Antivenom immunotherapy is the mainstay of treatment for snakebite envenoming. Most parts of the world affected by snakebite envenoming depend on broad-spectrum polyspecific antivenoms that are known to contain a low content of case-specific efficacious immunoglobulins. Thus, advances in toxin-specific antibodies production hold much promise in future therapeutic strategies of snakebite envenoming. We report anti-3FTxs monoclonal antibodies developed against N. ashei venom in mice. All the three test mAbs (P4G6a, P6D9a, and P6D9b) were found to be IgG antibodies, isotyped as IgG1. SDS-PAGE analysis of the test mAbs showed two major bands at approximately 55 and 29 kDa, suggestive of immunoglobulin heavy and light chain composition, respectively. The immunoaffinity-purified test mAbs demonstrated higher binding efficacy to the target antigen compared to negative control. Similarly, a cocktail of the test mAbs was found to induce a significantly higher inhibition (p-value < 0.0001) compared to two leading commercial brands of antivenoms on the Kenyan market, implying a higher specificity for the target antigen. Both the test mAbs and 3FTxs polyclonal antibodies induced comparable inhibition (p-value = 0.9029). The inhibition induced by the 3FTxs polyclonal antibodies was significantly different from the two antivenoms (p-value < 0.0001). Our results demonstrate the prospects of developing toxin-specific monoclonal-based antivenoms for snakebite immunotherapy. [ABSTRACT FROM AUTHOR]

The administration of toxin-specific therapy in snake envenoming is predicated on improved diagnostic techniques capable of detecting specific venom toxins. Various serological tests have been used in detecting snakebite envenoming. Comparatively, enzyme-linked immunosorbent assay (ELISA) has been shown to offer a wider practical application. We report an inhibition ELISA for detecting three-finger toxin (3FTx) proteins in venoms of African spitting cobras. The optimized assay detected 3FTxs in N. ashei (including other Naja sp.) venoms, spiked samples, and venom-challenged mice samples. In venoms of Naja sp., the assay showed inhibition, implying the detection of 3FTxs, but showed little or no inhibition in non-Naja sp. In mice-spiked samples, one-way ANOVA results showed that the observed inhibition was not statistically significant between spiked samples and negative control (p-value = 0.164). Similarly, the observed differences in inhibition between venom-challenged and negative control samples were not statistically significant (p-value = 0.9109). At an LOD of 0.01 µg/mL, the assay was able to confirm the presence of 3FTxs in the samples. Our results show a proof of concept for the use of an inhibition ELISA model as a tool for detecting 3FTxs in the venoms of African spitting cobra snakes. [ABSTRACT FROM AUTHOR]

In contrast to comprehensively investigated antibacterial activity of snake venoms, namely crude venoms and their selected components, little is known about antifungal properties of elapid snake venoms. In the present study, the proteome of two venoms of red spitting cobra Naja pallida (NPV) and Mozambique spitting cobra Naja mossambica (NMV) was characterized using LC-MS/MS approach, and the antifungal activity of crude venoms against three Candida species was established. A complex response to venom treatment was revealed. NPV and NMV, when used at relatively high concentrations, decreased cell viability of C. albicans and C. tropicalis, affected cell cycle of C. albicans, inhibited C. tropicalis-based biofilm formation and promoted oxidative stress in C. albicans, C. glabrata and C. tropicalis cells. NPV and NMV also modulated ammonia pulses during colony development and aging in three Candida species. All these observations provide evidence that NPV and NMV may diminish selected pathogenic features of Candida species. However, NPV and NMV also promoted the secretion of extracellular phospholipases that may facilitate Candida pathogenicity and limit their usefulness as anti-candidal agents. In conclusion, antifungal activity of snake venoms should be studied with great caution and a plethora of pathogenic biomarkers should be considered in the future experiments. [ABSTRACT FROM AUTHOR]

Simple Summary: Snake venom is mostly composed of proteins and peptides, which are of interest to many researchers due to their potential pharmacological properties. Due to their biochemical character, these components are analyzed using proteomic techniques such as electrophoresis, chromatography and mass spectrometry. A very important stage of such studies is the measurement of protein concentration in the sample, which is most often performed by colorimetric methods. In the presented article, we used five such techniques on venoms of two snake species, namely Agkistrodon contortrix and Naja ashei. In the case of A. contortrix venom, four methods provide similar concentration values, whereas, in the case of N. ashei, the differences between results are very significant. The source of these differences should probably be seen in the differences in amino acid composition of proteins of these two venoms. With this report, we would like to draw attention to the need to select an appropriate method for measuring the concentration of protein in the venom, especially in the case of Elapid species. Snake venom is an extremely interesting natural mixture of proteins and peptides, characterized by both high diversity and high pharmacological potential. Much attention has been paid to the study of venom composition of different species and also detailed analysis of the properties of individual components. Since proteins and peptides are the active ingredients in venom, rapidly developing proteomic techniques are used to analyze them. During such analyses, one of the routine operations is to measure the protein concentration in the sample. The aim of this study was to compare five methods used to measure protein content in venoms of two snake species: the Viperids representative, Agkistrodon contortrix, and the Elapids representative, Naja ashei. The study showed that for A. contortrix venom, the concentration of venom protein measured by four methods is very similar and only the NanoDrop method clearly stands out from the rest. However, in the case of N. ashei venom, each technique yields significantly different results. One of the key problems of modern infectious disease medicine is the growing number of drug-resistant and multi-drug-resistant bacterial strains. For this reason, many studies are devoted to the search for highly active antimicrobial substances that could be used in therapy against bacterial infections. As it turns out, snake venoms are a rich source of proteins that exert a strong antibacterial effect, and therefore they have become an interesting research material. We analyzed Naja ashei venom for such antibacterial properties, and we found that a specific composition of proteins can act to eliminate individual bacterial cells, as well as the entire biofilm of Staphylococcus epidermidis. In general, we used ion exchange chromatography (IEX) to obtain 10 protein fractions with different levels of complexity, which were then tested against certified and clinical strains of S. epidermidis. One of the fractions (F2) showed exceptional antimicrobial effects both alone and in combination with antibiotics. The protein composition of the obtained fractions was determined using mass spectrometry techniques, indicating a high proportion of phospholipases A2, three-finger toxins, and L-amino acids oxidases in F2 fraction, which are most likely responsible for the unique properties of this fraction. Moreover, we were able to identify a new group of low abundant proteins containing the Ig-like domain that have not been previously described in snake venoms. [ABSTRACT FROM AUTHOR]

This article presents a new approach to collision avoidance in drone swarms, designed for operations in large drone swarms and dynamic environments. The mechanism uses distributed communication, where drones share information about their positions and planned trajectories to predict and avoid collisions. The proposed mechanism enables drones to autonomously cooperate and maintain safe distances in complex scenarios. It is based on the concept of repulsion vectors. The avoidance response is determined by the level of immersion in the protective sphere of obstacles, including other drones. The advantage of the algorithm lies in its simplicity and low computational complexity, allowing it to be used even in small and inexpensive drones. The algorithm was tested in a developed simulation environment, created to handle swarms of over 20 drones and to demonstrate the scalability of the proposed solution. Two scenarios were analyzed: (i) two swarms, each with nine drones, flying on a collision course; (ii) a swarm of 25 drones changing formation. The results showed that the mechanism is effective in avoiding collisions, maintaining safe distances and adapting to changing conditions. The proposed mechanism represents a significant advancement in swarm coordination, offering a robust and scalable solution for real-world applications.

