Simple Summary: Goats are susceptible to ectoparasite infection, and lice control is based on studies conducted in cattle and sheep through the topical application of several synthetic insecticides such as organophosphates, carbamates, and pyrethroids. However, the overuse of these compounds has led to insect resistance phenomena. Thus, natural insecticides could be preferred, considering that synthetic insecticides and their metabolites negatively impact the environment and human and animal health. The aim of the present study was to evaluate the efficacy and safety of neem oil on caprine pediculosis and on kids' growth performances. The neem (Azadirachta indica) belongs to the Meliaceae family, and in Eastern countries it is mainly considered for the insecticidal activities of the kernel oil. The neem seeds contain bioactive principles, such as azadirachtin A, salannin, nimbin, and nimbolide. The trial was carried out on 24 kids, 120 days old, maintained in open yards. Animals were divided in 4 homogeneous groups (n = 6 animals/group) based on age, louse count, body condition score (BCS) and live body weight: Control Group (C, saline NaCl, 0.9%), Neem Group 1 (NO-100, 100 mL of neem oil per 10 kg), Neem Group 2 (NO-200, 200 mL/10 kg), Neem Group 3 (NO-300, 300 mL/10 kg). The treatments were performed by spraying the insecticide on the goat's body. The study lasted 56 days, and weekly, the kids underwent louse count, BCS and body weight determination, and FAMACHA score. Data were analyzed by ANOVA for repeated measures. The species of lice identified was Linognathus stenopsis. Kids belonging to NO-200 and NO-300 showed a stronger reduction of louse count throughout the study (>95%). The daily weight gain recorded was significantly higher (p < 0.05) in NO-300 than C. No differences were found for BCS and FAMACHA scores. Simple Summary: Artificial Insemination (AI) is mainly used after estrus synchronization in buffalo, and consecutive synchronization protocols are used to enhance reproductive efficiency. In this study, two different synchronization protocols have been used: Ovsynch vs. a P4-administration, and their efficiency in primiparous animals has been evaluated in different seasons for up to four cycles of re-synchronization protocols. Results show that the pregnancy rate upon the initial AI tends to be higher in P4 treated buffaloes, and that AI efficiency after re-synchronization through P4 is higher than the Ovsynch protocol. In conclusion, synchronization treatments must be selected according to the season of the year. The results derived from this study could be useful for buffalo breeders who want to improve the reproductive efficiency in primiparous animals in commercially managed buffalo herds. Primiparous buffaloes were tested in two periods of the year characterized, by either low or high reproductive efficiency. They were subjected to two protocols for synchronization of ovulation: (i) Ovsynch (OV) and (ii) progesterone based (P4) treatment. After calving, the animals underwent a series of four cycles of re-synchronization protocols. The season did not affect pregnancy rates when the results of the two treatments were pooled together with regard to the first synchronization protocol, followed by AI. Pregnancy rates were similar during the low breeding season (50.3% vs. 57.4% in OV and P4, respectively), but different during the high breeding season (50.4% vs. 67.7% in OV and P4, respectively; p = 0.000). Logistic regression confirmed a significant effect of treatment and season interaction on pregnancy (p = 0.003). Following re-synchronization, a treatment by season interaction was detected during the low breeding season (odds ratio = 2.233), in favor of P4. Finally, a survival analysis showed a better response of animals subjected to P4 treatment from the second AI onward. In conclusion, the pooled data of pregnancy rates from both treatments between seasons are not different following AIs. Better results, though, were obtained from the implementation of P4 treatment, and are recorded in a season-fashioned mode when the comparison is made following first or cumulative AIs. Simple Summary: Mastitis can be considered one of the costliest diseases of the dairy Mediterranean Buffalo. Early detection of the disease is thus of great importance to farmers, to reduce or prevent production losses. The measurement of electrical conductivity (EC) of milk is a relatively simple and inexpensive technique, and it has been studied as a routine method for the diagnosis of mastitis in dairy farm. Limited information is currently available on the relationships among EC, production traits and somatic cells count in Italian Mediterranean Buffalo. Hence, the aim of this study was to investigate those correlations using data collected at a commercial Italian buffalo farm. We can conclude that in buffalo, as in other species, there is a strong relationship between EC and somatic cells. Furthermore, we observed that, if the objective is to have an additional and informative parameter for early detection of mastitis, frequent EC recording, possibly performed over a longer period of