The Kaladawan iron deposit is located in the North Altyn Tagh and exhibits occurrences of iron ore bodies at the contact zone between Ordovician magmatic rocks (basalts, rhyolite, and granodiorite) and marble. However, controversy persists regarding the genetic classification and metallogenic mechanism of this deposit. Through a field investigation, single mineral in situ geochemical analysis, whole-rock geochemical analysis, and Fe isotope determination, the following conclusions are made: (1) Ti-(Ni/Cr) and (V/Ti)-Fe diagrams indicate that the magnetite from all studied rocks underwent hydrothermal metasomatism, while (Ni/(Cr + Mn))-(Ti + V) and (Ca + Al + Mn)-(Ti + V) diagrams suggest a skarn origin for these magnetites. Therefore, it can be inferred that the Kaladawan iron deposit is skarn-type. (2) The iron ore exhibits similar rare-earth-element characteristics to the altered basalt. Additionally, the altered basalts (δ56Fe = 0.024~0.100‰) are more enriched in light Fe isotopes than the unaltered basalts (δ56Fe = 0.129~0.197‰) at the same location, indicating that the ore-forming materials of the Kaladawan iron ore are mainly derived from basaltic rocks. (3) According to the law of mass conservation and the intermediate Fe isotopic composition of the iron ore between the granodiorite and basalt, the hydrothermal fluid for the formation of iron ores was inferred to be derived from the late intrusive granodiorite. To address the time-optimal trajectory planning (TOTP) problem with joint jerk constraints in a Cartesian coordinate system, we propose a time-optimal path-parameterization (TOPP) algorithm based on nonlinear optimization. The key insight of our approach is the presentation of a comprehensive and effective iterative optimization framework for solving the optimal control problem (OCP) formulation of the TOTP problem in the (s , s ˙) -phase plane. In particular, we identify two major difficulties: establishing TOPP in Cartesian space satisfying third-order constraints in joint space, and finding an efficient computational solution to TOPP, which includes nonlinear constraints. Experimental results demonstrate that the proposed method is an effective solution for time-optimal trajectory planning with joint jerk limits, and can be applied to a wide range of robotic systems. Alkaline pectate lyases have biotechnological applications in plant fiber processing, such as ramie degumming. Previously, we characterized an alkaline pectate lyase from Bacillus clausii S10, named BacPelA, which showed potential for enzymatic ramie degumming because of its high cleavage activity toward methylated pectins in alkaline conditions. However, BacPelA displayed poor thermo-alkaline stability. Here, we report the 1.78 Å resolution crystal structure of BacPelA in apo form. The enzyme has the characteristic right-handed β-helix fold of members of the polysaccharide lyase 1 family and shows overall structural similarity to them, but it displays some differences in the details of the secondary structure and Ca2+-binding site. On the basis of the structure, 10 sites located in flexible regions and showing high B-factor and positive ΔTm values were selected for mutation, aiming to improve the thermo-alkaline stability of the enzyme. Following site-directed saturation mutagenesis and screening, mutants A238C, R150G, and R216H showed an increase in the T5015 value at pH 10.0 of 3.0 °C, 6.5 °C, and 7.0 °C, respectively, compared with the wild-type enzyme, interestingly accompanied by a 24.5%, 46.6%, and 61.9% increase in activity. The combined mutant R150G/R216H/A238C showed an 8.5 °C increase in the T5015 value at pH 10.0, and an 86.1% increase in the specific activity at 60 °C, with approximately doubled catalytic efficiency, compared with the wild-type enzyme. Moreover, this mutant retained 86.2% activity after incubation in ramie degumming conditions (4 h, 60 °C, pH 10.0), compared with only 3.4% for wild-type BacPelA. The combined mutant increased the weight loss of ramie fibers in degumming by 30.2% compared with wild-type BacPelA. This work provides a thermo-alkaline stable, highly active pectate lyase with great potential for application in the textile industry, and also illustrates an effective strategy for rational design and improvement of pectate lyases. An ultrasensitive and renewable electrochemiluminescence (ECL) immunosensor