Inconel 718 (In718) is the most widely used nickel-based alloy in additive manufacturing due to its favorable processability. However, In718's high-temperature performance is not suited for the most demanding applications in the aerospace industry. Therefore, in this study, Inconel 718 powder was coated with 3% wt. rhenium (In718-Re) using AM's in situ alloying capabilities to improve high-temperature properties. The proposed alloy's mechanical performance was evaluated, focusing on the effects of post-process heat treatment and hot isostatic pressing following the laser-based powder bed fusion of metals (PBF-LB/M) processing. Static tensile tests conducted at room temperature and elevated temperatures (650 °C and 760 °C) demonstrated that the alloy has comparable strength to pure In718 according to ASTM F3055-14a-an ultimate tensile strength of 1247 MPa, yield strength of 909 MPa, and almost 2× higher elongation of 23.8%. Fatigue tests at room temperature indicated a fatigue limit below 400 MPa for 10 7 cycles. Fractographic analysis revealed that fatigue performance was primarily impacted by a lack of fusion defects inherent to the PBF-LB/M process, highlighting the need for optimized powder preparation and processing parameters to minimize defect formation. While rhenium addition shows limited benefits in Inconel 718, this study underscores the potential of in situ alloying through powder surface modification as a flexible method for incorporating high-melting-point elements into nickel-based alloys for tailored alloy design in additive manufacturing.

Drug development for human disease relies on preclinical model systems such as human cell cultures and animal experiments before therapeutic treatments can ultimately be tested on humans in clinical studies. We here describe the generation of a novel human cell line (HLMVEC/SVTERT289) that we generated by transfection of microvascular endothelial cells from healthy donor lung tissue with the catalytic domain of telomerase and the SV40 large T/small t-antigen. These cells exhibited satisfactory growth characteristics and largely maintained their native characteristics, including morphology, cell surface marker expression, angiogenic potential and the protein composition of secreted extracellular vesicles. In order to test their suitability as a disease model, we simulated mechanical stress induced by cyclic stretch as encountered in ventilator-induced lung injury using the FlexCell ® system and compared their performance to primary lung endothelial cells. In this setting, HLMVEC/SVTERT289 cells exhibited significantly higher neprilysin activity on the cell surface and extracellular vesicles secreted from the cell line exhibited higher Tissue Factor and ACE2 expression but lower ACE expression and ACE activity than vesicles released from the primary cells. This study provides an unprecedented and detailed characterization of the HLMVEC/SVTERT289 cell line, which should help to appraise its suitability in different molecular studies.

Introduction: In this single-center retrospective analysis, we present case data and insights gathered over the past eight years. Additionally, we computed postnatal, pre-therapy lesion-to-lung ratios of Congenital Pulmonary Airway Malformations (CPAMs) to retrospectively assess potential outcome prediction using lesion-to-lung ratios., Methods: Data were collected between 2015 and 2022. Information such as chosen therapy, surgical duration, postoperative hospital stay, and follow-up was obtained from electronic case records. Pre-therapy pulmonary lesion volumes [mm 3 ], lesion-to-ipsilateral-lung ratio, and lesion-to-both-lungs ratio of CPAMs were retrospectively calculated from computed tomography images using specialized software., Results: Of the 40 identified cases, 27 had CPAM, 7 had pulmonary sequestration, 4 had bronchogenic cysts, and 2 had congenital lobar emphysema. Histological examinations of resected specimens revealed no malignancy. For CPAMs, the median surgery age was 7 months (interquartile range (IQR): 0.45-11), averaging 9.54 ± 15.01 months. The CPAM surgery lasted on average 126 ± 53 min (median 124 min (IQR 108-172)). The mean length of hospital stay was 6 ± 1.41 days for thoracoscopic surgery and 17 ± 18.23 days for open surgery. No clear link was found between the lesion ratio and management choice or surgical length. Notably, larger lesions tended to result in longer postoperative stays. CPAMs with a lesion-to-ipsilateral-lung ratio of ≤0.11 were asymptomatic., Discussion and Conclusions: If patients present no symptoms, mild symptoms, or smaller CPAM lesions, "wait and watch" and a CT scan of the thorax up to approximately six months of age remain a reasonable approach. The true risk of malignancy remains ambiguous, especially since there was no evidence of malignancy in our biopsies. On the other hand, prophylactic surgery before symptoms arose led to earlier discharge and overall low intraoperative complications compared to symptomatic counterparts. Ultimately, the adopted therapy pathway remains a parental choice. For CPAMs, an increased lesion-to-lung ratio correlated with extended hospital stay and symptomatic presentation. However, there was no cut-off value for conservative or surgical treatment.

Packaging materials mainly serve the function of protecting products. The most common representative of this group is poly(ethylene terephthalate) (PET), which is not biodegradable and therefore, its waste might be burdensome to the environment. Thus, this work aims to develop outlines for obtaining polyester-based systems, preferably biobased ones, intended for the packaging industry and their detailed characterization. The obtained multilayer systems based on two biobased thermoplastic polyesters, i.e., poly(ethylene 2,5-furandicarboxylate) (PEF) and the "double green" polylactide (PLA), were subjected to various analyses, i.e., UV-Vis spectrophotometry, microscopic evaluation, tensile tests, differential scanning calorimetry (DSC), oxygen transmission rate (OTR), water absorption tests, surface analyses, and biofilm formation. The best possible arrangement was selected in terms of the packaging industry. It was proven that the elastic modulus was significantly higher for multilayer systems, whilst higher-strength parameters were observed for PLA single foils. Regardless of thickness, PLA and PEF foils exhibit similar absorption values in cold water. Moreover, PEF foils demonstrated significantly better barrier properties towards oxygen gas compared to PLA foils of the same thickness. However, a multilayer system composed of two PLA foils with a single inner PEF foil had an OTR value only slightly higher than that of the PEF foil alone. PEF was also found to be a material that exhibited a limited formation of bacterial biofilm, particularly strains of S. aureus and E. coli . All of the above findings clearly confirm the sensibility of the research topic undertaken in this work on the application of biobased thermoplastic polyesters in the packaging industry.

The development of new graphene-based materials necessitates the application of suitable material imaging techniques, especially for the identification of defects in the graphene structure and its continuity. For this purpose, it is natural to use one of the main properties of graphene-electrical conductivity. In this work, we prepare a 9 cm 2 large-area monolayer graphene membrane on porous scaffolding sealed with either GO or rGO. Then, we use electrostatic force microscopy (EFM) AFM mode along with SE and AEE SEM modes to characterize the as-prepared graphene membranes thoroughly. The combination of SEM-AEE and AFM-EFM techniques not only assesses the quality of graphene itself but also characterizes the selectivity and effectiveness of masking graphene layer defects by applying GO or rGO. This makes these methods valuable in optimizing the production of advanced graphene nanocomposites such as semipermeable membranes.

Background and Objectives : Toxic epidermal necrolysis (TEN) and Stevens-Johnson syndrome (SJS) are rare yet life-threatening dermatologic conditions characterized by severe skin and mucous membrane involvement. Accurate prognostic systems are crucial for clinical management to assess disease severity and predict outcomes. The primary objective of this study was to assess the epidemiological characteristics and clinical outcomes of patients with Stevens-Johnson syndrome (SJS), toxic epidermal necrolysis (TEN), and SJS/TEN overlap over a 17-year period at a specialized burn center. The secondary objectives were to evaluate the performance of existing prognostic scoring systems (SCORTEN, Re-SCORTEN, and ABCD-10) in predicting mortality and to propose a novel classification tree model to improve mortality prediction. Materials and Methods : A 17-year retrospective study at a burn center included 68 patients with SJS, SJS/TEN overlap, or TEN. Demographic, clinical, laboratory data, and prognostic scores (SCORTEN, Re-SCORTEN, ABCD-10) were collected and analyzed for associations with mortality. A classification tree was created to detect unknown determinants of SJS/TEN mortality. Results : The drug most frequently associated with the occurrence of SJS/TEN was metamizole. The mortality rate was 51%. Affected body surface area, platelet count, and serum blood urea nitrogen differed significantly between survivors and non-survivors. Regarding the scoring systems, only the Re-SCORTEN showed reliable differentiation for these groups. A classification tree model achieved an accuracy of 89% in predicting the mortality risk. In the ROC curve analysis, the AUC values were 0.88 for the classification tree, 0.66 for Re-SCORTEN, 0.61 for SCORTEN, and 0.56 for ABCD-10. Conclusions : This study explores mortality predictors in SJS/TEN via a classification tree model, highlighting potential factors for further investigation. While cautioning against immediate clinical application due to data constraints, the findings underscore the need for larger studies to validate and refine prediction models in this context.

