Simple Summary: In this work, we documented the presence of two neurotrophins NGF (Nerve Growth Factor) and BDNF (Brain-Derived Neurotrophic Factor) in the pancreas of two species of marine mammals, Tursiops truncatus (common bottlenose dolphin) and Otaria flavescens (South American sea lion). Pancreas samples were provided by the Mediterranean Marine Mammal Tissue Bank of the Department of Comparative Biomedicine and Food Science of the University of Padua (Italy). These two growth factors are well-known mediators for maintaining the survival of neurons but also exert a metabotrophic effect. The localization in the pancreas of bottlenose dolphins and sea lions of these neurotrophins could help to better understand how these marine animals handle metabolic challenges in their adaptation to ocean life. In this study, we have investigated the immunolocalization of NGF (Nerve Growth Factor) and BDNF (Brain-Derived Neurotrophic Factor) in the pancreas of two species of marine mammals: Tursiops truncatus (common bottlenose dolphin), belonging to the order of the Artiodactyla, and Otaria flavescens (South American sea lion), belonging to the order of the Carnivora. Our results demonstrated a significant presence of NGF and BDNF in the pancreas of both species with a wide distribution pattern observed in the exocrine and endocrine components. We identified some differences that can be attributed to the different feeding habits of the two species, which possess a different morphological organization of the digestive system. Altogether, these preliminary observations open new perspectives on the function of neurotrophins and the adaptive mechanisms of marine mammals in the aquatic environment, suggesting potential parallels between the physiology of marine and terrestrial mammals. Simple Summary: The use of by-products from the olive oil industry in livestock farming is an important resource due to their high content of biomolecules with strong antioxidant properties. In this study, 30 Italian Holstein–Friesian cows were fed with an Olea europaea L. polyphenol-enriched diet (500 mg/cow/day) to evaluate the effects of such supplementation on metabolic parameters and milk quality. The enriched diet helped (1) maintain cows' metabolic parameters in the physiological range producing hypoglycemic and hypolipidemic effects, and (2) improve the fatty acid profile of milk, paralleled by an increase in protein content and lactose. The obtained results show that in lactating dairy cows, supplementation with Olea europaea L. extracts can be a valid strategy to improve animal welfare and milk quality. Here, we evaluated the effect of dietary supplementation with an Olea europaea L. extract on the animal welfare and milk quality of dairy cows. Thirty Italian Holstein–Friesian dairy cows in the mid-lactation phase (90 to 210 days) were blocked into experimental groups based on parity class (namely, primiparous (P) (n = 10), secondiparous (S) (n = 10) and pluriparous (PL) (n = 10)) and received, for 60 days, Phenofeed Dry® at 500 mg/cow/day. Milk and blood samples were collected before the start of the treatment (T0), subsequently every 15 days (T1–T4) and at 45 days after the end of treatment (T5). In the serum, glucose and triglycerides, stress, the thyroid, lactation and sex hormones were measured; in the milk, lysozyme content as well as the fatty acid profile were assessed. In the whole animal, the enriched feed helped to maintain hormonal parameters in the physiological range while producing hypoglycemic (T4 vs. T0, for P and PL p < 0.001) and hypolipidemic effects (T4 vs. T0, for P p < 0.001 and for PL p < 0.01). At the milk level, it resulted in a reduction in total fat (T5 vs. T0, for P, S and PL p < 0.001) and in the saturated fatty acids (SFAs)/monounsaturated fatty acids (MUFAs) ratio paralleled by an increase in polyunsaturated fatty acids (PUFAs) (T5 vs. T0, for P, S and PL p < 0.001), protein content (lysozyme (T4 vs. T0, for P and PL p < 0.001)) and lactose (T5 vs. T0, for P, S and PL p < 0.001). This study aimed to examine the impact of different surface properties of poly(lactic-co-glycolic) acid (PLGA) nanoparticles (P NPs) and PLGA-Poloxamer nanoparticles (PP NPs) on their in vivo biodistribution. For this purpose, NPs were formulated via nanoprecipitation and loaded with diphenylhexatriene (DPH), a fluorescent dye. The obtained NPs underwent comprehensive characterization, encompassing their morphology, technological attributes, DPH release rate, and thermodynamic properties. The produced NPs were then administered to wild-type mice via intraperitoneal injection, and, at scheduled time intervals, the animals were euthanized. Blood samples, as well as the liver, lungs, and kidneys, were extracted for histological examination and biodistribution analysis. The findings of this investigation revealed that the presence of poloxamers led to smaller NP sizes and induced partial crystallinity in the NPs. The biodistribution and histological