Simple Summary: Despite remarkable progress in treating early-stage gastric cancer (GC), the clinical outcomes for patients with advanced disease remain very poor. Tissue invasion and metastasis constitute the major causes of cancer-related deaths, including GC. This highlights the urgent need to develop animal models that can recapitulate these processes to develop novel therapeutic strategies. We developed a highly reproducible and cost-effective procedure to establish orthotopic GC xenografts showing high engraftment and metastatic rates via the direct implantation of tumor cell suspensions. Compared with the routine method to establish orthotopic xenograft models by engrafting intact tumor fragments, our approach significantly shortens the experimental timeline and allows for the flexible adjustment of the number of tumor cells implanted to control the rate of tumor progression. Both dose- and time-dependent progressions of tumor invasion and metastasis were nicely recapitulated in our model. Our work provides valuable tools for studying GC progression and metastasis and developing effective therapies. Although the implantation of intact tumor fragments is a common practice to generate orthotopic xenografts to study tumor invasion and metastasis, the direct implantation of tumor cell suspensions is necessary when prior manipulations of tumor cells are required. However, the establishment of orthotopic xenografts using tumor cell suspensions is not mature, and a comparative study directly comparing their engraftment and metastatic capabilities is lacking. It is unclear whether tumor fragments are superior to cell suspensions for successful engraftment and metastasis. In this study, we employed three GC cell lines with varying metastatic capacities to stably express firefly luciferase for monitoring tumor progression in real time. We successfully minimized the risk of cell leakage during the orthotopic injection of tumor cell suspensions without Corning Matrigel by systematically optimizing the surgical procedure, injection volume, and needle size options. Comparable high engraftment and metastatic rates between these two methods were demonstrated using MKN-45 cells with a strong metastatic ability. Importantly, our approach can adjust the rate of tumor progression flexibly and cuts the experimental timeline from 10–12 weeks (for tumor fragments) to 4–5 weeks. Collectively, we provided a highly reproducible procedure with a shortened experimental timeline and low cost for establishing orthotopic GC xenografts via the direct implantation of tumor cell suspensions. The Dongying Depression (Bohai Bay Basin, eastern China) was widely filled with evaporite (anhydrite and halite) layers during the Paleogene period, especially the middle of the fourth member of the Shahejie Formation (Es4). Most evaporite layers are interbedded with mudstone strata. The strata of Es4 are divided into three sections, referred to as the upper layers, evaporite layers, and lower layers, respectively. The analysis of elemental concentrations, elemental ratios, and Pr/Ph suggests that the lower layers were deposited in an intermittent saline lake environment within a relatively dry climate. The evaporite layers were formed in a highly saline lake environment, whereas the upper layers were formed in a brackish-saline to fresh-water environment. Organic matter (OM) abundance indices, including total organic carbon (TOC), chloroform extracts, total hydrocarbon content (HC), hydrocarbon generation potential (S1 + S2), and OM type, show that the source rock potential for petroleum generation in the upper layers is best, that in the evaporite layers is fair, and in the lower layers it is poor. Carbon isotopes (δ13C) of hydrocarbons in the evaporite and lower layers were heavier than those in the upper layers. Thermal maturity parameters show that the thermal evolution process of OM in the upper layers was faster where evaporite were present compared with evaporite-free areas, while the thermal evolution of OM in the lower layers was slower in these regions. The high thermal conductivity of evaporites may have accelerated the thermal evolution of source rocks in upper layers and allowed hydrocarbon generation at a shallower burial depth. This resulted in the earlier appearance of the petroleum generation window compared to in evaporite-free areas. Meanwhile, the thermal evolution of OM in the lower layers was restrained, and consequently the hydrocarbon generation window was widened, which implies the potential for petroleum exploration in deep strata under the evaporite sequence. Background and Objectives: With the aging population, the incidence of degenerative lumbar spinal stenosis (LSS) is increasing. Sarcopenia is an age-related muscular decrease. Although epidural balloon neuroplasty is effective in patients with LSS refractory to conventional treatments, its effect has not been assessed in patients with sarcopenia. Therefore, this study evaluated the effect of epidural balloon neuroplasty in patients with LSS and sarcopenia. Materials and Methods: This retrospective study reviewed the following details from the electronic medical records: patient characteristics, including sex, age, body