Simple Summary: In the present study, we aimed to explore the potential local regulatory role of the cocaine- and amphetamine-regulated transcript (CART) signaling pathway in granulosa cell (GC) apoptosis, which is a key mechanism promoting follicular atresia in several animal species, including buffalo. Our results showed how CART activity adversely influences buffalo GC viability by affecting estradiol production and enhancing apoptosis. The regulatory mechanism by which CART can affect GC apoptosis entails the modulation of the AKT/GSK3β/β-catenin pathway, a key intracellular signaling pathway essential for cell viability. In conclusion, this study provides valuable insights into the intricate mechanisms governing ovarian follicle development and granulosa cell function. Our findings revealed increased expression of CARTPT and BAX and decreased levels of AKT, β-catenin, and CYP19A1 genes in atretic follicles compared to healthy follicles. Subsequently, CART treatment in the presence of FSH inhibited the FSH-induced increase in GC viability by reducing estradiol production and increasing apoptosis. This change was accompanied by an increase in the gene expression levels of both CARTPT and BAX. At the protein level, treatment with CART in the presence of FSH negatively affected the activity of AKT, β-catenin, and LEF1, while the activity of GSK3β was enhanced. In conclusion, our study shows how CART negatively influences buffalo GC viability, underlying the modulation of the AKT/GSK3β/β-catenin pathway and promoting apoptosis—a key factor in follicular atresia. Toxoplasmosis is a significant global zoonosis with devastating impacts, and an effective vaccine against toxoplasmosis for humans has not yet been developed. In this study, we designed and formulated a novel DNA vaccine encoding the inhibitor of STAT1 transcriptional activity (IST) of T. gondii utilizing the eukaryotic expression vector pEGFP-N1 for the first time, with CL264 being a molecular adjuvant. Following intramuscular injection of the vaccine into mice, the levels of antibodies and cytokines were assessed to evaluate the immune response. Additionally, mice were challenged with highly virulent RH-strain tachyzoites of T. gondii, and their survival time was observed. The results show that the levels of IgG in serum, the ratio of IgG2a/IgG1 and the levels of IFN-γ in splenocytes of mice were significantly higher in the pEGFP-TgIST group and the pEGFP-TgIST + CL264 group than in the control group. In addition, the proportion of CD4+/CD8+ T cells was higher in mice immunized with either the pEGFP-TgIST group (p < 0.001) or the pEGFP-TgIST + CL264 group (p < 0.05) compared to the three control groups. Notably, TgIST-immunized mice exhibited prolonged survival times after T. gondii RH strain infection (p < 0.05). Our findings collectively demonstrate that the TgIST DNA vaccine elicits a significant humoral and cellular immune response and offers partial protection against acute T. gondii infection in the immunized mice, which suggests that TgIST holds potential as a candidate for further development as a DNA vaccine. Red rice, a variety of pigmented grain, serves dual purposes as both a food and medicinal resource. In recent years, we have witnessed an increasing interest in the dermatological benefits of fermented rice extracts, particularly their whitening and hydrating effects. However, data on the skincare advantages derived from fermenting red rice with Aspergillus oryzae remain sparse. This study utilized red rice as a substrate for fermentation by Aspergillus oryzae, producing a substance known as red rice Aspergillus oryzae fermentation (RRFA). We conducted a preliminary analysis of RRFA's composition followed by an evaluation of its skincare potential through various in vitro tests. Our objective was to develop a safe and highly effective skincare component for potential cosmetic applications. RRFA's constituents were assessed using high-performance liquid chromatography (HPLC), Kjeldahl nitrogen determination, the phenol-sulfuric acid method, and enzyme-linked immunosorbent assay (ELISA). We employed human dermal fibroblasts (FB) to assess RRFA's anti-aging and antioxidative properties, immortalized keratinocytes (HaCaT cells) and 3D epidermal models to examine its moisturizing and reparative capabilities, and human primary melanocytes (MCs) to study its effects on skin lightening. Our findings revealed that RRFA encompasses several bioactive compounds beneficial for skin health. RRFA can significantly promote the proliferation of FB cells. And it markedly enhances the mRNA expression of ECM-related anti-aging genes and reduces reactive oxygen species production. Furthermore, RRFA significantly boosts the expression of Aquaporin 3 (AQP3), Filaggrin (FLG), and Hyaluronan Synthase 1 (HAS1) mRNA, alongside elevating moisture levels in a 3D epidermal model. Increases were also observed in the mRNA expression of Claudin 1 (CLDN1), Involucrin (IVL), and Zonula Occludens-1 (ZO-1) in keratinocytes. Additionally, RRFA demonstrated an inhibitory effect on melanin synthesis. Collectively, RRFA contains