Ablative composites serve as sacrificial materials, protecting underlying materials from high-temperature environments by endothermic reactions. These materials undergo various phenomena, including thermal degradation, pyrolysis, gas generation, char formation, erosion, gas flow, and different modes of heat transfer (such as conduction, convection, and radiation), all stemming from these endothermic reactions. These phenomena synergize to form a protective layer over the underlying materials. Carbon, with its superb mechanical properties and various available forms, is highlighted, alongside phenolics known for good adhesion and fabric ability and elastomers valued for flexibility and resilience. This study focuses on recent advancements in carbon-and-phenolic and carbon-and-elastomeric composites, considering factors such as erosion speed; high-temperature resistance; tensile, bending, and compressive strength; fiber–matrix interaction; and char formation. Various authors' calculations regarding the percentage reduction in linear ablation rate (LAR) and mass ablation rate (MAR) are discussed. Energy and water poverty are two main challenges of the modern world. Most developing and underdeveloped countries need more efficient electricity-producing sources to overcome the problem of potable water evaporation. At the same time, the traditional way to produce energy/electricity is also responsible for polluting the environment and damaging the ecosystem. Notably, many techniques have been used around the globe, such as a photovoltaic (PV) cooling (active, passive, and combined) process to reduce the working temperature of the PV panels (up to 60 °C) to improve the system efficiency. For floating photovoltaic (FPV), water cooling is mainly responsible for reducing the panel temperature to enhance the production capacity of the PV panels, while the system efficiency can increase up to around 30%. At the same time, due to the water surface covering, the water loss due to evaporation is also minimized, and the water evaporation could be minimized by up to 60% depending on the total area covered by the water surfaces. Therefore, it could be the right choice for generating clean and green energy, with dual positive effects. The first is to improve the efficiency of the PV panels to harness more energy and minimize water evaporation. This review article focuses mainly on various PV and FPV cooling methods and the use and advantages of FPV plants, particularly covering efficiency augmentation and reduction of water evaporation due to the installation of PV systems on the water bodies. This study explores the high-temperature (HT) tensile rupture characteristics of a dissimilar gas-tungsten-arc-welded (GTAW) joint between P92 steel and Alloy 617, fabricated using ER62S-B9 and ERNiCrCoMo-1 fillers. The high-temperature tensile tests were performed at elevated temperatures of 550 °C and 650 °C. An optical microscope (OM) and a field emission scanning electron microscope (FESEM) were utilized to characterize the joint. The high-temperature test results indicated that the specimen failed at the P92 base metal/intercritical heat-affected zone (ICHAZ) rather than the weld metal for the ERNiCrCoMo-1(IN617) filler. This finding confirmed the suitability of the joint for use in the Indian advanced ultra-supercritical (A-USC) program. The fracture surface morphology and presence of precipitates were analysed using an SEM equipped with energy dispersive spectroscopy (EDS). The appearance of the dimples and voids confirmed that both welded fillers underwent ductile–dominant fracture. EDS analysis revealed the presence of Cr-rich M23C6 phases, which was confirmed on the fracture surface of the ER62S-B9 weld (P92-weld). The capacity of aquifers to store water and the stability of infrastructure can each be adversely influenced by variations in groundwater levels and subsequent land subsidence. Along the south bank of the River Thames, the Battersea neighbourhood of London is renovating a vast 42-acre (over 8 million sq ft) former industrial brownfield site to become host to a community of homes, shops, bars, restaurants, cafes, offices, and over 19 acres of public space. For this renovation, between 2016 and 2020, a significant number of bearing piles and secant wall piles, with diameters ranging from 450 mm to 2000 mm and depths of up to 60 m, were erected inside the Battersea Power Station. Additionally, there was considerable groundwater removal that caused the water level to drop by 2.55 ± 0.4 m/year between 2016 and 2020, as shown by Environment Agency data. The study reported here used Sentinel-1 C-band radar images and the persistent scatterer interferometric synthetic aperture radar (PSInSAR) methodology to analyse the associated land movement for Battersea, London, during this period. The average land subsidence was found to occur at the rate of −6.8 ± 1.6 mm/year, which was attributed to large groundwater withdrawals and underground pile construction for the renovation work. Thus, this study underscores the critical interdependence between civil engineering construction, groundwater management, and land subsidence. It emphasises the need for holistic planning and sustainable development practices to mitigate the adverse effects of construction on groundwater resources and land stability. By considering the Sustainable Development Goals (SDGs) outlined by the United Nations, particularly Goal 11 (Sustainable Cities and Communities) and Goal 6 (Clean Water and Sanitation), city planners and stakeholders can proactively address these interrelated challenges. Power electronic systems have a great impact on modern society. Their applications target a more sustainable future by minimizing the negative impacts of industrialization on the environment, such as global warming effects and greenhouse gas emission. Power devices based on wide band gap (WBG) material have the potential to deliver a paradigm shift in regard to energy efficiency and working with respect to the devices based on mature silicon (Si). Gallium nitride (GaN) and silicon carbide (SiC) have been treated as one of the most promising WBG materials that allow the performance limits of matured Si switching devices to be significantly exceeded. WBG-based power devices enable fast switching with lower power losses at higher switching frequency and hence, allow the development of high power density and high efficiency power converters. Seaweeds are well known for having a wealth of nutritional benefits and providing ecological support to associated fauna. Seasonality influences the biochemical characteristics, affecting their ecological and economic values. In the present study, we evaluated pigments, primary and secondary metabolites, minerals, and antioxidant properties of green seaweed Chaetomorpha antennina growing on the intertidal rocks along the Covelong coast, India, in different seasons (from June 2019 to March 2020). Significant variations were found in the levels of antioxidants, minerals, and metabolites in different seasons, e.g., amino acid levels were the highest in post-monsoon and the lowest in summer. In monsoon, we found the highest concentration of fatty acids in the thalli. Lipid peroxidation and total antioxidant activity were at their maximum levels during post-monsoon, which indicated oxidative damage responses. No significant variations were found in the levels of photosynthetic pigments. The outcomes indeed suggested seasonal variations in the biochemical and nutrient profile of C. antennina. We suggest that the harvesting/collection of C. antennina for different nutrients and metabolites should be performed in the respective seasons. [ABSTRACT FROM AUTHOR]

