Due to its designable nanostructure and simple and inexpensive preparation process, electrospun nanofibers have important applications in energy collection, wearable sports health detection, environmental pollutant detection, pollutant filtration and degradation, and other fields. In recent years, a series of polymer-based fiber materials have been prepared using this method, and detailed research and discussion have been conducted on the material structure and performance factors. This article summarizes the effects of preparation parameters, environmental factors, a combination of other methods, and surface modification of electrospinning on the properties of composite nanofibers. Meanwhile, the effects of different collection devices and electrospinning preparation parameters on material properties were compared. Subsequently, it summarized the material structure design and specific applications in wearable device power supply, energy collection, environmental pollutant sensing, air quality detection, air pollution particle filtration, and environmental pollutant degradation. We aim to review the latest developments in electrospinning applications to inspire new energy collection, detection, and pollutant treatment equipment, and achieve the commercial promotion of polymer fibers in the fields of energy and environment. Finally, we have identified some unresolved issues in the detection and treatment of environmental issues with electrospun polymer fibers and proposed some suggestions and new ideas for these issues. Simple Summary: Commercial sand digging initially created suitable nesting habitats to attract blue-tailed bee-eaters but subsequently reduced their breeding success by destroying their nests. However, commercial sand digging can be modified to create suitable and stable nesting habitats for bee-eaters. The development of human society often interferes with wild animals and their natural habitats. Interference during resource exploitation is mostly negative; however, in some cases, it can be positive and even have significance for some species. In this study, we investigated the number of blue-tailed bee-eaters (Merops philippinus), a species under 'state protection category II' in China, between controlled and manipulated nesting habitats from 2017 to 2022. Our results indicated that commercial sand-digging activities, either illegal or approved, initially created suitable nesting habitats to attract blue-tailed bee-eaters but subsequently led to damage of nests or nesting habitats. However, sand digging can be modified by avoiding the breeding season to provide safe and suitable nesting habitats for bee-eaters. The number of breeding birds more than tripled when digging during the breeding season was avoided. We also found that conventional conservation strategies, which strictly prohibited sand-digging activities, did not contribute to the nesting habitats of bee-eaters. Iron-based particles loaded on porous carbon materials have attracted extensive attention as catalysts for denitration and desulfurization reactions. However, the carbon support of a high-temperature denitration catalyst is inevitably oxidized in the presence of H2O and O2. The mechanism of denitration catalyst oxidation and its influence on the catalytic reaction remain to be further explored. Fe2O3-loaded graphene models with carbon vacancy (Gdef), hydroxyl (HyG), and carboxyl (CyG) were constructed to investigate the effects of hydroxylation and carboxylation on the catalytic activity of Fe2O3/graphene for oxidative desulfurization and denitration by using density functional theory (DFT) calculations. According to the analysis of structural properties and adsorption energy, the adsorption process of Fe2O3 on HyG and CyG was observed to have proceeded more favorably than that on Gdef. The density-of-states (DOS) results also affirmed that HyG and CyG promote the electron delocalization of Fe2O3 around the Fermi level, enhancing the chemical activity of Fe2O3. Moreover, adsorption energy analysis indicates that hydroxylation and carboxylation enhanced the adsorption of SO2 and H2O2 on Fe2O3/graphene while also maintaining preferable adsorption stability of NO. Furthermore, mechanistic research explains that adsorbed H2O2 on HyG and CyG directly oxidizes NO and SO2 into HNO2 and H2SO4 following a one-step reaction. Polyacrylamide has promising applications in a wide variety of fields. However, conventional polyacrylamide is prone to hydrolysis and thermal degradation under high temperature conditions, resulting in a decrease in solution viscosity with increasing temperature, which limits its practical effect. Herein, combining molecular dynamics and practical experiments, we explored a facile and fast mixing strategy to enhance the thermal stability of polyacrylamide by adding common poloxamers to form the interpenetrating network hydrogel. The blending model of three synthetic polyacrylamides (cationic, anionic, and nonionic) and poloxamers was first established, and then the interaction process between them was simulated by all-atom molecular dynamics. In the results, it was found that the hydrogen bonding between the amide groups on all polymers and the oxygen-containing groups (ether and hydroxyl groups) on poloxamers is very strong, which may be the key to improve the high temperature resistance of the hydrogel. Subsequent rheological tests also showed that poloxamers can indeed significantly improve the stability and viscosity of nonionic polyacrylamide containing only amide groups at high temperatures and can maintain a high viscosity of 3550 mPa·S at 80 °C. Transmission electron microscopy further showed that the nonionic polyacrylamide/poloxamer