In recent years, the search for more sustainable fillers for elastomeric composites than silica and carbon black has been underway. In this work, silanized starch was used as an innovative filler for elastomeric composites. Corn starch was chemically modified by silanization (with n-octadecyltrimethoxysilane) via a condensation reaction to produce a hydrophobic starch. Starch/natural rubber composites were prepared by mixing the modified starch with elastomer. The morphology, hydrophobicity, and chemical structure of starch after and before modification were studied. The results showed that starch after silanization becomes hydrophobic (θ w = 117.3°) with a smaller particle size. In addition, FT-IR spectrum analysis confirmed the attachment of silane groups to the starch. The modified starch dispersed better in the natural rubber matrix and obtained a more homogeneous morphology. The composite achieved the best dynamic (ΔG' = 203.8 kPa) and mechanical properties (TS b = 11.4 MPa) for compositions with 15 phr of modified starch. In addition, the incorporation of silanized starch improved the hydrophobicity of the composite (θ w = 117.8°). The higher starch content allowed the composites to achieve a higher degree of cross-linking, resulting in better resistance to swelling in organic solvents. This improvement is due to enhanced elastomer-filler interactions and reduced spaces that prevent solvent penetration into the material's depths. The improved mechanical properties and good dynamic properties, as well as improved hydrophobicity, were mainly due to improved interfacial interactions between rubber and starch. This study highlights the potential and new approach of silane-modified starch as a sustainable filler, demonstrating its ability to enhance the mechanical, dynamic, and hydrophobic properties of elastomeric composites while supporting greener material solutions for the rubber industry.

Civil engineering structures are subject to both static and dynamic loadings. This applies especially to buildings in seismic areas as well as bridges, viaducts, and road and railway structures loaded with road or rail traffic. One of the solutions used to repair and strengthen such structures in the event of emergency damage are fibre-reinforced polyurethanes (FRPUs). The article proposes a laboratory method for determining the dynamic complex modulus of FRPU composite tape. The theoretical basis for determining the complex modulus for the tested material is presented. Laboratory tests were carried out using the tensile method for four cyclic loading frequencies and a cyclic load ratio equal to 0.5. Under the assumed test conditions, the material showed a viscoelastic performance with a dominant elastic part (storage modulus). For a frequency of 0.1 Hz, the viscous part (loss modulus) was about 8% of the storage modulus value, while for a frequency of 10 Hz, this value was about 5%. For a loading frequency of 0.1 Hz, the elastic part of the complex modulus was about 1160 MPa, while for a frequency of 10 Hz, it was about 1790 MPa. With the increase in loading frequency, the absolute value of the complex modulus increased.

This study aimed to investigate the properties of tin(II) oxide (SnO) as an unconventional cross-linking agent for chloroprene (CR) and styrene-butadiene (SBR) rubbers compositions. The use of tin(II) oxide results from the need to reduce the use of zinc oxide as a cross-linking agent due to environmental regulations and its toxic impact on aquatic environments. The studied elastomeric blends can be cross-linked with tin(II) oxide, and the results demonstrate the significant potential of this oxide in such applications. The CR/SBR vulcanizates cross-linked with SnO exhibit good mechanical properties and a high degree of cross-linking. The studies clearly show that the proportions of both rubbers as well as the amount of tin(II) oxide used influence the cross-linking of the CR/SBR blends and the properties of vulcanizates. FTIR spectrum analysis allowed the identification of the cross-linking mechanism, which followed the Friedel-Crafts alkylation reaction mechanism. The AFM analysis determined the miscibility of the rubbers and interelastomeric reactions, proving that the rubbers studied are partially miscible. The results of the oxygen index measurements indicated that the obtained vulcanizates showed flame resistance and self-extinguishing properties. Multivariate regression was performed to fit the models to the experimental value and to determine the influence of the content of the cross-linking agent and the CR and SBR proportions on the properties of the blends.

This study explores the potential use of mould biomass and waste fibres for the production of agrotextiles. First, 20 mould strains were screened for efficient mycelium growth, with optimized conditions of temperature, sources of carbon and nitrogen in the medium, and type of culture (submerged or surface). A method was developed for creating a biocomposite based on the mould mycelium, reinforced with commercial bleached softwood kraft (BSK) pulp and fibre additives (cotton, hemp). The best properties, including mechanical, water permeability, and air permeability, were shown by the biocomposites containing 10-20% Cladosporium cladosporioides mycelium grown in surface or submerged cultures, milled with BSK pulp, cotton, and hemp (10-20%). The mould mycelium was refined with cellulosic fibrous material, formed, pressed, and dried, resulting in a biomaterial with good mechanical parameters, low water permeability, and high air permeability. The biocomposite was fully biodegradable in soil after 10 days in field conditions. The use of the biocomposite as a crop cover shortened the germination time and increased the percentage of germinated onion, but had no effect on parsley seeds. This study shows the potential of using mould mycelium for the production of biomaterial with good properties for applications in horticulture.

RAB11, a pivotal RabGTPase, regulates essential cellular processes such as endocytic recycling, exocytosis, and autophagy. The protein was implicated in various human diseases, including cancer, neurodegenerative disorders, viral infections, and podocytopathies. However, a small-molecular inhibitor is lacking. The complexity and workload associated with potential assays make conducting large-scale screening for RAB11 challenging. We employed a tiered approach for drug discovery, utilizing deep learning-based computational screening to preselect compounds targeting a specific pocket of RAB11 protein with experimental validation by an in vitro platform reflecting RAB11 activity through the exocytosis of GFP. Further validation included the exposure of Drosophila by drug feeding. In silico pre-screening identified 94 candidates, of which 9 were confirmed using our in vitro platform for Rab11 activity. Focusing on compounds with high potency, we assessed autophagy, which independently requires RAB11, and validated three of these compounds. We further analyzed the dose-response relationship, observing a biphasic, potentially hormetic effect. Two candidate compounds specifically caused a shift in Rab11 vesicles to the cell periphery, without significant impact on Rab5 or Rab7. Drosophila larvae exposed to another candidate compound with predicted oral bioavailability exhibited minimal toxicity, subcellular dispersal of endogenous Rab11, and a decrease in RAB11-dependent nephrocyte function, further supporting an inhibitory role. Taken together, the combination of computational screening and experimental validation allowed the identification of small molecules that modify the function of Rab11. This discovery may further open avenues for treating RAB11-associated disorders.

Background: Hexokinase (HK) deficiency is a rare autosomal recessively inherited disease manifested by chronic nonspherocytic hemolytic anemia. Most patients present with a mild to severe course of the disease (fetal hydrocephalus, neonatal hyperbilirubinemia, severe anemia). We reviewed 37 cases of patients with hexokinase deficiency described so far, focusing on the severity of the disease, clinical presentation, treatment applied, and genetic test results., Methods: We present a 10-year-old girl who initially presented with symptoms of weakness, excessive fatigue, and yellowing of the skin and sclerae. Genetic testing detected the (TA)7 variant in both alleles of the UGT1A1 gene and diagnosed Gilbert's disease. In the follow-up, red cell hemolysis was observed. The diagnosis was extended, and tests for red cell enzymopathy were performed and a reduced level of hexokinase-0.65 IU/gHb (normal 0.78-1.57) was found. Next-generation sequencing revealed a new sense-change variant in exon 7 in the hexokinase gene not previously reported in databases., Results: Up to this date, only around 37 cases of hexokinase deficiency associated with hereditary nonspherocytic hemolytic anemia have been documented around the world. Diagnosing hexokinase deficiency involves clinical evaluation, laboratory testing, and genetic analysis. Management focuses on treating symptoms and preventing complications; there is no cure for the underlying enzyme deficiency. In patients with severe anemia, the treatment is multiple blood transfusions followed by iron chelation therapy., Conclusions: Understanding and diagnosing hexokinase deficiency is critical for providing appropriate care and improving the quality of life for affected individuals.

