hPSCs and the PET surface induces β-catenin-mediated wingless/integrated ( Wnt ) signaling, leading to upregulation of the expression of adhesion molecules in hPSCs. In this study, we sought to unveil mechanisms underlying the role of the PET membrane in hPSC self-renewal and metabolism. We discovered that physicochemical cues of the PET membrane considerably alter hPSC metabolism by increasing the cell yield and suppressing the generation of toxic byproduct, indicating an effective cell self-renewal and a less apoptotic culture environment in the membrane culture system. Furthermore, we discovered that a caspase-8 medicated apoptotic pathway plays a profound role in obstructing hPSCs grown on a traditional tissue culture plate (TCP). Treating hPSCs seeded on a TCP surface with a caspase-8 inhibitor significantly suppressed cellular apoptotic pathway and improved cell proliferation and metabolism. Our experimental results provided valuable insights into signal pathways influencing hPSC self-renewal during routine maintenance and expansion, which would shed light on large-scale preparation of hPSCs for clinical applications.