diverse ingredients which are beneficial for skin health and showcases multifaceted skincare effects in terms of anti-aging, antioxidant, moisturizing, repairing, and whitening capabilities in vitro, highlighting its potential for future cosmetic applications. This study presents a comprehensive investigation into the mechanical properties of Basic Magnesium Sulfate Cement Concrete (BMSC) in comparison to Ordinary Portland Cement Concrete (OPC) within reinforced concrete components. The main objective is to evaluate BMSC's applicability for practical engineering purposes, with a focus on its with early high strength, improved toughness, and superior crack resistance compared to conventional concrete. Experimental procedures involved fabricating beam specimens using OPC concrete with a C40 strength grade, alongside BMSC beams with varying strength grades (C30, C40, and C50). These specimens underwent bending resistance tests to analyze crack patterns and mechanical characteristics. The findings reveal that BMSC beams demonstrate enhanced bending and tensile properties at equivalent strength grades compared to OPC beams. Particularly, cracking mainly occurred at the mid-span region of BMSC beams, characterized by narrower cracks, indicating superior crack resistance. However, it was noted that the toughness of BMSC beams decreases as the strength grade increases. The maximum mid-span deflection of the BMSC test beam was smaller than that of the OPC test beam, which was 3.8 mm and 2.6 mm, respectively. The maximum crack width of the OPC beam was 4.7 times that of the BMSC beam. To facilitate practical implementation, the study developed calculation models for estimating the crack bending distance and ultimate bending distance in BMSC beams, offering valuable tools for engineering design and optimization. This article studies the relationship between renewable energy stocks' performance and climate risk. It shows that publicly held renewable energy stocks underperform as a reaction to climate policy information releases, modeled by feed-in tariff (FIT) legislation announcements. The study examined stock price behaviors 2 days before and 30 days after FIT policy announcements. The stock sample used in the study has 3702 firm-day combinations, which included 180 cleantech firms and 32 events from 2007 to 2017. Based on the residual analysis of the sample's abnormal return, it indicated that the FIT announcements are associated with significant declines in returns. The cumulative abnormal return until Day 18 was a significant −0.83%, while the average abnormal return on the day was −0.16% at normal levels. The study partially excluded the likelihood of a transitory result by varying the measurement horizon. It also adopted both the market model and the Fama–French three-factor models to rule out model misspecification when estimating abnormal returns and thus increased the robustness. In fact, the results were stable to changes in estimating the model's specifications. In addition, the study compared the portfolio's performance with mimicking portfolios in terms of size, book-to-market equity (BE/ME), and the firms' geographic location. Autonomous underwater manipulation is very important for the robotic and intelligence operations of oceanic engineering. However, a small target often involves limited features and results in inaccurate visual matching. In order to improve visual measurement accuracy, this paper has proposed an improved unsharp masking algorithm to further enhance the weak texture region of blurred and low contrast images. Moreover, an improved ORB feature-matching method with adaptive threshold, non-maximum suppression and improved random sample consensus has also been proposed. To overcome unknown underwater disturbances and uncertain system parameters in the underwater robotic manipulations, an adaptive non-singular terminal sliding mode controller has been proposed with a quasi-barrier function to suppress the chattering problem and improve grasp accuracy for small target. Volatile organic compounds (VOCs) emitted by plant foliage play an important role in ozone (O3) and secondary organic aerosol (SOA) formation. Their emissions can be influenced by the leaf age. We explored the VOCs emissions and their effects on the formation of O3 and SOA from plant foliage in different ages. VOCs emissions from the young, mature, and senescent leaves of Ginkgo biloba, Ligustrum lucidum, and Forsythia suspensa were measured using the dynamic enclosure system and the TD–GC–MS technique. Based on the emission rates of quantified compounds, their potential to form O3 and SOA was estimated. Results showed that there were significant differences in the VOCs emission rate and their composition among leaves in different ages. The emission rate of the total VOCs by young leaves was the highest, while the lowest by senescent leaves. Monoterpenes were the dominant VOCs category, and isoprene emission had the lowest contribution for the leaves at each age. With increasing leaf age, the proportion of monoterpenes emission increased, and the proportion of sesquiterpenes decreased. The