Simple Summary: The common bottlenose dolphin, Tursiops truncatus, is a worldwide cetacean species essential for marine ecosystems' health and balance. Understanding the genetic connectivity and structure of different populations is crucial for the correct management and conservation of a species, such as designing Special Areas of Conservation or Marine Protected Areas. In this study, we described the genetic composition of 49 bottlenose dolphins from the Canary Islands, which were previously unstudied, and compared them with individuals from the rest of the North-East Atlantic Ocean. The results showed that Canarian bottlenose dolphins have a remarkably diverse genetic composition, and this population is possibly part of a larger oceanic population in the North Atlantic. Therefore, the studied Special Areas of Conservation in the Canary Islands may correspond to a hotspot of genetic diversity and could be a strategic area for the conservation of the species. In recent decades, worldwide cetacean species have been protected, but they are still threatened. The bottlenose dolphin (Tursiops truncatus) is a vulnerable keystone species and a useful bioindicator of the health and balance of marine ecosystems in oceans all over the world. The genetic structure of the species is shaped by their niche specialization (along with other factors), leading to the classification of two ecotypes: coastal and pelagic. In this study, the genetic diversity, population structure, and ecotypes of bottlenose dolphins from the Canary Islands were assessed through the analysis of 49 new samples from biopsies and from stranded animals using the 636 bp portion of the mitochondrial control region and 343 individuals from databases (n = 392). The results reveal high genetic diversity in Canarian bottlenose dolphins (Hd = 0.969 and π = 0.0165) and the apparent lack of population genetic structure within this archipelago. High genetic structure (Fst, Φst) was found between the Canary Islands and coastal populations, while little to no structure was found with the pelagic populations. These results suggest that Canarian bottlenose dolphins are part of pelagic ecotype populations in the North Atlantic. The studied Special Areas of Conservation in the Canary Islands may correspond to a hotspot of genetic diversity of the species and could be a strategic area for the conservation of the oceanic ecotype of bottlenose dolphins. Geryon longipes is a crab species that inhabits the muddy bottoms of the middle and lower slopes, as well as bathyal bottoms ranging from 400 to 2000 m in depth. To assess its molecular diversity, a fragment of 572 bp of the COI (Cytochrome Oxidase subunit I) mitochondrial gene was sequenced in eight Western Mediterranean locations. Within the studied area, two oceanographic fronts are present (Almeria-Oran Front and Ibiza Channel). From the 124 sequences obtained, only 7 distinct haplotypes were identified. The population distribution indicated three well-differentiated regions: the Alboran Sea, the Gulf of Vera and the Levantine/Catalan coasts. The molecular diversity was compared with that obtained in the same year for the same gene in Liocarcinus depurator, a crab species that is captured on the continental shelf and upper slope (40 to 500 m). The estimates of molecular diversity parameters for the COI gene fragment were rather similar between both species, but the number of haplotypes was higher for L. depurator. Finally, the obtained COI sequences of G. longipes were compared to those from other populations of the species distribution range, recovered from the DNA repository. Only one additional, different haplotype was reported (Sicily), whereas all the rest were common with those described in our study. Therefore, the COI gene fragment would indicate that all the sequences analysed in the Mediterranean and NE Atlantic belong to the same species, G. longipes. [ABSTRACT FROM AUTHOR]

Pulmonary surfactant (PS) has been proposed as an efficient drug delivery vehicle for inhaled therapies. Its ability to adsorb and spread interfacially and transport different drugs associated with it has been studied mainly by different surface balance designs, typically interconnecting various compartments by interfacial paper bridges, mimicking in vitro the respiratory air–liquid interface. It has been demonstrated that only a monomolecular surface layer of PS/drug is able to cross this bridge. However, surfactant films are typically organized as multi-layered structures associated with the interface. The aim of this work was to explore the contribution of surface-associated structures to the spreading of PS and the transport of drugs. We have designed a novel vehiculization balance in which donor and recipient compartments are connected