In today's medicine, the celerity of the bio-assays analysis is crucial for the timely selection of the appropriate therapy and hence its effectiveness, especially in case of diseases characterized by the late onset of symptoms. In this paper, a lens-less fiber optics-based fluorescence sensor designed for the measurement of labeled bio-assays is presented and its potential for the early diagnosis of sepsis via C-reactive protein (CRP) detection is demonstrated. The sensor performance results from the combination of two key elements: a planar antenna that redirects fluorescence the marker emission and an automated fiber-based optical system for multi-spot analysis. First, the working principle of the device is demonstrated with a well-established antibody–antigen format (immunoglobulin IgG/anti-IgG assay), reporting more than one order of magnitude enhanced limit of detection (LOD) and limit of quantification (LOQ) for the planar antenna with respect to a standard glass substrate. The prototype is then tested against a sample mimicking a realistic case, prepared with commercially available human serum, showing a LOD and LOQ in the clinical range of interest (0.0015 μg/mL and 0.005 μg/mL, respectively) for the investigation of the sepsis biomarker CRP. These results validate the developed prototype as a simple and easy-to-operate device, compatible with standardized micro-well arrays, and potentially suitable for POC applications. A methodology to enhance the sensitivity of long-period fiber gratings (LPFGs) based on the combination of three different enhancement approaches is presented; the methods here adopted are the working near mode transition (MT) of a cladding mode (CM), working near the turn-around point of a CM and the enhancement of the evanescent field of CMs by reducing the cladding diameter or by increasing the order number of CMs. In order to combine these enhancement methodologies, an electrostatic self-assembly (ESA) process was used to deposit a polymeric overlay, with a chosen thickness, onto the etched fiber. The add-layer sensitivity of the sensor was theoretically calculated, and the demonstration of the real applicability of the developed LPFG as a biosensor was performed by means of an IgG/anti-IgG immunoassay in human serum in a thermostated microfluidic system. The limits of detection (LODs) calculated by following different procedures (three times the standard deviation of the blank and the mean value of the residuals) were 6.9 × 10−8 µg/mL and 4.5 × 10−6 µg/mL, respectively. A new methodology to enhance the sensitivity of a long period fiber grating sensor (LPFG) at the Turn Around Point (TAP) is here presented. The LPFG sensor has been fabricated by etching the fiber up to 20.4 µm, until the sidelobes of dispersed LP0,2 cladding mode appeared near TAP in aqueous medium. In this paper, the electronic transduction of DNA hybridization is presented by coupling organic charge-modulated field-effect transistors (OCMFETs) and hairpin-shaped probes. These probes have shown interesting properties in terms of sensitivity and selectivity in other kinds of assays, in the form of molecular beacons (MBs). Their integration with organic-transistor based sensors, never explored before, paves the way to a new class of low-cost, easy-to-use, and portable genetic sensors with enhanced performances. Thanks to the peculiar characteristics of the employed sensor, measurements can be performed at relatively high ionic strengths, thus optimizing the probes’ functionality without affecting the detection ability of the device. A complete electrical characterization of the sensor is reported, including calibration with different target concentrations in the measurement environment and selectivity evaluation. The design of Whispering Gallery Mode Resonators (WGMRs) used as an optical transducer for biosensing represents the first and crucial step towards the optimization of the final device performance in terms of sensitivity and Limit of Detection (LoD). Here, we propose an analytical method for the design of an optical microbubble resonator (OMBR)-based biosensor. In order to enhance the OMBR sensing performance, we consider a polymeric layer of high refractive index as an inner coating for the OMBR. The effect of this layer and other optical/geometrical parameters on the mode field distribution, sensitivity and LoD of the OMBR is assessed and discussed, both for transverse electric (TE) and transverse magnetic (TM) polarization. This work shows the improvements in the sensing capabilities and precision of an Optical Microbubble Resonator due to the introduction of an encaging poly(methyl methacrylate) (PMMA) box. A frequency fluctuation parameter σ was defined as a score of resonance stability and was evaluated in the presence and absence of the encaging system and in the case of air- or water-filling of the cavity. Furthermore, the noise interference introduced by the peristaltic and the syringe pumping system was studied. An optical sensor platform based on surface plasmon resonance (SPR) in a plastic optical fiber (POF) integrated into a thermo-stabilized flow cell for biochemical sensing applications is proposed. This device has been realized and experimentally tested by using a classic receptor-analyte assay. For this purpose, the gold surface of the POF was chemically modified through the formation of a self-assembling monolayer. The surface robustness of the POF-SPR platform has been tested for the first time thanks to the flow cell. The experimental results show that the proposed device can be successfully used for label-free biochemical sensing. The final goal of this work is to achieve a complete, small-size, simple to use and low cost optical sensor system. The whole system with the flow cell and the optical sensor are extensively described, together with the experimental results obtained with an immunoglobulin G (IgG)/anti-IgG assay. Optical fiber sensors, thanks to their compactness, fast response and real-time measurements, have a large impact in the fields of life science research, drug discovery and medical diagnostics. In recent years, advances in nanotechnology have resulted in the development of nanotools, capable of entering the single cell, resulting in new nanobiosensors useful for the detection of biomolecules inside living cells. In this paper, we provide an application of a nanotip coupled with molecular beacons (MBs) for the detection of DNA. The MBs were characterized by hybridization studies with a complementary target to prove their functionality both free in solution and immobilized onto a solid support. The solid support chosen as substrate for the