results from in vivo experiments evidenced that both, P and PP NPs, exhibited comparable concentrations in the bloodstream, while P NPs could not be detected in the other organs examined. Conversely, PP NPs were primarily sequestered by the lungs and, to a lesser extent, by the kidneys. Issues such as the design or reauditing of dams due to the occurrence of extreme events caused by climatic change are mandatory to address to ensure the safety of territories. These topics may be tackled numerically with Computational Fluid Dynamics and experimentally with physical models. This paper describes the 1:60 Froude-scaled numerical model of the Liscione (Guardialfiera, Molise, Italy) dam spillway and the downstream stilling basin. The k-ω SST turbulence model was chosen to close the Reynolds-averaged Navier–Stokes equations (RANS) implemented in the commercial software Ansys Fluent ®. The computation domain was discretized using a grid with hexagonal meshes. Experimental data for model validation were gathered from the 1:60 scale physical model of the Liscione dam spillways and the downstream riverbed of the Biferno river built at the Laboratory of Hydraulic and Maritime Constructions of the Sapienza University of Rome. The model was scaled according to the Froude number and fully developed turbulent flow conditions were reproduced at the model scale (Re > 10,000). From the analysis of the results of both the physical and the numerical models, it is clear that the stilling basin is undersized and therefore insufficient to manage the energy content of the fluid output to the river, with a significant impact on the erodible downstream river bottom in terms of scour depths. Simple Summary: Nesfatin-1 (Nesf-1) is a neuropeptide that plays important roles in regulating food intake, mainly related to its anorexigenic effect, and it is mainly distributed in the digestive systems of all vertebrates. With this study, we expand knowledge on the localization of Nesf-1 in the digestive tract of an aquatic mammalian species, the common bottlenose dolphin (Tursiops truncatus), allowing comparative study on terrestrial mammals. Dolphin tissue samples (three gastric chambers and intestine) were provided by the Mediterranean Marine Mammal Tissue Bank of the Department of Comparative Biomedicine and Food Science of the University of Padova (Italy). First identified as an anorexigenic peptide, in the last decades, several studies have suggested that Nesfatin-1 (Nesf-1) is a pleiotropic hormone implicated in numerous regulatory processes in peripheral organs and tissues. In vertebrates, Nesf-1 is indeed expressed in the central nervous system and peripheral organs. In this study, we characterized the pattern of Nesf-1 distribution within the digestive tract of the common bottlenose dolphin (Tursiops truncatus), composed of three gastric chambers and an intestine without a clear subdivision in the small and large intestine, also lacking a caecum. Our results indicated that Nesf-1 is widely distributed in cells of the mucosal epithelium of the gastric chambers. Most of the immunoreactivity was observed in the second chamber, compared to the first and third chambers. Immunopositivity was also found in nerve fibers and neurons, scattered or/and clustered in ganglion structures along all the examined gastrointestinal tracts. The invasive macroalga Caulerpa cylindracea has spread widely in the Mediterranean Sea, becoming a favorite food item for native fish for reasons yet unknown. By using a combination of behavioral, morphological, and molecular approaches, herein we provide evidence that the bisindole alkaloid caulerpin, a major secondary metabolite of C. cylindracea, significantly increases food intake in the model fish Danio rerio, influencing the regulation of genes involved in the orexigenic pathway. In addition, we found that the compound improves fish reproductive performance by affecting the hypothalamus–pituitary–gonadal axis. The obtained results pave the way for the possible valorization of C. cylindracea as a sustainable source of a functional feed additive of interest to face critical challenges both in aquaculture and in human nutrition. NPY is among the most abundant neuropeptides in vertebrate brain and is primarily involved in the regulation of food intake. The NPY system is also associated with the aging process showing beneficial effects on neuronal survival via autophagy modulation. Here, we explore the age-related regulation of NPY in the brain and foregut of the shortest- and longest-lived fish species, Nothobranchius furzeri and Danio rerio, respectively. These two research models, despite some similarities, display profound biological differences making them attractive vertebrates to elucidate the mechanisms underlying the regulation of neuropeptide synthesis and function. It is noteworthy that in both fish species only Npya has been identified, while in the other teleosts two classes of NPY (Npya and Npyb) have been annotated. Our