Camellia luteoflora is a rare and endangered plant endemic to China. It has high ornamental and potential economic and medicinal value, and is an important germplasm resource of Camellia. To understand the distributions and differences in metabolites from different parts of C. luteoflora, in this study, we used liquid chromatography–tandem mass spectrometry (LC–MS/MS) to examine the types and contents of chemical constituents in five organs of C. luteoflora: roots, stems, leaves, flowers, and fruits. The results showed that a total of 815 metabolites were identified in the five organs and were classified into 18 main categories, including terpenoids (17.1%), amino acids (10.4%), flavonoids (10.3%), sugars and alcohols (9.8%), organic acids (9.0%), lipids (7.1%), polyphenols (4.8%), alkaloids (4.8%), etc. A total of 684 differentially expressed metabolites (DEMs) in five organs were obtained and annotated into 217 KEGG metabolic pathways, among which metabolic pathways, ABC transporters, the biosynthesis of cofactors, and the biosynthesis of amino acids were significantly enriched. In DEMs, flowers are rich in flavonoids, polyphenols, organic acids, and steroids; fruits are rich in amino acids, alkaloids, vitamins, and xanthones; stems are rich in lignans; and leaves have the highest relative content of phenylpropanoids, ketoaldehydic acids, quinones, sugars and alcohols, terpenoids, coumarins, lipids, and others; meanwhile, the metabolite content is lower in roots. Among the dominant DEMs, 58 were in roots, including arachidonic acid, lucidone, isoliquiritigenin, etc.; 75 were in flowers, including mannose, shikimic acid, d-gluconic acid, kaempferol, etc.; 45 were in the fruit, including pterostilbene, l-ascorbic acid, riboflavin, etc.; 27 were in the stems, including salicylic acid, d-(-)-quinic acid, mannitol, (-)-catechin gallate, etc.; there was a maximum number of 119 dominant metabolites in the leaves, including oleanolic acid, l-glucose, d-arabitol, eugenol, etc. In sum, the rich chemical composition of C. luteoflora and the significant differences in the relative contents of metabolites in different organs will provide theoretical references for the study of tea, flower tea, edible oil, nutraceuticals, and the medicinal components of C. luteoflora. The tropical Garcinia genus of flowering plants is a prolific producer of aromatic natural products including polyphenols, flavonoids, and xanthones. In this study, we report the first phytochemical investigation of Garcinia caudiculata Ridl. from the island of Borneo. Fractionation, purification, and structure elucidation by MS and NMR resulted in the discovery of two meroterpenoids. One was a benzofuranone lactone previously isolated from Iryanthera grandis and Rhus chinensis, and the second was a new hydroquinone methyl ester that we named caudiquinol. The river valley forests of the Irtysh River Basin are a germplasm bank of Salicaceae species and rare plant resources in China, and the distribution varies with the river and is highly distinctive. However, there is a dearth of systematic research on the characteristics of plant resources. In this study, a comprehensive investigation was conducted in the trunk stream and six tributaries with valley forest distribution in the Irtysh River Basin, and 244 quadrats were set up. The analysis focused on the composition of the flora and resource characteristics. The results reveal the following: (1) The valley forests of the Irtysh River Basin contain 256 species of plants belonging to 57 families and 178 genera, among which 19 species of trees, 23 species of shrubs, and 214 species of herbs were investigated. (2) Among the identified species, 226 (88.67%) were recognized as resource plants, with medicinal plants being the most abundant (176 species, 68.75% of the total). (3) The distribution patterns of trees, shrubs, and herbs of each resource type vary across rivers. Elevation drop, river length, and river distance all significantly affect the number of specie. This study elucidated the current status and distributional characteristics of plant resources in the valley forests of the Irtysh River Basin, which is essential for both biodiversity conservation and sustainable resource utilization. The rare and endangered plant species Abies fanjingshanensis, which has a limited habitat, a limited distribution area, and a small population, is under severe threat, particularly due to poor leaf health. The plant endophytic microbiome is an integral part of the host, and increasing evidence indicates that the interplay between plants and endophytic microbes is a key determinant for sustaining plant fitness. However, little attention has been given to the differences in the endophytic microbial community structure, network complexity, and assembly processes in leaves with different leaf health statuses. Here, we investigated the endophytic bacterial and fungal communities in healthy leaves (HLs) and non-healthy leaves (NLs) of A. fanjingshanensis using 16S rRNA gene and internal transcribed spacer sequencing and evaluated how leaf health status affects the co-occurrence patterns and assembly processes of leaf endophytic microbial communities based on the co-occurrence networks, the niche breadth index, a neutral community model, and C-score metrics. HLs had significantly greater endophytic bacterial and fungal abundance and diversity than NLs, and there were significant differences in the endophytic microbial communities between HLs and NLs. Leaf-health-sensitive endophytic microbes were taxonomically diverse and were mainly grouped into four ecological clusters according to leaf health status. Poor leaf health reduced the complexity of the endophytic bacterial and fungal community networks, as reflected by a decrease in network nodes and edges and an increase in degrees of betweenness and assortativity. The stochastic processes of endophytic bacterial and fungal community assembly were weakened, and the deterministic processes became more important with declining leaf health. These results have important implications for understanding the ecological patterns and interactions of endophytic microbial communities in response to changing leaf health status and provide opportunities for further studies on exploiting plant endophytic microbes to conserve this endangered Abies species. The reproduction of rare and endangered plant species is one of the most important factors determining the stability and survival of their populations, and knowledge of the barriers to successful reproduction is essential for species conservation. Habitat loss and slow reproduction due to low fruit set are usually considered the main threats to Cephalanthera longifolia and C. rubra (Orchidaceae). The aim of this study was to analyse the natural fruit set of these species during three consecutive years in Lithuania in the northern part of the temperate zone of Europe. Six populations of C. longifolia and three populations of C. rubra were studied each year from 2021 to 2023. During the study period, 49.3% to 54.4% of C. longifolia and 40.0% to 54.3% of C. rubra individuals produced no fruit. Over the three-year period, fruit set in individual populations of C. longifolia ranged from 5.2% to 19.5%, whereas fruit set in populations of C. rubra ranged from 4.1% to 18.8%. Significant weak or moderate correlations were found between plant height, inflorescence length and the number of flowers in the inflorescence and fruit set of both species. Flower position in the inflorescence had a significant effect on fruit set in both species, and the fruit set rate of lower flowers was higher than that of upper flowers. Significant but weak correlations were found between the fruit set and most of the environmental factors analysed. The results of this study suggest that the fruit set of C. longifolia and C. rubra is dependent on insect pollination of the flowers, which in turn is affected by habitat conditions. Nymphoides coronata is an endangered aquatic plant species with significant medicinal and ecological importance. To preserve N. coronata from going extinct, we need to provide seedlings and efficient multiplication techniques so that it can be extensively studied. This study aimed to identify the most suitable sterilization treatment, growth medium, and substrate for the cultivation and propagation of N. coronata. Ethanol sterilization, fungicide treatment, and sterile water washing were the most important sterilization steps. A combination of 6-benzylaminopurine (6-BA) and indoleacetic acid (IAA) was the most suitable medium for bud induction and shoot proliferation. The use of α-naphthaleneacetic acid (NAA) increased the rooting rate and rooting time compared to indole-3-butyric acid (IBA). Increasing the concentration of NAA from 0.5 to 1.0 mg/L increased the rooting rate from 78 to 100% and reduced the rooting time from 7 to 5 days. The survival rate of N. coronata seedlings was 100% in a mixture of red soil and sand (1:1, w/w). As a result, the procedure mentioned above could potentially be used to safely propagate this rare species on a large scale. These findings provide valuable insights into the optimal conditions for the successful cultivation and propagation of N. coronata, which can contribute to the conservation and sustainable use of this important rare plant species. This study investigates the host diversity and characteristics of the rare parasitic plant Silverberry-like