Rare earth element and Yttrium (REY)-rich deep-sea sediments have been identified as significant potential resources. Although grain size analysis is important for the study of deposition environment and the future exploration of REY, related research is limited. Here, we have investigated the grain size and mineral composition of bulk pelagic sediments from Western North Pacific to indicate the evolutions of deposition environment. Grain size analysis results reveal that silt contents are dominant, and the grain size ranges between 20 and 40 μm and can be an additional grain characteristic indicator for REY-rich sediments. Furthermore, the better sorting coefficient of REY-rich sediments reflects the scouring action of bottom currents, which allows the preservation of coarse-grained bioapatite and the removal of clay fraction particles. ΣREY is negatively correlated with clay fractions but positively correlated with sand and silt fractions for the REY-rich sediments, and this is consistent with the dominant distribution of silt-sized biogenic apatite. In addition, grain size distribution (GSD) varies significantly after HCl treatment in the REY-rich sediment. Therefore, for deep-sea sediments collected below the carbonate compensation depth (CCD), an additional presence of a grain size peak at ~20–40 μm and distinct differences in GSD after HCl treatment can serve as important indicators for REY enrichment sediments. Tetrahydroprotoberberines (THPBs) are plant-specific alkaloids with significant medicinal value. They are present in trace amounts in plants and are difficult to chemically synthesize due to stereoselectivity and an unfavorable environment. In this study, a selective methylation strategy was developed for the biocatalysis of seven high-value-added THPB compounds using 4'-O-methyltransferase (Cj4'OMT), norcoclaurine 6-O-methyltransferase (Cj6OMT), and (S)-scoulerine 9-O-methyltransferase (SiSOMT and PsSOMT) in engineered E. coli. The methyltransferases Cj4'OMT, Cj6OMT, PsSOMT, and SiSOMT were expressed heterologously in E. coli. Compound 1 (10-methoxy-2,3,9-tetrahydroxyberbine) was synthesized using the recombinant E. coli strain Cj4'OMT and the substrate 2,3,9,10-tetrahydroxyberbine. Compound 2 (9-methoxy-2,3,10-tetrahydroxyberbine) was produced in the recombinant Escherichia coli (E. coli) strain PsSOMT, and compounds 2 and 3 (discretamine) were produced in the recombinant E. coli strain SiSOMT. Compounds 4 (9,10-methoxy-2,3-tetrahydroxyberbine) and 5 (corypalmine) were obtained by co-culturing the recombinant strains Cj4'OMT and SiSOMT with substrate. Compounds 6 (scoulerine) and 7 (isoscoulerine) were produced by co-culturing the substrate with the recombinant strains Cj4'OMT and Cj6OMT. To increase the yield of novel compound 2, the flask culture conditions of the engineered SiSOMT strain were optimized, resulting in the production of 165.74 mg/L of this compound. Simple Summary: The feeding relationship between copepods and phytoplankton has immense ecological significance. The feeding behavior of copepods may be affected by the ingestion of toxic algae. This study investigated the feeding behavior of copepods via study of the feeding selectivity of Paracalanus parvus, a key small copepod species, using a high-speed camera. The study demonstrates that toxic dinoflagellates alter the feeding behavior of copepods and describes the variations in their feeding response to different algal species and concentrations. The findings provide crucial insights for further studies on the feeding relationship between copepods and phytoplankton and the functional assessment of plankton ecosystems. This study investigated the feeding behavior of copepods by studying the feeding selectivity of Paracalanus parvus, a key small copepod species, using a high-speed camera. The feeding behavior of P. parvus separately fed on three algae, Prorocentrum minimum, Alexandrium minutum, and Thalassiosira weissflogii, was studied at five different concentrations. The factors characterizing feeding behavior, including the beating frequency (BF), beating time (BT), and rejection behavior, were analyzed. The average BT and BF of P. parvus fed on toxic algae were significantly lower than those of copepods fed on nontoxic algae, indicating that the toxic algae negatively affected their feeding behavior. There were no significant differences in feed rejection among the three algae during the short period of experimentation, indicating that the rejection behavior was insignificant in the early period (within 20 min) of feeding on toxic algae. The feeding behavior was inhibited when the concentration reached 250 cells/mL. The BT was initially affected at increasing concentrations followed by the BF, and P. minimum and A. minutum reduced the BF at concentrations of 250 and 1000 cells/mL, respectively. Analysis of the average BFs revealed that P. parvus was more