Background and Objectives: The ineffective combination of corticosteroids and antibiotics in treating some atopic dermatitis (AD) cases has been concerning. The skin barrier defects in AD ease the colonization of Staphylococcus aureus (S. aureus), which results in a rise in interleukin-31 (IL-31). Lumbricus rubellus (L. rubellus) has shown antimicrobial and antiallergic effects but has not been studied yet to decrease the growth of S. aureus and IL-31 levels in AD patients. This study aimed to analyze the effect of L. rubellus extract in reducing S. aureus colonization, the IL-31 level, and the severity of AD. Materials and Methods: A randomized controlled trial (RCT) (international registration number TCTR20231025004) was conducted on 40 AD patients attending Dermatology and Venereology Polyclinic, Mother and Child Hospital (RSIA), Aceh, Indonesia, from October 2021 to March 2022. AD patients aged 8–16 who had a Scoring Atopic Dermatitis (SCORAD) index > 25, with total IgE serum level > 100 IU/mL, and had healthy weight were randomly assigned into two groups: one received fluocinolone acetonide 0.025% and placebo (control group) and one received fluocinolone acetonide 0.025% combined with L. rubellus extract (Vermint®) (intervention group). The S. aureus colony was identified using a catalase test, coagulase test, and MSA media. The serum IL-31 levels were measured using ELISA assay, while the SCORAD index was used to assess the severity of and improvement in AD. Mean scores for measured variables were compared between the two groups using an unpaired t-test and Mann–Whitney U test. Results: A significant decline in S. aureus colonization (p = 0.001) and IL-31 (p = 0.013) in patients receiving L. rubellus extract was found in this study. Moreover, fourteen AD patients in the intervention group showed an improvement in the SCORAD index of more than 35% (p = 0.057). Conclusions: L. rubellus extract significantly decreases S. aureus colonization and the IL-31 level in AD patients, suggesting its potential as an adjuvant therapy for children with AD. Earthworm invasions often reduce biodiversity and affect the ability of ecosystems to perform ecosystem functions. Over the past few decades, European lumbricid species have spread widely in natural habitats in Western Siberia, without completely displacing the native species. The aim of the present experiment is to study the survival, reproductive potential, vertical distribution, abundance, and biomass of Aporrectodea caliginosa and Lumbricus rubellus invasive species for the region and the native species Eisenia nordenskioldi and their influence on each other. For this purpose, winter and vegetation experiments were conducted in mesocosms simulating the real situation of invasions in the same communities on the same types of soils with the same amount and composition of litter. The authors found that the native species was significantly inferior to A. caliginosa in reproduction rate, number of offspring, and final biomass, but had an advantage over L. rubellus. The native species was positively influenced by the presence of A. caliginosa. In the experiment, there was a significant rejuvenation of the populations of the studied species, as well as an increase in abundance and relative mass under interaction conditions compared to mono variants. The native species had advantages over the invasive species in terms of winter survival. The natural volume of litter used in the experiment was insufficient for L. rubellus. According to the results of the experiment, it can be assumed that the spread of L. rubellus will continue to be limited to non-freezing biotopes rich in organic matter. Metallothioneins (MTs) are multifunctional proteins occurring throughout the animal kingdom. While the expression and transcriptional regulation of MTs is well-studied in vertebrates, the mechanism of MT activation is still unknown for most invertebrates. Therefore, we examined wMT-2 gene regulation and expression patterns in Lumbricus rubellus and L. terrestris. Transcription levels, the occupation of DNA binding sites, the expression of putative transcriptional regulators, and promotor DNA methylation were determined. We found that wMT-2 expression does not follow a circadian pattern. However, Cd-induced wMT-2 induction was observed, and was, interestingly, suppressed by physical injury. Moreover, the promotor region that is responsible for the wMT-2 gene regulation was elucidated. ATF, a putative transcriptional regulator, showed increased phosphorylation upon Cd exposure, suggesting that it plays a major role in wMT-2 gene activation. The promotor methylation of wMT-2, on the other hand, is probably not involved in transcriptional regulation. Elucidating the regulatory mechanism of the earthworm MT gene activation might provide insights into the molecular coordination of the environmental stress response in invertebrates, and might also reveal a link to wound repair and, in a broader sense, to immunity. Earthworms express, as most animals, metallothioneins (MTs)—small, cysteine-rich proteins that bind d10 metal ions (Zn(II), Cd(II), or Cu(I)) in clusters. Three MT homologues are known for Lumbricus rubellus, the common red earthworm, one of which, wMT-2, is strongly induced by exposure of worms to cadmium. This study concerns composition, metal binding affinity and metal-dependent protein folding of wMT-2 expressed recombinantly and purified in the presence of Cd(II) and Zn(II). Crucially, whilst a single Cd7wMT-2 species was isolated from wMT-2-expressing E. coli cultures supplemented with Cd(II), expressions in the presence of Zn(II) yielded mixtures. The average affinities of wMT-2 determined for either Cd(II) or Zn(II) are both within normal ranges for MTs; hence, differential behaviour cannot be explained on the basis of overall affinity. Therefore, the protein folding properties of Cd- and Zn-wMT-2 were compared by 1H NMR spectroscopy. This comparison revealed that the protein fold is better defined in the presence of cadmium than in the presence of zinc. These differences in folding and dynamics may be at the root of the differential behaviour of the cadmium- and zinc-bound protein in vitro, and may ultimately also help in distinguishing zinc and cadmium in the earthworm in vivo. Exotic earthworms are drivers of biotic communities in invaded North American forest stands. Here we used ecologically important oribatid mite (Arachnida: Acari) communities, as model organisms to study the responses of litter- and soil-dwelling microarthropod communities to exotic earthworm invasion in a northern temperate forest. Litter- and soil-dwelling mites were sampled in 2008-2009 from forest areas: (1) with no earthworms; (2) those with epigeic and endogeic species, including Lumbricus rubellus Hoffmeister; and (3) those with epigeic, endogeic, and anecic earthworms including L. terrestris L. Species richness and diversity of litter- and soil-dwelling (0-2 cm soil depth) oribatid mites was 1-2 times higher in sites without earthworms than in sites with worms. Similarly, litter-dwelling oribatid mites were between 72 and 1,210 times more abundant in earthworm-free sites than in sites with worms. Among earthworm invaded sites, abundance of litter-dwelling oribatid mites in sites without the anecic L. terrestris was twice as high in May and 28 times higher in October, compared to sites with L. terrestris. Species richness, diversity, and abundance of oribatid mites were greater in litter-layers than in the soil-layers that showed a varied response to earthworm invasion. Species compositions of both litter- and soil-dwelling oribatid mite communities of forests with no earthworms were markedly different from those with earthworms. We conclude that exotic earthworm invasions are associated with significant declines of species diversity, numbers, and compositional shifts in litter- and soil-inhabiting communities. These faunal shifts may contribute to earthworm effects on soil processes and food web dynamics in historically earthworm-free, northern temperate forests. [ABSTRACT FROM AUTHOR]