This study investigates the dynamic stiffness and damping characteristics of three polyurethane materials-PM, PS, and PST-using a comprehensive vibroacoustic testing approach. The aim is to examine material parameters such as dynamic stiffness, Young's modulus, critical damping factor, and the influence of sample irregularities on the accuracy of measurements. The study employs both experimental testing, in which cuboidal and cylindrical polyurethane samples were subjected to sinusoidal excitation, and finite element modeling (FEM) to simulate the test conditions in sample without irregularities. Results indicate that sample contact surface irregularities (even as low as ~0.04 mm) significantly impact the measured dynamic stiffness, with the effect intensifying for materials with higher Young's modulus values (above 5 MPa). Furthermore, cylindrical samples demonstrated more stable and repeatable measurements compared to cuboidal samples, where surface irregularities were tested in a more controlled environment. The findings underscore the need to consider sample geometry and irregularities in dynamic stiffness assessments to ensure better material evaluations. This work contributes valuable insights for the accurate modeling and testing of materials used in vibration isolation and sound insulation contexts.

The growing antibiotic resistance of microorganisms causing postoperative infections following orthopedic surgeries underscores the urgent need for localized antiseptic and lavage delivery systems to enhance infection control. This study evaluates the in vitro effectiveness of antiseptic and lavage solutions-including polyhexanide, povidone-iodine, low-concentrated hypochlorite, Ringer's solution, and saline-against Staphylococcus epidermidis , Staphylococcus aureus MRSA, Cutibacterium acnes , Corynebacterium amycolatum , Pseudomonas aeruginosa , and Candida albicans . Using microplate models (Minimum Inhibitory Concentration, Minimum Biofilm Eradication Concentration, and Biofilm-Oriented Antiseptic Test assays), flow-based models (Bioflux system), and surfaces relevant to orthopedic implants (e.g., stainless steel disks/screws, Co-Cr-Mo, Ti-Al-Nb orthopedic alloys, and ultra-high-molecular-weight polyethylene), as well as a bio-nano-cellulose scaffold representing tissue, we assessed the solutions' activity. The cytotoxicity of the solutions was evaluated using osteoblast and keratinocyte cell lines, with additional in vivo insights gained through the Galleria mellonella larval model. The results show that polyhexanide-based solutions outperformed povidone-iodine in biofilm eradication in most tests applied, particularly on complex surfaces, whereas iodine demonstrated higher cytotoxicity in applied in vitro and in vivo tests. Low-concentration hypochlorite solutions exhibited minimal antibiofilm activity but also showed no cytotoxicity in cell line and G. mellonella larval models. These findings highlight the importance of careful antiseptic selection and rinsing protocols to balance infection control efficacy with tissue compatibility in orthopedic applications.

The aim of this study was to determine the frequency-temperature dependence of the AC conductivity and relaxation times in humid electrical pressboard used in the insulation of power transformers, impregnated with the innovative NYTRO ® BIO 300X bio-oil produced from plant raw materials. Tests were carried out for a composite of cellulose-bio-oil-water nanodroplets with a moisture content of 0.6% by weight to 5% by weight in the frequency range from 10 -4 Hz to 5·10 3 Hz. The measurement temperatures ranged from 20 °C to 70 °C. The current conductivity in percolation channels in cellulose-bio insulating oil-water nanodroplets nanocomposites was analyzed. In such nanocomposites, DC conduction takes place via electron tunneling between the potential wells formed by the water nanodroplets. It was found that the value of the percolation channel resistance is lowest in the case of a regular arrangement of the nanodroplets. As disorder increases, characterized by an increase in the standard deviation value, the percolation channel resistance increases. It was found that the experimental values of the activation energy of the conductivity and the relaxation time of the composite of cellulose-bio-oil-water nanodroplets are the same within the limits of uncertainty and do not depend on the moisture content. The value of the generalized activation energy is Δ E ≈ (1.026 ± 0.0160) eV and is constant over the frequency and temperature ranges investigated. This study shows that in the lowest frequency region, the conductivity value does not depend on frequency. As the frequency increases further, the relaxation time decreases; so, the effect of moisture on the conductivity value decreases. The dependence of the DC conductivity on the moisture content was determined. For low moisture contents, the DC conductivity is practically constant. With a further increase in water content, there is a sharp increase in DC conductivity. Such curves are characteristic of the dependence of the DC conductivity of composites and nanocomposites on the content of the conducting phase. A percolation threshold value of x c ≈ (1.4 ± 0.3)% by weight was determined from the intersection of flat and steeply sloping sections. The frequency dependence of the values of the relative relaxation times was determined for composites with moisture contents from 0.6% by weight to 5% by weight for a measurement temperature of 60 °C. The highest relative values of the relaxation time τ ref occur for direct current and for the lowest frequencies close to 10 -4 Hz. As the frequency increases further, the relaxation time decreases. The derivatives d ( logτ ref )/d(log f ) were calculated, from the analysis of which it was determined that there are three stages of relaxation time decrease in the nanocomposites studied. The first occurs in the frequency region from 10 -4 Hz to about 3·10 -1 Hz, and the second from about 3·10 -1 Hz to about 1.5·10 1 Hz. The beginning of the third stage is at a frequency of about 1.5·10 1 Hz. The end of this stage is above the upper range of the Frequency Domain Spectroscopy (FDS) meter, which is 5·10 3 Hz. It has been established that the nanodroplets are in the cellulose and not in the bio-oil. The occurrence of three stages on the frequency dependence of the relaxation time can be explained when the fibrous structure of the cellulose is taken into account. Nanodroplets, found in micelles, microfibrils and in the fibers of which cellulose is composed, can have varying distances between nanodroplets, determined by the dimensions of these cellulose components.

Salivary gland tumors are rare head and neck tumors constituting up to 6% of all head and neck neoplasms; despite being mostly benign, these tumors present in diverse histological subtypes, making them challenging to diagnose and treat. Our research aims to investigate the link between inflammation and tumorigenesis within the salivary glands based on the literature regarding the etiopathogenesis of benign salivary gland tumors. This scoping review was conducted following the PRISMA extension for scoping reviews and reporting guidelines. The search was conducted using the Pubmed and Embase databases. Articles published between 2004 and May 2024 were included in the review. A total of 1097 papers were collected and identified. After 271 duplicates were removed, 826 titles and abstracts were independently reviewed by two researchers. Based on the title and abstract, 735 citations were excluded, and 91 articles were assessed for eligibility. Data were extracted from 46 articles that met the inclusion criteria. The review highlights the significance of inflammation-related factors and its relations with benign salivary gland tumors (SGTs). Knowledge of the etiopathogenesis of these tumors remains insufficient, and their rich immunological background poses challenges in diagnosis. The findings also point to directions for further clinical research, which will be necessary to implement these molecules in clinical practice.

Disturbances in carbohydrate metabolism are suggested to be the early symptoms of pancreatic ductal adenocarcinoma (PDAC). The accumulated data suggests that endocrine function-related biomarkers may represent a breakthrough in the early detection of PDAC. Factors which may predispose one to the development of PDAC are insulin resistance and hyperinsulinemia. Elevated insulin levels induce the onset of carcinogenesis by altering the differentiation and function of islet cells through stimulating growth factors, including insulin-like growth factors (IGFs). Impaired β cell function, along with the impact of PDAC-released factors (e.g., adrenomedullin (ADM), IGF-1, and macrophage inhibitory factor (MIF) on pancreatic islets, may contribute to the induction of diabetes associated with PDAC. Recently, exosomes have attracted worldwide attention due to their role in varied features of cell function, particularly in cancer progression. Exosomes comprise of small extracellular vesicles produced by almost all cells. These vesicles contain a vast array of biomolecules, including proteins and microRNAs. Exosomes participate in cancer growth and promote angiogenesis. They promote tumorigenesis and metastasis, and are associated with the acquisition of cancer cells resistant to chemotherapy. Data have been accumulating recently on the role of exosomes in the rapid recognition, prognosis and potential therapy of pancreatic cancer.

