Simple Summary: This study addresses the suboptimal sensitivity of all the available ante mortem diagnostic techniques for paratuberculosis and evaluates the effect of a recent comparative intradermal tuberculin test on the humoral diagnosis of paratuberculosis in an infected caprine herd, as a potential diagnostic strategy to maximize the detection of infected animals. The results from our study showed an increase in the quantitative response detected using an ELISA technique (booster effect), which was translated into an increase, although not statistically significant, in the number of reactors. Paratuberculosis (PTB) and tuberculosis (TB) are two mycobacterial diseases with a severe economic and health impact on domestic ruminants. The ante mortem diagnosis of PTB is hampered, among other factors, by the limited sensitivity of all the available diagnostic techniques. Since TB-infected goats subjected to the comparative intradermal tuberculin test (CITT) may experience a booster effect on their antibody titer and a potential enhancement to the sensitivity of humoral techniques for tuberculosis, in the present study we aimed to evaluate this diagnostic strategy on the humoral diagnosis of PTB in serum and milk samples collected from a caprine herd that was TB free and PTB infected. The results from 120 goats indicated a significant increase (p < 0.001) in the quantitative response detected using an ELISA technique, conducted using serum and milk samples taken 15 and 30 days after performing a CITT (day 0 of the study); although, it did not translate into a significant increase in the number of reactors during any of the testing events (0, 3,15, 30 and 60 days post-CITT). Additionally, the number of ELISA-positive animals was higher for the serum versus the milk samples at both 15 and 30 days post-CITT. The increase in the quantitative ELISA result suggested a diagnostic strategy that maximizes ELISA sensitivity, mainly using serum samples, in PTB-infected herds; although, it may depend on individual differences and the interpretation criteria. Simple Summary: Acute stress can manifest physiologically through a "stress leukogram" (neutrophilia, eosinopenia, lymphopenia, and mild monocytosis), with eosinopenia as one of its main indicators. Changes in cortisol concentrations, the primary stress biomarker, whether measured in blood or saliva, strongly correlate with the severity of gastrointestinal diseases in equids. Recently, eosinophil count has been used to identify critically ill horses and as a prognostic marker in horses with Systemic Inflammatory Response Syndrome or SIRS. However, the relationship between both parameters has not been proven. Therefore, the objective of this study was to assess the possible differences in eosinophil count in horses with acute abdominal diseases that survived and non-survived upon hospitalization and its relationship with salivary cortisol. It was demonstrated that non-surviving horses showed the lowest eosinophil count, and a strong correlation (negative) was observed between cortisol and eosinophil count. Thus, eosinophil count could be a potential prognosis biomarker in horses with acute abdominal diseases. Stress leukogram includes eosinopenia as one of its main markers (neutrophilia, eosinopenia, lymphopenia, and mild monocytosis). Cortisol is the main stress biomarker, which is also strongly correlated with the severity of gastrointestinal diseases. This study aimed to determine the relationship between salivary cortisol and the eosinophil cell count (EC) in equids with abdominal pain. To do this, 39 horses with abdominal pain referred to an emergency service were included. All samples were taken on admission, and several parameters and clinical data were included. Equids were classified according to the outcome as survivors and non-survivors. Non-surviving equids presented higher salivary cortisol concentrations (Non-Survivors: 1.580 ± 0.816 µg/dL; Survivors 0.988 ± 0.653 µg/dL; p < 0.05) and lower EC (Non-Survivors: 0.0000 × 103/µL (0.000/0.0075); Survivors: 0.0450 × 103/µL (0.010/0.1825); p < 0.01). In addition, the relationship between salivary cortisol concentration, EC, and the WBC was determined. Only a strong correlation (negative) was observed between cortisol and EC (r = −0.523, p < 0.01). Since cortisol is not an analyte that can be measured routinely in clinical settings such as emergencies, the EC could be a good alternative. Introduction: Acquired brain injury (ABI) constitutes a significant and growing global public health concern. People with ABI often face a range of physical and psychosocial challenges that span the domains of "body structure and function", "activity", and "participation", as defined by the International Classification of Functioning, Disability, and Health. Multidisciplinary approaches based on exercise therapy with social leisure activities are essential to improve physical recovery and health-related quality of life after injury. Methods: Adults with ABI, aged > 18 years, in the subacute or chronic stage (within more than one month after the injury) will be recruited through a rehabilitation center. Adults will be randomized to receive