variations of isoprene and other VOCs were different. The potentials of total VOCs, isoprene, monoterpenes, sesquiterpenes, and other VOCs to form O3 (OFP) and SOA (SOAP) varied significantly among leaves at different ages. The total OFP and SOAP were the highest by young leaves, while the lowest by senescent leaves. With increasing leaf age, the contribution of monoterpenes to OFP and SOAP also increased, while that of sesquiterpenes decreased. Our study will provide support for the more accurate parameterization of the emission model and help to understand the VOCs emissions and study the precise prevention and control of complex air pollution at different times. While disclosing financial information has been widely proved to reduce the financing cost of a company, the impact of non-financial information, such as sustainability information, disclosing on the financing cost of the company is still in debate. The goal of this paper is to explore the impact of disclosing sustainability-related information on the cost of equity for firms. The paper first introduces the concept of sustainability information disclosure, and then exhibits its benefit through exploring its impact on reducing a firm's financing cost. It uses the Gartner supply chain top 50 rankings to construct the experiment environment to test for the effect of sustainability information disclosure on the cost of equity capital. The study uses the Gartner top 50 supply chain rankings from 2013 to 2017 to construct the experiment environment, and test for the sustainability information disclosure's impact on reducing the cost of equity capital. The regressions, which are based on the 350 firm-year sample of the United States and the 604 global firm-year sample, indicate that sustainability information disclosure significantly reduced the cost of equity capital. This paper uses a fixed effect regression method to analyze the impact of sustainability information disclosure. According to the regression result, the sustainability information disclosure variable has a significant negative coefficient. The result is robust under many settings. Thus, the paper finds that sustainability information disclosure significantly diminishes the cost of equity capital, controlling for ESG information disclosure. Taxus is a rare and endangered woody plant worldwide with important economic and ecological values. However, the weak environmental adaptability of Taxus species, in particular the unstable photosynthetic activity in different seasons, always affects its normal growth and development and limits its conservation and exploitation. To improve the survival of Taxus trees in cultivated areas, the seasonal dynamics of chlorophyll fluorescence (CF) and key physiological parameters were comprehensively investigated in T. media and T. mairei. The results demonstrated that the photosynthetic activity of both Taxus species was sensitive to local summer and winter environmental conditions, with the heterogeneity of fluorescence signatures intuitively presented on the needle surface by CF-Imaging detection, while images of maximum quantum efficiency of PSII photochemistry (Fv/Fm) demonstrated values below 0.7 in the blue–green sectors in winter. The distribution of light energy was regulated by the photosynthetic apparatus in both Taxus species to maintain a stable actual quantum yield of PSII photochemistry (φPSII), which was around 0.4–0.5. Based on a redundancy discriminant analysis, the interpretation rate of light intensity and air temperature ranked as the top two in both Taxus species, which were considered the main environmental factors affecting the photosynthetic performance of Taxus by disturbing the electron transport chain. In the winter, T. mairei exhibited weaker electron transport activity than T. media, thus caused lower photochemistry and more severe photosynthetic damages. Interestingly, both Taxus species demonstrated consistent response patterns, including diverse energy dissipation strategies and enhancement of osmoregulatory substances and antioxidative activities, thus maintaining stable photosynthetic functions in response to environmental changes. A biological process called epithelial-mesenchymal transition (EMT) allows epithelial cells to change into mesenchymal cells and acquire some cancer stem cell properties. EMT contributes significantly to the metastasis, invasion, and development of treatment resistance in cancer cells. Current research has demonstrated that phytochemicals are emerging as a potential source of safe and efficient anti-cancer medications. Phytochemicals could disrupt signaling pathways related to malignant cell metastasis and drug resistance by suppressing or reversing the EMT process. In this review, we briefly describe the pathophysiological properties and the molecular mechanisms of EMT in the progression of cancers, then summarize phytochemicals with diverse structures that could block the EMT process in different types of cancer. Soil salinization is an important factor limiting food security and ecological stability. As a commonly used greening tree species, Robinia