The 9% Cr steels were developed for ultra-supercritical (USC) power plants to meet the requirements of high operating temperature and pressure. These steels are produced to operate at high temperatures where impact toughness is not a concern; however, it becomes important for the welded joints to have good impact toughness at room temperature for manufacturing. The present work investigates the effect of the post-weld heat treatment (PWHT) parameters, i.e., temperature and time, on the impact toughness of multi-pass gas tungsten arc welded (GTAW) joints of ferritic/martensitic grade P92 steel. The microstructural evolution in welded joints given varying post-weld temperatures and times was studied. The lath martensitic structure of the weld metal for the as-welded joints resulted in high hardness and low impact toughness. The weld fusion zone toughness was 12 J, which was lower than the minimum specified values of 41 J (ASME standards) and 47 J (EN ISO 3580:2017). The PWHT temperature and time were found to have a significant effect on the impact toughness of the weld metal. A drastic increase in the impact toughness of the weld metal was noticed, which was attributed to lath break-up, reduction in dislocation density and reduction in solid solution hardening. The maximum impact toughness of 124 J was measured for PWHT temperature and time of 760 °C and 120 min, respectively. The effect of PWHT parameters on tensile strength was also investigated, and test results showed that the joint was safe for USC boiler application as it failed from the region of the P92 base metal. The variation in microstructural evolution along the weldments resulted in hardness variation. PWHT led to homogeneity in microstructure and, ultimately, reduction in hardness value. According to the study, the optimum temperature and time for PWHT of a GTAW joint of P92 steel were found to be 760 °C and 120 min, respectively. [ABSTRACT FROM AUTHOR]

The native function of amyloid-β (Aβ) peptides is still unexplored. However, several recent reports suggest a prominent role of Aβ peptides in acetylcholine homeostasis. To clarify this role of Aβ, we have reported that Aβ peptides at physiological concentrations can directly enhance the catalytic efficiency of the key cholinergic enzyme, choline acetyltransferase (ChAT), via an allosteric interaction. In the current study, we further aimed to elucidate the underlying ChAT-Aβ interaction mechanism using in silico molecular docking and dynamics analysis. Docking analysis suggested two most probable binding clusters on ChAT for Aβ40 and three for Aβ42. Most importantly, the docking results were challenged with molecular dynamic studies of 100 ns long simulation in triplicates (100 ns × 3 = 300 ns) and were analyzed for RMSD, RMSF, RoG, H-bond number and distance, SASA, and secondary structure assessment performed together with principal component analysis and the free-energy landscape diagram, which indicated that the ChAT-Aβ complex system was stable throughout the simulation time period with no abrupt motion during the evolution of the simulation across the triplicates, which also validated the robustness of the simulation study. Finally, the free-energy landscape analysis confirmed the docking results and demonstrated that the ChAT-Aβ complexes were energetically stable despite the unstructured nature of C- and N-terminals in Aβ peptides. Overall, this study supports the reported in vitro findings that Aβ peptides, particularly Aβ42, act as endogenous ChAT-Potentiating-Ligand (CPL), and thereby supports the hypothesis that one of the native biological functions of Aβ peptides is the regulation of acetylcholine homeostasis. Herein, we report the designing of a C3N4/BiOI heterostructure that is supported on gum acacia-crosslinked-poly(acrylamide) hydrogel to fabricate a novel nanocomposite hydrogel. The potential application of the obtained nanocomposite hydrogel to remediate crystal violet dye (CVD) in an aqueous solution was explored. The structural and functional analysis of the nanocomposite hydrogel was performed by FTIR (Fourier transform infrared spectroscopy), X-ray diffraction (XRD), transmission electron microscopy (TEM), and scanning electron microscopy (SEM). The different reaction parameters, such as CVD concentration, nanocomposite hydrogel dosage, and working pH, were optimized. The C3N4/BiOI heterostructure of the nanocomposite hydrogel depicts Z-scheme as the potential photocatalytic mechanism for the photodegradation of CVD. The effects of the sterilization process have been studied on medical grade thermoplastic polyetheretherketone (PEEK). For a reusable medical device, material reliability is an important parameter to decide its lifetime, as it will be subjected to the continuous steam sterilization process. A spring nature, clip component was selected out of a newly designed medical device (patented) to perform this reliability study. This clip component was sterilized for a predetermined number of cycles (2, 4, 6, 8, 10, 20 . . . 100) at 121 °C for 30 min. A significant decrease of ~20% in the compression force of the spring was observed after 30 cycles, and a ~6% decrease in the lateral dimension of the clip was observed after 50 cycles. No further significant change in the compression force or dimension was observed for the subsequent sterilization cycles. Vickers hardness and differential scanning calorimetry (DSC) techniques were used to characterize the effects of sterilization. DSC results exhibited no significant change in the degree of cure and melting behavior of PEEK before and after the sterilization. Hardness measurement exhibited an increase of ~49% in hardness after just 20 cycles. When an unsterilized sample was heated for repetitive cycles without the presence of moisture (121 °C, 10 and 20 cycles), only ~7% of the maximum change in hardness was observed. In the present study, the vertical and horizontal growth of Patna Urban Agglomeration was evaluated using the Persistent Scatterer Interferometry Synthetic Aperture Radar (PSInSAR) technique during 2015–2018. The vertical urban growth assessment of the city landscape was assessed using microwave time series (30 temporal) datasets of Single Look Complex (SLC) Sentinel-1A interferometric Synthetic Aperture Radar using SARPROZ software (ver. 2020). This study demonstrated that peripheral city regions experienced higher vertical growth (~4 m year−1) compared to the city core regions, owing to higher urban development opportunities leading to significant land use alterations, the development of high-rise buildings, and infrastructural development. While the city core of Patna observed an infill and densification process, as it was already saturated and highly densified. The rapidly urbanizing city in the developing region witnessed a considerable horizontal urban expansion as estimated through the normalized difference index for built-up areas (NDIB) and speckle divergence (SD) using optical Sentinel 2A and microwave Sentinel-1A ground range detected (GRD) satellite data, respectively. The speckle divergence-based method exhibited high urban growth (net growth of 11.28 km2) with moderate urban infill during 2015–2018 and reported a higher accuracy as compared to NDIB. This study highlights the application of SAR remote sensing for precise urban area delineation and temporal monitoring of urban growth considering horizontal and vertical expansion through processing a long series of InSAR datasets that provide valuable information for informed decision-making and support the development of sustainable and resilient cities. Several cellular functions such as apoptosis, cellular proliferation, inflammation, and immune regulation involve the tumor necrosis factor-α(TNF)/TNF receptor (TNFR) pathway. Human immunodeficiency virus 1 (HIV-1) interacts with the TNF/TNFR pathway. The activation of the TNF/TNFR pathway impacts HIV-1 replication, and the TNF/TNFR pathway is the target of HIV-1 proteins. A hallmark of HIV-1 infection is immune activation and inflammation with increased levels of TNF in the plasma and the tissues. Therefore, the control of the TNF/TNFR pathway by new therapeutic approaches could participate in the control of immune activation and impact both viral replication and viral persistence. In this review, we will describe the intricate interplay between HIV-1 proteins and TNF/TNFR signaling and how TNF/TNFR activation modulates HIV-1 replication and discuss new therapeutic approaches, especially anti-TNF therapy, that could control this pathway and ultimately favor the clearance of infected cells to cure HIV-infected patients. First-pass metabolism alters arsenic biotransformation and its immunomodulatory activities. This study aims to determine the mRNA expression of intestinal-immunity- and permeability-associated genes, levels of cytokine/chemokines and levels of immunoglobulin isotypes when CD-1 mice were exposed to a single dose of intravenous (IV) sodium arsenite (50 µg/kg body weight (BW)) and to compare these responses to exposure via oral gavage (OG) (50 µg/kg BW). Samples were collected at 1, 4, 24 and 48 h post IV exposure and 24 and 48 h post OG. Sodium arsenite IV exposure led to a transient modulation of mRNA expression and protein levels of immunity-related genes involved in inflammation/apoptotic pathways and production of cytokines/chemokines, whereas it also led to downregulated expression of genes encoding tight junction, focal adhesion, and gap junction proteins, which are responsible for maintaining cell permeability. Oral exposure perturbed fewer cell-permeability-related genes at 24 and 48 h post exposure. At 24 h post exposure, OG decreased IgA and IgG2b levels; however, IV exposure significantly increased IgG2b, IgG3 and IgA in ileal tissue. Earlier, we showed significant downregulation of mRNA expression of genes involved in the immune-related pathways during OG in the intestinal mucosa of the same animals. Cumulatively, these results provide evidence that the exposure route of a xenobiotic can differentially impact the intestinal responses due to the impact of first-pass metabolism. Antibiotic water contamination is a growing environmental problem in the present day. As a result, water treatment is required for its reduction and elimination. Due to their important role in resolving this issue, photocatalysts have drawn a great deal of interest over the past few decades. When non-biodegradable organic matter is present in polluted water, the photo catalytic process, which is both environmentally friendly and an improved oxidation method, can be an effective means of remediation. In this regard, we report the successful synthesis of pure phased rare earth doped ZnO nanoparticles for tetracycline degradation. The prepared catalysts were systematically characterized for structural, optical, and magnetic properties. The optical band gap was tailored by rare earth doping, with redshift for Sm and Dy doped nanoparticles and blueshift for Nd doped ZnO nanoparticles. The analysis of photoluminescence spectra revealed information about the defect chemistry of all synthesised nanoparticles. Magnetic studies revealed that all synthesized diluted magnetic semiconductors exhibit room temperature ferromagnetism and can be employed for spintronic applications. Moreover, Dy doped ZnO nanoparticles were found to exhibit a maximum degradation efficiency of 74.19% for tetracycline (TCN) removal. The synthesized catalysts were also employed for the degradation of Malachite green (MG), and Crystal violet (CV) dyes. The maximum degradation efficiency achieved was 97.18% for MG and 98% for CV for Dy doped ZnO nanoparticles. Human immunodeficiency virus type 1 (HIV-1) targets CD4+ T cells and cells of the monocyte/macrophage lineage. HIV pathogenesis is characterized by the depletion of T lymphocytes and by the presence of a population of cells in which latency has been established called the HIV-1 reservoir. Highly active antiretroviral therapy (HAART) has significantly improved the life of HIV-1 infected patients. However, complete eradication of HIV-1 from infected individuals is not possible without targeting latent sources of infection. HIV-1 establishes latent infection in resting CD4+ T cells and findings indicate that latency can also be established in the cells of monocyte/macrophage lineage. Monocyte/macrophage lineage includes among others, monocytes, macrophages and brain resident macrophages. These cells are relatively more resistant to apoptosis induced by HIV-1, thus are important stable hideouts of the virus. Much effort has been made in the direction of eliminating HIV-1 resting CD4+ T-cell reservoirs. However, it is impossible to achieve a cure for HIV-1 without considering these neglected latent reservoirs, the cells of monocyte/macrophage lineage. In this review we will describe our current understanding of the mechanism of latency in monocyte/macrophage lineage and how such cells can be specifically eliminated from the infected host. Groundwater variation can cause land-surface movement, which in turn can cause significant and recurrent harm to infrastructure and the water storage capacity of aquifers. The capital cities in the England (London) and India (Delhi) are witnessing an ever-increasing population that has resulted in excess pressure on groundwater resources. Thus, monitoring groundwater-induced land movement in both these cities is very important in terms of understanding the risk posed to assets. Here, Sentinel-1 C-band radar images and the persistent scatterer interferometric synthetic aperture radar (PSInSAR) methodology are used to study land movement for London and National Capital Territory (NCT)-Delhi from October 2016 to December 2020. The land movement velocities were found to vary between −24 and +24 mm/year for London and between −18 and +30 mm/year for NCT-Delhi. This land movement was compared with observed groundwater levels, and spatio-temporal variation of groundwater and land movement was studied in conjunction. It was broadly observed that the extraction of a large quantity of groundwater leads to land subsidence, whereas groundwater recharge leads to uplift. A mathematical model was used to quantify land subsidence/uplift which occurred due to groundwater depletion/rebound. This is the first study that compares C-band PSInSAR-derived land subsidence response to observed groundwater change for London and NCT-Delhi during this time-period. The results of this study could be helpful to examine the potential implications of ground-level movement on the resource management, safety, and economics of both these cities. Mitochondria regulate a myriad of cellular functions. Dysregulation of mitochondrial control within airway epithelial cells has been implicated in the pro-inflammatory response to allergens in asthma patients. Because of their multifaceted nature, mitochondrial structure must be tightly regulated through fission and fusion. Dynamin Related Protein 1 (DRP1) is a key driver of mitochondrial fission. During allergic asthma, airway epithelial mitochondria appear smaller and structurally altered. The role of DRP1-mediated mitochondrial fission, however, has not been fully elucidated in epithelial response to allergens. We used a Human Bronchial Epithelial Cell line (HBECs), primary Mouse Tracheal Epithelial Cells (MTECs), and conditional DRP1 ablation in lung epithelial cells to investigate the impact of mitochondrial fission on the pro-inflammatory response to house dust mite (HDM) in vitro and in vivo. Our data suggest that, following HDM challenge, mitochondrial fission is rapidly upregulated in airway epithelial cells and precedes production of pro-inflammatory cytokines and chemokines. Further, deletion of Drp1 in lung epithelial cells leads to decreased fission and enhanced pro-inflammatory signaling in response to HDM in vitro, as well as enhanced airway hyper-responsiveness (AHR), inflammation, differential mucin transcription, and epithelial cell death in vivo. We propose a device for monitoring the number of people who are physically present inside indoor environments. The device performs local processing of infrared array sensor data detecting people's direction, which allows monitoring users' occupancy in any space of the building and also respects people privacy. The device implements a novel real-time pattern recognition algorithm for processing data sensed by a low-cost infrared (IR) array sensor. The computed information is transferred through a Z-Wave network. On-field evaluation of the algorithm has been conducted by placing the device on top of doorways in offices and laboratory rooms. To evaluate the performance of the algorithm in varying ambient temperatures, two groups of stress tests have been designed and performed. These tests established the detection limits linked to the difference between the average ambient temperature and perturbation. For an in-depth analysis of the accuracy of the algorithm, synthetic data have been generated considering temperature ranges typical of a residential environment, different human walking speeds (normal, brisk, running), and distance between the person and the sensor (1.5 m, 5 m, 7.5 m). Ankyrin repeat proteins are found in all three kingdoms of life. Fundamentally, these proteins are involved in protein-protein interaction in order to activate or suppress biological processes. The basic architecture of these proteins