either a racket sports-based exercise therapy program combined with usual care (sET) or usual care alone (UC) using a random number sequence with a 1:1 allocation ratio. sET intervention consists of an 8-week exercise therapy program focusing on different racket sports skills, 1 h in duration, 2 days/week. It will be delivered by a physiotherapist in tailored, face-to-face, group-based sessions. Primary outcomes will be the health-related quality of life (SF-36) and upper extremity motor function (Fugl-Meyer Assessment-Upper Extremity Scale). Discussion: The study proposes an intervention that combines sports-based exercise therapy with usual care. It aims to determine whether this intervention improves the health-related quality of life and upper limb motor function in adults with ABI compared with usual care alone. Honey is considered one of the last untreated natural food substances, with a complex composition. It is produced by bees (Apis mellifera) from nectar. The glycemic index (GI) is a physiological assessment of a food's carbohydrate content via its effect on postprandial blood glucose concentrations. This study evaluated the GI and the satiety response to three Mexican types of honey administered to 26 healthy volunteers. The fructose values ranged from 272.40 g/kg to 395.10 g/kg, while the glucose value ranged from 232.20 g/kg to 355.50 g/kg. The fructose/glucose (F/G) ratio of honey was 1.45, 1.00, and 1.17 for highland, multifloral, and avocado honey, respectively. Highland and avocado honey were classified as medium-GI (69.20 ± 4.07 and 66.36 ± 5.74, respectively), while multifloral honey was classified as high-GI (74.24 ± 5.98). Highland honey presented a higher satiety values response than glucose. The difference in GI values and the satiety response effect of highland honey could be explained by its different carbohydrate composition and the possible presence of other honey components such as phytochemicals. Oleogelation is an alternative process to improve the nutritional properties of food by creating soft-matter structures with the same functionality as commercial fats (shortenings). In this study, oleogels were produced by adding organic candelilla wax at 3% (OC03), 6% (OC06), and 9% (OC09) to extra-virgin linseed oil, and then characterized by their physicochemical properties. Furthermore, the physicochemical and sensorial properties of five cookie formulations were evaluated. Organic candelilla wax influenced the oleogel formulations, giving higher values of color (L* and b*), texture, acidity index, and melting point. In the cookie formulations, the luminosity values decreased when the percentage of oleogel was increased; reddish trends were obtained (a* values) for the cookie where 70% of the fat was replaced by the oleogel (C70), and more yellow trends were obtained (b* values) for C100. The moisture content was higher in cookies with oleogels, but it was within quality limits. The percentage of fat migration was lower in cookies with a mixture of fats and oleogels. In terms of hardness, the substitution of oleogels resulted in softer cookies. In terms of the sensory evaluation, the most accepted cookie was C70. Therefore, this study demonstrates the possibility of using organic-candelilla-wax-based oleogels in a real food model rich in unsaturated fats. [ABSTRACT FROM AUTHOR]

Diffuse parenchymal lung diseases (DPLD) or Interstitial lung diseases (ILD) are a heterogeneous group of lung conditions with common characteristics that can progress to fibrosis. Within this group of pneumonias, idiopathic pulmonary fibrosis (IPF) is considered the most common. This disease has no known cause, is devastating and has no cure. Chronic lesion of alveolar type II (ATII) cells represents a key mechanism for the development of IPF. ATII cells are specialized in the biosynthesis and secretion of pulmonary surfactant (PS), a lipid-protein complex that reduces surface tension and minimizes breathing effort. Some differences in PS composition have been reported between patients with idiopathic pulmonary disease and healthy individuals, especially regarding some specific proteins in the PS; however, few reports have been conducted on the lipid components. The purpose of this research is to produce and characterize bacterial cellulose (BC) films coated with chitosan (BC-CH). BC films were produced in a fermentation medium based on Camellia sinensis tea and dextrose (12 days at 25 °C) and subsequently treated with coating-forming solutions (CFSs) based on chitosan (BC-CH 0.5%, BC-CH 1.0%, and BC-CH 1.5%). As a result, the FTIR spectra of BC and BC-CH 1.5% showed the main characteristic bands of cellulose and chitosan. In the physicochemical characterization of the films, it was found that the incorporation of the chitosan coatings did not affect the thickness; however, it decreased the luminosity (L*) and increased redness (a*), yellowness (b*), and opacity (75.24%). Additionally, the light absorption properties in the UV-Vis range were improved. Furthermore, the application of the CFSs increased: the solubility (64.91%), the antimicrobial activity against S. aureus (6.55 mm) and E. coli (8.25 mm), as well