time, is more effective than recording EC only a few times. Even if our results are encouraging, further studies are needed, in order to validate them, especially if the objective is the development of udder disease prediction models. The measurement of milk electrical conductivity (EC) is a relatively simple and inexpensive technique that has been evaluated as a routine method for the diagnosis of mastitis in dairy farms. The aim of this study was to obtain further knowledge on relationships between EC, production traits and somatic cell count (SCC) in Italian Mediterranean Buffalo. The original dataset included 5411 records collected from 808 buffalo cows. Two mixed models were used to evaluate both the effect of EC on MY, PP and FP and EC at test-day, and the effect of EC on somatic cell score (SCS) by using five different parameters (EC_param), namely: EC collected at the official milk recording test day (EC_day0), EC collected 3 days before official milk recording (EC_day3), and three statistics calculated from EC collected 1, 3 and 5 days before each test-day, respectively. All effects included in the model were significant for all traits, with the only exception of the effect of EC nested within parity for FP. The relationship between EC and SCS was always positive, but of different magnitude according to the parity. The regression of EC on SCS at test-day using different EC parameters was always significant except when the regression parameter was the slope obtained from a linear regression of EC collected over the 5-day period. Moreover, in order to evaluate how well the different models fit the data, three parameters were used: the Average Information Criteria (AIC), the marginal R2 and the conditional R2. During haymaking and ensilage, a significant loss of sugars occurs. The addition of a total mixed ratio (TMR) with a liquid feed might provide promptly utilisable energy and recover the nutrients lost during the conservation. Interesting results were already obtained by including liquid feed in a TMR in a dairy cow. However, the possibility to also utilize them in Italian Mediterranean buffalo is not yet supported by data. This study aimed to evaluate the in vitro fermentation characteristics and kinetics of different types of liquid feed, utilising bovine and buffalo rumen liquor as inocula. TMR supplemented with 0.025 g of four different liquid feeds was incubated with the TMR as control with buffalo and bovine rumen fluid using in vitro gas production technique. Considering bovine inoculum, all the experimental diets showed lower organic matter degradability and higher volatile fatty acid production than control TMR, while with buffalo rumen liquor, significant differences were observed between experimental and control diets in terms of gas production and fermentation kinetics. The tested liquid feeds can have different fermentation patterns depending on their ingredients and compositions. Supplementing liquid feeds to a standard diet seems to provide a source of energy that improves fermentation. No negative effects were observed on the in vitro fermentation at the dosage utilised. Recent pharmacological research on milk whey, a byproduct of the dairy industry, has identified several therapeutic properties that could be exploited in modern medicine. In the present study, we investigated the anticancer effects of whey from Mediterranean buffalo (Bubalus bubalis) milk. The antitumour effect of delactosed milk whey (DMW) was evaluated using the HCT116 xenograft mouse model of colorectal cancer (CRC). There were no discernible differences in tumour growth between treated and untreated groups. Nevertheless, haematoxylin and eosin staining of the xenograft tissues showed clearer signs of different cell death in DMW-treated mice compared to vehicle-treated mice. Detailed biochemical and molecular biological analyses revealed that DMW was able to downregulate the protein expression levels of c-myc, phospho-Histone H3 (ser 10) and p-ERK. Moreover, DMW also activated RIPK1, RIPK3, and MLKL axis in tumour tissues from xenograft mice, thus, suggesting a necroptotic effect. The necroptotic pathway was accompanied by activation of the apoptotic pathway as revealed by increased expression of both cleaved caspase-3 and PARP-1. At the molecular level, DMW-induced cell death was also associated with (i) upregulation of SIRT3, SIRT6, and PPAR-γ and (ii) downregulation of LDHA and PPAR-α. Recently, several concerns have been expressed on red meat quality and consumption. The aims of this study were to evaluate the influence of different breeding techniques and a prolonged post dry aging (PDA) maturation process on biomolecules level in raw buffalo meat. In the first experiment, two groups of animals were maintained with different space availability (15 vs. 10 m²/animal) for 90 days and biomolecules content was evaluated. In experiment 2, two diets (with or without ryegrass green forage) were used to assess the concentration of these biomolecules. Finally, in experiment 3, the meat of the animals that showed the highest content of biomolecules was chosen to assess the influence of the PDA