comprises repeating modules forming elongated structures. Due to the lack of long-range interactions, a graded stability among the repeats is the generic properties of this protein family determining both protein folding and biological function. Protein folding intermediates were frequently found to be key for the biological functions of repeat proteins. In this review, we discuss most recent findings addressing this close relation for ankyrin repeat proteins including DARPins, Notch receptor ankyrin repeat domain, IκBα inhibitor of NFκB, and CDK inhibitor p19INK4d. The role of local folding and unfolding and gradual stability of individual repeats will be discussed during protein folding, protein-protein interactions, and post-translational modifications. One-dimensional metal nanowires, with novel functionalities like electrical conductivity, optical transparency and high mechanical stiffness, have attracted widespread interest for use in applications such as transparent electrodes in optoelectronic devices and active components in nanoelectronics and nanophotonics. In particular, silver nanowires (AgNWs) have been widely researched owing to the superlative thermal and electrical conductivity of bulk silver. Herein, we present a detailed review of the synthesis of AgNWs and their utilization in fabricating improved transparent conducting electrodes (TCE). We discuss a range of AgNW synthesis protocols, including template assisted and wet chemical techniques, and their ability to control the morphology of the synthesized nanowires. Furthermore, the use of scalable and cost-effective solution deposition methods to fabricate AgNW based TCE, along with the numerous treatments used for enhancing their optoelectronic properties, are also discussed. New developments require innovative ecofriendly materials defined by their biocompatibility, biodegradability, and versatility. For that reason, the scientific society is focused on biopolymers such as chitosan, which is the second most abundant in the world after cellulose. These new materials should show good properties in terms of sustainability, circularity, and energy consumption during industrial applications. The idea is to replace traditional raw materials with new ecofriendly materials which contribute to keeping a high production rate but also reducing its environmental impact and the costs. The chitosan shows interesting and unique properties, thus it can be used for different purposes which contributes to the design and development of sustainable novel materials. This helps in promoting sustainability through the use of chitosan and diverse materials based on it. For example, it is a good sustainable alternative for food packaging or it can be used for sustainable agriculture. The chitosan can also reduce the pollution of other industrial processes such as paper production. This mini review collects some of the most important advances for the sustainable use of chitosan for promoting circular economy. First-generation cysteine-based site-specific antibody–drug conjugates (ADCs) are limited to one drug per cysteine. However, certain applications require a high drug to antibody ratio (DAR), such as when low-potency payloads are used. Higher drug load can be achieved using classical cysteine conjugation methods, but these result in heterogeneity, suboptimal efficacy and pharmacokinetics. Here, we describe the design, synthesis and validation of heterobifunctional linkers that can be used for the preparation of ADCs with a DAR of two, three and four in a site-specific manner per single cysteine conjugation site, resulting in site-specific ADCs with a DAR of four, six and eight. The designed linkers carry a sulfhydryl-specific iodoacetyl reactive group, and multiple cyclic diene moieties which can efficiently react with maleimide-carrying payloads through the Diels–Alder reaction. As a proof of concept, we synthesized site-specific DAR four, six and eight ADCs carrying tubulysin (AZ13601508) using engineered antibodies with a cysteine inserted after position 239 in the antibody CH2 domain. We evaluated and compared the in vitro cytotoxicity of ADCs obtained via the site-specific platform described herein, with ADCs prepared using classical cysteine conjugation. Simple Summary: The rat progression elevated gene-3 (PEG-3) promoter displays cancer-selective expression, whereas the rat growth arrest and DNA damage inducible gene-34 (GADD34) promoter lacks cancer specificity. PEG-3 and GADD34 minimal promoters display strong sequence homology except for two single point mutations. Since mutations are prevalent in many gene promoters resulting in significant alterations in promoter specificity and activity, we have explored the relevance of these two nucleotide alterations in determining cancer-selective gene expression. We demonstrate that these two point mutations are required to transform a non-cancer-specific promoter (pGADD) into a cancer-selective promoter (pGAPE). Additionally, we found GATA2 transcription factor binding sites in the GAPE-Prom, which regulates pGAPE activity selectively in cancer cells. This newly created pGAPE has all the necessary elements making it an appropriate genetic tool to noninvasively deliver imaging agents to follow tumor growth and progression to metastasis and for generating conditionally replicating adenoviruses that can express and deliver their payload exclusively in cancer. Progression-elevated gene-3 (PEG-3) and rat growth arrest and DNA damage-inducible gene-34 (GADD34) display significant sequence homology with regulation predominantly transcriptional. The rat full-length (FL) and minimal (min) PEG-3 promoter display cancer-selective expression in rodent and human tumors, allowing for cancer-directed regulation of transgenes, viral replication and in vivo imaging of tumors and metastases in animals, whereas the FL- and min-GADD34-Prom lack cancer specificity. Min-PEG-Prom and min-GADD34-Prom have identical sequences except for two single-point mutation differences (at −260 bp and +159 bp). Engineering double mutations in the min-GADD34-Prom produce the GAPE-Prom. Changing one base pair (+159) or both point mutations in the min-GADD34-Prom, but not the FL-GADD34-Prom, results in cancer-selective transgene expression in diverse cancer cells (including prostate, breast, pancreatic and neuroblastoma) vs. normal counterparts. Additionally, we identified a GATA2 transcription factor binding site, promoting cancer specificity when both min-PEG-Prom mutations are present in the GAPE-Prom. Taken together, introducing specific point mutations in a rat min-GADD34-Prom converts this non-cancer-specific promoter into a cancer-selective promoter, and the addition of GATA2 with existing AP1 and PEA3 transcription factors enhances further cancer-selective activity of the GAPE-Prom. Due to the increasing demand in industrial application, nanofluids have attracted the considerable attention of researchers in recent decades. The addition of nanocellulose (CNC) with water (W) and ethylene glycol (EG) to a coolant for a radiator application exhibits beneficial properties to improve the efficiency of the radiator. The focus of the present work was to investigate the performance of mono or hybrid metal oxide such as Al2O3 and TiO2 with or without plant base-extracted CNC with varying concentrations as a better heat transfer nanofluid in comparison to distilled water as a radiator coolant. The CNC is dispersed in the base fluid of EG and W with a 60:40 ratio. The highest absorption peak was noticed at 0.9% volume concentration of TiO2, Al2O3, CNC, Al2O3/TiO2, and Al2O3/CNC nanofluids which indicates a better stability of the nanofluids' suspension. Better thermal conductivity improvement was observed for the Al2O3 nanofluids in all mono nanofluids followed by the CNC and TiO2 nanofluids, respectively. The thermal conductivity of the Al2O3/CNC hybrid nanofluids with 0.9% volume concentration was found to be superior than that of the Al2O3/TiO2 hybrid nanofluids. Al2O3/CNC hybrid nanofluid dominates over other mono and hybrid nanofluids in terms of viscosity at all volume concentrations. CNC nanofluids (all volume concentrations) exhibited the highest specific heat capacity than other mono nanofluids. Additionally, in both hybrid nanofluids, Al2O3/CNC showed the lowest specific heat capacity. The optimized volume concentration from the