Simple Summary: This study, focusing on silkworms as the research subject, investigated the detrimental effects of gene mutants containing premature termination codons on organisms and the potential genetic compensation effects they may trigger. The research findings revealed the presence of genetic compensation effects in silkworms with a premature termination mutation in the BmTrpA1 gene, leading to a significant increase in the mRNA levels of BmTrpA subfamily paralogs and in Upf2, a key factor of the nonsense-mediated mRNA degradation (NMD) pathway. Knocking down the key factor could suppress the genetic compensation effects. These discoveries document genetic compensation and its role in Lepidoptera insects. Genetic mutations leading to premature termination codons are known to have detrimental effects. Using the Lepidoptera model insect, the silkworm (Bombyx mori), we explored the genetic compensatory response triggered by mutations with premature termination codons. Additionally, we delved into the molecular mechanisms associated with the nonsense-mediated mRNA degradation pathway. CRISPR/Cas9 technology was utilized to generate a homozygous bivoltine silkworm line BmTrpA1−/− with a premature termination. Transcript levels were assessed for the BmTrpA paralogs, BmPyrexia and BmPainless as well as for the essential factors Upf1, Upf2, and Upf3a involved in the nonsense-mediated mRNA degradation (NMD) pathway. Upf2 was specifically knocked down via RNA interference at the embryonic stage. The results comfirmed that the BmTrpA1 transcripts with a 2-base deletion generating a premature termination codon in the BmTrpA1−/− line. From day 6 of embryonic development, the mRNA levels of BmPyrexia, BmPainless, Upf1, and Upf2 were significantly elevated in the gene-edited line. Embryonic knockdown of Upf2 resulted in the suppression of the genetic compensation response in the mutant. As a result, the offspring silkworm eggs were able to hatch normally after 10 days of incubation, displaying a non-diapause phenotype. It was observed that a genetic compensation response does exist in BmTrpA1−/− B. mori. This study presents a novel discovery of the NMD-mediated genetic compensation response in B. mori. Salinity stands as a significant environmental stressor, severely impacting crop productivity. Plants exposed to salt stress undergo physiological alterations that influence their growth and development. Meanwhile, plants have also evolved mechanisms to endure the detrimental effects of salinity-induced salt stress. Within plants, Calcineurin B-like (CBL) proteins act as vital Ca2+ sensors, binding to Ca2+ and subsequently transmitting signals to downstream response pathways. CBLs engage with CBL-interacting protein kinases (CIPKs), forming complexes that regulate a multitude of plant growth and developmental processes, notably ion homeostasis in response to salinity conditions. This review introduces the repercussions of salt stress, including osmotic stress, diminished photosynthesis, and oxidative damage. It also explores how CBLs modulate the response to salt stress in plants, outlining the functions of the CBL-CIPK modules involved. Sudden infant death syndrome (SIDS) has always been a regrettable issue for families. After sleeping in the supine position was proposed, the incidence of SIDS declined dramatically worldwide. However, SIDS still accounts for the top 10 causes of infant deaths in Taiwan. Recognizing the risk factors and attempting to minimize these cases are imperative. We obtained information on cases with SIDS from the National Health Insurance Research Database in Taiwan and interconnected it with the Taiwan Maternal and Child Health Database to acquire infant–maternal basal characteristics between 2004 and 2017. The SIDS subjects were matched 1:10 considering gestational age to normal infants. After case selection, a total of 953 SIDS cases were included. Compared with healthy infants, SIDS infants had younger parents, lower birth weight, and lower Apgar scores. After adjusting for potential confounders, infants with mothers aged <20 years had 2.81 times higher risk of SIDS. Moreover, infants in the non-eastern region had a significantly lower risk of SIDS than those in the eastern region. We concluded that infants of young mothers (especially maternal age <20 years) and infants in the eastern region of Taiwan had a higher risk of SIDS than their counterparts. [ABSTRACT FROM AUTHOR]