significantly affected by P. minimum containing diarrheal shellfish poison than by A. minutum containing paralytic shellfish poison. The BF of copepods fed on P. minimum was significantly lower than that of copepods fed on A. minutum at 250–500 cells/mL but was not significantly different from that at 1000 cells/mL. This indicated that the inhibitory effect of P. minimum on the feeding behavior was more significant at concentrations observed at the onset of red tide blooms (0.25–0.5 × 102 cells/mL), but insignificant at concentrations reaching those in advanced red tides (>103 cells/mL). Cyanophages play important roles in regulating the population dynamics, community structure, metabolism, and evolution of cyanobacteria in aquatic ecosystems. Here, we report the genomic analysis of an estuarine cyanophage, S-CREM1, which represents a new genus of T4-like cyanomyovirus and exhibits new genetic characteristics. S-CREM1 is a lytic phage which infects estuarine Synechococcus sp. CB0101. In contrast to many cyanomyoviruses that usually have a broad host range, S-CREM1 only infected the original host strain. In addition to cyanophage-featured auxiliary metabolic genes (AMGs), S-CREM1 also contains unique AMGs, including three antitoxin genes, a MoxR family ATPase gene, and a pyrimidine dimer DNA glycosylase gene. The finding of three antitoxin genes in S-CREM1 implies a possible phage control of host cells during infection. One small RNA (sRNA) gene and three cis-regulatory RNA genes in the S-CREM1 genome suggest potential molecular regulations of host metabolism by the phage. In addition, S-CREM1 contains a large number of tRNA genes which may reflect a genomic adaption to the nutrient-rich environment. Our study suggests that we are still far from understanding the viral diversity in nature, and the complicated virus–host interactions remain to be discovered. Cyanobacteria can perform both anoxygenic and oxygenic photosynthesis, a characteristic which ensured that these organisms were crucial in the evolution of the early Earth and the biosphere. Reactive oxygen species (ROS) produced in oxygenic photosynthesis and reactive sulfur species (RSS) produced in anoxygenic photosynthesis are closely related to intracellular redox equilibrium. ROS comprise superoxide anion (O2●−), hydrogen peroxide (H2O2), and hydroxyl radicals (●OH). RSS comprise H2S and sulfane sulfur (persulfide, polysulfide, and S8). Although the sensing mechanism for ROS in cyanobacteria has been explored, that of RSS has not been elucidated. Here, we studied the function of the transcriptional repressor PerR in RSS sensing in Synechococcus sp. PCC7002 (PCC7002). PerR was previously reported to sense ROS; however, our results revealed that it also participated in RSS sensing. PerR repressed the expression of prxI and downregulated the tolerance of PCC7002 to polysulfide (H2Sn). The reporter system indicated that PerR sensed H2Sn. Cys121 of the Cys4:Zn2+ site, which contains four cysteines (Cys121, Cys124, Cys160, and Cys163) bound to one zinc atom, could be modified by H2Sn to Cys121-SSH, as a result of which the zinc atom was released from the site. Moreover, Cys19 could also be modified by polysulfide to Cys19-SSH. Thus, our results reveal that PerR, a representative of the Cys4 zinc finger proteins, senses H2Sn. Ulcerative colitis is a type of non-specific inflammatory bowel disease with unclear etiology. It is considered a progressive disease with risks of bowel motility disorders, anorectal dysfunction, and even colorectal cancer. Commonly used diagnostic markers have poor specificity and cannot predict the development of ulcerative colitis. In this study, 77 serum samples (31 patients, 46 healthy controls) were analyzed using high performance liquid chromatography-quadrupole time-of-flight mass spectrometry and 31 metabolites with significant level changes were found, revealing the relationship of ulcerative colitis to disturbed glutathione metabolism and caffeine metabolism. In addition, pyroglutamic acid, a biomarker of cervical cancer and gastric cancer, was identified with elevated levels in the serum of ulcerative colitis patients. The role of pyroglutamic acid was further analyzed, and the results indicated its positive correlation with the upregulation of inflammatory factors and increased levels of phosphorylated histone H2AX (γH2AX) in IEC-6 cells, which are related to DNA damage. All these results suggest that pyroglutamic acid is not only a biomarker for distinguishing ulcerative colitis status, but that it is also a potential effective metabolite that promotes the transformation of ulcerative colitis to colorectal cancer. Lytic and lysogenic infections are the main strategies used by viruses to interact with microbial hosts. The genetic information of prophages provides insights into the nature of phages and their potential influences on hosts. Here, the siphovirus vB_MoxS-R1 