mass index, diabetes, hypertension, stenosis grading, pain duration, location, pain intensity, and medications. Back and leg pain intensity was evaluated before and after the procedure at one, three, and six months during the follow-up period. A generalized estimating equations model was used at six months follow-up. Patients were divided into sarcopenia and non-sarcopenia groups using the cross-sectional area of the psoas muscle at the level of L3 on magnetic resonance imaging. Results: A total of 477 patients were included (sarcopenia group: 314 patients, 65.8%; non-sarcopenia group: 163 patients, 34.2%). Age, sex, body mass index, and medication quantification scale III were statistically different between both groups. The generalized estimating equations analyses—with unadjusted and adjusted estimation—revealed a significantly reduced pain intensity after the procedure compared to the baseline in both groups. The difference in pain intensity between both groups was not statistically different. Conclusions: Percutaneous epidural balloon neuroplasty may be considered for patients with chronic lumbar LSS regardless of accompanying sarcopenia. [ABSTRACT FROM AUTHOR]

Since 2005, there have been no systematic reviews on the effects of multiple manual therapies, including muscle energy technique (MET), on the hamstrings. Therefore, this systematic review aimed to provide clinical evidence for the effectiveness of the MET on hamstring flexibility. We queried 10 electronic databases (PubMed, EMBASE, The Cochrane Library, KISS, NDSL, KMBASE, KISTI, RISS, Dbpia, and OASIS) up to the end of March 2022. This study only included randomized controlled trials (RCTs) investigating the use of MET for the hamstring. The literature was organized using Endnote. Literature screening and data extraction were conducted by two researchers independently. The methodological quality of the included RCTs was evaluated using the Cochrane risk-of-bias tool 1.0, and the meta-analysis was performed using RevMan 5.4. In total, 949 patients from 19 RCTs were selected according to the inclusion criteria. During active knee extension tests, the efficacy between MET and other manipulations did not significantly differ. For sit and reach tests, MET groups had higher flexibility compared to stretching (MD = 1.69, 95% CI: 0.66 to 2.73, p = 0.001) and no treatment (MD = 2.02, 95% CI: 0.70 to 3.33, p = 0.003) groups. No significant differences were observed in the occurrence of adverse reactions. Overall, we found that MET is more efficacious for improving hamstring flexibility compared to stretching and having no treatment during sit and reach tests because it combines isometric contraction with stretching. The aim of this study was to assess the patterns and trends of pharmacological treatment for outpatients with postherpetic neuralgia (PHN) in China in the period 2015–2019. Prescription data for outpatients with PHN were extracted from the database of the Hospital Prescription Analysis Program of China according to the inclusion criteria. The trends in yearly prescriptions and corresponding costs were analyzed and stratified by drug class and specific drugs. A total of 19,196 prescriptions from 49 hospitals in 6 major regions of China were included for analysis. The yearly prescriptions increased from 2534 in 2015 to 5676 in 2019 (p = 0.027), and the corresponding expenditures increased from CNY 898,618 in 2015 to CNY 2,466,238 in 2019 (p = 0.027). Gabapentin and pregabalin are the most commonly used drugs for PHN, and more than 30% of these two drugs were combined with mecobalamin. Opioids were the second most frequently prescribed drug class, and oxycodone accounted for the largest share of the cost. Topical drugs and TCAs are rarely used. The frequent use of pregabalin and gabapentin was in accordance with current guidelines; however, the use of oxycodone raised concerns about rationality and economic burden. Extensive research has been conducted for decades to elucidate the molecular and regulatory mechanisms for phytochrome-mediated light signaling in plants. As a result, tens of downstream signaling components that physically interact with phytochromes are identified, among which negative transcription factors for photomorphogenesis, PHYTOCHROME-INTERACTING FACTORs (PIFs), are well known to be regulated by phytochromes. In addition, phytochromes are also shown to inactivate an important E3 ligase complex consisting of CONSTITUTIVELY PHOTOMORPHOGENIC 1 (COP1) and SUPPRESSORs OF phyA-105 (SPAs). This inactivation induces the accumulation of positive transcription factors for plant photomorphogenesis, such as ELONGATED HYPOCOTYL 5 (HY5). Although many downstream components of phytochrome signaling have been studied thus far, it is not fully elucidated which intrinsic activity of phytochromes is necessary for the regulation of these components. It should be noted that phytochromes are autophosphorylating protein kinases. Recently, the protein kinase activity of phytochrome A (phyA) has shown to be important for its function in plant light signaling using Avena sativa phyA mutants with reduced or increased kinase activity. Ephedra species were erect, branching shrubs found in desert