maturation process. Buffaloes reared at 15 m² showed a significantly (p < 0.05) higher content of the considered biomolecules compared with their counterparts. Similarly, buffaloes fed green forage showed higher content of biomolecules (p < 0.05) compared with the control group. The meat of the animals bred at 15 m² and fed green forage showed a significant (p < 0.01) increase of biomolecules content during the PDA maturation process up to 60 days without influence microbiological profile in terms of total aerobic bacterial counts, yeasts, and molds. Simple Summary: "Mozzarella di Bufala Campana DOP" (mozzarella cheese) is mainly produced and marketed during the spring and summer months. The buffalo is a seasonal species that increases its reproductive activity when daylight hours decrease. Therefore, to increase milk production in the favourable period, the so-called "Out of Breeding Season Mating" technique is applied. It consists of the interruption of sexual promiscuity during the naturally occurring breeding season and concentrating calving and milk production during periods of increasing daylight length. However, the application of this technique increases the incidence of anoestrus, as animals are forced to breed outside of their natural and favoured period of the year, although other factors can also increase the incidence of anoestrus, such as climate. A reduction of seasonal anoestrus can be achieved by using some hormonal treatments. In this study, primiparous acyclic buffaloes were selected and divided into three classes according to their days in milk. Animals were synchronized using P4 vaginal implants, and artificial insemination (AI) was performed according to protocol. The temperature–humidity index (THI) was recorded to evaluate its influence on anoestrus. Statistical analysis showed that the implemented P4-based treatments were highly effective in removing the anoestrus condition in buffaloes. On the contrary, no influence of the THI on the efficacy of the P4 synchronization treatment was observed. The aim of this study was to evaluate the efficacy of a progesterone-based treatment on anoestrus in buffaloes. Primiparous acyclic buffaloes (n = 276), were divided into three classes according to their days in milk (DIM): from 50 to 90 (Class I; n = 86), from 91 to 150 (Class II; n = 102) and from 150 to 200 (Class III; n = 88). Animals were synchronized using P4 vaginal implants, followed by timed artificial insemination (TAI). They were then allowed to enter into a larger group of buffaloes for natural mating 15 days after AI was performed, and pregnancy status was monitored from then on at 15-day intervals. Finally, the temperature–humidity index (THI) was calculated. Statistical analysis was performed by ANOVA by means and both multiple and linear regression. The total pregnancy rate (PR) was 87.7%, with no differences among DIM classes (88.0, 92.4, and 80.0% in Classes I, II, and III, respectively). However, the PR at TAI tended to be higher (p = 0.07) in buffaloes in Class II. The follicle (FL) area in Class II buffaloes was larger (p < 0.01) than that of the other classes. No influence of the THI on the total PR was recorded. The pregnancy outcome at TAI was affected by the FL area (odds ratio = 2.237; p < 0.05) and body condition score (BCS) (odds ratio = 1.256; p < 0.05). In conclusion, treatment with vaginal P4 optimizes pregnancy rates in anoestrus buffaloes, particularly when the animals are in mid-lactation and show an optimal BCS. Simple Summary: Colorectal cancer (CRC) is the second leading cause of cancer-related deaths and the most frequently diagnosed cancer type. CRC risk can be preventable by modifiable risk factors, including diet. This study provides evidence on the antiproliferative and pro-apoptotic effects of whey from Mediterranean water buffalo (Bubalus bubalis) milk in HT-29, HCT 116, LoVo, and SW480 cells. Results showed that whey induced metabolic dysfunctions and modulated the bioenergetic signature of CRC cells by targeting SIRT3 expression. These findings unveil the anti-neoplastic effects of whey and pave the way for the use of this by-product, rich in bioactive nutrients, in the setting of novel prevention strategies to reduce the risk of CRC. Emerging strategies to improve healthy aging include dietary interventions as a tool to promote health benefits and reduce the incidence of aging-related comorbidities. The health benefits of milk are also linked to its richness in betaines and short-chain acylcarnitines, which act synergistically in conferring anticancer, anti-inflammatory, and antioxidant properties. Whey, despite being a dairy by-product, still has a considerable content of bioactive betaines and acylcarnitines. Here, we investigated the anticancer properties of whey from Mediterranean water buffalo (Bubalus bubalis) milk by testing its antiproliferative effects in colorectal cancer (CRC) cells HT-29, HCT 116, LoVo and SW480. Results indicated that treatment with whey for 72 h inhibited cell proliferation (p < 0.001), induced cell cycle arrest, and apoptosis via caspase-3 activation, and modulated cell metabolism by limiting