was induced from a Microbacterium strain isolated from an estuarine Synechococcus culture. vB_MoxS-R1 has a high replication capability, with an estimated burst size of 2000 virions per cell. vB_MoxS-R1 represents a novel phage genus-based genomic analysis. Six transcriptional regulator (TR) genes were predicted in the vB_MoxS-R1 genome. Four of these TR genes are involved in stress responses, virulence and amino acid transportation in bacteria, suggesting that they may play roles in regulating the host cell metabolism in response to external environmental changes. A glycerophosphodiester phosphodiesterase gene related to phosphorus acquisition was also identified in the vB_MoxS-R1 genome. The presence of six TR genes and the phosphorus-acquisition gene suggests that prophage vB_MoxS-R1 has the potential to influence survival and adaptation of its host during lysogeny. Possession of four endonuclease genes in the prophage genome suggests that vB_MoxS-R1 is likely involved in DNA recombination or gene conversion and further influences host evolution. Exacerbating deoxygenation is extensively affecting marine organisms, with no exception for phytoplankton. To probe these effects, we comparably explored the growth, cell compositions, photosynthesis, and transcriptome of a diatom Thalassiosira pseudonana under a matrix of pO2 levels and Light:Dark cycles at an optimal growth light. The growth rate (μ) of T. pseudonana under a 8:16 L:D cycle was enhanced by 34% by low pO2 but reduced by 22% by hypoxia. Under a 16:8 L:D cycle, however, the μ decreased with decreasing pO2 level. The cellular Chl a content decreased with decreasing pO2 under a 8:16 L:D cycle, whereas the protein content decreased under a 16:8 L:D cycle. The prolonged photoperiod reduced the Chl a but enhanced the protein contents. The lowered pO2 reduced the maximal PSII photochemical quantum yield (FV/FM), photosynthetic oxygen evolution rate (Pn), and respiration rate (Rd) under the 8:16 or 16:8 L:D cycles. Cellular malondialdehyde (MDA) content and superoxide dismutase (SOD) activity were higher under low pO2 than ambient pO2 or hypoxia. Moreover, the prolonged photoperiod reduced the FV/FM and Pn among all three pO2 levels but enhanced the Rd, MDA, and SOD activity. Transcriptome data showed that most of 26 differentially expressed genes (DEGs) that mainly relate to photosynthesis, respiration, and metabolism were down-regulated by hypoxia, with varying expression degrees between the 8:16 and 16:8 L:D cycles. In addition, our results demonstrated that the positive or negative effect of lowering pO2 upon the growth of diatoms depends on the pO2 level and is mediated by the photoperiod. Ecological engineering by artificial upwelling is considered a promising way to improve water quality. Artificial upwelling could lift nutrient-rich bottom water to the surface, enhance seaweed growth and consequently increase nutrient removal from seawater. However, one of the major obstacles of the engineering application is to determine the suitable position of ecological engineering, which is critical for artificial upwelling's performance. In this paper, potential artificial upwelling positions in a semi-closed bay are simulated by using the unstructured-grid Finite-Volume Coastal Ocean Model (FVCOM). The results show that the upwelling position with relative small tidal current and close to corner will be helpful to increasing nutrient concentration of surface water, and be appropriate to build the ecological engineering. With proper design of the ecological engineering, it is possible to have a noticeable impact in semi-closed bay. The purpose of this work is to construct a tribo-fatigue damage map of high-speed railway wheel material under different tangential forces and contact pressure conditions through JD-1 testing equipment. The results indicate that the wear rate of the wheel material varies with tangential force and contact pressure. The wear mapping of the wheel material is constructed and divided into three regions: slight wear, severe wear, and destructive wear, based on the wear rate under each test condition. With an increase in tangential force and contact pressure, the maximum crack length and average crack length of the wheel material increases. According to the surface damage morphologies and corresponding statistical results of average crack length of wheel material under each experiment condition, a tribo-fatigue damage map is constructed and divided into three regions: slight fatigue damage region, fatigue damage region, and severe fatigue damage region. Fatigue cracks initiate on the wheel specimen surface. Some cracks may propagate into material and fracture under cyclic rolling contact; some cracks may grow into inner material with a certain depth, and then turn toward the surface to form material flaking; some cracks may always propagate parallel to the wheel roller