as the antioxidant activity (57.71% and 24.57% free radical scavenging activity), and the content of total phenols (2.45 mg GAE/g). Featured Application: The degradation in the Perovksite solar cells is one of the main problems to solve in order to achieve commercial devices. In this work influence of the active layer thickness in the degradation mechanisms is studied. Perovskite solar cells (PSCs) have become very popular due to the high efficiencies achieved. Nevertheless, one of the main challenges for their commercialization is to solve their instability issues. A thorough understanding of the processes taking place in the device is key for the development of this technology. Herein, J-V measurements have been performed to characterize PSCs with different active layer thicknesses. The solar cells' parameters in pristine devices show no significant dependence on the active layer thickness. However, the evolution of the solar cells' efficiency under ISOS-L1 protocol reveals a dramatic burn-in degradation, more pronounced for thicker devices. Samples were also characterized using impedance spectroscopy (IS) at different degradation stages, and data were fitted to a three RC/RCPE circuit. The low frequency capacitance in the thickest samples suffers a strong increase with time, which suggests a significant growth in the mobile ion population. This increase in the ion density partially screens the electric field, which yields a reduction in the extracted current and, consequently, the efficiency. This paper has been validated with two-dimensional numerical simulations that corroborate (i) the decrease in the internal electric field in dark conditions in 650 nm devices, and (ii) the consequent reduction in the carrier drift and, therefore, of the effective current extraction and efficiency. Visible light communication systems can be used in a wide variety of applications, from driving to home automation. The use of wearables can increase the potential applications in indoor systems to send and receive specific and customized information. We have designed and developed a fully organic and flexible Visible Light Communication system using a flexible OLED, a flexible P3HT:PCBM-based organic photodiode (OPD) and flexible PCBs for the emitter and receiver conditioning circuits. We have fabricated and characterized the I-V curve, modulation response and impedance of the flexible OPD. As emitter we have used a commercial flexible organic luminaire with dimensions 99 × 99 × 0.88 mm, and we have characterized its modulation response. All the devices show frequency responses that allow operation over 40 kHz, thus enabling the transmission of high quality audio. Finally, we integrated the emitter and receiver components and its electronic drivers, to build an all-organic flexible VLC system capable of transmitting an audio file in real-time, as a proof of concept of the indoor capabilities of such a system. Humanitarian organizations have a crucial role in response and relief efforts after floods. The effectiveness of disaster response is contingent on accurate and timely information regarding the location, timing and impacts of the event. Here we show how two near-real-time data sources, satellite observations of water coverage and flood-related social media activity from Twitter, can be used to support rapid disaster response, using case-studies in the Philippines and Pakistan. For these countries we analyze information from disaster response organizations, the Global Flood Detection System (GFDS) satellite flood signal, and flood-related Twitter activity analysis. The results demonstrate that these sources of near-real-time information can be used to gain a quicker understanding of the location, the timing, as well as the causes and impacts of floods. In terms of location, we produce daily impact maps based on both satellite information and social media, which can dynamically and rapidly outline the affected area during a disaster. In terms of timing, the results show that GFDS and/or Twitter signals flagging ongoing or upcoming flooding are regularly available one to several days before the event was reported to humanitarian organizations. In terms of event understanding, we show that both GFDS and social media can be used to detect and understand unexpected or controversial flood events, for example due to the sudden opening of hydropower dams or the breaching of flood protection. The performance of the GFDS and Twitter data for early detection and location mapping is mixed, depending on specific hydrological circumstances (GFDS) and social media penetration (Twitter). Further research is needed to improve the interpretation of the GFDS signal in different situations, and to improve the pre-processing of social media data for operational use. Early flood warning and real-time monitoring systems play a key role in flood risk reduction and disaster response decisions. Global-scale flood forecasting and satellite-based flood detection systems are currently operating, however their reliability for decision-making applications needs to be assessed. In this study, we performed comparative evaluations of several operational global