glucose uptake and interfering with mitochondrial energy metabolism with the highest effects observed in HT-29 and HCT 116 cells. At molecular level, these effects were accompanied by upregulation of sirtuin 3 (SIRT3) (p < 0.01) and peroxisome proliferator-activated receptor (PPAR)-γ expression (p < 0.001), and downregulation of lactate dehydrogenase A (LDHA) (p < 0.01), sterol regulatory-element binding protein 1 (SREBP1) (p < 0.05), and PPAR-α (p < 0.01). Transient SIRT3 gene silencing blocked the effects of whey on the LDHA, PPAR-γ, and PPAR-α protein expressions (p < 0.01) suggesting that the whey capacity of perturbating the metabolic homeostasis in CRC cell lines is mediated by SIRT3. Simple Summary: Considering the seasonal trend of milk production due to application of estrus synchronization treatments in autumn or in spring season, as well as to meet meat production and market requirements for religious holidays, one may find it useful to simplify the milk and milk constituent yield quantification during the entire period of lactation. The present work aimed to verify the application of Wood's model to a different number of test-day milk recording protocols in order to reduce the frequency of National Breeding Association test-day milking record protocol. Wood's model fit well on goat milk production, showing high R2 values. Moreover, the milk yield, fat-corrected milk yield, fat yield, and protein yield estimation applying Wood's model to four recordings (first, second, fourth, and sixth month records) showed results superimposable to International Committee of Animal Recording (ICAR) application and Wood's model application on test-day records every 15 days. Goats have important social and economic roles in many countries because of their ability to survive and be productive in marginal areas. The overarching aim of this study was to compare the application of Wood's model to different test-day milk recording protocols for estimation of total milk, fat, and protein yield in dairy goats. A total of 465 goats were used (Garganica, 78; Girgentana, 81; Jonica, 76; Maltese, 77; Red Mediterranean, 76; Saanen, 77). Milk yield was recorded every 15 days throughout lactation of 210 days, for a total of 14 collection days, during both morning and afternoon milking sessions. Milk samples were collected and analyzed for protein and fat. The fat-corrected milk was standardized at 35g fat/kg of milk. Wood models showed high R2 values, and thus good fitting, in all the considered breeds. Wood model applied to first, second, fourth, and sixth month recordings (C) and ICAR estimation showed total milk yield very close to Wood's model applied to all 14 recordings (A) (p > 0.38). Differently, Wood's model applied to the first, second, third, and fourth month recording (B) estimation showed great differences (p < 0.01). This could be applied for farms that had the necessity to synchronize flock groups for kidding in order to produce kid meat. In farms that apply the estrus induction and/or synchronization for kidding, it would be possible to perform only four test-day milk recordings and to apply the Wood's model on them in order to obtain the estimation of total milk, fat, and protein yield during lactation for animals inscribed, or to be inscribed, to the genealogical book. Simple Summary: Reproductive biotechnologies in beef farms are key issues, such as artificial insemination, fixed-time artificial insemination (FTAI), embryo transfer, and ovum pick up, among others. Artificial insemination takes the first place with these available tools. Thus, science keeps improving hormonal treatments and procedures to optimize pregnancies after insemination. A synchronization protocol commonly used in beef heifers is the 5d Co-synch protocol, which fits better in terms of ovarian synchronization and resulting conception rate. We propose a modification of this protocol, which consists of delaying by 24 h the removal of the intravaginal progesterone device, to decrease the percentage of heifers showing estrus before the date of FTAI. Moreover, this modification will make easier the workload when inseminating herds with heifers and cows at the same time. Based on our results, the protocol 5d Co-synch with the delayed removal of the intravaginal progesterone device fits as well as the original protocol in terms of synchronization and conception rate. Therefore, it can be implemented in the heifers of beef cattle commercial farms. In beef herds, increasing animal welfare, improving reproductive performance and easing animal management are key goals in farm economics. We explored whether delaying the removal of the intravaginal progesterone device by 24 h in heifers synchronized with a 5d Co-synch 72-h protocol could improve reproductive efficiency of fixed-time artificial insemination (FTAI). In experiment 1, we examined the total synchronization rate (TSR) in cycling Holstein heifers. Heifers (13.4 ± 0.69 mo.) were randomly assigned to the standard 5d Co-synch 56-h protocol (5dCo56; n = 10), 5d Co-synch 72-h (5dCo72; n = 17), or the modified 5d Co-synch 72-h protocol, in which removal of the