Sonochemical synthesis is becoming a popular method of preparing various nanomaterials, including metals, carbons, oxides, and chalcogenides. This method is relatively cheap and responds to the challenges of green chemistry as it typically does not involve high temperatures, high pressures, inert atmospheres, or long reaction times in comparison to other conventional methods. The utilization of ultrasound in synthesis makes the elimination of toxic solvents possible, as well as the execution of the synthesis without the use of reducing and stabilizing agents, while receiving products with the same or even better properties. The application of ultrasound allows for the synthesis of various nanomaterials with different properties for use in fields such as catalysis, electrochemistry, medicine, and biosensors. The final product is influenced by multiple variables such as temperature, pH, reagents, capping agents, time of reaction, and the addition of dopants.

Urban green exercise offers substantial physical and mental health benefits, especially in urban environments with limited natural access. This study analyzes the demand for urban year-round green exercise under various meteorological conditions. Using a primary empirical research design, data were collected from 408 active participants in Germany through an online survey. The survey consisted of one questionnaire, with multiple sections including demographics, green exercise, year-round green exercise, and nature-relatedness. Spearman rank correlations and Pearson's correlation were conducted for data analysis, and linear regressions calculated differences between groups. The results indicate a high demand for green exercise, with most participants engaging in endurance-based sports in urban green spaces at least once a week, regardless of weather conditions. However, adverse weather, such as precipitation and extreme cold, significantly reduces the likelihood of green exercise. Furthermore, the study identifies a positive correlation between participants' connection to nature and their likelihood of engaging in green exercise under different weather conditions. The findings suggest that urban year-round green exercise could be a viable public health intervention, accessible to a broad demographic, with the potential to improve overall well-being. However, further research is needed to explore the barriers to green exercise in adverse weather and to develop strategies to promote year-round green exercise.

This paper presents the experimental and numerical results of a study on producing axisymmetric parts made of the C60-grade steel by skew rolling. The experimental part of this study involved conducting the skew rolling process with varying parameters, including the forming angle α , tool angle θ , chuck velocity V u , and reduction ratio δ . Their effect on the quality of produced parts was examined and described by the roughness parameter Ra. Numerical calculations involved the use of machine learning models to predict the quality class of produced parts. The highest prediction accuracy of the results was obtained with the random forest and logistic regression models. Metrics such as precision, recall and accuracy were used to evaluate the performance of individual models. Confusion matrices and ROC curves were also employed to illustrate the performance of the classification models. The results of this study will make it possible to prevent the formation of spiral grooves on the circumference of steel parts during the rolling process.

Background: The evaluation of evidence, which frequently takes the form of scientific evidence, necessitates the input of experts in relevant fields. The results are presented as expert opinions or expert evaluations, which are generally accepted as a reliable representation of the facts. A further issue that remains unresolved though is the process of evaluating the expertise and knowledge of an expert in the first instance. In general, earned certificates, grades and other objective criteria are typically regarded as representative documentation to substantiate an expert status. However, there is a possibility that these may not always be sufficiently representative., Objectives: The goal of the present study was to provide evidence that the neural processing of law-relevant and law-irrelevant terms varies significantly between participants who have received training in the field of law (experts) and those who have not (novices)., Methods: To this end, changes in brain activity were recorded via electroencephalography (EEG) during visual presentations of terms belonging to five different categories (fake right, democracy, filler word, basic right and rule of law). Event-related potentials (ERPs) were subsequently averaged for each category and subjected to statistical analysis., Results: The results clearly demonstrate that participants trained in law processed fake rights and filler words in a similar manner. Furthermore, both of these conditions elicited different levels of brain activity compared to all law-relevant terms. This was not the case in participants who had not received legal training. The brains of untrained participants processed all five term categories in a strikingly similar manner. In light of prior knowledge regarding language processing, the primary focus was on two distinct electrode locations: one in the left posterior region, and the other in the left frontal region. In both locations, the most prominent differences in brain activity elicited by the aforementioned term categories in law-trained participants occurred approximately 450 milliseconds after stimulus onset. The results were further corroborated by a repeated-measures ANOVA and subsequent t -tests, which also demonstrated the absence of this effect in law-untrained participants., Conclusions: The findings of this study provide empirical evidence that brain activity measurements, in particular ERPs, can be used to distinguish between experts trained in a specific field of expertise and novices in that field. Such findings have the potential to facilitate objective assessments of expertise, enabling comparisons between experts and novices that extend beyond traditional criteria such as qualifications and experience. Instead, individuals can be evaluated based on their cognitive processes, as observed through brain activity.

Bone repair and regeneration require physiological cues, including mechanical, electrical, and biochemical activity. Many biomaterials have been investigated as bioactive scaffolds with excellent electrical properties. Amongst biomaterials, piezoelectric materials (PMs) are gaining attention in biomedicine, power harvesting, biomedical devices, and structural health monitoring. PMs have unique properties, such as the ability to affect physiological movements and deliver electrical stimuli to damaged bone or cells without an external power source. The crucial bone property is its piezoelectricity. Bones can generate electrical charges and potential in response to mechanical stimuli, as they influence bone growth and regeneration. Piezoelectric materials respond to human microenvironment stimuli and are an important factor in bone regeneration and repair. This manuscript is an overview of the fundamentals of the materials generating the piezoelectric effect and their influence on bone repair and regeneration. This paper focuses on the state of the art of piezoelectric materials, such as polymers, ceramics, and composites, and their application in bone tissue engineering. We present important information from the point of view of bone tissue engineering. We highlight promising upcoming approaches and new generations of piezoelectric materials.

Genome skimming is a novel approach that enables obtaining large-scale genomic information based on high-copy DNA fractions from shallow whole-genome sequencing. The simplicity of this method, low analysis costs, and large amounts of generated data have made it widely used in plant research, including species identification, especially in the case of protected or endangered taxa. This task is particularly difficult in the case of closely related taxa. The Pinus mugo complex includes several dozen closely related taxa occurring in the most important mountain ranges in Europe. The taxonomic rank, origin, or distribution of many of these taxa have been debated for years. In this study, we used genome skimming and multilocus DNA barcoding approaches to obtain different sequence data sets and also to determine their genetic diversity and suitability for distinguishing closely related taxa in the Pinus mugo complex. We generated seven different data sets, which were then analyzed using three discrimination methods, i.e., tree based, distance based, and assembling species by automatic partitioning. Genetic diversity among populations and taxa was also investigated using haplotype network analysis and principal coordinate analysis. The proposed data set based on divergence hotspots is even twenty-times more variable than the other analyzed sets and improves the phylogenetic resolution of the Pinus mugo complex. In light of the obtained results, Pinus × rhaetica does not belong to the Pinus mugo complex and should not be identified with either Pinus uliginosa or Pinus rotundata . It seems to represent a fixed hybrid or introgressant between Pinus sylvestris and Pinus mugo . In turn, Pinus mugo and Pinus uncinata apparently played an important role in the origins of Pinus uliginosa and Pinus rotundata .