statistical analytical tool was found to be 0.5%. The experimental results show that the heat transfer coefficient, convective heat transfer, Reynolds number and the Nusselt number have a proportional relationship with the volumetric flow rate. Hybrid nanofluids exhibit better thermal conductivity than mono nanofluids. For instance, a better thermal conductivity improvement was shown by the mono Al2O3 nanofluids than the CNC and TiO2 nanofluids. The properties of trehalose + water mixtures are studied as a function of mixture composition and temperature using molecular dynamics simulations. As trehalose disaccharide has been proposed for dry preservation purposes, the objective of this work is to analyse the nanoscopic properties of the considered mixtures, in terms of aggregation, clustering, interactions energies, and local dynamics, and their relationships with hydrogen bonding. The reported results allow a detailed characterization of hydrogen bonding and its evolution with mixture composition and thus inferring the effects of trehalose on water structuring providing results to justify the mechanisms of trehalose acting as preservation agent. Antibiotic resistance is one of the main public health concerns of this century. This resistance is also associated with oxidative stress, which could contribute to the selection of resistant bacterial strains. Bearing this in mind, and considering that flavonoid compounds are well known for displaying both activities, we investigated a series of hydroxy-3-arylcoumarins with structural features of flavonoids for their antibacterial activity against different bacterial strains. Active compounds showed selectivity against the studied Gram-positive bacteria compared to Gram-negative bacteria. 5,7-Dihydroxy-3-phenylcoumarin (compound 8) displayed the best antibacterial activity against Staphylococcus aureus and Bacillus cereus with minimum inhibitory concentrations (MICs) of 11 μg/mL, followed by Staphylococcus aureus (MRSA strain) and Listeria monocytogenes with MICs of 22 and 44 μg/mL, respectively. Moreover, molecular docking studies performed on the most active compounds against Staphylococcus aureus tyrosyl-tRNA synthetase and topoisomerase II DNA gyrase revealed the potential binding mode of the ligands to the site of the appropriate targets. Preliminary structure–activity relationship studies showed that the antibacterial activity can be modulated by the presence of the 3-phenyl ring and by the position of the hydroxyl groups at the coumarin scaffold. Zeolites are porous minerals with high absorbency and ion-exchange capacity. Their molecular structure is a dense network of AlO4 and SiO4 that generates cavities where water and other polar molecules or ions are inserted/exchanged. Even though there are several synthetic or natural occurring species of zeolites, the most widespread and studied is the naturally occurring zeolite clinoptilolite (ZC). ZC is an excellent detoxifying, antioxidant and anti-inflammatory agent. As a result, it is been used in many industrial applications ranging from environmental remediation to oral applications/supplementation in vivo in humans as food supplements or medical devices. Moreover, the modification as micronization of ZC (M-ZC) or tribomechanically activated zeolite clinoptilolite (TMAZ) or furthermore as double tribomechanically activated zeolite clinoptilolite (PMA-ZC) allows improving its benefits in preclinical and clinical models. Despite its extensive use, many underlying action mechanisms of ZC in its natural or modified forms are still unclear, especially in humans. The main aim of this review is to shed light on the geochemical aspects and therapeutic potentials of ZC with a vision of endorsing further preclinical and clinical research on zeolites, in specific on the ZC and its modified forms as a potential agent for promoting human brain health and overall well-being. Nuclear magnetic resonance (NMR) spectroscopy enables the non-invasive observation of biochemical processes, in living cells, at comparably high spectral and temporal resolution. Preferably, means of increasing the detection limit of this powerful analytical method need to be applied when observing cellular processes under physiological conditions, due to the low sensitivity inherent to the technique. In this review, a brief introduction to in-cell NMR, protein–small molecule interactions, posttranslational phosphorylation, and hyperpolarization NMR methods, used for the study of metabolites in cellulo, are presented. Akt signaling plays a central role in many biological processes that are key players in human immunodeficiency virus 1 (HIV-1) pathogenesis. The persistence of latent reservoirs in successfully treated patients, mainly located in macrophages and latently infected resting CD4+ T cells, remains a major obstacle in HIV-1 eradication. We assessed the in vitro effects of an HIV protease inhibitor (PI) and a non-nucleoside reverse transcriptase inhibitor (NNRTI) on HIV-1 Nef-induced Akt activation in macrophages and on HIV-1 reactivation in U1 monocytoid cells. Ex vivo, we investigated the impact of combination antiretroviral therapy (cART) on Akt activation, as measured by flow cytometry, and on the viral reservoir size, quantified by qPCR, in monocytes and autologous resting CD4+ T cells from HIV-infected individuals (Trial registration: NCT02858414). We found that, in myeloid cells, both Akt activation and HIV-1 reactivation were inhibited by PI but not by NNRTI in vitro. Our results indicate that cART decreases Akt activation and reduces the size of the HIV reservoir in both monocytes and resting CD4+ T cells. Our study indicates that Akt activation could play a role in HIV reservoir formation, indicating that drugs which target Akt could be efficient for limiting its size in aviremic chronically infected patients. Reactive astrogliosis is an early event in the continuum of Alzheimer's disease (AD). Current advances in positron emission tomography (PET) imaging provide ways of assessing reactive astrogliosis in the living brain. In this review, we revisit clinical PET imaging and in vitro findings using the multi-tracer approach, and point out that reactive astrogliosis precedes the deposition of Aβ plaques, tau pathology, and neurodegeneration in AD. Furthermore, considering the current view of reactive astrogliosis heterogeneity—more than one subtype of astrocyte involved—in AD, we discuss how astrocytic body fluid biomarkers might fit into trajectories different from that of astrocytic PET imaging. Future research focusing on the development of innovative astrocytic PET radiotracers and fluid biomarkers may provide further insights into the heterogeneity of reactive astrogliosis and improve the detection of AD in its early stages. Amyotrophic lateral sclerosis (ALS) is a neuronal degenerative condition identified via a build-up of mutant aberrantly folded proteins. The native folding of polypeptides is mediated by molecular chaperones, preventing their pathogenic aggregation. The mutant protein expression in ALS is linked with the entrapment and depletion of chaperone capacity. The lack of a thorough understanding of chaperones' involvement in ALS pathogenesis presents a significant challenge in its treatment. Here, we review how the accumulation of the ALS-linked mutant FUS, TDP-43, SOD1, and C9orf72 proteins damage cellular homeostasis mechanisms leading to neuronal loss. Further, we discuss how the HSP70 and DNAJ family co-chaperones can act as potential targets for reducing misfolded protein accumulation in ALS. Moreover, small HSPB1 and HSPB8 chaperones can facilitate neuroprotection and prevent stress-associated misfolded protein apoptosis. Designing therapeutic strategies by pharmacologically enhancing cellular chaperone capacity to reduce mutant protein proteotoxic effects on ALS pathomechanisms can be a considerable advancement. Chaperones, apart from directly interacting with misfolded proteins for protein quality control, can also filter their toxicity by initiating strong stress-response pathways, modulating transcriptional expression profiles, and promoting anti-apoptotic functions. Overall, these properties of chaperones make them an attractive target for gaining fundamental insights into misfolded protein disorders and designing more effective therapies against ALS. The surge in electric vehicles (EVs) and