findings document that in both species: (i) NPY is centrally regulated; (ii) NPY levels increase in the brain during aging; (iii) NPY is localized in the enteroendocrine cells as well as in the myenteric plexus and drastically decreases in old animals. According to our data, the age-related regulation in the gut resembles that described in other vertebrate species while the increased levels in the brain offer the unique possibility to explore the role of NPY in model organisms to develop future experimental and translatable approaches. Floods can transport debris of a very wide range of dimensions, from cohesive sediments to large floating debris, such as trees and cars. The latter increases the risk associated with floods by, for example, obstructing the flow or damaging structures due to impact. The transport of this type of debris and their interaction with structures are often studied experimentally in the context of tsunamis and flash floods. Numerical studies on this problem are rare, therefore the present study focuses on the numerical modelling of the flow-debris interaction. This is achieved by simulating multiple laboratory experiments, available in the literature, of a single buoyant container transported by a dam-break flow in order to validate the chosen numerical approach. The numerical simulations are carried using the open source DualSPHysics model based on the smoothed particle hydrodynamics method coupled with the multi-physics engine CHRONO, which handles the container–bottom interactions. The trajectory, as well as the velocity of the centroid of the container, were tracked throughout the simulation and compared with the same quantities measured in the laboratory. The agreement between the model and the experiment results is quantitatively assessed using the normalised root mean squared error and it is shown that the model is accurate in reproducing the floating container trajectory and velocity. Collagens are the most abundant proteins in vertebrates and constitute the major components of the extracellular matrix. Collagens play an important and multifaceted role in the development and functioning of the nervous system and undergo structural remodeling and quantitative modifications during aging. Here, we investigated the age-dependent regulation of col4a1 and col25a1 in the brain of the short-lived vertebrate Nothobranchius furzeri, a powerful model organism for aging research due to its natural fast-aging process and further characterized typical hallmarks of brain aging in this species. We showed that col4a1 and col25a1 are relatively well conserved during vertebrate evolution, and their expression significantly increases in the brain of N. furzeri upon aging. Noteworthy, we report that both col4a1 and col25a1 are expressed in cells with a neuronal phenotype, unlike what has already been documented in mammalian brain, in which only col25a1 is considered a neuronal marker, whereas col4a1 seems to be expressed only in endothelial cells. Overall, our findings encourage further investigation on the role of col4a1 and col25a1 in the biology of the vertebrate brain as well as the onset of aging and neurodegenerative diseases. Erosion at either dam or spillway foundations, destabilization in riverbanks, and damage in the natural environment located downstream of either dams or spillways represent crucial elements to be taken into account in the risk assessment of hydraulic structures. One of the main problems is related to the scouring that water flow may induce at the downstream boundary of spillways. This issue is exacerbated in the case of undersized stilling basins, i.e., when a significant level of energy migrates downstream by acting on unprotected natural riverbed. If the scour depths are large enough, the structural stability of the infrastructure will be threatened. This paper aims to illustrate an innovative technical solution suitable to protect the riverbed located just downstream of stilling basins by means of artificial Antifer blocks. These kinds of artificial blocks are widely used in the field of maritime construction, but in the literature, there are no theoretical formulations for their design within the frame of river engineering. In order to demonstrate the efficacy of the proposed technical solution, it is applied to a real case investigated by means of physical modeling. The riverbed located just downstream of the stilling basin of Liscione Dam (Campobasso, Italy) experienced scour due to high discharges during and after extreme rain events. Different protection strategies have been tested to assess the influence of different placement methods and packing densities on the stability of Antifer block armor layers. Simple Summary: In the last decades the European ban towards antibiotics resulted in an increase of the number of studies on the effects of natural feed additives, that can enhance the health of farm animals intended for human consumption. Polyphenols such as flavanones