In this paper, the laser beam oscillation welding (LBOW) was utilized to weld a 2 mm thick AA2060 aluminum-lithium (Al-Li) alloy plate. The weld pool behaviors under three scanning paths (pure laser, O-shaped, and ∞-shaped) were investigated. It was observed that the O-shaped scanning path resulted in the most stable welding process. In addition, the weld macroscopic formation, microstructure, and mechanical property between different paths were studied. The results showed that pure laser and ∞-shaped patterns produced welding defects such as spatters and collapse during the welding process, while the O-shaped pattern exhibited good macroscopic formation at varying laser powers. The O-shaped pattern promoted the finest grain in the weld center and reduced the heat input during the welding process. The equiaxed grain zone (EQZ) width of the O-shaped pattern is the smallest compared to the other two patterns at high laser power. In addition to this, the O-shaped pattern could effectively reduce the porosity in the weld. When an O-shaped scanning pattern was adopted at the ideal laser power parameter of 3000 W, the microhardness of the weld center increased by approximately 5.6% compared to pure laser mode. Background: Brown seaweed is promising for the treatment of type 2 diabetes mellitus (T2DM). Its bioactive constituents can positively affect plasma glucose homeostasis in healthy humans. We investigated the effect of the brown seaweeds Sargassum (S.) fusiforme and Fucus (F.) vesiculosus in their natural form on glucose regulation in patients with T2DM. Methods: We conducted a randomized, double-blind, placebo-controlled pilot trial. Thirty-six participants with T2DM received, on a daily basis, either 5 g of dried S. fusiforme, 5 g of dried F. vesiculosus, or 0.5 g of dried Porphyra (control) for 5 weeks, alongside regular treatment. The primary outcome was the between-group difference in the change in weekly average blood glucose levels (continuous glucose monitoring). The secondary outcomes were the changes in anthropometrics, plasma lipid levels, and dietary intake. The data were analyzed using a linear mixed-effects model. Results: The change in weekly average glucose levels was 8.2 ± 2.1 to 9.0 ± 0.7 mmol/L (p = 0.2) in the S. fusiforme group (n = 12) and 10.1 ± 3.3 to 9.2 ± 0.7 mmol/L (p = 0.9) in the F. vesiculosus group (n = 10). The between-group difference was non-significant. Similarly, no between-group differences were observed for the changes in the secondary outcomes. Discussion: A daily intake of 5 g of fresh, dried S. fusiforme or F. vesiculosus alongside regular treatment had no differential effect on weekly average blood glucose levels in T2DM. Improving the high-temperature stress rupture properties of Inconel 718 (IN718) alloys is crucial for enhancing aircraft engine performance. By using the laser powder bed fusion (LPBF) technique, IN718 alloys were crafted at varying volumetric energy densities (VED) in this study. The dendrite growth mode, reinforcing phase distribution and high temperature stress rupture properties of various VED samples were investigated. The results showed that the stress rupture life and the uniform elongation of the samples both first increased and then decreased with the increase in VED. When the VED was 60 J/mm3, the maximum rupture life and elongation of the sample were 43 h and 3.8%, respectively. As the VED increased, the angle of dislocation in the dendrite decreased while the spacing between primary dendrite arms increased, resulting in an increase in the size and volume fraction of the Laves phase. Following a heat treatment, the δ phase would nucleate preferentially around the dissolved Laves phase causing an increase in the volume fraction of the δ phase with the increase in VED. The creep voids readily formed around the δ phase are distributed along the grain boundaries, while the inhomogeneous δ phase and fine grains facilitated crack initiation and propagation. Furthermore, a significant quantity of the δ phase consumed the Nb element, thereby hindering adequate precipitation in the γ″ phase and causing cracks. [ABSTRACT FROM AUTHOR]

