We aimed to identify the mechanism underlying the preventive effects of non-alcoholic fatty liver disease (NAFLD) through Platycodi Radix consumption using liver proteomic and bioinformatic analysis. C57BL/6J mice were categorized into three groups: those receiving a standard chow diet (NCD), those on a high-fat diet (HFD), and those on an HFD supplemented with 5% Platycodi Radix extract (PRE). After a 12-week period, PRE-fed mice exhibited a noteworthy prevention of hepatic steatosis. Protein identification and quantification in liver samples were conducted using LC-MS/MS. The identified proteins were analyzed through Ingenuity Pathway Analysis software, revealing a decrease in proteins associated with FXR/RXR activation and a concurrent increase in cholesterol biosynthesis proteins in the PRE-treated mouse liver. Subsequent network analysis predicted enhanced bile acid synthesis from these proteins. Indeed, the quantity of bile acids, which was reduced in HFD conditions, increased in the PRE group, accompanied by an elevation in the expression of synthesis-related proteins. Our findings suggest that the beneficial effects of PRE in preventing hepatic steatosis may be mediated, at least in part, through the modulation of FXR/RXR activation, cholesterol biosynthesis, and bile acid synthesis pathways. CLPTM1L (Cleft Lip and Palate Transmembrane Protein 1-Like) has previously been implicated in tumorigenesis and drug resistance in cancer. However, the genetic link between CLPTM1L and bladder cancer remains uncertain. In this study, we investigated the genetic association of variable number of tandem repeats (VNTR; minisatellites, MS) regions within CLPTM1L with bladder cancer. We identified four CLPTM1L-MS regions (MS1~MS4) located in intron regions. To evaluate the VNTR polymorphic alleles, we analyzed 441 cancer-free controls and 181 bladder cancer patients. Our analysis revealed a higher frequency of specific repeat sizes within the MS2 region in bladder cancer cases compared to controls. Notably, 25 and 27 repeats were exclusively present in the bladder cancer group. Moreover, rare alleles within the medium-length repeat range (25–29 repeats) were associated with an elevated bladder cancer risk (odds ratio [OR] = 5.78, 95% confidence interval [CI]: 1.49–22.47, p = 0.004). We confirmed that all MS regions followed Mendelian inheritance, and demonstrated that MS2 alleles increased CLPTM1L promoter activity in the UM-UC3 bladder cancer cells through a luciferase assay. Our findings propose the utility of CLPTM1L-MS regions as DNA typing markers, particularly highlighting the potential of middle-length rare alleles within CLPTM1L-MS2 as predictive markers for bladder cancer risk. In this study, the oxidation characteristics of elemental mercury were assessed based on the input gas environment, temperature, and particle size distribution of the steel slag. Experiments were performed at room temperature, 100 °C and 200 °C, under air and simulated gas environments. The oxidation reaction of elemental mercury was conducted using steel slag samples of 1 mm, 2.36 mm, and 4.75 mm at various conditions. From the basic characteristic analysis of the steel slag, it was found that the steel slag exhibits a similar composition to that of fly ash, and it can be utilized as an oxidizing agent. Results show that regardless of the temperature and the particle size distribution of steel slag, the oxidation reaction of elemental mercury rarely occurred in the air environment. However, in the case of the HCl gas environment, it was observed that the smaller the steel slag particle size, the stronger the oxidation reaction. It is believed that the oxidation efficiency of the steel slag increased as the contact area between the gas and particles increased. The oxidation reactivity was nearly two times higher in the temperature range of 100 °C to 200 °C than it was at room temperature. In this study, we investigated the applicability of an automobile shredder residue (ASR) as an energy and recycling resource. First, ASR gasification was conducted in a fixed-bed reactor (throughput = 1 kg/h) at different temperatures (800, 1000, and 1200 °C) and an equivalence ratio of 0.1–0.5. Clay bricks were prepared with the solid residue obtained from the gasification process to address the issue of solid-residue production in pyrolysis. The syngas (H2 + CO) from ASR gasification had maximum and minimum yields of approximately 86 and 40 vol.%, respectively. Furthermore, the yield of syngas increased with the temperature and equivalence ratio (ER); therefore, the optimum conditions for the ASR gasification were determined to be a temperature of 1200 °C and an ER of 0.5. In addition, solid residues, such as char and ash, began to melt due to thermal heating in the range of 1300–1400 °C and were converted into melting slag, which was subsequently used to manufacture clay bricks. The absorption ratios and compressive strengths of the clay bricks were compared to those set by Korean Industrial Standards to evaluate the quality of the clay bricks. As a result, the manufactured clay bricks were estimated to have a compressive strength of over 22.54 N/mm2 and an absorption ratio of less than 10%. Additionally, greenhouse gas (GHG) emissions from the melting–gasification process were estimated and compared with those from ASR incineration, calculated using the greenhouse gas equations provided by the Korean Ministry of Environment. As a result, it was revealed that the GHG emissions from the combined melting–gasification process (gasification, melting system, and clay-brick manufacturing) were approximately ten times higher than those from the ASR-incineration process. Thus, in