Determining the optimal endotracheal tube (ETT) depth in neonates remains challenging for neonatologists. The guideline for optimal ETT depth is based on the patients' weight or gestational age. However, there is a discrepancy in the suggested ETT depth between these two parameters. The aim of this retrospective study was to compare the recommended weight-based and age-based formulas for optimal ETT depth and obtain the optimal reference before intubation. Participants were assigned to group 1 if the recommended ETT insertion depth based on weight was concordant with the recommended depth based on gestational age, and to group 2 if the weight and age-based depth recommendations were discordant. After exclusion, 180 patients were included in the analysis. Results indicated that the predicted ETT depth suggested by age required more adjustment than by weight (p < 0.05). Furthermore, the required adjustment in the weight-based formula was smaller than the age-based formula (p < 0.05). Multivariate linear regression analysis revealed that weight was the key factor affecting the optimal depth (p < 0.001). These results imply that when there is a discrepancy in ETT depth between the weight-based and age-based recommendation, the weight-based one will be more accurate than the age-based one. Combining research areas of biomechanics and pedestrian dead reckoning (PDR) provides a very promising way for pedestrian positioning in environments where Global Positioning System (GPS) signals are degraded or unavailable. In recent years, the PDR systems based on a smartphone’s built-in inertial sensors have attractedmuch attention in such environments. However, smartphone-basedPDR systems are facing various challenges, especially the heading drift, which leads to the phenomenon of estimated walking path passing through walls. In this paper, the 2D PDR system is implemented by using a pocket-worn smartphone, and then enhanced by introducing a map-matching algorithm that employs a particle filter to prevent the wall-crossing problem. In addition, to extend the PDR system for 3D applications, the smartphone’s built-in barometer is used to measure the pressure variation associated to the pedestrian’s vertical displacement. Experimental results show that the map-matching algorithm based on a particle filter can e ectively solve the wall-crossing problem and improve the accuracy of indoor PDR. By fusing the barometer readings, the vertical displacement can be calculated to derive the floor transition information. Despite the inherent sensor noises and complex pedestrian movements, smartphone-based 3D pedestrian positioning systems have considerable potential for indoor location-based services (LBS). Heterosis, a phenomenon characterized by the superior performance of hybrid individuals relative to their parents, has been widely utilized in livestock and crop breeding, while the underlying genetic basis remains elusive in chickens. Here, we performed a reciprocal crossing experiment with broiler and layer chickens and conducted RNA sequencing on liver tissues for reciprocal crosses and their parental lines to identify inheritance patterns of gene expression. Our results showed that heterosis of the abdominal fat percentage was 69.28%–154.71% in reciprocal crosses. Over-dominant genes of reciprocal crosses were significantly enriched in three biological pathways, namely, butanoate metabolism, the synthesis and degradation of ketone bodies, and valine, leucine, and isoleucine degradation. Among these shared over-dominant genes, we found that a lipid-related gene, HMGCL, was enriched in these pathways. Furthermore, we validated this gene as over-dominant using qRT-PCR. Although no shared significant pathway was detected in the high-parent dominant genes of reciprocal crosses, high-parent dominant gene expression was the major gene inheritance pattern in reciprocal crosses and we could not exclude the effect of high-parent dominant genes. These findings suggest that non-additive genes play important roles in the heterosis of important traits in chickens and have important implications regarding our understanding of heterosis. Anaplastic lymphoma kinase (ALK) is a receptor tyrosine kinase involved in various cancers. In its basal state, the structure of ALK is in an autoinhibitory form stabilized by its A-loop, which runs from the N-lobe to the C-lobe of the kinase. Specifically, the A-loop adopts an inhibitory pose with its proximal A-loop helix (αAL-helix) to anchor the αC-helix orientation in an inactive form in the N-lobe; the distal portion of the A-loop is packed against the C-lobe to block the peptide substrate from binding. Upon phosphorylation of the first A-loop tyrosine (Y1278), the αAL-helix unfolds; the distal A-loop detaches from the C-lobe and reveals the P+1 pocket that accommodates the residues immediately after their phosphorylation, and ALK is activated accordingly. Recently, two neuroblastoma mutants, F1174L and R1275Q, have been determined to cause ALK activation without phosphorylation on Y1278. Notably, F1174 is located on the C-terminus of the αC-helix and away from the A-loop, whereas R1275 sits on the αAL-helix. In this molecular modeling study, we investigated the structural impacts of F1174L and R1275Q that lead to the gain-of-function event. Wild-type ALK and ALK with phosphorylated Y1278 were also modeled for comparison. Our modeling suggests that the replacement of F1174 with a smaller residue, namely leucine, moves the αC-helix and αAL-helix into closer contact and further distorts the distal portion of the A-loop. In wild-type ALK, R1275 assumes the dual