pseudoacacia often suffers from salt stress that can manifest as leaf yellowing, decreased photosynthesis, disintegrated chloroplasts, growth stagnation, and even death. To elucidate how salt stress decreases photosynthesis and damages photosynthetic structures, we treated R. pseudoacacia seedlings with different concentrations of NaCl (0, 50, 100, 150, and 200 mM) for 2 weeks and then measured their biomass, ion content, organic soluble substance content, reactive oxygen species (ROS) content, antioxidant enzyme activity, photosynthetic parameters, chloroplast ultrastructure, and chloroplast development-related gene expression. NaCl treatment significantly decreased biomass and photosynthetic parameters, but increased ion content, organic soluble substances, and ROS content. High NaCl concentrations (100–200 mM) also led to distorted chloroplasts, scattered and deformed grana lamellae, disintegrated thylakoid structures, irregularly swollen starch granules, and larger, more numerous lipid spheres. Compared to control (0 mM NaCl), the 50 mM NaCl treatment significantly increased antioxidant enzyme activity while upregulating the expression of the ion transport-related genes Na+/H+ exchanger 1(NHX 1) and salt overly sensitive 1 (SOS 1) and the chloroplast development-related genes psaA, psbA, psaB, psbD, psaC, psbC, ndhH, ndhE, rps7, and ropA. Additionally, high concentrations of NaCl (100–200 mM) decreased antioxidant enzyme activity and downregulated the expression of ion transport- and chloroplast development-related genes. These results showed that although R. pseudoacacia can tolerate low concentrations of NaCl, high concentrations (100–200 mM) can damage chloroplast structure and disturb metabolic processes by downregulating gene expression. This paper explores the strategies and practices of capturing climate risk premia for venture capital (VC) fund managers and entrepreneurs in the private cleantech sector. It also examines the impact of the feed-in tariffs (FITs) policy on the management of cleantech investments. It is shown that a longer investment period, less investment capital in cleantech investment management strategies, and optimistic climate risk management practices will help investors to better capture climate risk premia. In fact, the FITs policy will give rise to VC fund managers and entrepreneurs having a positive view regarding the prospects of the cleantech sector, motivating them to make long-term investments. Furthermore, it is shown that the greater the impact of the FITs policy, the greater the climate risk premia to be captured. An FeCoCrAlCu HEA coating was prepared on the surface of 45# steel by cold-spray-assisted induction remelting. The results showed that the FeCoCrAlCu HEA coating was composed of BCC and FCC phases. The BCC phase possessed an amplitude-modulated structure consisting of a B1-disordered phase (FeCr) and a B2-ordered phase (AlCo), as well as a nanoscale BCC phase precipitated near grain boundaries. The FCC phase was composed of a solid solution of the Al–Cu matrix and manifested characteristics of a typical twin structure. In addition, the hardness of the FeCoCrAlCu HEA coating was 528.2 HV. The friction coefficient of the FeCoCrAlCu HEA-/Al2O3 pair was 0.379, and the wear rate was 3.96 × 10−5 mm3/(N × m). In 3.5 wt.%NaCl and 5.0 wt.%H2SO4 corrosive media, the FeCoCrAlCu HEA coating had a more positive self-corrosion potential (Ecorr) and a lower corrosion current density (Icorr) than the substrate. This study is aimed at developing novel analytical methods to accurately visualize the spatial distribution of various endogenous components in Arctium lappa L. (A. lappa) roots, and to precisely guide the setting of pre-treatment operations during processing technologies and understand plant metabolism process. The matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) imaging technology was used for visual demonstration of the in situ spatial distribution in A. lappa roots. This work consisted of four steps: matrix selection, section preparation, matrix coating, and MALDI-TOF MS imaging analysis. Consequently, eight saccharides, four caffeoylquinic acids, four flavonoids, six amino acids, one choline, and one phospholipid were imaged and four unidentified components were found. Saccharides were distributed in the center, whereas caffeoylquinic acids and flavonoids were mainly present in the epidermis and cortex. Furthermore, amino acids were mainly detected in the phloem, and choline in the cambium, while phosphatidylserine was found in the secondary phloem and cambium. This study demonstrated that MALDI-TOF MS imaging technology could provide a technical support to understand the spatial distribution of components in A. lappa roots, which would promote the processing technologies for A. lappa roots and help us to understand the plant metabolism process. The present study aimed to explore the improvement of the bioactivity of ginger peel polysaccharides (GPs) by the modification of zinc after structural characterization. The obtained GP–Zn (II) complexes consisted dominantly of glucose and galactose in a mass proportion