and anthocyanins (responsible of the red, purple or blue colors) are bioactive compounds found in fruits and vegetables. Polyphenols possess multiple pharmacological characteristics, like antioxidant, anti-inflammatory and immunostimulant properties. Although many of the biological effects of polyphenols are known, only a limited number of studies has been focused on the effects of their supplementation in ruminant diet. Therefore, we evaluated the effect of a diet supplemented with a standardized powder extract, red (blood) orange and lemon extract (RLE), rich in flavanones, anthocyanins and other polyphenols on the neuropeptide Y (NPY) distribution in the gastro–entero–pancreatic system of goat kids. In mammals, NPY occurs in both the central and peripheral nervous systems and it is involved in the control of different physiological processes, including food intake regulation. For the first time, we document that NPY is widely distributed in the abomasum, duodenum and pancreas of goat kids and that significantly increases in the abomasum and pancreas of RLE supplemented feed animals. The use of natural compounds as feed additive is also increasing in farm animals, thanks to the beneficial effect on both animals and consumers health. Here, we questioned whether natural extracts, such as red orange and lemon extract (RLE) rich in flavanones, anthocyanins, and other polyphenols, used as feed additives could display an effect on the neuropeptide Y (NPY) in the gastro–entero–pancreatic tract of goat kids. NPY is one of the most abundant neuropeptides in mammals, known for its orexigenic role although it is involved in many central and peripheral functions. We carried out immunohistochemical analyses on samples of abomasum, duodenum and pancreas collected from two experimental groups: one fed with standard diet and one with standard diet + RLE. For the first time we document NPY distribution in the abomasum, duodenum and pancreas of goats and observe the highest number of NPY positive cells in neuroendocrine cells of duodenum. Remarkably, upon RLE feed supplementation, NPY immunoreactive cells increased significantly in abomasal epithelium and pancreatic islets but not in duodenum, likely due to pH variation of abomasum and duodenum. Several failures of recurved concrete crownwalls have been observed in recent years. This work aims to get a better insight within the processes underlying the loading phase of these structures due to non-breaking wave impulsive loading conditions and to identify the dominant failure modes. The investigation is carried out through an offline one-way coupling of computational fluid dynamics (CFD) generated wave pressure time series and a time-varying structural Finite Element Analysis. The recent failure of the Civitavecchia (Italy) recurved parapet is adopted as an explanatory case study. Modal analysis aimed to identify the main modal parameters such as natural frequencies, modal masses and modal shapes is firstly performed to comprehensively describe the dynamic response of the investigated structure. Following, the CFD generated pressure field time-series is applied to linear and non-linear finite element model, the developed maximum stresses and the development of cracks are properly captured in both models. Three non-linear analyses are performed in order to investigate the performance of the crownwall concrete class. Starting with higher quality concrete class, it is decreased until the formation of cracks is reached under the action of the same regular wave condition. It is indeed shown that the concrete quality plays a dominant role for the survivability of the structure, even allowing the design of a recurved concrete parapet without reinforcing steel bars. Simple Summary: The obesity induced by overconsumption of nutrients leads to systemic inflammation and alters metabolic homeostasis by acting on central nervous system and peripheral tissues such as intestine. The 3,5-diiodo-L-thyronine (3,5-T2) is well-known for its positive role on fat mass and lipid metabolism, and at date, it is widely used as a drug for the treatment of obesity. However, the safe and effective dose as well as the possible adverse effects of this molecule have not been sufficiently explored. In this study, we analyzed the role of 3,5-T2 in regulating central and peripheral inflammation in diet-induced obese (D.I.O.) model of zebrafish. We found that 3,5-T2 sustained the intestinal alteration caused by D.I.O., as indicated by the high levels of pro-inflammatory cytokines, accompanied by a significant effect of 3,5-T2 on body weight and central inflammation in D.I.O. zebrafish. Therefore, the suggested potential use of 3,5-T2 to contrast obesity should be viewed with caution. We conclude that the zebrafish model can help to better understand the fundamental beneficial and side effects of 3,5-T2, which is of great importance to define the possible use of this metabolite of thyroid hormones as a drug in different diseases including obesity. The 3,5-diiodo-L-thyronine (3,5-T2) is an endogenous metabolite of thyroid hormones, whose administration to rodents fed high-fat diet (HFD) prevents body weight increase and reverts the expression pattern of pro-inflammatory factors associated to HFD. The diet-induced obese (D.I.O.) zebrafish (Danio rerio) has been recently used as an experimental model to investigate fundamental processes underlying central and peripheral obesity-driven inflammation. Herein, we aim to understand the role of 3,5-T2 in regulating central and peripheral inflammation in D.I.O. model of zebrafish. 