progesterone device was delayed by 24 h (6dCo48; n = 19). In experiment 2, 309 cycling beef heifers on 18 commercial farms were subjected to the 5d Co-synch 72-h or 6-d Co-synch 48-h protocol and conception rate (CR) studied. In experiment 1, the three protocols led no differences on TSRs of 80.0% (5dCo56), 88.2% (5dCo72), and 89.5% (6dCo48). In experiment 2, the CR from the beef heifers, observed during two consecutive reproductive seasons did not differ: 59.7% for 5dCo72 and 62.0% for 6dCo48 (p = 0.907). Simple Summary: Small ruminant farming can potentially adopt low-input strategies by using agro-industrial byproducts as alternative feeding sources. Byproducts are natural, and thus are preferred by consumers because most of them have antioxidant activity that may improve different aspects linked to meat quality and human health. For this reason, a red orange and lemon extract, (RLE) rich in anthocyanins, is tested as an oral additive on kids' meat. No differences were recorded on animal performance, but on meat, RLE inclusion improves juiciness and reduces color deterioration. Moreover, RLE reduced cooking loss, and meat quality is positively influenced, due to both delaying lipid oxidation and a better fatty acid profile resulting in healthier meat for human consumption. This study evaluates the animal performance and meat characteristics of 60 Saanen suckling kids daily fed a red orange and lemon extract (RLE), rich in anthocyanins. In our methodology, after colostrum administration, animals are randomly assigned to two treatments: Treatment group (Group RLE; n = 30) that received RLE (90 mg/kg live body weight) as oral food additive, and a control group (Group CON; n = 30) that received a standard diet. Animals are slaughtered after 40 days. The RLE administration did not influence daily weight gain, carcass measurements, or incidences (expressed as a percentage) of different anatomical regions on the whole carcass weight. On the contrary, RLE supplementation significantly improved the oxidative profile of the meat seven days after slaughtering, as demonstrated by the reduced levels of thiobarbituric acid reactive substances (TBARS; p < 0.01) and hydroperoxides (p < 0.01) in Group RLE compared to Group CON. A significant influence of RLE administration is observed on day 7 for yellowness (p < 0.01). There are also lower saturated and higher monounsaturated and polyunsaturated fatty acids concentration in Group RLE meat (p < 0.01), which also shows lower atherogenic and thrombogenic indexes (p < 0.01) compared to Group CON. Simple Summary: The advent of color Doppler ultrasonography promoted tremendous advances in research and clinical practice in animal reproduction, because it allowed noninvasive visualization of the vascularization in reproductive organs. In particular, the corpus luteum blood flow reflects luteal function better than luteal size in ruminants. Since buffaloes are a seasonal species, it is important to evaluate corpus luteum functionality also during the nonbreeding season, through blood flow examination and early pregnancy diagnosis. For this reason, we selected 29 Mediterranean buffaloes that had undergone synchronization and artificial insemination and were examined daily, from 5 to 10 days post-artificial insemination. Then, we retrospectively classified the buffaloes as pregnant or nonpregnant. Statistical analysis showed that pregnant animals had higher mean progesterone concentration and higher mean time average medium velocity values from Day 5 to Day 10 compared to nonpregnant buffaloes. Moreover, these two parameters could be used to predict the likelihood of pregnancy, starting on Day 6, although more reliable results could be obtained at Day 10 post-artificial insemination. In conclusion, a proper CL growth and development from Day 5 to Day 10 post-timed artificial insemination (TAI) is crucial for pregnancy maintenance during the transitional period. This study evaluated corpus luteum (CL) development in buffaloes out of breeding season and assessed an early pregnancy diagnosis. Mediterranean buffaloes (n = 29) were synchronized and artificially inseminated. CL B-mode/color Doppler ultrasonography examinations were performed daily from Days 5 to 10 post-synchronization, recording CL dimensions and blood flow parameters. Blood samples were collected on the same days for the progesterone (P4) assay. Data were grouped into pregnant or nonpregnant and retrospectively analyzed. The total pregnancy rate was 50.0% (13/26) on Day 45. A significant difference between CL average area in pregnant and nonpregnant buffaloes was recorded only on Day 10. Pregnant buffaloes showed a significantly higher mean P4 concentration and higher mean time average medium velocity (TAMV) values from Day 5 to Day 10 compared to nonpregnant buffaloes. Linear regression analysis showed a significant relationship between P4 levels and TAMV. Multiple logistic regression highlighted a significant influence of TAMV on pregnancy outcome, particularly on Day 8. This is probably due to the strong relationship between TAMV and P4 production. Both TAMV and P4 could be used to predict