flood forecasting and flood detection systems, using 10 major flood events recorded over 2012-2014. Specifically, we evaluated the spatial extent and temporal characteristics of flood detections from the Global Flood Detection System (GFDS) and the Global Flood Awareness System (GloFAS). Furthermore, we compared the GFDS flood maps with those from NASA's two Moderate Resolution Imaging Spectroradiometer (MODIS) sensors. Results reveal that: (1) general agreement was found between the GFDS and MODIS flood detection systems, (2) large differences exist in the spatio-temporal characteristics of the GFDS detections and GloFAS forecasts, and (3) the quantitative validation of global flood disasters in data-sparse regions is highly challenging. Overall, satellite remote sensing provides useful near real-time flood information that can be useful for risk management. We highlight the known limitations of global flood detection and forecasting systems, and propose ways forward to improve the reliability of large-scale flood monitoring tools. A visible light communication (VLC) system using an organic bulk heterojunction photodetector (OPD) is presented. The system has been successfully proven indoors with an audio signal. The emitter consists of three commercial high-power white LEDs connected in parallel. The receiver is based on an organic photodetector having as active layer a blend of poly(3-hexylthiophene) (P3HT) and phenyl C61-butyric acid methyl ester (PCBM). Lung cancer (LC) and chronic obstructive pulmonary disease (COPD) commonly coexist in smokers, and the presence of COPD increases the risk of developing LC. Cigarette smoke causes oxidative stress and an inflammatory response in lung cells, which in turn may be involved in COPD and lung cancer development. The aim of this study was to identify differential proteomic profiles related to oxidative stress response that were potentially involved in these two pathological entities. Protein content was assessed in the bronchoalveolar lavage (BAL) of 60 patients classified in four groups: COPD, COPD and LC, LC, and control (neither COPD nor LC). Proteins were separated into spots by two dimensional polyacrylamide gel electrophoresis (2D-PAGE) and examined by matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF/TOF). A total of 16 oxidative stress regulatory proteins were differentially expressed in BAL samples from LC and/or COPD patients as compared with the control group. A distinct proteomic reactive oxygen species (ROS) protein signature emerged that characterized lung cancer and COPD. In conclusion, our findings highlight the role of the oxidative stress response proteins in the pathogenic pathways of both diseases, and provide new candidate biomarkers and predictive tools for LC and COPD diagnosis. Simple Summary: Obesity and its associated complications, such as metabolic syndrome, are an increasing problem in both humans and horses in the developed world. Adipose tissue is a key endocrine organ that communicates with other organs by multiple endocrine substances called adipokines. There is evidence to suggest that adipokines may contribute to the regulation of biological processes, such as metabolism, immunity, and inflammation. The aim of this study was to investigate the usefulness of one of these adipokines in horses, resistin, and its relationship with insulin dysregulation (ID) and inflammation. Seventy-two horses, included in one of the four following groups, were studied: healthy controls, horses with inflammatory conditions, horses with mild, and horses with severe ID. Plasma resistin concentrations were significantly different between groups, and the highest values were recorded in the inflammatory and severe ID groups. The lack of correlation of resistin with basal insulin concentration and the significant correlation of resistin with the inflammatory marker serum amyloid A suggest that, as is the case in humans, plasma resistin concentrations in horses are predominantly related to inflammatory conditions and not to ID. Obesity and its associated complications, such as metabolic syndrome, are an increasing problem in both humans and horses in the developed world. The expression patterns of resistin differ considerably between species. In rodents, resistin is expressed by adipocytes and is related to obesity and ID. In humans, resistin is predominantly produced by inflammatory cells, and resistin concentrations do not reflect the degree of obesity, although they may predict cardiovascular outcomes. The aim of this study was to investigate the usefulness of resistin and its relationship with ID and selected indicators of inflammation in horses. Seventy-two horses, included in one of the four following groups, were studied: healthy controls (C, n = 14), horses with inflammatory conditions (I, n = 21), horses with mild ID (ID1, n = 18), and horses with severe ID (ID2, n = 19). Plasma resistin concentrations were significantly different between groups and the higher values were recorded in the I and ID2 groups (C: 2.38 ± 1.69 ng/mL; I: 6.85 ± 8.38 ng/mL; ID1: 2.41 ± 2.70 ng/mL; ID2: 4.49 ± 3.08 ng/mL). Plasma resistin was not correlated with basal