The nuclear liver X receptors (LXRα/β) and peroxisome proliferator-activated receptors (PPARα/γ) are involved in the regulation of multiple biological processes, including lipid metabolism and inflammation. The activation of these receptors has been found to have neuroprotective effects, making them interesting therapeutic targets for neurodegenerative disorders such as Alzheimer's Disease (AD). The Asian brown seaweed Sargassum fusiforme contains both LXR-activating (oxy)phytosterols and PPAR-activating fatty acids. We have previously shown that dietary supplementation with lipid extracts of Sargassum fusiforme prevents disease progression in a mouse model of AD, without inducing adverse effects associated with synthetic pan-LXR agonists. We now determined the LXRα/β- and PPARα/γ-activating capacity of lipid extracts of six European brown seaweed species (Alaria esculenta, Ascophyllum nodosum, Fucus vesiculosus, Himanthalia elongata, Saccharina latissima, and Sargassum muticum) and the Asian seaweed Sargassum fusiforme using a dual luciferase reporter assay. We analyzed the sterol and fatty acid profiles of the extracts by GC-MS and UPLC MS/MS, respectively, and determined their effects on the expression of LXR and PPAR target genes in several cell lines using quantitative PCR. All extracts were found to activate LXRs, with the Himanthalia elongata extract showing the most pronounced efficacy, comparable to Sargassum fusiforme, for LXR activation and transcriptional regulation of LXR-target genes. Extracts of Alaria esculenta, Fucus vesiculosus, and Saccharina latissima showed the highest capacity to activate PPARα, while extracts of Alaria esculenta, Ascophyllum nodosum, Fucus vesiculosus, and Sargassum muticum showed the highest capacity to activate PPARγ, comparable to Sargassum fusiforme extract. In CCF-STTG1 astrocytoma cells, all extracts induced expression of cholesterol efflux genes (ABCG1, ABCA1, and APOE) and suppressed expression of cholesterol and fatty acid synthesis genes (DHCR7, DHCR24, HMGCR and SREBF2, and SREBF1, ACACA, SCD1 and FASN, respectively). Our data show that lipophilic fractions of European brown seaweeds activate LXRs and PPARs and thereby modulate lipid metabolism. These results support the potential of brown seaweeds in the prevention and/or treatment of neurodegenerative diseases and possibly cardiometabolic and inflammatory diseases via concurrent activation of LXRs and PPARs. Patients undergoing bone fractures frequently suffer from irritating chronic pain after orthopedic repairs. Chemokine-mediated interactions between neurons and microglia are important steps for neuroinflammation and excitatory synaptic plasticity during the spinal transmission of pathological pain. Recently, glabridin, the main bioactive component of licorice, has been shown to exhibit anti-nociceptive and neuroprotective properties for inflammatory pain. This present study evaluated the therapeutic potential of glabridin and its analgesic mechanisms using a mouse model of tibial fracture-associated chronic pain. Repetitive injections of glabridin were delivered spinally daily for 4 continuous days from days 3 to 6 after the fractures. Herein, we discovered that repeated administrations of glabridin (10 and 50 μg, but not 1 μg) could prevent prolonged cold allodynia and mechanical allodynia following bone fractures. A single intrathecal intervention with glabridin (50 μg) relieved an existing chronic allodynia two weeks following the fracture surgeries. Systemic therapies with glabridin (intraperitoneal; 50 mg kg−1) were protective against long-lasting allodynia caused by fractures. Furthermore, glabridin restricted the fracture-caused spinal overexpressions of the chemokine fractalkine and its receptor CX3CR1, as well as the elevated number of microglial cells and dendritic spines. Strikingly, glabridin induced the inhibition of pain behaviors, microgliosis, and spine generation, which were abolished with the co-administration of exogenous fractalkine. Meanwhile, the exogenous fractalkine-evoked acute pain was compensated after microglia inhibition. Additionally, spinal neutralization of fractalkine/CX3CR1 signaling alleviated the intensity of postoperative allodynia after tibial fractures. These key findings identify that glabridin therapies confer protection against inducing and sustaining fracture-elicited chronic allodynia by suppressing