was developed for the detection of tumor markers by combining a newly designed trace tag and streptavidin-coated magnetic particles (SCMPs). The trace tag (RuL@SiO2-Au∼RuL-Ab2) was prepared by loading Ru(bpy)3 2+(RuL)-conjuged secondary antibodies (RuL-Ab2) on RuL@SiO2 (RuL-doped SiO2) doped Au (RuL@SiO2-Au). To fabricate the immunosensor, SCMPs were mixed with biotinylated AFP primary antibody (Biotin-Ab1), AFP, and RuL@SiO2-Au∼RuL-Ab2 complexes, then the resulting SCMP/Biotin-Ab1/AFP/RuL@SiO2-Au∼RuL-Ab2 (SBAR) sandwich-type immunocomplexes were absorbed on screen printed carbon electrode (SPCE) for detection. The immunocomplexes can be easily washed away from the surface of the SPCE when the magnetic field was removed, which made the immunosensor reusable. The present immunosensor showed a wide linear range of 0.05-100 ng mL-1 for detecting AFP, with a low detection limit of 0.02 ng mL-1 (defined as S/N = 3). The method takes advantage of three properties of the immunosensor: firstly, the RuL@SiO2-Au∼RuL-Ab2 composite exhibited dual amplification since SiO2 could load large amount of reporter molecules (RuL) for signal amplification. Gold particles could provide a large active surface to load more reporter molecules (RuL-Ab2). Accordingly, through the ECL response of RuL and tripropylamine (TPA), a strong ECL signal was obtained and an amplification analysis of protein interaction was achieved. Secondly, the sensor is renewable because the sandwichtype immunocomplexes can be readily absorbed or removed on the SPCE's surface in a magnetic field. Thirdly, the SCMP modified probes can perform the rapid separation and purification of signal antibodies in a magnetic field. The full-length cDNA sequence of F3′5′H gene from the Brunfelsia acuminata was obtained by RT-PCR and RACE, whose GenBank accession number is JQ678765. The sequence contains a 1521 bp open reading frame, 120 bp 5′UTR and 61 bp 3′UTR, encoding a total of 506 amino acids. The molecular mass of the predicted protein is 56.47 kDa with an estimated pI of 8.78, respectively. Sequence alignment showed that the amino acid sequence of F3′5′H was 91%, 87% and 84% with that of Petunia × hybrida, Nierembergia sp., Solanum tuberosum, respectively. Real-time quantitative PCR analysis showed that the expression of F3′5′H gene was different in petals of different days, which was the highest expression level on day 0 and significantly higher than other days. Tree growth in mountain ecosystems is affected by complex environments, and its relationship with climatic and environmental factors varying with elevation. In order to examine the spatial variation and temporal stability of the growth/climate relationship of Betula platyphylla (BP), the dendrochronological method was used to analyze the radial growth/climate relationship between 1946 and 2016 of the BP trees along an altitudinal gradient in the Changbai Mountain of northeast China. Our results showed that the mean sensitivity of BP was higher than that of other species studied in Changbai Mountain. The growth/climate relationship of BP trees varies with altitude, and this conclusion has reached a consensus from the study of tree growth response to climate change. More specifically, at low altitudes (550–995 m a.s.l.), the radial growth of BP is mainly affected by spring precipitation and temperature in May and October of the current year. However, at high-altitude areas (1210–1425 m a.s.l.), it is mainly affected by the temperature in September of the previous year and May of the current year. Furthermore, the growth/climate relationship of BP trees showed temporal instability. After 1970, the rise in temperature inhibited the growth of BP at low altitudes and promoted the growth of BP trees at high altitudes. The relationships between migration and housing congestion have attracted attention in engaging the public against the COVID-19 pandemic and some other public health crises. In recent years in China, promoting the citizenization ("shimin hua") of migrants and improving the quality of urbanization have become the focus of attention in the new-type urbanization today. The housing space consumption of migrants is one of the important indices to look into regarding their real living status in the receiving cities: how do the housing consumption behavior and residential quality vary between the local, inter- and