taxillus (Taxillus yadoriki). Seogwipo, Jeju Island, where T. yadoriki grows naturally, was examined to clarify the diversity of host species, their location, and the part of the tree where T. yadoriki is attached. A total of 687 host trees were found. Taxillus yadoriki was intensively distributed in the lowlands below 200 m a.s.l. The host trees belonged to 40 species, four varieties, three cultivars, and two hybrids from 32 genera and 21 families, of which 23 taxa were identified as host plants for the first time in this study. Incredibly, this plant was found to be able to parasitize a variety of plants from 12 orders. Taxillus yadoriki was found to be parasitic in three orders of Gymnosperms, Ginkgoales, Cupressales, and Pinales, as well as nine orders of Angiosperms, including Magnoliales, Rosales, and Ericales. Rosaceae was the most diverse at 18.0%, and most species were broad-leaved and arboreal. Among them, Cryptomeria japonica and Litsea japonica had the highest frequency of parasitization and T. yadoriki was mainly attached to the upper part of the main tree and the main branch; as a result, T. yadoriki was distributed and showed specificity in the position of disturbance on the host. T. yadoriki's specific distribution model was built as a Maxent program. The area under the curve of the receiver operation characteristic was 0.948 ± 0.026 (mean ± standard deviation), and the performance of the constructed model was excellent. Of the total eight environmental factors, density of artistic forest and meant temperature of coldest quarter accounted for 75.3% of the total importance, which was the main distribution determinant of species. In the average distribution probability map of T. yadoriki, the critical probability for determining distribution was calculated to be 0.2898 ± 0.1018. Neuwiedia singapureana is a rare and endangered plant of the Apostasioideae subfamily. The Apostasioideae subfamily has a unique evolutionary status, as it is considered to be the most primitive group forming the base of the Orchidaceae evolutionary tree. Therefore, N. singapureana has high scientific research and conservation value. The endophytic fungal communities associated with orchids are rich and diverse, but few studies have investigated the endophytic fungi of Neuwiedia orchid plants. In the present study, the aim was to examine the endophytic fungal community structures associated with wild N. singapureana rhizomes and normal roots in the ground and with bare prop roots in the air at two sampling sites in China. High-throughput sequencing of nuclear ribosomal DNA fragments of the internal transcribed spacer regions was conducted, and cultivable methods were adopted. A total of 2161 endophytic fungal operational taxonomic units (OTUs) were obtained at a 97% sequence similarity threshold. The endophytic fungal diversity differed among the samples but not significantly. There were many more non-mycorrhizal endophytic fungal than orchid mycorrhizal (OM) fungal species detected in the N. singapureana orchid, about 98.33% OTUs of non-mycorrhizal fungi contrasting with 1.67% OTUs of potential orchid mycorrhizal fungi, among which Ceratobasidiaceae, Russulaceae, and Thelephoraceae were the dominant orchid mycorrhizal fungi. One culturable OM fungal Epulorhiza sp. isolated from the rhizome was capable of significantly promoting the seed germination and seedling growth of Dendrobium officinale and Epidendrum secundum orchids, respectively, with different efficiencies. These endophytic fungal strains with growth-promoting functions will provide materials for orchid conservation and for the study of the mechanisms underlying orchid symbiotic associations. The screening of rare plants from the Yucatan region and the known native plants in Mexico, that have been successfully introduced worldwide, has been conducted. Based on a literature analysis and a search of English and Spanish scientific information regarding botanical, plant biochemical, and antioxidant potential in databases such as Google Scholar, Scopus, Web of Knowledge, as well as the national databases of Mexico (Flora: Yucatan Peninsula (cicy.mx) and Especies endémicas|Biodiversidad Mexicana), rare or underutilized plants from the Yucatan region with antioxidant potential have been selected. The formulas of the most studied secondary metabolites of these selected rare plants are shown. Among the selected rare plants with antioxidant potential, the families Sapidaceae and Anacardiaceae had the highest number of representatives. Additionally, representatives from the families Annonaceae, Moraceae, Malpighiaceae, Solanaceae, Ebenaceae, Asteraceae, Ranunculaceae, and Leguminosae were also presented. The current scientific data analysis of selected rare plants from the Yucatan region, Mexico, provides significant background for their further use and introduction in not only the Yucatan region of Mexico, but also worldwide. The genus Phagnalon Cass. (Asteraceae) is composed of widely distributed species and most of them, due to the medicinal properties they possess, are widely used in folk medicine but also as spices in the culinary field. The polar and non-polar extracts, as well as the complex mixtures of their essential oils, from several Phagnalon species and ssp., have shown antibiotic, antiviral, cytotoxic, and several other biological properties. In this work, the chemical composition and the antimicrobial, cytotoxic, and antioxidant properties of the Jordan accession of Phagnalon sinaicum Bornm. & Kneuck. essential oil (EO), an extremely rare plant that grows in ravines in the Middle East, were investigated. The EO, analyzed by GC-MS, was found to be rich in terpenoid compounds, and, in particular, in oxygenated monoterpenes, with the main compound being artemisia ketone (22.3%), followed by α-thujone (17.7%), and santolin alcohol (14.8%). The EO had good antimicrobial activity, especially against Escherichia coli Gram-negative bacterium (3 mg/mL MIC values) and was also effective in counteracting in vitro biofilm formation. Furthermore, this EO showed low cytotoxicity against immortalized human keratinocytes lines, but had good antioxidant activity on the same eukaryotic cellular models. [ABSTRACT FROM AUTHOR]

Mining activities create disturbed and polluted areas in which revegetation is complicated, especially in northern areas. For the first time, the state of the ecosystems in the impact zone of tailings formed during the processing of rare earth element deposits in the Subarctic have been studied. This work aimed to reveal aspects of accumulation and translocation of trace and biogenic elements in plants (Avenella flexuosa (L.) Drejer, Salix sp., and Betula pubescens Ehrh.) that are predominantly found in primary ecosystems on the tailings of loparite ores processing. The chemical composition of soil, initial and washed plant samples was analyzed using inductively coupled plasma mass spectrometry. Factor analysis revealed that anthropogenic and biogenic factors affected the plants' chemical composition. A deficiency of nutrients (Ca, Mg, Mn) in plants growing on tailings was found. The absorption of REE (Ce, La, Sm, Nd) by A. flexuosa roots correlated with the soil content of these elements and was maximal in the hydromorphic, which had a high content of organic matter. The content of these elements in leaves in the same site was minimal; the coefficient of REE bioaccumulation was two orders of magnitude less than in the other two sites. The high efficiency of dust capturing and the low translocation coefficient of trace elements allow us to advise A. flexuosa for remediation of REE-contained tailings and soils. The responses of rare plants to environmental stressors will determine their potential to adapt to a rapidly changing climate. We used a common garden approach to evaluate how six populations of the annual San Diego thornmint (Acanthomintha ilicifolia Lamiaceae; listed as endangered in the state of California and as threatened by the US Fish and Wildlife Service) from across the species range respond in terms of growth (biomass, height, and width) and reproduction (seed production, floral production, and next generation seed viability) to experimental differences in water availability. We found a significant irrigation-by-population interaction on the aboveground growth, wherein the differences in the magnitude and direction of treatment did not correlate directly with climate variables in natural populations. With respect to reproduction, the low-irrigation treatment produced more seeds per plant, more reproductive individuals, and a larger proportion of viable seed in most, but not all, populations. The seed production and the effect of irrigation on seed production correlated positively with rainfall at wild source populations. These results suggest that Acanthomintha ilicifolia responds to water limitation by creating more and higher-quality seed, and that plants locally adapted to a higher annual rainfall show a greater plasticity to differences in water availability than plants adapted to a lower annual rainfall, a finding that can inform the in situ demographic management and ex situ collection strategy for Acanthomintha ilicifolia and other rare California annuals. When working with rare plant species, applying morphometric techniques is one of the main ways to obtain a representative data set on plant individuals and the state of particular populations growing in different ecological