role of maintaining the αAL-helix–αC-helix interaction in an inactive form and securing αAL-helix conformation through the D1276–R1275 interaction. Accordingly, mutating R1275 to a glutamine reorients the αC-helix to an active form and deforms the entire A-loop. The new three-dimensional structure that the graphene connected with SWCNTs (G-CNTs, Graphene Single-Walled Carbon Nanotubes) can solve graphene and CNTs′ problems. A comprehensive study of the mechanical and electrical performance of the junctions was performed by first-principles theory. There were eight types of junctions that were constituted by armchair and zigzag graphene and (3,3), (4,0), (4,4), and (6,0) CNTs. First, the junction strength was investigated. Generally, the binding energy of armchair G-CNTs was stronger than that of zigzag G-CNTs, and it was the biggest in the armchair G-CNTs (6,0). Likewise, the electrical performance of armchair G-CNTs was better than that of zigzag G-CNTs. Charge density distribution of G-CNTs (6,0) was the most homogeneous. Next, the impact factors of the electronic properties of armchair G-CNTs were investigated. We suggest that the band gap is increased with the length of CNTs, and its value should be dependent on the combined effect of both the graphene's width and the CNTs' length. Last, the relationship between voltage and current (U/I) were studied. The U/I curve of armchair G-CNTs (6,0) possessed a good linearity and symmetry. Fluorine, a hazard that is associated with coal, has resulted in serious environmental issues during the production and utilization of coal. In this paper, we provide a detailed review of fluorine in Chinese coal, including the distribution, concentration, modes of occurrence, genetic factors, and environmental effects. The average concentration of fluorine in Chinese coal is 130.0 mg/kg, which is slightly higher than coal worldwide (88.0 mg/kg). The enrichment of fluorine in Chinese coal varies across different coal deposit regions, and it is especially high in Inner Mongolia (Junger coalfield, Daqingshan coalfield) and southwest China (coal mining regions in Yunnan, Guizhou province). The fluorine distribution is uneven, with a relatively high content in southwest coal (including Yunnan, Guizhou, Chongqing, and Sichuan provinces), very high content in the coal of North China (Inner Mongolia) and South China (Guangxi), and is occasionally found in the northwest (Qinghai). Fluorine occurs in various forms in coal, such as independent minerals (fluorine exists as fluorapatite or fluorite in coal from Muli of Qinghai, Taoshuping of Yunnan, Guiding of Guizhou, and Daqingshan of Inner Mongolia), adsorption on minerals (fluorine in coal from Nantong, Songzao of Chongqing, Guxu of Sichuan, and Shengli, Daqingshan, and Junger from Inner Mongolia), substitution in minerals (Wuda coal, Inner Mongolia), and a water-soluble form (Haerwusu coal, Inner Mongolia). The enrichment of fluorine is mainly attributed to the weathering of source rock and hydrothermal fluids; in addition to that, volcanic ash, marine water influence, and groundwater affect the fluorine enrichment in some cases. Some environmental and human health problems are related to fluorine in coal, such as damage to the surrounding environment and husbandry (poisoning of livestock) during the coal combustion process, and many people have suffered from fluorosis due to the burning of coal (endemic fluorosis in southwest China). In order to meet the higher accuracy and system reliability requirements, the information fusion for multi-sensor systems is an increasing concern. Dempster-Shafer evidence theory (D-S theory) has been investigated for many applications in multi-sensor information fusion due to its flexibility in uncertainty modeling. However, classical evidence theory assumes that the evidence is independent of each other, which is often unrealistic. Ignoring the relationship between the evidence may lead to unreasonable fusion results, and even lead to wrong decisions. This assumption severely prevents D-S evidence theory from practical application and further development. In this paper, an innovative evidence fusion model to deal with dependent evidence based on rank correlation coefficient is proposed. The model first uses rank correlation coefficient to measure the dependence degree between different evidence. Then, total discount coefficient is obtained based on the dependence degree, which also considers the impact of the reliability of evidence. Finally, the discount evidence fusion model is presented. Software DefinedWireless Rechargeable Sensor Networks (SDWRSNs) are an inexorable trend for Wireless Sensor Networks (WSNs), including Wireless Rechargeable Sensor Network (WRSNs). However, the traditional network devices cannot be completely substituted in the short term. Hybrid SDWRSNs, where software defined devices and traditional devices coexist, will last for a long time. Hybrid SDWRSNs bring new challenges as well as opportunities for energy saving issues, which is still a key problem