fractalkine/CX3CR1-dependent spinal microgliosis and spine morphogenesis, suggesting that glabridin is a promising candidate in the translational development of chronic fracture pain control. The nuclear receptors—liver X receptors (LXR α and β) are potential therapeutic targets in cardiovascular and neurodegenerative diseases because of their key role in the regulation of lipid homeostasis and inflammatory processes. Specific oxy(phyto)sterols differentially modulate the transcriptional activity of LXRs providing opportunities to develop compounds with improved therapeutic characteristics. We isolated oxyphytosterols from Sargassum fusiforme and synthesized sidechain oxidized sterol derivatives. Five 24-oxidized sterols demonstrated a high potency for LXRα/β activation in luciferase reporter assays and induction of LXR-target genes APOE, ABCA1 and ABCG1 involved in cellular cholesterol turnover in cultured cells: methyl 3β-hydroxychol-5-en-24-oate (S1), methyl (3β)-3-aldehydeoxychol-5-en-24-oate (S2), 24-ketocholesterol (S6), (3β,22E)-3-hydroxycholesta-5,22-dien-24-one (N10) and fucosterol-24,28 epoxide (N12). These compounds induced SREBF1 but not SREBP1c-mediated lipogenic genes such as SCD1, ACACA and FASN in HepG2 cells or astrocytoma cells. Moreover, S2 and S6 enhanced cholesterol efflux from HepG2 cells. All five oxysterols induced production of the endogenous LXR agonists 24(S)-hydroxycholesterol by upregulating the CYP46A1, encoding the enzyme converting cholesterol into 24(S)-hydroxycholesterol; S1 and S6 may also act via the upregulation of desmosterol production. Dissimilar materials brazing of Ti2AlNb alloy to GH99 superalloy is of great pragmatic importance in the aerospace field, especially the lightweight space aircraft components manufacturing. In this work, TiZrCuNi amorphous filler alloy was used as brazing filler, and experiments were carried out at different brazing temperatures and times to investigate the changes in interfacial structures and properties of the joints. The typical interfacial microstructure was Ti2AlNb alloy/B2/β/Ti2Ni (Al, Nb) + B2/β + (Ti, Zr)2(Ni, Cu) + (Ti, Zr)(Ni, Cu)/(Cr, Ni, Ti) solid solution + (Ni, Cr) solid solution/GH99 superalloy when being brazed at 1000 °C for 8 min. The interfacial microstructure of the joints was influenced by diffusion and reaction between the filler alloy and the parent metal. The prolongation of brazing process parameters accelerated the diffusion and reaction of the liquid brazing alloy into both parent metals, which eventually led to the aggregation of (Ti, Zr)2(Ni, Cu) brittle phase and increased thickness of Ti2Ni (Al, Nb) layer. According to fracture analyses, cracks began in the Ti2Ni (Al, Nb) phase and spread with it as well as the (Ti, Zr)2(Ni, Cu) phase. The joints that were brazed at 1000 °C for 8 min had a maximum shear strength of ~216.2 MPa. Furthermore, increasing the brazing temperature or extending the holding time decreased the shear strength due to the coarse Ti2Ni (Al, Nb) phase and the continuous (Ti, Zr)2(Ni, Cu) phase. A laser powder bed fusion (LPBF) Inconel 718 (IN718) alloy was fabricated under different process parameters, and the samples with optimal process parameters were treated by subsequent heat treatment. The outcomes demonstrated that the pores' size and shape were influenced by the process parameters. The porosity of the samples first grew and subsequently reduced as the volume energy density increased, and the types of pores changed from lack-of-fusion pores to gas holes and keyhole pores. The porosity of the samples with three process parameters were 3.46%, 0.15% and 1.26%, respectively. The sample of 60.0 J/mm3 was chosen for a single solution (heat treatment at 980 °C for 1 h, SS) and direct aging (heat treatment at 720 °C for 8 h and then 620 °C for 8 h, DA), and the microstructure evolution of the samples was examined. Because of the segregation of the elements Nb and Mo, the Laves phase was distributed among the dendrites in the as-built (AB) sample. The solution heat treatment dissolved the Laves phase and precipitated the δ phase, while aging heat treatment precipitated the γ′/γ" phases. Additionally, the impact of the second phase on the tensile characteristics was looked into. The DA sample had the