Traditional varieties are a valuable tool in modern apple breeding. However, the use of synonyms and missing source documentation hinder an effective identification and conservation of relevant cultivars. During several projects, the authenticity and diversity of the apple cultivar collection of the German Fruit Genebank (GFG) was evaluated extensively. The trueness-to-type of 7890 apple trees was assessed on a pomological and molecular level. Pomological evaluations were performed by at least two experienced experts to identify the original cultivar names. On the molecular level, a set of 17 SSR markers was used to determine a unique genetic profile for each apple cultivar. The pomological and molecular characterization was expressed in terms of a comprehensive trueness-to-type criterion and the results were previously published as a well-curated dataset. In this study, the published dataset was analyzed to evaluate the quality and diversity of the apple collection of the GFG and highlight new findings based on phylogenetic and parentage analysis. The dataset contains 1404 unique genetic profiles corresponding to unambiguous cultivar names. Of these 1404 cultivars, 74% were assessed as true-to-type. The collection of diploid apple cultivars showed a high degree of expected heterozygosity (Hexp = 0.84). Genetic diversity in terms of year and location of origin was investigated with a STRUCTURE analysis. It was hypothesized that genetic diversity might decline overtime due to restrictive breeding programs. The results showed a shift dynamic between older and newer cultivars in one specific cluster, but no significant decrease in genetic diversity was observed in this study. Lastly, a parentage analysis was performed to check parental relationships based on historical research. Out of 128 parent–child trios, 110 trios resulted in significant relationships and reconfirmed the information from the literature. In some cases, the information from the literature was disproven. This analysis also allowed for readjusting the trueness-to-type criteria for previously undetermined cultivars. Overall, the importance of authenticity evaluations for gene bank cultivars was highlighted. Among fruits, the apple is unique for producing large amounts of the dihydrochalcone phloridzin, which, together with phloretin, its aglycone, is valuable to the pharmaceutical and food industries for its antidiabetic, antioxidant, and anticarcinogenic properties, as well as its use as a sweetener. We analysed the phloridzin concentration, total phenolic content, and antioxidant activity in the peel, flesh, seeds, juice, and pomace of 13 international and local apple varieties. In the unprocessed fruit, the seeds had the highest phloridzin content, while the highest total phenolic contents were mostly found in the peel. In processed samples, phloridzin and the total phenolic compounds especially were higher mostly in juice than in pomace. Moreover, the total phenolic content was much higher than the phloridzin content. Juice showed the highest antioxidant activity, followed by the peel and flesh. Across all samples, antioxidant activity did not directly correlate with phloridzin concentrations, suggesting that the antioxidant activity ascribed to phloridzin may need re-evaluation. In the Ferric Reducing Antioxidant Power (FRAP) assay, phloridzin only showed antioxidant activity at high concentrations when compared to its aglycone, phloretin. Considering the large amounts of apple juice produced by the juice industry, residual pomace is a promising source of phloridzin. The apple tree (Malus × domestica Borkh.) belongs to the Rosaceae. Due to its adaptability and tolerance to different soil and climatic conditions, it is cultivated worldwide for fresh consumption. The priorities of apple growers are high-quality fruits and stable yield for high production. About 90 to 95 percent of fruits should fall or be eliminated from apple trees to avoid overcropping and poor-quality fruits. Apple trees engage in a complex biological process known as yield self-regulation, which is influenced by several internal and external factors. Apple buds develop in different stages along the branches, and they can potentially give rise to new shoots, leaves, flowers, or fruit clusters. The apple genotype determines how many buds will develop into fruit-bearing structures and the capacity for yield self-regulation. Plant hormones such as ethylene, cytokinins, auxins, and gibberellins play a crucial role in regulating the fruit set, growth, and development, and the balance of these hormones influences the flowering intensity, fruit size, and fruit number on the apple tree. Apple growers often interfere in the self-regulation process by manually thinning fruit clusters. Different thinning methods, such as by hand, mechanical thinning, or applying chemical substances, are used for flower and fruit thinning. The most profitable in commercial orchards is the use of chemicals for elimination, but more environmentally sustainable solutions are required due to the European Green Deal. This review focuses on the biological factors and genetic mechanisms in apple yield self-regulation for a better