by a whole three-dimensional layer of liquid and not only by an interfacial bridge. By combining different surfactant formulations and liposomes with a fluorescent lipid dye and a model hydrophobic drug, budesonide (BUD), we observed that the use of the bridge significantly reduced the transfer of lipids and drug through the air–liquid interface in comparison to what can be spread through a fully open interfacial liquid layer. We conclude that three-dimensional structures connected to the surfactant interfacial film can provide an important additional contribution to interfacial delivery, as they are able to transport significant amounts of lipids and drugs during surfactant spreading. Wine is a complex matrix composed of numerous substances and color has an important influence on its quality and consumer acceptance. Color is affected by numerous factors such as pre-fermentation and fermentation operations, ageing, contact or addition of certain substances. In this study, different chromatic parameters were determined in 99 wines (58 red and 41 white) made from Galician (Northwest Spain) grape varieties. These parameters were obtained by using simple, rapid, and inexpensive spectrophotometric methodologies: color intensity, hue, total polyphenols content (Total Polyphenol Index TPI, Folin–Ciocalteu index, FCI), total anthocyans, total tannins, and color coordinates measured by the CIELab system. The influence of ageing in barrels (red wines) or using chips (white and red wines) on these parameters was also studied. A principal component analysis (PCA) was carried out to characterize the wines according to their chromatic characteristics. Application of PCA to the experimental data resulted in satisfactory classifications of studied white and red wines according to the cited enological practices. [ABSTRACT FROM AUTHOR]

A fast skin clearance is the main goal to achieve in psoriasis treatment. Patients that present a fast and exceptional improvement with treatment are called super-responders (SR). There is no consensus on the definition of SR with respect to psoriasis. Included herein is a retrospective analysis of a multicenter, observational study of real clinical practices including patients with moderate-to-severe plaque PSO undergoing treatment with Guselkumab (GUS). This cross-sectional analysis includes information on patients between February 2019 to February 2022. A SR is a patient that achieved a PASI = 0 at weeks 12 and 24. Analyses have been performed "as observed" using GraphPad Prism version 8.3.0 for Windows (GraphPad Software, San Diego, CA, USA, At baseline, the PASI is significantly correlated with VAS_pruritus, BSA, and DLQI, while DLQI is significantly correlated with VAS_pruritus. Significant correlations increase in number and magnitude over the follow-up time. In relation to the univariate logistic models carried out, only three variables showed a significant association with the super-responder variable: depression, VAS_pruritus, and DLQI.SR patients, who show a faster evolution in PASI and BSA improvement than non-SRs. Simple Summary: Black-shouldered Kite (Elanus caeruleus) is a relative recent colonizer in Europe. The study of juvenile dispersal may help to better understand the patterns of range expansion and colonization. In this study, we provide some information about patterns of juvenile dispersal according to sex, habitat quality, timing of reproduction, and nesting hatching order to test two competing hypotheses about natal dispersal in this poorly studied raptor. We found some evidence supporting the Resources Competition Hypothesis since nestlings hatched from high quality territories and hatched first within the brood stayed closer from natal areas than nestlings hatched from low quality territories or later hatched nestlings. Knowledge of animal dispersal patterns is of great importance for the conservation and maintenance of natural populations. We here analyze juvenile dispersal of the poorly studied Black-shouldered Kite (Elanus caeruleus) monitored in southwestern Spain in an ongoing long-term study initiated in 2003. The European population of Black-shouldered kites is thought to be a recent one funded by colonizing African birds, as no kites have been found in the European fossil record, and the breeding population has progressively expanded to the North in the late 20th and 21st centuries. We obtained information on movements behavior during dispersal from 47 juveniles Kites after marking 384 nestlings with wing tags and three nestlings with radio transmitter. We have tested two competing hypotheses (i.e., the Resources Competition Hypothesis and the Wandering Hypothesis (WH)) that may explain the leptokurtic distribution of the natal dispersal distance in Elanus. After