of 95.10:2.10, with a molecular weight of 4.90 × 105 Da and a Zn content of 21.17 mg/g. The chelation of GPs and Zn (II) was mainly involved in the O–H of hydroxyl groups, and this interaction reduced the crystallinity and decreased the asymmetry of GPs, with a slight effect on the thermal stability. The administration of GPs and their Zn (II) complexes effectively alleviated CuSO4-induced inflammatory response in zebrafish (Tg: zlyz-EGFP) via down-regulating the mRNA expression levels of pro-inflammatory cytokines (IL-1β, IL-6, IL-8, IL-12 and TNF-α) and upregulating the expression of anti-inflammatory cytokine (IL-10). Furthermore, the modification of Zn (II) enhanced the inflammation-inhibiting effect of polysaccharides. A series of novel aloe-emodin–coumarin hybrids were designed and synthesized. The antitumor activity of these derivatives was evaluated against five human tumor cell lines (A549, SGC-7901, HepG2, MCF-7 and HCT-8). Some of the synthesized compounds exhibited moderate to good activity against one or more cell lines. Particularly, compound 5d exhibited more potent antiproliferative activity than the reference drug etoposide against all tested tumor cell lines, indicating that it had a broad spectrum of antitumor activity and that it may provide a promising lead compound for further development as an antitumor agent by structural modification. Single-cell RNA-sequencing (scRNA-seq) is a recent high-throughput technique that can measure gene expression, reveal cell heterogeneity, rare and complex cell populations, and discover cell types and their relationships. The analysis of scRNA-seq data is challenging because of transcripts sparsity, replication noise, and outlier cell populations. A gene coexpression network (GCN) analysis effectively deciphers phenotypic differences in specific states by describing gene–gene pairwise relationships. The underlying gene modules with different coexpression patterns partially bridge the gap between genotype and phenotype. This study presents a new framework called scGENA (single-cell gene coexpression network analysis) for GCN analysis based on scRNA-seq data. Although there are several methods for scRNA-seq data analysis, we aim to build an integrative pipeline for several purposes that cover primary data preprocessing, including data exploration, quality control, normalization, imputation, and dimensionality reduction of clustering as downstream of GCN analysis. To demonstrate this integrated workflow, an scRNA-seq dataset of the human diabetic pancreas with 1600 cells and 39,851 genes was implemented to perform all these processes in practice. As a result, scGENA is demonstrated to uncover interesting gene modules behind complex diseases, which reveal biological mechanisms. scGENA provides a state-of-the-art method for gene coexpression analysis for scRNA-seq data. The aim of our study was to synthesize a pumpkin peel polysaccharide (PPP)-Cr(III) complex and investigate its hypoglycemic activity. Firstly, a novel PPP-Cr(III) complex with a Cr content of 23.77 mg/g was synthesized and characterized. Physicochemical characterization indicated that PPP-Cr(III) had some changes in chemical composition, monosaccharide composition, and morphological structure compared with PPP. The molecular weights of PPP-Cr(III) and PPP were 1.398 × 106 g/mol and 3.386 × 106 g/mol, respectively, showing a lower molecular weight after the introduction of Cr(III). Fourier transform infrared spectroscopy showed that a new characteristic absorption peak of Cr-O appeared at 534 cm−1 in PPP-Cr(III), indicating that Cr(III) was successfully complexed with PPP. Secondly, the hypoglycemic activity of PPP-Cr(III) based on α-glucosidase inhibitory and insulin resistance (IR)-HepG2 cells was evaluated. Compared with PPP, PPP-Cr(III) exhibited a more significantly α-glucosidase inhibitory activity. The IR-HepG2 cells confirmed an obvious increase in glucose consumption. Western blot analysis demonstrated that the treated IR-HepG2 cells were able to increase the protein levels of p-AMPK and p-GSK-3β, indicating that IR-HepG2 cells exerted hypoglycemic activity via the AMPK/GSK-3β signaling pathway. Taxus mairei is an important source for industrial extraction of taxol in China. However, the standard and steps of extraction are currently not uniform, which seriously affects the taxol yield. In the present study, the influence of four factors (methanol concentration, solid-liquid ratio, ultrasonic extraction temperature, and ultrasonic extraction time) on the taxol yield was successively explored in T. mairei. A response surface methodology (RSM) was used to optimize the extraction process based on the single-factor experiments above. The optimal conditions were as follows: methanol concentration was 90%, solid-liquid ratio was 1:15 (g/mL), ultrasonic extraction temperature was 40 °C and ultrasonic extraction time was 60 min. Moreover, the twigs and needles from T. mairei with different tree ages were treated by the optimum extraction process, which further revealed temporal and spatial distribution of taxol in the reproducible tissues. Interestingly, the taxol content was relatively higher in needles of T. 