mixture further formed an interpenetrating network structure. In addition, the Fourier transform infrared test also proved the existence of strong hydrogen bonding between the two polymers. To adapt to the complex application of polyurethane bearings, it is feasible to improve the tribological performance of single polyurethane-based friction materials through the synergistic effect produced by multi-component-lubricating fillers. In this context, rather than using tungsten disulfide (WS2), which has demonstrated excellent self-lubricating performance as a lubricating oil additive, this paper proposes that WS2 and single-walled carbon nanotubes (SWNTs) can be designed for addition into a polyether 4,4′-diphenylmethane diisocyanate (MDI) polyurethane matrix as self-lubricating fillers so as to explore the synergistic effect of micro- and nano-lubricating fillers on the tribological performance of polyurethane matrix materials. Through a series of characterizations and tests, it was found that the dispersion of two-component-lubricating additives in a polyurethane matrix is improved when the ratio of WS2 to SWNTs is roughly 2:1. In this case, the tribological performance of polyurethane matrix composites is more satisfactory than at other ratios. In addition, compared with the blank sample, the tribological performance of the synergistically modified polyurethane composites under dry friction is more significantly improved with the increase in contact load, while there is no significant improvement under water lubrication. Aside from contributing to the idea of exploring the synergistic effect of WS2 and other micro or nanofillers, this method also opens up the possibility of practically applying WS2 in the field of friction. Simple Summary: Insect-specific viruses (ISVs) are increasingly recognised for their role in causing severe illnesses and mortality in both humans and animals. In this study, the full genome of ISV (Rice thrips ollusvirus 1, RTOV1) was revealed in rice thrips using metatranscriptome sequencing, RT-PCR, and RACE, respectively. RTOV1 has a typical linear G-N-L genome structure of the order Jingchuvirales. Phylogenetic analysis categorised this virus as an Ollusvirus. The infection of host insects with RTOV1 triggered antiviral RNA interference (RNAi), resulting in a significant accumulation of 22-nt virus-derived small interfering RNAs (vsiRNAs), with a notable bias towards the A/U content. Our study provides valuable information on ISVs in thrips that may be useful for pest management. Insects constitute the largest proportion of animals on Earth and act as significant reservoirs and vectors in disease transmission. Rice thrips (Haplothrips aculeatus, family Phlaeothripidae) are one of the most common pests in agriculture. In this study, the full genome sequence of a novel Ollusvirus, provisionally named "Rice thrips ollusvirus 1" (RTOV1), was elucidated using transcriptome sequencing and the rapid amplification of cDNA ends (RACE). A homology search and phylogenetic tree analysis revealed that the newly identified virus is a member of the family Aliusviridae (order Jingchuvirales). The genome of RTOV1 contains four predicted open reading frames (ORFs), including a polymerase protein (L, 7590 nt), a glycoprotein (G, 4206 nt), a nucleocapsid protein (N, 2415 nt) and a small protein of unknown function (291 nt). All of the ORFs are encoded by the complementary genome, suggesting that the virus is a negative-stranded RNA virus. Phylogenetic analysis using polymerase sequences suggested that RTOV1 was closely related to ollusvirus 1. Deep small RNA sequencing analysis reveals a significant accumulation of small RNAs derived from RTOV1, indicating that the virus replicated in the insect. According to our understanding, this is the first report of an Ollusvirus identified in a member of the insect family Phlaeothripidae. Amino acid (AA) analysis is important in biochemistry, food science, and clinical medicine. However, due to intrinsic limitations, AAs usually require derivatization to improve their separation and determination. Here, we present a liquid chromatography-mass spectrometry (LC-MS) method for the derivatization of AAs using the simple agent urea. The reactions proceed quantitatively under a wide range of conditions without any pretreatment steps. Urea-derivatized products (carbamoyl amino acids) of 20 AAs exhibit better separation on reversed-phase columns and increased response in a UV detector compared to underivatized ones. We applied this approach to AA analysis in complex samples using a cell culture media as a model, and it showed potential for the determination of oligopeptides. Galectin-2 is a prototype member of the galactoside-binding galectin family. It is predominately expressed in the gastrointestinal tract but is also detected in several other tissues such as the placenta and in the cardiovascular system. Galectin-2 expression and secretion by epithelial cells has been reported to contribute to the strength of the mucus layer, protect the integrity of epithelia. A number of studies have also suggested the involvement of galectin-2 in tissue inflammation, immune response and cell apoptosis. Alteration of galectin-2 expression occurs in inflammatory bowel disease, coronary artery diseases, rheumatoid arthritis, cancer, and pregnancy disorders and has been shown to be involved in disease pathogenesis. Simple Summary: C1GalT1 is one of the enzymes that catalyze the addition of sugar residues to proteins (protein glycosylation). It specifically controls the synthesis and formation of a special disaccharide structure Galβ1,3GalNAcα-, which occurs predominately in cancer but rarely in normal cells. Recent