3,5-T2 (10 nM and 100 nM) was administered with the obesity-inducing diet (D.I.O. with 3,5-T2) or after 4 weeks of obesity-inducing diet (D.I.O. flw 3,5-T2). 3,5-T2 significantly increased the body weight and serum triglyceride levels in D.I.O. zebrafish in both conditions. Moreover, 3,5-T2 sustained or increased inflammation in the anterior (AI) and mid (MI) intestine when administered with the obesity-inducing diet, as indicated by the immunoexpression of the inflammatory markers tumor-necrosis factor-α (TNFα), cyclooxygenase 2 (COX2), calnexin, caspase 3, and proliferating cell nuclear antigen (PCNA). On the contrary, when 3,5-T2 was administered after the obesity-inducing diet, partly reverted the intestinal alteration induced by D.I.O. In addition, brain inflammation, as indicated by the increase in the activation of microglia, was detected in D.I.O. zebrafish and D.I.O. treated with 3,5-T2. These findings reveal that the effects of 3,5-T2 on fish intestine and brain can deviate from those shown in obese mammals, opening new avenues to the investigation of the potential impact of this thyroid metabolite in different diseases including obesity. Nerve growth factor (NGF) receptors are evolutionary conserved molecules, and in mammals are considered necessary for ensuring the survival of cholinergic neurons. The age-dependent regulation of NTRK1/NTRKA and p75/NGFR in mammalian brain results in a reduced response of the cholinergic neurons to neurotrophic factors and is thought to play a role in the pathogenesis of neurodegenerative diseases. Here, we study the age-dependent expression of NGF receptors (NTRK1/NTRKA and p75/NGFR) in the brain of the short-lived teleost fish Nothobranchius furzeri. We observed that NTRK1/NTRKA is more expressed than p75/NGFR in young and old animals, although both receptors do not show a significant age-dependent change. We then study the neuroanatomical organization of the cholinergic system, observing that cholinergic fibers project over the entire neuroaxis while cholinergic neurons appear restricted to few nuclei situated in the equivalent of mammalian subpallium, preoptic area and rostral reticular formation. Finally, our experiments do not confirm that NTRK1/NTRKA and p75/NGFR are expressed in cholinergic neuronal populations in the adult brain of N. furzeri. To our knowledge, this is the first study where NGF receptors have been analyzed in relation to the cholinergic system in a fish species along with their age-dependent modulation. We observed differences between mammals and fish, which make the African turquoise killifish an attractive model to further investigate the fish specific NGF receptors regulation. Nesfatin-1 (Nesf-1) was identified as an anorexigenic and well conserved molecule in rodents and fish. While tissue distribution of NUCB2 (Nucleobindin 2)/Nesf-1 is discretely known in vertebrates, reports on ontogenetic expression are scarce. Here, we examine the age-related central and peripheral expression of NUCB2/Nesf-1 in the teleost African turquoise killifish Nothobranchius furzeri, a consolidated model organism for aging research. We focused our analysis on brain areas responsible for the regulation of food intake and the rostral intestinal bulb, which is analogous of the mammalian stomach. We hypothesize that in our model, the stomach equivalent structure is the main source of NUCB2 mRNA, displaying higher expression levels than those observed in the brain, mainly during aging. Remarkably, its expression significantly increased in the rostral intestinal bulb compared to the brain, which is likely due to the typical anorexia of aging. When analyzing the pattern of expression, we confirmed the distribution in diencephalic areas involved in food intake regulation at all age stages. Interestingly, in the rostral bulb, NUCB2 mRNA was localized in the lining epithelium of young and old animals, while Nesf-1 immunoreactive cells were distributed in the submucosae. Tissue engineering (TE) pursues the ambitious goal to heal damaged tissues. One of the most successful TE approaches relies on the use of scaffolds specifically designed and fabricated to promote tissue growth. During regeneration the guidance of biological events may be essential to sustain vasculature neoformation inside the engineered scaffold. In this context, one of the most effective strategies includes the incorporation