terms of operation cost on the carbon capture process for GHG reduction, the combined melting–gasification process would be a more efficient process than that of incineration. As laser refractive surgeries (LRS) have been widely performed to correct myopia, ophthalmologists easily encounter patients with glaucoma who have a history of LRS. It is well known that intraocular pressure (IOP) in eyes with glaucoma is not accurate when measured using Goldmann applanation tonometry. However, risk factors for glaucoma progression, particularly those associated with measured IOP, have rarely been studied. We analysed data for 40 patients with a history of LRS and 50 age-matched patients without a history of LRS. Structural progression was defined as significant changes in thickness in the peripapillary retinal nerve fibre layer as identified using optical coherence tomography event-based guided progression analysis. Risk factors were determined via Cox regression analysis. Disc haemorrhage (DH) was associated with glaucoma progression in both the non-LRS group and LRS group (hazard ratio (HR): 4.650, p = 0.012 and HR: 8.666, p = 0.019, respectively). However, IOP fluctuation was associated with glaucoma progression only in the LRS group (HR: 1.452, p = 0.023). Our results show that DH was a significant sign of progression in myopic glaucoma eyes. When treating patients with myopia and glaucoma, IOP fluctuation should be monitored more carefully, even if IOP seems to be well controlled. [ABSTRACT FROM AUTHOR]

This study aimed to investigate the effect of hypoxic preconditioning (HPC) on primary retinal ganglion cell (RGC) survival and the associated mechanism, including the role of vascular endothelial growth factor (VEGF). Retinas were separated from the enucleated eyeballs of Sprague–Dawley rats on postnatal days 1–4. RGCs were harvested using an immunopanning-magnetic separation system and maintained for 24 h in a defined medium. Hypoxic damage (0.3% O2) was inflicted on the cells using a CO₂ chamber. Anti-VEGF antibody (bevacizumab) was administered to RGCs exposed to hypoxic conditions, and RGC survival rate was compared to that of non-anti-VEGF antibody-treated RGCs. HPC lasting 4 h significantly increased RGC survival rate. In the RGCs exposed to hypoxic conditions for 4 h, VEGF mRNA and protein levels were significantly increased. Treatment with high dose bevacizumab (>1 mg/mL) countered HPC-mediated RGC survival. Protein kinase B and focal adhesion kinase levels were significantly increased in 4-h hypoxia-treated RGCs. HPC showed beneficial effects on primary RGC survival. However, only specifically controlled exposure to hypoxic conditions rendered neuroprotective effects. Strong inhibition of VEGF inhibited HPC-mediated RGC survival. These results indicate that VEGF may play an essential role in promoting cell survival under hypoxic conditions. [ABSTRACT FROM AUTHOR]

Purpose: The pattern electroretinogram (PERG) is useful to detect retinal ganglion cell (RGC) damage in patients with glaucoma. Optical coherence tomography angiography (OCTA) measures retinal vessel density (VD), which is known to be reduced in glaucoma. There may be correlations between parameters of the PERG and OCTA in open-angle glaucoma (OAG). Methods: In total, 95 eyes of 95 OAG patients and 102 eyes of 102 normal controls were included in this study. N35, P50, and N95 latency along with P50 and N95 amplitude were obtained using the PERG. Retinal VD was measured around the peripapillary and macular area according to the ETDRS grid (concentric circles with diameters of 1, 3, and 6 mm), which is named a center (≤1 mm), an inner (1–3 mm), an outer (3–6 mm), and a full (≤6 mm) area. Pearson correlation analysis was done between parameters, and partial correlation analysis was done after adjusting confounding factors. Results: P50 amplitude, N95 amplitude, and VD of most measured areas were significantly lower in the OAG group compared to the normal group. N95 amplitude showed a statistically significant correlation with parameters of optical coherence tomography and visual field, peripapillary outer and full VD, and macular outer and full VD even after adjusting confounding factors. There was no significant correlation between parameters in the normal group. Conclusions: N95 amplitude was associated with structural and functional change including VD reduction in OAG. Gaining energy independence by utilizing new and renewable energy resources has become imperative for Korea. Energy recovery from Korean municipal solid waste (MSW) could be a promising option to resolve the issue, as Korean MSW is highly recyclable due to its systematic separation, collection and volume-based waste disposal system. In this study, gasification experiments were conducted on Korean municipal waste-derived solid fuel (SRF) using a fixed bed reactor by varying the equivalence ratio (ER) to assess the viability of syngas production. Experiments were also conducted on coal and biomass under similar conditions to compare the experimental results, as the gasification applicability of coal and biomass are long-established. Experimental results showed that Korean SRF could be used to recover energy in form of syngas. In particular, 50.94% cold gas efficiency and 54.66% carbon conversion ratio with a lower heating value of 12.57 MJ/Nm3 can be achieved by gasifying the SRF at 0.4 ER and 900 °C. However, compared to coal and biomass, the syngas efficiency of Korean SRF was less, which can be resolved by operating the gasification processes at high temperatures. If proper research and development activities are conducted on Korean SRF, it could be a good substitute for fossil fuels in the future. [ABSTRACT FROM AUTHOR]