understanding of the regulatory mechanism of fruitlet abscission for future breeding programs targeted at self-regulating yield apple varieties. White haze, an emerging disorder caused by extensive fungal growth on the apple surface, results in a compromised fruit quality and decreased marketability. The use of biological control agents could be an interesting tool to reduce its development. This work aimed to test the efficacy of two Aureobasidium pullulans strains (AP2 and PL5) in controlling white haze on stored apples. An in vivo trial was conducted by inoculating fruits with white haze causal agents (Entyloma belangeri, Golubevia pallescens, Tilletiopsis washingtonensis) and treating them with the antagonistic yeasts. Three commercial biofungicides were also included in the trial. Both A. pullulans strains, along with the Metschnikowia fructicola-based product, reduced white haze incidence after 110 days of storage at 1 ± 1 °C and after 7 days of shelf life. Furthermore, the effect of A. pullulans application on the fruit fungal microbiome was assessed. A significant impact of apple matrix and treatment on the mycobiome composition was observed. Analyses showed a good colonization of A. pullulans on the treated apples, both epiphytically and endophytically. A decrease in white haze-related fungi abundance was observed in the treated fruits. Foliar fertilizing is very important to supply apple plants with calcium and micronutrients. The most cost-effective approach to this is the application of the fertilizers in tank mixtures with plant protection chemicals. Plasma-activated water (PAW) has great potential for the use in the agriculture. We used two type of PAWs, PAW1 (made using underwater electrical discharge in an aqueous KNO3 solution and includes reactive nitrogen species and platinum nanoparticles) and PAW2 (made using a plasma torch with nitrogen gas makeup and contains reactive nitrogen species but not metals). We studied the impact of two PAW types on the contents of Ca, B, Mn, Fe, and Co in leaves and Ca, Mn, Fe, Zn, and Mo in fruits sprayed with tank mixtures containing the fertilizers. We also tested the efficiency of PAW in the control of apple scab when applied as tank mixtures with plant protection chemicals. The application of the PAWs significantly increased foliar Ca when the PAW was mixed with Ca-containing formulations (spraying PAW1 containing Ca increased leaf Ca by up to 21%, and PAW2 up by to 9% compared to Ca spraying without PAW). The largest fruit Ca increase was in the variant treated with PAW1 with a micronutrient spraying program (up to 143%). The PAW treatments enhanced the baseline mineral contents of the plants even when they were not sprayed with the nutrients. PAW1 mainly increased the nutrient contents of the apple fruits. PAWs have proven to be efficient for the control of apple scab, thereby reducing the demand for fungicides. The scab damage to the leaves and fruits was similar in plants treated with PAWs without fungicides (1.7–1.9% on the leaves and 1.6–1.8% on the fruits) compared to the conventional chemical scab control (0.9% leaves and 0.6% fruits) and was significantly lower than in the untreated control (9.3% on leaves and 11.9 on fruits). The weather variations around the world are already having a profound impact on agricultural production. This impacts apple production and the quality of the product. Through agricultural precision, growers attempt to optimize both yield and fruit size and quality. Two experiments were conducted using field-grown "Gala" apple trees in Geneva, NY, USA, in 2021 and 2022. Mature apple trees (Malus × domestica Borkh. cv. Ultima "Gala") grafted onto G.11 rootstock planted in 2015 were used for the experiment. Our goal was to establish a relationship between stem water potential (Ψtrunk), which was continuously measured using microtensiometers, and the growth rate of apple fruits, measured continuously using dendrometers throughout the growing season. The second objective was to develop thresholds for Ψtrunk to determine when to irrigate apple trees. The economic impacts of different irrigation regimes were evaluated. Three different water regimes were compared (full irrigation, rainfed and rain exclusion to induce water stress). Trees subjected the rain-exclusion treatment were not irrigated during the whole season, except in the spring (April and May; 126 mm in 2021 and 100 mm in 2022); that is, these trees did not receive water during June, July, August and half of September. Trees subjected to the rainfed treatment received only rainwater (515 mm in 2021 and 382 mm in 2022). The fully irrigated trees received rain but were also irrigated by drip irrigation (515 mm in 2021 and 565 mm in 2022). Moreover, all trees received the same amount of water out of season in autumn and winter (245 mm in 2021 and 283 mm in 2022). The microtensiometer sensors detected differences in Ψtrunk among our treatments over the entire growing season. In both years, experimental trees with the same trunk cross-section area (TCSA) were selected (23–25 cm−2 TCSA), and crop load was adjusted to 7 fruits·cm−2 TCSA in 2021 and 8.5 fruits·cm−2 TCSA in 2022. However, the irrigated