insulin concentrations. A significant (r = 0.336, p = 0.002) correlation was found between resistin and serum amyloid A. Our results show that, as is the case in humans, plasma resistin concentrations in horses are predominantly related to inflammatory conditions and not to ID. Remotely sensed flood extents obtained in near real-time can be used for emergency flood incident management and as observations for assimilation into flood forecasting models. High-resolution synthetic aperture radar (SAR) sensors have the potential to detect flood extents in urban areas through clouds during both day- and night-time. This paper considers a method for detecting flooding in urban areas by merging near real-time SAR flood extents with model-derived flood hazard maps. This allows a two-way symbiosis, whereby currently available SAR urban flood extent improves future model flood predictions, while flood hazard maps obtained after the SAR overpasses improve the SAR estimate of urban flood extents. The method estimates urban flooding using SAR backscatter only in rural areas adjacent to urban ones. It was compared to an existing method using SAR returns in both rural and urban areas. The method using SAR solely in rural areas gave an average flood detection accuracy of 94% and a false positive rate of 9% in the urban areas and was more accurate than the existing method. The study objective was to assess the role of badgers as a reservoir of tuberculosis for cattle and other sympatric wild species in the region. During the follow-up, 27/639 badgers (4.23%) were positive for the Mycobacterium tuberculosis complex based on bacterial isolation, while 160/673 (23.77%) were positive based on P22 ELISA. Badger infection was spatially and temporally associated with cattle herd infection. The present work investigated the prevalence, spatial distribution, and temporal distribution of tuberculosis (TB) in free-ranging Eurasian badgers (Meles meles) and cattle in Asturias (Atlantic Spain) during a 13-year follow-up. The study objective was to assess the role of badgers as a TB reservoir for cattle and other sympatric wild species in the region. Between 2008 and 2020, 673 badgers (98 trapped and 575 killed in road traffic accidents) in Asturias were necropsied, and their tissue samples were cultured for the Mycobacterium tuberculosis complex (MTC) isolation. Serum samples were tested in an in-house indirect P22 ELISA to detect antibodies against the MTC. In parallel, data on MTC isolation and single intradermal tuberculin test results were extracted for cattle that were tested and culled as part of the Spanish National Program for the Eradication of Bovine TB. A total of 27/639 badgers (4.23%) were positive for MTC based on bacterial isolation, while 160/673 badgers (23.77%) were found to be positive with the P22 ELISA. The rate of seropositivity was higher among adult badgers than subadults. Badger TB status was spatially and temporally associated with cattle TB status. Our results cannot determine the direction of possible interspecies transmission, but they are consistent with the idea that the two hosts may exert infection pressure on each other. Over the last decades, climate change has triggered an increase in the frequency of spruce bark beetle (Ips typographus L.) in Central Europe. More than 50% of forests in the Czech Republic are seriously threatened by this pest, leading to high ecological and economic losses. The exponential increase of bark beetle infestation hinders the implementation of costly field campaigns to prevent and mitigate its effects. Remote sensing may help to overcome such limitations as it provides frequent and spatially continuous data on vegetation condition. Using Sentinel-2 images as main input, two models have been developed to test the ability of this data source to map bark beetle damage and severity. All models were based on a change detection approach, and required the generation of previous forest mask and dominant species maps. The first damage mapping model was developed for 2019 and 2020, and it was based on bi-temporal regressions in spruce areas to estimate forest vitality and bark beetle damage. A second model was developed for 2020 considering all forest area, but excluding clear-cuts and completely dead areas, in order to map only changes in stands dominated by alive trees. The three products were validated with in situ data. All the maps showed high accuracies (acc > 0.80). Accuracy was higher than 0.95 and F1-score was higher than 0.88 for areas with high severity, with omission errors under 0.09 in all cases. This confirmed the ability of all the models to detect bark beetle attack at the last phases. Areas with no damage or low severity showed more complex results. The no damage category yielded greater commission errors and relative bias (CEs = 0.30–0.42, relB = 0.42–0.51). The similar results obtained for 2020 leaving out clear-cuts and dead trees proved that the proposed methods could be used to help forest managers fight bark beetle pests. Eco-hydrological models can be used to support effective land management and planning of forest resources. These models require a Digital Elevation Model (DEM), in order to accurately represent the morphological surface and to simulate catchment responses. This is particularly relevant on low altimetry catchments, where a high resolution DEM can result in a more accurate representation of terrain morphology (e.g., slope, flow direction), and therefore a better prediction of hydrological responses. This work intended to use Soil and Water Assessment Tool (SWAT) to assess the influence of DEM resolutions (1 m, 10 m and 30 m) on the accuracy of catchment representations and hydrological responses on a low relief forest catchment with a dry and hot summer Mediterranean climate. The catchment responses were simulated using independent SWAT models built up using three DEMs. These resolutions resulted in marked differences regarding the total number of channels, their length as well as the hierarchy. Model performance was increasingly improved using fine resolutions DEM, revealing a bR2 (0.87, 0.85 and 0.85), NSE (0.84, 0.67 and 0.60) and Pbias (−14.1, −27.0 and −38.7), respectively, for 1 m, 10 m and 30 m resolutions. The spatial and temporal dynamics of the forest cover can be captured using remote sensing data. Forest masks are a valuable tool to monitor forest characteristics, such as biomass, deforestation, health condition and disturbances. This study was carried out under the umbrella of the EC H2020 MySustainableForest (MSF) project. A key achievement has been the development of supervised classification methods for delineating forest cover. The forest masks presented here are binary forest/non-forest classification maps obtained using Sentinel-2 data for 16 study areas across Europe with different forest types. Performance metrics can be selected to measure accuracy of forest mask. However, large-scale reference datasets are scarce and typically cannot be considered as ground truth. In this study, we implemented a stratified random sampling system and the generation of a reference dataset based on visual interpretation of satellite images. This dataset was used for validation of the forest masks, MSF and two other similar products: HRL by Copernicus and FNF by the DLR. MSF forest masks showed a good performance (OAMSF = 96.3%; DCMSF = 96.5), with high overall accuracy (88.7–99.5%) across all the areas, and omission and commission errors were low and balanced (OEMSF = 2.4%; CEMSF = 4.5%; relBMSF = 2%), while the other products showed on average lower accuracies (OAHRL = 89.2%; OAFNF = 76%). However, for all three products, the Mediterranean areas were challenging to model, where the complexity of forest structure led to relatively high omission errors (OEMSF = 9.5%; OEHRL = 59.5%; OEFNF = 71.4%). Comparing these results with the vision from external local stakeholders highlighted the need of establishing clear large-scale validation datasets and protocols for remote sensing-based forest products. The aim of this research was to develop and characterize five gelled double emulsions based on chia mucilage (CM) and different biopolymers (κ-carrageenan, C; locust bean gum, L; thixogum, T; and whey protein concentrate, W) loaded with green tea extract (GTE). Gelled double emulsions consisted of W1 (whey-protein-concentrate/sodium-azide/NaCl/GTE)/O and (PGPR/canola-oi)/W2 (CM, CMC, CML, CMT and CMW), and were characterized based on physicochemical properties during 35 days of storage. Optical microscopy clearly showed the drops of the internal phase surrounded by droplets of oil dispersed in the second aqueous phase; the droplet size was higher for CMT and lowest for CMW. In addition, all emulsions were highly stable at creaming and were effective in reducing the loss of antioxidant activity (88.82%) and total phenols (64.26%) during storage; CMT, CML and CM were the most effective. Furthermore, all emulsions showed a protective effect by modulating the release of the GTE in a simulated gastrointestinal environment, allowing a controlled release during the gastric-intestinal digestion phases and reaching its maximum release in the intestinal phase (64.57–83.31%). This investigation reports on a new national model to evaluate the effectiveness of catchment sensitive farming in England, and how pollution mitigation measures have improved water quality between 2006 and 2016. An adapted HYPE (HYdrological Predictions for the Environment) model was written to use accurate farm emissions data so that the pathway impact could be accounted for in the land phase of transport. Farm emissions were apportioned into different runoff fractions simulated in surface and soil layers, and travel time and losses were taken into account. These were derived from the regulator's 'catchment change matrix' and converted to monthly load time series, combined with extensive point source load datasets. Very large flow and water quality monitoring datasets were used to calibrate the model nationally for flow, nitrogen, phosphorus, suspended sediments and faecal indicator organisms. The model was simulated with and without estimated changes to farm emissions resulting from catchment measures, and spatial and temporal changes to water quality concentrations were then assessed. Lung surfactant is a