surface. With the rapid development of the energy internet, the transaction of distributed renewable energy (DRE) is playing an increasingly important role in the energy market. However, in the transaction model of distributed renewable energy combined with public blockchain technology, nodes in the trading network can join or leave the network at any time without any permission, which hinders the regulation of electricity institutions. Corresponding to the transaction principle, a distributed renewable energy transaction authentication mechanism based on consortium blockchain is proposed in this paper. First, certificate authority nodes were set in the transaction network to provide nodes with access authority by controlling the public keys and private keys of trading participants so that they can complete their identity authentication. Next, essential chaincodes in the transaction authentication were designed and deployed on a Hyperledger Fabric blockchain site, and a simulation experiment of a simple DRE transaction was used to elaborate the details on the transaction process. Ruscogenin (RUS) is a steroidal sapogenin found in Ruscus aculeatus and Ophiopogon japonicus with several pharmacological activities. In the work reported herein, a novel method termed competitive fluorescence-linked immunosorbent assay (cFLISA) based on monoclonal antibodies (mAbs) coupled with quantum dots (QDs) was developed for the quick and sensitive determination of RUS in biological samples. The mAbs against RUS were conjugated with CdSe/ZnS QDs by the crossing-linking reagents and an indirect cFLISA method was developed. There was a good linear relationship between inhibition efficiency and logarithm concentration of RUS which was varied from 0.1 to 1000 ng/mL. The IC 50 and limit of detection (LOD) were 9.59 ng/mL and 0.016 ng/mL respectively, which much lower than the enzyme-linked immunosorbent assay (ELISA) method. The recoveries in plasma and tissues were ranged from 82.3% to 107.0% and the intra- and inter-day precision values were below 15%. The developed cFLISA has been successfully applied to the measurement of the concentrations of RUS in biological samples of rats, and showed great potential for the tissue distribution study of RUS. Background : Tumor compression-induced pain (TCIP) is a complex pathological cancer pain. Spinal glial cells play a critical role in maintenance of cancer pain by releasing proinflammatory cytokines and chemokines. In this study, we verified the role of levo -corydalmine ( l -CDL) on TCIP. Methods : Spontaneous pain, paw withdrawal threshold and latency were assessed using TCIP mouse model. Immunofluorescence was used to identify the reactions of glia. RT-PCR and western blot or ELISA were used to determine mRNA or protein expression of tumor necrosis factor-α (TNF-α), interlukin-1β (IL-1β), CC chemokine ligand 2 (CCL2) and chemotactic cytokine receptor 2 (CCR2) in vivo and in vitro. Results : l -CDL significantly attenuated TCIP hypersensitivity, accompanying with downregulation of TNF-α and IL-1β expression levels and declined astrocytes and microglial activation. It also significantly decreased the expression of the mRNA and protein level for CCL2 and CCR2. Further, l -CDL could suppress TNF-α-induced astrocytes activation and IL-1β expression through downregulating the CCL2/CCR2. Besides, CCL2-induced BV-microglia activation and inflammatory factors secretion were suppressed by l -CDL via CCR2. Conclusions : Suppression of CCL2/CCR2 by l -CDL may contribute to alleviate TCIP, offering an alternative medication for TCIP., Competing Interests: The authors declare no conflict of interest.

As a network transmission protocol, Networked Transport of RTCM via Internet Protocol (NTRIP) is widely used in GPS and Global Orbiting Navigational Satellite System (GLONASS) Augmentation systems, such as Continuous Operational Reference System (CORS), Wide Area Augmentation System (WAAS) and Satellite Based Augmentation Systems (SBAS). With the deployment of BeiDou Navigation Satellite system(BDS) to serve the Asia-Pacific region, there are increasing needs for ground monitoring of the BeiDou Navigation Satellite system and the development of the high-precision real-time BeiDou products. This paper aims to optimize the decoding algorithm of NTRIP Client data streams and the user authentication strategies of the NTRIP Caster based on NTRIP. The proposed method greatly enhances the handling efficiency and significantly reduces the data transmission delay compared with the Federal Agency for Cartography and Geodesy (BKG) NTRIP. Meanwhile, a transcoding method is proposed to facilitate the data transformation from the BINary EXchange (BINEX) format to the RTCM format. The transformation scheme thus solves the problem of handing real-time data streams from Trimble receivers in the BeiDou Navigation Satellite System indigenously developed by China.