intra-provincial migratory patterns? This article uses the micro household data of the 1% population sampling survey conducted in 2015 by the National Bureau of Statistics of China to look into the spatial variance of the aggregate housing space consumption behaviors of the local and non-local population at the prefectural level and above in urban China. This study finds that: (a) the longer migratory pattern indicates a thriftier housing space consumption that implies a higher probability of residential overcrowding among the inter-provincial migrants; at the same time, the locals enjoy the greater living comfort than their migrant peers; (b) the spatial variance in terms of housing space consumption can be attributed to a series of destination city contexts, such as the geological background, city administrative rank, areal location, local-nonlocal demography, municipal economic growth, and the local residential development levels. The results show that the more "targeted" housing policies are needed to solve the housing difficulties with migrant workers for a goal of human-centered urbanization development. Although we lack the more detailed data-sets to examine the correlation between public health risks (like the COVID-19 pandemic) and housing congestion problems (especially with the population on the move), this research is still illuminating in terms of how to cut down the public health risk in a highly mobile and rapidly urbanizing context like China. Pinus massoniana and P. hwangshanensis are two conifer species located in southern China, which are of both economic and ornamental value. Around the middle and lower reaches of the Yangtze River, P. massoniana occurs mainly at altitudes below 700 m, while P. hwangshanensis can be found above 900 m. At altitudes where the distribution of both pines overlaps, a natural introgression hybrid exists, which we will further refer to as the Z pine. This pine has a morphological character that shares attributes of both P. massoniana and P. hwangshanensis. However, compared to the other two pines, its reproductive structure, the pinecone, has an ultra-low ripening rate with seeds that germinate poorly. In this study, we aimed to find the reason for the impaired cone maturation by comparing transcriptome libraries of P. massoniana and Z pine cones at seven successive growth stages. After sequencing and assembly, we obtained unigenes and then annotated them against NCBI's non-redundant nucleotide and protein sequences, Swiss-Prot, Clusters of Orthologous Groups, Gene Ontology and KEGG Orthology databases. Gene expression levels were estimated and differentially expressed genes (DEGs) of the two pines were mined and analyzed. We found that several of them indeed relate to reproductive process. At every growth stage, these genes are expressed at a higher level in P. massoniana than in the Z pine. These data provide insight into understanding which molecular mechanisms are altered between P. massoniana and the Z pine that might cause changes in the reproductive process. [ABSTRACT FROM AUTHOR]

The purpose of the study was to develop a facile method for the fabrication of a stable and reusable magnetic graphene composite absorbent to remove trace levels of polybrominated diphenyl ethers in water treatment. The poly cationic Fe₃O₄@PDDA (poly(diallyldimethyl ammonium chloride) (PDDA)) core-shell structured nanoparticles were first synthesized, and then, DNA was laid on the surface of graphene oxide (GO x ) to prepare the polyanionic GO x @DNA composite. The above materials were then mixed together and adhered together through sol-gel technology. Thus, the Fe₃O₄@PDDA/GO x @DNA composite absorbent was prepared. Its performance was tested by disperse solid phase extraction and gas chromatography/mass spectrometric (GC/MS) for removing six kinds of indicative polybrominated diphenyl ethers (BDEs) in water samples. The removal percentages of several real samples for six kinds of BDEs (BDE17, BDE28, BDE 71, BDE 47, BDE 66, BDE 100) at the ng/mL order of magnitude were in the range of 88.2%-99.1%. The removal percentage still reached 80.0% when the adsorbent was reused at least 20 times. The results suggested that the magnetic absorbent can obviously remove trace levels of BDEs from large volumes of aqueous solutions in environmental pollution cleanup with high removal efficiency.