highest strength, with a tensile strength of 1458 MPa and a yield strength of 1271 MPa. With tensile and yield strengths of 980 MPa and 620 MPa, respectively, the AB sample had the lowest strength due to the Laves phase's role as the crack initiator. This paper is dedicated to the calculation of the radiative properties of 82%argon-18%CO2 thermal plasmas with the addition of metallic vapors (iron, in the present case, due to workpiece and wire erosion), this mixture being representative of metal active gas (MAG) arc welding processes. These radiative properties are obtained in the frame of the net emission coefficient (NEC) theory, using the recent and accurate "line by line" method. All significant radiative contribution mechanisms are taken into account in the calculation: atomic lines, atomic continuum (radiative attachment, radiative recombination, and bremsstrahlung), molecular bands for diatomic and polyatomic molecules, and molecular continuum. Broadening phenomena (Doppler and pressure effects) are also carefully treated for bound-bound transitions (atomic lines and molecular bands). Regarding 82%Ar-18%CO2 plasma, the results obtained demonstrate the key role of molecular bands at low temperatures (T < 4 kK), whereas the atomic line and continuum prevailed at intermediate and high temperatures. With the addition of a few percentages of iron vapor, it was shown that the total NEC is significantly increased (especially at low temperatures) and that the atomic and ionic lines become dominant in all the studied temperature ranges (3–30 kK). This theoretical study will constitute a groundwork to build a diagnostic method (based on the calculation of partial NECs for accurately chosen spectral intervals) for the determination of plasma temperature and iron vapor concentration in welding arcs. High-efficiency and high-quality welding has always been the focus of welding research. This article proposes a novel double-pulse, triple-wire MIG welding process for the welding of 6082-T6 aluminum alloy. The process characteristics of welding arc and droplet transfer were studied, and the performances of weld formation, morphology, hardness, and tensile strength were tested for the 1 Hz, 3 Hz, and 5 Hz double-pulse welding and normal-pulse welding. It was found that in the welding process, the pulsed arc steadily alternated among three welding wires without arc interruption, and the arc length changed periodically with the double-pulse frequency. The droplets transferred with a stable one-pulse-one-drop mode. Besides, a proper double-pulse frequency, e.g., 3 Hz in this case, was conducive to forming good welds with regular fish-scale patterns and no pores. The tensile strength of the joint could reach 64% of the base material's tensile strength, and its fracture belonged to plastic fracture, which occurred in the HAZ. Dysregulation of cholesterol homeostasis is a major risk factor of atherosclerosis, which can lead to serious health problems, including heart attack and stroke. Liver X receptor (LXR) α and β are transcription factors belonging to the nuclear receptor superfamily, which play important roles in cholesterol homeostasis. Selectively activating LXRβ provides a promising strategy for the treatment of atherosclerosis. Here, we employed atherosclerotic apoE-knockout mice to evaluate the effects of saringosterol, a phytosterol with potent and selective action for LXRβ, which we identified previously in edible marine seaweed Sargassum fusiforme. We found that saringosterol treatment reduced the atherosclerotic plaque burden without having undesirable adverse hepatic effects in apoE-deficient mice fed an atherogenic diet. Meanwhile, reduced serum levels of cholesterol, accompanied by altered expression of LXR-regulated genes involved in cholesterol absorption, transport, efflux, excretion, and elimination, were observed in apoE-knockout mice after saringosterol treatment. Together, our study not only establishes saringosterol as an effective cholesterol-lowering and anti-atherogenic phytosterol but also provides insights into the underlying mechanism. In this paper, the microstructure analysis and performance research of dual laser beam welded 2060-T8/2099-T83 aluminum–lithium alloys were carried out. First, the macroscopic