studies have shown that C1GalT1 is overexpressed in many common cancers including colon, breast, gastric, lung, head and neck, pancreatic, esophageal, prostate, and hepatocellular cancer. C1GalT1 overexpression is also often associated with poorer prognosis and poorer patient survival. This review summarizes our current understanding of the expression of C1GalT1 in various cancers and discusses the impact of C1GalT change on cancer cell activities in cancer development and progression. C1GalT1 (T-synthase) is one of the key glycosyltransferases in the biosynthesis of O-linked mucin-type glycans of glycoproteins. It controls the formation of Core-1 disaccharide Galβ1,3GalNAcα- (Thomsen–Friedenreich oncofetal antigen, T or TF antigen) and Core-1-associated carbohydrate structures. Recent studies have shown that C1GalT1 is overexpressed in many cancers of epithelial origin including colon, breast, gastric, head and neck, pancreatic, esophageal, prostate, and hepatocellular cancer. Overexpression of C1GalT1 is often seen to also be associated with poorer prognosis and poorer patient survival. Change of C1GalT1 expression causes glycosylation changes of many cell membrane glycoproteins including mucin proteins, growth factor receptors, adhesion molecules, and death receptors. This leads to alteration of the interactions of these cell surface molecules with their binding ligands, resulting in changes of cancer cell activity and behaviors. Simple Summary: MMP-13 is an enzyme that can digest and disrupt the tissue structures surrounding epithelial cells that line the surface of many internal organs, as well as the tissue structures surrounding endothelial cells that line the surface of blood vessels. The production of MMP-13 is tightly controlled in physiological conditions but is increased in various cancers and plays multiple roles in tumour progression and metastasis. This review summarises the current understanding of the regulation of MMP-13 production and discusses the actions of MMP-13 in cancer progression and metastasis. Matrix metalloproteinase-13 (MMP-13) is a member of the Matrix metalloproteinases (MMPs) family of endopeptidases. MMP-13 is produced in low amounts and is well-regulated during normal physiological conditions. Its expression and secretion are, however, increased in various cancers, where it plays multiple roles in tumour progression and metastasis. As an interstitial collagenase, MMP-13 can proteolytically cleave not only collagens I, II and III, but also a range of extracellular matrix proteins (ECMs). Its action causes ECM remodelling and often leads to the release of various sequestered growth and angiogenetic factors that promote tumour cell growth, invasion and angiogenesis. In this paper, in order to improve the performance of the ZL205A castings, continuous ultrasonic and pulsed ultrasonic treatments were applied to the melted alloy to study the effect of ultrasound propagation distance on microstructure and microhardness. The results indicated that ZL205A grains were significantly refined by ultrasonic vibration, but the refinement effect became weak gradually with the increase of sampling depth. The minimum grain sizes were 103.2 μm and 122.5 μm respectively in continuous and pulsed ultrasonic treatment. Grain boundary segregation also became more serious and coarser with the increase of vertical depth. In addition, microhardness and vertical depth are not positively correlated linearly. As the vertical depth increased, microhardness first decreased and then increased, the maximum hardnesses were 73.9 HV and 72.84 HV, respectively, in the two process modes. According to the experiment results and available studies, the mechanism of ultrasonic treatment maybe that: the cooling rate of solidification interface front increased by cavitation and streaming, thus changing the solute redistribution behavior of the ZL205A melt. The effect of ultrasonic temperature on density, microstructure and mechanical properties of vacuum counter-pressure casting ZL114A alloy during solidification was investigated by optical microscopy (OM), scanning electron microscope (SEM) and a tensile test. The results show that compared with the traditional vacuum counter-pressure casting aluminum alloy, the primary phase and eutectic silicon of the alloy with ultrasonic treatment has been greatly refined due to the dendrites broken by ultrasonic vibration. However, the refining effect of ultrasonic treatment on vacuum counter-pressure casting aluminum alloy will be significantly affected by ultrasonic temperature. When the ultrasonic temperature increases from 680 °C to 720 °C, the primary phase is gradually refined, and the morphology of eutectic silicon also changes from coarse needle-like flakes to fine short rods. With a further increase in the ultrasonic temperature, the microstructure will coarse again. The tensile strength and elongation of vacuum counter-pressure casting ZL114A alloy increases first and then decreases with the increase of ultrasonic temperature. The optimal mechanical properties were achieved with tensile strength of 327 MPa and the elongation of 5.57% at ultrasonic temperature of 720 °C, which is 6.3% and 8.2%, respectively, higher than that of alloy without ultrasonic treatment. Optimal power flow (OPF) is a crucial tool in the operation and planning of modern power systems. However, as power system optimization shifts towards larger-scale frameworks, and with the growing integration of distributed generations, the computational time and memory requirements of solving the alternating current (AC) OPF problems can increase exponentially with system size, posing computational challenges. In recent years, machine learning (ML) has demonstrated notable advantages in efficient computation and has been extensively applied to tackle OPF challenges. This paper presents five commonly employed OPF transformation techniques that leverage ML, offering a critical overview of the latest applications of advanced ML in solving OPF problems. Simple Summary: The ubiquitin system plays an important role in defense against viruses. In this study, we cloned and characterized the polyubiquitin-C (UBC) gene in Laodelphax striatellus. Expression profile analysis showed that LsUBC was highly expressed in the salivary glands, midgut, and reproductive system. Moreover, the expression of LsUBC was upregulated during viral infection, and inhibition of LsUBC expression increased viral accumulation. These results indicated that LsUBC plays a vital role in inhibiting viral accumulation in L. striatellus. Many hosts utilize the ubiquitin system to defend against viral infection. As a key subunit of the ubiquitin system, the role of polyubiquitin in the viral infection of insects is unclear. Here, we identified the full-length cDNA of the polyubiquitin-C (UBC) gene in Laodelphax striatellus, the small brown planthopper (SBPH). LsUBC was expressed in various tissues and was highly expressed in salivary glands, midgut, and reproductive systems. Furthermore, the LsUBC expression profiles in the developmental stages showed that LsUBC was ubiquitously expressed in seven developmental stages and was highest expressed in female adults with SBPH. qRT-PCR analyses indicated that rice stripe virus (RSV) infection promoted the LsUBC expression. Negeviruses are a proposed group of insect-specific viruses that can be separated into two distinct phylogenetic clades, Nelorpivirus and Sandewavirus. Negeviruses are well-known for their wide geographic distribution and broad host range among hematophagous insects. In this study, the full genomes of two novel negeviruses from each of these clades were identified by RNA extraction and sequencing from a single dungfly (Scathophaga furcata) collected from the Arctic Yellow River Station, where these genomes are the first negeviruses from cold zone regions to be discovered. Nelorpivirus dungfly1 (NVD1) and Sandewavirus dungfly1 (SVD1) have the typical negevirus genome organization and there was a very high coverage of viral transcripts. Small interfering RNAs derived from both viruses were readily detected in S. furcata, clearly showing that negeviruses are targeted by the host antiviral RNA interference (RNAi) pathway. These results and subsequent in silico analysis (studies) of public database and published virome data showed that the hosts of nege-like viruses include insects belonging to many orders as well as various non-insects in addition to the hematophagous insects previously reported. Phylogenetic analysis reveals at least three further groups of negeviruses, as well as several poorly resolved solitary branches, filling in the gaps within the two sub-groups of negeviruses and plant-associated viruses in the Kitaviridae. In order to improve the performance of the investment casting shell, ceramic and nylon fiber was added to the silica sol slurry to study the effect of ceramic and nylon fiber on the liquidity of the silica sol slurry and the bending strength of the investment casting shell. Meanwhile, the fracture surface of shell sample was observed by SEM. The experiment results indicate that the movement viscosity of slurry increases with ceramic and nylon fiber content, increasing from 0 to 0.75 wt.%. The movement viscosity of ceramic fiber composite slurry is higher than nylon fiber composite slurry. The wet and high strength of shell firstly increases then decreases, with an increase of ceramic and nylon fiber content. When the ceramic and nylon fibers' contents are 0.6 wt.%, the maximum wet strengths are 3.56 MPa and 3.84 Mpa respectively, increasing by approximately 38% and 43%. Moreover, the high strength of shell achieves its highest value, 5.08 Mpa, when the ceramic fiber content is 0.6 wt.%; however, when the nylon fiber content was more than 0.3%, the high strength of the nylon fiber reinforced shell was decreased sharply. Therefore, an addition of ceramic and nylon fiber to silica sol slurry distinctly influences the wet and high strength of investment casting shell. [ABSTRACT FROM AUTHOR]

The newest member of the Hepacivirus genus, bovine hepacivirus (BovHepV), was first identified in cattle in 2015 and is a novel hepacivirus C virus (HCV)-like virus. This virus has been detected in five countries so far and is classified into four subtypes. Bovine serum is commonly used for cell cultures and is considered the major source of viral contamination of pharmaceutical products. In this study, bovine serum samples were collected from seven countries located in Asia, America, Oceania, and Europe and were tested for BovHepV RNA using nested PCR, in order to: (i) obtain more knowledge on the geographical distribution and subtypes of BovHepV; and (ii) detect the potential contamination of BovHepV in commercial bovine serum samples used for cell culture propagation. The results demonstrated that bovine serum samples from individual donor cattle in China contained BovHepV RNA. After PCR, sequencing, and assembly, the genomes of the Chinese BovHepV strains were obtained. Genetic analysis of the polyprotein gene revealed a protein identity of <77% and a nucleotide identity of <85% between the Chinese BovHepV strains and all other previously reported BovHepV strains. Using cut-off values for determination of HCV genotypes and subtypes, BovHepV strains worldwide were classified into one unique genotype and seven subtypes. The BovHepV strains identified