or arid regions worldwide as the source of ephedrine alkaloids. In this study, the complete chloroplast genome of Ephedra przewalskii and E. monosperma on the Qinghai-Tibet Plateau were sequenced, assembled, and annotated. Compared with the other four published Ephedra species, the chloroplast genomes of Ephedra species were highly conservative, with a quadripartite structure. The length of the chloroplast genome was 109,569 bp in E. przewalskii with 36.6% GC and 109,604 bp in E. monosperma with 36.6% GC. We detected 118 genes in both Ephedra species, including 73 PCGs, 37 tRNA genes, and eight rRNA genes. Among them, the ndh family genes were lost, which could be used to study the phylogeny and genetic diversity of the genus Ephedra, combined with multiple highly variable intergenic spacer (IGS) regions. Codon usage preference of Ephedra species was weak. The ratio of non-synonymous substitutions and synonymous substitutions was low, showing that the PCGs of Ephedra may be under the pressure of purifying selection. ML and BI analysis showed similar phylogenetic topologies. Ephedra species clustered together in a well-supported monophyletic clade. E. przewalskii and E. monosperma were not gathered in one clade, consistent with the classification system by Flora of China. This study reveals differences in the chloroplast genomes of Ephedra, providing valuable and abundant data for the phylogenetic analysis and species identification of Ephedra. [ABSTRACT FROM AUTHOR]

Contemporary urban design, requiring a deep understanding of urban form and its performance, has recently shifted its focus on the vibrancy of waterfronts in coastal cities. Based on analytical methods of quantitative urban morphology, this study aims to explore the common morphological features of waterfronts with high urban vibrancy. We selected vibrant waterfront cases from different countries as the benchmark and collected the multi-sourced urban data. The quantitative analysis extracts the common morphological characteristics of vibrant waterfront by calculating the range of those indicators in different cases. The results indicate that those successful waterfronts comprise compact street networks and are mostly dominated by building types favorable for urban vibrancy. They possess high development intensity and mixed functions. Consequently, the compact urban form and dense-mixed land use are recommended for developing vital waterfronts. Moreover, considering the problematic waterfront area of the Jinshatan area in Yantai, quantitative urban morphology methods can be adopted to develop precise urban design guidance for vibrancy-oriented design practice. Spent mushroom substrates (SMSs) can be developed as a biofertilizer through composting. Here, we investigated the dynamics of bacterial and fungal communities during commercial composting and the effect of swine and poultry manure on their communities through MiSeq pyrosequencing. Weissella paramesenteroides and Lactobacillus helveticus were dominant bacterial species in the composts with soy waste (SMS-SW), whereas Thermotogaceae sp. and Ureibacillus sp. were dominant in the composts with swine and poultry manure (SMS-PM). For the fungal community, Flammulina velutipes was dominant in SMS-SW, whereas Trichosporon asahii, Candida catenulate, Aspergillus fumigatus, and Candida tropicalis were dominant in SMS-PM. The addition of manure affected the bacterial community significantly. Redundancy analysis indicated that bacterial communities were affected by temperature, potassium, and potassium oxide and fungal communities by temperature, Kjeldahl nitrogen, organic matter, and ammonium nitrogen. Ultra-high-strength quenching and partitioning (Q&P) steels have achieved remarkable lightweight effect in automotive manufacture due to the excellent mechanical performances. However, the problem of sheared-edge cracking greatly limits their application. In this work, the damage generated in the shearing process of QP980 steel is experimentally investigated via microstructure characterization and micro-/macromechanical property evaluation. Moreover, the shearing deformation is simulated with six widely used damage models. The experimental results reveal that microvoids, microcracks, and work-hardening behavior are the main damage factors affecting the formability of sheared edges. Microvoids mainly formed at phase interfaces have a small size (≤5 μm), while microvoids generated from inclusions with a small number have a large size (>5 μm). As deformation continuously grows, microvoids distributed around the sheared surface are split into microcracks, which act as crack initiators in the subsequent forming step. Additionally, the highest microhardness in the fracture zone further enhances the susceptibility of edge cracking. Furthermore, the optimum damage model for QP980 steel was determined by developing user-defined subroutine VUSDFLD in Abaqus, which can be used in the prediction of fracture behavior of QP980 steel to reduce the risk of edge cracking. In this work, we proposed an efficient and straightforward up-conversion process to enhance the external quantum efficiency in a red-emission organic light-emitting diode (OLED). The carbon quantum