morphology and microstructure characteristics of T-joint aluminum–lithium alloys under different welding conditions were observed. Then the effect of welding parameters and pore defects on tensile and fatigue properties of the weld were carried out and the experimental results were analyzed. It was found that the weld heat input has a significant influence on the penetration of the welded aluminum–lithium alloys joint. When the laser power is too high, the weld will absorb more laser energy and the increase in the evaporation of magnesium will further increase the weld penetration. When the penetration depth increases, the transverse tensile strength tends to decrease. There is no obvious rule for the effect of pore defects on the tensile strength of the weld. At the same time, the heat input of the weld is inversely proportional to the porosity. When the weld heat input increases from 19.41 to 23.33 kJ/m, the porosity decreases from 5.35% to 2.08%. During the fatigue test, it was confirmed that the existence of pore defects would reduce the fatigue life of the weld. In addition, from the analysis of the fatigue fracture morphology it can be found that when the porosity is low, the weld toe is the main source of fatigue cracks. The crack propagation zone shows a typical beach pattern and the final fracture of the base metal presents the characteristics of a brittle fracture. While, when the porosity is high, the crack source is mainly located at the pore defects. Plant volatile organic compounds (VOCs) represent a relatively wide class of secondary metabolites. The VOC profiles of seven seaweeds (Grateloupia filicina, Polysiphonia senticulosa, Callithamnion corymbosum, Sargassum thunbergii, Dictyota dichotoma, Enteromorpha prolifera and Ulva lactuca) from the Yellow Sea of China were investigated using multifiber headspace solid phase microextraction coupled with gas chromatography–mass spectrometry (HS-SPME/GC–MS), among them, the VOCs of three red algae Grateloupia filicina, Polysiphonia senticulosa, and Callithamnion corymbosum were first reported. Principal component analysis (PCA) was used to disclose characteristic categories and molecules of VOCs and network pharmacology was performed to predict potential biomedical utilization of candidate seaweeds. Aldehyde was found to be the most abundant VOC category in the present study and (E)-β-ionone was the only compound found to exist in all seven seaweeds. The chemical diversity of aldehydes in E. prolifera suggest its potential application in chemotaxonomy and hinted that divinylbenzene/carboxen/polydimethylsiloxane (DVB/CAR/PDMS) fiber is more suitable for aldehyde extraction. VOCs in D. dichotoma were characterized as sesquiterpenes and diterpenes and the most relevant pharmacological pathway was the neuroactive ligand–receptor interaction pathway, which suggests that D. dichotoma may have certain preventive and therapeutic values in cancer, especially in lung cancer, in addition to neuropsychiatric diseases. We recently found that dietary supplementation with the seaweed Sargassum fusiforme, containing the preferential LXRβ-agonist 24(S)-saringosterol, prevented memory decline and reduced amyloid-β (Aβ) deposition in an Alzheimer's disease (AD) mouse model without inducing hepatic steatosis. Here, we examined the effects of 24(S)-saringosterol as a food additive on cognition and neuropathology in AD mice. Six-month-old male APPswePS1ΔE9 mice and wildtype C57BL/6J littermates received 24(S)-saringosterol (0.5 mg/25 g body weight/day) (APPswePS1ΔE9 n = 20; C57BL/6J n = 19) or vehicle (APPswePS1ΔE9 n = 17; C57BL/6J n = 19) for 10 weeks. Cognition was assessed using object recognition and object location tasks. Sterols were analyzed by gas chromatography/mass spectrometry, Aβ and inflammatory markers by immunohistochemistry, and gene expression by quantitative real-time PCR. Hepatic lipids were quantified after Oil-Red-O staining. Administration of 24(S)-saringosterol prevented cognitive decline in APPswePS1ΔE9 mice without affecting the Aβ plaque load. Moreover, 24(S)-saringosterol prevented the increase in the inflammatory marker Iba1 in the cortex of APPswePS1ΔE9 mice (p < 0.001). Furthermore, 24(S)-saringosterol did not affect the expression of lipid