Solid recovered fuel (SRF) residue, which is leftovers from the SRF manufacturing process, usually is discarded in landfill because of its low heating value and high ash and moisture content. However, it could be used as a fuel after mechanical and biological treatment. Gasification experiments were conducted on treated SRF residue (TSRFR) to assess the viability of syngas production. Efforts were also made to improve the gasification performance by adding low-cost natural minerals such as dolomite and lime as bed material, and by blending with biomass waste. In the case of additive mineral tests, dolomite showed better performance compared to lime, and in the case of biomass blends, a 25 wt% pine sawdust blend with TSRFR showed the best performance. Finally, as an appropriate condition, a combined experiment was conducted at an equivalence ratio (ER) of 0.2 using a 25 wt% pine sawdust blend with TSRFR as a feedstock and dolomite as the bed material. The highest dry gas yield (1.81 Nm3/kg), with the highest amount of syngas (56.72 vol%) and highest lower heating value (9.55 MJ/Nm3) was obtained in this condition. Furthermore, the highest cold gas efficiency (48.64%) and carbon conversion rate (98.87%), and the lowest residue yield (11.56%), tar (0.95 g/Nm3), and gas pollutants content was observed. Proper treatment and careful management of sewage sludge are essential because its disposal can lead to adverse environmental impacts such as public health hazards, as well as air, soil, and water pollution. Several efforts are being made currently not only to safely dispose of sewage sludge but also to utilize it as an energy source. Therefore, in this study, initiatives were taken to valorize sewage sludge cake by reducing the moisture content and increasing the calorific value by applying a hydrothermal treatment technique for efficient energy recovery. The sludge cake treated at 200 °C for 1 h was found to be the optimum condition for hydrothermal carbonization, as, in this condition, the caloric value of the treated sludge increased by 10% and the moisture content removed was 20 wt.%. To recover energy from the hydrothermally treated sludge, a gasification technology was applied at 900 °C. The results showed that the product gas from hydrothermally treated sludge cake had a higher lower heating value (0.98 MJ/Nm3) and higher cold gas efficiency (5.8%). Furthermore, compared with raw sludge cake, less tar was generated during the gasification of hydrothermally treated sludge cake. The removal efficiency was 28.2%. Thirteen types of fuel pellets were prepared from hydrothermally treated hospital solid waste, hydrothermally treated rice straw, pyrolytic plastic waste residue, rice straw, and Sakhalin fir residue using a flat die pellet machine. Different pellet properties such as pellet density, pellet durability, aspect ratio, physicochemical characteristics, and gross calorific value (GCV) were evaluated as well as compared concerning the European standard specification for residential/commercial densified fuels. In addition, the quality of pellets was compared with coal. The results showed that the pellets made only with hydrothermally treated hospital solid waste, hydrothermally treated rice straw, pyrolytic plastic waste residue, and rice straw failed to meet few individual criteria (<3 wt% ash content, <10 wt% moisture content, <0.03 wt% chlorine content, >96.5 wt% pellet durability, and >600 kg/m3 pellet density) of the European standard specifications. However, most of the mixed fuel pellets satisfied the requirement of pellet properties according to the European standard specification. In particular, up to 16.70 wt% hydrothermally treated rice straw, 1.50 wt% hydrothermally treated hospital solid waste, and 4.76 wt% of pyrolytic plastic waste residue can be blended with Shakhalin fir residue to produce low-chlorine fuel pellets. The gross calorific value of pellets made from the mixture of hydrothermally treated wastes and pyrolytic plastic waste residue (around 22 MJ/kg) showed similar results to that of coal. In the case of mixed pellets, the presence of these hydrothermally treated wastes and pyrolytic plastic waste residue valorized the fuel pellet quality. The main outcome of this study was the production of low chlorine biomass fuel pellets of high gross calorific values blended with hydrothermally treated wastes and pyrolytic waste residues, which opens a new door for utilizing waste in a better way, especially hospital solid waste. This study applied oxygen-enrichment conditions to remove tar (the main problem in biomass gasification) and increase gasification efficiency. Experiments on oxygen-enrichment conditions were conducted at oxygen concentrations of 21%, 25%, 30%, and 35% in oxidants. This was expected to increase the partial oxidation reaction in gasification reactions, thus leading to thermal decomposition of tar in producer gas. The decomposed tar was expected to be converted into syngas or combustible gases in the producer gas. The results were as follows: Tar-reduction efficiency was 72.46% at 30% oxygen enrichment compared to the standard 21% enrichment condition. In addition, the concentrations of syngas and combustible gases in the producer gas tended to increase. Therefore, the 30% oxygen-enrichment condition was optimal, resulting in 78.00% for cold gas efficiency and 80.24% for carbon conversion efficiency. The application of oxygen enrichment into the lab-scale gasification system clearly reduced the concentration of tar and tended to increase some indexes of gasification efficiency, thus suggesting the usefulness of this technique in large-scale biomass gasification operations.