in the present study were classified into a novel subtype, which was provisionally designated subtype G. The genetic relationships among the different BovHepV subtypes were further confirmed through phylogenetic analysis. Sperm motility and structural integrity are essential for successful fertilization in vivo, and any hindrance of the correct assembly of the axoneme and peri-axonemal structures in the sperm flagellum can lead to fertility problems. While there has been considerable advancement in studying diseases related to the flagellum, the underlying mechanisms that control sperm movement are not yet fully understood. In this study, we reveal that the tetratricopeptide repeat protein 6 (Ttc6) gene, expressed mainly in the testes, plays a crucial role in maintaining male fertility in mice. We further demonstrate that the knockout of Ttc6 in mice results in decreased sperm motility and induces an abnormal circular swimming pattern, consequently leading to male subfertility. Morphological analysis showed an atypical hairpin-like appearance of the spermatozoa, and ultrastructural studies showed unsheathed flagella at the juncture between the midpiece and principal piece. Collectively, these findings suggest that TTC6 plays an essential role in maintaining the stability of the annulus region of the sperm flagellum, thus ensuring the swift and directed motion of sperm. The axoneme and accessory structures of flagella are critical for sperm motility and male fertilization. Sperm production needs precise and highly ordered gene expression to initiate and sustain the many cellular processes that result in mature spermatozoa. Here, we identified a testis enriched gene transmembrane protein 232 (Tmem232), which is essential for the structural integrity of the spermatozoa flagella axoneme. Tmem232 knockout mice were generated for in vivo analyses of its functions in spermatogenesis. Phenotypic analysis showed that deletion of Tmem232 in mice causes male-specific infertility. Transmission electron microscopy together with scanning electron microscopy were applied to analyze the spermatozoa flagella and it was observed that the lack of TMEM232 caused failure of the cytoplasm removal and the absence of the 7th outer microtubule doublet with its corresponding outer dense fiber (ODF). Co-IP assays further identified that TMEM232 interacts with ODF family protein ODF1, which is essential to maintain sperm motility. In conclusion, our findings indicate that TMEM232 is a critical protein for male fertility and sperm motility by regulating sperm cytoplasm removal and maintaining axoneme integrity. Canine influenza virus (CIV) significantly threatens the canine population and public health. Tetherin, an innate immune factor, plays an important role in the defense against pathogen invasion and has been discovered to restrict the release of various enveloped viruses. Two isoforms of canine tetherin (tetherin-X1 and tetherin-X2) were identified in peripheral blood leukocytes of mixed-breed dogs using reverse transcription polymerase chain reaction (RT–PCR). Amino acid alignment revealed that relative to full-length tetherin (tetherin-X1) and truncated canine tetherin (tetherin-X2) exhibited deletion of 34 amino acids. The deletion occurred at the C-terminus of the coiled-coiled ectodomain and the N-terminus of the glycosylphosphatidylinositol (GPI)-anchor domain. Tetherin-X2 was localized subcellularly at the cell membrane, which was consistent with the localization of tetherin-X1. In addition, canine tetherin-X1 and tetherin-X2 restricted the release of H3N2 CIV. However, canine tetherin-X1 had higher antiviral activity than canine tetherin-X2, indicating that the C-terminus of the coiled-coiled ectodomain and the N-terminus of the GPI-anchor domain of canine tetherin (containing the amino acids deleted in tetherin-X2) are critical for its ability to restrict H3N2 CIV release. Simple Summary: The present work focused on the RNA virome analysis of an agricultural pest. In this study, the four novel RNA viruses in A. sinensis were reported. According to the phylogenetic analysis, they were classified as nege-like virus, iflavirus, chu-like virus, and ollusvirus. The complete genome sequence of four RNA viruses were obtained with RACE. Moreover, the analysis of virus-derived small interfering RNAs showed that most of the RNA viruses were targeted by the host antiviral RNA interference pathway. A large number of RNA viruses have been discovered in most insect orders using high-throughput sequencing (HTS) and advanced bioinformatics methods. In this study, an RNA virome of the grasshopper was systematically identified in Atractomorpha sinensis (Orthoptera: Pyrgomorphidae), an important agricultural pest known as the pink-winged grasshopper. These insect viruses were classified as the nege-like virus, iflavirus, ollusvirus, and chu-like virus using HTS and phylogenetic analyses. Meanwhile, the full sequences of four novel RNA viruses were obtained with RACE and named Atractomorpha sinensis nege-like virus 1 (ASNV1), Atractomorpha sinensis iflavirus 1 (ASIV1), Atractomorpha sinensis ollusvirus 1 (ASOV1), and Atractomorpha sinensis chu-like virus 1 (ASCV1), respectively. Moreover, the analysis of virus-derived small interfering RNAs showed that most of the RNA viruses were targeted by the host antiviral RNA interference pathway. Melanoma cell adhesion molecule (MCAM, CD146, MUC18) is a heavily glycosylated transmembrane protein and a marker of melanoma metastasis. It is expressed in advanced primary melanoma and metastasis but rarely in benign naevi or normal melanocytes. More and more evidence has shown that activation of the MCAM on cell surface plays a vital