metabolism-related LXR-response genes in the hippocampus nor the hepatic neutral lipid content. Thus, administration of 24(S)-saringosterol prevented cognitive decline in APPswePS1ΔE9 mice independent of effects on Aβ load and without adverse effects on liver fat content. Dietary fats are generally fatty acids that may play positive or negative roles in the prevention and treatment of diseases. In nature, fatty acids occur in the form of mixtures of saturated fatty acid (SFA), monounsaturated fatty acid (MUFA), and polyunsaturated fatty acid (PUFA), so their nutritional and/or medicinal values must be determined. Herein, we do not consider the classic indices, such as ∑SFA, ∑MUFA, ∑PUFA, ∑n-6 PUFA, ∑n-3 PUFA, and n-6 PUFA/n-3 PUFA; instead, we summarize and review the definitions, implications, and applications of indices used in recent years, including the PUFA/SFA, index of atherogenicity (IA), the index of thrombogenicity (IT), the hypocholesterolemic/hypercholesterolemic ratio (HH), the health-promoting index (HPI), the unsaturation index (UI), the sum of eicosapentaenoic acid and docosahexaenoic acid (EPA + DHA), fish lipid quality/flesh lipid quality (FLQ), the linoleic acid/α-linolenic acid (LA/ALA) ratio, and trans fatty acid (TFA). Of these nutritional indices, IA and IT are the most commonly used to assess the composition of fatty acids as they outline significant implications and provide clear evidence. EPA + DHA is commonly used to assess the nutritional quality of marine animal products. All indices have their advantages and disadvantages; hence, a rational choice of which to use is critical. Role-based access control (RBAC) is one of the most popular access-control mechanisms because of its convenience for management and various security policies, such as cardinality constraints, mutually exclusive constraints, and user-capability constraints. Role-engineering technology is an effective method to construct RBAC systems. However, mining scales are very large, and there are redundancies in the mining results. Furthermore, conventional role-engineering methods not only do not consider more than one cardinality constraint, but also cannot ensure authorization security. To address these issues, this paper proposes a novel method called role-engineering optimization with cardinality constraints and user-oriented mutually exclusive constraints (REO_CCUMEC). First, we convert the basic role mining into a clustering problem, based on the similarities between users and use-partitioning and compression technologies, in order to eliminate redundancies, while maintaining its usability for mining roles. Second, we present three role-optimization problems and the corresponding algorithms for satisfying single or double cardinality constraints. Third, in order to evaluate the performance of authorizations in a role-engineering system, the maximal role assignments are implemented, while satisfying multiple security constraints. Role-based access control (RBAC), which has been regarded as one of the most popular access-control mechanisms, is featured by the separation-of-duty constraints, mutually exclusive constraints, and the least-privileges principle. Role mining, a bottom-up role-engineering technology, is an effective method to migrate from a non-RBAC system to an RBAC system. However, conventional role-mining approaches not only do not consider the separation of duty constraints, but also cannot ensure the security of a constructed RBAC system when the corresponding mined results violate the separation of a duty constraint and/or the least-privileges principle. To solve these problems, this paper proposes a novel method called role-mining optimization with separation-of-duty constraints and security detections for authorizations (RMO_SODSDA), which mainly includes two aspects. First, we present a role-mining-optimization approach for satisfying the separation of duty constraints, and we constructed different variants of mutually exclusive constraints to correctly implement the given separation of duty constraints based on unconstrained role mining. Second, to ensure the security of the constructed system and evaluate authorization performance, we reduced the authorization-query problem to a maximal-satisfiability problem. Parametric