dots in the luminescent materials initiated the up-conversion by doping the (6,6)-phenyl-C61-butyric acid methyl ester (PCBM) in an amount of 0.001 wt. %, and the external quantum efficiency (EQE) increased from approximately 80% to 9.27% without spectrum change. The time-resolved photoluminescence was applied to understand the mechanism of EQE enhancement in the PCBM-doped OLED. Two decay-time constants fit the TRPL. After PCBM doping, the extended PL intensity indicated increased time constants. The time constants increased from 1.06 and 4.02 ns of the reference sample to 3.48 and 11.29 ns of the PCBM-doped material, respectively. The nonradiative energy transfer (NRET) mechanism was proposed responsible for the decay-time enhancement. The excitons in the PCBM, either by excitation or injection, will transfer to the phosphorescent material nonradiatively. As the PCBM has lower energy levels than the luminous material for electrons, the backward exciton transfer is a kind of up-conversion. With the increased amounts of excitons in the luminescent material, the luminescent external quantum efficiency and the decay-time increased. Stylosanthes guianensis is an excellent forage legume in subtropical and tropical regions with drought tolerance, but little is known about its drought tolerance mechanism. Dehydration responsive element binding proteins (DREBs) are responsive to abiotic stresses. A SgDREB2C was cloned from S. guianensis, while SgDREB2C protein was localized at nucleus. SgDREB2C transcript was induced by dehydration treatment. Transgenic Arabidopsis overexpressing SgDREB2C showed enhanced osmotic and drought tolerance with higher levels of relative germination rate, seedlings survival rate and Fv/Fm and lower levels of ion leakage compared with WT after osmotic and drought stress treatments. In addition, higher levels of superoxide dismutase (SOD) and ascorbate peroxidase (APX) activities and stress responsive gene (COR15A, COR47) transcripts were observed in transgenic Arabidopsis than in WT under drought stress. These results suggest that SgDREB2C regulated drought tolerance, which was associated with increased SOD and APX activities and stress-responsive gene expression under drought stress. Reduced-representation sequencing (RRS) has made it possible to identify hundreds to thousands of genetic markers for phylogenomic analysis for the testing of phylogenetic hypotheses in non-model taxa. The use of customized probes to capture genetic markers (i.e., ultraconserved element (UCE) approach) has further boosted the efficiency of collecting genetic markers. Three UCE probe sets pertaining to spiders (Araneae) have been published, including one for the suborder Mesothelae (an early diverged spider group), one for Araneae, and one for Arachnida. In the current study, we developed a probe set specifically for the superfamily Araneoidea in spiders. We then combined the three probe sets for Araneoidea, Araneae, and Arachnid into a fourth probe set. In testing the effectiveness of the 4 probe sets, we used the captured loci of the 15 spider genomes in silico (6 from Araneoidea). The combined probe set outperformed all other probe sets in terms of the number of captured loci. The Araneoidea probe set outperformed the Araneae and Arachnid probe sets in most of the included Araneoidea species. The reconstruction of phylogenomic trees using the loci captured from the four probe sets and the data matrices generated from 50% and 75% occupancies indicated that the node linked to the Stegodyphus + RTA (retrolateral tibial apophysis) clade has unstable nodal supports in the bootstrap values, gCFs, and sCFs. Our results strongly indicate that developing ad hoc probe sets for sub-lineages is important in the cases where the origins of a lineage are ancient (e.g., spiders ~380 MYA). [ABSTRACT FROM AUTHOR]

In this study, a multiplex detection system was proposed by integrating a localized surface plasmon resonance (LSPR) sensing array and parallel microfluidic channels. The LSPR sensing array was fabricated by nanoimprinting and gold sputter on a polycarbonate (PC) substrate. The polydimethylsiloxane (PDMS) microfluidic channels and PC LSPR sensing array were bound together through (3-aminopropyl)triethoxysilane (APTES) surface treatment and oxygen plasma treatment. The resonant spectrum of the LSPR sensing device was obtained by broadband white-light illumination and polarized wavelength measurements with a spectrometer. The sensitivity of the LSPR sensing device was measured using various ratios of glycerol to water solutions with different refractive indices. Multiplex detection was demonstrated using human immunoglobulin G (IgG), IgA, and IgM. The anti-IgG, anti-IgA, and anti-IgM were separately modified in each sensing region. Pulmonary arterial hypertension (PAH) is a debilitating condition of the pulmonary circulatory system that occurs in patients of all ages and if untreated, eventually leads to right heart failure and death. Despite existing medical treatment options that improve survival and quality of life, the disease remains incurable. Thus, there is an urgent need to develop novel therapies to treat this disease. Emerging evidence suggests that long