The permeation of cryoprotectants into insect embryos is critical for successful cryopreservation. However, the permeability of silkworm embryos to cryoprotectants and the effects of cryopreservation remain poorly studied. In this study, we evaluated the permeability and toxicity of four cryoprotective agents (CPAs) as well as the vitrification effect on the viability of silkworm embryos. Among the four CPAs, propylene glycol (PG) showed the best permeability. Ethylene glycol (EG) and PG were the least toxic CPAs, but glycerol (GLY) and dimethyl sulfoxide (DMSO) were more toxic. Moreover, we examined several factors including the kind and the concentration of CPAs, exposure time, embryonic stage, and silkworm strains. Embryos at the earlier phases of stage 25 were more tolerant to vitrification using EG. We found that over 21% of embryos treated with EG at the early 2 phase of stage 25: 163 h after egg laying (AEL) developed and progressed to serosa ingestion after vitrification and rewarming. The result was the same in other strains as well. Our results are valuable for the development of new cryopreservation protocols of silkworm embryos.

Flavivirus comprises globally emerging and re-emerging pathogens such as Zika virus (ZIKV), Dengue virus (DENV), and West Nile virus (WNV), among others. Although some vaccines are available, there is an unmet medical need as no effective antiviral treatment has been approved for flaviviral infections. The development of host-directed antivirals (HDAs) targeting host factors that are essential for viral replication cycle offers the opportunity for the development of broad-spectrum antivirals. In the case of flaviviruses, recent studies have revealed that neutral sphingomyelinase 2, (nSMase2), involved in lipid metabolism, plays a key role in WNV and ZIKV infection. As a proof of concept, we have determined the antiviral activity of the non-competitive nSMase2 inhibitor DPTIP against WNV and ZIKV virus. DPTIP showed potent antiviral activity with EC 50 values of 0.26 µM and 1.56 µM for WNV and ZIKV, respectively. In order to unravel the allosteric binding site of DPTIP in nSMase2 and the details of the interaction, computational studies have been carried out. These studies have revealed that DPTIP could block the DK switch in nSMase2. Moreover, the analysis of the residues contributing to the binding identified His463 as a crucial residue. Interestingly, the inhibitory activity of DPTIP on the H463A mutant protein supported our hypothesis. Thus, an allosteric cavity in nSMase2 has been identified that can be exploited for the development of new inhibitors with anti-flaviviral activity., Competing Interests: The authors declare no conflict of interest.

Platelets in atherosclerosis, bypass stenosis, and restenosis have been extensively assessed. However, a sequential ultrastructural study of platelets in angiogenesis during the early phases of these lesions has received less attention. Our objective was the study of platelets in angiogenesis and vessel regression during intimal thickening (IT) formation, a precursor process of these occlusive vascular diseases. For this purpose, we used an experimental model of rat occluded arteries and procedures for ultrastructural observation. The results show (a) the absence of platelet adhesion in the de-endothelialized occluded arterial segment isolated from the circulation, (b) that intraarterial myriad platelets contributed from neovessels originated by sprouting angiogenesis from the periarterial microvasculature, (c) the association of platelets with blood components (fibrin, neutrophils, macrophages, and eosinophils) and non-polarized endothelial cells (ECs) forming aggregates (spheroids) in the arterial lumen, (d) the establishment of peg-and-socket junctions between platelets and polarized Ecs during intussusceptive angiogenesis originated from the EC aggregates, with the initial formation of IT, and (e) the aggregation of platelets in regressing neovessels ('transitory paracrine organoid') and IT increases. In conclusion, in sprouting and intussusceptive angiogenesis and vessel regression during IT formation, we contribute sequential ultrastructural findings on platelet behavior and relationships, which can be the basis for further studies using other procedures.

The dissolution of both copper and arsenic from a copper concentrate was investigated in oxidative ammonia/ammonium solutions at moderate temperatures and atmospheric pressure. The main parameters studied were temperature, pH, concentrations of different ammonia salts, the presence of sodium hypochlorite, pretreatment with sodium chloride, and curing period. In all ammoniacal solutions studied, increasing the temperature enhanced the dissolution of copper, but the dissolution of arsenic remained marginal. Mixing the copper concentrate with sodium chloride and leaving it to rest for 72 h before leaching in ammoniacal solutions significantly increased the dissolution of copper and slightly increased the dissolution of arsenic from the concentrate. A maximum of 35% of Cu and 3.3% of As were extracted when ammonium carbonate was used as the lixiviant. The results show relatively rapid dissolution of the concentrate with the addition of sodium hypochlorite in ammonium carbonate solution, achieving a dissolution of up to 50% and 25% of copper and arsenic, respectively. A copper dissolution with a non-linear regression model was proposed, considering the effect of NaClO and NH 4 Cl at 25 °C. These findings highlight the importance of using the correct anionic ligands for the ammonium ions and temperature to obtain a high dissolution of copper or arsenic. The results also showed that the curing time of the packed bed before the commencement of leaching appeared to be an important parameter to enhance the dissolution of copper and leave the arsenic in the residues.

Mine water is normally considered as waste that has to be managed. However, new applications are increasingly being sought for the water that floods mining voids, especially in relation to its use as an energy resource. The worldwide energy market, within the current transition framework, is searching for creative approaches to produce and store clean energy. In particular, underground pumped hydroelectric energy storage systems (UPHS) constitute efficient and flexible alternatives to deal with intermittent renewable energy sources. In this work, a UPHS is designed using the mine water and the voids of a closed coal mine in Asturias (North-west Spain) as a lower reservoir. Moreover, this system is combined with a wind energy generation facility and the efficiency of the hybrid system is evaluated. With an investment cost of EUR 193 M, a 40 MW UPHES joined to a 60 MW wind farm would generate benefits of about EUR 54 M in 40 years. The reduction in CO 2 emissions (29,000 equivalent tons per year) and the social benefits in a traditional mining area are other intangible advantages of this system.

The emergence of SARS-CoV-2 variants, as observed with the D614G spike protein mutant and, more recently, with B.1.1.7 (501Y.V1), B.1.351 (501Y.V2) and B.1.1.28.1 (P.1) lineages, represent a continuous threat and might lead to strains of higher infectivity and/or virulence. We report on the occurrence of a SARS-CoV-2 haplotype with nine mutations including D614G/T307I double-mutation of the spike. This variant expanded and completely replaced previous lineages within a short period in the subantarctic Magallanes Region, southern Chile. The rapid lineage shift was accompanied by a significant increase of cases, resulting in one of the highest incidence rates worldwide. Comparative coarse-grained molecular dynamic simulations indicated that T307I and D614G belong to a previously unrecognized dynamic domain, interfering with the mobility of the receptor binding domain of the spike. The T307I mutation showed a synergistic effect with the D614G. Continuous surveillance of new mutations and molecular analyses of such variations are important tools to understand the molecular mechanisms defining infectivity and virulence of current and future SARS-CoV-2 strains.

This paper describes a method for calibrating multi camera and multi laser 3D triangulation systems, particularly for those using Scheimpflug adapters. Under this configuration, the focus plane of the camera is located at the laser plane, making it difficult to use traditional calibration methods, such as chessboard pattern-based strategies. Our method uses a conical calibration object whose intersections with the laser planes generate stepped line patterns that can be used to calculate the camera-laser homographies. The calibration object has been designed to calibrate scanners for revolving surfaces, but it can be easily extended to linear setups. The experiments carried out show that the proposed system has a precision of 0.1 mm.