immobilization of the MBs was a 30 nm tapered tip of an optical fiber, fabricated by chemical etching. With this set-up promising results were obtained and a limit of detection (LOD) of 0.57 nM was reached, opening up the possibility of using the proposed nanotip to detect mRNAs inside the cytoplasm of living cells. Abstract: Whispering gallery mode resonators (WGMR) are an efficient tool for the realization of optical biosensors. A high Q factor preservation is a crucial requirement for good biosensor performances. In this work we present an Eudragit®L100 coated microspherical WGMR as an efficient immunosensor. The developed resonator was morphologically characterized using fluorescence microscopy. The functionalization process was tuned to preserve the high Q factor of the resonator. Chemiluminescence assays have shown great advantages compared with other optical techniques. Gold nanoparticles have drawn much attention in chemiluminescence analysis systems as an enzyme-free catalyst. The catalytic activity of gold nanoparticles for chemiluminescence sensing depends on size, shape and the surface charge property, which is hard to characterize in batches. As there is no positive or negative correlation between chemiluminescence signals and sizes of gold nanoparticles, the best way to get optimal gold nanoparticles is to control the reaction conditions via online chemiluminescence sensing systems. Therefore, a new method was developed for online synthesis of gold nanoparticles with a three-dimension hydrodynamic focusing microreactor, directly coupled with a microfluidic chemiluminescence sensing chip, which was coupled to a charge-coupled device camera for direct catalytical characterization of gold nanoparticles. All operations were performed in an automatic way with a program controlled by Matlab. Gold nanoparticles were synthesized through a single-phase reaction using glucose as a reducing agent and stabilizer at room temperature. The property of gold nanoparticles was easily controlled with the three-dimension microreactor during synthesis. The catalyst property of synthesized gold nanoparticles was characterized in a luminol–NaOCl chemiluminescence system. After optimizing parameters of synthesis, the chemiluminescence signal was enhanced to a factor of 171. The gold nanoparticles synthesized under optimal conditions for the luminol–NaOCl system were stable for at least one month. To further investigate the catalytic activity of synthesized gold nanoparticles in various situations, two methods were used to change the property of gold nanoparticles. After adding a certain amount of salt (NaCl), gold nanoparticles aggregated with a changed surface charge property and the catalytic activity was greatly enhanced. Glutathione was used as an example of molecules with thiol groups which interact with gold nanoparticles and reduce the catalytic activity. The chemiluminescence intensity was reduced by 98.9%. Therefore, we could show that using a microreactor for gold nanoparticles synthesis and direct coupling with microfluidic chemiluminescence sensing offers a promising monitoring method to find the best synthesis condition of gold nanoparticles for catalytic activity. This work is dedicated to developing enzyme biosensor software to solve problems regarding soil pollution analysis. An algorithm and specialised software have been developed which stores, analyses and visualises data using JavaScript programming language. The developed software is based on matching data of 51 non-commercial standard soil samples and their inhibitory effects on three enzyme systems of varying complexity. This approach is able to identify the influence of chemical properties soil samples, without toxic agents, on enzyme biosensors. Such software may find wide use in environmental monitoring. Chemiluminescence (CL) detection is widely employed in biosensors and miniaturized analytical devices since it offers high detectability and flexible device design (there are no geometry requirements for the measurement cell, except the ability to collect the largest fraction of emitted photons). Although the emission anisotropy phenomenon for an emitting dipole bound to the interface between two media with different refractive index is well known for fluorescence, it is still poorly investigated for CL reactions, in which the excited-state reaction products can diffuse in solution before the photon emission event. In this paper, we propose a simple method for the real-time evaluation of the CL emission anisotropy based on a radial array of optical fibers, embedded in a poly(methyl methacrylate) semicylinder and coupled with a Charge-Coupled Device (CCD) camera through a suitable interface. The polar-time evolutions of the CL emission have been studied for catalyzing enzymes immobilized onto a solid surface (heterogeneous configuration) or free in solution (homogeneous configuration). Evidence of the anisotropy phenomenon is observed, indicating that the lifetime of the excited-state products of the enzyme-catalyzed reactions is shorter than the time required for their diffusion in solution at a distance at which the CL can be considered isotropic. Severe infection and sepsis are a common, expensive, and frequently fatal conditions in critically ill patients. The sepsis diagnosis is not trivial, since it is an extremely complex chain of events involving inflammatory and anti-inflammatory processes, cellular reactions, and circulatory disorders. For these reasons, delay in diagnosis and initiation of drug treatments have shown to be crucial for this pathology. Moreover, a multitude of biomarkers has been proposed, many more than for other pathologies. In order to select optimal treatments for the highly heterogeneous group of sepsis patients and to reduce costs, novel multiplexed tools that better characterize the patient and his or her specific immune response are highly desired. In order to achieve the fundament of drastically improved multi-analyte detection and to attain low limits of detection in diagnostics, the area of point-of-care testing (POCT) technology is developing quickly, leading to the production of instruments, the reliability of which is continuously increasing. For this purpose, a selection of two biomarkers—C-reactive protein (CRP) and neopterin (NP)—was studied in this paper and a fluorescence-based integrated optical system, suitable for future POCT applications, was implemented that is capable of performing the simultaneous measurement of the two different biomarkers in replicate. A limit of detection of 10 and 2.1 µg L−1 was achieved for CRP and NP spiked in commercially available human serum, respectively.