considering that the wireless chargers are also exhaustible, especially in some rigid environment out of the main supply. Numerous energy saving schemes for WSNs, or even some works for WRSNs, are no longer suitable for the new features of hybrid SDWRSNs. To solve this problem, this paper puts forward an Energy-saving Traffic Scheduling (ETS) algorithm. The ETS algorithm adequately considers the new characters in hybrid SDWRSNs, and takes advantage of the Software Defined Networking (SDN) controller’s direct control ability on SDN nodes and indirect control ability on normal nodes. The simulation results show that, comparing with traditional Minimum Transmission Energy (MTE) protocol, ETS can substantially improve the energy efficiency in hybrid SDWRSNs for up to 20–40% while ensuring feasible data delay. Fucoidan, a fucose-containing sulfated polysaccharide with diverse biological functions, is mainly recovered from brown algae. In this study, we utilized a compressional-puffing process (CPP) to pretreat Sargassum crassifolium (SC) and extracted fucoidans from SC by warm water. Three fucoidan extracts (SC1: puffing at 0 kg/cm2; SC2: puffing at 1.7 kg/cm2; and SC3: puffing at 6.3 kg/cm2) were obtained, and their composition, and antioxidant and neuroprotective activities were examined. The results suggest that CPP decreased the bulk density of algal samples, expanded the algal cellular structures, and eliminated the unpleasant algal odor. The extraction yields of fucoidans were increased and impurities of fucoidans were decreased by increasing the pressures used in CPP. The SC1-SC3 extracts displayed various characteristics of fucoidan as illustrated by the analyses of composition, Fourier transform infrared (FTIR) spectroscopy, and molecular weight. All three extracts SC1-SC3 showed antioxidant activity dose-dependently. Although both SC1 and SC2 possessed high and similar neuronal protective properties, SC2 showed a higher extraction yield, higher efficacy in the reversion of H2O2-induced cytotoxicity in rat pheochromocytoma PC-12 cells, and lower impurities compared with SC1, and thus SC2 is suggested as a good candidate for a therapeutic agent in the preventive treatment of neurodegenerative diseases. A fiber-optic delay based strain sensor with high precision and temperature insensitivity was reported, which works on detecting the delay induced by strain instead of spectrum. In order to analyze the working principle of this sensor, the elastic property of fiber-optic delay was theoretically researched and the elastic coefficient was measured as 3.78 ps/km.με. In this sensor, an extra reference path was introduced to simplify the measurement of delay and resist the cross-effect of environmental temperature. Utilizing an optical fiber stretcher driven by piezoelectric ceramics, the performance of this strain sensor was tested. The experimental results demonstrate that temperature fluctuations contribute little to the strain error and that the calculated strain sensitivity is as high as 4.75 με in the range of 350 με. As a result, this strain sensor is proved to be feasible and practical, which is appropriate for strain measurement in a simple and economical way. Furthermore, on basis of this sensor, the quasi-distributed measurement could be also easily realized by wavelength division multiplexing and wavelength addressing for long-distance structure health and security monitoring. Data from EV-D68-infected patients demonstrate that pathological changes in the lower respiratory tract are principally characterized by severe respiratory illness in children and acute flaccid myelitis. However, lack of a suitable animal model for EV-D68 infection has limited the study on the pathogenesis of this critical pathogen, and the development of a vaccine. Ferrets have been widely used to evaluate respiratory virus infections. In the current study, we used EV-D68-infected ferrets as a potential animal to identify impersonal indices, involving clinical features and histopathological changes in the upper and lower respiratory tract (URT and LRT). The research results demonstrate that the EV-D68 virus leads to minimal clinical symptoms in ferrets. According to the viral load detection in the feces, nasal, and respiratory tracts, the infection and shedding of EV-D68 in the ferret model was confirmed, and these results were supported by the EV-D68 VP1 immunofluorescence confocal imaging with 2,6-linked sialic acid (SA) in lung tissues. Furthermore, we detected the inflammatory cytokine/chemokine expression level, which implied high expression levels of interleukin (IL)-1a, IL-8, IL-5, IL-12, IL-13, and IL-17a in the lungs. This study was compared and characterized two different alkali (potassium carbonate (PC) and potassium acetate (PA))-catalyzed acetylations of slicewood with vinyl acetate (VA) by a vapor phase reaction. The results revealed that the esterification reaction between VA and the hydroxyl groups of slicewood could be improved by using PC or PA as a catalyst. Additionally, a