trees showed the highest fruit growth rates and final fruit weight (157 g and 70 mm), followed by the rainfed only treatment (132 g and 66 mm), while the rain-exclusion treatment had the lowest fruit growth rate and final fruit size (107 g and 61 mm). The hourly fruit shrinking and swelling rate (mm·h−1) measured with dendrometers and the hourly Ψtrunk (bar) measured with microtensiometers were correlated. We developed a logistic model to correlate Ψtrunk and fruit growth rate (g·h−1), which suggested a critical value of −9.7 bars for Ψtrunk, above which there were no negative effects on fruit growth rate due to water stress in the relatively humid conditions of New York State. A support vector machine model and a multiple regression model were developed to predict daytime hourly Ψtrunk with radiation and VPD as input variables. Yield and fruit size were converted to crop value, which showed that managing water stress with irrigation during dry periods improved crop value in the humid climate of New York State. MdPG1 encoding polygalacturonase in apple (Malus × domestica) is a key gene associated with fruit firmness and texture variations among apple cultivars. However, the causative variants of MdPG1 are still not known. In this study, we identified a SNPA/C variant within an ERF-binding element located in the promoter region of MdPG1. The promoter containing the ERF-binding element with SNPA, rather than the SNPC, could be strongly bound and activated by MdCBF2, a member of the AP2/ERF transcription factor family, as determined by yeast-one-hybrid and dual-luciferase reporter assays. We also demonstrated that the presence of a novel long non-coding RNA, lncRNAPG1, in the promoter of MdPG1 was a causative variant. lncRNAPG1 was specifically expressed in fruit tissues postharvest. lncRNAPG1 could reduce promoter activity when it was fused to the promoter of MdPG1 and a tobacco gene encoding Mg-chelatase H subunit (NtCHLH) in transgenic tobacco cells but could not reduce promoter activity when it was supplied in a separate gene construct, indicating a cis-regulatory effect. The 52 members of the Teosinte-Branched 1/Cycloidea/Proliferating Cell Factors (TCP) Transcription Factor gene family in Malus × domestica (M. × domestica) were identified in 2014 on the first genome assembly, which was released in 2010. In 2017, a higher quality genome assembly for apple was released and is now considered to be the reference genome. Moreover, as in several other species, the identified TCP genes were named based on the relative position of the genes on the chromosomes. The present work consists of an update of the TCP gene family based on the latest genome assembly of M. × domestica. Compared to the previous classification, the number of TCP genes decreased from 52 to 40 as a result of the addition of three sequences and the deduction of 15. An analysis of the intragenic identity led to the identification of 15 pairs of orthologs, shedding light on the forces that shaped the evolution of this gene family. Furthermore, a revised nomenclature system is proposed that is based both on the intragenic identity and the homology with Arabidopsis thaliana (A. thaliana) TCPs in an effort to set a common standard for the TCP classification that will facilitate any future interspecific analysis. Red fleshed apples (Malus × domestica Borkh.) differ in colour intensity between cultivars, seasons and sites. The objective of this study was to develop a procedure for predicting anthocyanin content from digital images of flesh discs. Flesh cylinders of uniform colour were excised, scanned and their colours determined in the R, G and B and the L*a*b* colour spaces. Anthocyanin content was also quantified chemically. A calibration line was constructed to predict anthocyanin content of flesh discs of varying colour from a scan or a photograph in the studio or outdoors. Anthocyanin concentration was linearly related to the logarithms of G, B and L*. From these relationships, the anthocyanin content of a flesh disc was predicted, pixel by pixel. Colour corrections were applied using a reference colour chart included in all images. The Finlayson algorithm was most effective for correcting the G parameter obtained by a flatbed scanner. For variable imaging methods (scanning or photography), the Vandermonde algorithm for correcting the L* parameter and the Finlayson algorithm for correcting the G parameter were most effective in predicting anthocyanin content. Regulatory compliance of experimental releases into the environment of not yet approved genetically modified plants often requires implementation of measures to reduce the dispersal of reproductive material. To study the impact of nets on pollen flow in an experimental field site in Switzerland, non-GM apple varieties 'Ladina' and 'Nicogreen' were planted inside and outside a netted plot, respectively. Seeds harvested from mature fruits were germinated and the paternal variety of the seedlings was determined using simple sequence repeat (SSR) molecular markers. We demonstrate that pollination frequency from trees inside the netted plot to trees outside over a two-year (2018 and 2019) study of 4500 seedlings is 