their electrical appliances requires highly efficient, lightweight electrical machines with better performance. However, conventional wire used for electrical machine windings have certain limits to the current requirements. Copper is a commonly used material in electrical windings, and due to its ohmic resistance, it causes 75% of total losses in electrical machines (copper losses). The high mass of the copper results in a bulky system size, and the winding temperature of copper is always maintained at less than 150 °C to preserve the thermal insulation of the electric machine of the windings. On the other hand, carbon nanotubes and carbon nanotube materials have superior electrical conductivity properties and mechanical properties. Carbon nanotubes ensure 100 MS/m of electrical conductivity, which is higher than the copper electrical conductivity of 59.6 MS/m. In the literature, various carbon nanotubes have been studied based on electrical conductivity, temperature co-efficient with resistivity, material thickness and strength, insulation, and efficiency of the materials. The inclusion of physiographic and atmospheric influences is critical for spatial modeling of orographic precipitation in complex terrains. However, attempts to incorporate cloud cover frequency (CCF) data when interpolating precipitation are limited. CCF considers the rain shadow effect during interpolation to avoid an overly strong relationship between elevation and precipitation in areas at equivalent altitudes across rain shadows. Conventional multivariate regression or geostatistical methods assume the precipitation–explanatory variable relationship to be steady, even though this relation is often non-stationarity in complex terrains. This study proposed a novel spatial mapping approach for precipitation that combines regression-kriging (RK) to leverage its advantages over conventional multivariate regression and the spatial autocorrelation structure of residuals via kriging. The proposed hybrid model, RK (GT + CCF), utilized CCF and other physiographic factors to enhance the accuracy of precipitation interpolation. The implementation of this approach was examined in a mountainous region of southern Syria using in situ monthly precipitation data from 57 rain gauges. The RK model's performance was compared with conventional multivariate regression models (CMRMs) that used geographical and topographical (GT) factors and CCF as predictors. The results indicated that the RK model outperformed the CMRMs with a root mean square error of <8 mm, a mean absolute percentage error range of 5–15%, and an R2 range of 0.75–0.96. The findings of this study showed that the incorporation of MODIS–CCF with physiographic variables as covariates significantly improved the interpolation accuracy by 5–20%, with the largest improvement in modeling precipitation in March. The development of potent non-nucleoside inhibitors (NNIs) could be an alternate strategy to combating infectious bovine viral diarrhea virus (BVDV), other than the traditional vaccination. RNA-dependent RNA polymerase (RdRp) is an essential enzyme for viral replication; therefore, it is one of the primary targets for countermeasures against infectious diseases. The reported NNIs, belonging to the classes of quinolines (2h: imidazo[4,5-g]quinolines and 5m: pyrido[2,3-g] quinoxalines), displayed activity in cell-based and enzyme-based assays. Nevertheless, the RdRp binding site and microscopic mechanistic action are still elusive, and can be explored at a molecular level. Here, we employed a varied computational arsenal, including conventional and accelerated methods, to identify quinoline compounds' most likely binding sites. Our study revealed A392 and I261 as the mutations that can render RdRp resistant against quinoline compounds. In particular, for ligand 2h, mutation of A392E is the most probable mutation. The loop L1 and linker of the fingertip is recognized as a pivotal structural determinant for the stability and escape of quinoline compounds. Overall, this work demonstrates that the quinoline inhibitors bind at the template entrance channel, which is governed by conformational dynamics of interactions with loops and linker residues, and reveals structural and mechanistic insights into inhibition phenomena, for the discovery of improved antivirals. Constructing visible-light-active Z-scheme heterojunctions has proven fruitful in enhancing the photocatalytic activity of photocatalysts for superior water clean-up. Herein, we report the fabrication of a CoFe2O4@Bi2O3/NiO (CBN) Z-scheme nanoheterojunction. The obtained CBN heterojunction was used for visible-light-assisted degradation of ofloxacin (OFL) in water. The OFL degradation efficiency achieved by the CBN heterojunction was 95.2% in 90 min with a rate constant of kapp = 0.03316 min−1, which was about eight times that of NiO and thirty times that of CoFe2O4. The photocatalytic activity of a Bi2O3/NiO Z-scheme heterojunction was greatly enhanced by the visible activity and redox mediator effect of the cobalt ferrite co-catalyst. Higher charge-carrier separation, more visible-light capture, and the Z-scheme mechanism in the Z-scheme system were the important reasons for the high performance of CBN. The scavenging experiments suggested ●O2− as an active species for superior OFL degradation. The possible OFL degradation pathway was predicted based on LC-MS findings of degradation intermediate products. The magnetic nature of the CBN helped in the recovery of the catalyst after reuse for six cycles. This work provides new insights into designing oxide-based heterojunctions with high visible-light activity, magnetic character, and high redox capabilities for potential practical applications in environmental treatment. In the present review, using an integrated approach based on the experimental and theoretical study of the processes of thermal decomposition and combustion of practically important polymers, such as polymethyl methacrylate, polyethylene, and glass-fiber-reinforced epoxy resin, the features of the mechanism for reducing the combustibility of these materials with phosphorus-containing flame-retardants (FR), as well as graphene, are identified. A set of original experimental methods was developed and applied that make it possible to study the kinetics of thermal decomposition and the thermal and chemical structure of the flames of the studied materials, including those with FR additives, as well as to measure the flame propagation velocity, the mass burning rate, and the heat fluxes from the flame on the surface of a material. Numerical models were developed and tested to describe the key parameters of the flames of the studied polymeric materials. An analysis of the experimental and numerical simulation data presented showed that the main effect of phosphorus-containing fire-retardants on reducing the combustibility of these materials is associated with the inhibition of combustion processes in the gas phase, and the effect of adding graphene manifests itself in both gas and condensed phases. Climate change is perceived to be the primary reason for the amplification of extreme climatic phenomena. Estimation of changes in extreme values under climate change thus plays an important role in disaster risk assessment and management. However, the different changes in extremes in two distinct regions: inland and coast under climate change are yet to be investigated meticulously. This study is intended to assess the changes in frequency of rainfall and temperature extremes under the impact of climate change in two distinct locations: coast and inland of Bengal delta, a region highly vulnerable to climate change. The multi-model ensemble (projections from CMIP6 framework) technique with the application of frequency analysis was employed to appraise the impact in two future time horizons. Results suggest that the inland estimate of extreme rainfall by the end of this century is barely able to exceed the coastal estimate of extreme rainfall in present conditions. The rate of increase of warm extremes is almost similar; however, with the cold extreme, the increase rate is a little higher inland than on the coast. In both regions, a greater rise in climate extremes is expected in the far future than in the near future. Skin aging is a progressive biological process of the human body, and it is not only