Brain tumor segmentation using Magnetic Resonance Imaging (MRI) is vital for clinical decision making. Traditional deep learning-based studies using convolutional neural networks have predominantly processed MRI data as two-dimensional slices, leading to the loss of contextual information. While three-dimensional (3D) convolutional layers represent an advancement, they have not fully exploited pathological information according to the three-axis nature of 3D MRI data—axial, coronal, and sagittal. Recognizing these limitations, we introduce a Tri-Axis based Context-Aware Reverse Network (TACA-RNet). This innovative approach leverages the unique 3D spatial orientations of MRI, learning crucial information on brain anatomy and pathology. We incorporated three specialized modules: a Tri-Axis Channel Reduction module for optimizing feature dimensions, a MultiScale Contextual Fusion module for aggregating multi-scale features and enhancing spatial discernment, and a 3D Axis Reverse Attention module for the precise delineation of tumor boundaries. The TACA-RNet leverages three specialized modules to enhance the understanding of tumor characteristics and spatial relationships within MRI data by fully utilizing its tri-axial structure. Validated on the Brain Tumor Segmentation Challenge 2018 and 2020 datasets, the TACA-RNet demonstrated superior performances over contemporary methodologies. Pralidoxime chloride, a highly hydrophilic antidote, cannot be effectively separated by reverse-phase high-performance liquid chromatography (RP-HPLC), unless the mobile-phase composition is varied. However, the use of ion-pairing reagents for pralidoxime separation is hindered by the persistent contamination of the stationary phase or chromatography system inside the HPLC system. Thus, this study aimed to develop a simple, rapid, and robust method based on RP-HPLC to determine pralidoxime chloride in antidote autoinjectors using a chaotropic salt as the mobile-phase additive. The use of UV detection at 270 nm allowed for the simultaneous detection of pralidoxime chloride and the internal standard, pyridine-2-aldoxime. The addition of chaotropic salts (NaPF6, NaBF4, and NaClO4) and an ionic liquid ([EMIM]PF6) increased the retention time of pralidoxime chloride. Among them, NaPF6 exhibited the highest capacity factor in the reverse-phase C18 column. Increasing the salt concentration increased the capacity factor and the number of theoretical plates. Analytical method validation was performed to assess the linearity, accuracy, precision, recovery, and repeatability, according to the Ministry of Food and Drug Safety guidelines. Additionally, this newly developed method exhibits an adequate separation capability, making it a potential substitute for the current method employed in the United States/Korean Pharmacopoeia, and it ensures the necessary durability to maintain the robustness and reliability of the analytical system. Acquired chemoresistance of tumor cells is an unwanted consequence of cancer treatment. Overcoming chemoresistance is particularly important for efficiently improving cancer therapies. Here, using multiple lines of evidence, we report the suppressive role of SERTAD1 in apoptosis/anoikis. Among various breast cancer cell lines, higher SERTAD1 expression was found in MCF7 and MDA-MB-231 in suspension than in adherent cell culture. We revealed an unexpected phenomenon that different types of cell deaths were induced in response to different doses of doxorubicin (Dox) in breast cancer cells, presumably via lysosomal membrane permeabilization. A low dose of Dox highly activated autophagy, while a high dose of the chemotherapy induced apoptosis. Inhibition of SERTAD1 promoted the sensitivity of breast cancer cells to Dox and paclitaxel, leading to a significant reduction in tumor volumes of xenograft mice. Simultaneously targeting cancer cells with Dox and autophagy inhibition successfully induced higher apoptosis/anoikis. The novel role of SERTAD1 in maintaining cellular homeostasis has also been suggested in which lysosomal contents, including LAMP1, LAMP2, CTSB, and CTSD, were reduced in SERTAD1-deficient cells. Anti-neutrophil cytoplasmic antibody (ANCA)-associated vasculitis is an autoimmune disorder which affects small- and, to a lesser degree, medium-sized vessels. ANCA-associated vasculitis encompasses three disease phenotypes: granulomatosis with polyangiitis (GPA), microscopic polyangiitis (MPA), and eosinophilic granulomatosis with polyangiitis (EGPA). This classification is largely based on clinical presentations and has several limitations. Recent research provided evidence that genetic background, risk of relapse, prognosis, and co-morbidities are more closely related to the ANCA serotype, proteinase 3 (PR3)-ANCA and myeloperoxidase (MPO)-ANCA, compared to the disease phenotypes GPA or MPA. This finding has been extended to the investigation of biomarkers predicting disease activity, which again more closely relate to the ANCA serotype. Discoveries related to the immunopathogenesis translated into clinical practice as targeted therapies are on the rise. This review will summarize the current understanding of the immunopathogenesis of ANCA-associated vasculitis and the interplay between ANCA serotype and proposed disease biomarkers and illustrate how the extending knowledge of the immunopathogenesis will likely translate into development of a personalized medicine approach in the management of ANCA-associated vasculitis. Rhizobia produces different types of surface polysaccharides. Among them, cyclic β-(1→2)-d-glucan is located in the periplasmic space of rhizobia and plays an important role in the adaptation of bacteria to osmotic adaptation. Cyclic β-(1→2)-d-glucan (CG), synthesized from Sinorhiozbbium meliloti 1021, has a neutral and anionic form. In the present study, we characterized the exact chemical structures of anionic CG after purification using size exclusion s (Bio-Gel P-6 and P-2) chromatography, and DEAE-Sephadex anion exchange chromatography. The exact structure of each isolated anionic CG was characterized using various analytical methods such as nuclear magnetic resonance (NMR), attenuated total reflection Fourier transform infrared (ATR-FTIR) spectroscopy and matrix associated laser desorption ionization-time of Flight (MALDI-TOF) mass spectrometry. The precise chemical structures of novel anionic CG molecules were elucidated by various NMR spectroscopic analyses, including 1H, 13C, 31P, and 2D HSQC NMR spectroscopy. As a result, we discovered that anionic CG molecules have either glycerophosphoryl or succinyl residues at C6 positions of a neutral CG. In addition, the results of MALDI-TOF mass spectrometric analysis confirmed that there are two types of patterns for anionic CG peaks, where one type of peak was the succinylated CG (SCG) and the other was glycerophospholated CG (GCG). In addition, it was revealed that each anionic CG has one to four substituents of the succinyl group of SCG and glycerophosphoryl group of GCG, respectively. Novel materials have been recently developed for coping with various environmental factors. Generally, to improve the thermal comfort to humans in cold environments, securing an air layer is important. Therefore, this study analyzed the thermal properties of 3D spacer technical materials, 3D printed using thermoplastic polyurethane, according to the structural changes. Four 3D spacer technical material structures were designed with varying pore size and thickness. These samples were moved into a cold climate chamber (temperature 5 ± 1 °C, relative humidity (60 ± 5)%, wind velocity ≤0.2 m/s) and placed on a heating plate set to 30 °C. The surface and internal temperatures were measured after 0, 10, 20, and 30 min and then 10 min after turning off the heating plate. When heat was continuously supplied, the 3D spacer technical material with large pores and a thick air layer showed superior insulation among the materials. However, when no heat was supplied, the air gap thickness dominantly affected thermal insulation, regardless of the pore size. Hence, increasing the air gap is more beneficial than increasing the pore size. Notably, we found that the air gap can increase insulation efficiency, which is of importance to the new concept of 3D printing an interlining. [ABSTRACT FROM AUTHOR]