0.6% (26 seedlings). Moreover, these outcrossing events decreased with increasing distance from the pollen donor. Over the study period, we found on average 0.9%, 0.5%, 0.4% and 0.09% of the seedlings derived from apples of 'Nicogreen' trees at 8 m, 15 m, 72 m and 117 m being generated by fertilisations of 'Ladina' pollen, respectively. In comparison, 48.3% (2018 season) and 75.1% (2019 season) of examined 'Ladina' seedlings in the netted plot originated from 'Nicogreen' tree pollen outside the netted plot. The results suggest that insect netting is effective in minimizing egress of apple pollen from an experimental site and that the likelihood of outcrossing is reduced further (<0.1%) when there are no compatible apple trees within a radius of 100 m of the pollen donor. The Annurca type is an historical Italian apple fruit probably native to the Campania region. These fruits are harvested before a full maturity stage and go through an open-field reddening process. Products based on 1-methylcyclopropene (1-MCP) are widely used in postharvest to improve apple quality and avoid disorders in controlled atmosphere. In this work, we tested the impact of 1-MCP in the traditional reddening system of Annurca type by analyzing three postharvest strategies, namely, fruits exposed for ten days to the traditional reddening and then cold-stored (RDG treatment); fruits treated in postharvest with 1-MCP and then cold-stored (MCP treatment); fruits treated in postharvest with 1-MCP, exposed for ten days to the open-field reddening and then cold-stored (MCP + RDG treatment). Using a full factorial design, we measured main physical and compositional traits and described the properties of the products by consumer testing at two storage times. The MCP + RDG treatment provided the most valuable results, by positively affecting fruit flesh firmness and several sensorial attributes (e.g., hardness, crunchiness, juiciness, sweetness), including a reduction of the mealiness. At the end of the cold-storage, MCP + RDG apples scored a higher consumer's overall liking. The work demonstrated that the 1-MCP technology can be integrated into a traditional open-field postharvest process to improve fruit quality and, potentially, to extend the shelf life of Annurca apples. Polyphenols within fruits and vegetables may contribute to health benefits due to their consumption, with the anthocyanin sub-set also adding colour. The Lemonade™ apple variety has green skin and white flesh, with low anthocyanin content, while some apple varieties have high anthocyanin content in both the skin and flesh. Effects of red compared with white-fleshed apples were studied in healthy human subjects in a randomized, placebo-controlled, cross-over intervention trial. Twenty-five healthy subjects consumed dried daily portions of the red-fleshed or placebo (white-fleshed) apple for two weeks, followed by one-week washout and further two-week crossover period. During the study, volunteers provided faecal samples for microbiota composition analysis and blood samples for peripheral blood mononuclear cell (PBMC) gene expression analysis. Subtle differences were observed in the faecal microbiota of subjects that were fed the different apples, with significant (p < 0.05) reductions in relative abundances of Streptococcus, Ruminococcus, Blautia, and Roseburia, and increased relative abundances of Sutterella, Butyricicoccus, and Lactobacillus in subjects after consuming the red apple. Changes in PBMC gene expression showed 18 mRNA transcripts were differentially expressed between the two groups, of which 16 were immunoglobulin related genes. Pathway analysis showed that these genes had roles in pathways such as immunoglobulin production, B cell-mediated immunity, complement activation, and phagocytosis. In conclusion, this study shows that anthocyanin-rich apples may influence immune function compared to control apples, with changes potentially associated with differences in the faecal microbiota. An intact skin is essential in high-quality apples. Ongoing deposition of cuticular material during fruit development may decrease microcracking. Our objective was to establish a system for quantifying cutin and wax deposition in developing apple fruit. Oleic acid (13C and 14C labelled) and palmitic acid (14C labelled) were fed to developing apples and the amounts incorporated in the cutin and wax fractions were quantified. The incorporation of 14C oleic acid (C18) was significantly higher than that of 14C palmitic acid (C16) and the incorporation in the cutin fraction exceeded that in the wax fraction. The amount of precursor incorporated in the cutin increased asymptotically with time, but the amount in the wax fraction remained about constant. Increasing the concentration of the precursor applied generally increased incorporation. Incorporation in the cutin fraction was high during early development (43 days after full bloom) and decreased towards maturity. Incorporation was higher from a dilute donor solution (infinite dose feeding) than from a donor solution subjected to drying (finite dose feeding) or from perfusion of the precursor by injection. Feeding the skin of a developing apple with oleic acid resulted in significant incorporation in the cutin fraction under both laboratory and field conditions. [ABSTRACT FROM AUTHOR]