pregnancy starting on Day 6, although a more reliable result was obtained at Day 10. Simple Summary: In goat breeding, artificial insemination has a strong economic impact: It provides genetic improvement in intensive production systems and guarantees breed preservation in extensive systems. Sperm freezability is affected by several factors, such as breed species and freezing procedures. In addition, in goats, the seminal plasma enzyme phospholipase A reacts with egg yolk (component of semen extender), compromising sperm viability. Thus, seminal plasma is removed before freezing. This removal causes a decreasing level of antioxidants that prevent formation of toxic lipid peroxides with deleterious effects on fertility. Crocin is a water-soluble carotenoid that acts as an antioxidant, protecting cells and tissues against oxidation. Therefore, the aim of the present study was to assess the effect of crocin in the semen extender before cryopreservation. Five different goat breeds (Garganica, Jonica, Maltese Mediterranean Red and Saanen) were chosen to evaluate sperm qualitative characteristics, such as post-thawing sperm motility, viability, morphology and membrane integrity, as well as DNA fragmentation and intracellular ROS levels. The results highlighted that crocin supplementation in the extender decreased oxidative stress and improved sperm motility and the DNA integrity of frozen-thawed sperm in different breeds. The effect of crocin in the semen extender before cryopreservation was evaluated on sperm parameters of 20 bucks of five different breeds: Garganica (GA), Jonica (JO), Maltese (MA), Mediterranean Red (MR) and Saanen (SA). Semen samples were centrifuged, to remove seminal plasma, divided in two aliquots and diluted with Tris-egg-yolk-based extender, containing 0 (control group) and 1 mM crocin. Crocin concentration was established after a preliminary dose trial. On fresh and frozen-thawed sperm, motility, viability, morphology, membrane integrity, DNA fragmentation and ROS levels were evaluated. The freezing process led to a decrease (p < 0.05) in all the sperm parameters recorded, confirming the deleterious effect of cryopreservation on goat semen. The most interesting result regarding the inclusion of crocin in the extender before cryopreservation was as follows: Crocin significantly improved (p < 0.05) sperm motility in all breeds, except for Mediterranean Red, compared to the control group. Furthermore, 1 mM crocin reduced percentage of spermatozoa with DNA fragmentation with a marked decrement (p < 0.05) in Garganica and Saanen, as compared to the control group. Finally, intracellular ROS decreased (p < 0.01) in the crocin-treated sperm of all breeds, as compared to the control. In conclusion, supplementation of 1 mM crocin in the extender decreased oxidative stress, improving sperm motility and the DNA integrity of frozen-thawed sperm in different breeds. Simple Summary: Today, the ability to determine the pregnancy status of cows as soon as possible after artificial insemination (AI) has become the most important thing to obtain in an ideal farm. Several efforts have been made to discover biomarkers of early pregnancy but, as of today, without any particular result. Most of the studies were carried out on non-invasive and cheap biological fluids, such as milk. Therefore, in order to identify potential biomarkers of early pregnancy a metabolomic approach on milk of 10 pregnant and 10 non-pregnant buffaloes was used. Milk was recovered in different days before and after the AI, and the data were analyzed retrospectively. The results revealed significant differences between pregnant and non-pregnant buffaloes, as well as in the expression of five metabolites. These data suggest the effectiveness of the metabolomic analysis for the identification of novel potential biomarkers in early prediction of pregnancy in buffaloes after AI, and these findings would give breeders the opportunity to rebreed animals at the next estrus event, saving most of the days as open. This study aimed to identify potential biomarkers for early pregnancy diagnosis in buffaloes subjected to artificial insemination (AI). The study was carried out on 10 pregnant and 10 non-pregnant buffaloes that were synchronized by Ovsynch-Timed Artificial Insemination Program and have undergone the first AI. Furthermore, milk samples were individually collected ten days before AI (the start of the synchronization treatment), on the day of AI, day 7 and 18 after AI, and were analyzed by LC–MS. Statistical analysis was carried out by using Mass Profile Professional (Agilent Technologies, Santa Clara, CA, USA). Metabolomic analysis revealed the presence of several metabolites differentially expressed between pregnant and non-pregnant buffaloes. Among these, a total of five metabolites were identified by comparison with an online database and a standard compound as acetylcarnitine (3-Acetoxy-4-(trimethylammonio)butanoate), arginine-succinic acid hydrate, 5′-O-{[3-({4-[(3aminopropyl)amino]butyl}amino)propyl]carbamoyl}-2′-deoxyadenosine, N-(1-Hydroxy-2-hexadecanyl)pentadecanamide, and