granular computing classification algorithms lead to difficulties in terms of parameter selection, the multiple performance times of algorithms, and increased algorithm complexity in comparison with nonparametric algorithms. We present nonparametric hyperbox granular computing classification algorithms (NPHBGrCs). Firstly, the granule has a hyperbox form, with the beginning point and the endpoint induced by any two vectors in N-dimensional (N-D) space. Secondly, the novel distance between the atomic hyperbox and the hyperbox granule is defined to determine the joining process between the atomic hyperbox and the hyperbox. Thirdly, classification problems are used to verify the designed NPHBGrC. The Invar alloy is widely used for aircraft wing mould manufacturing. In this work, keyhole-tungsten inert gas (K-TIG) butt welding was used to join 10 mm thick Invar 36 alloy plates. The effect of heat input on the microstructure, morphology and mechanical properties was studied by using scanning electron microscopy, high energy synchrotron X-ray diffraction, microhardness mapping, tensile and impact testing. It was shown that regardless of the selected heat input, the material was solely composed of austenite, although the grain size changed significantly. The change in heat input also led to texture changes in the fusion zone, as qualitatively determined with synchrotron radiation. With increases in heat input, the impact properties of the welded joints decreased. The coefficient of thermal expansion of the joints was measured, which demonstrated that the current process is suitable for aerospace applications.

Sargassum seaweeds produce abundant biomass in China and have long been used as herbal medicine and food. Their characteristic fatty acid (FA) profiles and related potential function in promoting cardiovascular health (CVH) have not been systematically investigated. In this study, FA profiles of four medicinal Sargassum were characterized using GC-MS. Principal component analysis was used to discriminate the four medicinal Sargassum, and orthogonal projection to latent structures discriminant analysis was carried out between the two official species HAI ZAO and between the two folk medicine species HAI QIAN. In all of the algae investigated, the major SFA and MUFA were palmitic and stearic acid, respectively, while the major PUFAs were linoleic, arachidonic, and eicosapentaenoic acid. S. fusiforme and S. horneri had higher concentrations of PUFAs. With respect to CVH, all of the studied species, particularly S. fusiforme, exhibited satisfactory levels such as PUFA/SFA ratio and n-6/n-3 ratio. Each species possesses a unique FA profile and is discriminated clearly. Potential key FA markers (between the two Chinese official species, and between the two folk species) are assessed. The study provides characteristic fatty acid profiles of four Chinese medicinal Sargassum and their related potential function in promoting CVH.

Productive transcription of the integrated HIV-1 provirus is restricted by cellular factors that inhibit RNA polymerase II elongation. The viral Tat protein overcomes this by recruiting a general elongation factor, P-TEFb, to the TAR RNA element that forms at the 5' end of nascent viral transcripts. P-TEFb exists in multiple complexes in cells, and its core consists of a kinase, Cdk9, and a regulatory subunit, either Cyclin T1 or Cyclin T2. Tat binds directly to Cyclin T1 and thereby targets the Cyclin T1/P-TEFb complex that phosphorylates the CTD of RNA polymerase II and the negative factors that inhibit elongation, resulting in efficient transcriptional elongation. P-TEFb is tightly regulated in cells infected by HIV-1-CD4+ T lymphocytes and monocytes/macrophages. A number of mechanisms have been identified that inhibit P-TEFb in resting CD4+ T lymphocytes and monocytes, including miRNAs that repress Cyclin T1 protein expression and dephosphorylation of residue Thr186 in the Cdk9 T-loop. These repressive mechanisms are overcome upon T cell activation and macrophage differentiation when the permissivity for HIV-1 replication is greatly increased. This review will summarize what is currently known about mechanisms that regulate P-TEFb and how this regulation impacts HIV-1 replication and latency.