conditions and experiencing different degrees of anthropogenic load. Zygophyllum pinnatum, an understudied species growing on the territory of the Republic of Bashkortostan, the Orenburg region, and the Republic of Kazakhstan, is referred as a rare species. The purpose of the work is to study morphometric parameters and reveal the vitality structure of the Z. pinnatum coenopopulations in two regions of the Russian Federation (the Republic of Bashkortostan and Orenburg region) and in the north-west of the Republic of Kazakhstan (the Aktobe Region). Applying standard morphometric techniques, we analyzed the state of 16 coenopopulations of the species. Judging by most morphometric parameters, the leadership belongs to the individuals from the "Troitsk Cretaceous Mountains" coenopopulation located in the Sol-Iletsky District of Orenburg oblast, where, apparently, the most optimal plant conditions are formed (the absence of human impact and low projective cover of the grass stand to have a favorable effect). The minimum values for most parameters are recorded in the coenopopulations located on the territory of the Republic of Kazakhstan, resulting from arid growing conditions. The variability of most characters is within the normal range of the species response (Cv--5.0-44.6%). The discriminant function analysis revealed the morphostructural similarity of individuals from most coenopopulations. Regarding vitality, eight coenopopulations are prosperous, and another eight are depressed. The condition of the species coenopopulations is stable; however, the species is not provided with proper protection measures. Magnolia wufengensis is a newly discovered rare and endangered species endemic to China. The primary objective of this study is to find the most suitable species distribution models (SDMs) by comparing the different SDMs to predict their habitat distribution for protection and introduction in China under climate change. SDMs are important tools for studying species distribution patterns under climate change, and different SDMs have different simulation effects. Thus, to identify the potential habitat for M. wufengensis currently and in the 2050s (2041–2060) and 2070s (2061–2080) under different climate change scenarios (representative concentration pathways RCP2.6, RCP4.5, RCP6.0, and RCP8.5) in China, four SDMs, Maxent, GARP, Bioclim, and Domain, were first used to compare the predicted habitat and explore the dominant environmental factors. The four SDMs predicted that the potential habitats were mainly south of 40° N and east of 97° E in China, with a high distribution potential under current climate conditions. The area under the receiver operating characteristic (ROC) curve (AUC) (0.9479 ± 0.0080) was the highest, and the Kappa value (0.8113 ± 0.0228) of the consistency test and its performance in predicting the potential suitable habitat were the best in the Maxent model. The minimum temperature of the coldest month (−13.36–9.84 °C), mean temperature of the coldest quarter (−6.06–12.66 °C), annual mean temperature (≥4.49 °C), and elevation (0–2803.93 m), were the dominant factors. In the current climate scenario, areas of 46.60 × 104 km2 (4.85%), 122.82 × 104 km2 (12.79%), and 96.36 × 104 km2 (10.03%), which were mainly in central and southeastern China, were predicted to be potential suitable habitats of high, moderate, and low suitability, respectively. The predicted suitable habitats will significantly change by the 2050s (2040–2060) and 2070s (2060–2080), suggesting that M. wufengensis will increase in high-elevation areas and shift northeast with future climate change. The comparison of current and future suitable habitats revealed declines of approximately 4.53%–29.98% in highly suitable habitats and increases of approximately 6.45%–27.09% and 0.77%–21.86% in moderately and lowly suitable habitats, respectively. In summary, these results provide a theoretical basis for the response to climate change, protection, precise introduction, cultivation, and rational site selection of M. wufengensis in the future. Daphne mezereum of the Thymelaeaceae family is a medicinal shrub occurring naturally in Europe and under legal protection in Poland. In the present study, a protocol developed for mass propagation of February daphne from nodal explants is presented. Micropropagation is one of the in vitro techniques that allow the preservation of rare and valuable plants by developing efficient methods for their propagation. In the proliferation stage, explants were cultured in the Woody Plant Medium (WPM) with different cytokinins, and in the rooting stage on the semi-solid WPM medium with perlite, indole-3-butyric acid (IBA) in various concentrations was used. The maximum proliferation rate (five shoots per explant) was observed on the medium containing 4.14 μM of meta-Topolin (mT). When the regenerated shoots were rooted in vitro in the presence of IBA in various concentrations, that of 19.68 μM induced the highest number of roots per shoot (6.63) and the maximal root length (2.15 cm). It is also worth remembering that plants are often colonized by different groups of microorganisms, which also affect the diversity of the ecosystem. Cymbidium goeringii Reichb. fil., locally known as the spring orchid in the Republic of Korea, is one of the most important and popular horticultural species in the family Orchidaceae. C. goeringii cultivars originated from plants with rare phenotypes in wild mountains where pine trees commonly grow. This study aimed to determine the cultivar-specific combined genotypes (CGs) of short sequence repeats (SSRs) by analyzing multiple samples per cultivar of C. goeringii. In this study, we collected more than 4000 samples from 67 cultivars and determined the genotypes of 12 SSRs. Based on the most frequent combined genotypes (CG1s), the average observed allele number and combined matching probability were 11.8 per marker and 3.118 × 10−11, respectively. Frequencies of the CG1 in 50 cultivars (n ≥ 10) ranged from 40.9% to 100.0%, with an average of 70.1%. Assuming that individuals with the CG1 are genuine in the corresponding cultivars, approximately 30% of C. goeringii on the farms and markets may be not genuine. The dendrogram of the phylogenetic tree and principal coordinate analysis largely divided the cultivars into three groups according to their countries of origin; however, the genetic distances were not great among the cultivars. In conclusion, this dataset of C. goeringii cultivar-specific SSR profiles could be used for ecogenetic studies and forensic authentication. This study suggests that genetic authentication should be introduced for the sale of expensive C. goeringii cultivars. The decline in semi-natural grasslands has resulted from reduced traditional use, leading to a marked decrease in their overall area. Despite being located within Japan's core protected area system, the effectiveness of natural parks in conserving grasslands remains unclear. This study aimed to determine whether natural parks successfully prevent the decline in grasslands. By analyzing historical changes in value perceptions toward grasslands in natural parks, natural parks' contribution to grassland conservation and the associated challenges were explored. The study focused on the Sengokuhara grassland in Fuji-Hakone-Izu National Park and incorporated previous studies on other grasslands. It revealed that grasslands have been valued for their scenic views and recreational potential since the establishment of the national park. As grassland scarcity increased nationwide, attention shifted toward rare plants found exclusively on grasslands, along with grasslands' value as semi-natural landscapes. Consequently, natural-park-based grasslands have primarily been conserved based on these two values. However, conservation efforts within natural parks have mainly focused on regulating human activities, with limited measures to sustain nature, such as grasslands, through livelihoods. To enhance grassland conservation, it is essential to recognize their cultural value and develop mechanisms that ensure economic benefits are reinvested in conservation efforts. Iris lacustris, a northern Great Lakes endemic, is a rare species known from 165 occurrences across Lakes Michigan and Huron in the United States and Canada. Due to multiple factors, including habitat loss, lack of seed dispersal, patterns of reproduction, and forest succession, the species is threatened. Early population genetic studies using isozymes and allozymes recovered no to limited genetic variation within the species. To better explore genetic variation across the geographic range of I. lacustris and to identify units for conservation, we used tunable Genotyping-by-Sequencing (tGBS) with 171 individuals across 24 populations from Michigan and Wisconsin, and because the species is polyploid, we filtered the single nucleotide polymorphism (SNP) matrices using polyRAD to recognize diploid and tetraploid loci. Based on multiple population genetic approaches, we resolved three to four population clusters that are geographically structured across the range of the species. The species migrated from west to east across its geographic range, and minimal genetic exchange has occurred among populations. Four units for conservation are recognized, but nine adaptive units were identified, providing evidence for local adaptation across the geographic range of the species. Population genetic analyses with all, diploid, and tetraploid loci recovered similar results, which suggests that methods may be robust to variation in ploidy level. [ABSTRACT FROM AUTHOR]