The CdTe quantum dots (QDs), graphene nanocomposite (CdTe-G) and dextran-Fe3O4 magnetic nanoparticles have been synthesized for developing an ultrasensitive electrochemiluminescence (ECL) immunoassay for Carcinoembryonic antigen 19-9 (CA 19-9) in serums. Firstly, the capture probes (CA 19-9 Ab1/Fe3O4) for enriching CA 19-9 were synthesized by immobilizing the CA 19-9's first antibody (CA 19-9 Ab1) on magnetic nanoparticles (dextran-Fe3O4). Secondly, the signal probes (CA 19-9 Ab2/CdTe-G), which can emit an ECL signal, were formed by attaching the secondary CA 19-9 antibody (CA 19-9 Ab2) to the surface of the CdTe-G. Thirdly, the above two probes were used for conjugating with a serial of CA 19-9 concentrations. Graphene can immobilize dozens of CdTe QDs on their surface, which can emit stronger ECL intensity than CdTe QDs. Based on the amplified signal, ultrasensitive antigen detection can be realized. Under the optimal conditions, the ECL signal depended linearly on the logarithm of CA 19-9 concentration from 0.005 to 100 pg/mL, and the detection limit was 0.002 pg/mL. Finally, five samples of human serum were tested, and the results were compared with a time-resolved fluorescence assay (TRFA). The novel immunoassay provides a stable, specific and highly sensitive immunoassay protocol for tumor marker detection at very low levels, which can be applied in early diagnosis of tumor.

An ultrasensitive electrochemiluminescent immunoassay (ECLIA) for aflatoxins M1 (ATM1) in milk using magnetic Fe3O4-graphene oxides (Fe-GO) as the absorbent and antibody-labeled cadmium telluride quantum dots (CdTe QDs) as the signal tag is presented. Firstly, Fe3O4 nanoparticles were immobilized on GO to fabricate the magnetic nanocomposites, which were used as absorbent to ATM1. Secondly, aflatoxin M1 antibody (primary antibody, ATM1 Ab1), was attached to the surface of the CdTe QDs-carbon nanotubes nanocomposite to form the signal tag (ATM1 Ab1/CdTe-CNT). The above materials were characterized. The optimal experimental conditions were obtained. Thirdly, Fe-GO was employed for extraction of ATM1 in milk. Results indicated that it can adsorb ATM1 efficiently and selectively within a large extent of pH from 3.0 to 8.0. Adsorption processes reached 95% of the equilibrium within 10 min. Lastly, the ATM1 with a serial of concentrations absorbed on Fe-GO was conjugated with ATM1 Ab1/CdTe-CNT signal tag based on sandwich immunoassay. The immunocomplex can emit a strong ECL signal whose intensity depended linearly on the logarithm of ATM1 concentration from 1.0 to 1.0 × 10(5) pg/mL, with the detection limit (LOD) of 0.3 pg/mL (S/N = 3). The method was more sensitive for ATM1 detection compared to the ELISA method. Finally, ten samples of milk were tested based on the immunoassay. The method is fast and requires very little sample preparation, which was suitable for high-throughput screening of mycotoxins in food.

An ultrasensitive portable electrochemical immunosensor for human immunodeficiency virus p24 (HIV p24) antigen detection has been developed, whereby the detection sensitivity was 1000 times higher than that of the ELISA method. Firstly, a novel HRP enzyme-antibody copolymer (EV-p24 Ab2) was synthesized through an EnVision regent (EV, a dextrin amine skeleton anchoring more than 100 molecules of HRP and 15 molecules of anti IgG), then incubated in the secondary antibody of p24. Secondly, the copolymer was immobilized on the gold nanocolloids (AuNPs) to fabricate a novel signal tag (AuNPs/EV-p24 Ab2). Subsequently, a sandwich-type immunoreaction would take place between the capture probe (silicon dioxide-coated magnetic Fe₃O₄ nanoparticles (MNPs) labeled with the primary p24 antibody (MNPs-p24 Ab1)), p24 (different concentrations) and the signal tag [AuNPs/EV-p24 Ab2)] to form the immunocomplex. Finally, the immunocomplex was absorbed on the surface of screen printed carbon electrode (SPCE) by a magnet and immersed in the o-hydroxyl phenol (HQ) and H₂O₂. The large amounts of HRP on the signal tag can catalyze the oxidation of HQ by H₂O₂, which can induce an amplified reductive current. Moreover, the capture probe could improve the accumulation ability of p24 and facilitate its separation from the substrate through the magnet. Under optimal conditions, the proposed immunoassay exhibited good sensitivity to p24 within a certain concentration range from 0.001 to 10.00 ng/mL, with a detection limit of 0.5 pg/mL (S/N = 3). The proposed method can be used for real-time and early detection of HIV-infected people.