significant weight percent gain was obtained after VA acetylation with 5% of catalyst. Furthermore, the reactivity of the cellulose hydroxyl groups for VA acetylation was more pronounced at the C2 reactive site compared to acetylation with acetic anhydride. On the other hand, the apparent activation energy of thermal decomposition between 10% and 70% conversion is 174-183, 194-200, and 183-186 kJ/mol for unmodified slicewood and VA-acetylated slicewood with PC and PA, respectively. Yields of rainfed potato (Solanum tuberosum L.) in China's semiarid region are restricted by limited precipitation. Climate change could cause significant fluctuation in the rain-fed agricultural production due to the spatiotemporal changes in temperature and precipitation. As adjusting sowing dates proved to be an important management technique for improving grain yields, we examined how potato production can be improved by altering planting date in China's semiarid region, thus improving potato adaptation to climate change. Field trials with five target sowing dates (from late April to early June) were carried out in Inner Mongolia, China during four growing seasons (2010-2013), and the effects of sowing dates on potato development, yield and water use efficiency (WUE) were estimated. Results showed that although delayed sowing shortened the duration of potato growth period, non-significant thermal time differences after flowering stage was found among the treatments. However, greater precipitation was shown in three intermediate treatments. Potato yield was significantly affected by sowing dates, and intermediate sowing dates showed greater yield compared to the earlier or later sowing dates. Delayed sowing dates significantly improved WUE in a drier year. As for a normal year, earlier sowing dates promoted WUE because of less water consumption, but these increases came at the cost of reducing yields. Under the current climate conditions in the study area, the flexible sowing time for potato was determined as from early May to early June, and optimum sowing time was between 10 May and 27 May. In conclusion, adjusting sowing date affected duration, thermal time, and precipitation over potato growth period, and the optimal sowing date exhibited higher yield by obtaining greater precipitation, which could improve potato adaption to climate change. In order to meet the requirements of cable fault detection, a new structure of piezoelectric accelerometer was designed and analyzed in detail. The structure was composed of a seismic mass, two sensitive beams, and two added beams. Then, simulations including the maximum stress, natural frequency, and output voltage were carried out. Moreover, comparisons with traditional structures of piezoelectric accelerometer were made. To verify which vibration mode is the dominant one on the acceleration and the space between the mass and glass, mode analysis and deflection analysis were carried out. Fabricated on an n-type single crystal silicon wafer, the sensor chips were wire-bonged to printed circuit boards (PCBs) and simply packaged for experiments. Finally, a vibration test was conducted. The Pingshuo Mine is an important coal mine of the Ningwu coalfield in northern Shanxi Province, China. To investigate the mineralogy and geochemistry of Pingshuo coals, core samples from the mineable No. 4 coals were collected. The minerals, major element oxides, and trace elements were analyzed by scanning electron microscopy (SEM), LTA-XRD in combination with Siroquant software, X-ray fluorescence (XRF), inductively coupled plasma mass spectrometry (ICP-MS) and ICP-CCT-MS (As and Se). The minerals in the Pennsylvanian coals from the Pingshuo Mine dominantly consist of kaolinite and boehmite, with minor amounts of siderite, anatase, goyazite, calcite, apatite and florencite. Major-element oxides including SiO2 (9.54 wt %), Al2O3 (9.68 wt %), and TiO2 (0.63 wt %), as well as trace elements including Hg (449.63 ng/g), Zr (285.95 µg/g), Cu (36.72 µg/g), Ga (18.47 µg/g), Se (5.99 µg/g), Cd (0.43 µg/g), Hf (7.14 µg/g), and Pb (40.63 µg/g) are enriched in the coal. Lithium and Hg present strong positive correlations with ash yield and SiO2, indicating an inorganic affinity. Elements Sr, Ba, Be, As and Ga have strong positive correlations with CaO and P2O5, indicating that most of these elements may be either associated with phosphates and carbonates or have an inorganic--organic affinity. A hybrid quantum mechanics (QM)/molecular dynamics (MD) simulation is performed to investigate the effect of an ionizable group (-SO3-Na+) on polyaniline as gas sensing materials. Polymers considered for this work include emeraldine base of polyaniline (EB-PANI) and its derivatives (Na-SPANI (I), (II) and (III)) whose rings are partly monosubstituted by -SO3-Na+. The hybrid simulation results show that the adsorption energy, Mulliken charge and band gap of analytes (CO2 and H2O) in polyaniline are relatively sensitive to the position and the amounts of -SO3-Na+, and these parameters would affect