The superfamily of soluble N-ethylmaleimide-sensitive factor attachment protein receptor (SNARE) proteins mediates membrane fusion during vesicular transport between endosomes and the plasma membrane in eukaryotic cells, playing a vital role in plant development and responses to biotic and abiotic stresses. Peanut (Arachis hypogaea L.) is a major oilseed crop worldwide that produces pods below ground, which is rare in flowering plants. To date, however, there has been no systematic study of SNARE family proteins in peanut. In this study, we identified 129 putative SNARE genes from cultivated peanut (A. hypogaea) and 127 from wild peanut (63 from Arachis duranensis, 64 from Arachis ipaensis). We sorted the encoded proteins into five subgroups (Qa-, Qb-, Qc-, Qb+c- and R-SNARE) based on their phylogenetic relationships with Arabidopsis SNAREs. The genes were unevenly distributed on all 20 chromosomes, exhibiting a high rate of homolog retention from their two ancestors. We identified cis-acting elements associated with development, biotic and abiotic stresses in the promoters of peanut SNARE genes. Transcriptomic data showed that expression of SNARE genes is tissue-specific and stress inducible. We hypothesize that AhVTI13b plays an important role in the storage of lipid proteins, while AhSYP122a, AhSNAP33a and AhVAMP721a might play an important role in development and stress responses. Furthermore, we showed that three AhSNARE genes (AhSYP122a, AhSNAP33a and AhVAMP721) enhance cold and NaCl tolerance in yeast (Saccharomyces cerevisiae), especially AhSNAP33a. This systematic study provides valuable information about the functional characteristics of AhSNARE genes in the development and regulation of abiotic stress responses in peanut. Many rare plant species lack up-to-date research about their reproductive ecology, which challenges effective in situ and ex situ conservation, particularly in the face of ongoing environmental and anthropogenic changes. For protected species, outdated and incomplete information also creates barriers to successful recovery planning and delisting. In this study, we gathered a range of reproductive metrics for the federally threatened and state endangered Florida endemic mint, Macbridea alba Chapman (Lamiaceae). We collected data at seven populations within Apalachicola National Forest (Florida, USA) and conducted germination trials to estimate reproductive potential. Additionally, we observed a previously undocumented lepidopteran seed predator for the species and confirmed the occurrence of vivipary. The seed set was low with less than 20% of flowers per inflorescence producing seed across populations; however, germination was high with more than 60% of seeds germinating in five of seven populations. When comparing our results to previous research conducted more than 20 years ago, the results were similar overall (i.e., germination, vivipary); however, new information emerged (i.e., herbivore pressure). As M. alba undergoes reassessment as a potential candidate for delisting from the Endangered Species Act (ESA) list, this information is critical for assessing recovery goals and decisions regarding the species' protected status. Titan arum (Amorphophallus titanum) is a rare and endangered plant in the world. It has a huge flower and releases a repulsive odor like a corpse. On the evening of 23 July 2022 (Beijing time), a titan arum in the north garden of the China National Botanical Garden in Beijing bloomed. In order to determine the components and contents of volatile compounds released by the titan arum during its flowering, the dynamic headspace adsorption method was utilized to collect the odor of the titan arum flower on the evening of 23 July (S1), the morning of 24 July (S2), the afternoon of 24 July (S3) and the evening of 24 July (S4). The volatile compounds were analyzed by automatic thermal desorption–gas chromatography/mass spectrometry. Sixty-three volatile compounds were detected in the titan arum flower in four periods. The comparison of the total volatile compounds released in four periods was S2 > S3 > S1 > S4. The highest content of volatile compounds in the S1 period were sulfur compounds (dimethyl disulfide and dimethyl trisulfide), and the sulfur compounds were released in large amounts only in the S1 period. Dimethyl disulfide was the volatile substance with the highest content in the S1 period (20.00%). The total volatile compounds content of titan arum flower in the S2 period was the highest among the four periods. From the S2 period, the relative content of sulfur compounds decreased significantly until the S4 period. Compared with the S1 period, 1-butanol and butyl acrylate increased significantly and 1-butanol became the highest relative component of volatile compounds in the S2 period. After the S3 period, the total amount of volatile compounds began to decline and reached the lowest level in the S4 period. It is worth noting that the contents of two terpenes, α-pinene and γ-terpinene, rose from the S1 period until their height in the S3 period. From the S4 period, the contents of most volatile compounds decreased significantly. This study revealed the varieties and contents of volatile compounds in the titan arum flower at different flowering periods. The changing trend and physiological significance of dimethyl oligosulfide from the evening of flowering (S1) to the second day (S2–S4) were emphatically discussed, and this research also provides a reference for the study of the release of volatile compounds and the molecular biology of the flower fragrance of titan arum. In recent times, less-known fruit species have increasingly attracted worldwide attention and their health benefits are at the forefront. The fruits of plants from the genus Prunus are good sources of nutrients due to their economic, agronomic, and healthy values. However, Prunus lusitanica L., commonly known as Portuguese laurel cherry is considered an endangered species. Thus, the present work aimed to monitor the nutritional components of P. lusitanica fruits grown in three locations in northern Portugal for four consecutive years (2016–2019), using AOAC (Association of Official Analytical Chemists), spectrophotometric, and chromatographic analysis. The results evidenced the abundance of phytonutrients in P. lusitanica, such as proteins, fat, carbohydrates, soluble sugars, dietary fibre, amino acids, and minerals. It was also highlighted that the variation of nutritional components was relatively linked to the year factor, being especially relevant in the frame of the current changing climate, among others. These findings suggest that P. lusitanica L. deserves to be conserved and planted because of its food and nutraceutical applications. However, more detailed information on this rare plant species, such as phytophysiology, phytochemistry, bioactivity, pharmacology, etc., is certainly required for the design and development of appropriate uses and valorization alternatives for this species. Many wild ornamental plant species have been introduced to improve the landscape of cities; however, until now, no study has been performed to explore the composition and function of foliar endophytes associated with cultivated rare plants in cities after their introduction. In this study, we collected the leaves of the healthy ornamental plant Lirianthe delavayi from wild and artificially cultivated habitats in Yunnan and compared their diversity, species composition, and functional predictions of their foliar endophytic fungal community based on high-throughput sequencing technology. In total, 3125 ASVs of fungi were obtained. The alpha diversity indices of wild L. delavayi populations are similar to those of cultivated samples; however, the species compositions of endophytic fungal ASVs were significantly varied in the two habitats. The dominant phylum is Ascomycota, accounting for more than 90% of foliar endophytes in both populations; relatively, artificial cultivation trends to increase the frequency of common phytopathogens of L. delavayi, such as Alternaria, Erysiphe. The relative abundance of 55 functional predictions is different between wild and cultivated L. delavayi leaves (p < 0.05); in particular, chromosome, purine metabolism, and peptidases are significantly increased in wild samples, while flagellar assembly, bacterial chemotaxis, and fatty acid metabolism are significantly enhanced in cultivated samples. Our results indicated that artificial cultivation can greatly change the foliar endophytic fungal community of L. delavayi, which is valuable for understanding the influence of the domestication process on the foliar fungal community associated with rare ornamental plants in urban environments. Giant genomes are rare across the plant kingdom and their study has focused almost exclusively on angiosperms and gymnosperms. The scarce genetic data that are available for ferns, however, indicate differences in their genome organization and a lower dynamism compared to other plant groups. Tmesipteris is a small genus of mainly epiphytic ferns that occur in Oceania and several Pacific Islands. So far, only two species with giant genomes have been reported in the genus, T. tannensis (1C = 73.19 Gbp) and T. obliqua (1C = 147.29 Gbp). Low-coverage genome skimming