Among the fungal diseases of apple trees, serious yield losses are due to an apple scab caused by Venturia inaequalis. Protection against this disease is based mainly on chemical treatments, which are currently very limited. Therefore, it is extremely important to introduce cultivars with reduced susceptibility to this pathogen. One of the important sources of variability for breeding is the process of polyploidization. Newly obtained polyploids may acquire new features, including increased resistance to diseases. In our earlier studies, numerous tetraploids have been obtained for several apple cultivars with 'Free Redstar' tetraploids manifesting enhanced resistance to apple scab. In the present study, tetraploids of 'Free Redstar' were assessed in terms of phenotype and genotype with particular emphasis on the genetic background of their increased resistance to apple scab. Compared to diploid plants, tetraploids (own-rooted plants) were characterized with poor growth, especially during first growing season. They had considerably shorter shoots, fewer branches, smaller stem diameter, and reshaped leaves. In contrast to own-rooted plants, in M9-grafted three-year old trees, no significant differences between diplo- and tetraploids were observed, either in morphological or physiological parameters, with the exceptions of the increased leaf thickness and chlorophyll content recorded in tetraploids. Significant differences between sibling tetraploid clones were recorded, particularly in leaf shape and some physiological parameters. The amplified fragment length polymorphism (AFLP) analysis confirmed genetic polymorphism of tetraploid clones. Methylation-sensitive amplification polymorphism (MSAP) analysis showed that the level of DNA methylation was twice as high in young tetraploid plants as in a diploid donor tree, which may explain the weaker vigour of neotetraploids in the early period of their growth in the juvenile phase. Molecular analysis showed that 'Free Redstar' cultivar and their tetraploids bear six Rvi genes (Rvi5, Rvi6, Rvi8, Rvi11, Rvi14 and Rvi17). Transcriptome analysis confirmed enhanced resistance to apple scab of 'Free Redstar' tetraploids since the expression levels of genes related to resistance were strongly enhanced in tetraploids compared to their diploid counterparts. Exposure of the fruit surface to moisture during early development is causal in russeting of apple (Malus × domestica Borkh.). Moisture exposure results in formation of microcracks and decreased cuticle thickness. Periderm differentiation begins in the hypodermis, but only after discontinuation of moisture exposure. Expressions of selected genes involved in cutin, wax and suberin synthesis were quantified, as were the wax, cutin and suberin compositions. Experiments were conducted in two phases. In Phase I (31 days after full bloom) the fruit surface was exposed to moisture for 6 or 12 d. Phase II was after moisture exposure had been discontinued. Unexposed areas on the same fruit served as unexposed controls. During Phase I, cutin and wax synthesis genes were down-regulated only in the moisture-exposed patches. During Phase II, suberin synthesis genes were up-regulated only in the moisture-exposed patches. The expressions of cutin and wax genes in the moisture-exposed patches increased slightly during Phase II, but the levels of expression were much lower than in the control patches. Amounts and compositions of cutin, wax and suberin were consistent with the gene expressions. Thus, moisture-induced russet is a two-step process: moisture exposure reduces cutin and wax synthesis, moisture removal triggers suberin synthesis. [ABSTRACT FROM AUTHOR]

Russeting is an economically important surface disorder in apple (Malus × domestica Borkh). Indirect evidence suggests an irregular skin structure may be the cause of the phenomenon. The objective of this study was to characterize epidermal and hypodermal cell morphology and the mechanical properties of the skins of apple cultivars of differing russet susceptibility. Dimensions of epidermal and hypodermal cells were determined using microscopy. Stiffness (S), maximum force ( F m a x ), and maximum strain ( ε m a x ) at failure were quantified using uniaxial tensile tests of skin strips. Particularly during early fruit development, epidermal cells (EC) and hypodermal cells (HC) in russet non-susceptible cultivars occurred in greater numbers per unit area than in russet-susceptible ones. The EC and HC were lower in height, shorter in length, and of reduced tangential surface area. There were little differences in S or F m a x between non-susceptible and susceptible cultivars. However, the ε m a x were higher for the skins of non-susceptible cultivars, than for those of susceptible ones. This difference was larger for the young than for the later growth stages. It is concluded that russet-susceptible cultivars generally have larger cells and a wider distribution of cell sizes for both EC and HC. These result in decreased ε m a x for the skin during early fruit development when