'Jinxishan' (a cultivar from T. mairei with yellow aril, FY), but was less in FY twigs. The accumulation of taxol in twigs and leaves of females (with red aril, FR) was significantly higher than that of males (M); however, the content showed a decreasing trend with the increasing tree ages. Therefore, it is suitable to increase the proportion of female trees especially the FY leaves as raw materials for the industrial production of taxol from T. mairei, and the tree ages should be better controlled at 3–7 years. Leaf coloration changes evoke different photosynthetic responses among different poplar cultivars. The aim of this study is to investigate the photosynthetic difference between a red leaf cultivar (ZHP) and a green leaf (L2025) cultivar of Populus deltoides. In this study, 'ZHP' exhibited wide ranges and huge potential for absorption and utilization of light energy and CO2 concentration which were similar to those in 'L2025' and even showed a stronger absorption for weak light. However, with the increasing light intensity and CO2 concentration, the photosynthetic capacity in both 'L2025' and 'ZHP' was gradually restricted, and the net photosynthetic rate (Pn) in 'ZHP' was significantly lower than that in 'L2025'under high light or high CO2 conditions, which was mainly attributed to stomatal regulation and different photosynthetic efficiency (including the light energy utilization efficiency and photosynthetic CO2 assimilation efficiency) in these two poplars. Moreover, the higher anthocyanin content in 'ZHP' than that in 'L2025' was considered to be closely related to the decreased photosynthetic efficiency in 'ZHP'. According to the results from the JIP-test, the capture efficiency of the reaction center for light energy in 'L2025' was significantly higher than that in 'ZHP'. Interestingly, the higher levels of light quantum caused relatively higher accumulation of QA- in 'L2025', which blocked the electron transport and weakened the photosystem II (PSII) performance as compared with 'ZHP'; however, the decreased capture of light quantum also could not promote the utilization of light energy, which was the key to the low photosynthetic efficiency in 'ZHP'. Taxol is one of the most effective anticancer drugs in the world that is widely used in the treatments of breast, lung and ovarian cancer. The elucidation of the taxol biosynthetic pathway is the key to solve the problem of taxol supply. So far, the taxol biosynthetic pathway has been reported to require an estimated 20 steps of enzymatic reactions, and sixteen enzymes involved in the taxol pathway have been well characterized, including a novel taxane-10β-hydroxylase (T10βOH) and a newly putative β-phenylalanyl-CoA ligase (PCL). Moreover, the source and formation of the taxane core and the details of the downstream synthetic pathway have been basically depicted, while the modification of the core taxane skeleton has not been fully reported, mainly concerning the developments from diol intermediates to 2-debenzoyltaxane. The acylation reaction mediated by specialized Taxus BAHD family acyltransferases (ACTs) is recognized as one of the most important steps in the modification of core taxane skeleton that contribute to the increase of taxol yield. Recently, the influence of acylation on the functional and structural diversity of taxanes has also been continuously revealed. This review summarizes the latest research advances of the taxol biosynthetic pathway and systematically discusses the acylation reactions supported by Taxus ACTs. This work aimed to investigate the effect of fucoidan (FPS) on urate transporters induced by uric acid (UA). The results showed that UA stimulated the expression of glucose transporter 9 (GLUT9) and urate transporter 1 (URAT1) in HK-2 cells, and FPS could reverse the effect. Moreover, UA could activate NF-κB, JNK and PI3K/Akt pathways, but both pathway inhibitors and FPS inhibited the UA-induced activation of these three pathways. These data suggested that FPS effectively inhibited the expression induction of reabsorption transporters URAT1 and GLUT9 by UA, through repressing the activation of NF-κB, JNK and PI3K/Akt signal pathways in HK-2 cells. The in vitro research findings support the in vivo results that FPS reduces serum uric acid content in hyperuricemia mice and rats through inhibiting the expression of URAT1 and GLUT9 in renal tubular epithelial cells. Flowering crabapples are a series of precious ornamental woody plants. However, their growth and development are inhibited in the subtropical regions due to the weak photosynthesis under high-temperature environment in the summer. Chlorophyll a fluorescence transient and 2-dimensional electrophoresis (2-DE) analyses were conducted to investigate the response characteristics of photosynthesis under simulated 38 °C heat stress in leaves of Malus. 