sequence data were generated in these two species and analyzed using the RepeatExplorer2 pipeline to identify and quantify the repetitive DNA fraction of these genomes. We found that both species share a similar genomic composition, with high repeat diversity compared to taxa with small (1C < 10 Gbp) genomes. We also found that, in general, characterized repetitive elements have relatively high heterogeneity scores, indicating ancient diverging evolutionary trajectories. Our results suggest that a whole genome multiplication event, accumulation of repetitive elements, and recent activation of those repeats have all played a role in shaping these genomes. It will be informative to compare these data in the future with data from the giant genome of the angiosperm Paris japonica, to determine if the structures observed here are an emergent property of massive genomic inflation or derived from lineage specific processes. The Palestinian environment suffered from abuse and neglect for decades. As a nascent state, Palestine aspired to protect and restore its environment, in line with international standards and conventions. Yet, many challenges remain, including a lack of data, the science–policy–practice gaps, and the lack of sovereignty. The management of protected areas (PAs) is particularly challenging under these circumstance. Two key protected areas in the northern West Bank (Wadi Qana and Wadi Al-Zarqa Al-Ulwi) were surveyed, with a focus on plant communities and dealing with threats. The two areas had similar Mediterranean flora with differences, including in rare plants. Both areas have important but different wetlands in their buffer zones. The data show that the protection of the two areas is important because of the differences noted, and both areas are valued as IUCN category IV (habitat/species management area) protection. Wetlands have an important ecological function and economic value. However, with economic development and urban expansion, wetland plants have suffered serious damage. Rare and endangered plants are "thermometers" that reflect the health processes of their ecosystems. To better protect the wetlands in Sanya, China, we systematically investigated and analyzed the species, quantities, distributions, and community characteristics of the rare and endangered plants in the Sanya River basin using the sample and sample strip methods. We established a total of 152 quadrats, of which 46 contained rare and endangered plants. We identified 27 rare and endangered plants that mainly appeared in the tree and shrub layers. The dominant families and genera of the community were evident. However, the proportion of families and genera with fewer or single species was high, indicating that the species composition of the community is complex, and the plant species diversity is rich. The dominant species in each layer of the community were evident, and the rare and endangered plants are occasional species of the community. The community similarity in the urban areas was high, indicating that the rare and endangered plants in these areas require highly homogenous habitats. The community similarity in the suburbs was low, indicating that the rare and endangered plants in these areas are highly adaptable to different habitats. Threat factors and vegetation coverage degree had a significant impact on the number of species and population size of rare and endangered plants. Finally, according to our study and IUCN classification criteria for the endangered levels, Sonneratia × gulngai meets the CR (Critical Endangered) assessment criteria, thus we recommend upgrading it to the endangered level from VU (Vulnerable). The conservation actions of endangered plant species require a clear knowledge of their habitats. Urtica rupestris Guss. (Urticaceae) is a rare endemic plant species occurring on shady cliffs in the southern-eastern part of Sicily. In the last century, the extreme anthropogenic alterations of Hyblaean plateau have caused the continuous and unrestrained fragmentation of natural habitats and consequently the reduction and disappearance of some plant species. A total of 52 vegetation plots, of which 34 are unpublished, were analyzed in order to characterize the floristic composition of the U. rupestris community. All the relevés were classified using classification and ordination methods. The species is mainly linked to shady and wet rock habitats, and only secondarily colonizes the undergrowth shrubs. According to IUCN criteria, we propose a new risk status for this species and the establishment of a new habitat (92/43CEE) for correct long-term conservation. Finally, a new association, Urtico rupestris-Adiantetum capilli-veneris, which falls within the Polysticho setiferi-Phyllitidion scolopendrii alliance (Adiantetea capilli-veneris class), was described. This study was conducted to examine the effect of plant growth regulators on the rooting and growth of Rhododendron micranthum Turcz., a rare plant in South Korea. Indole-3-butyric acid (IBA) 150, 250, 350 mg·L−1 and α-naphthaleneacetic acid (NAA) 50, 150, 250 mg·L−1 treatment groups were established and cutting was performed in a commercial growth medium in March 2021 with 3 replications of 10 plants per group. After 70 days, the longest initial root growth was observed under the NAA 150 and 250 mg·L−1 treatments. Moreover, the number of roots and percentage of rooting were slightly higher in NAA 150 and 250 mg·L−1, indicating that these treatments were more effective in promoting plant growth. The above-ground part of plants treated with NAA 150 and 250 mg·L−1 had a taller plant height, longer and wider leaves, and a greater number of leaves compared to other groups. Accordingly, the survival rate of plants treated with NAA 150 and 250 mg·L−1 was higher than 80%, suggesting well-established roots and excellent above-ground growth. Micropropagation is an effective way to preserve the gene pool of threatened plants. This study is devoted to the mathematical modeling of nutrient media and the study of the effect of mT (meta-topoline) on the multiplication of shoots of Hedysarum grandiflorum, Hyssopus cretaceus, and Matthiola fragrans in vitro in comparison with benzylaminopurine (BAP) and kinetin (KT). Initiation was performed on an MS medium with 0.5 mg/L BAP. For shoots multiplication, MS, B5, and WPM media were used with the addition of mT, BAP, KT. For H. grandiflorum, the multiplication coefficient of shoots was highest on medium B5 with the addition of mT at a concentration of 1 mg/L—2.90 shoots per plant, for H. cretaceus—B5 + 0.5 mg/L mT, and for M. fragrans—B5 + 1 mg/L KT. A positive effect of mT on H. grandiflorum and M. fragrans in vitro was found. The efficiency of using KT for H. cretaceus shoot multiplication is shown. The effectiveness of the B5 nutrient medium for H. grandiflorum and M. fragrans was determined. The positive effect of WPM for H. cretaceus micropropagation has been demonstrated. Global warming is one of the most serious problems leading to changes in the distribution areas of species and biodiversity. Rhodiola quadrifida is a rare plant with adaptogenic properties and grows in the highlands in a narrow temperature range of 2–15 °C. The aim of our work was to study the growth and content of the main metabolites in two in vitro cultures of Rhodiola quadrifida at temperatures of 5, 15 and 25 °C. Hairy roots and calli were cultivated on agar medium for 28 days. The maximum values of the growth index were observed at 25 °C (2.32 and 2.12 for calli and hairy roots, respectively). HPLC-MS showed the absence of tyrosol and rosarin in both cultures, and rosin in the root culture. The content of salidroside changed slightly in calli and roots. Cultivation at 5 °C significantly stimulated the formation of rosin in calli. Only a residual amount of rosavin was noted in the roots, regardless of temperature. The content of rosin was higher in calli at 15 °C with a maximum content at the end of the cultivation cycle 25 µg/g DW. Thus, Rhodiola quadrifida will be able to grow with an increase in temperature by 10 °C but this will be accompanied by a significant reduction in its medicinal value [ABSTRACT FROM AUTHOR]

