Drug use is increasing in South Korea, necessitating evidence-based policy interventions. However, there is a dearth of reliable tools for identifying the personal and psychological factors that drive drug use among Korean adults. In this study, we developed and validated an instrument based on a motivational model to measure drug use motivation in the Korean context. A survey was administered online to 250 Korean adults aged 19–59. Of the initial 37 potential items evaluated, 23 were retained after exploratory factor analysis. Based on their relevance and factor loadings, the final scale consisted of 15 items distributed across five distinct factors: enhancement, coping, social, positive expectancy, and negative expectancy. Confirmatory factor analysis validated the strong fit of the five-factor model. Criterion-related validity was demonstrated by the significant correlations between the five factors and the selected criterion variables. This instrument can be used in research initiatives related to drug addiction and can provide basic data for policy interventions intended to curb drug addiction problems in South Korea. Simple Summary: YAP-TEAD protein-protein interaction is a crucial feature in the Hippo pathway for cancer therapeutics. In this study, to investigate novel YAP-TEAD protein-protein interaction inhibitors, the fragment molecular orbital method for hot spot analysis of YAP-TEAD PPI and FMO-based pharmacophore construction were introduced. We performed pharmacophore based virtual screening through an in-house library. Virtual hit compounds were derived, and the biological activity of the hit compounds was confirmed, through in vitro experiments. Finally, we proposed a compound BY03 as a novel promising PPI inhibitor for the YAP-TEAD interaction. The Hippo pathway is an important signaling pathway modulating growth control and cancer cell proliferation. Dysregulation of the Hippo pathway is a common feature of several types of cancer cells. The modulation of the interaction between yes-associated protein (YAP) and transcriptional enhancer associated domain (TEAD) in the Hippo pathway is considered an attractive target for cancer therapeutic development, although the inhibition of PPI is a challenging task. In order to investigate the hot spots of the YAP and TEAD1 interacting complex, an ab initio Fragment Molecular Orbital (FMO) method was introduced. With the hot spots, pharmacophores for the inhibitor design were constructed, then virtual screening was performed to an in-house library. Next, we performed molecular docking simulations and FMO calculations for screening results to study the binding modes and affinities between PPI inhibitors and TEAD1. As a result of the virtual screening, three compounds were selected as virtual hit compounds. In order to confirm their biological activities, cellular (luciferase activity, proximity ligation assay and wound healing assay in A375 cells, qRT-PCR in HEK 293T cells) and biophysical assays (surface plasmon resonance assays) were performed. Based on the findings of the study, we propose a novel PPI inhibitor BY03 and demonstrate a profitable strategy to analyze YAP–TEAD PPI and discover novel PPI inhibitors. [ABSTRACT FROM AUTHOR]

This study investigated income-related health inequality at sub-national level, focusing on mortality inequality among middle-aged and older adults (MOAs). Specifically, we examined income-related mortality inequality and its social factors among MOAs across 25 districts in Seoul using administrative big data from the National Health Insurance Service (NHIS). We obtained access to the NHIS's full-population micro-data on both incomes and demographic variables for the entire residents of Seoul. Slope Index of Inequality (SII) and Relative Index of Inequality (RII) were calculated. The effects of social attributes of districts on SIIs and RIIs were examined through ordinary least squares and spatial regressions. There were clear income-related mortality gradients. Cross-district variance of mortality rates was greater among the lowest income group. SIIs were smaller in wealthier districts. Weak spatial correlation was found in SIIs among men. Lower RIIs were linked to lower Gini coefficients of income for both genders. SIIs (men) were associated with higher proportions of special occupational pensioners and working population. Lower SIIs and RIIs (women) were associated with higher proportions of female household heads. The results suggest that increasing economic activities, targeting households with female heads, reforming public pensions, and reducing income inequality among MOAs can be good policy directions.