'Prairifire', a spring-red leaf cultivar of flowering crabapple with strong thermal adaptability. In the present study, the net photosynthetic rate (Pn) was significantly decreased during the heat shock process, which showed a similar trend to the stomatal conductance (Gs), indicating a sensitive stomatal behavior to heat stress. Moreover, an efficient reaction center in photosystem II (PSII), and a functionally intact oxygen-evolving complex (OEC) conferred strong photosynthetic adaptability under heat stress. The higher level of transketolase (TK) under 48-h heat shock treatment was considered a protective mechanism of photosynthetic apparatus. However, heat stress inhibited the functions of light harvesting complex II (LHCII), electron transport in PSII, and the levels of key enzymes in the Calvin cycle, which were considered as the reasons causing an increase in the proportion of non-stomatal restrictions. Twenty-five sophora alkaloids-cinnamic acid hybrids (including matrine-cinnamic acid hybrids, sophoridine-cinnamic acid hybrids, and sophocarpine-cinnamic acid hybrids) were designed, synthesized, and evaluated in vitro against three human tumor cell lines (HeLa, HepG2 and A549) with cisplatin as a positive control. Some matrine-cinnamic acid and sophoridine-cinnamic acid compounds exhibited potent effect against all three cancer cell lines, such as compounds 5b, 5e, 5g, and 6d. The structure-activity relationship study of the synthesized compounds was also performed. Preliminary mechanistic studies indicated that compounds 5e and 6d could induce apoptosis in HepG2 cell line. Further, compounds 5e and 6d altered mitochondrial membrane potential and produced ROS leading to cell apoptosis of HepG2 cells. Under the context of climate change, studying CO2 emissions in alpine rivers is important because of the large carbon storage in these terrestrial ecosystems. In this study, riverine partial pressure of CO2 (pCO2) and CO2 emission flux (FCO2) in the Yellow River source region (YRSR) under different landcover types, including glaciers, permafrost, peatlands, and grasslands, were systematically investigated in April, June, August, and October 2016. Relevant chemical and environmental parameters were analyzed to explore the primary controlling factors. The results showed that most of the rivers in the YRSR were net CO2 source, with the pCO2 ranging from 181 to 2441 μatm and the FCO2 ranging from −50 to 1574 mmol m−2 d−1. Both pCO2 and FCO2 showed strong spatial and temporal variations. The highest average FCO2 was observed in August, while the lowest average was observed in June. Spatially, the lowest FCO2 were observed in the permafrost regions while the highest FCO2 were observed in peatland. By integrating seasonal changes of the water surface area, total CO2 efflux was estimated to be 0.30 Tg C year−1. This indicates that the YRSR was a net carbon source for the atmosphere, which contradicts previous studies that conclude the YRSR as a carbon sink. More frequent measurements of CO2 fluxes, particularly through several diel cycles, are necessary to confirm this conclusion. Furthermore, our study suggested that the riverine dissolved organic carbon (DOC) in permafrost (5.0 ± 2.4 mg L−1) is possibly derived from old carbon released from permafrost melting, which is equivalent to that in peatland regions (5.1 ± 3.7 mg L−1). The degradation of DOC may have played an important role in supporting riverine CO2, especially in permafrost and glacier-covered regions. The percent coverage of corresponding land cover types is a good indicator for estimating riverine pCO2 in the YRSR. In view of the extensive distribution of alpine rivers in the world and their sensitivity to climate change, future studies on dynamics of stream water pCO2 and CO2 outgassing are strongly needed to better understand the global carbon cycle. Melatonin, an indoleamine widely found in animals and plants, is considered as a candidate phytohormone that affects responses to a variety of biotic and abiotic stresses. In plants, melatonin has a similar action to that of the auxin indole-3-acetic acid (IAA), and IAA and melatonin have the same biosynthetic precursor, tryptophan. Salt stress results in the rapid accumulation of melatonin in plants. Melatonin enhances plant resistance to salt stress in two ways: one is via direct pathways, such as the direct clearance of reactive oxygen species; the other is via an indirect pathway by enhancing antioxidant enzyme activity, photosynthetic efficiency, and metabolite content, and by regulating transcription factors associated with stress. In addition, melatonin can affect the performance of plants by affecting the expression of genes. Interestingly, other precursors and metabolite molecules associated with melatonin can also increase the tolerance of plants to salt stress. A total of 18 matrine derivatives were designed, synthesized, and evaluated for their inhibitory effect against TGF-β1-induced total collagen accumulation in human fetal lung fibroblast MRC-5 cell lines. Among them, compound 3f displayed the most potent anti-fibrotic activity (IC50 = 3.3 ± 0.3 μM) which was 266-fold more potent than matrine. 