A lobed leaf is a common trait in plants, but it is very rare in Lauraceae plants, including species of Phoebe. In the study of germplasm resources of Phoebe neurantha, we found lobed leaf variant seedlings, and the variation could be inherited stably. Studying the lobed leaf mechanism of P. neurantha var. lobophylla can offer insight into the leaf development mechanism of woody plants. RNA-seq and small RNA-seq analysis results showed that a total of 8091 differentially expressed genes (DEGs) and 16 differentially expressed miRNAs were identified in P. neurantha var. lobophylla. Considering previous research results, a leaf margin morphological development related miRNA, pne-miRNA319a, was primary identified as a candidate miRNA. Target gene prediction showed that a total of 2070 genes were predicted to be the target genes of differentially expressed miRNAs. GO enrichment analysis of differentially expressed target genes suggested that PnTCP2 is related to lobed leaf formation. The TRV-VIGS gene silencing of PnTCP2 led to lobed leaves in P. neurantha seedlings. The downregulation of PnTCP2 led to lobed leaves. The yeast two-hybrid test and bimolecular fluorescence complementation test confirmed that the PnTCP2 protein interacted with the PnLBD41 protein. Based on the expression analysis of gene-silenced leaves and RNA-seq and small RNA-seq analysis results, pne- miRNA319a and PnLBD41 might also play important roles in this process. A rare plant species of the Orchidaceae family, Dendrobium officinale is considered among the top ten Chinese medicinal herbs for its polysaccharide. Since 2021, when the dieback disease of D. officinale was first reported in Yueqing City, Zhejiang Province, China, Fusarium isolates (number = 152) were obtained from 70 plants in commercial greenhouses. The disease incidence ranged from 40% to 60% in the surveyed areas. Multilocus sequence analysis (MLSA) coupled with morphological characterization revealed that the collected isolates belonged to five species (sp.), viz., Fusarium concentricum, F. fujikuroi, F. nirenbergiae, F. curvatum, and F. stilboides, with isolation frequencies of 34.6%, 22.3%, 18.4%, 13.8%, and 10.5%, respectively. Notably, at least two Fusarium species were simultaneously isolated and identified from the infected plants. Finally, the pathogenicity test results demonstrated that such species were responsible for the dieback disease of D. officinale. However, F. concentricum and F. fujikuroi were more invasive compared to the other species in this study. To the best of the authors' knowledge, this study was the first report of F. concentricum, F. curvatum, F. fujikuroi, F. nirenbergiae, and F. stilboides causing the dieback disease of D. officinale in China and worldwide. This work provides valuable data about the diversity and pathogenicity of Fusarium populations, which will help in formulating effective strategies and policies for better control of the dieback disease. Dunelands are one of the most dynamic environments on Earth, which greatly hinders their conservation and management. In the dune slacks along the Manawatū coast, New Zealand, lies a zone of small, temporary, freshwater wetlands that host early seral communities of rare turf plants. Analysis of historical aerial photos allowed determination of coastline movement, distance of the wetlands from the coast, and wetland movements through time. Study sites were around the coastal settlements of Tangimoana and Foxton Beach, both having major rivers debouching nearby, and Himatangi, amongst stabilising exotic pine plantations. The coastline is prograding (with seaward movement) generally by 0.7–1 m yr−1, but is more variable closer to river mouths, with episodes of movement of up to 15 m yr−1. Wetlands occur 200–400 m behind the strandline, are closer to the coast at Foxton Beach, and furthest away at Himatangi. Wetlands wobble in their position at 5.2 m yr−1 but migrate inland at an average of 2.7 m yr−1. Wetland size appears unrelated to rainfall, but may be related positively to coastal progradation rate, to which wetland movement is negatively related. Near the major rivers, dunes are less stable, and wetlands can be impacted both positively and negatively. Wetland existence and movement is balanced between stability and dynamism on the coast, and management will need to be proactive to maintain environments for early successional turfs. Obtaining accurate plant population estimates has been integral in listing, recovery, and delisting species under the U.S. Endangered Species Act of 1973 and for monitoring vegetation in response to livestock grazing. Obtaining accurate population estimates remains a daunting and labor-intensive task. Small unmanned aircraft systems (sUASs or drones) may provide an effective alternative to ground surveys for rare and endangered plants. The objective of our study was to evaluate the efficacy of sUASs (DJI Phantom 4 Pro) for surveying the Wright fishhook cactus (Sclerocactus wrightiae), a small (1–8 cm diameter) endangered species endemic to grazinglands in the southwest desert of Utah, USA. We assessed sUAS-based remotely sensed imagery to detect and count individual cacti compared to ground surveys and estimated optimal altitudes (10 m, 15 m, or 20 m) for collecting imagery. Our results demonstrated that low altitude flights provided the best detection rates (p < 0.001) and counts (p < 0.001) compared to 15 m and 20 m. We suggest that sUASs can effectively locate cactus within grazingland areas, but should be coupled with ground surveys for higher accuracy and reliability. We also acknowledge that these technologies may have limitations in effectively detecting small, low-growing individual plants such as the small and obscure fishhook cactus species. The leafless orchids are rare epiphytic plants with extremely reduced leaves, and their aerial roots adopted for photosynthesis. The beneficial plant–microbial interactions contribute significantly to host nutrition, fitness, and growth. However, there are no data available on the bacterial associations, inhabiting leafless orchids. Here, we describe the diversity of cyanobacteria, which colonize the roots of greenhouse Microcoelia moreauae and Chiloschista parishii. The biodiversity and structure of the cyanobacterial community were analyzed using a complex approach, comprising traditional cultivable techniques, denaturing gradient gel electrophoresis (DGGE), and phylogenetic analysis, as well as the light and scanning electron microscopy (SEM). A wide diversity of associated bacteria colonize the root surface, forming massive biofilms on the aerial roots. The dominant populations of filamentous nitrogen-fixing cyanobacteria belonged to the orders Oscillatoriales, Synechococcales, and Nostocales. The composition of the cyanobacterial community varied, depending on the nitrogen supply. Two major groups prevailed under nitrogen-limiting conditions, belonging to Leptolyngbya sp. and Komarekiella sp. The latter was characterized by DGGE profiling and sequencing, as well as by its distinctive features of morphological plasticity. The leading role of these phototrophophic and diazotrophic cyanobacteria is discussed in terms of the epiphytic lifestyle of the leafless orchids. [ABSTRACT FROM AUTHOR]

The use of plants as traditional medicines is common and has prevailed in many different cultures over time. Polymethoxyflavones (PMFs) are natural polyphenols from the group of flavonoids. Zapotin, a member of the PMFs, is found mainly in citrus plants and is almost exclusively limited to their peels. The chemical structure of zapotin has been questioned from the very beginning, since the structure of flavonoids with a single oxygen atom in the C2′ position is extremely rare in the plant kingdom. To clarify this, the structural determination and bio-inspired synthesis of zapotin are discussed in detail in this review. Due to the broad biological potential of PMFs, the complication in the isolation process and characterization of PMFs, as well as their purification, have been estimated by adapting various chromatographic methods. According to available data from the literature, zapotin may be a promising curative agent with extensive biological activities, especially as a chemopreventive factor. Apart from that, zapotin acts as an antidepressant-like, anticancer, antifungal, and antioxidant agent. Astragalus exscapus L. subsp. transsilvanicus (Schur) Nyár. (Fabaceae) is a rare plant endemic to the Transylvanian Plateau, represented by 24 identified populations. Limited information on the genetic variation and population structure is available, which obstructs efficient measures for conservation strategy. The present study aimed to analyze the genetic diversity and population structure of eight populations of A. exscapus subsp. transsilvanicus revealed by sequence-related amplified polymorphism (SRAP) markers. A total of 164 bands were amplified, 96.7% of which (159) were polymorphic. Nei's gene diversity index (He) was estimated to be 0.228 at the population level and 0.272 at the subspecies level. The genetic differentiation among populations (Gst) was 0.165, which indicated a low proportion of total genetic diversity. The analysis of molecular variance (AMOVA) indicated that 17% of the total variation of A. exscapus subsp. transsilvanicus is found among the populations, while 83% was found within the populations. A UPGMA dendrogram, principal coordinate analysis, and the STRUCTURE software grouped the populations into two clusters uncorrelated with the provenience of the 125 individuals, which might be attributed to fragmentation processes, insect pollination, population size, and specific environmental conditions of the habitats. Stinging trichomes are rare in plants, occurring only in angiosperms, where they are reported for a few genera belonging to six families. Although there is no report of stinging trichomes in Apocynaceae, previous fieldwork collections of Fischeria and Matelea caused us a mild allergic reaction on the skin when we contacted the dense indumentum of the plants. This fact associated with the well-known presence of glandular trichomes with acute apex in both genera raised suspicions that stinging trichomes could be present in the family. Hence, this study aimed to investigate the likely occurrence of stinging trichomes in Fischeria and Matelea. We analyzed vegetative shoots and leaves of Fischeria stellata and Matelea denticulata through the usual procedures of light and scanning electron microscopy. We also performed several histochemical tests to investigate the chemical composition of trichome secretion. We detected that glandular trichomes occur throughout the surface of the leaf and stem. They are