of vasculature forming cells, namely endothelial cells (EC), into engineered constructs. However, the most common EC sources currently available, intended as primary cells, are affected by several limitations that make them inappropriate to personalized medicine. Human induced Pluripotent Stem Cells (hiPSC), since the time of their discovery, represent an unprecedented opportunity for regenerative medicine applications. Unfortunately, human induced Pluripotent Stem Cells-Endothelial Cells (hiPSC-ECs) still display significant safety issues. In this work, we reviewed the most effective protocols to induce pluripotency, to generate cells displaying the endothelial phenotype and to perform an efficient and safe cell selection. We also provide noteworthy examples of both in vitro and in vivo applications of hiPSC-ECs in order to highlight their ability to form functional blood vessels. The paper investigates on the hydro-acoustic waves propagation caused by the underwater earthquake, occurred on 6 February 2012, between the Negros and Cebu islands, in the Philippines. Hydro-acoustic waves are pressure waves that propagate at the sound celerity in water. These waves can be triggered by the sudden vertical sea-bed movement, due to underwater earthquakes. The results of three dimensional numerical simulations, which solve the wave equation in a weakly compressible sea water domain are presented. The hydro-acoustic signal is compared to an underwater acoustic signal recorded during the event by a scuba diver, who was about 12 km far from the earthquake epicenter. [ABSTRACT FROM AUTHOR]

This paper illustrates a methodology to get a reliable estimation of the local wave properties, based on the reconstruction of the motion of a moving sailboat by means of GNSS receivers installed on board and an original kinematic positioning approach. The wave parameters reconstruction may be used for many useful practical purposes, e.g., to improve of autopilots, for real-time control systems of ships, to analyze and improve the performance of race sailboats, and to estimate the local properties of the waves. A Class 40 oceanic vessel (ECO40) left from the port of "Riva di Traiano" located close to Rome (Italy) on 19 October 2014 to perform a non-stop sailing alone around the world in energy and food self-sufficiency. The proposed system was installed on ECO40 and the proposed method was applied to estimate the wave properties during a storm in the Western Mediterranean Sea. Neurotrophins contribute to the complexity of vertebrate nervous system, being involved in cognition and memory. Abnormalities associated with neurotrophin synthesis may lead to neuropathies, neurodegenerative disorders and age-associated cognitive decline. The genome of teleost fishes contains homologs of some mammalian neurotrophins as well as a gene coding for an additional neurotrophin (NT-6). In this study, we characterized this specific neurotrophin in the short-lived fish Nothobranchius furzeri, a relatively new model for aging studies. Thus, we report herein for the first time the age-related expression of a neurotrophin in a non-mammalian vertebrate. Interestingly, we found comparable expression levels of NT-6 in the brain of both young and old animals. More in detail, we used a locked nucleic acid probe and a riboprobe to investigate the neuroanatomical distribution of NT-6 mRNA revealing a significant expression of the neurotrophin in neurons of the forebrain (olfactory bulbs, dorsal and ventral telencephalon, and several diencephalic nuclei), midbrain (optic tectum, longitudinal tori, and semicircular tori), and hindbrain (valvula and body of cerebellum, reticular formation and octavolateral area of medulla oblongata). By combining in situ hybridization and immunohistochemistry, we showed that NT-6 mRNA is synthesized in mature neurons. Zebrafish (Danio rerio) is a teleost fish widely accepted as a model organism for neuroscientific studies. The adults show common basic vertebrate brain structures, together with similar key neuroanatomical and neurochemical pathways of relevance to human diseases. However, the brain of adult zebrafish possesses, differently from mammals, intense neurogenic activity, which can be correlated with high regenerative properties. Brain derived neurotrophic factor (BDNF), a member of the neurotrophin family, has multiple roles in the brain, due also to the existence of several biologically active isoforms, that interact with different types of receptors. BDNF is well conserved in the vertebrate evolution, with the primary amino acid sequences of zebrafish and human BDNF being 91% identical. Here, we review the available literature regarding BDNF in the vertebrate brain and the potential involvement of BDNF in telencephalic regeneration after injury, with particular emphasis to the zebrafish. Finally, we highlight the potential of the zebrafish brain as a valuable model to add new insights on future BDNF studies. [ABSTRACT FROM AUTHOR]