In this paper, Fe(3)O(4) nanoparticles (Fe(3)O(4) NPs) grafted carboxyl groups of multiwalled carbon nanotubes with cationic polyelectrolyte poly (dimethyldiallylammonium chloride) (PDDA) (MWCNTs-COO(-)/PDDA@Fe(3)O(4)), are successfully synthesized and used for the extraction of six kinds of major toxic polychorinated biphenyls (PCBs) from a large volume of water solution. The hydrophilicity of the PDDA cage can enhance the dispersibility of sorbents in water samples, and the superparamagnetism of the Fe(3)O(4) NPs facilitate magnetic separation which directly led to the simplification of the extraction procedure. With the magnetic solid-phase extraction (MSPE) technique based on the MWCNTs-COO(-)/PDDA@Fe(3)O(4) sorbents, it requires only 30 min to extract trace levels of PCBs from 500 mL water samples. When the eluate condensed to 1.0 mL, concentration factors for PCBs became over 500. The spiked recoveries of several real water samples for PCBs were in the range of 73.3-98.9% with relative standard deviations varying from 3.8% to 9.4%, reflecting good accuracy of the method. Therefore, preconcentration of trace level of PCBs by using this MWCNTs-COO(-)/PDDA@Fe(3)O(4) sorbent, which are stable for multiple reuses, from water solution can be performed.

A novel electrochemical immunosensor for tumor biomarker detection based on three-dimensional, magnetic and electroactive nanoprobes was developed in this study. To fabricate the nanoprobes, negatively charged Fe(3)O(4) nanoparticles (Fe(3)O(4) NPs) and gold nanoparticles (Au NPs) were first loaded on the surface of multiple wall carbon nanotubes (MCNTs) which were functioned with redox-active hemin and cationic polyelectrolyte poly(dimethyldiallylammonium chloride) (PDDA). Using alpha fetoprotein (AFP) as a model analyte, AFP antibody (anti-AFP) was absorbed on the surface of Au NPs, bovine serum albumin (BSA) was then used to block sites against non-specific binding, and finally formed anti-AFP/Au NPs/Fe(3)O(4)/hemin/MCNTs named anti-AFP nanoprobes. When the target antigen AFP was present, it interacted with anti-AFP and formed an antigen-antibody complex on the nanoprobe interface. This resulted in a decreased electrochemical signal of hemin for quantitative determination of AFP when immobilized onto the screen-printed working electrode (SPCE). The results showed that the nanoprobe-based electrochemical immunosensor was sensitive to AFP detection at a concentration of 0.1 to 200 ng·mL(-1) with a detection limit of 0.04 ng·mL(-1), it also demonstrated good selectivity against other interferential substances. The electroactive nanoprobes can be massively prepared, easily immobilized on the SPCE for target detection and rapidly renewed with a magnet. The proposed immunosensor is fast, simple, sensitive, stable, magnet-controlled, nontoxic, label-free and reproducible.

A novel supramolecular amperometric immunosensor for the determination of Human Immunodeficiency Virus antigen p24 (HIV p24) was built up using the electrostatic layer-by-layer self-assembly technique upon a gold electrode with HIV p24 antibody (anti-p24) being immobilized on polyelectrolyte/gold nanoparticle multilayer films. The multilayer films were composed of poly(L-lysine) (pLys) and mercaptosuccinic acid (MSA) stabilized Fe(3)O(4)(core)/gold(shell) nano particles (GMPs).The immunosensor preparation steps were monitored by X-ray fluorescence spectrometry (XRFS), scanning electron microscopy (SEM) and transmission electron microscopy (TEM). In pH 6.5 PBS, after the immunosensor was incubated with HIV p24 solution at 25 degrees C for 5 min, the electron transfer access of FeCN is partially inhibited, which leads to a linear decrease of peak current. In addition, the performance of the immunosensor was studied in detail. It offers high-sensitivity for the detection of p24 and has good correlation for the detection of p24 in the range of 0.1 to 100.0 ng/mL with a detection limit of 0.05 ng/mL estimated at a signal-to-noise ratio of 3. The proposed immunosensor was used to analyze p24 in human serum specimens and the results showed the developed immunosensor provides a promising alternative approach for detecting p24 in the early diagnosis of AIDS patients.