multicellular, uniseriate with an apical secretory cell, which has a dilated base and a needle-shaped apex. The secretion is compressed into the acuminate portion of the apical cell by a large vacuole, and crystals are deposited in the cell wall in a subapical position, providing a preferential site of rupture. The secretion, composed of amino acids and/or proteins, is released under mechanical action, causing skin irritation. Based on our detailed morphological and anatomical analyses, and in the functional aspects observed, we concluded that the glandular trichomes in Fischeria and Matelea can indeed be classified as stinging. Thus, Apocynaceae is the seventh family for which this type of trichome has been reported. We also compiled information on stinging trichomes in all families of angiosperms. Their phylogenetic distribution indicates that they have evolved at least 12 times during angiosperm evolution and may represent an evolutionary convergence of plant defense against herbivory. Daegwallyeong is a mountain pass at an altitude of 832 m, which has been designated a conservation area because of its essential role in Korea's forest ecosystem. Simultaneously, this area is considered a suitable place for forest grazing due to the cool temperature during the summer. Some areas have been converted to grassland for livestock feeding, and the scale has continued increasing. Although livestock in a forest area is more ecofriendly than industrialized facilities, it could impact the native ecosystem, especially in terms of the flora and vegetation. We investigated the changes in flora and vegetation of Daegwallyeong before and after the grassland formation. The total number of vascular plant species changed throughout the survey period. It was decreased by thinning and forest floor removal in 2015. However, it bounced back to the original number in 2016, even after grazing. However, there was a dramatic decrease after the second forest floor removal and 3 months of grazing in 2017. The number of flora slightly increased after the fallow of grazing in 2019, but it did not fully recover. Although the number of flora seemed back to normal, the composition of the flora in 2019 was significantly changed from the forest without disturbance in 2014. First, there was the invasion of naturalized plants such as Taraxacum officinale and Barbarea vularis, as well as ecosystem-disturbing flora such as Carex callitrichos var. nana and Rumex acetosa. Second, the coverage of those species expanded after thinning and grazing. Most importantly, we lost five valuable rare species, Anemone koraiensis, Viola diamantiaca, Chionanthus retusus, Scopolia japonica, and Streptopus ovalis, from the area. Additionally, the ground condition of the area was severely damaged, and plants no longer grow in some areas. The survey and analysis of plants in this study showed the adverse effects of forest grazing practices on rare plants in Daegwallyeong. Desert vegetation is an important part of arid and semi-arid areas, which plays an important role in preventing wind and fixing sand, conserving water and soil, maintaining the balanced ecosystem. Therefore, mapping the vegetation accurately is necessary to conserve rare desert plants in the fragile ecosystems that are easily damaged and slow to recover. In mapping desert vegetation, there are some weaknesses by using traditional digital classification algorithms from high resolution data. The traditional approach is to use spectral features alone, without spatial information. With the rapid development of drones, cost-effective visible light data is easily available, and the data would be non-spectral but with spatial information. In this study, a method of mapping the desert rare vegetation was developed based on the pixel classifiers and use of Random Forest (RF) algorithm with the feature of VDVI and texture. The results indicated the accuracy of mapping the desert rare vegetation were different with different methods and the accuracy of the method proposed was higher than the traditional method. The most commonly used decision rule in the traditional method, named Maximum Likelihood classifier, produced overall accuracy (76.69%). The inclusion of texture and VDVI features with RGB (Red Green Blue) data could increase the separability, thus improved the precision. The overall accuracy could be up to 84.19%, and the Kappa index with 79.96%. From the perspective of features, VDVI is less important than texture features. The texture features appeared more important than spectral features in desert vegetation mapping. The RF method with the RGB+VDVI+TEXTURE would be better method for desert vegetation mapping compared with the common method. This study is the first attempt of classifying the desert vegetation based on the RGB data, which will help to inform management and conservation of Ulan Buh desert vegetation. Rare carnivorous plants representing the genus Sarracenia are perceived as very interesting to scientists involved in various fields of botany, ethnobotany, entomology, phytochemistry and others. Such high interest is caused mainly by the unique capacity of Sarracenia spp. to attract insects. Therefore, an attempt to develop a protocol for micropropagation of the Sarracenia alata (Alph.Wood) Alph.Wood, commonly named yellow trumpets, and to identify the specific chemical composition of volatile compounds of this plant in vitro and ex vivo was undertaken. Thus, the chemical volatile compounds excreted by the studied plant to attract insects were recognized with the application of the headspace solid-phase microextraction (HS-SPME) coupled with the GC-MS technique. As the major volatile compounds (Z)-3-hexen-1-ol (16.48% ± 0.31), (E)-3-hexen-1-ol acetate (19.99% ± 0.01) and β-caryophyllene (11.30% ± 0.27) were identified. Further, both the chemical assumed to be responsible for attracting insects, i.e., pyridine (3.10% ± 0.07), and whole plants were used in in vivo bioassays with two insect species, namely Drosophila hydei and Acyrthosiphon pisum. The obtained results bring a new perspective on the possibilities of cultivating rare carnivorous plants in vitro since they are regarded as a valuable source of bioactive volatile compounds, as including ones with repellent or attractant activity. Simple Summary: Lauraceae, as an angiosperm group with high ecological and economic value, has been widely studied. With the development of science and technology, the research of Lauraceae has changed from morphology to molecular systematics. Our paper reviewed the molecular phylogeny of Lauraceae in recent years. From the aspects of gene fragments, chloroplast genome and DNA barcodes, we mainly discussed the establishment of Cinnamomeae in the 'Core Lauraceae' and the tribal controversial genera (Actinodaphne, Sassafras, Caryodaphnopsis, Neocinnamomum and Cassytha). We think that the whole genome and inflorescence characteristics are the breakthrough to solve the tribal problem of Lauraceae. Using reliable molecular and morphological evidence to reconstruct the phylogenetic relationship of Lauraceae will provide an important theoretical basis for the rational utilization of Lauraceae resources, the development of potential resources and the protection of rare plants. Lauraceae is a large family of woody plants with high ecological and economic value. The tribal and generic division and phylogenetic relationship of Lauraceae have long been controversial. Based on morphological and molecular evidence, phylogenetic relationships within the Cinnamomeae, Laureae and Perseeae tribes, also called 'the Core Lauraceae', have arisen particular attention. In this review, we comprehensively collated the literatures on the phylogeny of Lauraceae published in recent years and summarized progress made in molecular systematic researches employing gene fragments, chloroplast genomes and DNA barcodings analyses. We clarified the phylogenetic relationships and main controversies of 'the Core Lauraceae', the systemic position of fuzzy genera (Neocinnamomum, Caryodaphnopsis and Cassytha) and the development of chloroplast genome and DNA barcodes. We further suggested and proposed the whole genome analysis and different inflorescence types would be possible to provide more information for further research on phylogenetic relationships and taxonomy of Lauraceae. Rindera graeca is a rare endemic plant where in vitro culture has been used in order to investigate bioactive metabolites. Phytochemical study of the in vitro shoots and hairy roots led to the isolation of seven phenolic derivatives and the unusual furano-naphthoquinone rinderol. R. graeca was also analyzed for its pyrrolizidine alkaloids content by LC-MS, and it was found to contain echinatine together with echinatine and rinderine N-oxides. Rinderol, isolated only from in vitro hairy root culture for the first time in the genus, revealed promising bioactivities. It was evaluated in vitro against a panel of microorganisms, showing very strong activity specifically against Gram-positive bacteria (MIC values 0.98 × 10−2–1.18 µg/mL) as well as very interesting antiproliferative effect against the human non-small-cell bronchopulmonary carcinoma cell line NSCLC-N6-L16 and the epidermoid lung cancer cell line A549. These findings were compared with the chemical profile of the plant from nature, while this study is the first to report on the effects of R. graeca extracts obtained from in vitro culture, providing a