russet susceptibility is high. This increases the chances of skin failures which is known to trigger russeting. Nutritional unbalances, such as calcium deficiency at the fruit level, are generally the causative agent of post-harvest disorders in apples. Foliar application of Ca as calcium chloride is the current solution to increase Ca concentration in apples, even though the effectiveness of this approach is often not satisfactory. In this research, we tested the efficacy of a combined application of Ca with selected biostimulants to improve apple quality and to reduce the incidence of storage disorders. The experiment was conducted in two "Jonathan" apple orchards that differed in management systems and characteristics. Tree canopies were sprayed with calcium chloride alone and in combination with a commercial product containing zinc and silicon or a seaweed extract. The seaweed extract increased apple quality by boosting the reddish coloration (+32% of color index) and by enhancing final anthocyanin concentration of fruit skin. Both biostimulants significantly reduced (by 20%) the incidence of the physiological disorder, known as "Jonathan spot", after 160 days of storage. Increased concentration of nutrients (Ca, Zn, and Mn) in the skin of apples after biostimulant applications, together with changes of the phenolic profile during the storage, are discussed as the possible causes of the reduced fruit susceptibility to post-harvest disorders. The effects of harvest time, 1-methylcyclopropene (1-MCP) and air storage time on the susceptibility of flesh browning in 'Empire' apples were studied during a seven-day shelf life period after air storage at 0.5 °C for seven months. Early- or late-harvested 'Empire' apples without 1-MCP increased production of ethylene, respiration rates and internal ethylene concentration during the shelf life. Respiration rates increased in the late-harvested fruit for the shelf life period with/without 1-MCP treatment. The 1-MCP-treated fruit was approximately 10 N firmer than fruit not treated with 1-MCP at an early harvest for the entire shelf life duration. Peroxidase activity and percent change in electrical conductivity in the flesh were elevated in late-harvested fruit for the shelf life duration, whereas polyphenol oxidase activities were found to be stimulated by 1-MCP treatment, regardless of harvest time. Fungal storage rots like blue mould, grey mould, bull's eye rot, bitter rot and brown rot destroy large amounts of the harvested apple crop around the world. Application of fungicides is nowadays severely restricted in many countries and production systems, and these problems are therefore likely to increase. Considerable variation among apple cultivars in resistance/susceptibility has been reported, suggesting that efficient defence mechanisms can be selected for and used in plant breeding. These are, however, likely to vary between pathogens, since some fungi are mainly wound-mediated while others attack through lenticels or by infecting blossoms. Since mature fruits are considerably more susceptible than immature fruits, mechanisms involving fruit-ripening processes are likely to play an important role. Significant associations have been detected between the susceptibility to rots in harvested fruit and various fruit maturation-related traits like ripening time, fruit firmness at harvest and rate of fruit softening during storage, as well as fruit biochemical contents like acidity, sugars and polyphenols. Some sources of resistance to blue mould have been described, but more research is needed on the development of spore inoculation methods that produce reproducible data and can be used for large screenings, especially for lenticel-infecting fungi. Grafting can improve the agricultural traits of crop plants, especially fruit trees. However, the regulatory networks and differentially expressed microRNAs (miRNAs) related to grafting in apple remain unclear. Herein, we conducted high-throughput sequencing and identified differentially expressed miRNAs among self-rooted Fuji, self-rooted M9, and grafted Fuji/M9. We analyzed the flowering rate, leaf morphology, and nutrient and carbohydrate contents in the three materials. The flowering rate, element and carbohydrate contents, and expression levels of flowering genes were higher in Fuji/M9 than in Fuji. We detected 206 known miRNAs and 976 novel miRNAs in the three materials, and identified those that were up- or downregulated in response to grafting. miR156 was most abundant in Fuji, followed by Fuji/M9, and then self-rooted M9, while miR172 was most abundant in M9, followed by Fuji/M9, and then Fuji. These expression patterns suggest that that these miRNAs were related to grafting. A Gene Ontology (GO) analysis showed that the differentially expressed miRNAs controlled genes involved in various biological processes, including cellular biosynthesis and metabolism. The expression of differentially expressed miRNAs and flowering-related genes was verified by qRT-PCR. Altogether, this comprehensive analysis of miRNAs related to grafting provides valuable information for breeding and grafting of apple and other fruit trees. [ABSTRACT FROM AUTHOR]