time-dependent. Differently substituted 3-phenylcoumarins proved to efficiently inhibit tyrosinase. In the current work, new substitution patterns have been explored, and the biological studies were extended to other important enzymes involved in the processes of skin aging, as elastase, collagenase and hyaluronidase. From the studied series, five compounds presented inhibitory activity against tyrosinase, one compound against elastase, eight compounds against collagenase and two compounds against hyaluronidase, being five compounds dual inhibitors. The 3-(4′-Bromophenyl)-5,7-dihydroxycoumarin (1) and 3-(3′-bromophenyl)-5,7-dihydroxycoumarin (2) presented the best profiles against tyrosinase (IC50 = 1.05 µM and 7.03 µM) and collagenase (IC50 = 123.4 µM and 110.4 µM); the 3-(4′-bromophenyl)-6,7-dihydroxycoumarin (4) presented a good inhibition against tyrosinase and hyaluronidase; the 3-(3′-bromophenyl)-6,7-dihydroxycoumarin (5) showed an effective tyrosinase and elastase inhibition; and 6,7-dihydroxy-3-(3′-hydroxyphenyl)coumarin (11) presented a dual profile inhibition against collagenase and hyaluronidase. Furthermore, considering the overall activities tested, compounds 1 and 2 proved to be the most promising anti-aging compounds. These compounds also showed to have a photo-protective effect, without being cytotoxic to human skin keratinocyte cells. Water pollution has been a prevalent issue globally for some time. Some pollutants are released into the water system without treatment, making the water not suitable for consumption. This problem may lead to more grave problems in the future including the destruction of the ecosystem along with the organisms inhabiting it, and illness and diseases endangering human health. Conventional methods have been implemented to remove hazardous pollutants such as dyes, heavy metals, and oil but are incapable of doing so due to economic restraints and the inability to degrade the pollutants, leading to secondary pollution. Photocatalysis is a more recently applied concept and is proven to be able to completely remove and degrade pollutants into simpler organic compounds. Titanium dioxide (TiO2) is a fine example of a photocatalyst owing to its cost-effectiveness and superb efficiency. However, issues such as the high recombination rate of photogenerated electrons along with positive holes while being only limited to UV irradiation need to be addressed. Carbonaceous materials such as graphene oxide (GO) can overcome such issues by reducing the recombination rate and providing a platform for adsorption accompanied by photocatalytic degradation of TiO2. The history and development of the synthesis of GO will be discussed, followed by the methods used for GO/TiO2 synthesis. The hybrid of GO/TiO2 as a photocatalyst has received some attention in the application of wastewater treatment due to its efficiency and it being environmentally benign. This review paper thereby aims to identify the origins of different pollutants followed by the sickness they may potentially inflict. Recent findings, including that GO/TiO2-related nanocomposites can remove pollutants from the water system, and on the photodegradation mechanism for pollutants including aromatic dyes, heavy metal and crude oil, will be briefly discussed in this review. Moreover, several crucial factors that affect the performance of photocatalysis in pollutant removal will be discussed as well. Therefore, this paper presents a critical review of recent achievements in the use of GO/TiO2-related nanocomposites and photocatalysis for removing various pollutants in wastewater treatment. The study evaluates the impacts of India's COVID-19 lockdown and unlocking periods on the country's ambient air quality. India experienced three strictly enforced lockdowns followed by unlocking periods where economic and social restrictions were gradually lifted. We have examined the in situ and satellite data of NO2 emissions for several Indian cities to assess the impacts of the lockdowns in India. Additionally, we analyzed NO2 data acquired from the Sentinel-5P TROPOMI sensor over a few districts of the Punjab state, as well as the National Capital Region. The comparisons between the in situ and satellite NO2 emissions were performed for the years 2019, 2020 and up to July 2021. Further analysis was conducted on the satellite data to map the NO2 emissions over India during March to July for the years of 2019, 2020 and 2021. Based on the in situ and satellite observations, we observed that the NO2 emissions significantly decreased by 45–55% in the first wave and 30% in the second wave, especially over the Northern Indian cities during the lockdown periods. The improved air quality over India is indicative of reduced pollution in the atmosphere due to the lockdown process, which slowed down the industrial and commercial activities, including the migration of humans from one place to another. Overall, the present study contributes to the understanding of the trends of the ambient air quality over large geographical areas using the Sentinel-5P satellite data and provides valuable information for regulatory bodies to design a better decision support system to improve air quality. The determination of crop water requirements and crop coefficient (Kc) of agricultural crops helps to create an appropriate irrigation schedule for the effective management of irrigation water. A portable smart weighing lysimeter (1000 × 1000 mm and 600 mm depth) was developed at CPCT, IARI, New Delhi for real-time measurement of Crop Coefficient (Kc) and water requirement of chrysanthemum crop and bulk data storage. The paper discusses the assembly, structural and operational design of the portable smart weighting lysimeter. The performance characteristics of the developed lysimeter were evaluated under different load conditions. The Kc values of the chrysanthemum crop obtained from the lysimeter installed inside the greenhouse were Kc ini. 0.43 and 0.38, Kc mid-1.27 and 1.25, and Kc end-0.67 and 0.59 for the years 2019–2020 and 2020–2021, respectively, which apprehensively corroborated with the FAO 56 paper for determination of crop coefficient. The Kc values decreased progressively at the late-season stage because of the maturity and aging of the leaves. The lysimeter's edge temperature was somewhat higher, whereas the center temperature closely matched the field temperature. The temperature difference between the center and the edge increased as the ambient temperature rose. The developed smart lysimeter system has unique applications due to its real-time measurement, portable attribute, and ability to produce accurate results for determining crop water use and crop coefficient for greenhouse chrysanthemum crops. In the present work, we use a merger of computational and biochemical techniques as a rational guideline for structural modification of benzofuran derivatives to find pertinent structural features for the butyrylcholinesterase inhibitory activity and selectivity. Previously, we revealed a series of 2-phenylbenzofuran compounds that displayed a selective inhibitory activity for BChE. Here, in an effort to discover novel selective BChE inhibitors with favorable physicochemical and pharmacokinetic profiles, 2-benzylbenzofurans were designed, synthesized, and evaluated as BChE inhibitors. The 2-phenylbenzofuran scaffold structure is modified by introducing one methylene spacer between the benzofuran core and the 2-phenyl ring with a hydroxyl substituent in the para or meta position. Either position 5 or 7 of the benzofuran scaffold was substituted with a bromine or chlorine atom. Further assessment of the selected list of compounds indicated that the substituent's nature and position determined their activity and selectivity. 