valuable contribution to the scientific community towards this sustainable method of production of potential bioactive molecules. Salvia ceratophylloides (Ard.) is an endemic and rare plant species recently rediscovered as very few individuals at two different Southern Italy sites. The study of within-plant variation is fundamental to understand the plant adaptation to the local conditions, especially in rare species, and consequently to preserve plant biodiversity. Here, we reported the variation of the morpho-ecophysiological and metabolic traits between the sessile and petiolate leaf of S. ceratophylloides plants at two different sites for understanding the adaptation strategies for surviving in these habitats. The S. ceratophylloides individuals exhibited different net photosynthetic rate, maximum quantum yield, light intensity for the saturation of the photosynthetic machinery, stomatal conductance, transpiration rate, leaf area, fractal dimension, and some volatile organic compounds (VOCs) between the different leaf types. This within-plant morpho-physiological and metabolic variation was dependent on the site. These results provide empirical evidence of sharply within-plant variation of the morpho-physiological traits and VOCs profiles in S. ceratophylloides, explaining the adaptation to the local conditions. The effects of herbivorous insects on a plant population are not always well tolerated. This is especially true if the herbivorous actions are directed toward rare plant species. Salvia ceratophylloides Ard. is a rare endemism of southern Italy. Observations of the plants in situ revealed that many of them were under severe stress and did not produce seeds. Therefore, to find out which factors affect the reproductive activity as a whole, an observational study was carried out. We found bottom-up and top-down effects on plant health and reproduction associated with herbivorous action. Squamapion elongatum (Coleoptera, Curculionoidea, Apionidae), in all monitored sites, infested plants non-uniformly but was able to threaten the health condition, flowering, and seed production of sage by digging tunnels into the sage branches (bottom-up action), and then secondarily by seed feeder Systole salvia Zerova (Hymenoptera, Eurytomidae) predating sage seeds (top-down action). Mainly, chalcid parasitoid wasps such as Trichomalus spp. (Hymenoptera, Pteromalidae), as well as Eupelmus vesicularis and E. muellneri (Hymenoptera, Eupelmidae), limited the herbivorous S. elongatum population and the seed herbivore S. salviae emerged with its parasitoid Ormyrus diffinis (Hymenoptera, Ormyridae). Overall, this study showed how ecological interactions among herbivores, their host, and their natural enemies act on this sage species in all sites investigated. Among the herbivores, mainly S. elongatum affected this rare sage species, which should be taken into consideration, especially in the formulation of biological control solutions and for improving operating practice aimed at reproducing the species. This study provides the molecular characterization of the herbivorous species involved, in order to support future projects to evaluate the intra- and interspecific genetic variability of insects, their evolutionary relationships, and phylogeny studies. There is growing attention given to gene flow between crops and the wild relatives as global landscapes have been rapidly converted into agricultural farm fields over the past century. Crop-to-wild introgression may advance the extinction risks of rare plants through demographic swamping and/or genetic swamping. Malus sieversii, the progenitor of the apple, is exclusively distributed along the Tien Shan mountains. Habitat fragmentation and hybridization between M. sieversii and the cultivated apples have been proposed to be the causal mechanism of the accelerated extinction risk. We examined the genetic diversity pattern of eleven wild and domesticated apple populations and assessed the gene flow between M. sieversii and the cultivated apples in Kazakhstan using thirteen nuclear microsatellite loci. On average, apple populations harbored fairly high within-population diversity, whereas population divergences were very low suggesting likely influence of human-mediated dispersal. Assignment results showed a split pattern between the cultivated and wild apples and frequent admixture among the apple populations. Coupled with the inflated contemporary migration rates, the admixture pattern might be the signature of increased human intervention within the recent past. Our study highlighted the prevalent crop to wild gene flow of apples occurring in Kazakhstan, proposing an accelerated risk of genetic swamping. [ABSTRACT FROM AUTHOR]

The current investigation aimed to present an overview of the conservation of biological diversity of rare and endangered plant species. Methods of biodiversity conservation as well as several overview recommendations for the preservation of various rare species have been considered. An overview of the taxa included in the red book has been presented on the example of the Russian Federation. Global and local codes and classifiers of plant rarity were also presented. Xerolekia speciosissima (L.) Anderb., a rare plant from the north of Italy, is a member of the Inuleae-Inulinae subtribe of the Asteraceae. Despite its close taxonomic relationship with many species possessing medicinal properties, the chemical composition of the plant has remained unknown until now. A hydroalcoholic extract from the aerial parts of X. speciosissima was analyzed by HPLC-DAD-MSn, revealing the presence of caffeic acid derivatives and flavonoids. In all, 19 compounds, including commonly found chlorogenic acids and less frequently occurring butyryl and methylbutyryl conjugates of dicaffeoylquinic and tricaffeoylhexaric acids, plus two flavonoids, were tentatively identified. Chromatographic separation of a hydroalcoholic extract from the capitula of the plant led to the isolation of (+)-dehydrodiconiferyl alcohol 4-O-β-glucopyranoside, quercimeritrin, astragalin, isoquercitrin, 6-hydroxykaempferol-7-O-β-glucoside, quercetagitrin, methyl caffeate, caffeic acid, protocatechuic acid, chlorogenic acid and 1,5-dicaffeoylquinic acid. Composition of a nonpolar extract from the aerial parts of the plant was analyzed by chromatographic methods supported with 1H-NMR spectroscopy. The analysis revealed the presence of loliolide, reynosin, samtamarine, 2,3-dihydroaromaticin, 2-deoxy-4-epi-pulchellin and thymol derivatives as terpenoid constituents of the plant. One of the latter compounds—7,10-diisobutyryloxy-8,9-epoxythymyl isobutyrate—at concentrations 0.5, 1.0 and 2.5 μM, significantly reduced IL-8, IL-1β and CCL2 excretion by LPS-stimulated human neutrophils. Efforts to conserve rare plant species can be limited by a lack of time and funding for monitoring. Understanding species occurrence and distribution patterns within existing protected habitat and throughout the entire species range can help stewards prioritize rare plant monitoring. We created a database of rare plant occurrences from public, private, and research sources to analyze the distribution of rare plant species throughout the existing protected area within the Nature Reserve of Orange County in California, USA. We analyzed species occurrence relative to the urban edge, roads, trails, and mean high tide line. We also determined the vegetation community with the highest number of rare plant species to help prioritize habitats for conservation and restoration. We found that some parts of protected areas have more rare plant species and we also found sampling biases on the location of occurrence data. We found that rare species occur close to roads and trails and the mean high tide line. Rare species were in all vegetation communities within the reserve, including degraded areas. Using patterns of distribution and considering the immediate threats to a rare species population can help land managers and stewards prioritize monitoring toward the most threatened species. Translocation is increasingly being used to supplement existing occurrences and establish new occurrences of rare plant species, but translocation success is dependent on understanding responses to habitat conditions and management. Platanthera integrilabia (white fringeless orchid) is a rare terrestrial orchid species presently found in mostly small occurrences that comprise a fraction of its historical distribution and abundance in the southeastern United States. We investigated the influence of shade and white-tailed deer herbivory, as cited concerns for this species, on the early success of its translocation from tubers as determined through measures of emergence, survival, growth, and reproduction of two cohorts. Our findings suggest that translocation from tubers could be a viable option to assist the conservation of P. integrilabia relative to its propagation from seed, but that low early emergence, survival, and flowering rates should be considered in translocation plans. Our results also indicate that translocation and ongoing habitat management should consider the potential for light availability to differentially impact distinct plant life stages and influence deer herbivory. We recommend that additional translocation studies designed to investigate the influence of site conditions on outcomes could improve the success of such efforts as well as inform the management of extant occurrences.