N-[2,3-Bis(dodecyloxy)propyl]-L-lysinamide). Interestingly, acetylcarnitine was dominant in milk samples collected from non-pregnant buffaloes. The results obtained from milk metabolic profile and hierarchical clustering analysis revealed significant differences between pregnant and non-pregnant buffaloes, as well as in the metabolite expression. Simple Summary: The study aims to compare physiological and productive responses of five Mediterranean heritage goat breeds with a high production breed, spread worldwide, the Saanen breed. The overall objective of the paper was to highlight that in difficult environmental conditions, although not so extreme, a high production breed does not actually represent the best solution, both from animals' welfare conditions and for production. This study aimed to describe metabolic, oxidative, and mineral blood profiles of Saanen does through lactation compared with Mediterranean breed clusters (Maltese and Rossa Mediterranea, and Jonica, Garganica, and Girgentana). Milk and blood samples of 57 dairy goats (9–10 goats per breed) were collected from the 2nd to the 30th week of lactation every 2–3 weeks. Saanen showed greater milk yield and somatic cell score, and lower fat and protein percentage through lactation (p < 0.05) than the Mediterranean breed clusters. Blood analysis revealed that stage of lactation had a greater impact than breed cluster, except for uric acid, alkaline phosphatase, and aspartate aminotransferase. Plasmatic non-esterified fatty acids indicated a greater negative energy balance in Saanen than the other breed clusters during early and medium lactation stages (p < 0.05). Serum Cl, Mg, and Ca increased in all the breed clusters from early to the following stages of lactation (p < 0.05). No significant prooxidant/antioxidant imbalances were detected in any of the three clusters during the entire lactation. In conclusion, Mediterranean breeds tended to recover earlier from negative energy balance than Saanen, but effects of breed or stage of lactation on long-term oxidative stress indicators were not evident. Simple Summary: The buffalo dairy industry has deep roots in Southern Italy, due to the traditional link with Mozzarella di Bufala, a Protected Designation of Origin cheese with high economic value and market demand. At farm level, strategies aiming to improve milk yield and quality are essential to maximize profitability and dairy chain efficiency. In this study, we analyzed a large data set of Italian Buffaloes (around 70 thousands animals) in order to detect the phenotypic sources of variation of milk yield and quality traits, disclose exploitable favorable correlations among milk traits, and provide useful information for dairy buffalo chain stakeholders. The buffalo milk industry has economic and social relevance in Italy, as linked to the manufacture of traditional dairy products. To provide an overview of the current status of buffaloes' performances on a large scale, almost 1 million milk test-day records from 72,294 buffaloes were available to investigate milk yield, energy corrected milk, fat, protein, and lactose content, and somatic cell score (SCS). Phenotypic correlations between milk traits were calculated and analysis of variance was carried out through a mixed model approach including fixed effect of parity, stage of lactation, sampling time, month of calving, and all their interactions and random effects of buffalo, herd-test-date, and residual. Third-parity buffaloes were the most productive in terms of milk yield, while the lowest solid content was detected in sixth parity buffaloes. A considerable gap between primiparous and multiparous buffaloes was observed for milk yield, especially in early- and mid-lactation. Overall, SCS progressively increased with parity and showed a negative correlation with milk yield in both primiparous (−0.12) and multiparous (−0.14) buffaloes. Results suggested that, at the industrial level, milk of primiparous buffaloes may be preferred for transformation purposes, since it was characterized by greater solid content and lower SCS. Results of this study provide a picture of the Italian population of buffaloes under systematic performance records and might be beneficial to both dairy industry and breeding organizations. The aim of this study is to combine advanced GC-MS and metabolite identification in a robust and repeatable technology platform to characterize the metabolome of buffalo milk and mozzarella cheese. The study utilized eleven dairies located in a protected designation of origin (PDO) region and nine dairies located in non-PDO region in Italy. Samples of raw milk (100 mL) and mozzarella cheese (100 g) were obtained from each dairy. A total of 185 metabolites were consistently detected in both milk and mozzarella cheese. The PLS-DA score plots clearly differentiated PDO and non-PDO milk and mozzarella samples. For milk samples, it was possible to divide metabolites into two classes according to region: those with lower concentrations in PDO samples (galactopyranoside, hydroxybuthyric acid, allose, citric acid) and those with lower concentrations in non-PDO samples (talopyranose, pantothenic