role in melanoma progression and metastasis. However, the natural MCAM binding ligand that initiates MCAM activation in melanoma so far remains elusive. This study revealed that galectin-3, a galactoside-binding protein that is commonly overexpressed in many cancers including melanoma, is naturally associated with MCAM on the surface of both skin and uveal melanoma cells. Binding of galectin-3 to MCAM, via O-linked glycans on the MCAM, induces MCAM dimerization and clustering on cell surface and subsequent activation of downstream AKT signalling. This leads to the increases of a number of important steps in melanoma progression of cell proliferation, adhesion, migration, and invasion. Thus, galectin-3 is a natural binding ligand of MCAM in melanoma, and their interaction activates MCAM and promotes MCAM-mediated melanoma progression. Small ships in remote sensing images have blurred details and are difficult to detect. Existing algorithms usually detect small ships based on predefined anchors with different sizes. However, limited by the number of different sizes, it is difficult for anchor-based methods to match small ships of different sizes and structures during training, as they can easily cause misdetections. In this paper, we propose a hybrid anchor structure to generate region proposals for small ships, so as to take full advantage of both anchor-based methods with high localization accuracy and anchor-free methods with fewer misdetections. To unify the output evaluation and obtain the best output, a label reassignment strategy is proposed, which reassigns the sample labels according to the harmonic intersection-over-union (IoU) before and after regression. In addition, an adaptive feature pyramid structure is proposed to enhance the features of important locations on the feature map, so that the features of small ship targets are more prominent and easier to identify. Moreover, feature super-resolution technology is introduced for the region of interest (RoI) features of small ships to generate super-resolution feature representations with a small computational cost, as well as generative adversarial training to improve the realism of super-resolution features. Based on the super-resolution feature, ship proposals are further classified and regressed by using super-resolution features to obtain more accurate detection results. The sperm flagellum is essential for male fertility. Multiple morphological abnormalities of the sperm flagella (MMAF) is a severe form of asthenoteratozoospermia. MMAF phenotypes are understood to result from pathogenic variants of genes from multiple families including AKAP, DANI, DNAH, RSPH, CCDC, CFAP, TTC, and LRRC, among others. The Leucine-rich repeat protein (LRRC) family includes two members reported to cause MMAF phenotypes: Lrrc6 and Lrrc50. Despite vigorous research towards understanding the pathogenesis of MMAF-related diseases, many genes remain unknown underlying the flagellum biogenesis. Here, we found that Leucine-rich repeat containing 46 (LRRC46) is specifically expressed in the testes of adult mice, and show that LRRC46 is essential for sperm flagellum biogenesis. Both scanning electron microscopy (SEM) and Papanicolaou staining (PS) presents that the knockout of Lrrc46 in mice resulted in typical MMAF phenotypes, including sperm with short, coiled, and irregular flagella. The male KO mice had reduced total sperm counts, impaired sperm motility, and were completely infertile. No reproductive phenotypes were detected in Lrrc46−/− female mice. Immunofluorescence (IF) assays showed that LRRC46 was present throughout the entire flagella of control sperm, albeit with evident concentration at the mid-piece. Transmission electron microscopy (TEM) demonstrated striking flagellar defects with axonemal and mitochondrial sheath malformations. About the important part of the Materials and Methods, SEM and PS were used to observe the typical MMAF-related irregular flagella morphological phenotypes, TEM was used to further inspect the sperm flagellum defects in ultrastructure, and IF was chosen to confirm the location of protein. Our study suggests that LRRC46 is an essential protein for sperm flagellum biogenesis, and its mutations might be associated with MMAF that causes male infertility. Thus, our study provides insights for understanding developmental processes underlying sperm flagellum formation and contribute to further observe the pathogenic genes that cause male infertility. It has recently been claimed that the outstanding performance of machine-learning scoring functions (SFs) is exclusively due to the presence of training complexes with highly similar proteins to those in the test set. Here, we revisit this question using 24 similarity-based training sets, a widely used test set, and four SFs. Three of these SFs employ machine learning instead of the classical linear regression approach of the fourth SF (X-Score which has the best test set performance out of 16 classical SFs). We have found that random forest (RF)-based RF-Score-v3 outperforms X-Score even when 68% of the most similar proteins are removed from the training set. In addition, unlike X-Score, RF-Score-v3 is able to keep learning with an increasing training set size, becoming substantially more predictive than X-Score when the full 1105 complexes are used for training. These results show that machine-learning SFs owe a substantial part of their performance to training on complexes with dissimilar proteins to those in the test set, against what has been previously concluded using the same data. Given that a growing amount of structural and interaction data will be available from academic and industrial sources, this performance gap