complex mixture of phospholipids and specific proteins but its role in the pathogenesis of interstitial lung diseases is not established. Herein, we analyzed the effects of three representative phospholipid components, that is, dipalmitoilphosphatidylcoline (DPPC), phosphatidylglycerol (PG) and phosphatidylethanolamine (PE), on collagen expression, apoptosis and Ca2+ signaling in normal human lung fibroblasts (NHLF) and probed their effect in an experimental model of lung fibrosis. Collagen expression was measured with RT-PCR, apoptosis was measured by using either the APOPercentage assay kit (Biocolor Ltd., Northern Ireland, UK) or the Caspase-Glo 3/7 assay (Promega, Madison, WI, USA) and Ca2+ signaling by conventional epifluorescence imaging. The effect in vivo was tested in bleomycin-induced lung fibrosis in mice. DPPC and PG did not affect collagen expression, which was downregulated by PE. Furthermore, PE promoted apoptosis and induced a dose-dependent Ca2+ signal. PE-induced Ca2+ signal and apoptosis were both blocked by phospholipase C, endoplasmic reticulum pump and store-operated Ca2+ entry inhibition. PE-induced decrease in collagen expression was attenuated by blocking phospholipase C. Finally, surfactant enriched with PE and PE itself attenuated bleomycin-induced lung fibrosis and decreased the soluble collagen concentration in mice lungs. This study demonstrates that PE strongly contributes to the surfactant-induced inhibition of collagen expression in NHLF through a Ca2+ signal and that early administration of Beractant enriched with PE diminishes lung fibrosis in vivo. Diffuse parenchymal lung diseases (DPLD) or Interstitial lung diseases (ILD) are a heterogeneous group of lung conditions with common characteristics that can progress to fibrosis. Within this group of pneumonias, idiopathic pulmonary fibrosis (IPF) is considered the most common. This disease has no known cause, is devastating and has no cure. Chronic lesion of alveolar type II (ATII) cells represents a key mechanism for the development of IPF. ATII cells are specialized in the biosynthesis and secretion of pulmonary surfactant (PS), a lipid-protein complex that reduces surface tension and minimizes breathing effort. Some differences in PS composition have been reported between patients with idiopathic pulmonary disease and healthy individuals, especially regarding some specific proteins in the PS; however, few reports have been conducted on the lipid components. This review focuses on the mechanisms by which phospholipids (PLs) could be involved in the development of the fibroproliferative response.

This study with 40 office workers investigated (a) the effect of time spent standing on low- back and lower limb pain during a 1-h laboratory-based task; (b) the standing time after which a significant increase in pain is likely; and (c) the individual, physical and psychosocial factors that predict pain. The primary outcome was bodily location of pain and pain intensity on a 100-mm Visual Analogue Scale recorded at baseline and every 15 min. Physical measures included trunk and hip motor control and endurance. Self-report history of pain, physical activity, psychosocial job characteristics, pain catastrophizing and general health status were collected. Univariate analysis and regression models were included. The prevalence of low-back pain increased from 15% to 40% after 30 min while feet pain increased to 25% from 0 at baseline. The intensity of low-back and lower limb pain also increased over time. A thirty-minute interval was identified as the threshold for the development and increase in low-back and feet pain. Modifiable factors were associated with low-back pain intensity-lower hip abductor muscle endurance and poorer physical health, and with feet symptoms-greater body mass index and less core stability.

This study examines demographic, physical and psychosocial factors associated with an increase in low back pain (LBP) during a one-hour standing task. A cross-sectional survey with 40 office workers was conducted. The primary outcome was pain severity during a one-hour standing task recorded every 15 min using a 100 mm Visual Analogue Scale (VAS). Participants were defined as pain developers (PD), if they reported a change in pain of ≥10 mm from baseline, or non-pain developers (NPD). Physical outcomes included participant-rated and examiner-rated trunk and hip motor control and endurance. Self-report history of LBP, physical activity, psychosocial job characteristics, general health and pain catastrophising were collected. Fourteen participants were PD. Hip abduction, abdominal and spinal muscle endurance was lower for PD ( p ≤ 0.05). PD had greater self-reported difficulty performing active hip abduction and active straight leg raise tests ( p ≤ 0.04). Those reporting a lifetime, 12 month or 7-day history of LBP ( p < 0.05) and lower self-reported physical function ( p = 0.01) were more likely to develop LBP during the standing task. In conclusion, a history of LBP, reduced trunk and hip muscle endurance and deficits in lumbopelvic/hip motor control may be important to consider in office workers experiencing standing-induced LBP.