3f significantly inhibited the fibroblast-to-myofibroblast transition and extracellular matrix production of MRC-5 cells. The TGF-β/small mothers against decapentaplegic homologs (Smad) signaling was also inhibited by 3f, as evidenced by inhibition of cytoplasm-to-nuclear translocation of Smad2/3 and suppression of TGF-β1-induced upregulation of TGF-β receptor type I (TGFβRI). Additionally, 3f exhibited potent inhibitory effects against TGF-β1-induced fibroblasts migration. The recombinant baculovirus has been widely used as an efficient tool to mediate gene delivery into mammalian cells but has barely been used in fish cells. In the present study, we constructed a recombinant baculovirus containing the dual-promoter cytomegalovirus (CMV) and white spot syndrome virus (WSSV) immediate-early gene 1 (ie1) (WSSV ie1), followed by a puromycin–green fluorescent protein (Puro-GFP, pf) or puromycin–red fluorescent protein (Puro-RFP, pr) cassette, which simultaneously allowed for easy observation, rapid titer determination, drug selection, and exogenous gene expression. This recombinant baculovirus was successfully transduced into fish cells, including Mylopharyngodon piceus bladder (MPB), fin (MPF), and kidney (MPK); Oryzias latipes spermatogonia (SG3); and Danio rerio embryonic fibroblast (ZF4) cells. Stable transgenic cell lines were generated after drug selection, which was further verified by Western blot. A cell monoclonal formation assay proved the stable heredity of transgenic MPB cells. In addition, a recombinant baculovirus containing a pr cassette and four transcription factors for induced pluripotent stem cells (iPSC) was constructed and transduced into ZF4 cells, and these exogenous genes were simultaneously delivered and transcribed efficiently in drug-selected ZF4 cells, proving the practicability of this modified recombinant baculovirus system. We also proved that the WSSV ie1 promoter had robust activity in fish cells in vitro and in vivo. City sustainability is an important issue in the urbanization process. In this paper, the sustainability of 14 cities in Liaoning province in China is evaluated and predicted. The process of evaluating city sustainability is viewed as a multi-criteria decision-making problem. A simple additive weighting method is used for aggregating the normalized sustainability criteria data, built based on the three-pillar model and the associated weights. The results indicate that although the sustainability of the cities in Liaoning province is not perfect, the cities show better development momentum. For example, only two cities' (Shenyang and Dalian) average performance scores in 2010–2016 were over 0.5, but all the cities' sustainability improved in 2016 compared to 2010. We develop a stochastic simulation procedure used for predicting a city's sustainability in future years. Many prediction results were obtained, including the maximum, minimum, and average performance scores of the cities, the pairwise priority matrix representing the ratio of one city outperforming another, and the ranking of cities' sustainability with probabilities. The prediction results indicate various degrees of increase for almost all the cities' sustainability in the future, which is consistent with the judgment of better development momentum determined from the evaluation results. Shape-memory polymers (SMPs) selectively induced by near-infrared lights of 980 or 808 nm were synthesized via free radical copolymerization. Methyl methacrylate (MMA) monomer, ethylene glycol dimethylacrylate (EGDMA) as a cross-linker, and organic complexes of Yb(TTA)2AAPhen or Nd(TTA)2AAPhen containing a reactive ligand of acrylic acid (AA) were copolymerized in situ. The dispersion of the organic complexes in the copolymer matrix was highly improved, while the transparency of the copolymers was negligibly influenced in comparison with the pristine cross-linked PMMA. In addition, the thermal resistance of the copolymers was enhanced with the complex loading, while their glass transition temperature, cross-linking level, and mechanical properties were to some extent reduced. Yb(TTA)2AAPhen and Nd(TTA)2AAPhen provided the prepared copolymers with selective photothermal effects and shape-memory functions for 980 and 808 nm NIR lights, respectively. Early embryonic development is a multi-step process that is intensively regulated by various signaling pathways. Because of the complexity of the embryo and the interactions between the germ layers, it is very difficult to fully understand how these signals regulate embryo patterning. Recently, pluripotent stem cell lines derived from different developmental stages have provided an in vitro system for investigating molecular mechanisms regulating cell fate decisions. In this review, we summarize the major functions of the BMP, FGF, Nodal and Wnt signaling pathways, which have well-established roles in vertebrate embryogenesis. Then, we highlight recent studies in pluripotent stem cells that have revealed the stage-specific roles of BMP，FGF and Nodal pathways during neural differentiation. These findings enhance our understanding of the stepwise regulation of embryo patterning by particular signaling pathways and provide new insight into the mechanisms underlying early embryonic development.