between machine-learning SFs and classical SFs is expected to enlarge in the future. YABBY gene family plays an important role in the polarity development of lateral organs. We isolated the BraYAB1-702 gene, a member of the YABBY gene family, from young leaves of Chinese cabbage line 06J45. The full-length gene has a 937 bp CDNA sequence and contains an open reading frame (ORF) of 702 bp. The subcellular localization analysis showed that the expression product of the gene was localized in the nucleus. Ectopic expression of BraYAB1-702 in Arabidopsis thaliana caused leaf curling from the adaxial epidermises to abaxial epidermises; the partial abaxialization of the adaxial epidermises of leaves; leaf trichomes and stomata numbers being significantly increased; the plants being severely stunted; the flowering stage being remarkably delayed and inhibiting the development of shoot apical meristem (SAM) with the down-regulation of the expression of SHOOT MERISTEMLESS (STM), Brevipedicellus (BP) and KNAT2 which were related to the development of shoot apical meristem. These results from the present research help to reveal the molecular mechanism of BraYAB1-702 gene in the establishment of adaxial-abaxial polarity of the lateral organs in Chinese cabbage. Radish floral bud abortion (FBA) is an adverse biological phenomenon that occurs during reproduction. Although FBA is a frequent occurrence, its molecular mechanism remains unknown. A transcript-derived fragment (TDF72), which was obtained by cDNA amplified fragment length polymorphism (cDNA-AFLP), was up-regulated in the aborted buds and exhibited 89% sequence homology with the At?VPE gene. In this study, TDF72 was used to clarify the role of VPE in FBA by isolation of the VPE gene RsVPE1 from radish flower buds. The full-length genomic DNA was 2346 bp including nine exons and eight introns. The full-length cDNA was 1825 bp, containing a complete open reading frame (ORF) of 1470 bp, which encoded a predicted protein containing 489 amino acid residues, with a calculated molecular mass of 53.735 kDa. Expression analysis demonstrated that RsVPE1 was expressed in all tested organs of radish at different levels. Highest expression was detected in aborted flower buds, suggesting that RsVPE1 has a role in FBA. In order to analyze the role of RsVPE1 in FBA, RsVPE1 was overexpressed in transgenic Arabidopsis thaliana plants. Aborted flower buds appeared in transgenic plants subjected to heat stress. In addition, RsVPE1 expression in the transgenic plants reached a maximum when subjected to heat stress for 24 h and increased by 2.1-fold to 2.8-fold in three homozygous transgenic lines. These results indicated that RsVPE1 led to FBA when its expression levels exceeded a particular threshold, and provided evidence for the involvement of RsVPE1 in promoting FBA under heat stress. RIG-I functions as a virus sensor that induces a cellular antiviral response. Although it has been investigated in other species, there have been no further studies to date on canine RIG-I against canine influenza virus (CIV). In the present study, we cloned the RIG-I gene of beagle dogs and characterized its expression, subcellular localization, antiviral response, and interactions with CIV proteins. RIG-I was highly expressed and mainly localized in the cytoplasm, with low levels detected in the nucleus. The results revealed that overexpression of the CARD domain of RIG-I and knockdown of RIG-I showed its ability to activate the RLR pathway and induced the expression of downstream interferon-stimulated genes. Moreover, overexpression of canine RIG-I suppressed the replication of CIV. The association between RIG-I and CIV was evaluated with the luciferase assay and by indirect immunofluorescence and bimolecular fluorescence complementation analyses. The results showed that CIV nonstructural protein 1 (NS1) can strongly suppress the RIG-I–mediated innate immune response, and the novel interactions between CIV matrix proteins (M1 and M2) and canine RIG-I were disclosed. These findings provide a basis for investigating the antiviral mechanism of canine RIG-I against CIV, which can lead to effective strategies for preventing CIV infection in dogs. Severe fever with thrombocytopenia syndrome virus (SFTSV) is an emerging tick-borne bunyavirus in Asia that causes severe disease. Despite its clinical importance, treatment options for SFTSV infection remains limited. The SFTSV glycoprotein Gn plays a major role in mediating virus entry into host cells and is therefore a potential antiviral target. In this study, we employed an in silico structure-based strategy to design novel cyclic antiviral peptides that target the SFTSV glycoprotein Gn. Among the cyclic peptides, HKU-P1 potently neutralizes the SFTSV virion. Combinatorial treatment with HKU-P1 and the broad-spectrum viral RNA-dependent RNA polymerase inhibitor favipiravir exhibited synergistic antiviral effects in vitro. Atomic valence state regulation is an advantageous approach for improving photocatalytic efficiency and product selectivity. However, it is difficult to precisely control the ratio of the different valence states on the surface and the relationship between the surface valence change and catalytic efficiency in the photocatalytic reaction process is unclear. Herein, CeVO4 ultrathin nanosheets were fabricated by one-step solvothermal method with ethanolamine (MEA) as the structure-directing agent. The ratio of the concentrations of intrinsic Ce4+ and Ce3+ ions is precisely modulated from 19.82:100 to 13.33:100 changed by the volume of MEA added without morphology modification. The photocatalytic efficiency increases as the concentrations of intrinsic Ce4+ and Ce3+ ions