The aim of this study was to identify miRNAs that could potentially influence the activity of SMAD proteins involved in TGFβ signal transduction in five types of breast cancer in Polish women. Patients with five breast cancer subtypes were included in the study: luminal A ( n = 130), luminal B HER2- ( n = 100), luminal B HER2+ ( n = 96), non-luminal HER2+ ( n = 36), and TNBC ( n = 43). During surgery, tumor tissue was removed along with a margin of healthy tissue (control). Molecular analysis included determination of the expression of genes related to SMAD protein signal transduction using mRNA microarrays and reverse transcription quantitative polymerase chain reaction (RT-qPCR). Protein expression was determined using an enzyme-linked immunosorbent assay (ELISA). The miRNA profiling was performed using miRNA microarrays and the miRDB database. SMAD3 and SMAD5 were overexpressed in all types of breast cancer, which could be related to the reduced expression of miR-145, and the findings for SMAD4 and miR-155 were similar. Additionally, the level of SMAD7 was reduced, which may be due to the low activity of miR-15b and miR21b. This study determined the gene expression profiles involved in SMAD protein signal transduction across five different types of breast cancer and identified the miRNAs potentially regulating their activity. Overexpression of SMAD3, SMAD4, and SMAD5 suggests excessive activation of the TGFβ pathway, potentially promoting tumor growth and development. Concurrently, a significant reduction in SMAD7 expression removes inhibitory control in the TGFβ pathway, a phenomenon that is particularly evident in more aggressive breast cancer types.

A nonradiative recombination channel is proposed, which does not vanish at low temperatures. Defect-mediated nonradiative recombination, known as Shockley-Read-Hall (SRH) recombination, is reformulated to accommodate Coulomb attraction between the charged deep defect and the approaching free carrier. It is demonstrated that this effect may cause a considerable increase in the carrier velocity approaching the recombination center. The effect considerably increases the carrier capture rates. It is demonstrated that, in a typical semiconductor device or semiconductor medium, the SRH recombination rate at low temperatures is much higher and cannot be neglected. This effect renders invalid the standard procedure of estimating the radiative recombination rate by measuring the light output in cryogenic temperatures, as a significant nonradiative recombination channel is still present. We also show that SRH is more effective in the case of low-doped semiconductors, as effective screening by mobile carrier density could reduce the effect.

Dynamic recrystallisation (DRX) is one of the fundamental phenomena in materials science, significantly impacting the microstructure and mechanical properties of components subjected to large plastic deformations. Experimental research on that topic carried out for a wide range of new metallic materials is often supported by computational materials science. A direct consideration and detailed understanding of this phenomenon are possible with a class of full-field numerical models based on the cellular automata (CA) method. However, the classical CA approach is based on a regular, fixed computational space and has limitations in capturing large deformations of the computational domain. Therefore, the main goal of the work is to develop and implement an alternative solution to overcome this limitation. The proposed solution is based on coupling the finite element (FE) method with the random cellular automata (RCA) approach. Such a model can directly consider the influence of geometrical changes in microstructure during large plastic deformation on recrystallisation progress. Details of the developed RCA DRX model assumptions and coupling issues with FE mesh are discussed. Particular attention is also paid to increasing model efficiency and robustness studies.

Processing of cast-iron castings by machining is associated with a large amount of post-production waste in the form of cast-iron chips, which constitute up to about 5% of the weight of the entire casting. In the case of serial production, this generates large amounts of post-production waste, constituting a constantly growing scale of environmental problems. The aim of this research was to develop a simple and cheap method of utilizing post-production waste of gray cast-iron chips from the machining process for the production of small structural elements of water supply fittings. The analysis of the state of knowledge indicates that the simplest method of managing waste chips is to use them as a starting material in the process of manufacturing polymer composites. The most frequently chosen material for the matrix of polymer composites reinforced with metal powders is epoxy resin. The epoxy composite was produced by the vacuum-assisted casting method. This paper presents the results of tests of morphological, mechanical, and corrosion properties of epoxy composites filled with grey cast-iron powder with a grain size below 0.075 mm and a mass content in the composite of 65%. The composite cured at 130 °C for 90 min had the best mechanical properties. The sample cured at 130 °C for 90 min was observed to have the optimum effect, with a tensile strength of 28.35 MPa, a flexural strength of 55.4 MPa, and a compressive strength of 53.8 MPa. All tested composites were characterized by very good thermal resistance and, in comparison to gray cast iron, over 2.5 times lower weight and an over three times lower corrosion rate in the tap water environment.

The present study explores the synthesis and bio-safety evaluation of gadolinium-doped carbon quantum dots (GCQDs) as a potential dual-contrast agent for diagnostic imaging. GCQDs exhibit both fluorescent and magnetic properties, making them suitable for UV-Vis and magnetic resonance imaging (MRI). The synthesis of GCQDs was achieved via hydrothermal treatment, incorporating gadolinium into the carbon quantum dot matrix. The magnetic properties of GCQDs were analyzed, showing significantly enhanced values compared to gadobutrol, a common MRI contrast agent. However, synthesis constraints limit the gadolinium content achievable in nanodots. To assess the safety of GCQDs, their effects on the embryonic development of zebrafish ( Danio rerio ) were examined. Various concentrations of GCQDs were tested, observing mortality rates, hatchability, malformations, heartbeats, spontaneous movement, and GCQDs uptake. Dialysis studies indicated that gadolinium ions are incorporated into the internal structure of the carbon nanodots. Zebrafish toxicity tests revealed that while survival rates were comparable to control groups, hatchability decreased significantly with higher gadolinium concentrations in GCQDs. Fluorescence microscopy showed no statistical differences in the fluorescence intensity between groups. These findings suggest that GCQDs could serve as an effective dual-contrast agent, combining the optical imaging capabilities of CQDs with the enhanced MRI contrast provided by gadolinium. This study underscores the need for further research on the synthesis methods and biological interactions of GCQDs to ensure their safety and efficacy in medical applications.

We consider three different systems in a heat flow: an ideal gas, a van der Waals gas, and a binary mixture of ideal gases. We divide each system internally into two subsystems by a movable wall. We show that the direction of the motion of the wall, after release, under constant boundary conditions, is determined by the same inequality as in equilibrium thermodynamics dU-đQ≤0. The only difference between the equilibrium and non-equilibrium laws is the dependence of the net heat change, đQ, on the state parameters of the system. We show that the same inequality is valid when introducing the gravitational field in the case of both the ideal gas and the van der Waals gas in the heat flow. It remains true when we consider a thick wall permeable to gas particles and derive Archimedes' principle in the heat flow. Finally, we consider the Couette (shear) flow of the ideal gas. In this system, the direction of the motion of the internal wall follows from the inequality dE-đQ-đWs≤0, where dE is the infinitesimal change in total energy (internal plus kinetic) and đWs is the infinitesimal work exchanged with the environment due to the shear force imposed on the flowing gas. Ultimately, we synthesize all these cases within a general framework of the second law of non-equilibrium thermodynamics.

A study of the combustion processes of Ti/CuO and Ti/CuO/NC nanothermites prepared via electrospraying was conducted in this work. For this purpose, the compositions were thermally conditioned at 350, 550 and 750 °C, as selected based on our initial differential scanning calorimetry-thermogravimetry (DSC/TG) investigations. The tested compositions were analysed for chemical composition and morphology using SEM-EDS, Raman spectroscopy and XRD measurements. Additionally, the thermal behaviour and decomposition kinetics of compositions were explored by means of DSC/TG. The Kissinger and Ozawa methods were applied to the DSC curves to calculate the reaction activation energy. SEM-EDS analyses indicated that sintering accelerated with increasing equivalence ratio and there was a strong effect on the sintering process due to cellulose nitrate (NC) addition. The main combustion reaction was found to start at 420-450 °C, as confirmed by XRD and Raman study of samples annealed at 350 °C and 550 °C. Moreover, increasing the fuel content in the composition led to lower E a , higher reaction heats and a more violent combustion process. Conversely, the addition of NC had an ambiguous effect on E a . Finally, a multi-step combustion mechanism was proposed and is to some extent in line with the more general reactive sintering (RS) mechanism. However, unusual mass transfer was observed, i.e., to the fuel core, rather than the opposite, which is typically observed for Al-based nanothermites.