independence, juvenile females dispersed farther from the natal areas than males, as is common in birds. On average, males and females dispersed from their natal areas over 9 (i.e., 26.15 km) and 15 (i.e., 43.79 km) breeding territories, respectively. A male and two females dispersed further than 100 km from their natal nest. Our results indicated some evidence supporting the competition-for-resources hypotheses since nestlings hatched from high quality territories stayed closer from natal areas than nestlings hatched from low quality territories and also nestlings hatched first within the brood also tend to recruit closer to their natal area than later hatched nestlings which tend to disperse further away from their natal area. The information provided by these crucial demographic parameters will be used for the elaboration of future conservation plans for the management of this colonizing species in Europe. Polyhydroxyalkanoates (PHA) are polyesters produced intracellularly by many bacterial species as energy storage materials, which are used in biomedical applications, including drug delivery systems, due to their biocompatibility and biodegradability. In this study, we evaluated the potential application of this nanomaterial as a basis of inhaled drug delivery systems. To that end, we assessed the possible interaction between PHA nanoparticles (NPs) and pulmonary surfactant using dynamic light scattering, Langmuir balances, and epifluorescence microscopy. Our results demonstrate that NPs deposited onto preformed monolayers of DPPC or DPPC/POPG bind these surfactant lipids. This interaction facilitated the translocation of the nanomaterial towards the aqueous subphase, with the subsequent loss of lipid from the interface. NPs that remained at the interface associated with liquid expanded (LE)/tilted condensed (TC) phase boundaries, decreasing the size of condensed domains and promoting the intermixing of TC and LE phases at submicroscopic scale. This provided the stability necessary for attaining high surface pressures upon compression, countering the destabilization induced by lipid loss. These effects were observed only for high NP loads, suggesting a limit for the use of these NPs in pulmonary drug delivery. [ABSTRACT FROM AUTHOR]

Pulmonary surfactant is a lipid/protein complex synthesized by the alveolar epithelium and secreted into the airspaces, where it coats and protects the large respiratory air–liquid interface. Surfactant, assembled as a complex network of membranous structures, integrates elements in charge of reducing surface tension to a minimum along the breathing cycle, thus maintaining a large surface open to gas exchange and also protecting the lung and the body from the entrance of a myriad of potentially pathogenic entities. Different molecules in the surfactant establish a multivalent crosstalk with the epithelium, the immune system and the lung microbiota, constituting a crucial platform to sustain homeostasis, under health and disease. This review summarizes some of the most important molecules and interactions within lung surfactant and how multiple lipid–protein and protein–protein interactions contribute to the proper maintenance of an operative respiratory surface. Two decades since the discovery of the RNA interference (RNAi) pathway, we are now witnessing the approval of the first RNAi-based treatments with small interfering RNA (siRNA) drugs. Nevertheless, the widespread use of siRNA is limited by various extra- and intracellular barriers, requiring its encapsulation in a suitable (nanosized) delivery system. On the intracellular level, the endosomal membrane is a major barrier following endocytosis of siRNA-loaded nanoparticles in target cells and innovative materials to promote cytosolic siRNA delivery are highly sought after. We previously identified the endogenous lung surfactant protein B (SP-B) as siRNA delivery enhancer when reconstituted in (proteo) lipid-coated nanogels. It is known that the surface-active function of SP-B in the lung is influenced by the lipid composition of the lung surfactant. Here, we investigated the role of the lipid component on the siRNA delivery-promoting activity of SP-B proteolipid-coated nanogels in more detail. Our results clearly indicate that SP-B prefers fluid membranes with cholesterol not exceeding physiological levels. In addition, SP-B retains its activity in the presence of different classes of anionic lipids. In contrast, comparable fractions of SP-B did not promote the siRNA delivery potential of DOTAP:DOPE cationic liposomes. Finally, we demonstrate that the beneficial effect of lung surfactant on siRNA delivery is not limited to lung-related cell types, providing broader therapeutic opportunities in other tissues as well.