the sensitivity of Na-SPANI/EB-PANI towards CO2. The sensitivity of Na-SPANI (III)/EB-PANI towards CO2 can be greatly improved by two orders of magnitude, which is in agreement with the experimental study. In addition, we also demonstrate that introducing -SO3-Na+ groups at the rings can notably affect the gas transport properties of polyaniline. Comparative studies indicate that the effect of ionizable group on polyaniline as gas sensing materials for the polar gas molecule (H2O) is more significant than that for the nonpolar gas molecule (CO2). These findings contribute in the functionalization-induced variations of the material properties of polyaniline for CO2 sensing and the design of new polyaniline with desired sensing properties. The impacts of high technology industries have been growing increasingly to technological innovations and global economic developments, while the concerns in sustainability are calling for facilitating green materials and cleaner production in the industrial value chains. Today's manufacturing companies are not striving for individual capacities but for the effective working with green supply chains. However, in addition to environmental and social objectives, cost and economic feasibility has become one of the most critical success factors for improving supply chain management with green component procurement collaboration, especially for the electronics OEM (original equipment manufacturing) companies whose procurement costs often make up a very high proportion of final product prices. This paper presents a case study from the systems perspective by using System Dynamics simulation analysis and statistical validations with empirical data. Empirical data were collected from Taiwanese manufacturing chains--among the world's largest manufacturing clusters of high technology components and products--and their global green suppliers to examine the benefits of green component procurement collaborations in terms of shared costs and improved shipping time performance. Two different supply chain collaboration models, from multi-layer ceramic capacitor (MLCC) and universal serial bus 3.0 (USB 3.0) cable procurements, were benchmarked and statistically validated. The results suggest that the practices of collaborative planning for procurement quantity and accurate fulfillment by suppliers are significantly related to cost effectiveness and shipping time efficiency. Although the price negotiation of upstream raw materials for the collaborative suppliers has no statistically significant benefit to the shipping time efficiency, the shared cost reduction of component procurement is significantly positive for supply chain collaboration among green manufacturers. This paper reports the mineralogy and geochemistry of the Early Permian No. 5 coal from the Chuancaogedan Mine, Junger Coalfield, China, using optical microscopy, scanning electron microscopy (SEM), Low-temperature ashing X-ray diffraction (LTA-XRD) in combination with Siroquant software, X-ray fluorescence (XRF), and inductively coupled plasma mass spectrometry (ICP-MS). The minerals in the No. 5 coal from the Chuancaogedan Mine dominantly consist of kaolinite, with minor amounts of quartz, pyrite, magnetite, gypsum, calcite, jarosite and mixed-layer illite/smectite (I/S). The most abundant species within high-temperature plasma-derived coals were SiO2 (averaging 16.90%), Al2O3 (13.87%), TiO2 (0.55%) and P2O5 (0.05%). Notable minor and trace elements of the coal include Zr (245.89 mg/kg), Li (78.54 mg/kg), Hg (65.42 mg/kg), Pb (38.95 mg/kg), U (7.85 mg/kg) and Se (6.69 mg/kg). The coal has an ultra-low sulfur content (0.40%). Lithium, Ga, Se, Zr and Hf present strongly positive correlation with ash yield, Si and Al, suggesting they are associated with aluminosilicate minerals in the No. 5 coal. Arsenic is only weakly associated with mineral matter and Ge in the No. 5 coals might be of organic and/or sulfide affinity. [ABSTRACT FROM AUTHOR]

Lipases from microorganisms have multi-faceted properties and play an important role in ever-growing modern biotechnology and, consequently, it is of great significance to develop new ones. In the present work, a lipase gene from Candida albicans (CaLIP10) was cloned and two non-unusual CUG serine codons were mutated into universal codons, and its expression in Pichia pastoris performed optimally, as shown by response surface methodology. Optimal conditions were: initial pH of culture 6.86, temperature 25.53 °C, 3.48% of glucose and 1.32% of yeast extract. The corresponding maximal lipolytic activity of CaLIP10 was 8.06 U/mL. The purified CaLIP10 showed maximal activity at pH 8.0 and 25 °C, and a good resistance to non-ionic surfactants and polar organic solvent was noticed. CaLIP10 could effectively hydrolyze coconut oil, but exhibited no obvious preference to the fatty acids with different carbon length, and diacylglycerol was accumulated in the reaction products, suggesting that CaLIP10 is a potential lipase for the oil industry. A novel lipase gene lip5 from the yeast Candida albicans was cloned and sequenced. Alignment of amino acid sequences revealed that 86-34% identity exists with lipases from other Candida species. The lipase and its mutants were expressed in the yeast Pichia pastoris, where alternative codon usage caused the mistranslation of 154-Ser and 293-Ser as leucine. 