The objective of this study was to compile the currently available evidence regarding the functional and morphologic outcomes of functional orthodontic therapy for mandibular condyle fracture. We performed searches in PubMed and Google Scholar as well as manually (IOK issues 2008-2019) using the keywords "trauma", "TMJ", "activator", "condylar fracture", "fracture", "mandibular condylar fracture", "occlusal splint" and "functional appliance". Screening and analysis of study eligibility were performed using the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA). The search strategy identified 198 studies published between 1971 and 2018, with 93 studies remaining after removing duplicate hits. Of the 93 studies, 19 were included in this study, considering the inclusion criteria. There were 12 follow-up, 4 prospective, and 3 purely retrospective studies. Some of the studies showed good functional results of mandibular condyle fracture treatment, in addition to subjective patient satisfaction. The incisal edge difference could be increased to physiological ranges of >35 mm by means of activator therapy. Partial mandibular deviations to the fractured side remained post-therapy, especially after unilateral fractures. Fractures without significant dislocation and luxation showed radiographic changes in shape, described as hypoplastic and ellipsoid, in addition to good morphologic results. One study found that collum length shortened twofold after a luxated fracture compared with fractures without significant dislocation, despite activator therapy. Straightening of the fragment occurred only in the low-dislocated fragments. Overall, children showed a higher remodeling potential than adult patients. Several studies observed an improved clinical outcome for functional therapy after mandibular condyle fracture. The outcome is essentially determined by fracture type, fracture height, and age. Further studies, especially prospective studies, are necessary to improve the evidence of functional orthodontic therapy for mandibular condyle fractures.

Salivary gland tumors are highly variable in clinical presentation and histology. The World Health Organization (WHO) classifies 22 types of malignant and 11 types of benign tumors of the salivary glands. Diagnosis of salivary gland tumors is based on imaging (ultrasound, magnetic resonance imaging) and fine-needle aspiration biopsy, but the final diagnosis is based on histopathological examination of the removed tumor tissue. In this pilot study, we are testing a new approach to identifying peptide biomarkers in saliva that can be used to diagnose salivary gland tumors. The research material for the peptidomic studies was extracts from washings of neoplastic tissues and healthy tissues (control samples). At the same time, saliva samples from patients and healthy individuals were analyzed. The comparison of the peptidome composition of tissue extracts and saliva samples may allow the identification of potential peptide markers of salivary gland tumors in patients' saliva. The peptidome compositions extracted from 18 tumor and 18 healthy tissue samples, patients' saliva samples (11 samples), and healthy saliva samples (8 samples) were analyzed by LC-MS tandem mass spectrometry. A group of 109 peptides was identified that were present only in the tumor tissue extracts and in the patients' saliva samples. Some of the identified peptides were derived from proteins previously suggested as potential biomarkers of salivary gland tumors (ANXA1, BPIFA2, FGB, GAPDH, HSPB1, IGHG1, VIM) or tumors of other tissues or organs (SERPINA1, APOA2, CSTB, GSTP1, S100A8, S100A9, TPI1). Unfortunately, none of the identified peptides were present in all samples analyzed. This may be due to the high heterogeneity of this type of cancer. The surprising result was that extracts from tumor tissue did not contain peptides derived from salivary gland-specific proteins (STATH, SMR3B, HTN1, HTN3). These results could suggest that the developing tumor suppresses the production of proteins that are essential components of saliva.

In lung cancer patients, two complementary abnormalities were found that can cause disruption of the mitochondrial network: increased fusion and impaired fission, manifested by reduced levels of FIS1, a mitochondrial division regulator, and increased expression of MFN1, a mitochondrial fusion mediator. Immunoexpression studies of MFN1 and FIS1 proteins were performed in serum samples obtained from 47 patients with non-small cell lung cancer (NSCLC) and 21 controls. In the NSCLC patients, the immunoexpression of the MFN1 protein was significantly higher, and the FIS1 protein level was significantly lower than in the control group ( p < 0.01; p < 0.001; UMW test). Patients with early, operable lung cancer had significantly lower levels of MFN1 immunoexpression compared to patients with advanced, metastatic lung cancer ( p < 0.05; UMW test). This suggests that early stages of the disease are characterized by greater fragmentation of damaged mitochondria and apoptosis. In contrast, lower FIS1 protein levels were associated with a worse prognosis. Increased mitochondrial fusion in the blood of lung cancer patients may suggest an increase in protective and repair mechanisms. This opens up questions about why these mechanisms fail in the context of existing advanced cancer disease and is a starting point for further research into why protective mechanisms fail in lung cancer patients.

In this work, energetic coordination compounds (ECCs) of transition metals (Fe, Ni, Cu, Zn) containing aliphatic amines as ligands were synthesized: ethylenediamine; 1,3-diaminopropane; tris(2-aminoethyl)amine; tris(3-aminopropyl)amine. The compounds were investigated in terms of ignition/explosion temperature, friction and impact sensitivity. For selected compounds, structural characterisation was presented (IR-ATR spectroscopy, Raman spectroscopy) and their morphology was determined (SEM, powder XRD). They were also investigated by differential scanning calorimetry (DSC). In order to assess the potential application of selected ECCs in detonators, underwater explosion tests were carried out, determining energetic performance. The results achieved for detonators containing ECCs were compared with those for reference detonators (containing pentaerythritol tetranitrate, PETN), indicating their potential use as a "green" alternative to nitric acid esters.

Bi-allelic disruptive variants (nonsense, frameshift, and splicing variants) in KDM5B have been identified as causative for autosomal recessive intellectual developmental disorder type 65. In contrast, dominant variants, usually disruptive as well, have been more difficult to implicate in a specific phenotype, since some of them have been found in unaffected controls or relatives. Here, we describe individuals with likely pathogenic variants in KDM5B , including eight individuals with dominant missense variants. This study is a retrospective case series of 21 individuals with variants in KDM5B . We performed deep phenotyping and collected the clinical information and molecular data of these individuals' family members. We compared the phenotypes according to variant type and to those previously described in the literature. The most common features were developmental delay, impaired intellectual development, behavioral problems, autistic behaviors, sleep disorders, facial dysmorphism, and overgrowth. DD, ASD behaviors, and sleep disorders were more common in individuals with dominant disruptive KDM5B variants, while individuals with dominant missense variants presented more frequently with renal and skin anomalies. This study extends our understanding of the KDM5B -related neurodevelopmental disorder and suggests the pathogenicity of certain dominant KDM5B missense variants.

Nanothermites have found broad applications; however, due to being systems largely reacting in condensed phases, their performance is somewhat limited by heat and mass transfer. In order to alleviate this issue, nanothermites doped with gas-generating energetic materials have been developed. In this work, we present an investigation of a model Ti/CuO nanothermite doped by four classical energetic materials and investigate their properties and combustion performance. Mechanical and laser irradiation sensitivity, as well as ignition/explosion temperatures have been determined for the studied systems to establish their safety features. In terms of combustion performance, thrust force parameters and linear combustion velocity have been determined and the structure of the evolving flame front was recorded during open-air combustion experiments. The obtained results indicate that the developed doped nanothermite formulations are extremely promising materials for future applications.

Titanium dioxide was synthesized via hydrolysis of titanium (IV) isopropoxide using a sol-gel method, under neutral or basic conditions, and heated in the microwave-assisted solvothermal reactor and/or high-temperature furnace. The phase composition of the prepared samples was determined using the X-ray diffraction method. The specific surface area and pore volumes were determined through low-temperature nitrogen adsorption/desorption studies. The photoactivity of the samples was tested through photocatalytic reduction of carbon dioxide. The composition of the gas phase was analyzed using gas chromatography, and hydrogen, carbon oxide, and methane were identified. The influence of pH and heat treatment on the physicochemical properties of titania-based materials during photoreduction of carbon dioxide have been studied. It was found that the photocatalysts prepared in neutral environment were shown to result in a higher content of hydrogen, carbon monoxide, and methane in the gas phase compared to photocatalysts obtained under basic conditions. The highest amounts of hydrogen were detected in the processes using photocatalysts heated in the microwave reactor, and double-heated photocatalysts.