5-bromo-2-(4-hydroxybenzyl)benzofuran 9B proved to be the most potent butyrylcholinesterase inhibitor (IC50 = 2.93 µM) of the studied series. Abating the approaching yield plateau in rice requires taking advantage of potential technologies that requires knowledge on genetic diversity. Hybrid breeding, particularly in indica rice, requires the recruitment of large genetic variability from outside because the available genetic diversity of the cultivated pool has already been utilized to a great extent. In this study, we examined an assembly of 200 tropical japonica lines collected worldwide for population genetic structure and variability in yield-associated traits. Tested along with 30 indica and six wild rice lines belonging to India, the tropical japonica lines indicated great phenotypic variability, particularly related to new plant type (NPT) phenology, and formed six clusters. Furthermore, a marker-based characterization using a universal diversity marker panel classified the genotype assembly into four clusters, of which three encompassed tropical japonica lines, while the last cluster included mostly indica lines. The population structure of the panel also revealed a similar pattern, with tropical japonica lines forming three subpopulations. Remarkable variation in the allelic distribution was observed between the subpopulations. Superimposing the geographical sources of the genotypes over the population structure did not reveal any pattern. The genotypes sourced closer to the center of origin of rice showed relatively little diversity compared with the ones obtained from other parts of the world, suggesting migration from a common region of origin. The present work aims to understand the crystallographic basis of the mechanical behavior of rivaroxaban-malonic acid cocrystal (RIV-MAL Co) in comparison to its parent constituents, i.e., rivaroxaban (RIV) and malonic acid (MAL). The mechanical behavior was evaluated at the bulk level by performing "out of die" bulk compaction and at the particle level by nanoindentation. The tabletability order for the three solids was MAL < RIV < RIV-MAL Co. MAL demonstrated "lower" tabletability because of its lower plasticity, despite it having reasonably good bonding strength (BS). The absence of a slip plane and "intermediate" BS contributed to this behavior. The "intermediate" tabletability of RIV was primarily attributed to the differential surface topologies of the slip planes. The presence of a primary slip plane (0 1 1) with flat-layered topology can favor the plastic deformation of RIV, whereas the corrugated topology of secondary slip planes (1 0 2) could adversely affect the plasticity. In addition, the higher elastic recovery of RIV crystal also contributed to its tabletability. The significantly "higher" tabletability of RIV-MAL Co among the three molecular solids was the result of its higher plasticity and BS. Flat-layered topology slip across the (0 0 1) plane, the higher degree of intermolecular interactions, and the larger separation between adjacent crystallographic layers contributed to improved mechanical behavior of RIV-MAL Co. Interestingly, a particle level deformation parameter H/E (i.e., ratio of mechanical hardness H to elastic modulus E) was found to inversely correlate with a bulk level deformation parameter σ0 (i.e., tensile strength at zero porosity). Human immunodeficiency virus type 1 (HIV-1) establishes latency in resting memory CD4+ T cells and cells of myeloid lineage. In contrast to the T cells, cells of myeloid lineage are resistant to the HIV-1 induced cytopathic effect. Cells of myeloid lineage including macrophages are present in anatomical sanctuaries making them a difficult drug target. In addition, the long life span of macrophages as compared to the CD4+ T cells make them important viral reservoirs in infected individuals especially in the late stage of viral infection where CD4+ T cells are largely depleted. In the past decade, HIV-1 persistence in resting CD4+ T cells has gained considerable attention. It is currently believed that rebound viremia following cessation of combination anti-retroviral therapy (cART) originates from this source. However, the clinical relevance of this reservoir has been questioned. It is suggested that the resting CD4+ T cells are only one source of residual viremia and other viral reservoirs such as tissue macrophages should be seriously considered. In the present review we will discuss how macrophages contribute to the development of long-lived latent reservoirs and how macrophages can be used as a therapeutic target in eradicating latent reservoir. Biopolymers are a leading class of functional material suitable for high-value applications and are of great interest to researchers and professionals across various disciplines. Interdisciplinary research is important to understand the basic and applied aspects of biopolymers to address several complex problems associated with good health and well-being. To reduce the environmental impact and dependence on fossil fuels, a lot of effort has gone into replacing synthetic polymers with biodegradable materials, especially those derived from natural resources. In this regard, many types of natural or biopolymers have been developed to meet the needs of ever-expanding applications. These biopolymers are currently used in food applications and are expanding their use in the pharmaceutical and medical industries due to their unique properties. Plants have evolved several adaptive strategies through physiological changes in response to herbivore attacks. Plant secondary metabolites (PSMs) are synthesized to provide defensive functions and regulate defense signaling pathways to safeguard plants against herbivores. Herbivore injury initiates complex reactions which ultimately lead to synthesis and accumulation of PSMs. The biosynthesis of these metabolites is regulated by the interplay of signaling molecules comprising phytohormones. Plant volatile metabolites are released upon herbivore attack and are capable of directly inducing or priming hormonal defense signaling pathways. Secondary metabolites enable plants to quickly detect herbivore attacks and respond in a timely way in a rapidly changing scenario of pest and environment. Several studies have suggested that the potential for adaptation and/or resistance by insect herbivores to secondary metabolites is limited. These metabolites cause direct toxicity to insect pests, stimulate antixenosis mechanisms in plants to insect herbivores, and, by recruiting herbivore natural enemies, indirectly protect the plants. Herbivores adapt to secondary metabolites by the up/down regulation of sensory genes, and sequestration or detoxification of toxic metabolites. PSMs modulate multi-trophic interactions involving host plants, herbivores, natural enemies and pollinators. Although the role of secondary metabolites in plant-pollinator interplay has been little explored, several reports suggest that both plants and pollinators are mutually benefited. Molecular insights into the regulatory proteins and genes involved in the biosynthesis of secondary metabolites will pave the way for the metabolic engineering of biosynthetic pathway intermediates for improving plant tolerance to herbivores. This review throws light on the role of PSMs in modulating multi-trophic interactions, contributing to the knowledge of plant-herbivore interactions to enable their management in an eco-friendly and sustainable manner. Influenza (IAV) neuraminidase (NA) is a glycoprotein required for the viral exit from the cell. NA requires disulfide bonds for proper function. We have recently demonstrated that protein disulfide isomerase (PDI)A3 is required for oxidative folding of IAV hemagglutinin (HA), and viral propagation. However, it not known whether PDIs are required for NA maturation or if these interactions represent a putative target for the treatment of influenza infection. We sought to determine whether PDIA3 is required for disulfide bonds of NA, its activity, and propagation of the virus. Requirement of disulfides for NA oligomerization and activity were determined using biotin switch and redox assays in WT and PDIA3−/− in A549 cells. A PDI specific inhibitor (LOC14) was utilized to determine the requirement of PDIs in NA activity, IAV burden, and inflammatory response in A549 and primary mouse tracheal epithelial cells. Mice were treated with the inhibitor LOC14 and subsequently examined for IAV burden, NA activity, cytokine, and immune response. IAV-NA interacts with PDIA3 and this interaction is required for NA activity. PDIA3 ablation or inhibition decreased NA activity, viral burden, and inflammatory response in lung epithelial cells. LOC14 treatment significantly attenuated the influenza-induced inflammatory response in mice including the overall viral burden. These results provide evidence for PDIA3 inhibition suppressing NA activity, potentially providing a novel platform for host-targeted antiviral therapies. [ABSTRACT FROM AUTHOR]