non-coding RNAs (lncRNAs) play critical roles in pulmonary vascular remodeling and PAH. LncRNAs are implicated in pulmonary arterial endothelial dysfunction by modulating endothelial cell proliferation, angiogenesis, endothelial mesenchymal transition, and metabolism. LncRNAs are also involved in inducing different pulmonary arterial vascular smooth muscle cell phenotypes, such as cell proliferation, apoptosis, migration, regulation of the phenotypic switching, and cell cycle. LncRNAs are essential regulators of gene expression that affect various diseases at the chromatin, transcriptional, post-translational, and even post-translational levels. Here, we focus on the role of LncRNAs and their molecular mechanisms in the pathogenesis of PAH. This study discusses convolutional neural networks (CNNs) for vibration signals analysis, including applications in machining surface roughness estimation, bearing faults diagnosis, and tool wear detection. The one-dimensional CNNs (1DCNN) and two-dimensional CNNs (2DCNN) are applied for regression and classification applications using different types of inputs, e.g., raw signals, and time-frequency spectra images by short time Fourier transform. In the application of regression and the estimation of machining surface roughness, the 1DCNN is utilized and the corresponding CNN structure (hyper parameters) optimization is proposed by using uniform experimental design (UED), neural network, multiple regression, and particle swarm optimization. It demonstrates the effectiveness of the proposed approach to obtain a structure with better performance. In applications of classification, bearing faults and tool wear classification are carried out by vibration signals analysis and CNN. In this study, a portable and large-area blackbody system was developed following a series of processes including design, computational analysis, fabrication, and experimental analysis and evaluation. The blackbody system was designed to be lightweight (5 kg), and its temperature could exceed the ambient temperature by up to 15 °C under operation. A carbon-fiber-based heat source was used to achieve a uniform temperature distribution. A heat shield fabricated from an insulation material was embedded at the opposite side of the heating element to minimize heat loss. A prototype of the blackbody system was fabricated based on the design and transient coupled electro-thermal simulation results. The operation performance of this system, such as the thermal response, signal transfer function, and noise equivalent temperature difference, was evaluated by employing an infrared imaging system. In addition, emissivity was measured during operation. The results of this study show that the developed portable and large-area blackbody system can be expected to serve as a reliable reference source for the calibration of aerial infrared images for the application of aerial infrared techniques to remote sensing. Photomorphogenesis and skotomorphogenesis are two key events that control plant development, from seed germination to flowering and senescence. A group of wavelength-specific photoreceptors, E3 ubiquitin ligases, and various transcription factors work together to regulate these two critical processes. Phytochromes are the main photoreceptors in plants for perceiving red/far-red light and transducing the light signals to downstream factors that regulate the gene expression network for photomorphogenic development. In this review, we highlight key developmental stages in the life cycle of plants and how phytochromes and other components in the phytochrome signaling pathway play roles in plant growth and development. The morphology and distribution of the antennal sensilla of Megalurothrips usitatus Bagnall and Thrips palmi Karny were examined using scanning electron microscopy (SEM). These are serious pests of various economically important crops, and their antennae are important in chemical communication. The antennae of both species consist of a scape, pedicel, and flagellum, but the flagellum of M. usitatus is made up of six sub-segments, whereas that of T. palmi consists of five sub-segments. Seven morphological sensilla types, including Böhm bristle (BB), sensilla campaniformia (Sca), three types of sensilla basiconica (Sb1, Sb2 and Sb3), two types of sensilla chaetica (Sch1 and Sch2), sensilla styloconica (Sst), sensilla trichodea (St), and sensilla cavity (Scav), were recorded in both species. The scape and pedicels exhibited Sch1, BB and Sca. The flagellum exhibited two types of Sch, three types of Sb, St, Sst and Scav. Extensive research over several decades in plant light signaling mediated by photoreceptors has identified the molecular mechanisms for how phytochromes regulate photomorphogenic development, which includes degradation of phytochrome-interacting factors (PIFs) and inactivation of COP1-SPA complexes with the accumulation of master transcription factors for photomorphogenesis, such as HY5. However, the initial biochemical mechanism for the function of phytochromes has not been fully elucidated. Plant phytochromes have long been known as phosphoproteins, and a few protein phosphatases that directly interact with and dephosphorylate phytochromes