acid, mannobiose, etc.,). The same was observed for mozzarella samples with the proportion of some metabolites (talopyranose, 2, 3-dihydroxypropyl icosanoate, etc.,) higher in PDO samples while others (tagatose, lactic acid dimer, ribitol, etc.,) higher in non-PDO samples. Simple Summary: Milk fatty acid composition affects human health and dairy products flavor. In particular, some saturated fatty acids increase the risk of cardiovascular diseases, whereas conjugated linoleic acid inhibits carcinogenesis and reduces atherosclerosis and diabetes. Moreover, a greater amount of some short chain fatty acids increase the goaty flavor of dairy products. The objective of this study was to evaluate the breed and week of lactation effects on milk fatty acid profile of 5 Italian local goat breeds and a cosmopolitan breed reared in the same farm. Results showed that the fatty acid profile was mainly affected by the week of lactation. Saturated fatty acids were abundant in early lactation and unsaturated fatty acids were abundant in late lactation. Local goat breeds produced milk with lower concentration of saturated fatty acids than the cosmopolitan breed. This study may contribute to valorize milk of Italian local goat breeds which seems to have a healthier profile than milk of the cosmopolitan breed. A total of 252 milk samples were collected monthly from 42 goats (7 goats per breed) and analyzed for gross composition and FA profile. Individual FA was determined using gas-chromatography. Data were analyzed using a mixed model with repeated measures with breed and week of lactation as fixed effects. Results showed that the FA profile was significantly affected by week of lactation and only few FA by breed effect (p < 0.05). Overall, the main differences were found between Saanen and local breeds. This study contributed to the characterization of goat milk FA profile, and it may be of interest for the valorization of milk from local goat breeds which seem to have a healthier profile than milk of the cosmopolitan breed. Week of lactation affected mineral contents, except for B, being lower in early than late lactation, whereas, breed affected only P, Mg and Zn. Mediterranean Red and Jonica breeds' milk was richer in P than Maltese, and in Zn than Maltese, Girgentana and Saanen breeds. For Mg, only Saanen differed from Maltese. Simple Summary: In the last several years, the animal production sector has been very concerned about the overuse of antibiotics and the concomitant increase in antimicrobial resistance. The objective of this paper was the evaluation of the immune properties of different extracts isolated from Chlorella sorokiniana applied to sheep peripheral blood mononuclear cells (PBMCs). In particular, the effects of the unsaponified fraction (UP), the acetylated unsaponified fraction (AUP), and the total lipid fraction (TL) at two concentrations (0.4 and 0.8 mg/mL) on the proliferative response of sheep cells and their cytokine secretion were tested. All the extracts showed an inhibitory effect on cell proliferation, particularly in the presence of the UP fraction at a 0.4 mg/mL concentration. As regards cytokine production, the UP extract tested at a 0.8 mg/mL concentration increased interleukin(IL)-10 production, whereas the TL fraction at 0.4 mg/mL showed a cytokine profile characterized by an increase in both IL-10 secretion and the anti-inflammatory cytokine IL-6, and, to a lesser extent, IL-1β secretions, which are proinflammatory cytokines. These results supported the hypothesis that extracts from Chlorella sorokiniana could be considered useful ingredients to be integrated into animal feed with the aim to control immune responses during inflammation and minimize the use of antibiotics. The objective of this experiment was to study the effects of the unsaponified fraction (UP), the acetylated unsaponified fraction (AUP), and the total lipid fraction (TL) extracted and purified from Chlorella sorokiniana (CS) on the proliferation and cytokine profile of sheep peripheral blood mononuclear cells (PBMCs). Cells were cultured with 0.4 mg/mL and 0.8 mg/mL concentrations of each extract (UP, AUP, and TL fractions) and activated with 5 μg/mL concanavalin A (ConA) and 1 μg/mL lipopolysaccharide (LPS) at 37 °C for 24 h. PBMCs cultured with ConA and LPS represented the stimulated cells (SC), and PBMCs without ConA and LPS represented the unstimulated cells (USC). Cell-free supernatants were collected to determine IL-10, IL-1β, and IL-6 secretions; on cells, measurement of proliferation was performed. All the extracts tested significantly decreased the cell proliferation; in particular, the UP fraction at 0.4 mg/mL showed the lowest proliferative response. Furthermore, at 0.8 mg/mL, the UP fraction enhanced IL-10 secretion. On the contrary, the TL fraction at 0.4 mg/mL induced an increase in IL-10, IL-6, and, to a lesser extent, IL-1β secretions by cells. The AUP fraction did not change cytokine secretion. The results demonstrated that CS extracts could be useful ingredients in animal feed in order to minimize the use of antibiotics by modulating cell proliferation and cytokine response.