154-Ser to leucine resulted in loss of expression of Lip5, and 293-Ser to leucine caused a marked reduction in the lipase activity. Lip5-DM, which has double mutations that revert 154 and 293 to serine residues, showed good lipase activity, and was overexpressed and purified by (NH4)2SO4 precipitation and ion-exchange chromatography. The pure Lip5-DM was stable at low temperatures ranging from 15-35 °C and pH 5-9, with the optimal conditions being 15-25 °C and pH 5-6. The activation energy of recombinant lipase was 8.5 Kcal/mol between 5 and 25 °C, suggesting that Lip5-DM was a cold-active lipase. Its activity was found to increase in the presence of Zn2+, but it was strongly inhibited by Fe2+, Fe3+, Hg2+ and some surfactants. In addition, the Lip5-DM could not tolerate water-miscible organic solvents. Lip5-DM exhibited a preference for the short- and medium-chain length p-nitrophenyl (C4 and C8 acyl group) esters rather than the long chain length p-nitrophenyl esters (C12, C16 and C18 acyl group) with highest activity observed with the C8 derivatives. The subsea observation network has become an indispensable means of ocean exploration worldwide. However, the scale of the subsea observation network is limited by the power supply voltage and power level. Hence, to promote the development of a subsea observation network, this paper investigates the underwater high voltage DC-DC converter (HVC), which greatly improves the voltage and power level of the subsea observation network. The traditional series-parallel converter based on multi-module is faced with many technical problems, such as difficult transformer isolation, many fault points, low power density under higher input voltage level, and higher output power. The subsea HVC of this paper adopts a modular multilevel resonant DC-DC converter. The main circuit of HVC is designed in detail, including a module circuit, a resonant circuit, and a control scheme. Through the combination of the sub-module removal voltage regulation and closed-loop control, the converter can still output a stable voltage of 375 V when the input voltage changes. The modular sub-module and centralized transformer structure enables the converter to isolate high voltage easily, small volume, and high power density. The simulation and experiment results show the proposed HVC meets the design requirements and has good application prospects. It can be applied to submarine power transmission and distribution needs because of its wide range, large transformation ratio, and high efficiency. [ABSTRACT FROM AUTHOR]

Trafficability gives tracked vehicles adaptability, stability, and propulsion for various purposes, including deep-sea research in rough terrain. Terrain characteristics affect tracked vehicle mobility. This paper investigates the soil mechanical interaction dynamics between rubber-tracked vehicles and sedimental soils through controlled laboratory-simulated experiments. Focusing on Bentonite and Diatom sedimental soils, which possess distinct shear properties from typical land soils, the study employs innovative user-written subroutines to characterize mechanical models linked to the RecurDyn simulation environment. The experiment is centered around a dual-tracked crawler, which in itself represents a fully independent vehicle. A new three-dimensional multi-body dynamic simulation model of the tracked vehicle is developed, integrating the moist terrain's mechanical model. Simulations assess the vehicle's trafficability and performance, revealing optimal slip ratios for maximum traction force. Additionally, a mathematical model evaluates the vehicle's tractive trafficability based on slip ratio and primary design parameters. The study offers valuable insights and a practical simulation modeling approach for assessing trafficability, predicting locomotion, optimizing design, and controlling the motion of tracked vehicles across diverse moist terrain conditions. The focus is on the critical factors influencing the mobility of tracked vehicles, precisely the sinkage speed and its relationship with pressure. The study introduces a rubber-tracked vehicle, pressure, and moisture sensors to monitor pressure sinkage and moisture, evaluating cohesive soils (Bentonite/Diatom) in combination with sand and gravel mixtures. Findings reveal that higher moisture content in Bentonite correlates with increased track slippage and sinkage, contrasting with Diatom's notable compaction and sinkage characteristics. This research enhances precision in terrain assessment, improves tracked vehicle design, and advances terrain mechanics comprehension for off-road exploration, offering valuable insights for vehicle design practices and exploration endeavors.