have been identified. However, there is no report thus far of a protein kinase that acts on phytochromes. On the other hand, plant phytochromes have been suggested as autophosphorylating serine/threonine protein kinases, proposing that the kinase activity might be important for their functions. Indeed, the autophosphorylation of phytochromes has been reported to play an important role in the regulation of plant light signaling. More recently, evidence that phytochromes function as protein kinases in plant light signaling has been provided using phytochrome mutants displaying reduced kinase activities. Mycoplasma gallisepticum (MG), a pathogen that infects chickens and some other birds, triggers chronic respiratory disease (CRD) in chickens, which is characterized by inflammation. The investigation of microbial pathogenesis would contribute to the deep understanding of infection control. Since microribonucleic acids (miRNAs) play a key role in this process, gga-mir-146c, an upregulated miRNA upon MG infection, was selected according to our previous RNA-sequencing data. In this paper, we predicted and validated that MMP16 is one of gga-miR-146c target genes. Results show that MMP16 is the target of gga-miR-146c and gga-miR-146c can downregulate MMP16 expression within limits. gga-miR-146c upregulation significantly increased the expression of TLR6, NF-κB p65, MyD88, and TNF-α, whereas the gga-miR-146c inhibitor led to an opposite result. gga-miR-146c upregulation effectively decreased apoptosis and stimulated DF-1 cells proliferation upon MG infection. On the contrary, gga-miR-146c inhibitor promoted apoptosis and repressed the proliferation. Collectively, our results suggest that gga-miR-146c upregulation upon MG infection represses MMP16 expression, activating TLR6/MyD88/NF-κB pathway, promoting cell proliferation by inhibiting cell apoptosis, and, finally, enhancing cell cycle progression to defend against host MG infection. Mycoplasma gallisepticum (MG) mainly infects chickens to initiate chronic respiratory disease (CRD). microRNAs (miRNAs) play vital roles according to previously reported studies. Our previous study showed that gga-miR-16-5p, in MG-infected lungs of chicken embryo, was upregulated by Illumina sequencing. The study aimed to reveal what role gga-miR-16-5p plays in CRD progression. gga-miR-16-5p was upregulated in MG-infected fibroblast cells (DF-1). Phosphoinositide-3-kinase regulatory subunit 1 (PIK3R1) was demonstrated as the target gene of gga-miR-16-5p. Furthermore, PIK3R1 expression was lower in MG-infected groups than it in noninfected controls measured by qPCR. Additionally, overexpressed gga-miR-16-5p could downregulate PIK3R1 and phosphorylated serine/threonine kinase (p-Akt) to express protein, whereas there is an opposite effect on inhibition. Overexpressed gga-miR-16-5p resulted in decreased activity of tumor necrosis factor alpha (TNF-α) and the nuclear factor-kappaB (NF-κB) by qPCR. Furthermore, overexpressed gga-miR-16-5p restricted cell multiplication, cycle progression, and increased apoptosis of MG-infected DF-1 cells, whereas inhibited gga-miR-16-5p led to the opposite effect. Collectively, upregulated gga-miR-16-5p could decrease multiplication, cycle progression, and increase apoptosis of MG-infected DF-1 cells, at least partly through directly targeting PIK3R1 and inhibiting PI3K/Akt/NF-κB pathway to exert an anti-inflammatory effect. The effects of structural design parameters on the performance of nano-replicated photonic crystal (PC) label-free biosensors were examined by the analysis of simulated reflection spectra of PC structures. The grating pitch, duty, scaled grating height and scaled TiO2 layer thickness were selected as the design factors to optimize the PC structure. The peak wavelength value (PWV), full width at half maximum of the peak, figure of merit for the bulk and surface sensitivities, and surface/bulk sensitivity ratio were also selected as the responses to optimize the PC label-free biosensor performance. A parametric study showed that the grating pitch was the dominant factor for PWV, and that it had low interaction effects with other scaled design factors. Therefore, we can isolate the effect of grating pitch using scaled design factors. For the design of PC-label free biosensor, one should consider that: (1) the PWV can be measured by the reflection peak measurement instruments, (2) the grating pitch and duty can be manufactured using conventional lithography systems, and (3) the optimum design is less sensitive to the grating height and TiO2 layer thickness variations in the fabrication process. In this paper, we suggested a design guide for highly sensitive PC biosensor in which one select the grating pitch and duty based on the limitations of the lithography and measurement system, and conduct a multi objective optimization of the grating height and TiO2 layer thickness for maximizing performance and minimizing the influence of parameter variation. Through multi-objective optimization of a PC structure with a fixed grating height of 550 nm and a duty of 50%, we obtained a surface FOM of 66.18 RIU-1 and an S/B ratio of 34.8%, with a grating height of 117 nm and TiO2 height of 210 nm.