decrease and CV3 (prepared with 3 mL of MEA) shows the highest CO generation rate approximately 6 and 14 times larger than CV (prepared without MEA) and CV1 (prepared with 1 mL of MEA), respectively, in the photocatalytic CO2 reduction. Interestingly, about 6.8% photo-induced Ce4+ ions were generated on the surface of the catalysts during the photocatalytic CO2 reduction without any phase and morphology changes for CV3. Drought limits the growth and productivity of plants. Reproductive development is sensitive to drought but the underlying physiological and molecular mechanisms remain unclear in tomatoes. Here, we investigated the effect of drought on tomato floral development using morpho-physiological and transcriptome analyses. Drought-induced male sterility through abnormal anther development includes pollen abortion, inadequate pollen starch accumulation and anther indehiscence which caused floral bud and opened flower abortions and reduced fruit set/yield. Under drought stress (DS), pollen mother cell to meiotic (PMC-MEI) anthers survived whereas tetrad to vacuolated uninucleate microspore (TED-VUM) anthers aborted. PMC-MEI anthers had lower ABA increase, reduced IAA and elevated sugar contents under DS relative to well-watered tomato plants. However, TED-VUM anthers had higher ABA increase and IAA levels, and lower accumulation of soluble sugars, indicating abnormal carbohydrate and hormone metabolisms when exposed to drought-stress conditions. Moreover, RNA-Seq analysis identified altogether >15,000 differentially expressed genes that were assigned to multiple pathways, suggesting that tomato anthers utilize complicated mechanisms to cope with drought. In particular, we found that tapetum development and ABA homeostasis genes were drought-induced while sugar utilization and IAA metabolic genes were drought-repressed in PMC-MEI anthers. Our results suggest an important role of phytohormones metabolisms in anther development under DS and provide novel insight into the molecular mechanism underlying drought resistance in tomatoes. A better understanding of the mechanism of primordial follicle activation will help us better understand the causes of premature ovarian insufficiency (POI), and will help us identify new drugs that can be applied to the clinical treatment of infertility. In this study, single oocytes were isolated from primordial and primary follicles, and were used for gene profiling with TaqMan array cards. Bioinformatics analysis was performed on the gene expression data, and Ingenuity Pathway Analysis was used to analyze and predict drugs that affect follicle activation. An ovarian in vitro culture system was used to verify the function of the drug candidates, and we found that curcumin maintains the ovarian reserve. Long-term treatment with 100 mg/kg curcumin improved the ovarian reserve indicators of AMH, FSH, and estradiol in aging mice. Mechanistic studies show that curcumin can affect the translocation of FOXO3, thereby inhibiting the PTEN-AKT-FOXO3a pathway and protecting primordial follicles from overactivation. Semantic segmentation has been widely used in the basic task of extracting information from images. Despite this progress, there are still two challenges: (1) it is difficult for a single-size receptive field to acquire sufficiently strong representational features, and (2) the traditional encoder-decoder structure directly integrates the shallow features with the deep features. However, due to the small number of network layers that shallow features pass through, the feature representation ability is weak, and noise information will be introduced to affect the segmentation performance. In this paper, an Adaptive Multi-Scale Module (AMSM) and Adaptive Fuse Module (AFM) are proposed to solve these two problems. AMSM adopts the idea of channel and spatial attention and adaptively fuses three-channel branches by setting branching structures with different void rates, and flexibly generates weights according to the content of the image. AFM uses deep feature maps to filter shallow feature maps and obtains the weight of deep and shallow feature maps to filter noise information in shallow feature maps effectively. Based on these two symmetrical modules, we have carried out extensive experiments. On the ISPRS Vaihingen dataset, the F1-score and Overall Accuracy (OA) reached 86.79% and 88.35%, respectively. Anthracnose caused by Colletotrichum acutatum is one of the most devastating fungal diseases of pepper (Capsicum annuum L.). The utilization of chitin-binding proteins or chitinase genes is the best option to control this disease. A chitin-binding domain (CBD) has been shown to be crucial for the innate immunity of plants and activates the hypersensitive response (HR). The CaChiIII7 chitinase gene has been identified and isolated from pepper plants. CaChiIII7 has repeated CBDs that encode a chitinase enzyme that is transcriptionally stimulated by C. acutatum infection. The knockdown of CaChiIII7 in pepper plants confers increased hypersensitivity to C. acutatum, resulting in its proliferation in infected leaves and an attenuation of the defense response genes CaPR1, CaPR5, and SAR8.2 in the CaChiIII7-silenced pepper plants. Additionally, H2O2 accumulation, conductivity, proline biosynthesis, and root activity were distinctly reduced in CaChiIII7-silenced plants. Subcellular localization analyses indicated that the CaChiIII7 protein is located in the plasma membrane and cytoplasm of plant cells. The transient expression of CaChiIII7 increases the basal resistance to C. acutatum by significantly expressing several defense response genes and the HR in pepper leaves, accompanied by an induction of H2O2 biosynthesis.