Although a significant body of evidence has attributed certain occupational exposures with leukemia, such as benzene, formaldehyde, 1,3-butadiene and ionizing radiation, more research is needed to identify work environments at increased risk for this disease. Our study aimed to identify occupational and industry groups associated with an elevated incidence of leukemia using a diverse cohort of workers' compensation claimants from Ontario, Canada. A total of 2,363,818 workers in the Occupational Disease Surveillance System (ODSS) cohort, with claims between 1983-2019, were followed for malignant leukemia diagnoses up to 31 December 2019. We used a Cox proportional-hazards model to estimate the relative incidence of leukemia in specific occupation and industry groups. After adjusting for age and birth year, males in protective services (HR = 1.17, 95% CI = 1.02-1.35), metal machining (HR = 1.23, 95% CI = 1.07-1.41), transport (HR = 1.15, 95% CI = 1.06-1.25), and mining occupations (HR = 1.28, 95% CI = 1.02-1.60) had elevated risks of leukemia compared to other workers in the ODSS, with comparable findings by industry. Among female workers, slight risk elevations were observed among product fabricating, assembling, and repairing occupations, with other increased risks seen in furniture and fixture manufacturing, storage, and retail industries. These findings underscore the need for exposure-based studies to better understand occupational hazards in these settings.

This was a retrospective study of the profile and initial treatments of adults diagnosed with early-stage (ES) non-small cell lung cancer (NSCLC) during January 2018-December 2021 at 16 leading hospital institutions in Austria, excluding patients enrolled in clinical trials. In total, 319 patients were enrolled at a planned ~1:1:1 ratio across StI:II:III. Most tested biomarkers were programmed death ligand 1 (PD-L1; 58% expressing), Kirsten rat sarcoma virus (KRAS; 22% positive), and epidermal growth factor receptor (EGFR; 18% positive). Of 115/98/106 StI/II/III patients, 82%/85%/36% underwent surgery, followed by systemic therapy in 9%/45%/47% of those [mostly chemotherapy (ChT)]. Unresected treated StIII patients received ChT + radiotherapy [43%; followed by immune checkpoint inhibitors (ICIs) in 39% of those], ICI ± ChT (35%), and ChT-alone/radiotherapy-alone (22%). Treatment was initiated a median (interquartile range) of 24 (7-39) days after histological confirmation, and 55 (38-81) days after first medical visit. Based on exploratory analyses of all patients newly diagnosed with any stage NSCLC during 2018-2021 at 14 of the sites (N = 7846), 22%/10%/25%/43% had StI/II/III/IV. The total number was not significantly different between pre-COVID-19 (2018-2019) and study-specific COVID-19 (2020-2021) periods, while StI proportion increased (21% vs. 23%; p = 0.012). Small differences were noted in treatments. In conclusion, treatments were aligned with guideline recommendations at a time which preceded the era of ICIs and targeted therapies in the (neo)adjuvant setting.

The replacement of polymers derived from petrochemical resources has been a prominent area of focus in recent decades. Polymers used in engineering materials must exhibit mechanical strength and stiffness while maintaining performance through a broad temperature range. Most of the polyesters used as engineering materials are based on terephthalic acid (TPA) and its derivatives, which provide necessary rigidity to molecular chains due to an aromatic ring. Bio-based alternatives for TPA-based polyesters that are gaining popularity are the polyesters derived from 2,5-furandicarboxylic acid (FDCA). To broaden applicational possibilities, one effective way to achieve specific properties in targeted applications is to adjust the composition and structure of polymers using advanced polymer chemistry techniques. The incorporation of rigid diols such as isosorbide, 1,4-cyclohexanedimethanol (CHDM), and 2,2,4,4-tetramethyl-1,3-cyclobutanediol (CBDO) should result in a greater stiffness of the molecular chains. This review extensively explores the effect of incorporating rigid diols on material properties through a review of research articles as well as patents. Moreover, this review mainly focuses on the polyesters and copolyesters synthesized via two-step melt polycondensation and its alterations due to the industrial importance of this method. Innovative synthesis strategies and the resulting material properties are presented.

Wood characterized by desired mechanical properties and wood joining material is essential for creating wooden structures. The polymer adhesives are suitable for such applications due to the possibility of energy dissipation from stresses generated by wooden structures and the elimination of thermal bridging, which are common problems in metal joining materials. This research focuses on the thermophysical properties of the laboratory-prepared flexible and rigid polyurethanes to select an appropriate polymer adhesive. Our results showed that the highest thermal stability was in the case of the new PSTF-S adhesive, which reached 230 °C, but the lowest mass loss in the air environment was around 54% for the PS material. The mean thermal expansion coefficient for F&R PU adhesives was 124-164∙10 -6 K -1 . The thermal diffusivity of examined adhesives varied between 0.100 and 0.180 mm 2 s -1 . The thermal conductivity, depending on the type of polyurethane, was in the 0.13-0.29 W∙m -1 ∙K -1 range. The relative decrease in thermal diffusivity after heating the adhesives to 150 °C was from 2% for materials with the lowest diffusivity to 23% for the PU with the highest value of heat transfer. This paper presents the synthesis and characterization of graphene-polymer composites, focusing on their mechanical and antibacterial properties. Graphene flakes were obtained via an electrochemical method and integrated into polyamide 6 (PA6) matrices using melt intercalation. Various characterization techniques confirmed the quality of the graphene flakes, including X-ray diffraction (XRD), Raman spectroscopy, and infrared (IR) spectroscopy, as well as scanning and transmission electron microscopy (SEM and TEM) imaging. Mechanical tests showed an increase in the elastic modulus with graphene incorporation, while the impact strength decreased. The SEM analysis highlighted the dispersion of the graphene flakes within the composites and their impact on fracture behavior. Antimicrobial tests demonstrated significant antibacterial properties of the composites, attributed to both oxidative stress and mechanical damage induced by the graphene flakes. The results suggest promising applications for graphene-polymer composites in advanced antimicrobial materials.

The combination of Direct Metal Deposition (DMD) with milling offers numerous advantages for the manufacturing of complex geometry parts demanding high dimensional accuracy and surface quality. To reach this, a process strategy alternation between both processes is often required, leaving the milled surface with a layer of cooling fluid before adding material by DMD. This paper investigates the effect of cooling liquid on the milled interface in the combined process of milling and DMD. Five different interface conditions were examined, employing four distinct cleaning techniques to assess their impact on the quality of the interface. Key metrics analysed included hydrogen content, carbon content, and porosity levels at the interface. Cleaning techniques were evaluated to determine their necessity in enhancing the interface quality in the combined DMD and milling production process. Results from this study provide essential insights into the optimal cleaning requirements for improving the interface integrity in hybrid manufacturing processes, which could lead to more reliable and efficient production methods in industrial applications.

Leukemias are cancers of the blood-forming system, representing a significant challenge in medical science. The development of leukemia cells involves substantial disturbances within the cellular machinery, offering hope in the search for effective selective treatments that could improve the 5-year survival rate. Consequently, the pathophysiological processes within leukemia cells are the focus of critical research. Enzymes such as cystathionine beta-synthase and sulfurtransferases like thiosulfate sulfurtransferase, 3-mercaptopyruvate sulfurtransferase, and cystathionine gamma-lyase play a vital role in cellular sulfur metabolism. These enzymes are essential to maintaining cellular homeostasis, providing robust antioxidant defenses, and supporting cell division. Numerous studies have demonstrated that cancerous processes can alter the expression and activity of these enzymes, uncovering potential vulnerabilities or molecular targets for cancer therapy. Recent laboratory research has indicated that certain leukemia cell lines may exhibit significant changes in the expression patterns of these enzymes. Analysis of the scientific literature and online datasets has confirmed variations in sulfur enzyme function in specific leukemic cell lines compared to normal leukocytes. This comprehensive review collects and analyzes available information on sulfur enzymes in normal and leukemic cell lines, providing valuable insights and identifying new research pathways in this field.
