Background and Objectives: This study aimed to investigate the relationship between neuropathic pain and CREB-binding protein (CBP) and methyl-CpG-binding protein 2 (MeCP2) expression levels in a rat model with spared nerve injury (SNI). Materials and Methods: Rat (male Sprague-Dawley white rats) models with surgical SNI (n = 6) were prepared, and naive rats (n = 5) were used as controls. The expression levels of CBP and MeCP2 in the spinal cord and dorsal root ganglion (DRG) were compared through immunohistochemistry at 7 and 14 days after surgery. The relationship between neuropathic pain and CBP/MeCP2 was also analyzed through intrathecal siRNA administration. Results: SNI induced a significant increase in the number of CBPs in L4 compared with contralateral DRG as well as with naive rats. The number of MeCP2 cells in the dorsal horn on the ipsilateral side decreased significantly compared with the contralateral dorsal horn and the control group. SNI induced a significant decrease in the number of MeCP2 neurons in the L4 ipsilateral DRG compared with the contralateral DRG and naive rats. The intrathecal injection of CBP siRNA significantly inhibited mechanical allodynia induced by SNI compared with non-targeting siRNA treatment. MeCP2 siRNA injection showed no significant effect on mechanical allodynia. Conclusions: The results suggest that CBP and MeCP2 may play an important role in the generation of neuropathic pain following peripheral nerve injury. [ABSTRACT FROM AUTHOR]

Blended cement is commonly used for producing sustainable concretes. This paper presents an experimental study and an optimization design of a low-CO2 quaternary binder containing calcined clay, slag, and limestone using the response surface method. First, a Box–Behnken design with three influencing factors and three levels was used for the combination design of the quaternary composite cement. The lower limit of the mineral admixtures was 0%. The upper limits of slag, calcined clay, and limestone powder were 30%, 20%, and 10%, respectively. The water-to-binder ratio (water/binder) was 0.5. Experimental works to examine workability and strength (at 3 and 28 days) were performed for the composite cement. The CO2 emissions were calculated considering binder compositions. A second-order polynomial regression was used to evaluate the experimental results. In addition, a low-CO2 optimization design was conducted for the composite cement using a composite desirability function. The objectives of the optimization design were the target 28-day strength (30, 35, 40, and 45 MPa), target workability (160 mm flow), and low CO2 emissions. The trends of the properties of optimal combinations were consistent with those in the test results. Studies regarding the variables that could predict the success of conservative treatment for knee hemarthrosis are lacking. This retrospective study evaluated the laboratory variables of patients who had unsatisfactory results from conservative treatment for knee hemarthrosis. Twenty-nine patients conservatively treated for knee hemarthrosis were included and divided into two groups: group A comprised 14 patients who underwent interventional angiography and selective embolization due to failed conservative treatment, and group B comprised 15 patients with successful results after conservative treatment. The results of the serological and synovial fluid tests were evaluated. The mean number of synovial red blood cells (RBCs) was 1,905,857 cells/µL and 7730 cells/µL in groups A and B, respectively (p = 0.01), while the mean number of RBCs per high-power field (HPF) was 68.9 and 3.2, respectively (p < 0.01). Patients who underwent interventional angiography and selective embolization after failed conservative treatment for knee hemarthrosis had higher synovial RBC counts and RBC counts per HPF than those with successful outcomes after conservative treatment. It is necessary to carefully interpret the results of the synovial fluid analysis in patients with knee hemarthrosis; if the synovial fluid analysis shows a synovial RBC count greater than 81,500 and RBC count per HPF greater than 16.3, we recommend immediate interventional angiography rather than continuing conservative treatment. Metakaolin is reactive and is widely used in the modern concrete industry. This study presents an integrated strength–sustainability evaluation framework, which we employed in the context of metakaolin content in concrete. First, a composite hydration model was employed to calculate reactivity of metakaolin and cement. Furthermore, a hydration-based linear equation was designed to evaluate the compressive strength development of metakaolin composite concrete. The coefficients of the strength evaluation model are constants for different mixtures and ages. Second, the sustainability factors—CO2 emissions, resource consumption, and energy consumption—were determined based on concrete mixtures. Moreover, the sustainability factors normalized for unit strength were obtained based on the ratios of total CO2 emissions, energy consumption, and resource consumption to concrete strength. The results of our analysis showed the following: (1) As the metakaolin content increased, the normalized CO2 emissions and resource consumption decreased, and the normalized energy first decreased and then slightly increased. (2) As the concrete aged from 28 days to three months, the normalized CO2 emissions, resource consumption, and energy consumption decreased. (3) As the water/binder ratio decreased, the normalized CO2 emissions, resource consumption, and energy consumption decreased. Slag is increasingly unitized for the production of sustainable concrete. This paper presents a procedure with which to analyze the property development of slag composite concrete. Experimental studies of the hydration heat and compressive strength development and simulation studies using a kinetic hydration model and a thermodynamic model were performed. First, we performed an experimental study of the isothermal hydration heat of cement–slag blends. Based on the results of the experimental study on cumulative hydration heat, the reaction degree of slag was determined. We found that the reaction degree of slag decreased as the slag content increased. Second, the reaction degree of slag and cement were used as the input parameters for the Gibbs energy minimization (GEM) thermodynamic equilibrium model. Moreover, the phase assemblage of hydrating cement–slag was determined. The trends of calcium silicate hydrate (CSH) are similar to those of strength. Based on the CSH content, the strength of hardening cement–slag blends was determined. In addition, the calcium hydroxide (CH) content resulting from the thermodynamic model shows good agreement with the experimental results. High-calcium fly ash (FH) is the combustion residue from electric power plants burning lignite or sub-bituminous coal. As a mineral admixture, FH can be used to produce high-strength concrete and high-performance concrete. The development of chemical and mechanical properties is a crucial factor for appropriately using FH in the concrete industry. To achieve sustainable development in the concrete industry, this paper presents a theoretical model to systematically evaluate the property developments of FH blended concrete. The proposed model analyzes the cement hydration, the reaction of free CaO in FH, and the reaction of phases in FH other than free CaO. The mutual interactions among cement hydration, the reaction of free CaO in FH, and the reaction of other phases in FH are also considered through the calcium hydroxide contents and the capillary water contents. Using the hydration degree of cement, the reaction degree of free CaO in FH, and the reaction degree of other phases in FH, the proposed model evaluates the calcium hydroxide contents, the reaction degree of FH, chemically bound water, porosity, and the compressive strength of hardening concrete with different water to binder ratios and FH replacement ratios. The evaluated results are compared to experimental results, and good consistencies are found.
